Developing orally administered pediatric formulations presents significant challenges due to the unique characteristics of pediatric patients. Terbinafine hydrochloride (TER), a powerful antifungal agent, is effective against various fungal infections, including Tinea capitis, which is common in children. However, its low aqueous solubility necessitates innovative pharmaceutical strategies to enhance its effectiveness. This study describes a rational approach to selecting suitable carriers, approved for use in children, to increase the apparent solubility of TER and to guide the development of amorphous solid dispersions containing this drug. Assessments of solubility parameters, equilibrium solubility measurements, and calculations of pediatric dose numbers guided formulation development using theoretical and experimental methodologies. Carriers like Plasdone S-360 ULTRA®, HPMCAS L, and Soluplus® demonstrated favorable solubility parameter values with TER, indicating potential for drug solubilization. The solubility of TER was strongly dependent on pH. In buffer pH 6.5 containing 10% (w/v) of Soluplus®, TER presented the highest solubility value. The solid-state characterization techniques employed to assess the precipitate formed after equilibrium solubility studies during preformulation demonstrated that there were no phase transitions and no significant interactions between the drug and the evaluated carriers. Furthermore, the results demonstrate that Soluplus® achieved the lowest dose number (0.23) for pediatric patients over 6 years old. So, it was selected for preparing the amorphous solid dispersion via spray drying, which significantly enhanced the apparent solubility of TER while maintaining prolonged supersaturation, offering a promising alternative for developing solid formulations of this drug, particularly for pediatric patients, as it aims to improve oral bioavailability. This study designed a hydrogel patch assisted by an iontophoresis system to enhance the transdermal permeability of TEB, enabling deeper penetration into the skin layers. Methods: The influences of current intensity, pH levels, and drug concentration on the TEB hydrogel patch's permeability were explored using an adaptive ion electroosmosis system. The pharmacokinetic profile, facilitated by iontophoresis for transdermal permeation, was analyzed through the application of microdialysis technology. Scanning electron microscopy and transmission electron microscopy were employed to assess the impact of ion electroosmotic systems on skin integrity. Results: The cumulative drug accumulation within 8 h of the TEB hydrogel patches, assisted by iontophoresis, was 2.9 and 7.9 times higher than without iontophoresis assistance and TEB cream in the control group, respectively. TEB hydrogel patches assisted by iontophoresis can significantly increase the permeability of TEB, and the AUC(0–8 h) was 3.4 and 5.4 times higher, while the Cmax was 4.2 and 7.3 times higher than the TEB hydrogel patches without iontophoresis, respectively. This system has no significant impact on deep-layer cells. Conclusions: This system may offer a safe and effective clinical strategy for the local treatment of deep antifungal infections. Aim/Background: According to estimates, about a billion people in the world have fungal infections of the skin, nails, and hair. Skin disorders are a significant contributor to disability, deformity, and misery among people. In the present study, a versatile vesicular delivery of terbinafine hydrochloride through the dermal route in a biocompatible platform consisting of glycerine, phospholipids, and cholesterol is proposed for the treatment of mycoses. Materials and Methods: The glycerosomes of terbinafine hydrochloride were formulated by thin film hydration technique using central composite design to explore the composition effects on the formation of nanosized stable glycerosomes and drug loading. The optimized formulation was evaluated for deformability, antifungal activity, compatibility by FTIR, surface morphology, in vitro and ex vivo drug diffusion, skin irritation, histopathology, and cytotoxicity studies on HaCaT cell lines. Results: Numerical optimization of the design revealed that the particle size and entrapment efficiency of the drug were remarkably affected by the composition. Surface imaging by TEM revealed the formation of well-defined vesicles with no aggregation. The elasticity of the vesicles was established by determining their deformability index. The in vitro and ex vivo drug release study revealed better permeation of the drug compared to the marketed formulation. The histopathology and cytotoxicity study conceded the suitability of the glycerosomal carrier system for the transdermal delivery of terbinafine hydrochloride. This article presents the findings of a clinical trial that compared the effectiveness of two antifungal creams, ciclopirox olamine and terbinafine hydrochloride, in treating dermatophytosis. The study involved 60 patients with tinea corporis, tinea cruris, or tinea faciei, who were divided into three groups and treated with different creams for 6 weeks. The results showed that both creams were effective in treating the condition, but the combination therapy of both creams had a better response and lower relapse rate. The study acknowledges its limitations, such as a small sample size and short follow-up period, but concludes that the combination therapy is effective with no significant side effects. Targeted drug delivery is achieving great success in cancer therapy due to its potential to deliver drugs directly to the action site. Terbinafine hydrochloride (TER) is a broad-spectrum anti-fungal drug that has been found to have some potential anti-tumor effects in the treatment of colon cancer. We aimed here to design and develop pH-sensitive Eudragit (Eud)-coated mesoporous silica nanostructures (MSNs) to control drug release in response to changes in pH. The diffusion-supported loading (DiSupLo) technique was applied for loading TER into the MSNs. The formulation was optimized by a D-optimal design, which permits the concurrent assessment of the influence of drug/MSN%, coat concentration, and MSN type on the drug entrapment efficiency (EE) and its release performance. The optimal formula displayed a high EE of 96.49%, minimizing the release in pH 1.2 to 16.15% and maximizing the release in pH 7.4 to 78.09%. The cytotoxicity of the optimal formula on the colon cancer cells HT-29 was higher than it was with TER alone by 2.8-fold. Apoptosis in cancer cells exposed to the optimum formula was boosted as compared to what it was with the plain TER by 1.2-fold and it was more efficient in arresting cells during the G0/G1 and S stages of the cell cycle. Spinal infections, notably those induced by Aspergillus flavus (A. flavus), represent a complex and uncommon clinical challenge. In individuals with diabetes mellitus, the risk is exacerbated due to a compromised immune response and a heightened vulnerability to non-standard pathogens. This case report chronicles the intricate diagnostic and treatment journey of a 59-year-old diabetic patient grappling with a spinal infection attributed to A. flavus. The diagnosis was delayed due to non-specific symptoms and unclear radiological signs. The administration of voriconazole, a targeted antifungal treatment, resulted in a significant clinical and radiological improvement, underscoring its effectiveness in treating such unusual fungal spinal infections; meanwhile, we found that terbinafine hydrochloride also has a similar effect in treating fungal spinal infections. This case underscores the importance of considering fungal causes in spinal infections among diabetic patients and highlights prompt diagnosis and individualized targeted antifungal therapy.

Dermatophytosis, the most prevalent fungal infection, is witnessing a rising incidence annually. To address this challenge, we developed a terbinafine-loaded oil-in-water nanoemulsion (TH-NE) through the aqueous microtitration method. The formulation comprised olive oil (oil phase), Span 80 (surfactant), and propylene glycol (co-surfactant). Pseudo-phase ternary diagrams and thermodynamic studies underscored the stability of TH-NE. Employing the Box–Behnken design (BBD), we optimized TH-NE, which resulted in a remarkable particle size of 28.07 nm ± 0.5, a low polydispersity index (PDI) of 0.1922 ± 0.1, and a substantial negative zeta potential of −41.87 mV ± 1. Subsequently, TH-NE was integrated into a 1.5% carbopol matrix, yielding a nanoemulgel (TH-NEG). Texture analysis of TH-NEG demonstrated a firmness of 168.00 g, a consistency of 229.81 g/s, negative cohesiveness (−83.36 g), and a work of cohesion at −107.02 g/s. In vitro drug release studies revealed an initial burst effect followed by sustained release, with TH-NEG achieving an impressive 88% release over 48 h, outperforming TH-NE (74%) and the marketed formulation (66%). Ex vivo release studies mirrored these results, with TH-NEG (86%) and TH-NE (71%) showcasing sustained drug release in comparison to the marketed formulation (67%). Confocal microscopy illustrated that TH-NEG and TH-NE penetrated to depths of 30 µm and 25 µm, respectively, into the epidermal layer. Furthermore, dermatokinetic studies highlighted the enhanced drug penetration of TH-NEG compared to TH-NE through mouse skin. Terbinafine hydrochloride is a synthetic allylamine whose mechanism of action consists of inhibiting the enzyme squalene epoxidase that participates in the first stage of ergosterol synthesis, interfering with fungal membrane function. Ozonated oils are used for topical application of ozone, producing reactive oxygen species that cause cellular damage in microorganisms, therefore being an alternative treatment for acute and chronic skin infections. This study aimed to develop and characterize Eudragit® RS100 nanocapsules, obtained by interfacial deposition of preformed polymer method, containing 0.5% terbinafine hydrochloride and 5% ozonated sunflower seed oil as a potential treatment against dermatophytes. The polymeric nanocapsules were characterized regarding particle size, zeta potential, pH, drug content, encapsulation efficiency, and stability. The in vitro drug release, in vitro skin permeation, and in vitro antifungal activity were also evaluated. The particle size was around 150 nm with a narrow size distribution, the zeta potential was around + 6 mV, and the pH was 2.2. The drug content was close to 95% with an encapsulation efficiency of 53%. The nanocapsules were capable to control the drug release and the skin permeation. The in vitro susceptibility test showed greater antifungal activity for the developed nanocapsules, against all dermatophyte strains tested, compared to the drug solution. Therefore, the polymeric nanocapsules suspension containing terbinafine hydrochloride and ozonated oil can be considered a potential high-efficacy candidate for the treatment of dermatophytosis, with a possible reduction in the drug dose and frequency of applications. Droplet sizes of optimized formulations were in colloidal range. PDI values below 0.35 indicated considerably homogeneous nanoemulsions. Zeta potential values were from 13.2 to 18.1 mV indicating good stability, which was also confirmed by dispersion stability studies. Ex vivo permeation studies revealed almost total skin permeation of terbinafine hydrochloride from the nanoemulsions (96–98%) in 6 hours whereas commercial product reached only 57% permeation at the same time. Maximum drug amounts were seen in epidermis and dermis layers. Skin irritation and histopathological examination demonstrated dermatologically safe formulations. In conclusion, olive oil and clove oil-based nanoemulsion systems have potential to serve as promising carriers for topical terbinafine hydrochloride delivery.

Abstract Background Worldwide fungal infection cases are increasing by leaps and bounds. The patients who are immunocompromised, i.e., cancer and AIDS, are more susceptible to different types of fungal infections like cutaneous candidiasis and its associate infections. The available treatment for such a disease is creams, gels, etc. However, due to the lack of penetrability and higher systematic absorption, these formulations have reported many side effects. To overcome such challenges, various novel drug delivery systems were introduced. The present research focused on the preparation of glyceryl oleate ufasomes of terbinafine hydrochloride using the film hydration method. Result The prepared formulations were characterized for globular size (nm), zeta potential (mV), PDI, morphological characteristics, thermal behavior, in vitro drug release, in vitro antifungal activity, and in vitro skin permeation retention studies. After suitable formulation optimization using thin-film hydration method, 3:7 drug to glyceryl oleate ratio, UF3 formulation was found to produce higher drug entrapment efficacy (52.45 ± 0.56%), stable anionic zeta potential (− 33.37 ± 0.231 mV), desired globular size (376.5 ± 0.42 nm), and decent polydispersity index (0.348 ± 0.0345). Diffusion-controlled and zero-order sustained release profile was observed in the optimized UF3 batch. From the 5 days in vitro antifungal activity studies, it confirmed that UF3 ufasomes possessed good applicability in more prolonged therapy. Conclusion From the current investigation, it can be concluded that glyceryl oleate ufasomes of terbinafine hydrochloride could be an excellent approach to treat topical fungal infections.

Buccal administration of different active ingredients as an alternative to oral or dermal routes has been widely studied. The films are one of the most investigated dosage forms regarding the buccal formulations developed using bioadhesive polymers. Having thin and flexible structures, the films remain in the mouth for their duration of action without causing any discomfort. There are many studies conducted to develop buccal films for local treatment of oral fungal infections. In this research, buccal films were prepared using terbinafine hydrochloride, which is frequently used orally and systemically in the treatment of fungal infections and has low water solubility. The films were prepared by solvent casting method using hydroxypropyl methyl cellulose (HPMC) and polyvinyl pyrrolidone K30 (PVP) as the bioadhesive polymers and glycerin (2-3%) as the plasticizer. Characterization properties including thickness, weight uniformity, flexibility, tensile strength, swelling capacity were examined and the bioadhesive characteristics were determined by Texture Analyzer device using bovine buccal tissue. According to the findings of bioadhesion studies, the highest bioadhesive properties were detected in F1 and F7 formulations, which contained 5% HPMC. In vitro release studies exhibited that F1 and F7 film formulations with 5% HPMC represented slower and more controlled release compared to F2 and F8 film formulations having 4% HPMC + 1% PVP. Terbinafine hydrochloride is a potent antifungal drug indicated for oral and topical treatment of mycoses. A resonance Rayleigh scattering (RRS) method was developed for the determination of terbinafine hydrochloride through a feasible complexation reaction with erythrosine B. In a weakly acidic medium (acetate buffer, pH 5.0), terbinafine hydrochloride can react with erythrosine B through the electrostatic attraction and virtue of hydrophobic force to form an ion-association complex. The reaction resulted in the appearance of a new RRS peak at 369 nm. The RRS peak was increased by increasing the concentration of terbinafine hydrochloride in the linear range of 0.1–1.5 µg ml−1. All the reaction conditions (erythrosine B concentration, buffer volume, diluting solvent and pH) were optimized. The detection limit was 0.029 µg ml−1 while the quantitation limit was 0.089 µg ml−1. The suggested method after its validation was successfully applied for the determination of terbinafine hydrochloride in different pharmaceutical formulations (tablets and cream) with sufficient recovery.

Bigels with antifungal substances, ciclopirox olamine and terbinafine hydrochloride, were made of hydrogel (poloxamer 407 gel) and oleogel (polyethylene and liquid paraffin mixture). Prepared bigels were found physically stable at room temperature for six months and at least four months at 40 °C. Released amount of drug decreased when oleogel concentration in the formulation increased. Release test results depended on the insertion place of active substances. The amount of released substance was highest when ciclopirox olamine was incorporated in both phases in an equal quantity, and terbinafine hydrochloride in oleogel or in hydrogel. All formulations showed great inhibition of Microsporum canis. Thus, bigels with ciclopirox olamine and terbinafine hydrochloride are a promising dosage form for topical use.

Terbinafine is a new powerful antifungal agent indicated for both oral and topical treatment of mycosessince. It is highly effective in the treatment of determatomycoses. The chemical and pharmaceutical analysis of the drug requires effective analytical methods for quality control and pharmacodynamic and pharmacokinetic studies. Ever since it was introduced as an effective antifungal agent, many methods have been developed and validated for its assay in pharmaceuticals and biological materials. This article reviews the various methods reported during the last 25 years.

The physicochemical properties (pH, yield value, and squeeze force) of a drug for dermatomycosis, a terbinafine hydrochloride-containing cream (a brand-name product), and 12 over-the-counter drugs (OTCs) were measured and compared to ascertain the characteristics of each product. The pH of the brand-name product, Lamisil, was 4.1, and that of the OTC products ranged from 4.2 to 7.6; Lamisil Plus (7.6) had a significantly higher pH. Moreover, the yield value for Lamisil, as an index of cream ductility, was 128 dyn/cm2, and that for the OTC products ranged from 110 to 887 dyn/cm2. In particular, the OTC products Damalin (887 dyn/cm2), Barriact (512 dyn/cm2), and Exiv Deep (663 dyn/ cm2) had a significantly higher yield value. In addition, the squeeze force was measured by attaching a HapLog® to the thumb and second finger. The squeeze force for Lamisil was 12.9 N, and that for the OTC products ranged from 1.8 to 14.6 N. The OTC product Bilumon (1.8 N) had a significantly lower squeeze force. These results indicated that there were marked differences in the pharmaceutical properties of brand-name and OTC products. External preparations are characterized by their feel during use. Based on the current results, the pharmaceutical characteristics of drugs resulted in differences in their feel during use, suggesting that products appropriate for individual patients can be recommended. We measured and compared the physicochemical properties (pH, yield value, and squeeze force) of a drug for dermatomycosis, terbinafine hydrochloride-containing cream (brand-name product), and 12 generic products to clarify the characteristics of each product. On pH measurement, the pH value of the brand-name product, Lamisil, was 4.8, and those of the generic products ranged from 4.3 to 5.5, showing no marked difference. Furthermore, the yield value of Lamisil, as an index of cream ductility, was 122.2 dyn/cm2, and those of the generic products ranged from 42.1 to 1,621.5 dyn/cm2. In particular, the value of a generic product, Taiyo (42.1 dyn/cm2), was significantly lower, whereas that of another one, Viras (1,621.0 dyn/cm2), was significantly higher. In addition, the squeeze force was measured by attaching a HapLog® to the thumb and second finger. The value of Lamisil was 12.9 N, and those of the generic products ranged from 8.0 to 15.4 N. The values of generic products, Mylan (8.6 N), Tebinaceil (9.0 N), and Kelger (8.0 N), were significantly lower, whereas that of another one, Viras (15.4 N), was significantly higher. These results showed that there were marked differences in the pharmaceutical properties between the generic and brand-name products. The above pharmaceutical characteristics of drugs facilitated the presentation of reasons for differences in the sense of use, which characterizes external preparations, suggesting that products appropriate for individual patients can be recommended. Non-ionic surfactant micelles are helpful for improving the diffusion of topically delivered drugs through the cornea. This study aimed to develop terbinafine hydrochloride (TH)-loaded micelles based on a soft non-ionic surfactant-macrogol 15 hydroxystearate (HS 15) and to investigate their in vivo cornea permeation. Briefly, 0.25% TH-loaded HS 15 micelles (TH-HNMs) were developed using a simple co-solvent method. Characterization of the TH-HNMs by Zetasizer and transmission electron microscopy (TEM) revealed that the spherical and discrete micellar droplets with a small size (13.22 ± 0.73 nm) and an electrically neutral surface (− 2.15 ± 0.39 mV) were achieved. The drug entrapment efficiency of TH-HNMs was almost 100%. The release of TH from the micelles was pH dependent. 93.2 ± 3.4% of encapsulated TH was released from the micelles in the PBS at pH 5.0 within 6 h, but only 0.122 ± 0.020% of encapsulated TH was released in the PBS at pH 7.4 within the same release time. TH-HNMs possessed good physical stability in the pH neutral medium (pH 7.0).No obvious irritations were observed in rabbit eyes after ocular instillation of TH-HNMs. The in vivo corneal permeation study revealed that the TH-HNMs can penetrate into the corneal epithelium quickly and efficiently in mouse eyes. Good permeability was also noted in the stroma of mouse corneas with de-epithelialization. Compared with the TH oily solution, TH-HNMs delivered considerably increased levels of TH into rabbit corneas with or without de-epithelialization. The solubility of terbinafine hydrochloride in four pure solvents, including methanol, ethanol, n -propanol and ethyl acetate and their binary solvent mixtures, including (methanol + ethyl acetate), (ethanol + ethyl acetate) and ( n -propanol + ethyl acetate) performed at different mass fractions of methanol, ethanol or n -propanol ranging from 0.1 to 0.9, was determined by using isothermal saturation method with temperatures ranging from (278.15 to 313.15) K. The descending order of the mole fraction solubility in pure solvents was as follow: methanol > ethanol > n -propanol > ethyl acetate, and for the three mixture with given initial composition, the solubility of terbinafine hydrochloride increased with increasing temperature and mass fraction of alcohol for the three systems including (methanol + ethyl acetate), (ethanol + ethyl acetate) and ( n -propanol + ethyl acetate). At the same mass fraction of methanol, ethanol or n -propanol and temperature, the solubility of terbinafine hydrochloride was greater in (methanol + ethyl acetate) than in the other mixed solvents. The maximum mole fraction solubility of terbinafine hydrochloride was observed in methanol (6.297 × 10 − 2 at 313.15 K), followed by that in ethanol (3.785 × 10 − 2 at 313.15 K), n- propanol (3.007 × 10 − 2 at 313.15 K) and ethyl acetate (1.497 × 10 − 2 at 313.15 K)·The obtained solubility data were correlated with Jouyban-Acree model, van't Hoff-Jouyban-Acree model, modified Apelblat-Jouyban-Acree model and CNIBS/R-K model. The correlation showed good agreement with experimental results, the largest values of relative average deviations ( RAD ) and the root-mean-square deviations ( RMSD ) between the experimental and calculated solubility were 4.32 × 10 − 2 and 15.28 × 10 − 4 , respectively. On the basis of the obtained solubility, the standard enthalpy of solution ( ΔH sol o ), the standard Gibbs energy ( ΔG sol o ) of solution and the standard entropy of solution ( ΔS sol o ) of terbinafine hydrochloride dissolved in pure and mixed solvents were obtained by the famous van't Hoff calculations. Thermodynamic treatment of solubility data of terbinafine hydrochloride in these pure and mixed solvents by “Apparent thermodynamic analysis” indicated that the dissolution is an endothermic and entropy-driven process. The present study was aimed to optimize, develop, and evaluate microemulsion and microemulsion-based gel as a vehicle for transungual drug delivery of terbinafine hydrochloride for the treatment of onychomycosis. D-optimal mixture experimental design was adopted to optimize the composition of microemulsion having amount of oil ( X ), Smix (mixture of surfactant and cosurfactant; X ), and water ( X ) as the independent variables. The formulations were assessed for permeation (micrograms per square centimeter per hour; Y ), particle size (nanometer; Y ), and solubility of the drug in the formulation (milligrams per milliliter; Y ). The microemulsion containing 3.05% oil, 24.98% Smix, and 71.96% water was selected as the optimized formulation. The microemulsion-based gel showed better penetration (∼5 folds) as well as more retention (∼9 fold) in the animal hoof as compared to the commercial cream. The techniques used to screen penetration enhancers (hydration enhancement factor, ATR-FTIR, SEM, and DSC) revealed the synergistic effect of combination of urea and n-acetyl cysteine in disruption of the structure of hoof and hence, leading to enhanced penetration of drug. In spite of developing new drugs and modern formulations, the treatments of chronic fungal infections are still challenging. Fibrous wound dressings are new suggestions for the treatment of chronic superficial infections. In the present study, we formulated an antifungal agent, terbinafine hydrochloride (TFH), which is a hydrophobic drug, in wound dressings prepared by electrospun polycaprolactone, polycaprolactone/gelatin (50:50 w/w) and gelatin. To obtain more water-stable meshes, the preparations were treated by glutaraldehyde and their properties were determined before and after treatment. The morphology of fibrous meshes was observed by scanning electron microscopy. Drug loading efficiency and release rate were measured by high performance liquid chromatography (HPLC) and the release rate was monitored for 144 h. Antifungal tests were performed on Trichophyton mentagrophytes , Aspergillus fumigatus and Candida albicans cultured on Muller-Hinton agar. The toxicity of the meshes was measured after 24 h and 14 days by MTT assay. Terbinafine loading of polycaprolactone/gelatin (50:50) was 100% and it released the highest amount of TFH too. In antifungal tests, all samples were able to hinder T. mentagrophytes and A. fumigatus but not C. albicans growth among them, polycaprolactone fibers made the largest inhibition zone. In MTT assay, none of prepared samples showed toxicity against L929 cells. Teken together, the prepared TFH-loaded PCL/gelatin electrospun meshes were able to release TFH slowly and in a steady state in time. With respect to no obvious cytotoxicity in MTT assay and stong antifungal activity toward T. mentagrophytes in vitro , these TFH-based meshes could be considered as potential candidates in clinical application as wound dressing for treatment of chronic dermatophytosis. In this paper, the first part the chemical co-precipitation method was used in the preparation of magnetic nanoparticles and then application of SDS-coated Fe3O4 NPs as an adsorbent for extraction of trace amounts of terbinafine hydrochloride from pharmaceutical company wastewater was investigated. Then the drug was determined by high performance liquid chromatography with UV detection (HPLC/UV). The effects of various parameters on the extraction efficiency such as pH of solution, amount of SDS surfactant, extraction time, eluent type and desorption condithions were studied and optimized. Under the optimal conditions, the calibration curve was linear in the range of 1.0â400 µgL-1 with a correlation coefficient of 0.9998. Enrichment factor of 74 and limit of detection of 0.5µgL-1 were obtained. Also, the percent relative intra-day and inter-day standard deviation (R.S.D %) based on three replicate determinations were 1.0 and 3.5%, respectively. According to the results, the proposed method was successfully applied to extraction of terbinafine from environmental samples and satisfactory results were obtained.

Onychomycosis affects about 15% of the population. This disease causes physical and psychosocial discomfort to infected patients. Topical treatment (creams, solutions, gels, colloidal carriers, and nail lacquers) is usually the most commonly required due to the high toxicity of oral drugs. Currently, the most common topical formulations (creams and lotions) present a low drug delivery to the nail infection. Nail lacquers appear to increase drug delivery and simultaneously improve the effectiveness of treatment with increased patient compliance. These formulations leave a polymer film on the nail plate after solvent evaporation. The duration of the film residence in the nail constitutes an important property of nail lacquer formulation. In this study, a polyurethane polymer was used to delivery antifungals drugs, such as terbinafine hydrochloride (TH) and ciclopirox olamine (CPX) and the influence of its concentration on the properties of nail lacquer formulations was assessed. The nail lacquer containing the lowest polymer concentration (10%) was the most effective regarding the in vitro release, permeation, and antifungal activity. It has also been demonstrated that the application of PU-based nail lacquer improves the nail plate, making it smooth and uniform and reduces the porosity contributing to the greater effectiveness of these vehicles. To conclude, the use of polyurethane in nail formulations is promising for nail therapeutics.

Terbinafine hydrochloride-loaded nanofibrous buccal films were formulated with the aim to improve the solubility and dissolution behavior; thus, the local effectiveness of the antifungal agent. Poly(vinyl alcohol) and chitosan polymer composites were selected as delivery base in order to enhance the mucoadhesion of the fibrous films. The dissolution of terbinafine hydrochloride was carried out applying a stainless steel disc assembly and the terbinafine concentration was determined by HPLC–MS in selective ion monitoring mode. The prediction of the absorption behavior of the prepared fibrous samples in the human oral cavity was modeled using GastroPlus™ software. The result indicates that the fibrous films enabled fast and complete dissolution of the active agent. The drug absorption from the oral cavity could be minimized by the employment of the proper oral transit model. Because of the limited absorption of terbinafine hydrochloride from the oral mucosa the formulation can be beneficial in local administration in the case of hold and expectorate administration mode. Onychomycosis is a fungal infection of nail unit that is caused by dermatophytes. Oral Terbinafine hydrochloride (TBF-HCl) is being used for the treatment of onychomycosis since 24 years. The side effects caused by the systemic application and limitations of topical administration of this drug regarding the diffusion through nail lead to the development of a new formulation based on, TBF-HCl-loaded liposome. The newly obtained film formulations were prepared and characterized via several parameters, such as physical appearance, drug content, thickness, bioadhesive properties and tensile strength.In vitroandex vivopermeation studies were performed to select an optimum film formulation for antifungal activity to show the efficiency of formulations regarding the treatment of onychomycosis. Thein vitrorelease percentages of drug were found 71.6 ± 3.28, 54.4 ± 4.26, 56.1 ± 7.48 and 46.0 ± 2.43 for liposome loaded pullulan films (LI-P, LII-P) and liposome loaded Eudragit films (LI-E, LII-E), respectively. The accumulated drug in the nail plates were found 31.16 ± 4.22, 24.81 ± 5.35, 8.17 ± 1.81 and 8.92 ± 3.37 for LI-P, LII-P, LI-E and LII-E, respectively, which within therapeutic range for all film formulations. The accumulated drug in the nail plate was found within therapeutic range for all film formulations. The efficacy of the selected TBF-HCl-loaded liposome film formulation was compared with TBF-HCl-loaded liposome, ethosome, liposome poloxamer gel and ethosome chitosan gel formulations. It was found that TBF-HCl-loaded liposome film formulation had better antifungal activity on fungal nails which make this liposome film formulation promising for ungual therapy of fungal nail infection. The present study investigates the entrapment of terbinafine hydrochloride (TH) in ethosomal vesicles via unsonicated and sonication method. Carbopol 934P was incorporated in the best formulation, F6, obtained by sonication method. The formulated ethosomal gel obtained as such i.e. F6 ∗ was exploited to achieve a zero order release profile of TH. The composition includes phospholipid, ethanol and propylene glycol. Drug entrapment efficiency (DEE), in-vitro and ex-vivo drug diffusion studies, FT-IR and stability studies of the prepared ethosomes were investigated. The size and shape of F6 ethosomes vesicles were characterized by SEM. In-vitro drug release studies were performed using sigma dialysis membrane in phosphate buffer, pH 7.4 for 12 h while drug content was determined by HPLC. DEE was ranked from 55.33 ± 1.32% to 69.11 ± 2.11%. Highest DEE was seen with F6 ethosomal formulation with a vesicle size of 248 ± 1.02 nm. FT-IR studies confirmed that there was no chemical interaction between drug and excipients used in the formulation. Ex-vivo result suggested that drug diffusion observed after 12 h from F6 ∗ and marketed cream (MR) formulations was 74.01 ± 0.62% and 61.45 ± 0.86%, respectively. The results of similarity factor ( f 2 values) for MR and F6 ∗ ethosomal gel were 85.14 and 42.63, respectively. It revealed that F6 ∗ showed dissimilar dissolution profiles. Transdermal flux value for F6 ∗ and MR was found to be 144.61 ± 1.28 μg/cm 2 /h and 121.6 ± 1.16 μg/cm 2 /h, respectively. We describe the scroll system as a new microparticulate structured delivery system for enhanced delivery to/across the skin. The basic components of the scroll system are non-ionic surface active of the type of alkyl polyglycol ethers and a glycol. The unique structures are preserved with addition of various ingredients such as polymers, vegetable oils, pharmaceuticals, and permeation enhancers but are dismissed when amphiphile is withdrawn. The microparticles have a unique scroll structure with multiple 'wrapping.' Besides enabling superior permeation of drugs into/across the skin, the drugs delivered by scroll systems were more effective in vitro and in vivo compared to controls. Model drugs presented high entrapment capacity in scroll systems. The systems are stable over time and are safe for skin application. In order to form, they require a small number of ingredients, simple preparation method, and are environment friendly. Context: Terbinafine hydrochloride is an antifungal drug for onychomycosis. Poor permeability of its external preparation leads to poor curative effect. Transfersomes, also known as flexible liposome, could improve transmission of drug for local external use. Terbinafine hydrochloride-loaded liposome is expected to become a breakthrough on the treatment of onychomycosis. Objective: This study is aimed to prepare high skin penetration terbinafine hydrochloride transfersomes with high encapsulation efficiency, appropriate drug loading and good stability. Materials and methods: Taking entrapment efficiency as the main indicator, the formulations and the processes of preparation were investigated. Transfersomes with different surfactants were prepared in the optimization processes, and the formulations were optimized through the transdermal testin vitro. As a result, a gel contained transfersomes was obtained with a brief evaluation. Its pharmacokinetic properties of going through the skin were studied by using the micro dialysis technology and liquid chromatography-mass spectrometry to assay the penetration behavior of terbinafine. Results: Mean particle size of the terbinafine hydrochloride transfersomes was 69.6 ± 1.23 nm, and the entrapment efficiency was 95.4% ± 0.51. The content of the gel was 4.45 ± 0.15 mg/g. The accumulated permeation of the transfersomes gel in 12 h was 88.52 ± 4.06 µg cm−2and the intracutaneous drug detention was 94.38 ± 5.26 µg cm–2. The results of pharmacokinetic studies showed theCmaxand area under the curve (AUC) were apparently higher than the commercial cream. Discussion and conclusion: The terbinafine hydrochloride transfersomes was highly absorbed by the skin. The purpose of this study was to prepare hydrogels and microemulsion (ME)-based gel formulations containing 1% terbinafine hydrochloride (TER-HCL) and to evaluate the use of these formulations for the antifungal treatment of fungal infections. Three different hydrogel formulations were prepared using chitosan, Carbopol® 974 and Natrosol® 250 polymers. A pseudo-ternary phase diagram was constructed, and starting from ME formulation, a ME gel form containing 1% Carbopol 974 was prepared. We also examined the characteristic properties of the prepared hyrogels. The physical stability of hydrogels and the ME -based gels were evaluated after storage at different temperatures for a period of 3 months. The release of TER-HCL from the gels and the commercial product (Lamisil®) was carried out by using a standard dialysis membrane in phosphate buffer (pH 5.2) at 32 °C. The results of the in vitro release study showed that the Natrosol gel released the highest amount of drug, followed by Carbopol gel, chitosan gel, commercial product, and the microoemulsion-based gel in that order. In vitro examination of antifungal activity revealed that all the prepared and commercial products were effective against Candida parapsilosis, Penicillium, Aspergillus niger and Microsporum. Terbinafine hydrochloride (TBH) is a new antifungal agent widely used for the treatment of many contagious diseases. Two spectrophotometric methods based on ion-pair complexation reaction are described for the determination of TBH. The first method is based on the formation of ion-pair complex between TBH and alizarin red S (ARS) dye in HCl medium followed by its extraction into methylene chloride and measurement at 450 nm (method A). The other method entails breaking of drug-dye complex in alcoholic KOH medium and measuring the absorbance of the blue dye at 570 nm (method B). The experimental conditions for selective and sensitive assay were optimized, and the drug-dye stoichiometry was found to be 1:1. Beer's law was obeyed over the concentration ranges 2.5-60, and 4-24 µg mL-1 for method A and method B, respectively, and the corresponding molar absorptivity values were 5.92x10³ and 4.93x10³ L mol-1 cm-1, respectively. The limits detection (LOD) and quantification (LOQ) values were calculated to be 0.22 & 0.66 (method A) and 0.35 & 1.05 (method B). In addition, the methods were also validated with respect to accuracy, precision, robustness, ruggedness and selectivity. Selection of excipients used is a critical step in the design of a pharmaceutical dosage form as it affects its behavior upon application, as during storage. The purpose of the present study is to evaluate and compare the behavior of six liposomal formulations intended for topical application composed of two widely used phospholipids 1,2-diacyl-sn-glycero-3-phosphocholine and 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine with and without incorporation of cholesterol. Liposomal hydrogels made of hydroxyethylcellulose 3% and incorporating the anti-fungal agent terbinafine hydrochloride (E)-N-(6,6-dimethyl-2-hepten-4-inyl)-N-methyl-1- naphthalene-methanamine (-hydrochloride) were prepared, their viscosity was measured and in vitro drug release was studied. Moreover, physical stability and drug retention during storage at two different temperatures (2-8 °C and RT) were examined over time. The results showed differences in the behavior between the two phospholipids while incorporation of cholesterol at the studied concentrations was found to be of minor importance. Drug release was found to be favorable from 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine (POPC) liposomal hydrogels and drug retention was found to be higher at lower storage temperature for all batches. ABSTRACT In this paper, the application of sodium dodecylsulfate-coated Fe3O4 nanoparticles as an adsorbent was evaluated for the extraction and preconcentration of trace amounts of terbinafine hydrochloride in biological fluids before its determination by high-performance liquid chromatography with ultraviolet detection. Effects of various parameters on the extraction efficiency such as pH of solution, amount of sodium dodecylsulfate surfactant, extraction time, and desorption conditions were studied and optimized. Under the optimal conditions, the calibration curve was linear in the range of 2.5 to 500 µg L−1 with the correlation coefficient of 0.9998. Moreover, the calibration graphs were linear in the range of 5 to 500 µg L−1 (seven standards) for both urine and plasma samples and the limits of detection were found to be 0.3, 0.7, and 1.5 µg L−1in aqueous, urine, and plasma samples, respectively. Also, the percent of relative standard deviation (RSD%) based on three replicate determinations was in the range of 0.9% to 6.8%. Direct (A) and indirect (B) spectrophotometric methods are proposed for the assay of Terbinafine HCl, Telmisartan and Ramipril in bulk drugs and in their dosage forms using ceric (IV) sulphate and ceric (IV) sulphate-chromatrope 2R as reagents. Method (A) involves the addition of certain amount of ceric (IV) sulphate to drugs in acid medium, followed by determination of the decrease in absorbance measured at 319 nm using the experiment as a blank. Method (B) involves the addition of a known excess of ceric (IV) sulphate to drugs in acid medium followed by the determination of unreacted ceric by reacting with a fixed amount of chromatrope 2R then the decrease in color intensity of C2R was measured spectrophotometrically at λmax 507 nm against the reagent blank. The Beer's law is obeyed in the concentration range of (1-9) µg/ml (A), (1-7) µg/ml (B) of Terbinafine hydrochloride, (1-5) µg/ml(A), (B) of Telmisartan and (10-80) µg/ml (A), (10-60) µg/ml (B) of Ramipril. The validity of these methods have been successfully applied for the determination of the drugs in there pharmaceutical preparations. The present research work aimed at the impact of reduced vesicular size on the characteristics of ethosomes by comparing with the regular vesicular size of ethosomes as topical drug delivery vehicle to achieve optimal localized drug concentration and reduced dose frequency of the Terbinafine hydrochloride (TH), an antifungal drug. Oral use of TH contraindicated resulting from sever side effect, thus topical administration is recommended. Commercially available TH creams, lotions and sprays, have limitation of relatively short residual period at target site. The entrapment of drug in vesicles improves localization, solubility and availability of drug at the site; resulting in reduction of the dose. Ethosomes containing drug were prepared by employing higher concentration of alcohol in the form of hydroalcoholic or hydroglycolic phospholipid. Sonicated and unsonicated ethosomes were investigated for shape, particle size, and entrapment efficiency. Electronic microscope investigation not only revealed, vital evidence for presence of phospholipid vesicles in TH ethosomal systems but also displayed greater uniformity in size and shape of sonicated ethosomes than unsonicated ethosomes. Furthermore, the Comparative investigation was carried out for ex vivo skin permeation, ex vivo drug release and entrapment efficiency studies. Drug release followed zero order release rate kinetics. Drug accumulation study showed more than 19.01% of drug was deposited into skin by sonicated ethosomal formulation as compared to 2.57% by unsonicated ethosomal formulation. Sonicated and unsonicated ethosomes were found stable at refrigeration and room temperature conditions during stability studies. Drug accumulation studies in deep skin strata was found to be comparatively greater in sonicated ethosomes, which indicates higher localized drug and that in turn reduces dose frequency. This study determined the onychopharmacokinetics, nail absorption, distribution, and penetration of [14C]-terbinafine HCl in a new topical formulation into/through the human finger nail using the in vitro finite dose model. This study determined the penetration rate of terbinafine HCl from multiple doses of topical formulation applied daily for 14 days. Results showed that the total dose recovery (mass balance) was almost 100%. The concentration of terbinafine HCl in the deeper nail plate (ventral/intermediate layers) and the cotton-pad nail bed samples after the 14-day treatment were 613 ± 145 and (±S.D.) and 27 ± 1.2 µg/cm3 (or 1.9 ± 0.6 µg/cm3 daily) on average, respectively. In comparison with nail concentration data from the literature for other topical terbinatine formulations, our results show that higher amounts of terbinafine HCl reached the deep nail plate and/or the nail bed after a 14-day topical treatment with this topical formulation in vitro. Frequent instillation of terbinafine hydrochloride (T HCl) eye drops (0.25%, w/v) is necessary to maintain effective aqueous humor concentrations for treatment of fungal keratitis. The current approach aimed at developing potential positively charged controlled-release polymeric nanoparticles (NPs) of T HCl. The estimation of the drug pharmacokinetics in the aqueous humor following ocular instillation of the best-achieved NPs in rabbits was another goal. Eighteen drug-loaded (0.50%, w/v) formulae were fabricated by the nanopreciptation method using Eudragit® RS100 and chitosan (0.25%, 0.5%, and 1%, w/v). Soybean lecithin (1%, w/v) and Pluronic® F68 (0.5%, 1%, and 1.5%, w/v) were incorporated in the alcoholic and aqueous phases, respectively. The NPs were evaluated for particle size, zeta potential, entrapment efficiency percentage (EE%), morphological examination, drug release in simulated tear fluid (pH 7.4), Fourier-transform IR (FT-IR), X-ray diffraction (XRD), physical stability (2 months, 4°C and 25°C), and drug pharmacokinetics in the rabbit aqueous humor relative to an oily drug solution. Spherical, discrete NPs were successfully developed with mean particle size and zeta potential ranging from 73.29 to 320.15 nm and +20.51 to +40.32 mV, respectively. Higher EE% were achieved with Eudragit® RS100-based NPs. The duration of drug release was extended to more than 8 h. FT-IR and XRD revealed compatibility between inactive formulation ingredients and T HCl and permanence of the latter’s crystallinity, respectively. The NPs were physically stable, for at least 2 months, when refrigerated. Abstract: The study describes the development and evaluation of solid lipid nanoparticles (SLNs) of terbinafine hydrochloride (TH) for sustained release and skin targeting. TH-loaded SLNs were prepared by solvent-injection technique and optimized using 33 full-factorial design. Effect of drug:lipid ratio, surfactant concentration and volume of organic solvent were studied on % entrapment efficiency (%EE) and particle size (PS). The optimum formulation based on desirability (0.945) exhibited %EE of 73.74% and PS of 300nm. Optimized SLNs were incorporated into Carbopol gel and evaluated for drug content, pH, in vitro release, ex vivo retention, in vivo pharmacodynamic and stability studies. Drug release from SLNs dispersion followed Korsmeyer–Peppas model, indicating Fickian drug release, while that from the gel followed Higuchi model. The ex vivo studies through rat abdominal skin indicated skin retention ability of SLNs as compared to commercial product. In vivo pharmacodynamic studies showed that the SLNs based gel reduced fungal burden of Candida albicans in rats as compared to commercial product in shorter duration of time. The SLNs dispersion and gel exhibited physicochemical stability under refrigeration upto 3months. It was concluded that SLNs incorporated Carbopol gel had skin targeting ability and may serve as a promising carrier in treatment of fungal skin infections. [Copyright &y& Elsevier]

Abstract: Terbinafine hydrochloride (TH), a poorly water soluble antifungal agent, was characterized by solid state techniques including differential scanning calorimetry, thermogravimetry, X-ray powder diffraction, optical and electron microscopies, Fourier transform infrared, Raman and solid-state nuclear magnetic resonance spectroscopies and intrinsic dissolution rate (IDR). A colorless single crystal of TH was grown from an ethanol:water solution and its crystalline structure was determined through X-ray single crystal diffraction. Also, a new crystal habit of TH was obtained through the slow solvent evaporation technique revealing a needle-like shape. A comparison between the IDR results for the TH raw material and TH needle-like crystal revealed lower values for the new crystal habit, which can be attributed to the preferential orientation of the crystals in the compressed disks. Abstract: Terbinafine hydrochloride (T-HCl) is recommended for the management of fungal keratitis. To maintain effective aqueous humor concentrations, frequent instillation of T-HCl drops is necessary. This work aimed to develop alternative controlled-release in situ ocular drug-loaded nanoemulsion (NE) gels. Twelve pseudoternary-phase diagrams were constructed using oils (isopropyl myristate/Miglyol® 812), surfactants (Tween® 80/Cremophor® EL), a co-surfactant (polyethylene glycol 400) and water. Eight drug-loaded (0.5%, w/v) NEs were evaluated for thermodynamic stability, morphology, droplet size and drug release in simulated tear fluid (pH 7.4). Following dispersion in gellan gum solution (0.2%, w/w), the in situ NE gels were characterized for transparency, rheological behavior, mucoadhesive force, drug release and histopathological assessment of ocular irritation. Drug pharmacokinetics of sterilized F31 [Miglyol® 812, Cremophor® EL: polyethylene glycol 400 (1:2) and water (5, 55 and 40%, w/w, respectively)] in situ NE gel and oily drug solution were evaluated in rabbit aqueous humor. The NEs were thermodynamically stable and have spherical droplets (<30nm). The gels were transparent, pseudoplastic, mucoadhesive and showed more retarded zero-order drug release rates. F31 in situ NE gel showed the least ocular irritation potential and significantly (P <0.01) higher C max, delayed T max, prolonged mean residence time and increased bioavailability. The novel antifungal hybrid of terbinafine hydrochloride (TER-HCl)/montmorillonite was synthesized by the intercalation method under mechanical stirring. Intercalation of TER-HCl in the MMT galleries was characterized by X-ray diffraction (XRD), Fourier transform infrared spectra, elemental analysis (EA). and thermogravimetric analysis (TGA). The results from IR, TGA, and EA showed a difference in chemical composition of the MMT and the TER-HCl/MMT. XRD analysis showed that the basal spacing of montmorillonite significantly expanded from 1.53 to 2.79 nm. TER-HCl was successfully intercalated into the interlayer of MMT, and 28 % of TER-HCl was released after 48 h in 0.9 % (w/v) NaCl aqueous solution (pH 7) at 37 ± 0.5 °C. The antifungal activity of the hybrid against Candida albicans was evaluated using the inhibitory zone method and the minimum inhibitory concentration. The TER-HCl/MMT strongly inhibited C. albicans. Abstract: Terbinafine hydrochloride (TerbHCl) is an allylamine derivative with fungicidal action, especially against dermatophytes. Different analytical methods have been reported for quantifying TerbHCl in different samples. These procedures require time-consuming sample preparation or expensive instrumentation. In this paper, electrochemical methods involving capillary electrophoresis with contactless conductivity detection, and amperometry associated with batch injection analysis, are described for the determination of TerbHCl in pharmaceutical products. In the capillary electrophoresis experiments, terbinafine was protonated and analyzed in the cationic form in less than 1min. A linear range from 1.46 to 36.4μgmL−1 in acetate buffer solution and a detection limit of 0.11μgmL−1 were achieved. In the amperometric studies, terbinafine was oxidized at +0.85V with high throughput (225 injectionh−1) and good linear range (10–100μmolL−1). It was also possible to determine the antifungal agent using simultaneous conductometric and potentiometric titrations in the presence of 5% ethanol. The electrochemical methods were applied to the quantification of TerbHCl in different tablet samples; the results were comparable with values indicated by the manufacturer and those found using titrimetry according to the Pharmacopoeia. The electrochemical methods are simple, rapid and an appropriate alternative for quantifying this drug in real samples. [Copyright &y& Elsevier]

The main objective of the present study was to investigate the plausibility of iontophoretic delivery of drugs to the nail matrix via proximal nail fold. The in vitro drug transport studies were performed in Franz diffusion cells across folded epidermis, which is used as a model for the proximal nail fold. The amount of drug transported into the receiver compartment following iontophoresis for 3 h at 0.5 mA/cm2 was 150-fold higher than the control (0.008 ± 0.002 μg/cm2). The amount of drug present in the skin after iontophoresis (0.45 ± 0.12 μg/mg) was approximately fivefold higher as compared with that of the control (0.08 ± 0.01 μg/mg). Iontophoresis of terbinafine across the proximal nail fold was assessed using excised cadaver toe model as well. A custom-designed foam-pad-type patch system was used for iontophoresis in cadaver toes. The amount of the drug delivered into the nail matrix following iontophoresis for 3 h was significantly higher than the minimum inhibition concentration of terbinafine. However, on the contrary, passive delivery for about 24 h did not result in any detectable drug levels in the nail matrix. The aim of this study was to compare the skin permeation of ethosomes, binary ethosomes and transfersomes of Terbinafine Hydrochloride (TH) under non-occlusive conditions. These lipid vesicles were prepared and characterized for shape, size, zeta-potential and entrapment efficiency. Franz diffusion cells and confocal laser scanning microscopy (CLSM) were used for the percutaneous absorption studies. The quantity of drug in the skin from ethosomes, binary ethosomes (the weight ratio of ethanol to propylene glycol 7:3, ethanol-PG = 7:3, w/w), and transfersomes was 1.26, 1.51 ( p <0.05), 1.56 ( p <0.01) times higher than that of TH from traditional liposomes (control). The skin deposition of the applied dose (DD%) of TH from ethosomes, binary ethosomes, and transfersomes was 3.34 ( p < 0.05), 9.88 ( p < 0.01), 2.52 times higher than that of TH from control. The results of CLSM experiments showed that penetration depth and fluorescence intensity of Rhodamine B from binary ethosomes was much greater than that from ethosomes and transfersomes. These results indicated the binary ethosomes (ethanol-PG = 7:3, w/w) most effectively permitted drug penetration through skin; transfersomes made drug easiest to accumulate in the skin.
