Many-analysts studies explore how well an empirical claim withstands plausible alternative analyses of the same dataset by multiple, independent analysis teams. Conclusions from these studies typically rely on a single outcome metric (e.g. effect size) provided by each analysis team. Although informative about the range of plausible effects in a dataset, a single effect size from each team does not provide a complete, nuanced understanding of how analysis choices are related to the outcome. We used the Delphi consensus technique with input from 37 experts to develop an 18-item subjective evidence evaluation survey (SEES) to evaluate how each analysis team views the methodological appropriateness of the research design and the strength of evidence for the hypothesis. We illustrate the usefulness of the SEES in providing richer evidence assessment with pilot data from a previous many-analysts study.

Abstract Background Prostatic carcinoma (PCA) is a rare but severe condition in dogs that is similar to the androgen-independent form of PCA in men. In contrast to humans, PCA is difficult to diagnose in dogs as reliable biomarkers, available for PCA screening in human medicine, are currently lacking in small animal oncology. Calprotectin (S100A8/A9) and S100A12 are Ca2+-binding proteins of the innate immune system with promising potential to distinguish malignant from benign urogenital tract conditions, similar to the blood neutrophil-to-lymphocyte-ratio (NLR). However, both have not yet been extensively investigated in dogs with PCA. Thus, this study aimed to evaluate the expression of the S100/calgranulins (calprotectin, S100A12, and their ratio [Cal-ratio]) in prostatic biopsies from nine dogs with PCA and compare them to those in dogs with benign prostatic lesions (eight dogs with prostatitis and ten dogs with benign prostatic hyperplasia [BPH]) as well as five healthy controls. In addition, blood NLRs were investigated in twelve dogs with PCA and 22 dogs with benign prostatic conditions. Results Tissue S100A8/A9+ cell counts did not differ significantly between tissue from PCA and prostatitis cases (P = 0.0659) but were significantly higher in dogs with prostatitis than BPH (P = 0.0013) or controls (P = 0.0033). S100A12+ cell counts were significantly lower in PCA tissues than in prostatitis tissue (P = 0.0458) but did not differ compared to BPH tissue (P = 0.6499) or tissue from controls (P = 0.0622). Cal-ratios did not differ significantly among the groups but were highest in prostatitis tissues and significantly higher in those dogs with poor prostatitis outcomes than in patients that were still alive at the end of the study (P = 0.0455). Blood NLR strongly correlated with prostatic tissue S100A8/A9+ cell counts in dogs with PCA (ρ = 0.81, P = 0.0499) but did not differ among the disease groups of dogs. Conclusions This study suggests that the S100/calgranulins play a role in malignant (PCA) and benign (prostatic inflammation) prostatic conditions and supports previous results in lower urinary tract conditions in dogs. These molecules might be linked to the inflammatory environment with potential effects on the inflammasome. The blood NLR does not appear to aid in distinguishing prostatic conditions in dogs. Further investigation of the S100/calgranulin pathways and their role in modulation of tumor development, progression, and metastasis in PCA is warranted.

Many scholars argue that the artistic merit of films is diminishing as directors opt for producers’ more business-oriented approach. However, research discussing the impact of commercial success on filmmaking lacks focus on directors and their perspectives. This study seeks to understand the effect of commercial success on directors’ artistic visions for films. Using semi-structured interviews with Canadian filmmakers, this study aimed to learn from filmmakers by gaining direct access to their perspectives. This study found that while many directors are keen to add “commercial elements” to their films, this is generally in the hopes of appealing to a wider audience, not to generate commercial success alone. Moreover, most directors value artistic recognition from critics above the desire to garner commercial success. The findings of this study can be used to inform critics and audiences about how commercial pressures impact the artistic elements in a film.

Lake Sevan is the largest freshwater lake in the Caucasus region and one of Eurasia's largest alpine lakes. The lake has been experiencing growing cyanobacteria blooms; however, the relevance of external and internal phosphorus (P) loading to its eutrophication is still not known. We carried out a sediment investigation of Lake Sevan to estimate the potentially mobile P, which could be a source of internal total phosphorus (TP)-loading; we also estimated external TP-loading and retention. The study was carried out at four sampling points of different depths to determine the spatial variability of P in 2018. The sediments had elevated TP contents at the sediment surface; potentially mobile P ranged from 20 to 60% in the top 2 cm. The upper 4 cm sediment had an elevated content of TN (8 to 16.1 mg N g-1 dw) and TP (1.2 to 1.7 mg P g-1 dw). Spatial variability of most of the measured parameters was more prominent in the upper 3 cm. External TP loading was estimated to be 110 tons annually, and the TP retention was 85%. The estimated TP stored in the top cm of the sediment is 1,500 tons. The potential for P release is high; the short-term exchange between oxic and anoxic overlying water could release 0.01 to 0.02 mg P L-1 from the top cm of the sediment, and long-term diagenesis and burial could release about 0.12 mg P cm-2. Internal P-loading in Lake Sevan may play an essential role in eutrophication, especially given the long flushing time of Lake Sevan.

Background: Even though tumours are considered to occur frequently in pet hamsters, there is only a small number of scientific reports in current literature. Methods: Pathological reports from 177 hamsters were evaluated. Results: Of these, 78 were male and 75 were female. Median age of affected hamsters was 12 months (range 2–34). Integumental tumours were the most common neoplasms (62%, 109/177). As far as species was known, the number of Syrian hamsters (52%, 30/58) affected by tumours seemed to be lower than the number of affected dwarf hamsters (85%, 47/55). Tumours of the hematopoietic system were the second most frequently neoplasms (17%, 30/177). Relative number of neoplasms of the endocrine system, tumours of the digestive system (1.7%, 3/177) and other tumours (4%, 7/177 each) was low. The majority of integumental tumours were epithelial (66%; 91/126). Conclusion: This study aimed to analyze data from veterinary surgeries and pathological institutes about the occurrence of spontaneous tumours in Syrian hamsters and dwarf hamsters to give information about the frequency of tumours, prognosis and survival time. The neutrophil-to-lymphocyte ratio (NLR) can help in assessing inflammatory diseases, sepsis, and chronic hepatic conditions in humans. Dogs with congenital portosystemic shunts (PSSs) have signs of generalized inflammation, and the clinical signs can overlap with other conditions, including hypoadrenocorticism (HOC). Thus, the potential diagnostic and prognostic value of leukocyte ratios as surrogate markers was assessed in a retrospective case–control study including 106 dogs diagnosed with PSSs. The disease control groups were dogs with parenchymal hepatopathy (PH; n = 22) or HOC (n = 31). In the PSS dogs, the blood NLRs were associated with the severity of systemic inflammation but not with the shunt type, hepatoencephalopathy, systemic infection, or hypoglycemia. The baseline NLRs did not differ between the three disease groups, between medically and surgically treated PSS dogs, or between those with successful PSS ligation and dogs experiencing peri-/post-surgical complications. Abstract Background Urothelial carcinoma (UC) is the most common neoplasm of the canine lower urinary tract, affecting approximately 2% of dogs. Elderly female patients of certain breeds are predisposed, and clinical signs of UC can easily be confused with urinary tract infection or urolithiasis. Diagnosis and treatment are challenging given the lack of disease-specific markers and treatments. The S100A8/A9 complex and S100A12 protein are Ca2+-binding proteins expressed by cells of the innate immune system and have shown promise as urinary screening markers for UC. The neutrophil-to-lymphocyte ratio (NLR) can also aid in distinguishing certain neoplastic from inflammatory conditions. Our study aimed to evaluate the tissue expression of S100/calgranulins and the blood NLR in dogs with UC. Urinary bladder and/or urethral tissue samples from dogs with UC (n = 10), non-neoplastic inflammatory lesions (NNUTD; n = 6), and no histologic changes (n = 11) were evaluated using immunohistochemistry. Blood NLRs were analyzed in dogs with UC (n = 22) or NNUTD (n = 26). Results Tissue S100A12-positive cell counts were significantly higher in dogs with lower urinary tract disease than healthy controls (P = 0.0267 for UC, P = 0.0049 for NNUTD), with no significant difference between UC and NNUTD patients. Tissue S100A8/A9-positivity appeared to be higher with NNUTD than UC, but this difference did not reach statistical significance. The S100A8/A9+-to-S100A12+ ratio was significantly decreased in neoplastic and inflamed lower urinary tract tissue compared to histologically normal specimens (P = 0.0062 for UC, P = 0.0030 for NNUTD). NLRs were significantly higher in dogs with UC than in dogs with NNUTD, and a cut-off NLR of ≤ 2.83 distinguished UC from NNUTD with 41% sensitivity and 100% specificity. Higher NLRs were also associated with a poor overall survival time (P = 0.0417). Conclusions These results confirm that the S100/calgranulins play a role in the immune response to inflammatory and neoplastic lower urinary tract diseases in dogs, but the tissue expression of these proteins appears to differ from their concentrations reported in urine samples. Further investigations of the S100/calgranulin pathways in UC and their potential as diagnostic or prognostic tools and potential therapeutic targets are warranted. The NLR as a routinely available marker might be a useful surrogate to distinguish UC from inflammatory conditions.

Lake Sevan (Armenia) is one of the large freshwater high-mountain lakes of Eurasia. Detailed information about the extent and fate of trace elements on lake sediment and water quality has not been published yet. For this reason, surface sediment and water samples were collected from the southern and northern basins of Lake Sevan to determine trace element concentrations and assess the trace element behaviour. Geo-accumulation index, potential ecological risk index, and hazard index were calculated to estimate the environmental risk potential. In comparison to reference values, the investigated sediment samples contained elevated concentrations frequently for V, Cr, Co, Ni, Mo, Cd, Be, Ti, Rb, Sr, Se, Hf, and Th and occasionally for Cu, As, Li, B, Ag, Sb, Tl, Bi, U, La, Ce, Pr, Nd, Sm, Eu, Gd, Tb, Dy, Ho, Er, and Tm. An overall potential ecological risk posed by all the investigated trace elements in the sediments was assessed to be moderate-considerable, however, Mo, Hg, and Cd were the elements with the highest ecological risk potential. The two basins of the lake showed significantly different behaviour according to the investigated trace element contents in the sediments to be higher in the bigger basin compared to the smaller basin. In comparison to reference concentrations in water samples according to the use of the adapted geo-accumulation index, elevated values for Ti, Cr, Cu, Cd, and Pb were observed. Elevated concentration was also observed in the case of B in nearly all water samples in comparison with literature values. Nevertheless, several water samples can be seen as not strongly anthropogenic influenced by Co, Ni, Sn, Sb, Ag, Hg, and Bi. The concentrations of trace elements in the lake water caused health risks to humans particularly children in the case of lake water used for drinking purposes, moreover, As was the main element posing health hazards. The results point out further attention to the sources of elevated trace elements in Lake Sevan, including anthropogenic influences and geological characteristics.

Sharing research data allows the scientific community to verify and build upon published work. However, data sharing is not common practice yet. The reasons for not sharing data are myriad: Some are practical, others are more fear-related. One particular fear is that a reanalysis may expose errors. For this explanation, it would be interesting to know whether authors that do not share data genuinely made more errors than authors who do share data. (Wicherts, Bakker and Molenaar 2011) examined errors that can be discovered based on the published manuscript only, because it is impossible to reanalyze unavailable data. They found a higher prevalence of such errors in papers for which the data were not shared. However, (Nuijten et al. 2017) did not find support for this finding in three large studies. To shed more light on this relation, we conducted a replication of the study by (Wicherts et al. 2011). Our study consisted of two parts. In the first part, we reproduced the analyses from (Wicherts et al. 2011) to verify the results, and we carried out several alternative analytical approaches to evaluate the robustness of the results against other analytical decisions. In the second part, we used a unique and larger data set that originated from (Vanpaemel et al. 2015) on data sharing upon request for reanalysis, to replicate the findings in (Wicherts et al. 2011). We applied statcheck for the detection of consistency errors in all included papers and manually corrected false positives. Finally, we again assessed the robustness of the replication results against other analytical decisions. Everything taken together, we found no robust empirical evidence for the claim that not sharing research data for reanalysis is associated with consistency errors.

In a previous study, we identified candidate causative variants located in 24 functional candidate genes for milk protein and fatty acid composition in Montbéliarde, Normande, and Holstein cows. We designed these variants on the custom part of the EuroG10K BeadChip (Illumina Inc., San Diego, CA), which is routinely used for genomic selection analyses in French dairy cattle. To validate the effects of these candidate variants on milk composition and to estimate their effects on cheesemaking properties, a genome-wide association study was performed on milk protein, fatty acid and mineral composition, as well as on 9 cheesemaking traits (3 laboratory cheese yields, 5 coagulation traits, and milk pH). All the traits were predicted from midinfrared spectra in the Montbéliarde cow population of the Franche-Comté region. A total of 194 candidate variants located in 24 genes and 17 genomic regions were imputed on 19,862 cows with phenotypes and genotyped with either the BovineSNP50 (Illumina Inc.) or the EuroG10K BeadChip. We then tested the effect of each SNP in a mixed linear model including random polygenic effects estimated with a genomic relationship matrix. We confirm here the effects of candidate causative variants located in 17 functional candidate genes on both cheesemaking properties and milk composition traits. In each candidate gene, we identified the most plausible causative variant: 4 are missense in the ALPL, SLC26A4, CSN3, and SCD genes, 7 are located in 5'UTR (AGPAT6), 3' untranslated region (GPT), or upstream (CSN1S1, CSN1S2, PAEP, DGAT1, and PICALM) regions, and 6 are located in introns of the SLC37A1, MGST1, CSN2, BRI3BP, FASN, and ANKH genes. In dogs, the BRAF mutation (V595E) is common in bladder and prostate cancer and represents a specific diagnostic marker. Recent advantages in artificial intelligence (AI) offer new opportunities in the field of tumour marker detection. While AI histology studies have been conducted in humans to detect BRAF mutation in cancer, comparable studies in animals are lacking. In this study, we used commercially available AI histology software to predict BRAF mutation in whole slide images (WSI) of bladder urothelial carcinomas (UC) stained with haematoxylin and eosin (HE), based on a training (n = 81) and a validation set (n = 96). Among 96 WSI, 57 showed identical PCR and AI-based BRAF predictions, resulting in a sensitivity of 58% and a specificity of 63%. The sensitivity increased substantially to 89% when excluding small or poor-quality tissue sections. Test reliability depended on tumour differentiation (p < 0.01), presence of inflammation (p < 0.01), slide quality (p < 0.02) and sample size (p < 0.02). Based on a small subset of cases with available adjacent non-neoplastic urothelium, AI was able to distinguish malignant from benign epithelium. This is the first study to demonstrate the use of AI histology to predict BRAF mutation status in canine UC. Despite certain limitations, the results highlight the potential of AI in predicting molecular alterations in routine tissue sections.

Headwater streams play a major role for provision of ecosystem services, e.g. drinking water. We investigated a high-altitude headwater catchment of the Kharaa River (including 41 1st-order rivers) to understand the impact of land cover (especially forest cover), environment and human usage on runoff, chemical water quality and macroinvertebrate fauna in a river basin under discontinuous permafrost conditions in an arid, sparsely populated region of Mongolia. To verify our hypotheses that different landuses and environmental impacts in permafrost headwaters influence water quality, we investigated 105 sampling sites, 37 of them at intermittent stream sections without water flow. Discharge was positively impacted by land cover types steppe, grassland and forest and negatively by shrubland, forest burnt by wild fires (indicating a reduction of permafrost) and slope. Water quality was affected by altitude, longitude and latitude, shrub growth and water temperature. Shannon diversity of macroinvertebrates was driven by water temperature, iron content of the water, flow velocity, and subbasin size (adjusted R2 = 0.54). Sample plots clustered in three groups that differed in water chemistry, macroinvertebrate diversity, species composition and bio-indicators. Our study confirms that steppes and grasslands have a higher contribution to runoff than forests, forest cover has a positive impact on water quality, and diversity of macroinvertebrates is higher in sites with less nutrients and pollutants. The excellent ecological status of the upper reaches of the Kharaa is severely threatened by forest fires and human-induced climate change and urgently needs to be conserved.

Preregistration is a method to increase research transparency by documenting research decisions on a public, third-party repository prior to any influence by data. It is becoming increasingly popular in all subfields of psychology and beyond. Adherence to the preregistration plan may not always be feasible and even is not necessarily desirable, but without disclosure of deviations, readers who do not carefully consult the preregistration plan might get the incorrect impression that the study was exactly conducted and reported as planned. In this paper, we have investigated adherence and disclosure of deviations for all articles published with the Preregistered badge in Psychological Science between February 2015 and November 2017 and shared our findings with the corresponding authors for feedback. Two out of 27 preregistered studies contained no deviations from the preregistration plan. In one study, all deviations were disclosed. Nine studies disclosed none of the deviations. We mainly observed (un)disclosed deviations from the plan regarding the reported sample size, exclusion criteria and statistical analysis. This closer look at preregistrations of the first generation reveals possible hurdles for reporting preregistered studies and provides input for future reporting guidelines. We discuss the results and possible explanations, and provide recommendations for preregistered research.

Diagnosing chronic inflammatory enteropathies (CIE) in cats and differentiation from intestinal lymphoma (IL) using currently available diagnostics is challenging. Intestinally expressed S100/calgranulins, measured in fecal samples, appear to be useful non-invasive biomarkers for canine CIE but have not been evaluated in cats. We hypothesized S100/calgranulins to play a role in the pathogenesis of feline chronic enteropathies (FCE) and to correlate with clinical and/or histologic disease severity. This retrospective case-control study included patient data and gastrointestinal (GI) tissues from 16 cats with CIE, 8 cats with IL, and 16 controls with no clinical signs of GI disease. GI tissue biopsies were immunohistochemically stained using polyclonal α-S100A8/A9 and α-S100A12 antibodies. S100A8/A9+ and S100A12+ cells were detected in all GI segments, with few significant differences between CIE, IL, and controls and no difference between diseased groups. Segmental inflammatory lesions were moderately to strongly correlated with increased S100/calgranulin-positive cell counts. Clinical disease severity correlated with S100A12+ cell counts in cats with IL (ρ = 0.69, p = 0.042) and more severe diarrhea with colonic lamina propria S100A12+ cells with CIE (ρ = 0.78, p = 0.021) and duodenal S100A8/A9+ cells with IL (ρ = 0.71, p = 0.032). These findings suggest a role of the S100/calgranulins in the pathogenesis of the spectrum of FCE, including CIE and IL.

Histoplasmosis has been previously diagnosed in animals from Europe. The aim of this study is to review the literature on these reports, to analyze cases diagnosed at our laboratory (2000–2022) and to improve molecular typing of Histoplasma capsulatum directly from tissue to study the molecular epidemiology of Histoplasma capsulatum causing animal infections in Europe. Including 15 cases studied in our laboratory, we identified 39 cases of animal histoplasmosis between 1968 and 2022. They were diagnosed mostly in superficial tissue biopsies from cats and badgers from Central Europe. Using phylogenetic analyses of six partial genes, we were able to classify eight of the etiological agents as belonging to a highly supported lineage within the Eurasian clade. This study confirms the occurrence of autochthonous histoplasmosis in animals in Central Europe and proposes the addition of new loci to the MLST scheme to study the molecular epidemiology of histoplasmosis using either formalin-fixed paraffin-embedded tissue and fresh or cadaveric biopsies.

The analysis of dissolved organic matter (DOM) using high-field Fourier transform ion cyclotron resonance mass spectrometry (FTICR-MS) poses challenges in molecular formula assignment. The consideration of 13C isotopes provides new insights into the consistent elemental formula solutions. Modem software helps to overcome misinterpretation, but false assignments of molecular classes to mass peaks have rarely been elucidated until now. It will be demonstrated that this can be important with formula assignments comprising exactly five nitrogen and two sulfur atoms in DOM data sets: the molecular class CHON5S2. The existence of such components in DOM under tripeptide Met-His-Cys formed with the formula C14H23O4N5S2 cannot be excluded; however, components containing 5 N and 2 S should be suspected to not be highly abundant The true elemental compositions of such unusual "N5S2 moieties" were calculated using Suwannee River fulvic acid (SRFA) data from the literature and one data set from acidic pit lake pore water. The replacement of a H3N5S2 moiety with a 13C112C5O4 moiety explained more than 95% of the questionable "N5S2 moieties". Abstract Background The toxicological characterization of sediments is an essential task to monitor the quality of aquatic environments. Many hazardous pollutants may accumulate in sediments and pose a risk to the aquatic community. The present study provides an attempt to integrate a diagnostic whole mixture assessment workflow based on a slightly modified Danio rerio embryo acute toxicity test with chemical characterization. Danio rerio embryos were directly exposed to sieved sediment (≤ 63 μm) for 96 h. Sediment samples were collected from three polluted sites (Kramfors, Sundsvall and Örnsköldsvik) in the Gulf of Bothnia (Sweden) which are characterized by a long history of pulp and paper industry impact. Effect data were supported by chemical analyses of 237 organic pollutants and 30 trace elements. Results The results show that malformations and neurotoxic compounds are the main drivers of differentiation in chemical and effects analyses, respectively. Specific spinal cord malformations and delayed hatching were observed only in sediments from Kramfors while light hyperactivity was seen only after exposure to sediments from Sundsvall. Conclusions Our experiments demonstrate that specific chemical profiles lead to specific effect patterns in Danio rerio embryos. In fact, behavioral endpoints could help detect the exposure to neurotoxins, and the observation of body malformations seems to be a potential tool for the identification of site-specific pollutants as polychlorinated biphenyl (PCBs), brominated flame retardants (BFRs) and several pesticides. Overall, results show the suitability of Danio rerio embryos for the fast screening of sediment samples.

Lake Kivu, East Africa, is well known for its huge reservoir of dissolved methane (CH4) and carbon dioxide (CO2) in the stratified deep waters (below 250 m). The methane concentrations of up to ~ 20 mmol/l are sufficiently high for commercial gas extraction and power production. In view of the projected extraction capacity of up to several hundred MW in the next decades, reliable and accurate gas measurement techniques are required to closely monitor the evolution of gas concentrations. For this purpose, an intercomparison campaign for dissolved gas measurements was planned and conducted in March 2018. The applied measurement techniques included on-site mass spectrometry of continuously pumped sample water, gas chromatography of in-situ filled gas bags, an in-situ membrane inlet laser spectrometer sensor and a prototype sensor for total dissolved gas pressure (TDGP). We present the results of three datasets for CH4, two for CO2 and one for TDGP. The resulting methane profiles show a good agreement within a range of around 5-10% in the deep water. We also observe that TDGP measurements in the deep waters are systematically around 5 to 10% lower than TDGP computed from gas concentrations. Part of this difference may be attributed to the non-trivial conversion of concentration to partial pressure in gas-rich Lake Kivu. When comparing our data to past measurements, we cannot verify the previously suggested increase in methane concentrations since 1974. We therefore conclude that the methane and carbon dioxide concentrations in Lake Kivu are currently close to a steady state.

Within systems biology there is an increasing interest in the stochastic behaviour of biochemical reaction networks. An appropriate stochastic description is provided by the chemical master equation, which represents a continuous-time Markov chain (CTMC). The uniformisation technique is an efficient method to compute probability distributions of a CTMC if the number of states is manageable. However, the size of a CTMC that represents a biochemical reaction network is usually far beyond what is feasible. In this study, the authors present an on-the-fly variant of uniformisation, where they improve the original algorithm at the cost of a small approximation error. Abstract: Between 1999 and 2002, a former open-cast mine was filled with river water forming the recent Lake Goitsche. During filling initially acid water was neutralised. Phosphorus (P) imported from Mulde River was nearly completely removed from the water column by co-precipitation with iron (Fe) and aluminium (Al) and deposited in the sediment. During extremely high waters of the Mulde River in 2002, a dike breach facilitated a second high import of P into Lake Goitsche with suspended and dissolved matter. The analysis of total phosphorus (TP), however, showed that P again had been eliminated from the water body a few months after the flood event. Sediment investigations before filling with river water, during filling, and after the flood event were used to analyse the process of P immobilisation in a lake with acid mine drainage history. The ratios of Fe to soluble reactive P (SRP) of sediment pore water were up to three orders of magnitudes higher than in natural lakes and can serve as an indicator for potential internal P loading from sediments. The SRP concentrations at the oxic/anoxic boundary were near or below the limit of quantification (< 0.2μmol/L). Fe and manganese (Mn) redox cycling were responsible for hindering P dissolution from sediment to lake water. Finally it can be stated, that the risk of eutrophication for such a lake seems to be low. [Copyright &y& Elsevier]

Long-term changes in dissolved organic matter (DOM) quality, especially in humic-rich raw waters, may lead to intensive adaptions in drinking-water processing. However, seasonal DOM quality changes in standing waters are poorly understood. To fill this gap, the DOM quality of a German drinking water reservoir was investigated on a monthly basis by Fourier-transform ion cyclotron resonance mass spectrometry (FTICR MS) measurements and 2D fluorescence for 18 months. FTICR MS results showed seasonal changes of molecular formula (MF) intensities, indicating photochemical transformation of DOM as a significant process for DOM quality variation. For an assessment of the two humic-like components, identified by parallel factor analysis (PARAFAC) of excitation–emission matrices (EEM), their loadings were Spearman’s rank-correlated with the intensities of the FTICR MS-derived MF. One of the two PARAFAC components correlated to oxygen-rich and relatively unsaturated MF identified as easily photo-degradable, also known as coagulants in flocculation processes. The other PARAFAC component showed opposite seasonal fluctuations and correlated with more saturated MF identified as photo-products with some of them being potential precursors of disinfection byproducts. Our study indicated the importance of elucidating both the chemical background and seasonal behavior of DOM if raw water-quality control is implemented by bulk optical parameters.

Pore water of sediments plays an important role in aquatic systems as mediator and as the reactive zone between the sediment and surface water. Sediment pore waters with high ionic strength from acidified pit lakes were investigated to obtain information about the influence on the lake water quality. The analysis of soluble reactive phosphorus, nitrate, ammonium, silica, dissolved organic carbon, ferrous iron, sulfate, chloride, sodium, potassium, calcium, magnesium, manganese, and total dissolved iron was predetermined as the dataset required for evaluation of water quality. The data collection procedure was optimised by designing a methodology for stabilisation, dilution of pore water samples and adaptation of analytical methods. The developed methodology was evaluated with respect to the effort required in the laboratory under routine conditions. In the first round of analyses, 72% of 638 individual analyses from a random selection of pore water profiles were found to be within the calibration ranges. Remedial actions to handle the remaining 28% of invalid analytical results are exemplified. Differences between comparative analyses of some ions by continuous flow analysis, ion chromatography, and atomic emission spectroscopy were evaluated. The majority of results measured by ion chromatography differed on a highly significant level from results measured by atomic emission spectroscopy. Possible reasons, originating from the extreme sample matrix, are discussed. Finally, the designed methodology and the results of the method comparison are used to recommend the selection of analytical methods under specific conditions. [ABSTRACT FROM AUTHOR]

The objective of this study was to analyse the heritabilitiy of the lactational incidence of displaced abomasum (DA) and the relationships of DA with milk production traits in German Holstein cows. Data were recorded between February 1999 and January 2000 in cooperation with five veterinary practitioners. Their veterinary practices were located in the northern part of Lower Saxony. The investigation included 160 dairy farms under the official milk-recording scheme with 9315 cows. The lactational incidence of the left abomasal displacement amounted to 1.21%, and of the right abomasal displacement to 0.41%, respectively. The linear heritability estimates for the lactational incidences of left and right DA were h2 =0.05 ± 0.012 and h2 = 0.004 ± 0.005, respectively. Using the Dempster–Lerner-transformation the corresponding heritabilities were h2 = 0.53 and 0.09, respectively. Milk losses for the lactation when DA was diagnosed were significant and reached 1016 kg milk, 41 kg fat, 36 kg protein and 0.07% protein. Fat content significantly increased by 0.18%. The analysis could not show significant differences between cows diagnosed with DA and cows not diagnosed with DA in the 305-day milk production traits of the lactation preceding the diagnosis of DA. There was also no indication for an unequal distribution of breeding values for milk performance traits between cows with and without DA. The additive genetic correlations between 305-day milk performance and left DA were low. The results indicated that cows with a high milk production and superior breeding values for milk performance were not exposed to an increased risk for DA. [ABSTRACT FROM AUTHOR]
