- Abstract
Metastatic disease is a major source of cancer morbidity and mortality. Understanding the molecular basis of cancer metastasis will have a considerable impact on cancer therapy. We now understand that uncontrolled cell signaling is of great importance in oncogenesis and tumor progression. A major role in the assembly of signaling proteins into biochemical pathways and networks is played by adaptor proteins. In this chapter we describe specifically the role of these proteins in the process of tumor metastasis and the potential for cancer therapy. Historically considered undruggable targets, protein-protein interactions and adaptor proteins are emerging as rational and viable targets, as exemplified by the development of selective antagonist of the adaptor protein Grb2. Targeting adaptor proteins and cellular miswiring is an emerging and exciting cancer therapeutic opportunity. [ABSTRACT FROM AUTHOR]
