The temporal and spatial resolutions of rainfall data directly affect the accuracy of hydrological simulation. Weather radar has been used in business in China, but the uncertainty of data is large. At present, research on radar data and fusion in small and medium-sized basins in China is very weak. In this paper, taking the Duanzhuang watershed as an example, based on station data, Shijiazhuang's radar data are preprocessed, optimized and fused. Eleven rainfall events are selected for fusion by three methods and quality evaluation, and three flood simulations are used to test their effect. The results show that preprocessing and initial optimization have poor effects on radar data improvement. The rainfall proportional coefficient fusion method performs best in rainfall spatial estimation, where the R2 values of the three inspection stations are increased to 0.51, 0.78 and 0.82. Three fusion datasets in the peak flow and flood volume of flood simulation perform better than station data. For example, in the No.20210721 flood, the NSE of the three fusion data increased by 39, 30 and 48%. This shows that the fusion method can effectively improve the data accuracy of radar and can obtain high temporal and spatial resolution rainfall data. Gases are the vital nutrition of all organisms as the precursor of metabolism pathways. As a potential biological process, protein synthesis is inevitably regulated by gas transport and utilization. However, the effect of carbon dioxide (CO2) present in many metabolic pathways on protein synthesis has not been studied well. In this work, carbon dioxide combined with oxygen was employed for cell-free protein synthesis (CFPS) in the tube-in-tube reactor with precise control of gas concentration. In this in vitro system, gases could directly affect the protein synthesis process without transmembrane transport. Varied concentrations of carbon dioxide (0–1%) and constant oxygen concentration (21%) were employed for CFPS to assess the effects. The cell-free reactions with 0.3% CO2 and 21% O2 showed the highest protein yields. The combined effect of CO2 and O2 also resulted in relatively high protein expression under high oxygen conditions (0.3% CO2 and 100% O2). Moreover, metabolomics assays were performed to gain insight into metabolic changes, which showed that CO2 slightly improved energy metabolism and redox balance. In particular, the extra supplied CO2 activated the decarboxylating reactions and removed toxic metabolites to recover the protein synthesis activity. The exploration of CO2 on protein synthesis could provide guiding implications for basic studies and biomanufacturing.

Abstract Protein-based biomaterials have the characteristics of stability and biocompatibility. Based on these advantages, various bionic materials have been manufactured and used in different fields. However, current protein-based biomaterials generally need to form monomers in cells and be purified before being assembled in vitro. The preparation process takes a long time, and the complex cellular environment is challenging to be optimized for producing the target protein product. Here this study proposed technology for in situ synthesis and assembly of the target protein, namely the cell-free protein synthesis (CFPS), which allowed to shorten the synthesis time and increase the flexibility of adding or removing natural or synthetic components. In this study, successful expression and self-assembly of the dihedral symmetric proteins proved the applicability of the CFPS system for biomaterials production. Furthermore, the fusion of different functional proteins to these six scaffold proteins could form active polymers in the CFPS system. Given the flexibility, CFPS is expected to become a powerful tool as the prototyping and manufacturing technology for protein-based biomaterials in the future. Graphical Abstract

Pesticide residues in kumquat fruits from China, and the quality and chronic/acute intake risks in Chinese consumers, were assessed using the QuEChERS procedure and UHPLC-MS/MS and GC-MS/MS methods. Our 5-year monitoring and survey showed 90% of the 573 samples of kumquat fruits collected from two main production areas contained one or multiple residual pesticides. Overall, 30 pesticides were detected, including 16 insecticides, 7 fungicides, 5 acaricides, and 2 plant growth modulators, of which 2 pesticides were already banned. Two or more residual pesticides were discovered in 81% of the samples, and pesticide residues in 9.4% of the samples surpassed the MRLs, such as profenofos, bifenthrin, triazophos, avermectin, spirodiclofen, difenoconazole, and methidathion. The major risk factors on the safety of kumquat fruits before 2019 were profenofos, bifenthrin, and triazophos, but their over-standard frequencies significantly declined after 2019, which was credited to the stricter supervision and management policies by local governments. Despite the high detection rates and multi-residue occurrence of pesticides in kumquat fruits, about 81% of the samples were assessed as qualified. Moreover, the accumulative chronic diet risk determined from ADI is very low. To better protect the health of customers, we shall formulate stricter organic phosphorus pesticide control measures and stricter use guidelines, especially for methidathion, triazophos, chlorpyrifos, and profenofos. This study provides potential data for the design of kumquat fruit quality and safety control guidelines and for the reduction in health risks to humans.

Macrocycles are molecular structures extensively used in the design of catalysts, therapeutics and supramolecular assemblies but synthesis procedures that can produce macrocycles in high yield under high reaction concentrations are rare. Here the authors report the use of dynamic hindered urea bond for the construction of urea macrocycles with high efficiency.

Live attenuated influenza vaccines offer broader and longer-lasting protection in comparison to inactivated influenza vaccines. The neuraminidase (NA) surface glycoprotein of influenza A virus is essential for the release and spread of progeny viral particles from infected cells. In this study, we de novo synthesized the NA gene, in which 62% of codons were synonymously changed based on mammalian codon bias usage. The codon-reprogrammed NA (repNA) gene failed to be packaged into the viral genome, which was achievable with partial restoration of wild-type NA sequence nucleotides at the 3′ and 5′ termini. Among a series of rescued recombinant viruses, we selected 20/13repNA, which contained 20 and 13 nucleotides of wild-type NA at the 3′ and 5′ termini of repNA, respectively, and evaluated its potential as a live attenuated influenza vaccine. The 20/13repNA is highly attenuated in mice, and the calculated LD50 was about 10,000-fold higher than that of the wild-type (WT) virus. Intranasal inoculation of the 20/13repNA virus in mice induced viral-specific humoral, cell-mediated, and mucosal immune responses. Mice vaccinated with the 20/13repNA virus were protected from the lethal challenge of both homologous and heterologous viruses. This strategy may provide a new method for the development of live, attenuated influenza vaccines for a better and more rapid response to influenza threats.

Abstract Background This study aimed to determine the effect of PKM2 knockout in STZ induced type 1 diabetes mellitus (T1D) mouse models and to explore the possible mechanism. Method PKM2fl/fl C57BL/6 mouse was backcrossed with Ins‐1cre C57BL/6 mouse to generate β‐cell‐specific PKM2 knockout mouse after tamoxifen administration. The expression level of PKM2 in pancreas tissues was detected by quantitative reverse‐transcription polymerase chain reaction and western blot analysis. The blood glucose levels in STZ induced T1D mouse models were measured to validate the establishment of T1D models. The pathological changes of T1D mouse were examined by hematoxylin and eosin. The oxidative stress (OS) and inflammatory response in T1D mouse were determined by measuring the expression levels of malondialdehyde, superoxide dismutase, and 8‐OHdG in pancreatic tissues and the serum levels of interleukin‐6 and tumor necrosis factor‐α. The ability to catabolize glucose was assessed through intraperitoneal glucose tolerance test and insulin tolerance test. Results β‐cell‐specific PKM2 knockout was successfully achieved in PKM2fl/flcre+ mouse. T1D mouse with PKM2 knockdown had decreased blood glucose level and suppressed cell apoptosis. PKM2 knockout in T1D mouse attenuated β cell injury. OS and inflammatory response in T1D mouse with PKM2 knockout were also suppressed compared with T1D mouse without PKM2 knockout. Conclusion PKM2 knockout in T1D mouse can attenuate OS and inflammatory response as well as decrease blood glucose level, suggesting the potential of PKM2 as a drug target for T1D treatment.

Dissipation of imidacloprid (IMI) and its metabolites (urea, olefin, 5-hydroxy, guanidine, 6-chloronicotinic acid) in Chinese prickly ash (CPA) was investigated using QuEChERS combined with UPLC-MS/MS. Good linearity (r2 ≥0.9963), accuracy (recoveries of 71.8–104.3%), precision (relative standard deviations of 0.9–9.4%), and sensitivity (limit of quantification ≤0.05 mg kg−1) were obtained. After application of IMI at dosage of 467 mg a.i. L−1 for three times with interval of 7 d, the dissipation dynamics of IMI in CPA followed first-order kinetics, with half-life of 6.48–7.29 d. IMI was the main compound in CPA, followed by urea and guanidine with small amounts of olefin, 5-hydroxy, and 6-chloronicotinic acid. The terminal residues of total IMI and its metabolites at PHI of 14–21 d were 0.16–7.80 mg kg−1 in fresh CPA and 0.41–10.44 mg kg−1 in dried CPA, with the median processing factor of 3.62. Risk assessment showed the acute (RQa) and chronic dietary risk quotients (RQc) of IMI in CPA were 0.020–0.083% and 0.052–0.334%, respectively. Based on the dietary structures of different genders and ages of Chinese people, the whole dietary risk assessment indicated that RQc was less than 100% for the general population except for 2- to 7-year-old children (RQc of 109.9%), implying the long-term risks of IMI were acceptable to common consumers except for children.

The present study was carried out to profile the dissipation patterns and residues of five pesticides (triazophos, profenofos, chlorpyrifos, etoxazole and bifenthrin) on kumquat using QuEChERS method coupled with HPLC-MS/MS. The corresponding dietary health risks were also estimated. In the method validation, satisfactory results of good linearity (r2 ≥ 0.9956), sensitivity (limits of quantification ≤0.01 mg/kg), recoveries (71.0–95.7%) with relative standard deviations (0.70–9.4%) were obtained. The half-lives of the five pesticides in kumquat were 13.6–38.5 d under field conditions according to first-order kinetics. Based on the final residue experiment, dietary exposure risks of profenofos, chlorpyrifos, etoxazole and bifenthrin were all acceptably low, with RQc and RQa values of 0.00199–0.122 and 0.00145–0.200, respectively. However, exposure intake of triazophos posed unacceptable acute and chronic health risks for Chinese residents, especially for children with RQa and RQc up to 4.25 and 2.19. Forbidden use suggestion of triazophos and recommended MRLs of profenofos and bifenthrin were put forward in kumquat for safe production and consumption. This work was significant in providing guidance on appropriate application and MRL establishment of pesticides in kumquat.

With the rapid advancement of connected and autonomous vehicles (CAV) in terms of testing and usage, the implementation of the autonomous interchange in the real world has become more realistic than before. Since the beginning of the last decade, the concept of autonomous intersection has been developed with slightly different techniques. This paper introduces a next-generation interchange control algorithm (NIC) that deals with service interchange terminals for the CAV with the ability to adjust the dimensions and geometric designs. It proposes six different geometric designs that have been modeled in a simulation software package (VISSIM) to employ the NIC algorithm. The system depicted a real-world scenario of an interchange with a slight modification on the geometric design to provide smoother entry to the interchange terminal. The analysis of the proposed designs in terms of throughput, capacity, delay, and volume-to-capacity (V/C) ratio indicated different performance measurements based on the analyzed traffic movements. Tight turn with one left dedicated turn provided the highest performance, while wide turn with two left shared turns indicated the lowest performance. Both NIC designs demonstrated significantly higher throughputs and significantly lower delays compared to a current traffic signal system. By applying the NIC with the existence of the CAV, the operation of highway interchange can be significantly improved.

Rule-based and optimization-based autonomous intersection management (AIM) policies have been evaluated against traditional signal controls in terms of intersection safety, efficiency and emission. As one of AIM policies, reservation-based control has further taken advantage of the benefits of AIM, especially via optimization approaches. This paper presents a time-independent trajectory optimization approach for connected and autonomous vehicles under reservation-based intersection control. The existing approaches assign an arrival time and speed to vehicles ahead of entering the intersection. However, the vehicles may not follow the planned trajectory once the traffic condition varies sharply and thus the trajectory solution becomes infeasible with respect to the assigned arrival time and speed. The proposed approach aims to solve the fail-follow problem by separating the optimization between arrival time, speed, and trajectory planning by optimizing the trajectory without arrival time and speed predetermined. The approach finds the optimal solution in terms of the intersection efficiency meanwhile keeps the feasibility of trajectory planning by formulating the variation of acceleration rate and breaking a whole trajectory into an enlarged set of segments. Two different control strategies, BATCH and ZONE, are also proposed to test the performance of the optimization approach in comparison with another Dynamic Batch strategy. The results validate that the proposed approach can adapt to extremely high traffic demand scenario. Sensitivity analyses also evaluate the performance of the proposed approach under different problem settings in terms of intersection efficiency.

Typical traffic control and operations at work zones are not effective in mitigating the work zone bottleneck, as the common early merge behavior limits utilizing the available capacity in the closed lane. Recently, using late merge strategy is encouraged to fully utilize the closed lane, therefore increasing efficiency of the work zone. In practice, the late merge strategy does not work effectively due to the lack of compliance by drivers. This paper is aimed at exploring two work zone late merge strategies with and without enabling connected vehicles (CV) technology. This first strategy is the late merge without CV enabled. The second strategy incorporates CV-enabled cooperative merge to enhance the work zone efficiency through communications between vehicles in the open and closed lanes. The CV-enabled strategy employs a rule based decentralized control algorithm. The paper has implemented both late merge strategies in microsimulation and evaluated the operational performances. Three major performance measures, which are throughput, delay and queue length, are used in the evaluation. The results indicate that the operational performance of the CV-enabled cooperative late merge is superior to the late merge strategy without CV in most cases, especially when traffic demand is moderate, while both late merge strategies outperform the traditional early merge strategy. Moreover, effects of the control variables including the traffic demand, heavy vehicle percentage, lane distribution percentage, late merge length, reduction of gap acceptance factor, and compliance rate, have been analyzed. Ultimately, sensitivity analysis has been performed to investigate the trend of effects from the significant variables control.

Waste cooking oil (WCO) and stearic acid (SA) were considered to be renewable and readily available bio-oil resources to produce bio-fuel via catalytic hydrodeoxygenation. The most often used catalysts for hydrodeoxygenation were the vulcanized NiMo catalysts, however, the vulcanized catalysts tend to deactivate due to the loss and oxidation of sulfur upon treating bio-oil with high oxygen content. Therefore, it is necessary to develop sulfur-free catalysts. In present study, the reduced NiMo catalysts were prepared by loading Ni and Mo on different supports (γ-Al2O3, HZSM-5, β-zeolite, activated carbon and bentonite) using incipient-wetness impregnation method, and followed by H2 reduction. The effects of the properties of supports (acidity, porosity) and H2 reduction on the dispersity and valence state of metal components were investigated. The hydrodeoxygenation activities of the reduced catalysts were evaluated by catalytic WCO and SA. It was found that when Ni2+ was loaded on a slightly acidic supports such as γ-Al2O3 and activated carbon, Ni2+ was easier to be reduced, and aggregate to form larger particles. When Ni2+ was loaded on a strongly acid supports e.g. HZSM-5, β-zeolite or bentonite, Ni2+ was more difficult to be reduced but could uniformly disperse on the supports. The aggregated Ni atoms could promote the formation of C17, while the dispersed Ni atoms could promote the formation of C18. γ-Al2O3 supported catalyst achieved the highest SA (97.9%) and WCO (80%) conversion under the mild reaction conditions of 300 °C and 2 MPa H2 for 3 h, exhibiting the superior catalytic activity.

Autonomous Vehicles (AV) technology is emerging. Field tests on public roads have been on going in several states in the US as well as in Europe and Asia. During the US public road tests, crashes with AV involved happened, which becomes a concern to the public. Most previous studies on AV safety relied heavily on assessing drivers' performance and behaviors in a simulation environment and developing automated driving system performance in a closed field environment. However, contributing factors and the mechanism of AV-related crashes have not been comprehensively and quantitatively investigated due to the lack of field AV crash data. By harnessing California's Report of Traffic Collision Involving an Autonomous Vehicle Database, which includes the AV crash data from 2014 to 2018, this paper investigates by far the most current and complete AV crash database in the US using statistical modeling approaches that involve both ordinal logistic regression and CART classification tree. The quantitative analysis based on ordinal logistic regression and CART models has successfully explored the mechanism of AV-related crash, via both perspectives of crash severity and collision types. Particularly, the CART model reveals and visualize the hierarchical structure of the AV crash mechanism with knowledge of how these traffic, roadway, and environmental contributing factors can lead to crashes of various serveries and collision types. Statistical analysis results indicate that crash severity significantly increases if the AV is responsible for the crash. The highway is identified as the location where severe injuries are likely to happen. AV collision types are affected by whether the vehicle is on automated driving mode, whether the crashes involve pedestrians/cyclists, as well as the roadway environment. The method used in this research provides a proven approach to statistically analyze and understand AV safety issues. And this benefit is potential be even enhanced with an increasing sample size of AV-related crashes records in the future. The comprehensive knowledge obtained ultimately facilitates assessing and improving safety performance of automated vehicles.

Six plant growth regulators frequently used in fruit were detected using UPLC–MS/MS including chlormequat chloride, GA3, forchlorfenuron, paclobutrazol, thidiazuron and 2,4-D. A modified QuEChERS method was used in pretreatment: each compound was extracted with acetonitrile containing 1% acetic acid and cleaned with a mixture of ostade-cylsilane (ODS) and MgSO4. The detection was performed using electrospray ionization (ESI) and multiple reaction monitoring mode (MRM) and quantified using the external standard method. Results indicated that the calibration curves of six plant growth regulators were linear within the range of 1–100 µg⋅kg-1 with correlation coefficients over 0.999. The recovery at four spiked concentrations, 1, 20, 50, and 100 µg⋅kg-1, ranged from 60.77% to 119.43% and relative standard deviation (RSD) were between 0.18% and 32.94%. The limits of detection (LOD) and limits of quantitation (LOQ) were 0.01–0.14 µg⋅kg-1 and 0.02–0.46 µg⋅kg-1, respectively, for apples, 0.01–0.18 µg⋅kg-1 and 0.03–0.59 µg⋅kg-1 for grapes, 0.01–0.08 µg⋅kg-1 and 0.04–0.28 µg⋅kg-1 for kiwis, 0.003–0.110 µg⋅kg-1 and 0.01–0.38 µg⋅kg-1 for peaches, and 0.01–0.08 µg⋅kg-1 and 0.03–0.25 µg⋅kg-1 for oranges.

In order to understand how the uncertainties in the output can be apportioned to different sources of uncertainties in its inputs, it is critical to investigate the sensitivity of MOVES model. The MOVES model sensitivity for regional level has been well studied. However, the uncertainty analysis for project level running emissions has not been well understood. In this research, the MOVES model project level sensitivity tests on running emissions were conducted thru the analysis of vehicle specific power (VSP), scaled tractive power (STP), and MOVES emission rates versus speed curves. This study tested the speed, acceleration, and grade-three most critical variables for vehicle specific power for light duty vehicles and scaled tractive power for heavy duty vehicles. For the testing of STP, four regulatory classes of heavy duty vehicles including light heavy duty (LHD), medium heavy duty (MHD), heavy heavy duty (HHD) and bus were selected. MOVES project running emission rates were also tested for CO, PM2. 5, NOx and VOC versus the operating speeds. A Latin Hypercube (LH) sampling based on method for estimation of the “Sobal” sensitivity indices shows that the speed is the most critical variable among the three inputs for both VSP and STP. Acceleration and grades show lower response to the main effects and sensitivity indices. MOVES emission rates versus speeds curves for light duty vehicles show that highest emission occurs at lower speed range. No significant differences on emission rates among the regulatory classes of heavy duty vehicles are identified.

In this study, we investigated the influence of rhizosphere microorganisms on seed germination and root metabolism in Cerasus sachalinensis Kom. We inoculated C. sachalinensis plants with suspensions of dominant bacterial strains isolated from their rhizosphere. Four bacterial strains each with significant growth-promoting or growth-inhibiting effects were screened from the efficient root-colonizing microorganisms. The number of actinomycetes increased and that of fungi decreased significantly in the seedling rhizospheres after rhizobacteria treatment. The growth-promoting bacteria slightly affected the respiration rates and respiratory pathway enzymes, but significantly improved root viability, root carbohydrate concentration and seedling growth. Bacillus cereus, Staphylococcus sp. and Pseudomonas fluorescens were identified as the growth-promoting rhizobacteria; one strain could not be identified. After inoculation with the growth-inhibiting bacteria, the number of fungal colonies in the seedling rhizospheres increased and root viability and respiration rate as well as starch and sucrose accumulation in the roots significantly decreased. The glycolysis, pentose phosphate and alternative oxidase pathways became the major pathways of respiratory metabolism after inoculation with the growth-inhibiting bacteria. The height, leaf number, growth and dry weight of the seedlings decreased significantly in plants inoculated with the growth-inhibiting bacteria. Inoculation of C. sachalinensis rhizosphere with growth-promoting and growth-inhibiting bacteria affected the soil environmental factors such as microbial group composition, nutrient concentration and seedling biomass.

AIM:The aim was to evaluate the impact of metabolic syndrome (MS), MS individual components and 32 kinds of MS specific component combinations on all-cause mortality risk in a fixed cohort of MJ check-up population. METHODS:We observed the events of death in a fixed cohort, where the population was composed of 45,542 individuals aged 35-74 who were examined at MJ Health check-up Center in 1997 as baseline examination, and were followed up to 2005. Median duration of follow-up was 7.44 years. MS was defined according to the National Cholesterol Educational Program (the revised NCEP-ATPIII for Asian in 2004), the prevalence of MS was standardized according to China's fifth census data. We constructed common Cox regression model, simultaneously adjusting the classic risk factors (such as age, sex, smoking, alcohol drinking, physical activity, family history, etc.) to examine the relationship between MS, MS individual components and 32 kinds of MS specific component combinations on the occurrence of death with the fixed cohort. RESULTS:The standardized prevalence of MS was 29.75% (male: 30.36%, female: 29.51%). There were 1,749 persons who died during the median 7.44-years follow-up, the mortality rate was 46 per 10,000 person years. The mortality rates were 71 and 35 per 10,000 person years for those with and without MS, respectively. After adjustment for age, sex and classical risk factors, compared with subjects without MS, the hazard ratio of all-cause mortality was 1.26 (95% CI: 1.14-1.40). The appropriately landscaped highway scenes may not only help improve road safety and comfort but also help protect ecological environment. Yet there is very little research data on highway length threshold with consideration of distinctive landscape patterns. Against this backdrop, the paper aims to quantitatively analyze highway landscape’s effect on driving behavior based on drivers’ physiological performance and quantify highway length thresholds under three typical landscape patterns, namely, “open,” “semiopen,” and “vertical” ones. The statistical analysis was based on data collected in a driving simulator and electrocardiograph. Specifically, vehicle-related data, ECG data, and supplemental subjective stress perception were collected. The study extracted two characteristic indices, lane deviation and LF/HF, and extrapolated the drivers’ U-shaped physiological response to landscape patterns. Models on highway length were built based on LF/HF’s variation trend with highway length. The results revealed that the theoretical highway length threshold tended to increase when the landscape pattern was switched to open, semiopen, and vertical ones. And the reliability and accuracy of the results were validated by questionnaires and field operational tests. Findings from this research will assist practitioners in taking active environmental countermeasures pertaining to different roadside landscape patterns.

The combined effect of CO2 and O2 also resulted in relatively high protein expression under high oxygen conditions (0.3% CO2 and 100% O2). Moreover, metabolomics assays were performed to gain insight into metabolic changes, which showed that CO2 slightly improved energy metabolism and redox balance. In particular, the extra supplied CO2 activated the decarboxylating reactions and removed toxic metabolites to recover the protein synthesis activity. The origin and destination (O-D) of public transit passengers are important for the planning and operation of the transit system. However, only 46% of public transit agencies have a smart card system in the US, and most of them require an entry-only tap, which prohibits identifying passenger destinations unless utilizing an estimation model. Therefore, there is a need for a cost-effective and automated solution to facilitate the majority of the US transit agencies in recognizing the origin and destination of passengers as well as capturing passenger transfers. This paper created a novel algorithm for transit agencies to count passengers with the consideration of transfers using a costeffective Wi-Fi sensing--based approach. Two pilot studies were conducted in the city of Louisville, Kentucky on three different bus routes to explore the feasibility of the method. AWi-Fi detector was installed in the bus to detect passengers, and a manual counting was performed to be used as ground truth data. After data collection, the proposed algorithms were applied to optimize the detection radius and to eventually find origins, destinations, and transfers. Analysis revealed that the proposed Wi-Fi--based approach is capable of recognizing 78.7% of the total passengers as well as detecting their boarding and alighting activities. The paper demonstrates the ability of the proposed method to detect passengers with a reasonable detection rate by using Wi-Fi technology on bus routes, which makes it feasible for transit agencies to conduct frequent and low-cost network-level passenger O-D studies. Two-stage harvesting is the main method used for the mechanized harvesting of peanuts in China, in which the pickup device is a core part of the combine harvester. In order to solve the problem of pod loss caused by “stack” and “loss picking” from peanut plants when using a traditional spring-finger cylinder pickup device, an optimal spring-finger cylinder peanut pickup mechanism was designed and its picking properties were tested. Based on the picking characteristics and picking force analysis when considering a peanut plant windrow, the ideal picking attitude and swing rule of the spring-finger were determined, and the cam slide, as the core element of the spring-finger cylinder peanut pickup device, was optimized. A mathematical model of the cam cylinder center line was established according to the swing law of the four picking stations utilizing the spring-finger. MATLAB and ADAMS were used to establish a simulation of the pickup mechanism, and kinematics and dynamics simulation analyses of the pickup mechanism were carried out. According to the design results, a prototype was constructed and a running pickup test was carried out. The peanut plant picking experiments indicated that the phenomenon of peanut plant stacking had significantly disappeared. Furthermore, through response surface analysis, the optimal working parameters of the picking device were obtained as follows: the forward speed Vm was 48.0 m/min, the rotational speed N was 50 r/min, and the ground height H was −16.8 mm. The picking rate of peanut plants was 99.21% and the pod loss rate was 1.79% under two harvesting conditions, with a peanut plant moisture content of 15% to 17%. This study provides technical support for the future design of picking devices for two-stage peanut pickup harvesters.

This paper explores the efficiency of a novel merging system based on a cooperative late merge strategy (CLMS) to mitigate the capacity reduction in work zones due to lane closure. Cooperative late merge strategies in connected vehicles (CV) and connected and autonomous vehicles (CAV) environments are formulated to enhance throughput by reducing gaps and increasing the synchronized speed in the work zone. We propose decentralized and centralized systems based on vehicle-to-vehicle and vehicle-to-infrastructure communication. The decentralized CLMS incorporates a modified lane-changing model to reflect the cooperative feature under the CV environment. The centralized CLMS is developed to further optimize the work zone throughput based on gap reduction and speed harmonization features enabled by CAV. The results prove that the decentralized CLMS outperforms other systems by increasing throughput as well as reducing delay and queue length. The centralized CLMS demonstrated substantial improvements compared to other systems. The simulation results prove that the decentralized CLMS improves capacity by 17% and the centralized CLMS by 45%, when compared to a traditional work zone system. [ABSTRACT FROM AUTHOR]

Cadmium (Cd) accumulation in rice grain is of health concern. Identifying genes involved in grain Cd accumulation and performing molecular breeding may reduce it. In this study, knockout of OsNRAMP2, a member of the NRAMP family, reduced grain Cd concentrations by more than 38%, and overexpressing OsNRAMP2 increased grain Cd concentrations by more than 50%. Physiological experiments showed that OsNRAMP2 facilitated Cd translocation from root to shoot by positively regulating Cd efflux from the vacuoles. At filling stage, OsNRAMP2 was highly expressed in all tissues except for husk, suggesting its role in Cd remobilization. Changes in OsNRAMP2 expression affected the concentrations of Fe, Mn, Zn, and Cu in grain and also affected rice growth. Phylogenetic analysis showed that the distribution of OsNRAMP2 haplotypes between japonica and indica was different. Among the four haplotypes of OsNRAMP2, Hap 1, with a 6-bp nucleotide insertion in exon 1, had grain Cd concentration at least 45.3% lower than any of the other three haplotypes. Almost all (99.3%) japonica accessions but rare indica accessions (4.44%) from the 3K sequenced rice genomes carry Hap 1 of OsNRAMP2. Our study sheds light on the molecular mechanism of grain Cd accumulation and provides a promising target for low-Cd rice breeding.

ABSTRACT Two-stage harvesting is the main method for performing the mechanized harvesting of peanuts, and the picking device is a core part of the combined harvester in China. In order to solve the problem of pod loss caused by the "stacking", "impact" and "throwing up" of peanut plants by a traditional cam-slide spring-finger cylinder picking device, the shovel-finger and cylinder peanut-picking device was developed and used in a picking-up performance test based on the study of peanut-plant strip-picking characteristics. The mathematical model of the mechanism was established by analyzing the structure of the mechanism and the peanut-plant strip-picking characteristics, and the parameters of the mechanism were optimized using the objective function method. The prototype was developed and tested. The experiments in which the prototype was used to collect peanut plants indicated that the phenomenon of peanut plants being stacked and thrown disappeared. Through a response surface analysis and prototype test, the optimal working parameters of the picking device were obtained: the forward speed V was 48.0 m/minute, the rotational speed N was 45.3 r/minute and the ground height H was -18 mm. The peanut-plant picking rate was 98.9% and the fruit loss rate was 2.8% under two different harvesting conditions for which the peanut-plant moisture content was 15~17%.

As a model crop for monocots research, rice is also one of the most important food crops in the world. Rice plays an extremely important role in ensuring food security, since China is the world's largest producer and consumer of rice. Upgrading and improvement of rice varieties is an effective way to ensure the continuous increase of rice yield. As the heavy reliance on the experience of breeders, the traditional breeding method is time-consuming, poor in precision and low in predictability, which can no longer meet the needs of modern breeding. As a new generation of gene editing technology after zinc finger nuclease (ZFN) and transcription-activating-like effector nuclease (TALEN) technologies, CRISPR/Cas9, which can accurately and directionally modify the functional genes that control important traits without changing the genetic background of the receptor to achieve the rapid upgrading and improvement of traits, has gradually become an important tool for animal and plant scientific research and crop breeding. At present, CRISPR/Cas9 gene editing technology has been widely used in targeted improvement of important traits and germplasm resource innovation of rice. In the study, the working principle of CRISPR/Cas9 gene editing technology and its research progress in rice disease resistance, quality, fertility and abiotic stress were reviewed. It will provide a new insight for application prospect of gene editing in rice breeding and provide a reference for the further application of CRISPR/Cas9 gene editing technology in crop targeted improvement in the future.

Type 1 diabetes (T1D) is an autoimmune disease characterized by the immune-mediated destruction of insulin-producing β cells. Recent studies showed that in addition to malaria, artemisinin and its derivative, artesunate (AS), could alleviate several autoimmune diseases. However, whether AS has a role in the prevention or treatment of T1D is still unknown. Therefore, in this study we administrated AS or DMSO in the drinking water of nonobese diabetic (NOD) mice, a mouse model of T1D. We found that AS administration significantly prevented the incidence of T1D. The frequency of IL-4-producing CD4+ single-positive T cells and CD8+ T cells was significantly elevated, and IFN-γ-producing T cells were reduced in the spleen and pancreatic lymph nodes. In the pancreas, the skewing to IL-4-producing T cells was also observed. In addition, more regulatory T cells were found in the pancreas. mRNA levels of proinflammatory cytokines, including TNF-α and IL-6, were decreased. In addition, AS administration promoted the functional maturity of β cells in vitro. Our findings demonstrate that AS administration can prevent T1D in NOD mice mainly by reducing autoimmune T cells and increasing protective T cells. This study was conducted to clarify the roles of macronutrients in regulating plant primary and secondary metabolism of Scutellaria baicalensis Georgi. #Methods] The contents of chlorophyll (Chl) and soluble sugar were detected with ultraviolet spectrophotometry. The activities of phenylalanine ammonia-lyase (PAL) , cinnamate 4-hydroxylae (C4H) and chalcone synthase (CHS) were determined with ultraviolet spectrophotometry. Secondary metabolites were detected by high performance liquid chromatography (HPLC). # Results ] The content of chlorophyll in treatments N, P and K was increased, showing significant diference from that of the control (P < 0. 05) . Among them, Treatment N has the highest content of chlorophyll. Soluble sugar content in treatments N , P and K (K>N>P> CK) was increased considerably and had significant difference from that of the control (P < 0. 05). Compared with the control, PAL and C4 H ac-tivity in treatments N, P and K were increased dramatically by 186. 57% , 134. 09% , 306.91% and 73. 21% , 28. 91% , 247. 57% , respectively. CHS activity was also increased sharply in treatments N, P and K. Scutellarin, baicalin, wogonoside, baicalein, wogonin and oroxylin A contents in all treatments N, P and K (K > N >P) were increased, and had significant difference from that of the control (P<0.05). # Conclusions] The results showed that macronutrients can increase leaf chlorophyll content, soluble sugar content, PAL activity, C4H activity and secondary metabolites of S. baicalensis. To improve its cleaning performance, the peanut sheller’s air–screen cleaning device was used as the research object, and a numerical simulation was carried out. A scheme was proposed to optimize the installation angle of the screen surface of the air–screen cleaning device, and the influence of the airflow velocity of the sieve surface before and after the structural improvement of the machine on the airflow distribution of the whole chamber was evaluated. At the same time, the computational fluid dynamics–discrete element method (CFD–DEM) of coupling simulation was used to carry out single-factor simulation tests on the improved air–screen cleaning device, analyze the movement trajectory, velocity, and displacement of the extrudates in the cleaning chamber, characterize the separation law of extrudates, and determine the parameter ranges for each factor. In addition, based on the central composite Box–Behnken design, an orthogonal simulation test was carried out with three factors, including fan speed, amplitude, and vibration frequency, and the influence of each factor on the kernel loss rate and impurity rate was investigated. Furthermore, the influencing factors were optimized, and the optimal parameter combination was obtained; when the fan speed was 1682.72 r/min, the amplitude was 3.74 mm, and the vibration frequency was 492.86 Hz, it was more conducive to the cleaning of kernels. Finally, the accuracy of the simulation and optimization methods was verified via the machine test, and the optimal parameter combination was obtained: at a fan speed of 1680 r/min, an amplitude of 3.7 mm and a vibration frequency of 490 Hz were achieved, and the kernel loss rate and impurity rate were 2.01% and 2.42%, 3.71 and 4.42 percentage points lower than those before optimization, respectively.

Studying the evolutionary patterns of rice agronomic traits in South China and analyzing the characteristics of rice improvement can provide insights into the developmental trajectory of rice breeding in South China and can guide further enhancement of variety yield. In this study, widely promoted varieties and core parents developed through dwarf breeding in the southern region, as well as landraces, were collected and planted in three different ecological regions. A total of 18 agronomic traits were investigated related to heading date, plant type, panicle type, grain type, and yield, and multiple comparisons, a correlation analysis, and a path analysis were conducted. The results indicate that dwarf breeding has significantly increased the yield of inbred indica rice varieties in South China. However, a reduction in plant height has also resulted in a reduction in flag leaf, shorter panicles, and decreased biomass, which have led to metabolic source and storage capacity deficiencies and limited yield potential. To address these limitations, breeders have employed strategies such as increasing flag leaf width, spikelet density, number of primary branches, and grain number per panicle. These measures have led to a gradual increase in yield. Additionally, starting from the 1980s, high-quality rice breeding has been pursued in South China, resulting in slender grain shape and reduced thousand grain weight. Given that total grain number per panicle has already increased significantly and the thousand grain weight cannot be reduced further, enhancing the effective tiller number, which decreases year by year, becomes an important approach to increasing the yield of inbred indica rice varieties in South China.

A magnetic cation-exchange resin (MCER) was prepared by copolymerization of oleic acid-grafted magnetite with styrene, divinylbenzene (DVB), and triallylisocyanurate (TAIC) for removing Cd(II) from wastewater. A non-magnetic cation-exchange polystyrene resin (CEPR) was also prepared as a reference. Structural and morphological analyses revealed that the MCER and CEPR were mesoporous microspheres; the MCER contained about 25% Fe3O4. The influence of temperature, pH, contact time, and the initial concentration of Cd(II) on the adsorption of Cd(II) was investigated. The maximum adsorption capacity of the MCER reached 88.56 mg/g, which was achieved at 343 K using a Cd(II) initial concentration of 200 mg/L. The adsorption processes attained equilibrium within 120 min for the MCER and 300 min for the CEPR, and were well described by a pseudo-second-order kinetic model. Furthermore, the equilibrium adsorption data fitted the Freundlich isotherm model better than the Langmuir model. The superior magnetic response and regeneration of the MCER make it a good candidate as an adsorbent for removing Cd(II) from wastewater. [ABSTRACT FROM AUTHOR]

Abstract Existing research suggests that the human immune system and immune cells are involved in the pathogenesis of nephrotic syndrome, but there is still a lack of direct evidence. This study tried to analyze the profiling of immune cells in the peripheral blood of steroid-sensitive nephrotic syndrome (SSNS) patients and steroid-resistant nephrotic syndrome (SRNS) patients before and after standard steroid treatment to clarify the immunological mechanism of nephrotic syndrome patients. The number and proportion of CD4 + T cells in patients with nephrotic syndrome remained unchanged. However, there is an imbalance of Th1 and Th2 and an excessive increase of Th17 cells. The number of CD8 + T cells and the number of effector CD8 + T cells in them increased significantly, but only in SSNS, the number of activated CD8 + T cells increased, and the number of activated Treg cells decreased significantly. Nephrotic syndrome patients also have B cell disorder, and it is more prominent in SSNS patients. Compared with the normal control, only the number of B cells and plasmablast in SSNS patients increased significantly (Z = − 2.20, P = 0.028). This study also observed that transitional B cells decreased in both SSNS and SRNS patients, but SSNS patients' decrease was lower than in SRNS patients. Compared with normal controls, monocytes in patients with nephrotic syndrome decreased significantly. The main reason was that Non-classical Monocyte decreased, while Classical Monocyte increased slightly. The total number of NK cells did not change, but the internal cell subgroups' composition occurred. Changes, realized as CD56hi NK cells increased, CD56low NK cells decreased; and the above trend is more evident in SSNS patients. Patients with nephrotic syndrome have immune disorders, including T cells, B cells, Monocytes, and NK cells. It can be confirmed that immune factors are involved in the pathogenesis of the nephrotic syndrome.

Opisthopappus longilobus (Opisthopappus) and its descendant species, Opisthopappus taihangensis, commonly thrive on the Taihang Mountains of China. Being typical cliff plants, both O. longilobus and O. taihangensis release unique aromatics. To determine the potential differentiation and environmental response patterns, comparative metabolic analysis was performed on O. longilobus wild flower (CLW), O. longilobus transplant flower (CLT), and O. taihangensis wild flower (TH) groups. Significant differences in the metabolic profiles were found, not within O. longilobus, but between O. longilobus and O. taihangensis flowers. Within these metabolites, twenty-eight substances related to the scents were obtained (one alkene, two aldehydes, three esters, eight phenols, three acids, three ketones, three alcohols, and five flavonoids), of which eugenol and chlorogenic were the primary aromatic molecules and enriched in the phenylpropane pathway. Network analysis showed that close relationships occurred among identified aromatic substances. The variation coefficient (CV) of aromatic metabolites in O. longilobus was lower than O. taihangensis. The aromatic related compounds were significantly correlated with the lowest temperatures in October and in December of the sampled sites. The results indicated that phenylpropane, particularly eugenol and chlorogenic, played important roles in the responses of O. longilobus species to environmental changes.

In order to explore possible development process of red tides caused by Prorocentrum donghaiense, effects of concentrations of P. donghaiense and Oxyrrhis marina on the feeding behaviour of Oithona brevicornis were investigated. The results showed that within the concentration range of P. donghaiense, 1.0~5.0×104 cells·mL-1, ingestion rates (IRs) and faecal pellet production rates (FPPRs) of O. brevicornis on P. donghaiense increased with increasing concentrations of P. donghaiense, the maximum IR and FPPR were 620 cells·ind-1·h-1 and 31.67 pellet·copepod-1·d-1, respectively. When the concentration of P. donghaiense was 10.0×104 cells·mL-1, the IR value decreased to 400 cells·ind-1·h-1 and the FPPR value decreased to 13.33 pellet·copepod-1·d-1, respectively. Within the concentration range of P. donghaiense, 1.0~10.0×104 cells·mL-1, filtration rates (FRs) of O. brevicornis decreased with increasing concentrations of P. donghaiense. The results also showed that O. brevicornis could ingest O. marina fed P. donghaiense, and within the concentration range of O. marina, IRs of O. brevicornis on O. marina increased with increasing concentrations of O. marina, while its FRs decreased, the maximum IR value and FR value were 300 cells·ind-1·h-1 and 0.23 ml·ind·h-1, respectively. Within the concentration range of O. marina, FPPRs of O. brevicornis increased with increasing concentrations of O. marina, the maximum FPPR was 21.67 pellet·copepod-1·d-1, and FPPRs had a good linear relationship with IRs. In this study, "Copepodsred tide algae" and "Copepods-protozoa-red tide algae" food chain models can provide references for the development process and regulating method of red tides caused by P. donghaiense. Abstract Background 13q33–q34 microdeletions are rare chromosomal aberrations associated with a high risk of developmental disability, facial dysmorphism, cardiac defects and other malformation of organs. It is necessary to collect and report evidence of this rare chromosome mutation to improve the prognosis of this rare disease. Case presentation We report a patient harboring an 11.56 Mb microdeletion at 13q33.1–34 region, which contains about 30 OMIM genes. Besides the common clinical manifestations such as facial dysmorphism, developmental delay, intellectual disability, epilepsy, and congenital heart disease, she also suffered from a reduced anogenital distance, hematuria and left renal hypoplasia. Most related cases were characterized by facial deformity and heart defects, but there were few reports on renal malformation, especially regarding renal hypoplasia with hematuria. Conclusion We have reported a patient suffering from a reduced anogenital distance, hematuria and left renal hypoplasia. A de novo 11.56 Mb deletion ranging from 13q33.1 to 13q34 (Chr13:103542220–115,106,996) was found by SNP-array analysis. It might be the first time for hematuria and renal hypoplasia to be reported as symptoms of 13q33-q34 deletion syndrome Neurodevelopmental disability, heart defects and urogenital/anorectal anomalies may be resulted from common or overlapping regions of deletion in chromosome bands 13q33.1-q34 and may share a common molecular mechanism.

This study was aimed to investigate the growth characteristics of living diet Platymonas subcordiformis for aquaculture animals and its harmful organism Oxyrrhis marina in co-culture systems to provide experimental evidences for discussing their succession processes. The colour changes of culture media of P. subcordiformis and growth characteristics of O. marina and P. subcordiformis in co-culture were analysed by the combined methods of macro-observation, microscopic examination and microscopic counting. The results showed that with the decrease of the initial density of O. marina cell (0.65×104 cells mL-1, 0.37×104 cells mL-1 and 0.11×104 cells mL-1 in P. subcordiformis culture media), the time that the population of O. marina reached the stationary phase required longer, were 4d, 5d and 7d after inoculated by O. marina, respectively, and the death time of all cells of P. subcordiformis became longer, were 9d, 10d and 12d after inoculated by O. marina, respectively. After inoculation 9d, pale pink appeared in upper layer of culture media of P. subcordiformis in flasks and the density of O. marina was 2.10×105 cells mL-1. The results also indicated that the variation tendency of cell densities of O. marina and P. subcordiformis cultured in aquariums were consistent with that in flasks basically, pink flocs appeared in upper layer of culture media of P. subcordiformis after inoculation 12d and the density of O. marina was 2.10×106 cells mL-1. The experimental results showed that the feeding of O. marina on P. subcordiformis was affected by the initial concentration of O. marina, and P. subcordiformis populations were evolved to O. marina populations during the 15 days culture period, the colour of culture media appeared pale pink to pink depends on density of O. marina was confirmed, too. Also, the colour changes of P. subcordiformis culture media are indications of pollution by O. marina, which will be important for culture process of P. subcordiformis. [ABSTRACT FROM AUTHOR]

This report describes an 8-year-old male who presented with clinical manifestations including systemic edema, heavy proteinuria, hypoproteinemia, and persistent hypocomplementemia. Arachnoid cysts and focal nodular hyperplasia were also detected. Imaging examination and renal biopsy were performed, and Abernethy malformation type II with immune complex-mediated membranoproliferative glomerulonephritis was considered the diagnosis. Due to the persistence of embryonic vessels, Abernethy malformation is a rare congenital vascular malformation of the splanchnic venous system, which can be classified as type I (end-to-side shunt) and type II (side-to-side shunt). Abernethy malformation with glomerulonephritis remains extremely rare. In the patient described, glomerulonephritis mediated by immune complex with “full-house” positive immunohistochemistry was confirmed on renal biopsy. In addition, he was treated with glucocorticoids and tacrolimus. Whether surgical treatment is necessary should be determined according to the state of the disease in the later stages. The present case reflects the association between the congenital portosystemic shunt and the renal region and, to the authors' knowledge, may be the first report to describe arachnoid cysts as a symptom of Abernethy malformation.

This study was aimed to investigate the growth characteristics of Oxyrrhis marina and Chattonella marina in co-culture to provide experimental evidences for discussing successions of harmful algal blooms (HABs) and coastal biological communities. The colour changes of culture media of C. marina and growth characteristics of O. marina and C. marina in co-culture were analysed by the combined methods of macroobservation, microscopic examination and counting. In co-culture, the colours of culture media of C. marina had changed and their transparencies had increased with increasing elapsed incubation time after inoculated by O. marina under different initial cell densities. With the increase of the initial density of O. marina (0.17x104 cells/mL, 0.50x104 cells/mL and 0.64x104 cells/mL in C. marina culture media), the time required, that the populations of O. marina reached the stationary phases, was shorter i.e. 6d, 5d and 3d after inoculated by O. marina, respectively, and the death time of all cells of C. marina became shorter, i.e. 7d, 6d and 4d after inoculated by O. marina, respectively. During the 15 days culture period, all C. marina populations were evolved to O. marina populations. Residues of C. marina adhering to precipitates and chromatophores scattering in the culture media could strengthen the colour of culture media, C. marina populations were evolved to O. marina populations respectively within the concentrations designed in co-culture in this experiment. Disturbance feeding was one of the reasons for successions, and the results provide experimental evidences for discussing successions of red tides and coastal biological communities. Porous Y2O3 microparticles 500 μm in size were obtained, when 1 wt%-ammonium alginate aqueous solution was dropped into 0.5 M-YCl3 aqueous solution by a Pasteur pipette and the resultant gel microparticles were heat-treated at 1100°C. Small pores less than 1 μm were formed in the microparticles by the heat treatment. The bulk density of the heat-treated microparticle was as low as 0.66 g cm−3. The chemical durability of the heat-treated microparticles in simulated body fluid at pH = 6 and 7 was high enough for clinical application of in situ radiotherapy. Although the size of the microparticles should be decreased to around 25 μm using atomizing device such as spray gun for clinical application, we found that the porous Y2O3 microparticles with high chemical durability and low density can be obtained by utilizing gelation of ammonium alginate in YCl3 aqueous solution in this study. Yellow phase dilemma zone is dynamically distributed at high-speed signalized intersections because of varying driving behaviors in response to yellow indications. This article presents an observation-based study of the natures of dynamic dilemma zones. A case study was conducted at a high-speed intersection in Fairfield, Ohio. Time-based yellow-onset trajectories were obtained using the video-capture-based technique and then they were used to analyze three types of dynamic dilemma zone models: that is, Type I and II dilemma zones, and option zone. The results reveal that the contributing factors of Type I dilemma zone and option zone are not constant but dynamic at different speeds. Vehicle type has been proved a factor that significantly affects drivers' stopping behavior. Drivers of trucks are more likely to make pass decisions than drivers of passenger vehicles. Cars, sport utility vehicles, vans, and light trucks have similar downstream boundaries of Type II dilemma zone whereas heavy trucks have the furthest upstream boundary of Type II dilemma zone. The article presents a study that seeks to model the effects of the yellow-phase dilemma, which is one of the factors that lead to intersection-signal-related incidents. It utilizes the vehicle trajectory data to come up with a strategy that measures the values of the causal subjects in varied speeds. Video-capturing methods are used to structure the dynamic zone model of the said study that was conducted in Fairfield, Ohio. Information on dilemma zone (DZ) and option zone (OZ) are offered.

A new kind of molecularly imprinted polymer (MIP) microspheres for the selective extraction of kaempferol was prepared by precipitation polymerization using 4-vinylpridine (4-VP) and ethylene glycol dimethacrylate (EDMA) as functional monomer and cross-linker respectively. The synthesis conditions, such as ratios of 4-VP/EDMA and polymerization time were discussed in detail. Results showed that the 2% was the optimal concentration of co-monomers to obtain monodisperse MIP microspheres, the best ratio of 4-VP/EDMA was 1:2, and 24 h was considered as the proper polymerization time. Compared with the MIP agglomeration or coagulum particles, monodisperse MIP microspheres showed the better adsorption capacity: the saturated adsorption capacity of monodisperse MIP microspheres was 7.47 mg g−1, the adsorption equilibrium could be obtained in 30 min. Finally, the adsorption performances of the optimal MIP microspheres were evaluated by kinetic adsorption, adsorption isotherm, and selective adsorption experiments, which indicated that the adsorption mechanism were chemical single layer adsorption and the separation factor was up to 3.91 by comparing with the structure similar compound (quercetin). The MIP microspheres exhibit prospects in the kaempferol efficient and selective separation.

The involvement of secreted phospholipase A2s in respiratory diseases, such as asthma and respiratory viral infections, is well-established. However, the specific role of secreted phospholipase A2 group IIE (PLA2G2E) during influenza virus infection remains unexplored. Here, we investigated the role of PLA2G2E during H1N1 influenza virus infection using a targeted mouse model lacking Pla2g2e gene (Pla2g2e-/-). Our findings demonstrated that Pla2g2e-/- mice had significantly lower survival rates and higher viral loads in lungs compared to wild-type mice following influenza virus infection. While Pla2g2e-/- mice displayed comparable innate and humoral immune responses to influenza virus challenge, the animals showed impaired influenzaspecific cellular immunity and reduced T cell-mediated cytotoxicity. This indicates that PLA2G2E is involved in regulating specific T cell responses during influenza virus infection. Furthermore, transgenic mice expressing the human PLA2G2E gene exhibited resistance to influenza virus infection along with enhanced influenzaspecific cellular immunity and T cell-mediated cytotoxicity. Pla2g2e deficiency resulted in perturbation of lipid mediators in the lung and T cells, potentially contributing to its impact on the antiinfluenza immune response. Objective To explore the efficacy of atorvastatin combined with compound probiotics in the treatment of hyperlipidemia. Methods From January 2019 to December 2019, 33 patients with hyperlipidemia were recruited from the People‘s Hospital of Inner Mongolia Autonomous Region and were randomly divided into the test group（n=18） and the control group（n=15） using a randomized, double-blind, controlled trial design. The 20mg atorvastatin （once a day） plus compound probiotics（once a day, 2g each time） was given in test group, and the same dose of atorvastatin plus placebo was given in control group. The blood lipid level was measured once every 4 weeks for 12 weeks to compare the blood lipid levels at each time point between two groups. Results There were no significant differences in the baseline data between two groups (P＞0.05). There were no significant differences in the levels of TC, TG, LDL-C and other blood lipid levels related to the treatment effects between two groups(P＞0.05). Since the first outbreak in 2013, the influenza A (H7N9) virus has continued emerging and has caused over five epidemic waves. Suspected antigenic changes of the H7N9 virus based on hemagglutination inhibition (HI) assay during the fifth outbreak have prompted the update of H7N9 candidate vaccine viruses (CVVs). In this study, we comprehensively compared the serological cross-reactivities induced by the hemagglutinins (HAs) of the earlier CVV A/Anhui/1/2013 (H7/AH13) and the updated A/Guangdong/17SF003/2016 (H7/GD16). We found that although H7/GD16 showed poor HI cross-reactivity to immune sera from mice and rhesus macaques vaccinated with either H7/AH13 or H7/GD16, the cross-reactive neutralizing antibodies between H7/AH13 and H7/GD16 were comparably high. Passive transfer of H7/AH13 immune sera also provided complete protection against the lethal challenge of H7N9/GD16 virus in mice. Analysis of amino acid mutations in the HAs between H7/AH13 and H7/GD16 revealed that L226Q substitution increases the HA binding avidity to sialic acid receptors on red blood cells, leading to decreased HI titers against viruses containing HA Q226 and thus resulting in a biased antigenic evaluation based on HI assay. These results suggest that amino acids located in the receptor-binding site could mislead the evaluation of antigenic variation by solely impacting the receptor-binding avidity to red blood cells without genuine contribution to antigenic drift. Our study highlighted that viral receptor-binding avidity and combination of multiple serological assays should be taken into consideration in evaluating and selecting a candidate vaccine virus of H7N9 and other subtypes of influenza viruses.
