Background: Amalgamation of metal-tolerant plant growth promoting rhizobacteria (PGPR) with biochar is a promising direction for the development of chemical-free biofertilizers that can mitigate environmental risks, enhance crop productivity and their biological value. The main objective of the work includes the evaluation of the influence of prepared bacterial biofertilizer (BF) on biometric growth parameters as well as physiological and biochemical characteristics of rapeseed (Brassica napus L.) at copper action. Methods: The prepared BF was based on novel metal tolerant strain of PGPR Buttiauxella sp. EA20 isolated from the rhizosphere of orchid Epipactis atrorubens and birch wood biochar (BC). The pot-scale experiments included six treatments: peat-containing control substrate (CS); CS + 200Cu (200 mg Cu/kg of soil); CS + 5% BC (v/v); CS + 5% BC + 200Cu; CS + 5% BF (v/v); CS + 5% BF + 200Cu. Results: Single Cu treatment caused the decrease in rapeseed leaf area, shoot and root length, fresh and dry biomass, as well as an increase in water saturation deficit, possibly due to damage of cell membranes by lipid peroxidation. Addition of BF or BC alone mitigated these harmful effects of copper. Application of BF, regardless of Cu addition, increased the rapeseed leaf area (1.6 times on average), plant fresh and dry biomass (2.5 times on average), and photosynthetic pigment content (1.8 times on average). In addition, BF treatment along with Cu enhanced the antioxidant activity of B. napus due to the accumulation of non-enzymatic antioxidants such as carotenoids, free proline and soluble phenolic compounds, including flavonoids. Moreover, plant enrichment with copper and essential macronutrients such as nitrogen, phosphorus and potassium was observed. Conclusions: The study concludes that application of complex biofertilizer based on metal tolerant PGPR strain Buttiauxella sp. EA20 and birch wood biochar mitigated the harmful effects of copper, enhanced the rapeseed growth and increased its biological value. Future perspective includes evaluation of the potential for using the resulting biofertilizer to improve the growth and biofortification of other crop species.

Lignification of the stem in zinnia provides its mechanical properties due to xylem formation, which depends on the stage of plant development and is responsible for the transport of water and minerals. The study was aimed at the lignin deposition, anatomical traits, biochemical markers of lignification, as well as the genetic regulation of this process in zinnia stem cross sections of different age during their radial growth. The anatomical traits were assessed on cross sections. The content of lignin (Cysteine-assisted sulfuric method (CASA) and the thioglycolic acid (TGA) methods), the spectrum of phenolics (by thin layer chromatography (TLC)), the total activity and the variety of class III peroxidases were determined. The expression level of genes regulating phenylpropanoids and lignin biosynthesis were assessed. We suggest that time-specific and organ-specific lignification is determined by the metabolism of phenolic compounds and depends on the expression of genes of the phenylpropanoid pathway. It was shown that in the hypocotyl, during xylem ring formation, lignification was associated with increased expression of phenylalanine ammonia-lyase (PAL) and cinnamyl alcohol dehydrogenase (CAD) genes responsible for the early stages of the phenylpropanoid pathway, and with the rise of class III peroxidases activity, including cationic isoforms. This caused increased content and diversity of phenolics in mature hypocotyl. In epicotyl, which is younger than the hypocotyl, the proportion of ferulic acid among phenolics increased, which could be considered as a marker of lignification in it. The high expression level of CAD and the activity of peroxidases, including anionic isoforms, led to accumulation of lignin. Thus, the hypocotyl and epicotyl, being characterized by different ages, differed in spectrum of phenolics, isoforms of class III peroxidases, expression of the PAL, cinnamate 4-hydroxylase (C4H), peroxidases III class (PRX), and laccase (LAC) genes, and lignin content.

Peroxidases of class III are common in various organisms. They are involved in lignin biosynthesis and plant protection against stressors. Peroxidases are presented in many isoforms, whose role is not always clear. The aim of this study is to analyze the amino acid sequences of reference peroxidases with known functions and peroxidases from Arabidopsis thaliana L. whose functions are unknown and to consider their putative roles in lignin biosynthesis. The structural and functional organization of peroxidases was analyzed by bioinformatical methods applied to open Internet sources. Seven reference peroxidases were chosen from four plant species: Zinnia sp., Armoracia rusticana P.G. Gaertn., Lycopersicon esculentum L. и Populus alba L. Twenty-four amino acid sequences of homologous peroxidases from A. thaliana were selected for the analyses with the BLAST service. Their molecular weights and isoelectric points were calculated. Multiple alignments of amino acid sequences and phylogenetic analysis were done. Sites of binding to monolignol substrates were identified in seven peroxidases from A. thaliana, and the enzymes were assigned to the groups of Sor G-peroxidases. Amino acid replacements in the primary structures of peroxidases were analyzed. Peroxidases from A. thaliana were clustered with reference peroxidases. They formed six clusters on the phylogenetic tree, three of which contained only A. thaliana peroxidases. Peroxidases within each cluster had similar molecular weights and isoelectric points, common localization of expression, and similar functions. Thus, the use of bioinformatics, databases, and published data bring us to assumptions as to the functions of several A. thaliana class III peroxidases. AtPrx39 peroxidase was shown to be affine to sinapyl alcohol; AtPrx54, to p-coumaryl and coniferyl alcohols. They are likely to participate in lignin biosynthesis.

Metal tolerant plant growth-promoting (PGP) rhizobacteria are promising for enhancing plant productivity under copper (Cu) stress. Present pot scale experiment was conducted on Brassica napus L. to check the efficiency of rhizobacteria isolated from the rhizosphere of Tussilago farfara L. growing on Cu-contaminated soils. Out of fifty Cu tolerant strains, three isolates which showed multiple PGP traits such as indole-3-acetic acid (IAA) synthesis, phosphate (PS) solubilization, siderophore and ammonia production were identified preliminarily by morphological and physiological characteristics followed by 16S rRNA gene sequencing. The best Bacillus altitudinis strain TF16a which showed IAA: 15.5 mg L−1, PS: 215 mg L−1, siderophore halo zone ratio of 3.0 with high ammonia production was selected to prepare a biochar-based biofertilizer (BF). Seedling test showed maximum growth of B. napus shoot and root in presence of 5% of BF and this concentration was selected for further experiment. The pot experiment included four treatments: control (soil), 100Cu (100 mg Cu kg−1 soil), 5%BF (v/v), and 5%BF+100Cu, which were carried out for 30 days, after which the morphological, physiological, and biochemical parameters of B. napus were studied. The Cu treatment caused its accumulation in shoot and root up to 16.9 and 30.4 mg kg−1 DW, respectively, and increased malondialdehyde (MDA) content by 20%. Application of BF with copper led to the decrease in the Cu accumulation by 20% for shoot and 28% for root while MDA content was the same as in the control. Both treatments of BF with and without Cu increased chlorophyll a and b content by 1.3 times on average as well as non-enzymatic antioxidants such as soluble phenolic compounds (1.3 times) and free proline (1.6 times). Moreover, BF + Cu led to the increase in the biomass of shoot and root by 30 and 60%, respectively, while there was no significant effect on the growth characteristics of plants after the addition of BF without Cu. The study elucidates that BF based on B.altitudinis strain TF16a and biochar can be a promising bioformulation which could increase rapeseed growth under the moderate Cu concentration in soil.

Epipactis atrorubens (Hoffm.) Besser. is a regionally rare orchid species with highly ornamental properties due to its very beautiful bright flowers, therefore it is of considerable interest as a horticultural plant for use in botanical gardens and greenhouses. The objective of the research was to assess metal accumulation and some pro- and antioxidant reactions in E. atrorubens, colonizing serpentine dumps post asbestos mining. Additionally, some physicochemical properties of substrates, microbiotic characteristics and water status were investigated in orchids growing on two serpentine dumps and in a natural forest habitat of the Middle Urals, Russia. The dump substrates were characterized by the strong stoniness and the high content of Mg, Ni, Cr and Co (by 1.8 times on average) compared to the natural habitat. In these sites, E. atrorubens was characterized by increased mycorrhization. In the rhizome and roots of E. atrorubens the concentrations of most metals studied were considerably higher (more than 4 times on average) than in the leaves. It was found that orchids colonizing serpentine dumps produced more lipid peroxidation products (by 1.4 times on average) in the leaves which was accompanied by the more active synthesis of such non-enzymatic antioxidants as ascorbate, free proline, soluble phenolic compounds (including flavonoids) and non-protein thiols. The study suggests that non-enzymatic antioxidants increased the adaptive potential of E. atrorubens and contributed to its naturalization on serpentine dumps post asbestos mining.

Zinnia elegans Jacq. is an ornamental plant, widely used in landscaping. Heavy-metal pollution in urban and rural areas is still increasing, which determines the actuality of studying plants’ reactions to pollutants. Zinnia was not sufficiently studied in this regard, so the aim of our research was to identify morphophysiological changes in this species under excess copper concentration in the soil. For this, we treated a growth substrate with 200 µM CuSO4 solution for 20 days. At the end of the treatment, several morphological, biochemical, and molecular genetic traits were evaluated: the root and the shoot size; the concentration of H2O2 and malondialdehyde (MDA), as indicators of stress; the amount of the phenolic compounds and lignin; and the level of the expression of genes, which encoded their biosynthesis. The Cu amount in the substrate and zinnia organs was quantified using atomic-absorption spectroscopy; hydrogen peroxide, MDA, and phenolic compounds were determined spectrophotometrically, while the amount of lignin was determined according to Klason. Real-time PCR was used for estimation of the gene-transcription level. Lignin in tissues was visualized by fluorescent microscopy. In experimental plants, Cu accumulation was higher in the root than in the stem. This caused an increase in stress markers and a decrease in the root and stem lengths. For the first time for zinnia, it was shown that for several genes—4-coumarate-CoA ligase (4CL), cinnamoyl alcohol dehydrogenase (CAD), and class III peroxidase (PRX)—the level of expression increased under copper treatment. The rise of the transcripts’ amount of these genes was accompanied by a thickening and lignification of the cell walls in the metaxylem vessels. Thus, the adaptation of zinnia to the excess Cu in the growth medium was associated with the metabolic changes in the phenylpropanoid pathway. As a result, the lignification increased in the root, which led to the accumulation of Cu in this organ and limited its translocation through the xylem to the stem, which provided plant growth.

Biofortification of plant food products including microgreens is one of the most promising approaches to combat with the problem of balanced nutrition for the human population. This study introduces the microbiological biofortification using plant growth-promoting (PGP) bacteria, specifically three strains of zincsolubilizing Pseudomonas sp. (STF10, STF14, STF16) isolated from the rhizosphere of Tussilago farfara L. The research explores their impact on growth parameters and photosynthetic pigments in pea (Pisum sativum L.) microgreens following iodine spraying, providing groundbreaking insights. The experimental results indicate that inoculation of pea seeds with these rhizobacteria significantly increased the fresh and dry biomass of 14-day-old microgreens (by 22 and 15%, respectively, on average) compared to non-inoculated plants. At the same time the content of chlorophylls and carotenoids in seedlings increased on average by 18 and 36%, respectively. Spraying with one form of iodine (0.01% KI or KIO3) slightly increased fresh and dry biomass (by 8% on average) and did not alter the content of photosynthetic pigments. Wherein iodine content in pea microgreens increased by 4.4 and 3.8 times, after KI and KIO3 spraying, respectively. By conducting experiments in a hydroponic nutrient solution, this study provides a robust platform for evaluating the biofortification potential of the three Pseudomonas strains along with foliar iodine feeding, marking a significant stride towards enhancing the nutritional profile of pea microgreens. Metabolic changes under stress are often studied in short-term experiments, revealing rapid responses in gene expression, enzyme activity, and the amount of antioxidants. In a long-term experiment, it is possible to identify adaptive changes in both primary and secondary metabolism. In this study, we characterized the physiological state of tobacco plants and assessed the amount and spectrum of phenolic compounds and the lignification of axial organs under excess copper stress in a long-term experiment (40 days). Plants were treated with 100 and 300 μM CuSO4, as well as a control (Knop solution). Copper accumulation, the size and anatomical structure of organs, stress markers, and the activity of antioxidant enzymes were studied. Lignin content was determined with the cysteine-assisted sulfuric method (CASA), and the metabolite profile and phenolic spectrum were determined with UHPLC-MS and thin-layer chromatography (TLC). Cu2+ mainly accumulated in the roots and, to a lesser extent, in the shoots. Copper sulfate (100 μM) slightly stimulated stem and leaf growth. A higher concentration (300 μM) caused oxidative stress; H2O2 content, superoxide dismutase (SOD), and guaiacol peroxidase (GPOX) activity increased in roots, and malondialdehyde (MDA) increased in all organs. The deposition of lignin increased in the roots and stems compared with the control. The content of free phenolics, which could be used as substrates for lignification, declined. The proportions of ferulic, cinnamic, and p-coumaric acids in the hydrolysate of bound phenolics were higher, and they tended toward additional lignification. The metabolic profile changed in both roots and stems at both concentrations, and changed in leaves only at a concentration of 300 μM. Thus, changes in the phenolic spectrum and the enhanced lignification of cell walls in the metaxylem of axial (root and stem) organs in tobacco can be considered important metabolic responses to stress caused by excess CuSO4. This article studies the binding of the antitumor drug doxorubicin (DOX) with the nanocluster polyoxometalate (POM) {Mo72Fe30}, the latter serving as the drug carrier. The formation of a stoichiometric complex POM@(DOX)12 was observed in aqueous media. Using the Stern‐Volmer technique, we found the binding constant and free Gibbs energy for this interaction. The produced complex POM@(DOX)12 was examined with ultraviolet‐visible (UV‐Vis), fluorescence (steady state and life‐time), infrared (IR) and X‐ray photoelectron spectroscopy (XPS), which revealed the stoichiometry of complex and a key role of {Mo3Fe3} host pores on the POM surface to DOX binding. DOX's ability to intercalate into DNA after binding with the {Mo72Fe30} was investigated using a pH typical for blood (7.4). A pH‐dependent release of DOX from POM@(DOX)12 was demonstrated (rate constant is 3.3×10−4 s−1).
