In this study, we employed calcium imaging to investigate the dynamics of intracellular calcium levels in embryonic chicken cardiomyocytes upon extracellular, optoelectronic stimulation. A photovoltaic layer of donoracceptor pigments on a flexible PET substrate was used as a wireless stimulation electrode. Our findings revealed a distinct change in their spontaneous activity pattern in cardiac cells following asynchronous light stimulation. A short pause in cellular activity, indicative of overdrive suppression, was observed in recordings from several different cells. The pause in activity signifies a transient refractory period induced by stimulation of the photovoltaic device with red light. These findings suggest that photovoltaic electrodes can be used to effectively modulate the electrical activity of cardiac cells in a wireless, non-pharmacological manner. This opens new avenues for non-invasive and precise light-modulated control of cellular electrophysiology as well as potential therapeutic applications for cardiac rhythm disorders.

Fluid flow in the human body can be modeled using a water-level tank, a commonly used mechanistic approach in mechanical engineering for fluid transport processes. Postoperative fluid data from patients undergoing cardiac surgery is used to estimate fluid flow dynamics and total body water in the human body. This simplified model provide a basic understanding of the dynamics of fluid flow processes in the human body and could aid in modeling distribution of fluids in compartments.

This paper discusses possibilities to explain which Kv channels can be considered as first-order systems. This means that the dynamic behavior of these channels can be described with a first-order differential equation and are the most straightforward dynamical systems. For example, a typical first-order system is an RC circuit, and analogously, Kv channels can be considered with the dynamic behavior of first-order systems following RC circuits. This work represents a family of KV channels that can be described as first-order systems, providing an efficient method of ion channel modeling in computational electrophysiology.

Background: Peripheral nerve injuries affect over 2% of trauma patients and can lead to severe functional impairment and permanent disability. Autologous nerve transplantation is still the gold standard in the reconstruction of nerve defects. For small defects, conduits can be considered for bridging. Lately, the combined use of conduits and electrical stimulation has gained attention in the treatment of peripheral nerve injury. This review aimed to present the currently available data on this topic. Methods: PubMed, Embase, Medline and the Cochrane Library were searched for studies on electrical stimulation through nerve conduits for nerve defects in in vivo studies. Results: Fifteen studies fit the inclusion criteria. All of them reported on the application of nerve conduits combined with stimulation for sciatic nerve gaps in rats. Functional, electrophysiological and histological evaluations showed improved nerve regeneration after electrical stimulation. High variation was observed in the treatment protocols. Conclusion: Electrically stimulated conduits could improve peripheral nerve regeneration in rat models. The combined application of nerve guidance conduits and electrical stimulation shows promising results and should be further evaluated under standardized conditions. [ABSTRACT FROM AUTHOR]

This work introduces a low-cost open-source electrocardiography (ECG) simulator comprising both MATLAB software for signal generation and a dedicated circuit board for signal output via a commercial sound card. Synthetic, rate-dependent ECG simulation is based on third-order polynomials that are calculated in sections for the main waves and spikes, respectively. Besides the heart rate, the output profile is fully adjustable with respect to Einthoven lead signals I-III, the amplitudes of the individual ECG waves and spikes, as well as the constitution and intensity of common distortions. The underlying coefficients for the synthetic ECG profile are obtained experimentally by analysing recordings of 22 healthy individuals with heart rates in the range of 40-180 bpm. Eight of these recordings are selected to determine the coefficients for the polynomials (training set) while the remaining 14 serve as test set to evaluate their applicability and accuracy. Thereby, a mean correlation of 98.57% is found which is superior in comparison with a widely accepted rate-dependent ECG profile that is generated from square root and linear terms (correlation score: 91.46%). Although other use-cases are feasible, the focus of this work is the development of an ECG simulator for academic research and university education. Neurostimulation employing photoactive organic semiconductors offers an appealing alternative to conventional techniques, enabling targeted action and wireless control through light. In this study, organic electrolytic photocapacitors (OEPC) are employed to investigate the effects of light-controlled electric stimulation on neuronal networks in vitro and in vivo. The interactions between the devices and biological systems are characterized. Stimulation of primary rat cortical neurons results in an elevated expression of c-Fos within a mature neuronal network. OEPC implantation for three weeks and subsequent stimulation of the somatosensory cortex leads to an increase of c-Fos in neurons at the stimulation site and in connected brain regions (entorhinal cortex, hippocampus), both in the ipsi- and contralateral hemispheres. Reactivity of glial and immune cells after semi-chronic implantation of OEPC in the rat brain is comparable to that of surgical controls, indicating minimal foreign body response. Device functionality is further substantiated through retained charging dynamics following explantation. OEPC-based, light-controlled electric stimulation has a significant impact on neural responsiveness. The absence of detrimental effects on both the brain and device encourages further use of OEPC as cortical implants. Objective: This study aims to explore the potential of organic electrolytic photocapacitors (OEPCs), an innovative photovoltaic device, in mediating the activation of native voltage-gated Cav1.2 channels (I Ca,L ) in Guinea pig ventricular cardiomyocytes., Methods: Whole-cell patch-clamp recordings were employed to examine light-triggered OEPC mediated I Ca,L activation, integrating the channel's kinetic properties into a multicompartment cell model to take intracellular ion concentrations into account. A multidomain model was additionally incorporated to evaluate effects of OEPC-mediated stimulation. The final model combines external stimulation, multicompartmental cell simulation, and a patch-clamp amplifier equivalent circuit to assess the impact on achievable intracellular voltage changes., Results: Light pulses activated I Ca,L , with amplitudes similar to voltage-clamp activation and high sensitivity to the L-type Ca 2+ channel blocker, nifedipine. Light-triggered I Ca,L inactivation exhibited kinetic parameters comparable to voltage-induced inactivation., Conclusion: OEPC-mediated activation of I Ca,L demonstrates their potential for nongenetic optical modulation of cellular physiology potentially paving the way for the development of innovative therapies in cardiovascular health. The integrated model proves the light-mediated activation of I Ca,L and advances the understanding of the interplay between the patch-clamp amplifier and external stimulation devices., Significance: Treating cardiac conduction disorders by minimal-invasive means without genetic modifications could advance therapeutic approaches increasing patients' quality of life compared with conventional methods employing electronic devices.
