Abstract

Polysaccharides, extracted from Lycium barbarum (LBPs), display a wide variety of biological activities. However, the neuroprotective effects of LBPs remain poorly understood. The present study aimed to investigate the inhibitory effects of LBPs on production of lipopolysaccharide (LPS)--induced proinflammatory mediators in BV2 microglia. Our data showed that LPS induced the activation of NFκB and its upstream caspase 3 and upregulated the expression of another apoptosis-inducing factor, HSP60 in BV2 microglia cells and increased the release of TNF-α and HSP60 in culture media. After LBPs treatment, activated NFκB and caspase 3 were significantly suppressed. The enhanced expression of HSP60 was reduced and LPS induced release of TNF-α and HSP60 were inhibited. These results suggest that LBPs may offer substantial therapeutic potential for treatment of neurodegenerative diseases that are accompanied by microglia activation. [ABSTRACT FROM AUTHOR]
