ObjectiveTo study the change trend of blood pressure among rural adults in Shanxi province from 2002 to 2019, and to provide evidence for the prevention and control of hypertension in the population. MethodsThe study included 1 095 rural adult residents (≥ 18 years) with normal blood pressure at baseline in four counties of Shanxi province as part of the Provincial Nutrition and Chronic Diseases Household Cohort Study, which began in 2002. Data were collected from participants′ blood pressure measurements in 2015 and 2019, and changes in participants′ blood pressure between 2002 and 2019 were analyzed. ResultsAmong the participants, systolic and diastolic blood pressure (both in mm Hg) showed a significant increasing trend during the period (both P < 0.05). Compared to 122.9 ± 10.4 in 2002, systolic blood pressure increased by 14.8 to 137.7 ± 18.8 in 2015 and by 15.8 to 138.7 ± 18.1 in 2019, with an increase percentage of 12.0% and 12. 9%; while compared to 72.6 ± 6.9 in 2002, diastolic blood pressure increased by 7.3 to 79.9 ± 10.9 in 2015 and by 7.5 to 80.1 ± 10.9 in 2019, with increase percentages of 10.1% and 10.3%, respectively. ConclusionThere was a significant increasing trend in both systolic and diastolic blood pressure levels among rural adults in Shanxi province from 2002 to 2019.

Arteriosclerosis (AS) is a chronic inflammatory disease and Buyang Huanwu decoction (BHD) has been identified as an anti-atherosclerosis effect, and the study is aimed to investigate the underlying mechanism. The E4 allele of Apolipoprotein E (ApoE) is associated with both metabolic dysfunction and an enhanced pro-inflammatory response, ApoE-knockout (ApoE-/-) mice were fed with a high-fat diet to establish an arteriosclerosis model and treated with BHD or atorvastatin (as a positive control). The atherosclerotic plaque in each mouse was evaluated using Oil red O Staining. Elisa kits were used to evaluate blood lipid, interleukin-6 (IL-6), IL-1 beta (IL-1β), tumor necrosis factor alpha (TNF-α), IL-4, IL-10, and tumor growth factor beta (TGF-β) contents, while Western blot was applicated to measure inducible nitric oxide synthase (iNOS), arginase I (Arg-1) expression. Meanwhile, pyruvate kinase M2 (PKM2), hypoxia-inducible factor-1 alpha (HIF-1α) and its target genes glucose transporter type 1 (GLUT1), lactate dehydrogenase A (LDHA), and 3-phosphoinositide-dependent kinase 1 (PDK1), as well as IL-6, IL-1β, TNF-α, IL-4, IL-10, and TGF-β were evaluated by the quantitative reverse transcription-polymerase chain reaction. BHD treatment significantly reduced body weight and arteriosclerosis plaque area and blood lipid levels including total cholesterol (TC), triglyceride (TG), low-density lipoprotein cholesterol (LDL-C), and high-density lipoprotein cholesterol (HDL-C). Meanwhile, BHD demonstrated a significant suppression of M1 polarization, by decreased secretion of iNOS and pro-inflammatory factors (IL-6, IL-1β, and TNF-α) in ApoE-/- mice. The present study also revealed that BHD promotes the activation of M2 polarization, characterized by the expression of Arg-1 and anti-inflammatory factors (IL-4 and IL-10). In addition, PKM2/HIF-1α signaling was improved by M1/M2 macrophages polarization induced by BHD. The downstream target genes (GLUT1, LDHA, and PDK1) expression was significantly increased in high fat feeding ApoE-/- mice, and those of which were recused by BHD and Atorvastatin. These results suggested that M1/M2 macrophages polarization produce the inflammatory response against AS progress after BHD exposure.

Abstract Halorhodopsin, a light-driven chloride pump, utilizes photonic energy to drive chloride ions across biological membranes, regulating the ion balance and conveying biological information. In the light-driven chloride pump process, the chloride-binding chromophore (protonated Schiff base) is crucial, able to form the active center by absorbing light and triggering the transport cycle. Inspired by halorhodopsin, we demonstrate an artificial light-driven chloride pump using a helical porphyrin channel array with excellent photoactivity and specific chloride selectivity. The helical porphyrin channels are formed by a porphyrin-core star block copolymer, and the defects along the channels can be effectively repaired by doping a small number of porphyrins. The well-repaired porphyrin channel exhibits the light-driven Cl− migration against a 3-fold concentration gradient, showing the ion pumping behavior. The bio-inspired artificial light-driven chloride pump provides a prospect for designing bioinspired responsive ion channel systems and high-performance optogenetics.

Nanoporous membranes exhibit wide applications in the fields of energy, environmental, and analytical science. Among all nanoporous membrane materials, degradable block copolymers (DBCP) have attracted attention due to their well-defined nanopores, high porosity, narrow pore size distributions, and abundant functional end-groups. In this review, the fabrication strategies of DBCP-derived nanoporous membranes are described first, including degradation of sacrificial blocks, cleavage of block-linkers, and the removal of pore-forming agents. Secondly, the extensive applications of DBCP-derived nanoporous membranes are discussed. Generally, they have been used for separation membranes, nanotemplates, etched masks, catalysis, electronics, biomedicine, etc. Finally, the possible development directions and challenges of DBCP-derived nanoporous membranes in various applications are proposed.

Motile and non-motile cilia have distinct functions and protein complexes associated with them. Here, the authors show the conserved protein CFAP20 is important for both motile and non-motile cilia and is distinct from other ciliopathy-associated domains or macromolecular complexes.

BackgroundSleep disturbance is one of the most prominent complaints of patients with alcohol use disorder (AUD), with more than 70% of patients with AUD reporting an inability to resolve sleep problems during abstinence. Mindfulness-based stress reduction (MBSR) has been shown to improve sleep quality and as an alternative therapy to hypnotics for sleep disorders.ObjectiveThe aim of the present study was to evaluate the effect of short-term MBSR on sleep quality in male patients with AUD after withdrawal.MethodsA total of 91 male patients with AUD after 2 weeks of routine withdrawal therapy were randomly divided into two groups using a coin toss: the treatment group (n = 50) and the control group (n = 41). The control group was received supportive therapy, and the intervention group added with MBSR for 2 weeks on the basis of supportive therapy. Objective sleep quality was measured at baseline and 2 weeks after treatment using the cardiopulmonary coupling (CPC). Indicators related to sleep quality include total sleep time, stable sleep time, unstable sleep time, rapid eye movement (REM) sleep time, wake-up time, stable sleep latency, sleep efficiency, and apnea index. These indicators were compared by an analysis of covariance (ANCOVA) between the two groups, controlling for individual differences in the respective measures at baseline.ResultsThe results showed that there were no significant differences in the age [t (89) = –0.541, P = 0.590), BMI [t (89) = –0.925, P = 0.357], educational status [t (89) = 1.802, P = 0.076], years of drinking [t (89) = –0.472, P = 0.638), daily intake [t (89) = 0.892, P = 0.376], types of alcohol [χ2 (1) = 0.071, P = 0.789], scores of CIWA-AR [t (89) = 0.595, P = 0.554], scores of SDS [t (89) = –1.151, P = 0.253), or scores of SAS [t (89) = –1.209, P = 0.230] between the two groups. Moreover, compared with the control group, the total sleep time [F (1.88) = 4.788, P = 0.031) and stable sleep time [F (1.88) = 6.975, P = 0.010] were significantly increased in the treatment group. Furthermore, the average apnea index in the patients who received MBSR was significantly decreased than in the control group [F (1.88) = 5.284, P = 0.024].ConclusionThese results suggest that short-term MBSR could improve sleep quality and may serve as an alternative treatment to hypnotics for sleep disturbance in patients with AUD after withdrawal.

Oxygen-containing functional groups play crucial roles in graphene oxide due to their enhanced processability, stability, and functionalization. However, achieving precise control over the oxidation degrees of graphene oxide through a straightforward and effective method remains a significant challenge. Herein, we report a two-step electrochemical approach encompassing pre-intercalation and post-exfoliation/oxidation, enabling the mass exfoliation of graphene oxide with customizable oxidation levels. Initially, the pre-intercalation of concentrated sulfuric acid into graphite foil promotes uniform expansion, transforming it into a quasi-monolayer graphene structure. Subsequently, post-exfoliation in reductive/oxidative electrolytes triggers the simultaneous detachment and oxidation process, resulting in well-dispersed graphene nanosheets with quantified oxidation levels on a timescale of minutes. Comprehensive characterization studies confirm the varied oxidation levels of the exfoliated graphene oxide, spanning conventional oxidation degrees obtained via Staudenmaier's, Hofmann's, and Hummers' methods. Furthermore, we evaluate the scalability of this method and the solution processability of exfoliated graphene nanosheets, demonstrating the continuous production of graphene oxide at the kilogram scale and the fabrication of meter-length nanocomposite films with exceptional mechanical properties. ABSTRACT: Natural gas hydrate, oil and gas were all found together in the Qilian Mountain permafrost area, northeast of Qinghai-Tibet Plateau, China. They are closely associated with each other in space, but whether they are in any genetic relations are unknown yet. In this paper, a hydrocarbon gas-generation series, gas-fluid migration series and hydrocarbon gas-accumulation series are analyzed to probe the spatial, temporal and genetic relationships among natural natural gas hydrate, oil and gas. The subsequent results show that natural gas hydrate, oil and gas actually form a natural gas hydrate-oil-gas system. Based on the Middle Jurassic and the Upper Triassic hydrocarbon gas-generation series, it is divided into four major sub-systems in the study area: (1) A conventional Upper Triassic gas-bearing sub-system with peak hydrocarbon gas-generation in the late Middle Jurassic; (2) a conventional Middle Jurassic oil-bearing sub-system with low to mature hydrocarbon gas-generation in the late Middle Jurassic; (3) a natural gas hydrate sub-system with main gas source from the Upper Triassic gas-bearing sub-system and minor gas source from the Middle Jurassic oil-bearing sub-system as well as little gas source from the Middle Jurassic coal-bed gas and the microbial gas; (4) a shallower gas sub-system with microbial alteration of the main gas source from the Upper Triassic gas-bearing sub-system. This natural gas hydrate-oil-gas system and its sub-systems are not only theoretical but also practical, and thus they will play an important role in the further exploration of natural gas hydrate, oil and gas, even other energy resources in the study area.

PurposePost hemorrhagic shock mesenteric lymph (PHSML) return contributes to CD4+ T cell dysfunction, which leads to immune dysfunction and uncontrolled inflammatory response. Tumor necrosis factor α induced protein 8 like-2 (TIPE2) is one of the essential proteins to maintain the immune homeostasis. This study investigated the role of TIPE2 in regulation of CD4+ T lymphocyte function in interaction of PHSML and TLR2/TLR4.MethodsThe splenic CD4+ T cells were isolated from various mice (WT, TLR2-/-, TLR4-/-) by immunomagnetic beads, and stimulated with PHSML, normal lymphatic fluid (NML), respectively. Application of TIPE2-carrying interfering fragments of lentivirus were transfected to WT, TLR4-/-, and TLR2-/- CD4+ T cells, respectively. After interference of TIPE2, they were stimulated with PHSML and NML for the examinations of TIPE2, TLR2, and TLR4 mRNA expressions, proliferation, activation molecules on surface, and cytokine secretion function.ResultsPHSML stimulation significantly upregulated TIPE2, TLR2, and TLR4 mRNA expressions, decreased proliferation, CD25 expression, and IFN-γ secretion, and increased the secretion ability of IL-4 in WT CD4+ T cells. TIPE2 silencing enhanced proliferative capacity, upregulated CD25 expression, and increased IFNγ secretion in CD4+ T cells. PHSML stimulated TLR2-/-CD4+ T or TLR4-/-CD4+ T cells of which TIPE2 were silenced. TLR2 or TLR4 knockout attenuated PHSML-induced CD4+ T cells dysfunction; PHSML stimulation of silent TIPE2-expressing TLR2-/-CD4+ T or TLR4-/-CD4+ T revealed that the coexistence of low TIPE2 expression with lack of TLR2 or TLR4 eliminated this beneficial effect.ConclusionTIPE2 improves the PHSML-mediated CD4+T cells dysfunction by regulating TLR2/TLR4 pathway, providing a new intervention target following hemorrhagic shock-induced immune dysfunction.

ABSTRACT: Significant breakthroughs of shale gas exploration have been made in Lower Cambrian and Sinian shale in the north margin of the Yangtze Block, South China. The drill wells with industrial gas flow located in the southern margin of the Huangling dome. Base on the geological survey, 2D seismic, geochronological and drill wells data, the tectonic evolution history of Huangling dome was studied, and its control effect on the preservation condition of shale gas was discussed. The result shows that the Huangling dome might undergo four tectonic stages: (1) About 800 Ma, granite intrusion in the Huangling dome basement, primarily of granites replaced metamorphism rocks; (2) 800–200 Ma, no significant tectonic movement with slowly buried history; (3) From 200 Ma, multi-phase uplift and the sedimentary rocks was eroded in the core of the Huangling dome. Shale gas in the Cambrian and Sinian strata was well preserved in the margin of the Huangling dome as the following reasons: (1) The Sinian shale was buried about 7.8 km in-depth during Middle Jurassic, source rocks have a suitable thermal maturity for shale gas; (2) The rigid basement of the Huangling dome was mainly composed by homogeneity granite, without intensive deformation. As the main challenges of the widely distributed Lower Cambrian and Sinian shale are high-maturity and intensive deformation, a geological unit with a dome probably is a favorable zone for the old age shale gas. Therefore, it indicates that the adjacent zone of the Xuefengshan, Shennongjia and Hannan are the geological units with a dome and probably have potentials for the exploration of shale in the Lower Cambrian and Sinian.

ABSTRACT: In this paper, substantial domestic and foreign research results of microscopic shale reservoir space were systemically reviewed, the research history consisting of simple observation and qualitative classification, quantitative research, the combination of qualitative and quantitative research successively as well as the characteristics of each research stage were summarized. In addition, the current problems existing in the characterization methods of shale reservoir space were also analyzed. Furthermore, based on massive actual detection of typical core samples obtained from more than 50 global shale gas wells and relevant practical experience, a comprehensive characterization method of combining qualitative with the semi-quantitative characterization was put forward. In detail, the indicators of the qualitative characterization include pore combination type and organic-matter microscopic morphology type, while the core elements of the semi-quantitative characterization include the percentage of the organic-matter area and the plane porosity of the pores of different types. Based on the reference of the naming and classification of rocks, the three-end-member diagram method was used to characterize microscopic shale reservoir space. This is achieved by plotting the three end-member diagram of 3 kinds of first-order critical reservoir spaces, i.e., organic-matter pores, matrix pores, and micro-fractures, in order to intuitively present the features of the microscopic pore combination. Meanwhile, statistic histograms of organic-matter microscopic morphology type and the plane porosity of different types of pores were adopted to characterize the development degree of second-order pores quantitatively. By this comprehensive characterization method, the importance of both pore combination and the microscopic morphology of organic matter were emphasized, revealing the control of organic-matter microscopic morphology over the organic-matter pores. What is more, high-resolution FE-SEM was adopted to obtain semi-quantitative statistics results. In this way, the features of pore development and pore combination were quantified, not only reflecting the types and storage capacity of the microscopic shale reservoir space, but also presenting the hydrocarbon-generating potential of organic matter in shale. Therefore, the results of this research are capable of providing in-depth microscopic information for the assessment and exploration and development of shale gas resources.

Objective: The aim of this study was to clarify the role of autophagy in stellate ganglion block (SGB) reversing posthemorrhagic shock mesenteric lymph (PHSML)-mediated vascular hyporeactivity.Methods: Hemorrhagic shock model in conscious rats was employed to observe the effects of SGB (0.2 ml of 0.25% ropivacaine hydrochloride hydrate) and autophagy inhibitor 3-methyladenine (3-MA; 30 mg/kg) on the vascular reactivity of second-order rat mesenteric arteries in vitro, while the effects of PHSML (1 ml/kg) and autophagy agonist rapamycin (Rapa, 10 mg/kg) on the beneficial effect of SGB were investigated. The cellular viability, contractility, and autophagy-related protein expressions in vascular smooth muscle cells (VSMCs) were detected following treatments of PHSML, PHSML obtained from the rats that underwent hemorrhagic shock plus SGB (PHSML-SGB), and PHSML plus 3-MA (5 mM), respectively.Results: Hemorrhagic shock significantly decreased the vascular reactivity to gradient norepinephrine (NE), which is reversed by the SGB treatment and 3-MA administration. On the contrary, PHSML intravenous infusion and Rapa administration inhibited the vascular contractile responses in rats that underwent hemorrhagic shock plus SGB treatment. PHSML treatment significantly inhibited the cellular viability and contractility in VSMCs, increased the expressions of LC3-II and Beclin 1, and decreased the expression of p62, along with opposite appearances in these indices following PHSML-SGB treatment. In addition, 3-MA counteracted the adverse roles of PHSML in these indices in VSMCs.Conclusion: SGB inhibits PHSML-mediated vascular hyporeactivity by reducing the excessive autophagy in VSMCs.

Protracted alcohol withdrawal symptoms (PAWS), characterized by the presence of substance-specific signs and symptoms (including anxiety, irritability, mood instability, insomnia, and cravings), make alcohol abstinence difficult and increase the risk of relapse in recovering alcoholics. The goal of this study was to evaluate the effect of transcutaneous auricular vagus nerve stimulation (taVNS) on PAWS and plasma brain-derived neurotrophic factor (BDNF), interleukin-6 (IL-6), tumor necrosis factor-α (TNF-α), and leptin levels in patients with alcohol dependency. A total of 114 patients with alcohol dependence were randomly divided into two groups: the treatment group and the control group. The patients in the treatment group were treated with taVNS of the bilateral auricular concha using an ear vagus nerve stimulator. The Pennsylvania Alcohol Craving Scale was used to evaluate the extent of craving for alcohol. The Self-Rating Anxiety Scale and Self-Rating Depression Scale (SDS) were used to evaluate the extent of anxiety and depression symptoms, respectively. The Pittsburgh Sleep Quality Index (PSQI) was used to assess sleep quality. Enzyme-linked immunosorbent assay was used to measure plasma BDNF, IL-6, TNF-α, and leptin levels. The results showed that the SDS and PSQI scores were significantly lower in the treatment group than in the control group. Moreover, compared with the control group, the average BDNF levels in the treatment group were significantly increased. These results suggest that taVNS could improve the depression symptoms and sleep quality in alcohol-dependent patients after withdrawal, which might be related to the upregulation of plasma BDNF levels.

ABSTRACT: The Sinian Doushantuo Formation is the oldest shale gas reservoir discovered in the world, which contains good shale gas shows as the Eyangye-1 and Zidi-1 wells in the Yichang Region of western Hubei province, China. The shales in the Doushantuo Formation feature considerable thickness (135 m), high gas content (4.83 m3/t) and high fragile mineral content (up to 75%). Due to the influences of the sedimentary environment, the shale reservoirs here have high dolomitic content (54%) and abruptly changes in vertical lithofacies. Moreover, the characteristics of the shale sweet-spot differ significantly from that in the Wufeng Formation and Longmaxi Formation in southeast Sichuan Basin. A high-resolution sequence stratigraphic correlation was performed on the shales of the Doushantuo Formation at the Eyangye-1 well, Zidi-1 well, Zidi-2 well and some outcrop profiles in Yichang area for the identification of their sedimentary microfacies characteristics. A comprehensive comparative analysis was made by incorporating the elemental geochemistry, wireline and mud logging data of the well to further identify the sweet-spot interval therein. With the analysis of Eyangye-1 well, the sweet-spot interval of the Doushantuo Formation is 3360−3408 m, of which sedimentary microfacies is deepwater reduced environment with many pyrite laminaes, and the TOC is high to 3.42%, the Ro is 3.3%, the organic and matrix pore are well developed and the aperture more than 50 nm, the porosity is 2.7%, the desorption gas content average is 2.16%, and the main mineral is dolomitic (54%) and siliceous quartz (21%). Additionally, these parameters combined with previous studies will shed light on evaluating and characterizing the layers therein, also provide referential geological data for the following exploration and development activities of this shale system.

Abstract Background Sympathetic overactivation and inflammation are two major mediators to post-myocardial ischemia-reperfusion (I/R)-induced ventricular arrhythmia (VA). The vicious cycle between microglia and sympathetic activation plays an important role in sympathetic hyperactivity related to cardiovascular diseases. Recently, studies have shown that microglial activation might be attenuated by light-emitting diode (LED) therapy. Therefore, we hypothesized that LED therapy might protect against myocardial I/R-induced VAs by attenuating microglial and sympathetic activation. Methods Thirty-six male anesthetized rats were randomized into four groups: control group (n = 6), LED group (n = 6), I/R group (n = 12), and LED+I/R group (n = 12). I/R was generated by left anterior descending artery occlusion for 30 min followed by 3 h reperfusion. ECG and left stellate ganglion (LSG) neural activity were recorded continuously. After 3 h reperfusion, a programmed stimulation protocol was conducted to test the inducibility of VA. Furthermore, we extracted the brain tissue to examine the microglial activation, and the peri-ischemic myocardium to examine the expression of NGF and inflammatory cytokines (IL-1β, IL-18, IL-6, and TNF-α). ABSTRACT: At present, most shale gas exploration and development areas in China are difficult to provide sufficient and effective production data to support economic evaluation, since they are still in the initial stage of low exploration level. In addition, ecological and environmental factors are not taken into account in the evaluation process, which does not meet the needs of green energy development of China. Aiming at above problems, the dynamic economic evaluation method of shale gas resources based on calculus principle is proposed. The Arps hyperbolic decreasing curve model will be used in the evaluation of single shale gas well production, which can evaluate single well production of shale gas by fitting the existing dynamic production data to generate the production decreasing curve. Therefore, the variation regularity of the cumulative production of single well shale gas within the study area can be obtained by the model mentioned above. According to the variation regularity of the cumulative production obtained from the Arps hyperbolic decreasing curve model, the recovery period of single well cost, ultimate economic life and the ultimate economic resource can be evaluated dynamically by analyzing the variation regularity of the cumulative sales revenue and cumulative input cost of single shale gas well. Then the evaluation result can be further extend to the whole evaluation areas, in order to analyze shale gas resources ’ economic value in evaluation regions under different shale gas price conditions. The results of the above evaluation methods are not only conducive to improving the economic benefits of relative shale gas development enterprises, but also provide a basis for the national energy strategy deployment.© 2019 China Geology Editorial Office.

Summary: Background: Myasthenia gravis (MG) is the most common primary disorder of neuromuscular transmission, but the incidence of MG in China is unknown. We conducted the first nationwide study to determine the incidence and mortality rates of MG in all age groups at the national level in China. Methods: This national population-based registry study is based on the database of the Hospital Quality Monitoring System of National Health Commission, which covers 1665 hospitals providing myasthenia gravis care in 31 provinces and municipalities across China. 94,638 hospital admissions for 59,243 myasthenia gravis patients were identified from January 1st, 2016 to December 31st, 2018. Myasthenia gravis was identified by ICD-10 codes (G70). Incidence of myasthenia gravis was stratified by age, sex, and province. Findings: Of 59,243 patients, 30,503 individuals with myasthenia gravis were newly diagnosed. Age and sex adjusted incidence of myasthenia gravis was 0.68 per 100,000 person-years, with highest in the age group of 70–74 years. The incidence in females was 0.76 per 100,000 and 0.60 per 100,000 in males. The admission mortality rate was 14.69‰. Respiratory failure was the leading cause of death in patients with myasthenic crisis. There were 14,840 patients with thymomas, encompassing 14,636 (26.5%) adults and 204 (7.1%) juveniles. 9453 (63.7%) patients with thymomas underwent thymomectomy. The median length of hospital stay was 8 days (interquartile range (IQR) 4 to 15 days) with median hospitalization cost $1037 (IQR $493 to $2925). The Basic Medical Insurance was the most common payment method, covering 67.4% of patients. Interpretation: The age and sex adjusted incidence of MG was 0.68 per 100,000 person-years in China. The admission mortality rate was 14.69‰. Most cases of new onset MG occurred in the seventh decade of life. BackgroundAlisma orientalis beverage (AOB) is a Chinese traditional medicine formulated with a diversity of medicinal plants and used for treating metabolic syndrome and atherosclerosis (AS) since time ago. Given the current limited biological research on AOB, the mechanism by which AOB treats AS is unknown. This study investigats the role of AOB-induced gut microbiota regulation in the expansion of AS.MethodsWe established an AS model in male apolipoprotein E-deficient (ApoE−/−) mice that are fed with a high-fat diet (HFD), treated with numerous interventions, and evaluated the inflammatory cytokines and serum biochemical indices. The root of the aorta was stained with oil red O, and the proportion of the lesion area was quantified. Trimethylamine N-oxide (TMAO) and trimethylamine (TMA) levels in serum were evaluated through liquid chromatography with mass spectrometry. Flavin−containing monooxygenase 3 (FMO3) liver protein expression was assessed by Western blotting. 16S rDNA sequencing technique was adopted to establish the changes in the microbiota structure.ResultsAfter 8 weeks of HFD feeding, an inflammatory cytokine, and AS development expression were significantly decreased in mice treated with AOB; the same parameters in the mice treated with the antibiotics cocktail did not change. In the gut microbiota study, mice treated with AOB had a markedly different gut microbiota than the HFD-fed mice. Additionally, AOB also decreased serum TMAO and hepatic FMO3 expression.ConclusionThe antiatherosclerotic effects of AOB were found associated with changes in the content of gut microbiota and a reduction in TMAO, a gut microbiota metabolite, suggesting that AOB has potential therapeutic value in the treatment of AS.

ABSTRACT: After the breakthrough of shale gas exploration and development in the Ordovician Wufeng Formation (Fm.) and Silurian Longmaxi Fm. of Chongqing Jiaoshiba area, Changning-Weiyuan area, etc. in Sichuan basin, a series of discovery and breakthrough were obtained by China Geological Survey in the Cambrian Niutitang Fm. and Sinian Doushantuo Fm. shale of the areas with complicated structure outside Sichuan basin. Based on the understanding of the law of shale gas enrichment in Longmaxi Fm. in the basin, this paper puts forward three elements of the formation and enrichment of shale gas, which are the “Source”, the “Diagenesis” and the “Accumulation”, after deeply studying the law shale gas enrichment and accumulation in Sinian-Cambrian reservoir of the complex structure area outside the basin. The “Source” means the sedimentary environment and petrological characteristics of organic shale. The “Diagenesis” means the basin tectonic subsidence and hydrocarbon generation and expulsion process of organic matter. The “Accumulation” means the tectonic uplift and shale gas preservation. It is proposed that the Sinian-Cambrian and Ordovician-Silurian black shale series in the middle and upper Yangtze region of southern China were both formed in the deep-water shelf environment of rift trough and foreland basin respectively. The dessert intervals were formed in the strong reduction environment under transgressive system tract. The shale lithology belongs to calcium-siliceous and charcoal-siliceous respectively. Based on the summary of structural evolution in Yangtze area, the correlation of structural burial depth with shale diagenesis and the coupling evolution of organic matter with pore structure are discussed. Combining with structural styles, the preservation conditions of shale gas are discussed. Five types of shale gas reservoir control models are further described. Two types of future exploration directions, which are reverse fault syncline and paleo-uplift margin in complex structural area outside the basin, are proposed.

ABSTRACT: Focusing on the Yangtze River economic zone, the previous geological researches are systematically summarized, resources and environment conditions and major geological problems which are needing to be concerned in land planning and construction are studied. The results show that the resource conditions of cultivated land, shale gas, geotherm, lithium and so on are superior in the Yangtze River economic zone, and the resources and environment conditions are conducive to develop the modern agriculture, clean energy industry and strategic emerging industries. 3×1013 m2 farmlands without heavy metal pollution are concentrated; there are three national level shale gas exploration and development bases with explored reserves of 5.441×1011 m3; geothermal availability is 2.4×109 t of standard coal each year, equivalent to 19% of the amount of coal in 2014; Asia’s largest energy lithium metal ore deposit is found. In some parts of Yangtze River economic zone, there are some major geological problems such as active faults, karst collapse, ground subsidence, landslide-collapse-debris flow, affecting the river-crossing channels, high-speed railway, urban agglomeration and green ecological corridor planning and construction. Those problems should be concerned, and the relevant suggestions and countermeasures are put forward. Meanwhile, the ideas to further support the development of the Yangtze River economic zone are put forward.

ABSTRACT: There are plenty of Sinian and Cambrian potential shale gas resources in South China, which is characterized by high thermal evolution degrees, poor drilling performances and only occurs in local areas. Taking the principle “high to find low” is the key issue to achieving a breakthrough in older shale. China Geological Survey drilled in the periphery of the Proterozoic basement, i.e. the Huangling anticline, in the western Hubei, and Hannan paleocontinent in the southern Shanxi. It received high-quality gas-bearing shale with relatively low Ro in the in Lower Cambrian Niutitang formation and Sinian Doushantuo formation. Based on geological conditions of shale gas reservoirs in the Huangling anticline, this paper puts forward the new model named “Control over reservoirs by periphery of basement” about shale gas accumulation, suggesting that the shale deposited in a deepwater continental shelf in the periphery of the basement is characterized by shallow burial, a short burial time, stable tectonics, relatively low thermal evolution degrees, and shale gas reservoirs in a good condition. The shale of the Sinian-Cambrian strata deposited in deepwater continental shelves in the periphery of Chuanzhong paleo-uplift in Sichuan, Hannan paleocontinent in the southern Shanxi, Huangling anticline in western Hubei and Jiangnang-Xuefeng paleo-uplift in Hunan and Guizhou province have good shale gas exploration potential.

ABSTRACT: There are three types of shale gas resources in China. The resources are present in large amounts and are widely distributed. Marine facies, transitional facies and continental facies resources each account for a third. Based on resource distributions, there are many wells penetrated into the Sinian, Cambrian, Ordovician, Silurian, Devonian, Carboniferous and Permian strata of the Yangtze plate and its periphery, the North China Craton and the Tarim Basin. Many years of exploration have indicated that the marine Silurian Longmaxi shale gas is widely distributed in south China and has been industrialized in its production in the Sichuan basin. The shale gas from the Cambrian Niutitang Formation and the Sinian Doushantuo Formation are important discoveries in Yichang, Hubei and Zhenba, Shanxi. There are also shale gas resources found within transitional facies and continental facies in different areas in China. The “two element enrichment theory” has been summarized during the exploration process of Silurian marine shale gas in the Sichuan Basin. In addition, horizontal drilling and fracturing technologies up to 3500 m in depth have been developed. Based on the understanding of shale gas accumulation in a complex tectonic zone outside the Sichuan basin, a preliminary summary of the formation of the “converse fault syncline control reservoir” and “paleo uplift control reservoir” model has been constructed. The dominant theory of “Trinity” shale gas enrichment and the high yield of the “deep water Lu Pengxiang sedimentary facies belt, structural preservation conditions and overpressure” is summarized. Guided by the above theories. Anye1 well in Guizhou and Eyangye1 well in Hubei were drilled. “Four storey” oil and shale gas is found in the Permian Qixia group, the Silurian Shiniulan Formation, the Longmaxi Formation and the Ordovician Baota Formation in Anye1 well. Good shale gas has been gound in the Cambrian Niutitang formation inian Doushantuo formation in Eyangye 1well. This paper aims to summarize and review the main progress, theoretical technology and problems of shale gas exploration and development in recent years in China, and predicts the future exploration and development direction for shale gas and possible exploration areas.

Background: The incidence of noncommunicable diseases (NCDs) is rising dramatically throughout the world. Aspects of researches concerned with the improvement and development of prevention and control of NCDs have been conducted. Furthermore, the influence of most determinants of the major NCDs has showed that a broad and deep response involving stakeholders in different sectors is required in the prevention and control of NCDs. Objective: China has experienced an increase in NCDs in a short period compared with many countries. To address the burden of NCDs in China, it is important to learn about the progress that has been made in prevention and control of NCDs in China and worldwide, informed by opinions of stakeholders in different areas. Methods: In 2014, GRAND South developed the NCD Scorecard instrument to evaluate progress of NCD prevention and control in 23 countries through a 2-round Delphi process. The scorecard included 51 indicators in 4 domains: governance, surveillance and research, prevention and risk factors, and health system response. Stakeholders were then selected in the areas of government, nongovernmental organizations, private sectors, and academia to join the NCD Scorecard survey. Indicators of progress were scored by stakeholders from 0 (no activity), 1 (present but not adequate), and 2 (adequate) to 3 (highly adequate) and then the percentage of progress in each domain was calculated, representing the current situation in each country. Findings: There were 14 indicators in the domains of governance and surveillance and research. Of 429 stakeholders worldwide, 41 in China participated in the survey. China scored in the top 5 out of all participating countries in those 2 domains, scoring 67% in governance and 64% in surveillance and research. Indicators on which China scored particularly well included having a well-resourced unit or department responsible for NCDs, having a strong national system for recording the cause of all deaths, and having a system of NCD surveillance. Areas where China had the greatest need for improvement included increasing taxes on tobacco and addressing the needs of the population older than age 70 dying from major NCDs. ConclusionIn China the burden of disease of NCDs and disabilities remains serious, although China has put significant efforts into its governance and surveillance and research. To improve, further action is needed on reducing tobacco consumption, increasing investment in the national health budget, and increasing the focus on system construction.

Abstract Congenital cataract is the most frequent inherited ocular disorder and the most leading cause of lifelong visual loss. The screening of pathogenic mutations can be very challenging in some cases, for congenital cataracts are clinically and genetically heterogeneous diseases. The aim of this study is to investigate the mutation spectrum and frequency of 54 cartaract-associated genes in 27 Chinese families with congenital cataracts. Variants in 54 cataract-associated genes were screened by targeted next-generation sequencing (NGS) and then validated by Sanger sequencing. We identified pathogenic variants in 62.96% (17/27) of families, and over 52.94% (9/17) of these variants were novel. Among them, three are splicing site mutations, four are nonsense mutations, seven are missense mutations, two are frame shift mutations and one is intronic mutation. This included identification of: complex ocular phenotypes due to two novel PAX6 mutations; progressive cortical cataract and lamellar cataract with lens subluxation due to two novel CRYGS mutations. Mutations were also found in rarely reported genes including CRYBA4, CRYBA2, BFSP1, VIM, HSF4, and EZR. Our study expands the mutation spectrum and frequency of genes responsible for congenital cataracts. Targeted next-generation sequencing in inherited congenital cataract patients provided significant diagnostic information.

Abstract Purpose: To investigate the effect of mesenteric lymph drainage on the spleen injury and the expressions of inflammatory cytokines in splenic tissue in mice following hemorrhagic shock. Methods: Male C57 mice were randomly divided into the sham shock, shock and shock+drainage groups. The mice in both shock and shock+drainage groups suffered femoral artery bleeding, maintained mean arterial pressure (MAP) of 40±2 mmHg for 90 min, and were resuscitated. And mesenteric lymph drainage was performed in the shock+drainage group at the time of resuscitation. After three hours of resuscitation, the splenic tissues were harvested for the histological observation and protein and mRNA expression analysis of cytokines. Results: The spleen in the shock group revealed a significantly structural damage and increased mRNA expressions of MyD88 and TRAF6 and protein expressions of TIPE2, MyD88, TRIF and TRAF3 compared to the sham group. By contrast, the splenic pathological injury in the shock+drainage group was alleviated significantly, and the mRNA and protein expressions of TIPE2, MyD88, TRIF, TRAF3 and TRAF6 were significantly lower than those in the shock group. Conclusion: These results indicate that post-hemorrhagic shock mesenteric lymph drainage alleviates hemorrhagic shock-induced spleen injury and the expressions of inflammatory cytokines.

AIM:To characterize the disease-causing mutations in a Chinese family with ectopia lentis syndrome (ELS).METHODS:Patients and their family members were given complete physical, ophthalmic, and cardiovascular examinations. Genomic DNA samples were extracted from the peripheral blood of the pedigree members and 100 healthy controls. Mutation screening was performed in the fibrillin-1 (FBN1) gene by bi-directional sequencing of the amplified products. The mutation was analyzed using two bioinformatics methods.RESULTS:A novel heterozygous c.305G>A mutation in exon 3 of FBN1 was detected. As a result of this change, a highly conserved cysteine residue was replaced by a tyrosine residue (p.C102Y). Another mutation was found in the same exon (c.303T>C), which did not change the amino acid sequence. Both mutations were discovered in each affected individual, but not in the unaffected family members, or in 100 ethnically matched controls. A bioinformatics analysis predicted that mutation p.C102Y would affect protein function.CONCLUSION:In the first epidermal growth factor-like module, we identified a novel FBN1 mutation (p.C102Y), which caused ELS in the family. Our study presented a unique phenotype, including some distinct ophthalmic findings, such as hypoplasia of the iris and anisometropia. Our results expanded the mutation spectrum of FBN1 and enriched the overall knowledge of genotype-phenotype correlations due to FBN1 mutations.

Oxidative damage is a major cause of lung diseases, including pulmonary fibrosis. Laminarin is a kind of polysaccharide extracted from brown algae and plays vital roles in various biological processes. However, the functions and mechanisms of laminarin in pulmonary oxidative damage are poorly understood. This study aimed at investigating the protective effect of laminarin against pulmonary oxidative damage and underlying mechanisms. Human lung fibroblasts MRC-5 cells were treated with hydrogen peroxide to induce oxidative damage. Laminarin treatment was performed before or after hydrogen peroxide treatment, and then major indexes of oxidative damage, including superoxide dismutase (SOD), malondialdehyde (MDA), reduced glutathione (GSH) and catalase (CAT), were quantified by biochemical assays. The expression of oxidation-related factor, nuclear factor erythroid 2 like 2 (NRF2) was analyzed by qPCR, Western blot and immunofluorescence assay. NRF2 knockdown and overexpression were performed by cell transfection to reveal possible mechanisms. Results showed that laminarin treatment of 0.020 mg/mL for 24 h, especially the pre-treatment, could significantly relieve changes in SOD, MDA, GSH and CAT that were altered by hydrogen peroxide, and promote NRF2 mRNA (P

Although dietary patterns are crucial to cognitive function, associations of dietary patterns with cognitive function have not yet been fully understood. This cross-sectional study explored dietary patterns associated with cognitive function among the older adults in underdeveloped regions, using 1504 community-dwelling older adults aged 60 and over. Diet was assessed using a food frequency questionnaire and 24-h dietary recall. Factor analysis was used to extract dietary patterns. Global cognitive function was assessed using the Mini-Mental State Examination (MMSE). Two dietary patterns, a “mushroom, vegetable, and fruits” (MVF) pattern and a “meat and soybean products” (MS) pattern, were identified. The MVF pattern, characterized by high consumption of mushrooms, vegetables, and fruits was significantly positively associated with cognitive function (p < 0.05), with an odds ratio of (95% CIs) 0.60 (0.38, 0.94) for cognitive impairment and β (95% CIs) 0.15 (0.02, 0.29) for –log (31-MMSE score). The MS pattern, characterized by high consumption of soybean products and meat, was also associated with better cognitive function, with an odds ratio of 0.47 (95% CIs 0.30, 0.74) for cognitive impairment and β (95% CIs) 0.34 (0.21, 0.47) for –log (31-MMSE score). Our results suggested that both the MVF and MS patterns were positively associated with better cognitive function among older adults in underdeveloped regions.

Quantitative real time RT-PCR has been described as the most sensitive method for the detection of low abundance mRNA. To date, no reference genes have been screened in the half-smooth tongue sole (Cynoglossus semilaevis). The aim of this study was to select the most stable genes for quantitative real-time RT-PCR. Eight housekeeping genes (18S, TUBA, B2M, ACTB, EF1A, GAPDH, RPL17 and UBCE) were tested at different developmental stages, in different tissues, and following exposure to the drug SB-431542. Using geNorm, BestKeeper and NormFinder software, GAPDH/B2M, GAPDH/18S and UBCE/GAPDH were identified as the most suitable genes from samples taken of different developmental stages while 18S/RPL17 were consistently ranked as the best reference genes for different tissue types. Furthermore, TUBA/B2M, TUBA/UBCE and B2M/TUBA were found to be the most suitable genes in samples treated with the drug, SB-431542 by geNorm, BestKeeper and NormFinder respectively. Across both different developmental stages and tissue types, the combination of 18S and GAPDH was the most stable reference gene analyzed by Ref-Finder. To test and verify the screened reference genes, the expression profiles of LEFTY-normalized to the combination of GAPDH/18S and ACTB were presented. These results will be useful for future gene-expression studies in the half-smooth tongue sole.

A novel pyrethroid-degrading esterase gene pytY was isolated from the genomic library of Ochrobactrum anthropi YZ-1. It possesses an open reading frame (ORF) of 897 bp. Blast search showed that its deduced amino acid sequence shares moderate identities (30% to 46%) with most homologous esterases. Phylogenetic analysis revealed that PytY is a member of the esterase VI family. pytY showed very low sequence similarity compared with reported pyrethroid-degrading genes. PytY was expressed, purified, and characterized. Enzyme assay revealed that PytY is a broad-spectrum degrading enzyme that can degrade various pyrethroids. It is a new pyrethroid-degrading gene and enriches genetic resource. Kinetic constants of Km and Vmax were 2.34 mmol·L(-1) and 56.33 nmol min(-1), respectively, with lambda-cyhalothrin as substrate. PytY displayed good degrading ability and stability over a broad range of temperature and pH. The optimal temperature and pH were of 35°C and 7.5. No cofactors were required for enzyme activity. The results highlighted the potential use of PytY in the elimination of pyrethroid residuals from contaminated environments.

Phytophthora are eukaryotic pathogens that cause enormous losses in agriculture and forestry. Each Phytophthora species encodes hundreds of effector proteins that collectively have essential roles in manipulating host cellular processes and facilitating disease development. Here we report the crystal structure of the effector Phytophthora suppressor of RNA silencing 2 (PSR2). PSR2 produced by the soybean pathogen Phytophthora sojae (PsPSR2) consists of seven tandem repeat units, including one W-Y motif and six L-W-Y motifs. Each L-W-Y motif forms a highly conserved fold consisting of five α-helices. Adjacent units are connected through stable, directional linkages between an internal loop at the C terminus of one unit and a hydrophobic pocket at the N terminus of the following unit. This unique concatenation results in an overall stick-like structure of PsPSR2. Genome-wide analyses reveal 293 effectors from five Phytophthora species that have the PsPSR2-like arrangement, that is, containing a W-Y motif as the "start" unit, various numbers of L-W-Ymotifs as the "middle" units, and a degenerate L-W-Y as the "end" unit. Residues involved in the interunit interactions show significant conservation, suggesting that these effectors also use the conserved concatenation mechanism. Furthermore, functional analysis demonstrates differential contributions of individual units to the virulence activity of PsPSR2. These findings suggest that the L-W-Y fold is a basic structural and functional module that may serve as a "building block" to accelerate effector evolution in Phytophthora. [ABSTRACT FROM AUTHOR]

Estrogens have great harmfulness to living organisms. Adsorption is a key step to estrogen removal in biological process. As a typical estrogen with high estrogen activity, 17 beta-estradiol (E2) adsorption behavior by anaerobic granular sludge (AnGS) combined with zero-valent iron (ZVI) was researched here. The effect of pH on E2 adsorption by ZVI/AnGS was investigated and analyzed through adsorption equilibrium, adsorption kinetics, adsorption thermodynamics, and zeta potential. ZVI/AnGS had high E2 adsorption capacity. The optimized condition was pH 6 with 92.78% E2 adsorption efficiency, 4.95 μg·gVSS–1 equilibrium adsorption capacity, and 0.07 g·μg–1·min–1 adsorption rate. E2 was adsorbed as cationic forms or neutral inner-sphere surface complexes at a pH range of 2–10. The adsorption rate was controlled by film diffusion or intraparticle diffusion. Freundlich and Langmuir isotherms were both suitable for describing E2 adsorption process, which was exothermic and spontaneous. [ABSTRACT FROM AUTHOR]

[Objectives] To obtain a resveratrol synthase gene of Vitis vinifera and make bioinformatics analysis. [Methods] Taking total RNA of V. visifera as the template, by RT-PCR method, a complete cDNA sequence of resveratrol synthase gene was amplified from V. vinifera, and the resveratrol synthase gene was named as RS. The nucleic acid and protein sequences were analyzed using bioinformatics software. [Results] This sequence was 1179 bp in length, the similarity with reported resveratrol synthase gene reached 94% - 99%, and the similarity with amino acid sequence reached 96% - 99%; the RS gene encoded 392 amino acids, and amino acid sequence contained complete characteristic sequence GVLFGPGLT and active center sequence GCYAGGTVLR of stilbene synthase family; the predicted molecular weight was 42.78 kDa, the theoretical isoelectric point was 6.57, the instability parameter was 35.92, and it belonged to stable protein in the classification; the secondary structure was mainly α-helix, random coil and (folding, α-helix content was 44.13%, the random coil content was 26.53%, and β-folding content was 17.66%. [Conclusions] The isolated RS gene is a resveratrol synthase gene from V. vinifera. Clustering 16S rRNA sequences into operational taxonomic units (OTUs) is a crucial step in analyzing metagenomic data. Although many methods have been developed, how to obtain an appropriate balance between clustering accuracy and computational efficiency is still a major challenge. A novel density-based modularity clustering method, called DMclust, is proposed in this paper to bin 16S rRNA sequences into OTUs with high clustering accuracy. The DMclust algorithm consists of four main phases. It first searches for the sequence dense group defined as n-sequence community, in which the distance between any two sequences is less than a threshold. Then these dense groups are used to construct a weighted network, where dense groups are viewed as nodes, each pair of dense groups is connected by an edge, and the distance of pairwise groups represents the weight of the edge. Then, a modularity-based community detection method is employed to generate the preclusters. Finally, the remaining sequences are assigned to their nearest preclusters to form OTUs. Compared with existing widely used methods, the experimental results on several metagenomic datasets show that DMclust has higher accurate clustering performance with acceptable memory usage. The effects of adsorbate concentration, adsorbent concentration, temperature, pH, and ion strength on the 17beta-estradiol (E2) adsorption process by anaerobic granular sludge (AnGS) were investigated in this study. The adsorption process was analyzed according to adsorption isotherm, adsorption thermodynamics and adsorption kinetics. It was found that E2 was quickly and effectively adsorbed by AnGS. The adsorption capacity and adsorption removal efficiency of E2 increased as E2 concentration increased, and decreased as AnGS concentration increased. Low temperature, low pH, and low ionic strength promoted E2 adsorption by AnGS. The Freundlich absorption isotherm accurately described the E2 adsorption equilibrium process by AnGS and the pseudo-second-order model accurately described the adsorption kinetics. The overall adsorption process of E2 by AnGS was a physical adsorption process of spontaneous heat release controlled both by film diffusion and intraparticle diffusion. [ABSTRACT FROM AUTHOR]

China faces the greatest challenge from stroke in the world. The death rate for cerebrovascular diseases in China was 149.49 per 100 000, accounting for 1.57 million deaths in 2018. It ranked third among the leading causes of death behind malignant tumours and heart disease. The age-standardised prevalence and incidence of stroke in 2013 were 1114.8 per 100 000 population and 246.8 per 100 000 person-years, respectively. According to the Global Burden of Disease Study 2017, the years of life lost (YLLs) per 100 000 population for stroke increased by 14.6%; YLLs due to stroke rose from third highest among all causes in 1990 to the highest in 2017. The absolute numbers and rates per 100 000 population for all-age disability-adjusted life years (DALYs) for stroke increased substantially between 1990 and 2017, and stroke was the leading cause of all-age DALYs in 2017. The main contributors to cerebrovascular diseases include behavioural risk factors (smoking and alcohol use) and pre-existing conditions (hypertension, diabetes mellitus, dyslipidaemia and atrial fibrillation (AF)). The most prevalent risk factors among stroke survivors were hypertension (63.0%-84.2%) and smoking (31.7%-47.6%). The least prevalent was AF (2.7%-7.4%). The prevalences for major risk factors for stroke are high and most have increased over time. Based on the latest national epidemiological data, 26.6% of adults aged ≥15 years (307.6 million adults) smoked tobacco products. For those aged ≥18 years, age-adjusted prevalence of hypertension was 25.2%; adjusted prevalence of hypercholesterolaemia was 5.8%; and the standardised prevalence of diabetes was 10.9%. For those aged ≥40 years, the standardised prevalence of AF was 2.31%. Data from the Hospital Quality Monitoring System showed that 3 010 204 inpatients with stroke were admitted to 1853 tertiary care hospitals during 2018. Of those, 2 466 785 (81.9%) were ischaemic strokes (ISs); 447 609 (14.9%) were intracerebral haemorrhages (ICHs); and 95 810 (3.2%) were subarachnoid haemorrhages (SAHs).
