Background. Vascular calcification is widespread and clinically significant, contributing to substantial morbidity and mortality. Calcifying vascular cells are partly derived from local vascular smooth muscle cells (VSMCs), which can undergo chondrogenic or osteogenic differentiation under inflammatory environment. Recently, we have found activation of CD137 signaling accelerated vascular calcification. However, the underlying mechanism remains unknown. This study aims to identify key mediators involved in CD137 signaling-induced vascular calcification in vivo and in vitro. Methods. Autophagy flux was measured through mRFP-GFP-LC3 adenovirus and transmission electron microscopy. Von Kossa assay and alkaline phosphatase (ALP) activity were used to observe calcification in vivo and in vitro, respectively. Autophagosome-containing vesicles were collected and identified by flow cytometry and Western blot. Autophagy or calcification-associated targets were measured by Western blot, quantitative real-time PCR, and immunohistochemistry. Results. Treatment with the agonist-CD137 displayed c-Jun N-terminal kinase- (JNK-) dependent increase in the expression of various markers of autophagy and the number of autophagosomes relative to the control group. Autophagy flux experiments suggested that agonist-CD137 blocked the fusion of autophagosomes with lysosomes in cultured VSMCs. Calcium deposition, ALP activity, and the expression of calcification-associated proteins also increased in agonist-CD137 group compared with anti-CD137 group, which could be recovered by autophagy stimulator rapamycin. Autophagosome-containing vesicles collected from agonist-CD137 VSMCs supernatant promoted VSMC calcification. Conclusion. The present study identified a new pathway in which CD137 promotes VSMC calcification through the activation of JNK signaling, subsequently leading to the disruption of autophagic flux, which is responsible for CD137-induced acceleration of vascular calcification.

Previous studies showed little CD137 expressed in normal vascular smooth muscle cells (VSMCs) and it is important to find a valid way to elevate it before studying its function. The level of CD137 was detected by RT-PCR, western blot, and flow cytometry, respectively. CD137 signaling activation was activated by agonist antibody and measured through phenotype transformation indicators and cell functions. Proteins in supernatants were detected by ELISA. The total CD137 elevates under different concentrations of CM treatment. Among these, 25 ng/ml CM treatment increases the CD137 expression mostly. However, flow cytometry demonstrates that 10 ng/ml CM elevates surface CD137 more significantly than other concentrations and reaches the peak at 36 h. At 10 ng/ml, but not 25 ng/ml CM pretreatment, the levels of phenotype related proteins such as SM-MHC, α-SMA, and calponin decrease while vimentin and NFATc1 increase, suggesting that VSMCs undergo phenotype transformation. Transwell, CCK-8 assay, and ELISA showed that the ability of VSMCs viability, migration, and IL-2 and IL-6 secretion induced by CD137 signaling was significantly enhanced by the pretreatment of 10 ng/ml CM. This research suggested that 10 ng/ml CM pretreatment is more reasonable than other concentrations when exploring CD137 function in VSMCs.

Optimizing product selectivity and conversion efficiency are primary goals in catalysis. However, efficiency and selectivity are often mutually antagonistic, so that high selectivity is accompanied by low efficiency and vice versa. Also, just increasing the temperature is very unlikely to change the reaction pathway. Here, by constructing hierarchical plasmonic nanoreactors, we show that nanoconfined thermal fields and energetic electrons, a combination of attributes that coexist almost uniquely in plasmonic nanostructures, can overcome the antagonism by regulating selectivity and promoting conversion rate concurrently. For propylene partial oxidation, they drive chemical reactions by not only regulating parallel reaction pathways to selectively produce acrolein but also reducing consecutive process to inhibit the overoxidation to CO2, resulting in valuable products different from thermal catalysis. This suggests a strategy to rationally use plasmonic nanostructures to optimize chemical processes, thereby achieving high yield with high selectivity at lower temperature under visible light illumination.

Plasmon-mediated chemical reactions (PMCRs) constitute a vibrant research field, advancing such goals as using sunlight to convert abundant precursors such as CO2 and water to useful fuels and chemicals. A key question in this burgeoning field which has not, as yet, been fully resolved, relates to the precise mechanism through which the energy absorbed through plasmonic excitation, ultimately drives such reactions. Among the multiple processes proposed, two have risen to the forefront: plasmon-increased temperature and generation of energetic charge carriers. However, it is still a great challenge to confidently separate these two effects and quantify their relative contribution to chemical reactions. Here, we describe a strategy based on the construction of a plasmonic electrode coupled with photoelectrochemistry, to quantitatively disentangle increased temperature from energetic charge carriers effects. A clear separation of the two effects facilitates the rational design of plasmonic nanostructures for efficient photochemical applications and solar energy utilization.

The plasmon-exciton interaction between gap-mode nanocavities and A excitons of MoS2 is investigated, and the longitudinal distribution of the plasmon-exciton interactions is obtained at ~1 nm spatial resolution.

Mid-infrared antennas (MIRAs) support highly-efficient optical resonance in the infrared, enabling multiple applications, such as surface-enhanced infrared absorption (SEIRA) spectroscopy and ultrasensitive mid-infrared detection. However, most MIRAs such as dipolar-antenna structures support only narrow-band dipolar-mode resonances while high-order modes are usually too weak to be observed, severely limiting other useful applications that broadband resonances make possible. In this study, we report a multiscale nanobridged rhombic antenna (NBRA) that supports two dominant resonances in the MIR, including a charge-transfer plasmon (CTP) band and a bridged dipolar plasmon (BDP) band which looks like a quadruple resonance. These assignments are evidenced by scattering-type scanning near-field optical microscopy (s-SNOM) imaging and electromagnetic simulations. The high-order mode only occurs with nanometer-sized bridge (nanobridge) linked to the one end of the rhombic arm which mainly acts as the inductance and the resistance by the circuit analysis. Moreover, the main hotspots associated with the two resonant bands are spatially superimposed, enabling boosting up the local field for both bands by multiscale coupling. With large field enhancements, multiband detection with high sensitivity to a monolayer of molecules is achieved when using SEIRA. Our work provides a new strategy possible to activate high-order modes for designing multiband MIRAs with both nanobridges and nanogaps for such MIR applications as multiband SEIRAs, IR detectors, and beam-shaping of quantum cascade lasers in the future.

This review focuses on the advance of optical design from nano/micro to macro in SERS and SEIRA, especially on the optical coupling between nano/micro and macro scales.
