The use of indigenous endophytic bacteria (IEB) isolates as PGPR and resistance induction has been conducted to wilt in tomatoes (Lycopersicum esculentum Mill). This study aimed to obtain the best endophytic bacteria isolates in increasing the resistance of tomato plants to B. tabaci. The research used a completely randomized design (CRD) with ten treatments and three replications. The treatment consisted of eight isolates of IEB, positive controls (without using insecticides), and negative controls (using insecticides). The study was conducted at the Microbiology and Greenhouse Laboratory, Faculty of Agriculture, Universitas Andalas from May to July 2018. The parameters observed were the number of individuals who survived per day and the length of days needed to develop per stage. The results showed that EPL 1.1.4 and KLE 3.3 isolates inhibited egg laying, whereas EI.AB 2.1, EI.AB 1.2, KLE 3.3, and SNE 2.2 inhibited the development of B. tabaci. There was no difference in the length of day between IEB isolates and controls. EI.AB 2.1 showed the most significant adverse effect on the success of B. tabaci life with a value of lx = 0.0 achieved on nymph of instar 3. The experiment was carried out to examine the organic acids production of rice straw fermented with some types of microorganisms at different temperatures. The experiment was designed as Split Plot-Completely Randomized Design. The main plot was temperatures treatments (25, 35, 45°C) and the sub plot were microorganisms (Control, Control+Mollases, Lactobacillus fermentum, Bacillus subtilis, Bacillus coagulant, Saccharomyces cerevisiae, Aspergillus niger). The highest lactic acid productions was in B. coagulans treatment at 35°C (53.79 g/kg DM). The highest acetic acid productions was in L. fermentum at 35°C (13.20 g/kg DM), while the highest propionic acid productions were in Control treatment at 35°C (0.37 g/kg DM). [ABSTRACT FROM AUTHOR]

Purpose: Interleukin-18, transforming growth factor-β, and superoxide dismutase are important cytokines and antioxidants in protecting the body from damage caused by radiation exposure through an immune response mechanism. Genetic polymorphisms -607 C/A and -137 G/C are thought to affect the IL-18 cytokine in carrying out its function as a biomarker to indicate adverse conditions due to radiation. The purposes of this study were to investigate the association between 607 C/A and -137 G/C SNPs on the concentrations of IL-18, and to measure TGF-β and SOD activity in radiation workers and control group., Material and Method: We enrolled 40 radiation workers and 40 non-radiation workers as a control group. We determined genotype distribution of -607 C/A and -137 G/C SNPs and their correlation with IL-18 concentration by using PCR-RFLP method. We also measured the IL-18, TGF-β concentration, and SOD activity by using Elisa assay., Results and Conclusion: No relationship was found between -607 C/A and -137 G/C on IL-18 concentrations in all genotype groups, and no significant difference in IL-18 and TGF-β concentrations in the radiation worker and control groups. Significant differences were found only in lower SOD activity in radiation workers compared to controls. The -607 C/A and -137 G/C did not significantly correlate with IL-18 cytokine production in all genotypes. There was no significant difference between IL-18 and TGF-β concentrations in the radiation worker and control groups. However, there was a very significant decrease in the SOD activity of the radiation workers by 3.31 times compared to the controls.

Background: Adolescent pregnancy is prevalent in Asian-African countries. Hence, it is critical to track the progress of research and development trends related to the topic. The study aimed to characterize published articles on teen pregnancy by measuring the authors' country and affiliation, most relevant and cited journals, thematic research, and growth trends., Materials and Methods: Descriptive statistics and retrospective bibliometric analysis were used. Using the Scopus database, we collected published articles from 2010 to 2023. Titles and abstracts were screened. Eligible papers were reviewed based on co-occurrence analysis. Classification and visualization of results were conducted using VOSviewer software version 1.6.17., Results: A total of 369 articles were relevant and included in the review. The countries that produced the most publications were South Africa and the United States. The most relevant affiliation was with the University of Cape Town, the University of the Witwatersrand, and the University of Kwazulu-Natal. The most relevant source journals were PLOS One, Reproductive Health, and BMC Public Health. The most cited sources were Lancet, PLOS One, and AIDS Journal. Four clusters were obtained that reflect the main topics, including human immunodeficiency virus (HIV) infection, child, health services, and pregnancy. The current research terms include HIV infection and maternal health services., Conclusions: This study suggests more research on the term "vertical transmission, breastfeeding, and partner violence" related to teen pregnancy. Objective: The study aimed to conduct a comparative analysis of the effects of incorporating hydrazinyl coumarin derivative (HCD) in resin-modified (RMGIC) and conventional glass ionomer cement (cGIC) on their release profiles and antibacterial properties., Method: Resin-modified GIC, Fuji II LC (F2) and high-fluoride cGIC, Fuji VII (F7) were used as controls. HCD was synthesized in-house, incorporated into both RMGIC and cGICs at 1 % and 2 % weight percentages (w/w), and chemically analyzed using Fourier transform infrared (FTIR) spectroscopy. Then, the F2 containing HCD (GIC-HCD F2) and F7 containing HCD (GIC-HCD F7) were evaluated for HCD and fluoride release profiles using UV Visible spectrophotometer and pH/ISE benchtop fluoridemeter, respectively. The antibacterial properties were assessed against Streptococcus sanguinis using the agar well diffusion method and measurement of bacterial growth turbidity, followed by the observation of the bacterial morphology using scanning electron microscope. The data were statistically analyzed using one-way ANOVA and Bonferroni post-hoc tests., Results: The FTIR spectra confirmed the presence of HCD in the GIC-HCD matrices. HCD was successfully released from both GIC-HCD F2 and GIC-HCD F7 matrices at both weight percentages. Higher fluoride release and inhibitory zones were observed compared to the control groups, with GIC-HCD F2 having a more significant effect than GIC-HCD F7. Additionally, the incorporation of HCD slowed down the growth of Streptococcus sanguinis and showed remarkable changes in bacterial shape specifically on GIC-HCD F2., Conclusion: The incorporation of HCD into both RMGIC and cGIC improved fluoride release and enhanced the antibacterial activities, with a more significant effect observed in RMGIC compared to cGIC., Competing Interests: The authors have no conflict of interest to declare., (© 2024 The Authors.)

This study investigates the factors associated with stunting prevalence in Indonesia, utilizing a generalized lasso framework with modified penalty matrices to accommodate spatio-temporal data structures. Novel approaches are introduced to construct the penalty matrices, with particular focus on defining neighborhood structures. The proposed method is applied to data from 34 Indonesian provinces, covering the years 2019 to 2023. The primary outcome is stunting prevalence, modeled against nine predictor variables: poverty, exclusive breastfeeding, low birth weight (LBW), high school completion, access to proper sanitation, unmet health service needs, Gross Domestic Product (GDP), calorie consumption, and protein consumption. A total of nine spatio-temporal models were compared, including a modified generalized lasso with three distinct penalty matrices for each two tuning selection methods and a generalized ridge regression with three penalty matrices. Results indicate that the generalized lasso model with a 3-nearest neighbor adjacency matrix outperformed the alternatives. Temporal variations were observed in the effects of exclusive breastfeeding, LBW, high school completion, and unmet health service needs. Positive associations with stunting prevalence were identified for poverty, exclusive breastfeeding, LBW, and unmet health service needs, while negative associations were found for high school completion rates, access to proper sanitation, GDP, calorie intake, and protein consumption. The strongest associations were observed in parts of Sumatra, Sulawesi, and Jakarta. Three cyclic diarylheptanoids, myricanol ( 1 ), myricanone ( 2 ), and porson ( 3 ), were isolated from Myrica javanica (Myricaceae). As a major component, myricanol ( 1 ) was obtained from dry powdered bark and twigs (up to 1.6%). Transformation of myricanol ( 1 ) afforded 5-prenylmyricanol ( 4 ) and 5-benzylmyricanol ( 5 ) in 84.5% and 65% yields, respectively. The bioactivities of the isolated cyclic diarylheptanoids and their derivatives were investigated to determine their cytotoxicity and DPPH (2,2-diphenyl-1-picrylhydrazyl) scavenging activities. The cytotoxicity assay against murine leukaemia P-388 cells demonstrated that compounds 4 and 5 showed an almost two-fold increase in the activity of their parent molecule ( 1 ), with an IC 50 value of 12 µM. Furthermore, the free radical scavenging assay showed that myricanol ( 1 ) had the highest radical scavenging activity, revealing the importance of the free phenolic group (IC 50 39.3 µM).

Purpose: This study aims to determine the effects of Islamic Mindfulness-based Caring (IMC) on fatigue among breast cancer patients. Design: A quasi-experimental study was undertaken in the chemotherapy unit of a hospital, in Bandung, Indonesia. Methods: A total of 112 females with stage III breast cancer undertaking chemotherapy treatment were recruited by purposive sampling technique. The experimental group (n = 53) was given both IMC and usual care, whereas the control group (n = 59) received only the usual care. Data were collected on days 1, 3, 23, and 44 using the Functional Assessment of Chronic Illness Therapy-Fatigue (FACIT-F) and demographic information tools. Data analyses were carried out with the Mixed-Effects Linear Regression. Findings: We found a significant difference in fatigue between the control and experimental groups on days 23 (mean: 29.34-34.92; p  = .001) and 44 (mean: 28.68-37.89; p  < .001) after IMC intervention. The increase of time was significantly associated with a lower fatigue score ( p  < .05). Mixed-Effect Linear Regression showed a significant interaction between time and intervention of IMC effect on fatigue score on time-4 (adjusted β coefficient: 6.62; 95% CI: 2.17-11.06). Conclusions: Nurse-administered IMC is capable of reducing fatigue among breast cancer patients who underwent chemotherapy.

Statement of Problem: The stability of mandibular removable partial dentures with bilateral distal extensions may be improved with the controlled tissue support achieved by using the altered cast impression technique, although this process is time-consuming and technique-sensitive., Purpose: The purpose of this pilot clinical study was to compare casts generated from a conventional definitive impression with casts generated from an altered cast impression using a 3-dimensional (3D) analysis software program., Material and Methods: Three partially edentulous participants with mandibular Kennedy Class I were enrolled, and impressions were made with the 2 techniques and poured in stone. The casts were scanned, aligned, and superimposed by using a 3D analysis software program. Surface deviations were measured to evaluate the differences in displacement induced by the impression on the tissue surface. Five observations were made in 4 different areas on each partially edentulous side. Means from these observations were generated, and the Wilcoxon and Mann Whitney tests were performed for all data to assess the differences between the right and left sides in the same participant and among the 3 participants (α=.05)., Results: The casts made from the altered cast impression had an overall mean ±standard deviation displacement of -0.05 ±1.25 mm on the right and left sides of the mandibular buccal shelf area. Moreover, the greatest overall difference of about 0.45 ±0.41 mm occurred on the lingual slope of the residual ridge, and the differences in the other areas were 0.10 ±0.99 mm (crest of the residual ridge) and 0.16 ±0.66 mm (buccal slope of the residual ridge). The overall differences varied statistically between significance and nonsignificance for the same participant and among the 3 participants., Conclusions: A digital comparative analysis of the conventional and altered cast methods of recording the bilateral distal extension areas in partially edentulous participants showed that the altered cast method exhibited more displacement on the buccal vestibule or buccal shelf area compared with other examined areas. Aims: With improving cancer survivorship, cardiovascular disease (CVD) has become a leading cause of death in breast cancer (BC) survivors. At present, there is no prospectively validated, contemporary risk assessment tool specific to this patient cohort. Accordingly, we sought to investigate long-term cardiovascular outcomes in early-stage BC patients utilising a well characterised database at a quaternary referral centre. With the assembly of this cohort, we have derived a BC cardiovascular risk index titled the 'CRIB (Cardiovascular Risk Index in Breast Cancer)' to estimate the risk of a major adverse cardiovascular event (MACE) in women undergoing treatment for BC., Methods: A retrospective cohort study was conducted examining all female patients aged ≥18 years of age who underwent treatment for early-stage BC at a cancer centre in Melbourne, Australia, between 2009 and 2019. The primary aim of this study was to assess causes and predictors of MACE., Results: A total of 1,173 women with early-stage BC were included. During a median follow-up of 4.4 (1.8-6.7) years, 80 (6.8%) women experienced a MACE. These women were more likely to be older, with a high burden of cardiovascular risk factors and were more likely to have a history of established coronary artery disease (CAD) (p≤0.001 for all). A CRIB ≥3 (2 points: renal impairment, 1 point: age ≥65 years, body mass index [BMI]>27, diabetes, hypertension, history of smoking) demonstrated moderate discrimination (c-statistic 0.75) with appropriate calibration. A CRIB ≥3, which represented 23.9% of our cohort, was associated with a high risk of MACE (odds ratio [OR] 17.85, 95% confidence interval [CI] 6.36-50.05; p<0.001). A total of 138 (11.8%) women died during the study period. &lt;b&gt;Background and Objective:&lt;/b&gt; Gamma irradiation induces genotoxicity, characterized by the formation of extra-nuclear bodies and left behind during the anaphase stage of cell division, often referred to as a micronucleus (MN). The present work aims to monitor exposure to ionizing radiation as a genotoxic agent in the lymphocytes of workers at radiation energy centers. &lt;b&gt;Materials and Methods:&lt;/b&gt; The lymphocyte cytokinesis block micronucleus assay used and analyzed the correlation between the Nuclear Division Index (NDI), age, blood type and the number of micronuclei (MN). Blood samples were collected from 20 volunteers in heparin tubes, exposed to 2 Gy gamma rays and cultured &lt;i&gt;in vitro&lt;/i&gt;. &lt;b&gt;Results:&lt;/b&gt; A significant difference in the number of micronuclei between blood group A and blood groups A, B and AB. The Nuclear Division Index (NDI) value for lymphocytes of radiation energy center workers after gamma radiation was significant (1.74±0.1) but still within the normal range. Neither MN frequency nor NDI values correlated with age, but MN frequency showed a correlation with blood type. &lt;b&gt;Conclusion:&lt;/b&gt; The gamma irradiation did not induce a cytostatic effect but proved genotoxic to the lymphocytes of radiation energy center workers. Notably, blood type A demonstrated higher sensitivity to gamma radiation.

Background: Moringa oleifera fruit extract contains beneficial chemical compounds. This study was conducted to observed the power of antioxidant against liver injury by 2-Nitropropane induction in an obese male mice model. Methods: This research was in vivo laboratory experimental study with a post-test control design group only. The population was obese male mice models, Swiss strain, aged 6-8 weeks, weighing between 60-80 gr. The research sample was determined by Federer's formula for a complete randomized design experimental test, group N (control), O1 (induced by 2-Nitropropane intraperitoneal (i.p) once), O2 (induced by 2-Nitropropane i.p twice), P1 (induced by 2-Nitropropane i.p. once and gavage with M. oleifera fruit extract 500mg/kg bodyweight (BW) once a day), P2 (induced by 2-Nitropropane i.p. twice and gavage of M. oleifera fruit extract 500mg/kg BW once a day), and P3 (induced by 2-Nitropropane i.p. twice and gavage of vitamin C 500mg/kg BW once a day). Antioxidant potential parameters were measured by levels of malondialdehide (MDA), glutation (GSH), 8-hydroxy-2'-deoxyguanosine (8-OHdG), catalase activity, manganese superoxide dismutase (MnSOD), serum glutamic pyruvic transaminase (SGPT), serum glutamic oxaloacetic transaminase (SGOT). This research was held at the Biochemistry laboratory of Medicine Faculty, UPN Veteran Jakarta in May-September 2020. Analysis was carried out using SPSS version 20.0. The parameters were tested using ANOVA. Results: MDA levels decreased, GSH increased, 8-OHdG decreased, catalase activity increased, MnSOD activity increased and SGOT, SGPT levels decreased. M. oleifera fruit extract was statistically proven to be a candidate for potential antioxidant against liver injury of 2-Nitropropane induction in obese male mice model. Background: Child sexual abuse (CSA) has emerged as a global concern, particularly affecting children in Indonesia. However, there remains a scarcity of research on CSA within the Indonesian context. Objective: This study aimed to explore the educational needs of primary school teachers in preventing child sexual abuse., Methods: A qualitative descriptive study design was employed, with eight teachers and school principals selected through purposive sampling. Data were collected through focus group discussions and analyzed using content analysis., Results: Six categories emerged: 1) The risk of sexual abuse, 2) The necessity for clear CSA preventive regulations and sanctions for abusers, 3) Lack of CSA program socialization, 4) The need for structured CSA prevention education for children, 5) The importance of effective coordination with various relevant stakeholders, and 6) The presence of barriers and obstacles., Conclusion: This study provides valuable insights into the educational prerequisites for teachers to implement CSA prevention measures effectively. The findings emphasize the pressing need for school teachers to develop and implement CSA prevention programs, with the government's and nursing professionals' support, to enhance educators' abilities in combating CSA. This study evaluated the use of urethane dimethacrylate (UDMA) as a base monomer to prepare the newly developed flowable composite (FC) using nanohybrid silica derived from rice husk in comparison to bisphenol A-glycidyl methacrylate (Bis-GMA) on the degree of conversion and physicomechanical properties. The different loadings of base monomer to diluent monomer were used at the ratio of 40:60, 50:50, and 60:40. The bonding analysis confirmed the presence of nanohybrid silica in the newly developed FC. Independent t -test revealed a statistically significant increase in the degree of conversion, depth of cure and Vickers hardness of the UDMA-based FC, while surface roughness showed comparable results between the two base monomers. In conclusion, UDMA-based FC demonstrated superior performance with 60%-65% conversions, a significantly higher depth of cure exceeding 1 mm which complies with the Internal Standard of Organization 4049 (ISO 4049), and a substantial increase in Vickers hardness numbers compared to Bis-GMA-based FC, making UDMA a suitable alternative to Bis-GMA as a base monomer in the formulation of this newly developed FC derived from rice husk., Competing Interests: Declaration of conflicting interestsThe author(s) declared no potential conflicts of interest with respect to the research, authorship, and/or publication of this article.

Background: Stunting is a common nutritional problem in children. Many risk factors contributing to stunting are identified in the literature. Vitamin D supplementation during pregnancy is useful for preventing impairment of fetal bone growth in the womb. However, lack of review in exploring supplementation of prenatal vitamin D is related to stunting in children., Purpose: This study aims to identify the effectiveness of prenatal vitamin D supplementation in preventing stunting., Methods: A literature review was conducted using PubMed, CINAHL, ScienceDirect, and Cochrane Library. Inclusion criteria were a randomized controlled trial study, published between 2011-2023, a full-text article for pregnant women and an independent variable supplement vitamin D to prevent children's stunting. The keywords used in English were "children OR child" AND "pregnant women" AND "preventive stunting" AND "vitamin D". Study quality was assessed using the Joanna Briggs Institute (JBI) appraisal tool., Results: From the results of the study selection of 511 articles, 11 articles met the inclusion and eligibility criteria for study analysis. Four studies indicated that the administration of vitamin D did not influence the length of the neonate's body or the Length for Age Z-Score (LAZ), used as an indicator of stunting. However, seven articles demonstrated that providing vitamin D supplements to pregnant women impacted various aspects of anthropometry, including the length of the neonate's body. Our findings show that the dosage used of vitamin D in pregnancy varied between 1400-60,000 per week. Overall, results of this study analysis show that the intervention of prenatal vitamin D supplementation has an impact on the prevention of stunting., Conclusion: This literature review highlighted the benefits of maternal vitamin D during pregnancy and for the children's growth. The purpose of this study was to systematically review the impact of nanofillers on the physicomechanical properties of resin-based pit and fissure sealants (RBS). This review included in vitro studies with full-length English-language articles reporting on the physicomechanical properties of nanofilled RBS until February 2023. PubMed, Web of Sciences, Scopus, and LILACS databases were accessed for literature searches. The review was formulated based on the Preferred Reporting Items for Systematic Review and Meta-Analyses (PRISMA) guidelines and used the Consolidated Standards of Reporting Trials (CONSORT) guidelines and risk of bias Cochrane tool for quality assessment. The search resulted in 539 papers, of which 22 were eligible to be included in the review. Inorganic, polymeric, core-shell, and composite nanomaterials were used to reinforce the studied RBS. The inherent nature of the nanomaterial used, its morphology, concentration, and volume used were the primary parameters that determined the nanomaterial's success as a filler in RBS. These parameters also influenced their interaction with the resin matrix, which influenced the final physicomechanical properties of RBS. Introduction: It is widely acknowledged that the socioeconomic circumstances of a family income are correctly reflected in that family expenditures. The dietary habits of families are influenced by socioeconomic circumstances that may affect stunting. Therefore, this study aims to investigate the household economic predictor stunting (mother's employment, family income, and family expenditure) in children under five., Methods: This study used a cross-sectional design. Six districts and cities have been chosen as study areas based on the areas with the highest incidence of stunting in West Java, Indonesia. This study conducted specifically from October to December 2018. Data were examined using descriptive statistics (frequency distribution) and a Spearman Rank bivariate test. A logistic regression was employed to determine the elements that serve as predictors of stunting., Results: A total of 731 women with children under five participated in the study. A bivariate analysis showed that mother's employment had an association with stunting among children under five ( p = 0.014). Meanwhile, family income and expenditure are not statistically significant associated with stunting ( p > 0.05). A multivariate analysis showed that mother's employment was a stunting predictor with odd ratio (OR) 1.810 ( p = 0.017). The OR value means that mothers who do not work have a 1.810 chance for their children to experience stunting compared with mothers who have work. However, family income and expenditure have negative projected values ( p = 0.580 and p = 0.398, respectively)., Conclusion: Children under five who are stunted are potentially predictive with mothers' employment, with a chance is higher in mother who do not work. Low-income communities have limited capital and access to money or loans from formal financial institutions. To solve the problems, the government provides solutions, one of them is by forming a microfinance program, namely Rural Agribusiness Business Development (PUAP). PUAP program is one of the grant activities to farmer group association (Gapoktan) with a total capital assistance of IDR 100 million. The problem with the 52,186 Gapoktan units that participated in PUAP activities, only 7,703 units (15%) were transformed into Agribusiness Microfinance Institutions (LKMA). This paper differs from others as it briefly explains the PUAP/MFI's institutional transformation and the factors that affect its sustainability, which is so far still limited discussed. The paper aims to see what transformations Gapoktan becomes an inclusive LKMA and the level of sustainability of the LKMA. The research was conducted in Kendal Regency, Central Java- Indonesia, in 2022 on 5 LKMA. The process of transforming LKMA into an inclusive financial institution is analyzed descriptively. LKMA sustainability levels were analyzed using a multidimensional scaling (MDS) approach with the Rapfish application. So far, MDS with the Rapfish application is still very limited for microfinance analysis. MDS analysis is employed because it is relatively simple and effective for looking at sensitive attributes in improving sustainability and generating leverage attributes that can be used for policy-making. The result study shows that the transformation of PUAP into LKMA is driven by the ability to improve legality, financial governance and diversify the customer's business field. The five LKMAs have a sustainability status of 'sufficient' in running their business, with an index value of more than 50%. Objective: The purpose of this study was to assess the immune response and malondialdehyde levels in irradiated rats supplemented with Curcuma xanthorriza Roxb extract as a candidate for mitigating radiation exposure., Methods: Twenty-four male Wistar rats were grouped into eight treatment groups, then Curcuma xanthorrhiza Roxb extract was administered orally and irradiated at 6 Gy. Measurement of rats IL-6 and INF-γ was performed using a sandwich ELISA Kit, while the MDA concentration was quantified according to the method of Wills (1971). The statistical test is determined by one way ANOVA test. P-value <0.05 was considered statistically significant., Result: The concentration of IL-6 in all groups showed no statistically significant difference (P=0.18). There was an increase in the concentration of IL-6 in the group of rats irradiated with 6 Gy for 7 days and 14 days. Meanwhile, the INF-γ concentration also showed no significant results in all treatment groups (P=0.28). The average of MDA concentration showed a significant difference in the liver and spleen of irradiated rats at 6 Gy for 14 days compared to the control (0.044 nmol/mg vs 0.008 nmol/mg, P=0.03 and 0.032 nmol/mg vs 0.014 nmol/mg, P=0.05, respectively)., Conclusion: The administration of Curcuma xanthorriza Xorb extract was able to reduce MDA concentrations in the liver and spleen although not statistically significant. In addition, exposure to ionizing radiation at a dose of 6 Gy significantly increased lipid peroxidation in the liver and spleen by 5.5 times and 2.3 times, respectively.

This clinical report describes how a hollow obturator prosthesis was designed and fabricated for an 82-year-old partially edentulous patient with a large palatal defect. Computer-aided design (CAD) was used to design, articulate, and align the mandibular denture with the obturator prosthesis. The prosthesis was printed, adjusted chairside, rescanned, and made hollow by using a CAD software program. The prosthesis was printed in resin with a dental 3D printer. Quantitative evaluations of clinical (prosthesis dimensions, rest, and occlusal vertical dimensions) and virtual (surface area, volume, weight, interpoint mismatches, spatial overlap) parameters found that the 3D-printed prosthesis required an additional 5% chairside modification. The greatest differences in volume (24.7% less) and weight (22.2% less) were observed when the modified obturator bulb was made hollow via CAD. The innovation of nanocellulose as reinforcement filler in composites has been a topic of interest in the development of new biomaterials. The objective of this study was to investigate the mechanical properties of a nanohybrid dental composite made of rice husk silica and loaded with different percentages of kenaf nanocellulose. Kenaf cellulose nanocrystals (CNC) were isolated and characterized using a transmission electron microscope (TEM) (Libra 120, Carl Zeiss, Germany). The experimental composite was fabricated with fiber loadings of 1 wt%, 2 wt%, 3 wt%, 4 wt%, and 6 wt% silane-treated kenaf CNC, and subjected to a flexural and compressive strength test ( n = 7) using an Instron Universal Testing Machine (Shimadzu, Kyoto, Japan), followed by a scanning electron microscopic assessment of the flexural specimen's fracture surface using a scanning electron microscope (SEM) (FEI Quanta FEG 450, Hillsborough, OR, USA). Commercial composites Filtek Z350XT (3M ESPE, St. Paul, MN, USA), Neofil (Kerr Corporation, Orange, CA, USA) and Ever-X Posterior (GC Corporation, Tokyo, Japan) were used as a comparison. The average diameter of kenaf CNC under TEM was 6 nm. For flexural and compressive strength tests, one-way ANOVA showed a statistically significant difference ( p < 0.05) between all groups. Compared to the control group (0 wt%), the incorporation of kenaf CNC (1 wt%) into rice husk silica nanohybrid dental composite showed a slight improvement in mechanical properties and modes of reinforcement, which was reflected in SEM images of the fracture surface. The optimum dental composite reinforcement made of rice husk was 1 wt% kenaf CNC. Excessive fiber loading results in a decline in mechanical properties. CNC derived from natural sources may be a viable alternative as a reinforcement co-filler at low concentrations.

The Maluku Province is an underdeveloped region in Indonesia with over 1,340 scattered islands. Due to the limited health facilities and transportation infrastructure, access to healthcare is very challenging. Here, we combined data from various sources to locate the population clusters, health facilities, roads, and ports/docks, and then utilize geographic information systems (GIS) to estimate distances from residents to health facilities. Health workforce distribution data was then integrated to elucidate overall healthcare equity among districts in the province. The average distances to puskesmas (primary health clinics) were 8.89 km (by land) and 18.43 km (by land and water) respectively, and the average distances to hospitals were 56.19 km (by land) and 73.09 km (by land and water), with large disparities within and among districts. Analysis of health workforce data shows that 65% of 207 puskesmas lack physicians, while 49% lack midwives. Ambon, Tual, and Southeast Maluku have the highest health equity, while East Ceram, Buru, and South Buru have the lowest. In general, this study demonstrates the utility of GIS and spatial analyses, which can help identify problem areas in healthcare accessibility and equity in archipelago settings, and provide recommendations to stakeholders such as public health officials and district administrators., Competing Interests: The authors have declared that no competing interests exist., (Copyright: © 2023 Leosari et al. Heart failure has reached epidemic proportions in a progressively ageing population. The molecular mechanisms underlying heart failure remain elusive, but evidence indicates that DNA damage is enhanced in failing hearts. Here, we tested the hypothesis that endogenous DNA repair in cardiomyocytes is critical for maintaining normal cardiac function, so that perturbed repair of spontaneous DNA damage drives early onset of heart failure. To increase the burden of spontaneous DNA damage, we knocked out the DNA repair endonucleases xeroderma pigmentosum complementation group G (XPG) and excision repair cross-complementation group 1 (ERCC1), either systemically or cardiomyocyte-restricted, and studied the effects on cardiac function and structure. Loss of DNA repair permitted normal heart development but subsequently caused progressive deterioration of cardiac function, resulting in overt congestive heart failure and premature death within 6 months. Cardiac biopsies revealed increased oxidative stress associated with increased fibrosis and apoptosis. Moreover, gene set enrichment analysis showed enrichment of pathways associated with impaired DNA repair and apoptosis, and identified TP53 as one of the top active upstream transcription regulators. In support of the observed cardiac phenotype in mutant mice, several genetic variants in the ERCC1 and XPG gene in human GWAS data were found to be associated with cardiac remodelling and dysfunction. In conclusion, unrepaired spontaneous DNA damage in differentiated cardiomyocytes drives early onset of cardiac failure. These observations implicate DNA damage as a potential novel therapeutic target and highlight systemic and cardiomyocyte-restricted DNA repair-deficient mouse mutants as bona fide models of heart failure., (© 2023 The Authors. Asparagine is a non-essential amino acid since it can either be taken up via the diet or synthesized by asparagine synthetase. Acute lymphoblastic leukemia (ALL) cells do not express asparagine synthetase or express it only minimally, which makes them completely dependent on extracellular asparagine for their growth and survival. This dependency makes ALL cells vulnerable to treatment with L-asparaginase, an enzyme that hydrolyzes asparagine. To date, all clinically approved L-asparaginases have significant L-glutaminase co-activity, associated with non-immune related toxic side effects observed during therapy. Therefore, reduction of L-glutaminase co-activity with concomitant maintenance of its anticancer L-asparaginase effect may effectively improve the tolerability of this unique drug. Previously, we designed a new alternative variant of Erwinia chrysanthemi (ErA; Erwinaze) with decreased L-glutaminase co-activity, while maintaining its L-asparaginase activity, by the introduction of three key mutations around the active site (ErA-TM). However, Erwinaze and our ErA-TM variant have very short half-lives in vivo. Here, we show that the fusion of ErA-TM with an albumin binding domain (ABD)-tag significantly increases its in vivo persistence. In addition, we evaluated the in vivo therapeutic efficacy of ABD-ErA-TM in a B-ALL xenograft model of SUP-B15. Our results show a comparable long-lasting durable antileukemic effect between the standard-of-care pegylated-asparaginase and ABD-ErA-TM L-asparaginase, but with fewer co-glutaminase-related acute side effects. Since the toxic side effects of current L-asparaginases often result in treatment discontinuation in ALL patients, this novel ErA-TM variant with ultra-low L-glutaminase co-activity and long in vivo persistence may have great clinical potential.
