Summary: Background: The Global Program to Eliminate Lymphatic Filariasis (GPELF) is the largest public health program based on mass drug administration (MDA). Despite decades of MDA, ongoing transmission in some countries remains a challenge. To optimise interventions, it is critical to differentiate between recrudescence and new infections. Since adult filariae are inaccessible in humans, deriving a method that relies on the offspring microfilariae (mf) is necessary. Methods: We developed a genome amplification and kinship analysis-based approach using Brugia malayi samples from gerbils, and applied it to analyse Wuchereria bancrofti mf from humans in Côte d’Ivoire. We examined the pre-treatment genetic diversity in 269 mf collected from 18 participants, and further analysed 1-year post-treatment samples of 74 mf from 4 participants. Hemizygosity of the male X-chromosome allowed for direct inference of haplotypes, facilitating robust maternal parentage inference. To enrich parasite DNA from samples contaminated with host DNA, a whole-exome capture panel was created for W. bancrofti. Findings: By reconstructing and temporally tracking sibling relationships across pre- and post-treatment samples, we differentiated between new and established maternal families, suggesting reinfection in one participant and recrudescence in three participants. The estimated number of reproductively active adult females ranged between 3 and 11 in the studied participants. Population structure analysis revealed genetically distinct parasites in Côte d’Ivoire compared to samples from other countries. Exome capture identified protein-coding variants with ∼95% genotype concordance rate. Interpretation: We have generated resources to facilitate the development of molecular genetic tools that can estimate adult worm burdens and monitor parasite populations, thus providing essential information for the successful implementation of GPELF. Parasitic gastrointestinal nematodes pose significant health risks to humans, livestock, and companion animals, and their control relies heavily on the use of anthelmintic drugs. Overuse of these drugs has led to the emergence of resistant nematode populations. Herein, a naturally occurring isolate (referred to as BCR) of the dog hookworm, Ancylostoma caninum, that is resistant to 3 major classes of anthelmintics is characterized. Various drug assays were used to determine the resistance of BCR to thiabendazole, ivermectin, moxidectin and pyrantel pamoate. When compared to a drug-susceptible isolate of A. caninum, BCR was shown to be significantly resistant to all 4 of the drugs tested. Multiple single nucleotide polymorphisms have been shown to impart benzimidazole resistance, including the F167Y mutation in the β-tubulin isotype 1 gene, which was confirmed to be present in BCR through molecular analysis. The frequency of the resistant allele in BCR was 76.3% following its first passage in the lab, which represented an increase from approximately 50% in the founding hookworm population. A second, recently described mutation in codon 134 (Q134H) was also detected at lower frequency in the BCR population. Additionally, BCR exhibits an altered larval activation phenotype compared to the susceptible isolate, suggesting differences in the signalling pathways involved in the activation process which may be associated with resistance. Further characterization of this isolate will provide insights into the mechanisms of resistance to macrocyclic lactones and tetrahydropyrimidine anthelmintics.

We propose an overall procedure for measuring and unfolding fast neutron spectra using a trans-stilbene scintillation detector. Detector characterization was described, including the information on energy calibration, detector resolution, and nonproportionality response. The digital charge comparison method was used for the investigation of neutron-gamma Pulse Shape Discrimination (PSD). A pair of values of 600 ns pulse width and 24 ns delay time was found as the optimized conditions for PSD. A fitting technique was introduced to increase the trans-stilbene Proton Response Function (PRF) by 28% based on comparison of the simulated and experimental electron-equivalent distributions by the Cf-252 source. The detector response matrix was constructed by Monte-Carlo simulation and the spectrum unfolding was implemented using the iterative Bayesian method. The unfolding of simulated and measured spectra of Cf-252 and AmBe neutron sources indicates reliable, stable and no-bias results. The unfolding technique was also validated by the measured cosmic-ray induced neutron flux. Our approach is promising for fast neutron detection and spectroscopy.

Background & Aims: Dyskeratosis congenita (DC) is a telomere biology disorder caused primarily by mutations in the DKC1 gene. Patients with DC and related telomeropathies resulting from premature telomere dysfunction experience multiorgan failure. In the liver, DC patients present with nodular hyperplasia, steatosis, inflammation, and cirrhosis. However, the mechanism responsible for telomere dysfunction–induced liver disease remains unclear. Methods: We used isogenic human induced pluripotent stem cells (iPSCs) harboring a causal DC mutation in DKC1 or a CRISPR/Cas9 (clustered regularly interspaced short palindromic repeats/Cas9)–corrected control allele to model DC liver pathologies. We differentiated these iPSCs into hepatocytes (HEPs) or hepatic stellate cells (HSCs) followed by generation of genotype-admixed hepatostellate organoids. Single-cell transcriptomics were applied to hepatostellate organoids to understand cell type–specific genotype-phenotype relationships. Results: Directed differentiation of iPSCs into HEPs and stellate cells and subsequent hepatostellate organoid formation revealed a dominant phenotype in the parenchyma, with DC HEPs becoming hyperplastic and also eliciting a pathogenic hyperplastic, proinflammatory response in stellate cells independent of stellate cell genotype. Pathogenic phenotypes in DKC1-mutant HEPs and hepatostellate organoids could be rescued via suppression of serine/threonine kinase AKT (protein kinase B) activity, a central regulator of MYC-driven hyperplasia downstream of DKC1 mutation. Conclusions: Isogenic iPSC-derived admixed hepatostellate organoids offer insight into the liver pathologies in telomeropathies and provide a framework for evaluating emerging therapies.

On 2022 September 26, the Double Asteroid Redirection Test (DART) spacecraft impacted Dimorphos, the satellite of binary near-Earth asteroid (65803) Didymos. This demonstrated the efficacy of a kinetic impactor for planetary defense by changing the orbital period of Dimorphos by 33 minutes. Measuring the period change relied heavily on a coordinated campaign of lightcurve photometry designed to detect mutual events (occultations and eclipses) as a direct probe of the satellite’s orbital period. A total of 28 telescopes contributed 224 individual lightcurves during the impact apparition from 2022 July to 2023 February. We focus here on decomposable lightcurves, i.e., those from which mutual events could be extracted. We describe our process of lightcurve decomposition and use that to release the full data set for future analysis. We leverage these data to place constraints on the postimpact evolution of ejecta. The measured depths of mutual events relative to models showed that the ejecta became optically thin within the first ∼1 day after impact and then faded with a decay time of about 25 days. The bulk magnitude of the system showed that ejecta no longer contributed measurable brightness enhancement after about 20 days postimpact. This bulk photometric behavior was not well represented by an HG photometric model. An HG _1 G _2 model did fit the data well across a wide range of phase angles. Lastly, we note the presence of an ejecta tail through at least 2023 March. Its persistence implied ongoing escape of ejecta from the system many months after DART impact.

The Korean Pathfinder Lunar Orbiter (KPLO)-MAGnetometer (KMAG) consists of three triaxial fluxgate sensors (MAG1, MAG2, and MAG3) that measure the magnetic field around the Moon. The three sensors are mounted in the order MAG3, MAG2, and MAG1 inside a 1.2 m long boom, away from the satellite body. Before it arrived on the Moon, we compared the magnetic field measurements taken by DSCOVR and KPLO in solar wind to verify the measurement performance of the KMAG instrument. We found that there were artificial disturbances in the KMAG measurement data, such as step-like and spike-like disturbances, which were produced by the spacecraft body. To remove spacecraft-generated disturbances, we applied a multi-sensor method, employing the gradiometer technique and principal component analysis, using KMAG magnetic field data, and confirmed the successful elimination of spacecraft-generated disturbances. In the future, the proposed multi-sensor method is expected to clean the magnetic field data measured onboard the KPLO from the lunar orbit.

Abstract The objective of this retrospective study is to verify the effectiveness and safety of patient-specific titanium implants on maxillofacial bones, with a long-term follow-up. Total 16 patients with various maxillofacial defects underwent reconstruction using patient-specific titanium implants. Titanium implants, manufactured by electron beam melting, selective laser sintering, or milling, were inserted into the maxilla, mandible, or zygoma. Long-term follow‐up (36.7 ± 20.1 months) was conducted after the surgery. Bone fusion of the titanium implant body, postoperative infection, implant malunion, functional results, patient satisfaction, subsidence, osteolysis around the implants, and complications were recorded and analyzed at the last follow-up. Of the 28 implants, only one failed to unite with the bone; therefore, revision surgery was performed. No osteolysis or subsidence around the titanium implants nor adverse events were observed; the mean VAS score for satisfaction was 9. All patients enrolled in this trial were esthetically and functionally satisfied with their surgical results, and fixation failure and esthetic dissatisfaction complications were well resolved. Patient-specific titanium showed satisfactory outcomes when used to treat various oral and maxillofacial defects. A 3D printed titanium implant can be effectively used in the reconstruction of the zygoma and mandible instead of autogenous bone without donor site morbidity.

Onchocerciasis is a neglected tropical disease targeted for elimination using ivermectin mass administration. Ivermectin kills the microfilariae and temporarily arrests microfilariae production by the macrofilariae. We genotyped 436 microfilariae from 10 people each in Ituri, Democratic Republic of the Congo (DRC), and Maridi County, South Sudan, collected before and 4–5 months after ivermectin treatment. Population genetic analyses identified 52 and 103 mitochondrial DNA haplotypes among the microfilariae from DRC and South Sudan, respectively, with few haplotypes shared between people. The percentage of genotype-based correct assignment to person within DRC was ~88% and within South Sudan ~64%. Rarefaction and extrapolation analysis showed that the genetic diversity in DRC, and even more so in South Sudan, was captured incompletely. The results indicate that the per-person adult worm burden is likely higher in South Sudan than DRC. Analyses of haplotype data from a subsample (n = 4) did not discriminate genetically between pre- and post-treatment microfilariae, confirming that post-treatment microfilariae are not the result of new infections. With appropriate sampling, mitochondrial haplotype analysis could help monitor changes in the number of macrofilariae in a population as a result of treatment, identify cases of potential treatment failure, and detect new infections as an indicator of continuing transmission.

Abstract Objectives The purpose of this study was to investigate the condylar morphology and position of Koreans using cone-beam computed tomography (CBCT) images. Analyzing the mean values of this study with reference to left and right sides, gender, and age will help to understand the size of the condyle and glenoid fossa, condylar morphology, and temporomandibular joint (TMJ) symmetry for establishing the standard temporomandibular joint structures of Koreans and then design the standard temporomandibular joint prosthesis for Koreans. Results There was no significant result in the condyle size, condyle axis angle, joint space, fossa depth, and mandibular body size between the left and right sides (p > 0.05). On the other hand, the mediolateral width of the condyle and mandibular body size show significantly different with the gender (P < 0.05). Also, significant differences were found in condyle size, joint space, fossa depth, and mandibular body size according to age groups (p < 0.05). Conclusions Condylar position and morphology vary according to side, age, and gender. The results of this study are expected to help in customizing a treatment for the patients who need TMJ reconstruction by predicting the TMJ morphology according to age and gender and design the standard temporomandibular joint prosthesis for the Koreans.

BackgroundLymphatic filariasis (LF) is a neglected tropical disease caused by the filarial nematodes Wuchereria bancrofti, Brugia malayi and Brugia timori. The Global Program to Eliminate LF uses mass drug administration (MDA) of anti-filarial drugs that clear microfilariae (Mf) from blood to interrupt transmission by mosquitos. New diagnostic tools are needed to assess the impact of MDA on bancroftian filariasis, because available serologic tests can remain positive after successful treatment.Methodology/principal findingsWe identified Wb-bhp-1, which encodes a W. bancrofti homologue of BmR1, the B. malayi protein used in the Brugia Rapid antibody test for brugian filariasis. Wb-bhp-1 has a single exon that encodes a 16.3 kD protein (Wb-Bhp-1) with 45% amino acid identity to BmR1. Immunohistology shows that anti-Wb-Bhp-1 antibodies primarily bind to Mf. Plasma from 124 of 224 (55%) microfilaremic individuals had IgG4 antibodies to Wb-Bhp-1 by ELISA. Serologic reactivity to Wb-Bhp-1 varied widely with samples from different regions (sensitivity range 32-92%), with 77% sensitivity for 116 samples collected from microfilaremic individuals outside of sub-Saharan Africa. This variable sensitivity highlights the importance of validating new diagnostic tests for parasitic diseases with samples from different geographical regions. Individuals with higher Mf counts were more likely to have anti-Wb-Bhp-1 antibodies. Cross-reactivity was observed with a minority of plasma samples from people with onchocerciasis (17%) or loiasis (10%). We also identified, cloned and characterized BmR1 homologues from O. volvulus and L. loa that have 41% and 38% identity to BmR1, respectively. However, antibody assays with these antigens were not sensitive for onchocerciasis or loiasis.ConclusionsWb-Bhp-1 is a novel antigen that is useful for serologic diagnosis of bancroftian filariasis. Additional studies are needed to assess the value of this antigen for monitoring the success of filariasis elimination programs.

From the first phase of the high-cadence Formation of the Outer Solar System: an Icy Legacy (FOSSIL) survey, we analyzed lightcurves, ranging from one to four nights in length, of 371 trans-Neptunian objects (TNOs) for periodicity. We found 29 TNOs with periodic lightcurves, one of which is a good candidate for a close/contact binary. Another of the periodic FOSSIL TNOs could potentially have the fastest of all known TNO spin rates, with a period of 1.3 hr. We do not have total confidence in the period and thus plan to obtain a more detailed lightcurve for confirmation. The periodic TNOs have an average rotation period of 11.2 hr, close to the value obtained by Alexandersen et al., which had similar cadence, but different from other surveys. In regards to contention in the literature about whether smaller TNOs are more irregular in shape and thus have larger lightcurve amplitudes, we found that there is a weak correlation between absolute magnitude and lightcurve amplitude in a subset of 194 FOSSIL TNOs, even when using the more appropriate brightest (minimum) absolute magnitude instead of the time-averaged value.

The dynamic host-parasite mechanisms underlying hookworm infection establishment and maintenance in mammalian hosts remain poorly understood but are primarily mediated by hookworm’s excretory/secretory products (ESPs), which have a wide spectrum of biological functions. We used ultra-high performance mass spectrometry to comprehensively profile and compare female and male ESPs from the zoonotic human hookworm Ancylostoma ceylanicum, which is a natural parasite of dogs, cats, and humans. We improved the genome annotation, decreasing the number of protein-coding genes by 49% while improving completeness from 92 to 96%. Compared to the previous genome annotation, we detected 11% and 10% more spectra in female and male ESPs, respectively, using this improved version, identifying a total of 795 ESPs (70% in both sexes, with the remaining sex-specific). Using functional databases (KEGG, GO and Interpro), common and sex-specific enriched functions were identified. Comparisons with the exclusively human-infective hookworm Necator americanus identified species-specific and conserved ESPs. This is the first study identifying ESPs from female and male A. ceylanicum. The findings provide a deeper understanding of hookworm protein functions that assure long-term host survival and facilitate future engineering of transgenic hookworms and analysis of regulatory elements mediating the high-level expression of ESPs. Furthermore, the findings expand the list of potential vaccine and diagnostic targets and identify biologics that can be explored for anti-inflammatory potential.

RNA sequencing (RNA-Seq) and mass-spectrometry-based proteomics data are often integrated in proteogenomic studies to assist in the prediction of eukaryote genome features, such as genes, splicing, single-nucleotide (SNVs), and single-amino-acid variants (SAAVs). Most genomes of parasite nematodes are draft versions that lack transcript- and protein-level information and whose gene annotations rely only on computational predictions. Angiostrongylus costaricensis is a roundworm species that causes an intestinal inflammatory disease, known as abdominal angiostrongyliasis (AA). Currently, there is no drug available that acts directly on this parasite, mostly due to the sparse understanding of its molecular characteristics. The available genome of A. costaricensis, specific to the Costa Rica strain, is a draft version that is not supported by transcript- or protein-level evidence. This study used RNA-Seq and MS/MS data to perform an in-depth annotation of the A. costaricensis genome. Our prediction improved the reference annotation with (a) novel coding and non-coding genes; (b) pieces of evidence of alternative splicing generating new proteoforms; and (c) a list of SNVs between the Brazilian (Crissiumal) and the Costa Rica strain. To the best of our knowledge, this is the first time that a multi-omics approach has been used to improve the genome annotation of A. costaricensis. We hope this improved genome annotation can assist in the future development of drugs, kits, and vaccines to treat, diagnose, and prevent AA caused by either the Brazil strain (Crissiumal) or the Costa Rica strain.

Triclabendazole (TCBZ) resistance is an emerging problem in fascioliasis that is not well understood. Studies including small numbers of parasites fail to capture the complexity of susceptibility variations between and within Fasciolahepatica populations. As the first step to studying the complex resistant phenotype–genotype associations, we characterized a large sample of adult F. hepatica with diverging TCBZ susceptibility. We collected parasites from naturally infected livestock slaughtered in the Cusco and Cajamarca regions of Peru. These parasites were exposed to TCBZ sulfoxide (TCBZ.SO) in vitro to determine their susceptibility. We used a motility score to determine the parasite’s viability. We titrated drug concentrations and times to detect 20% non-viable (susceptible conditions) or 80% non-viable (resistant conditions) parasites. We exposed 3348 fully motile parasites to susceptible (n = 1565) or resistant (n = 1783) conditions. Three hundred and forty-one (21.8%) were classified as susceptible and 462 (25.9%) were classified as resistant. More resistant parasites were found in Cusco than in Cajamarca (p < 0.001). Resistant parasites varied by slaughterhouse (p < 0.001), month of the year (p = 0.008), fluke length (p = 0.016), and year of collection (p < 0.001). The in vitro susceptibility to TCBZ.SO in wildtype F. hepatica was associated with geography, season, and morphometry.

PURPOSE The purpose of this study was to analysis the effective short foot exercise methods and verify the effect of short foot exercise (SFE) on medial longitudinal arch (MLA) and balance in order to utilize it as foundation data for clinical fields. METHODS In this study, research papers were collected by using RISS, KISS and PubMed as a search term for SFE, foot intrinsic muscle exercise (FIME) and MLA. RESULTS A total of 30 research were gathered, and 16 papers of data were related effect of SFE, and 9 papers was related to effective exercise methods, and 5 papers was related to a comparison with other FIME. The collected data were classified into the influence of SFE on the MLA, balance, and the efficient exercise methods. CONCLUSIONS The result of this study confirmed that SFE was very effective to maintain MLA and improve foot stability, dynamic balance and gait. In addition to, imagery training and biofeedback with SFE was efficient methods, and SFE applied with weight bearing, or windlass effect, or 30° ankle dorsiflexion strengthened abductor halluces muscle effectively. Therefore, SFE will be useful for rehabilitation of pes planus patients or athletes.

Wolbachia are endosymbionts of numerous arthropod and some nematode species, are important for their development and if present can cause distinct phenotypes of their hosts. Prophage DNA has been frequently detected in Wolbachia, but particles of Wolbachia bacteriophages (phage WO) have been only occasionally isolated. Here, we report the characterization and isolation of a phage WO of the southern ground cricket, Allonemobius socius, and provided the first whole-genome sequence of phage WO from this arthropod family outside of Asia. We screened A. socius abdomen DNA extracts from a cricket population in eastern Missouri by quantitative PCR for Wolbachia surface protein and phage WO capsid protein and found a prevalence of 55% and 50%, respectively, with many crickets positive for both. Immunohistochemistry using antibodies against Wolbachia surface protein showed many Wolbachia clusters in the reproductive system of female crickets. Whole-genome sequencing using Oxford Nanopore MinION and Illumina technology allowed for the assembly of a high-quality, 55 kb phage genome containing 63 open reading frames (ORF) encoding for phage WO structural proteins and host lysis and transcriptional manipulation. Taxonomically important regions of the assembled phage genome were validated by Sanger sequencing of PCR amplicons. Analysis of the nucleotides sequences of the ORFs encoding the large terminase subunit (ORF2) and minor capsid (ORF7) frequently used for phage WO phylogenetics showed highest homology to phage WOAu of Drosophila simulans (94.46% identity) and WOCin2USA1 of the cherry fruit fly, Rhagoletis cingulata (99.33% identity), respectively. Transmission electron microscopy examination of cricket ovaries showed a high density of phage particles within Wolbachia cells. Isolation of phage WO revealed particles characterized by 40-62 nm diameter heads and up to 190 nm long tails. This study provides the first detailed description and genomic characterization of phage WO from North America that is easily accessible in a widely distributed cricket species.

Haemonchus contortus and Teladorsagia circumcincta are among the two most pathogenic internal parasitic nematodes infecting small ruminants, such as sheep and goats, and are a global animal health issue. Accurate identification and delineation of Haemonchidae species is essential for development of diagnostic and control strategies with high resolution for Trichostrongyloidea infection in ruminants. Here, we describe in detail and compare the complete mitochondrial (mt) genomes of the New Zealand H. contortus and T. circumcincta field strains to improve our understanding of species- and strain-level evolution in these closely related roundworms. In the present study, we performed extensive comparative bioinformatics analyses on the recently sequenced complete mt genomes of the New Zealand H. contortus NZ_Hco_NP and T. circumcincta NZ_Teci_NP field strains. Amino acid sequences inferred from individual genes of each of the two mt genomes were compared, concatenated and subjected to phylogenetic analysis using Bayesian inference (BI), Maximum Likelihood (ML), and Maximum Parsimony (MP). The AT-rich mt genomes of H. contortus NZ_Hco_NP and T. circumcincta NZ_Teci_NP are 14,001 bp (A+T content of 77.4%) and 14,081 bp (A+T content of 77.3%) in size, respectively. All 36 of the typical nematode mt genes are transcribed in the forward direction in both species and comprise of 12 protein-encoding genes (PCGs), 2 ribosomal RNA (rrn) genes, and 22 transfer RNA (trn) genes. The secondary structures for the 22 trn genes and two rrn genes differ between H. contortus NZ_Hco_NP and T. circumcincta NZ_Teci_NP, however the gene arrangements of both are consistent with other Trichostrongylidea sequenced to date. Comparative analyses of the complete mitochondrial nucleotide sequences, PCGs, A+T rich and non-coding repeat regions of H. contortus NZ_Hco_NP and T. circumcincta NZ_Teci_NP further reinforces the high levels of diversity and gene flow observed among Trichostrongylidea, and supports their potential as ideal markers for strain-level identification from different hosts and geographical regions with high resolution for future studies. The complete mt genomes of H. contortus NZ_Hco_NP and T. circumcincta NZ_Teci_NP presented here provide useful novel markers for further studies of the meta-population connectivity and the genetic mechanisms driving evolution in nematode species.

Flumazenil, a gamma-aminobutyric acid receptor antagonist, can promote arousal even under general anesthesia without the use of benzodiazepines. We hypothesized that flumazenil could promote arousal and reduce emergence agitation in patients undergoing orthognathic surgery with sevoflurane anesthesia. One hundred and two patients were randomly allocated to the control or flumazenil group. Saline or flumazenil was administered at the end of the surgery. The incidence of emergence agitation was measured by using Aono’s four-point scale, with scores of 3 and 4 indicating emergence agitation. The primary outcome was the incidence of emergence agitation. Secondary outcomes included duration of emergence agitation and time intervals between the discontinuation of anesthetics, first response, extubation, and post-anesthesia care-unit discharge readiness. The incidence of emergence agitation was 58.3% and 38.9% in the control and flumazenil groups, respectively, but it was not statistically significant. However, the duration of emergence agitation was shorter in the flumazenil group (p = 0.012). There were no significant differences in the time intervals between the discontinuation of anesthetics, first response, and extubation. Although flumazenil did not reduce the incidence of emergence agitation in patients undergoing orthognathic surgery with sevoflurane anesthesia, it can be considered as an option for awakening patients in terms of improving emergence profiles.

The purpose of this study is to present a novel maxillary sinus ventilation drainage (MSVD) device which facilitates blood drainage and nasal breathing after Le Fort I osteotomy. One hundred patients who underwent bimaxillary orthognathic surgery from January 2016 to June 2016 at the Department of Oral and Maxillofacial Surgery, Chung-Ang University Hospital were retrospectively selected and divided into two groups. MSVD was applied in 50 patients, who were allocated to the MSVD group, while the remaining 50 patients, in whom MSVD was not applied, were allocated to the non-MSVD group. All patients underwent a cone-beam computed tomography (CBCT) scan before and 2 days after surgery. CBCT was used to analyze middle meatus patency and the percentage of hematoma volume per entire maxillary sinus volume. Statistical comparisons between the two groups were performed using the Chi-squared and Mann–Whitney U tests to investigate the clinical effectiveness of MSVD. The MSVD group showed significantly higher maintenance ratio of the middle meatus patency and a higher percentage of maxillary sinus air volume (p < 0.05) than the non-MSVD group. MSVD facilitated nasal breathing after Le Fort I osteotomy by reducing hematoma inside the maxillary sinus and promoting middle meatal patency.

Antibiotic treatment has emerged as a promising strategy to sterilize and kill filarial nematodes due to their dependence on their endosymbiotic bacteria, Wolbachia. Several studies have shown that novel and FDA-approved antibiotics are efficacious at depleting the filarial nematodes of their endosymbiont, thus reducing female fecundity. However, it remains unclear if antibiotics can permanently deplete Wolbachia and cause sterility for the lifespan of the adult worms. Concerns about resistance arising from mass drug administration necessitate a careful exploration of potential Wolbachia recrudescence. In the present study, we investigated the long-term effects of the FDA-approved antibiotic, rifampicin, in the Brugia pahangi jird model of infection. Initially, rifampicin treatment depleted Wolbachia in adult worms and simultaneously impaired female worm fecundity. However, during an 8-month washout period, Wolbachia titers rebounded and embryogenesis returned to normal. Genome sequence analyses of Wolbachia revealed that despite the population bottleneck and recovery, no genetic changes occurred that could account for the rebound. Clusters of densely packed Wolbachia within the worm's ovarian tissues were observed by confocal microscopy and remained in worms treated with rifampicin, suggesting that they may serve as privileged sites that allow Wolbachia to persist in worms while treated with antibiotic. To our knowledge, these clusters have not been previously described and may be the source of the Wolbachia rebound.

The human hookworm Necator americanus infects more than 400 million people worldwide, contributing substantially to the poverty in these regions. Adult stage N. americanus live in the small intestine of the human host where they inject excretory/secretory (ES) products into the mucosa. ES products have been characterized at the proteome level for a number of animal hookworm species, but until now, the difficulty in obtaining sufficient live N. americanus has been an obstacle in characterizing the secretome of this important human pathogen. Herein we describe the ES proteome of N. americanus and utilize this information along with RNA Seq data to conduct the first proteogenomic analysis of a parasitic helminth, significantly improving the available genome and thereby generating a robust description of the parasite secretome. The genome annotation resulted in a revised prediction of 3,425 fewer genes than initially reported, accompanied by a significant increase in the number of exons and introns, total gene length and the percentage of the genome covered by genes. Almost 200 ES proteins were identified by LC-MS/MS with SCP/TAPS proteins, 'hypothetical' proteins and proteases among the most abundant families. These proteins were compared to commonly used model species of human parasitic infections, including Ancylostoma caninum, Nippostrongylus brasiliensis and Heligmosomoides polygyrus. SCP/TAPS proteins are immunogenic in nematode infections, so we expressed four of those identified in this study in recombinant form and showed that they are all recognized to varying degrees by serum antibodies from hookworm-infected subjects from a disease-endemic area of Brazil. Our findings provide valuable information on important families of proteins with both known and unknown functions that could be instrumental in host-parasite interactions, including protein families that might be key for parasite survival in the onslaught of robust immune responses, as well as vaccine and diagnostic targets.

Pregnane X receptor (PXR) is a key nuclear receptor that mediates drug metabolism and stimulates hepatocyte proliferation. However, the lack of PXR expression in human pluripotent stem cell-derived hepatocytes limits their application for drug screening and toxicity testing. Here, we generated a PXR-mCherry reporter human induced pluripotent stem cell (hiPSC) line using the CRISPR/Cas9 system. PXR-mCherry hiPSCs were pluripotent and had differentiation potential and a normal karyotype. This cell line is an important tool for identifying factors that increase PXR-mediated drug metabolism and hepatocyte proliferation.

A lunar vehicle radiation dosimeter (LVRAD) has been proposed for studying the radiation environment on the lunar surface and evaluating its impact on human health. The LVRAD payload comprises four systems: a particle dosimeter and spectrometer (PDS), a tissue-equivalent dosimeter, a fast neutron spectrometer, and an epithermal neutron spectrometer. A silicon photodiode sensor with compact readout electronics was proposed for the PDS. The PDS system aims to measure protons with 10–100 MeV of energy and assess dose in the lunar space environment. The manufactured silicon photodiode sensor has an effective area of 20 mm × 20 mm and thickness of 650 μm; the electronics consist of an amplifier, analog pulse processor, and a 12-bit analog-to-digital converter for signal readout. We studied the responses of silicon sensors which were manufactured with self-made electronics to gamma rays with a wide range of energies and proton beams.

In porous titanium scaffolds manufactured via 3D printing, the differences in bone formation according to pore design and implantation period were studied. Titanium scaffolds with three types of different pore structures (Octadense, Gyroid, and Dode) were fabricated via 3D printing using the selective laser melting method. Mechanical properties of scaffolds were investigated. Prepared specimens were inserted into both femurs of nine rabbits and their clinical characteristics were observed. Three animals were sacrificed at the 2nd, 4th, and 6th weeks, and the differences in bone formation were radiologically and histologically analyzed. The percentage of new bone and surface density in the pore structure were observed to be approximately 25% and 8 mm2/mm3, respectively. There was no difference in the amount of newly formed bone according to the pore design at 2, 4, and 6 weeks. In addition, no differences in the amount of newly formed bone were observed with increasing time within the same pore design for all three designs. During the 6-week observation period, the proportion of new bones in the 3D-printed titanium scaffold was approximately 25%. Differences in bone formation according to the pore design or implantation period were not observed.

Abstract Background Two-jaw surgery including mandibular and maxillary backward movement procedures are commonly performed to correct class III malocclusion. Bimaxillary surgery can reposition the maxillofacial bone together with soft tissue, such as the soft palate and the tongue base. We analyzed changes of pharyngeal airway narrowing to ascertain clinical correlations with the prevalence of snoring after two-jaw surgery. Methods A prospective clinical study was designed including a survey on snoring and three-dimensional (3D) computed tomography (CT) in class III malocclusion subjects before and after bimaxillary surgery. We conducted an analysis on changes of the posterior pharyngeal space find out clinical correlations with the prevalence of snoring. Results Among 67 subjects, 12 subjects complained about snoring 5 weeks after the surgical correction, and examining the 12 subjects after 6 months, 6 patients complained about the snoring. The current findings demonstrated the attenuation of the largest transverse width (LTW), anteroposterior length (APL), and cross-sectional area (CSA) following bimaxillary surgery given to class III malocclusion patients, particularly at the retropalatal level. The average distance of maxillary posterior movements were measured to be relatively higher (horizontal distance 3.9 mm, vertical distance 2.6 mm) in case of new snorers. Conclusions This study found that bimaxillary surgery could lead to the narrowing of upper airway at the retropalatal or retroglossal level as well as triggering snoring in subjects with class III malocclusion. Based on the current clinical findings, we also found that upper airway narrowing at retropalatal level may contribute to increasing the probability of snoring and that polysonography may need to be performed before orthognathic surgery in subjects with class III malocclusion.

Purpose – This study aims to analyze the relationship between global value chain (GVC) participation and poverty levels. Additionally, it investigates the impact of education levels, specifically analyzing literacy rates and tertiary education rates, on the correlation between GVC participation and poverty in Organization for Economic Co-operation and Development (OECD) and Association of Southeast Asian Nations (ASEAN) countries. Design/methodology/approach – Fixed effect and random effect models will be employed to quantify the relationships between the dependent and independent variables. The Hausman test is applied to determine the appropriate estimator between fixed and random effects. Also, in the model, time-fixed effect or two-way fixed effect has been used to control for unobserved heterogeneity both across entities and over time in panel data analysis. Findings – The findings demonstrate that engagement in GVCs presents a promising avenue for stimulating development, advancing income per capita growth and facilitating job creation. Notably, the results illuminate that the poverty-alleviating impacts of GVC participation are most conspicuous in nations boasting elevated levels of educational attainment among their populace. Originality/value – This research aims to promote a better understanding of the connection between GVC participation and the level of poverty, with GVC participation decomposed into forward participation and backward participation.

As described in the previous paper (Park et al. 2013), the detector subsystem of optical wide-field patrol (OWL) provides many observational data points of a single artificial satellite or space debris in the form of small streaks, using a chopper system and a time tagger. The position and the corresponding time data are matched assuming that the length of a streak on the CCD frame is proportional to the time duration of the exposure during which the chopper blades do not obscure the CCD window. In the previous study, however, the length was measured using the diagonal of the rectangle of the image area containing the streak; the results were quite ambiguous and inaccurate, allowing possible matching error of positions and time data. Furthermore, because only one (position, time) data point is created from one streak, the efficiency of the observation decreases. To define the length of a streak correctly, it is important to locate the endpoints of a streak. In this paper, a method using a differential convolution mask pattern is tested. This method can be used to obtain the positions where the pixel values are changed sharply. These endpoints can be regarded as directly detected positional data, and the number of data points is doubled by this result.

The correlation between meteorological data collected at the optical wide-field patrol network (OWL-Net) Station No. 1 and the seeing of satellite optical observation data was analyzed. Meteorological data and satellite optical observation data from June 2014 to November 2015 were analyzed. The analyzed meteorological data were the outdoor air temperature, relative humidity, wind speed, and cloud index data, and the analyzed satellite optical observation data were the seeing full-width at half-maximum (FWHM) data. The annual meteorological pattern for Mongolia was analyzed by collecting meteorological data over four seasons, with data collection beginning after the installation and initial set-up of the OWL-Net Station No. 1 in Mongolia. A comparison of the meteorological data and the seeing of the satellite optical observation data showed that the seeing degrades as the wind strength increases and as the cloud cover decreases. This finding is explained by the bias effect, which is caused by the fact that the number of images taken on the less cloudy days was relatively small. The seeing FWHM showed no clear correlation with either temperature or relative humidity.

Inactive space objects are usually rotating and tumbling as a result of internal or external forces. KOREASAT 1 has been inactive since 2005, and its drift trajectory has been monitored with the optical wide-field patrol network (OWL-Net). However, a quantitative analysis of KOREASAT 1 in regard to the attitude evolution has never been performed. Here, two optical tracking systems were used to acquire raw measurements to analyze the rotation period of two inactive satellites. During the optical campaign in 2013, KOREASAT 1 was observed by a 0.6 m class optical telescope operated by the Korea Astronomy and Space Science Institute (KASI). The rotation period of KOREASAT 1 was analyzed with the light curves from the photometry results. The rotation periods of the low Earth orbit (LEO) satellite ASTRO-H after break-up were detected by OWL-Net on April 7, 2016. We analyzed the magnitude variation of each satellite by differential photometry and made comparisons with the star catalog. The illumination effect caused by the phase angle between the Sun and the target satellite was corrected with the system tool kit (STK) and two line element (TLE) technique. Finally, we determined the rotation period of two inactive satellites on LEO and geostationary Earth orbit (GEO) with light curves from the photometry. The main rotation periods were determined to be 5.2 sec for ASTRO-H and 74 sec for KOREASAT 1.

Several optical monitoring strategies by a ground-based telescope to protect a Geostationary Earth Orbit (GEO) satellite from collisions with close approaching objects were investigated. Geostationary Transfer Orbit (GTO) objects, Inclined GeoSynchronous Orbit (IGSO) objects, and drifted GEO objects forced by natural perturbations are hazardous to operational GEO satellites regarding issues related to close approaches. The status of these objects was analyzed on the basis of their orbital characteristics in Two-Line Element (TLE) data from the Joint Space Operation Center (JSpOC). We confirmed the conjunction probability with all catalogued objects for the domestic operational GEO satellite, Communication, Ocean and Meteorological Satellite (COMS) using the Conjunction Analysis Tools by Analytical Graphics, Inc (AGI). The longitudinal drift rates of GeoSynchronous Orbit (GSO) objects were calculated, with an analytic method and they were confirmed using the Systems Tool Kit by AGI. The required monitoring area was determined from the expected drift duration and inclination of the simulated target. The optical monitoring strategy for the target area was analyzed through the orbit determination accuracy. For this purpose, the close approach of Russian satellite Raduga 1-7 to Korean COMS in 2011 was selected.

By using the Optical Wide-field Patrol (OWL) network developed by the Korea Astronomy and Space Science Institute (KASI) we generated the right ascension and declination angle data from optical observation of Low Earth Orbit (LEO) satellites. We performed an analysis to verify the optimum number of observations needed per arc for successful estimation of orbit. The currently functioning OWL observatories are located in Daejeon (South Korea), Songino (Mongolia), and Oukaïmeden (Morocco). The Daejeon Observatory is functioning as a test bed. In this study, the observed targets were Gravity Probe B, COSMOS 1455, COSMOS 1726, COSMOS 2428, SEASAT 1, ATV-5, and CryoSat-2 (all in LEO). These satellites were observed from the test bed and the Songino Observatory of the OWL network during 21 nights in 2014 and 2015. After we estimated the orbit from systematically selected sets of observation points (20, 50, 100, and 150) for each pass, we compared the difference between the orbit estimates for each case, and the Two Line Element set (TLE) from the Joint Space Operation Center (JSpOC). Then, we determined the average of the difference and selected the optimal observation points by comparing the average values.

As a governmentally approved domestic entity for Space Situational Awareness, Korea Astronomy and Space Science Institute (KASI) is developing and operating an optical telescopes system, Optical Wide-field PatroL (OWL) Network. During the test phase of this system, it is necessary to determine the range of brightness of the observable satellites. We have defined standard magnitude for Low Earth Orbit (LEO) satellites to calibrate their luminosity in terms of standard parameters such as distance, phase angle, and angular rate. In this work, we report the optical brightness range of five LEO Satellites using OWL-Net.

We estimated the orbit of the Communication, Ocean and Meteorological Satellite (COMS), a Geostationary Earth Orbit (GEO) satellite, through data from actual optical observations using telescopes at the Sobaeksan Optical Astronomy Observatory (SOAO) of the Korea Astronomy and Space Science Institute (KASI), Optical Wide field Patrol (OWL) at KASI, and the Chungbuk National University Observatory (CNUO) from August 1, 2014, to January 13, 2015. The astrometric data of the satellite were extracted from the World Coordinate System (WCS) in the obtained images, and geometrically distorted errors were corrected. To handle the optically observed data, corrections were made for the observation time, light-travel time delay, shutter speed delay, and aberration. For final product, the sequential filter within the Orbit Determination Tool Kit (ODTK) was used for orbit estimation based on the results of optical observation. In addition, a comparative analysis was conducted between the precise orbit from the ephemeris of the COMS maintained by the satellite operator and the results of orbit estimation using optical observation. The orbits estimated in simulation agree with those estimated with actual optical observation data. The error in the results using optical observation data decreased with increasing number of observatories. Our results are useful for optimizing observation data for orbit estimation.

An algorithm to automatically extract coordinate and time information from optical observation data of geostationary orbit satellites (GEO satellites) or geosynchronous orbit satellites (GOS satellites) is developed. The optical wide-field patrol system is capable of automatic observation using a pre-arranged schedule. Therefore, if this type of automatic analysis algorithm is available, daily unmanned monitoring of GEO satellites can be possible. For data acquisition for development, the COMS1 satellite was observed with 1-s exposure time and 1-m interval. The images were grouped and processed in terms of “action”, and each action was composed of six or nine successive images. First, a reference image with the best quality in one action was selected. Next, the rest of the images in the action were geometrically transformed to fit in the horizontal coordinate system (expressed in azimuthal angle and elevation) of the reference image. Then, these images were median-combined to retain only the possible non-moving GEO candidates. By reverting the coordinate transformation of the positions of these GEO satellite candidates, the final coordinates could be calculated.

A strategy is needed for a regional survey of geosynchronous orbits (GSOs) to monitor known space objects and detect uncataloged space objects. On the basis of the Inter-Agency Debris Committee’s recommendation regarding the protected region of geosynchronous Earth orbit (GEO), target satellites with perigee and apogee of GEO ± 200 km and various inclinations are selected for analysis. The status of the GSO region was analyzed using the satellite distribution based on the orbital characteristics in publicly available two-line element data. Natural perturbation effects cause inactive satellites to drift to two stable longitudinal points. Active satellites usually maintain the designed positions as a result of regular or irregular maneuver operations against their natural drift. To analyze the detection rate of a single optical telescope, 152 out of 412 active satellites and 135 out of 288 inactive satellites in the GSO region were selected on the basis of their visibility at the observation site in Daejeon, Korea. By using various vertical view ranges and various numbers of observations of the GSO region, the detection efficiencies were analyzed for a single night, and the numbers of follow-up observations were determined. The orbital estimation accuracies were also checked using the arc length and number of observed data points to maintain the GSO satellite catalog.

Preventive chemotherapy has long been practiced against nematode parasites of livestock, leading to widespread drug resistance, and is increasingly being adopted for eradication of human parasitic nematodes even though it is similarly likely to lead to drug resistance. Given that the genetic architecture of resistance is poorly understood for any nematode, we have analyzed multidrug resistant Teladorsagia circumcincta, a major parasite of sheep, as a model for analysis of resistance selection. We introgressed a field-derived multiresistant genotype into a partially inbred susceptible genetic background (through repeated backcrossing and drug selection) and performed genome-wide scans in the backcross progeny and drug-selected F2 populations to identify the major genes responsible for the multidrug resistance. We identified variation linking candidate resistance genes to each drug class. Putative mechanisms included target site polymorphism, changes in likely regulatory regions and copy number variation in efflux transporters. This work elucidates the genetic architecture of multiple anthelmintic resistance in a parasitic nematode for the first time and establishes a framework for future studies of anthelmintic resistance in nematode parasites of humans.

Food borne trematodes (FBTs) are an assemblage of platyhelminth parasites transmitted through the food chain, four of which are recognized as neglected tropical diseases (NTDs). Fascioliasis stands out among the other NTDs due to its broad and significant impact on both human and animal health, as Fasciola sp., are also considered major pathogens of domesticated ruminants. Here we present a reference genome sequence of the common liver fluke, Fasciola hepatica isolated from sheep, complementing previously reported isolate from cattle. A total of 14,642 genes were predicted from the 1.14 GB genome of the liver fluke. Comparative genomics indicated that F. hepatica Oregon and related food-borne trematodes are metabolically less constrained than schistosomes and cestodes, taking advantage of the richer millieux offered by the hepatobiliary organs. Protease families differentially expanded between diverse trematodes may facilitate migration and survival within the heterogeneous environments and niches within the mammalian host. Surprisingly, the sequencing of Oregon and Uruguay F. hepatica isolates led to the first discovery of an endobacteria in this species. Two contigs from the F. hepatica Oregon assembly were joined to complete the 859,205 bp genome of a novel Neorickettsia endobacterium (nFh) closely related to the etiological agents of human Sennetsu and Potomac horse fevers. Immunohistochemical studies targeting a Neorickettsia surface protein found nFh in specific organs and tissues of the adult trematode including the female reproductive tract, eggs, the Mehlis' gland, seminal vesicle, and oral suckers, suggesting putative routes for fluke-to-fluke and fluke-to-host transmission. The genomes of F. hepatica and nFh will serve as a resource for further exploration of the biology of F. hepatica, and specifically its newly discovered trans-kingdom interaction with nFh and the impact of both species on disease in ruminants and humans.

The detector subsystem of the Optical Wide-field Patrol (OWL) network efficiently acquires the position and time information of moving objects such as artificial satellites through its chopper system, which consists of 4 blades in front of the CCD camera. Using this system, it is possible to get more position data with the same exposure time by changing the streaks of the moving objects into many pieces with the fast rotating blades during sidereal tracking. At the same time, the time data from the rotating chopper can be acquired by the time tagger connected to the photo diode. To analyze the orbits of the targets detected in the image data of such a system, a sequential procedure of determining the positions of separated streak lines was developed that involved calculating the World Coordinate System (WCS) solution to transform the positions into equatorial coordinate systems, and finally combining the time log records from the time tagger with the transformed position data. We introduce this procedure and the preliminary results of the application of this procedure to the test observation images.

An optical tracking system has advantages for observing geostationary earth orbit (GEO) satellites relatively over other types of observation system. Regular surveying for unidentified space objects with the optical tracking system can be an early warning tool for the safety of five Korean active GEO satellites. Two strategies of positioning on the observed image of Communication, Ocean and Meteorological Satellite 1 are tested and compared. Photometric method has a half root mean square error against streak method. Also null eccentricity method for initial orbit determination (IOD) is tested with simulation data and real observation data. Under 10 minutes observation time interval, null eccentricity method shows relatively better IOD results than the other time interval. For follow-up observation of unidentified space objects, at least two consecutive observations are needed in 5 minutes to determine orbit for geosynchronous orbit space objects.

The objectives of this study are to analyze the satellite visibility at the randomly established ground sites, to determine the five optimal ground sites to perform the optical surveillance and tracking of domestic satellites, and to verify the acquisition of the optical observation time sufficient to maintain the precise ephemeris at optimal ground sites that have been already determined. In order to accomplish these objectives, we analyzed the visibility for sun-synchronous orbit satellites, low earth orbit satellites, middle earth orbit satellites and domestic satellites as well as the continuous visibility along with the fictitious satellite ground track, and calculate the effective visibility. For the analysis, we carried out a series of repetitive process using the satellite tool kit simulation software developed by Analytical Graphics Incorporated. The lighting states of the penumbra and direct sun were set as the key constraints of the optical observation. The minimum of the observation satellite elevation angle was set to be 20 degree, whereas the maximum of the sun elevation angle was set to be -10 degree which is within the range of the nautical twilight. To select the candidates for the optimal optical observation, the entire globe was divided into 84 sectors in a constant interval, the visibility characteristics of the individual sectors were analyzed, and 17 ground sites were arbitrarily selected and analyzed further. Finally, five optimal ground sites (Khurel Togoot Observatory, Assy-Turgen Observatory, Tubitak National Observatory, Bisdee Tier Optical Astronomy Observatory, and South Africa Astronomical Observatory) were determined. The total observation period was decided as one year. To examine the seasonal variation, the simulation was performed for the period of three days or less with respect to spring, summer, fall and winter. In conclusion, we decided the optimal ground sites to perform the optical surveillance and tracking of domestic satellites and verified that optical observation time sufficient to maintain the precise ephemeris could be acquired at the determined observatories.

Optical observation system provides angle-only measurement for orbit determination of space object. Range measurement can be directly acquired using laser ranging or tone ranging system. Initial orbit determination (IOD) by using angle- only data set shows discrepancy according to the measurement time interval. To solve this problem, range measurement data should be added for IOD. In this study, two-site optical observation was used to derive the range information. We have observed nine geostationary earth orbit satellites by using two-site optical observation system. The determination result of the range shows the accuracy over 99.5% compared to the results from the satellite tool kit simulation. And we confirmed that the orbit determination by the Herrick-Gibbs method with the range information obtained from the two-site observation is more accurate than the orbit determination by Gauss method with the one-site observation. For more accurate two-site optical observation, a baseline should satisfy an optimal condition of length and more precise observation system needed.

We modified the optical system of 500 mm wide-field telescope of which point spread function showed an irregularity. The telescope has been operated for Near Earth Space Survey (NESS) located at Siding Spring Observatory (SSO) in Australia, and the optical system was brought back to Korea in January 2008. After performing a numerical simulation with the tested value of surface figure error of the primary mirror using optical design program, we found that the surface figure error of the mirror should be fabricated less than root mean square (RMS) λ/10 in order to obtain a stellar full width at half maximum (FWHM) below 28 μm. However, we started to figure the mirror for the target value of RMS λ/20, because system surface figure error would be increased by the error induced by the optical axis adjustment, mirror cell installation, and others. The radius of curvature of the primary mirror was 1,946 mm after the correction. Its measured surface figure error was less than RMS λ/20 on the table of polishing machine, and RMS λ/15 after installation in the primary mirror cell. A test observation performed at Daeduk Observatory at Korea Astronomy and Space Science Institute by utilizing the exiting mount, and resulted in 39.8 μm of stellar FWHM. It was larger than the value from numerical simulation, and showed wing-shaped stellar image. It turned out that the measured-curvature of the secondary mirror, 1,820 mm, was not the same as the designed one, 1,795.977 mm. We fabricated the secondary mirror to the designed value, and finally obtained a stellar FWHM of 27 μm after re-installation of the optical system into SSO NESS Observatory in Australia.

Human pluripotent stem cell-derived hepatocytes have the potential to provide in vitro model systems for drug discovery and hepatotoxicity testing. However, these cells are currently unsuitable for drug toxicity and efficacy testing because of their limited expression of genes encoding drug-metabolizing enzymes, especially cytochrome P450 (CYP) enzymes. Transcript levels of major CYP genes were much lower in human embryonic stem cell-derived hepatocytes (hESC-Hep) than in human primary hepatocytes (hPH). To verify the mechanism underlying this reduced expression of CYP genes, including CYP1A1, CYP1A2, CYP1B1, CYP2D6, and CYP2E1, we investigated their epigenetic regulation in terms of DNA methylation and histone modifications in hESC-Hep and hPH. CpG islands of CYP genes were hypermethylated in hESC-Hep, whereas they had an open chromatin structure, as represented by hypomethylation of CpG sites and permissive histone modifications, in hPH. Inhibition of DNA methyltransferases (DNMTs) during hepatic maturation induced demethylation of the CpG sites of CYP1A1 and CYP1A2, leading to the up-regulation of their transcription. Combinatorial inhibition of DNMTs and histone deacetylases (HDACs) increased the transcript levels of CYP1A1, CYP1A2, CYP1B1, and CYP2D6. Our findings suggest that limited expression of CYP genes in hESC-Hep is modulated by epigenetic regulatory factors such as DNMTs and HDACs.

Parasite biology, by its very nature, cannot be understood without integrating it with that of the host, nor can the host response be adequately explained without considering the activity of the parasite. However, due to experimental limitations, molecular studies of parasite-host systems have been predominantly one-sided investigations focusing on either of the partners involved. Here, we conducted a dual RNA-seq time course analysis of filarial worm parasite and host mosquito to better understand the parasite processes underlying development in and interaction with the host tissue, from the establishment of infection to the development of infective-stage larva.Using the Brugia malayi-Aedes aegypti system, we report parasite gene transcription dynamics, which exhibited a highly ordered developmental program consisting of a series of cyclical and state-transitioning temporal patterns. In addition, we contextualized these parasite data in relation to the concurrent dynamics of the host transcriptome. Comparative analyses using uninfected tissues and different host strains revealed the influence of parasite development on host gene transcription as well as the influence of the host environment on parasite gene transcription. We also critically evaluated the life-cycle transcriptome of B. malayi by comparing developmental stages in the mosquito relative to those in the mammalian host, providing insight into gene expression changes underpinning the mosquito-borne parasitic lifestyle of this heteroxenous parasite.The data presented herein provide the research community with information to design wet lab experiments and select candidates for future study to more fully dissect the whole set of molecular interactions of both organisms in this mosquito-filarial worm symbiotic relationship. Furthermore, characterization of the transcriptional program over the complete life cycle of the parasite, including stages within the mosquito, could help devise novel targets for control strategies.

An energy storage system (ESS) in a power system facilitates tasks such as renewable integration, peak shaving, and the use of ancillary services. Among the various functions of an ESS, this study focused on frequency regulation (or secondary reserve). This paper presents an optimal scheduling algorithm for frequency regulation by an ESS. This algorithm determines the bidding capacity and base point of an ESS in each operational period to achieve the maximum profit within a stable state-of-charge (SOC) range. However, the charging/discharging efficiency of an ESS causes SOC errors whenever the ESS performs frequency regulation. With an increase in SOC errors, the ESS cannot respond to an automatic generation control (AGC) signal. This situation results in low ESS performance scores, and finally, the ESS is disqualified from performing frequency regulation. This paper also presents a real-time SOC management algorithm aimed at solving the SOC error problem in real-time operations. This algorithm compensates for SOC errors by changing the base point of the ESS. The optimal scheduling algorithm is implemented in MATLAB by using the particle swarm optimization (PSO) method. In addition, changes in the SOC when the ESS performs frequency regulation in a real-time operation are confirmed using the PSCAD/EMTDC tool. The simulation results show that the optimal scheduling algorithm manages the SOC more efficiently than a commonly employed planning method. In addition, the proposed real-time SOC management algorithm is confirmed to be capable of performing SOC recovery.

We present the results of a model calculation of the stellar distribution in a UV and centered at 2175Å corresponding to the well-known bump in the interstellar extinction curve. The stellar distribution model used here is based on the Bahcall-Soneira galaxy model (1980). The source code for model calculation was designed by Brosch (1991) and modified to investigate various designing factors for UV satellite payload. The model predicts UV stellar densities in different sky directions, and its results are compared with the TD-1 star counts for a number of sky regions. From this study, we can determine the field of view, size of optics, angular resolution, and number of stars in one orbit. There will provide the basic constrains in designing a satellite payload for UV observations.

In order to derive the UV color - effective temperature relation, we calculated the effective temperature and ultraviolet color index for about 118 IUE standard stars. Using differential correction method, the effective temperature was obtained by the best fittings of IUE low-dispersion spectra to the Kurucz model (1993), and the UV color index was calculated from the magnitude differences of the UV wavelength between 2750 and 2950 . The temperature determined by the Kurucz model fittings was consistent with that obtained by other investigators (Malagnini & Morossi 1990, Malagnini et al. 1986), except for the low temperature stars. The relations between UV color - effective temperature also was similar to that in Kurucz model.

Color indexes and effective temperatures for 25 standard stars have been determined as a pilot project which show a relationship between color index and effective temperature in ultraviolet region. The effective temperature was determined by comparing energy distribution curves derived from the IUE low dispersion spectra with Kurucz atmosphere model. The UV color index was deduced by integrating fluxes in 300A interval of the IUE low dispersion spectra. The relation between color index and effective temperature in ultraviolet is similar with that of optical region.

Developing intervention strategies for the control of parasitic nematodes continues to be a significant challenge. Genomic and post-genomic approaches play an increasingly important role for providing fundamental molecular information about these parasites, thus enhancing basic as well as translational research. Here we report a comprehensive genome-wide survey of the developmental transcriptome of the human filarial parasite Brugia malayi.Using deep sequencing, we profiled the transcriptome of eggs and embryos, immature (≤3 days of age) and mature microfilariae (MF), third- and fourth-stage larvae (L3 and L4), and adult male and female worms. Comparative analysis across these stages provided a detailed overview of the molecular repertoires that define and differentiate distinct lifecycle stages of the parasite. Genome-wide assessment of the overall transcriptional variability indicated that the cuticle collagen family and those implicated in molting exhibit noticeably dynamic stage-dependent patterns. Of particular interest was the identification of genes displaying sex-biased or germline-enriched profiles due to their potential involvement in reproductive processes. The study also revealed discrete transcriptional changes during larval development, namely those accompanying the maturation of MF and the L3 to L4 transition that are vital in establishing successful infection in mosquito vectors and vertebrate hosts, respectively.Characterization of the transcriptional program of the parasite's lifecycle is an important step toward understanding the developmental processes required for the infectious cycle. We find that the transcriptional program has a number of stage-specific pathways activated during worm development. In addition to advancing our understanding of transcriptome dynamics, these data will aid in the study of genome structure and organization by facilitating the identification of novel transcribed elements and splice variants.
