Vitamin D3 (Vit D3) and 25(OH)D3 are used as dietary sources of active vitamin D (1,25(OH)2D3) in pig husbandry. Although acting primarily on intestine, kidney and bone, their use in pig nutrition has shown a wide range of effects also in peripheral tissues. However, there is an ambiguity in the existing literature about whether the effects of Vit D3 and 25(OH)D3 differ in attributing the molecular and phenotypic outcomes in pigs. We searched Web of Science and PubMed databases concerning the efficacy of Vit D3 in comparison with 25(OH)D3 on pig physiology, i.e. reproductive capacities, growth performance, immunity and bone development. Dietary intake of Vit D3 or 25(OH)D3 did not influence the reproductive capacity of sows. Unlike Vit D3, the maternal intake of 25(OH)D3 significantly improved the growth performance of piglets, which might be attributed to maternally induced micronutrient efficiency. Consequently, even in the absence of maternal vitamin D supplementation, 25(OH)D3-fed offspring also demonstrated better growth than the offspring received Vit D3. Moreover, a similar superior impact of 25(OH)D3 was seen with respect to serum markers of innate and humoral immunity. Last but not least, supplements containing 25(OH)D3 were found to be more effective than Vit D3 to improve bone mineralisation and formation, especially in pigs receiving basal diets low in Ca and phosphorus. The insights are of particular value in determining the principal dietary source of vitamin D to achieve its optimum utilisation efficiency, nutritional benefits and therapeutic potency and to further improve animal welfare across different management types. The neonatal increase in circulating luteinizing hormone (LH) is crucial for testicular development. In male pigs, blood LH levels start to increase approximately 1 week after birth and return to basal level by 5-6 weeks of age. This study tested the hypothesis that neonatal treatment with a combination of estrogens and androgens suppresses LH secretion and thereby inhibits testicular development. On Day 1 after birth, piglets received a slow-release implant containing estradiol (E 2 , 8-40 mg) and trenbolone acetate (TBA, 40-200 mg) or remained intact. At 4 weeks of age, mean serum LH concentrations were ∼ 7 ng/mL in untreated males, whereas pigs with implants had serum LH concentrations < 1 ng/mL. Despite this reduction, LH was still detected in the pituitary glands of treated pigs. Interestingly, neonatal castration also lowered circulating LH, highlighting the importance of testis physiology in the early establishment of the reproductive axis. The higher dose (20 mg E2 + 100 mg TBA) inhibited testis function more effectively, as evidenced by lower circulating testosterone concentrations compared to intact pigs. Furthermore, E2 + TBA treatment had a lasting impact on testicular growth, resulting in smaller testes at 26 weeks of age and the presence of immature Leydig cells. Pig production, a vital sector of the meat industry, faces demands for improved quality, efficiency, and sustainability. Advancements in breeding, disease control, and artificial insemination have enhanced production, while biotechnologies such as in vitro embryo production (IVP) and genetic engineering offer further progress. In vitro embryo production could facilitate the global exchange of valuable genetic material, accelerate breeding programs, and improve productivity, and it is essential for generating genetically modified (GM) pigs. These GM pigs have two main applications: first, they allow for targeted modifications aimed at improving production traits relevant to pig production in agriculture, such as meat quality and disease resistance. Second, they serve as valuable biomedical models for human disease research, regenerative medicine, and organ transplantation. Yet, despite notable advancements in recent decades, the efficiency of the current IVP systems for porcine embryos remains a challenge. Compared to the in vivo environment, suboptimal culture conditions lead to issues such as elevated polyspermy, poor embryo development, and the production of low-quality blastocysts. This review provides an overview of the key steps and recent advancements in porcine IVP technology. We will emphasize the promising utilization of oocytes from live females of high genetic value through ovum pick-up and the incorporation of extracellular vesicles and cytokines into IVP media. We performed a plateau-linear reaction norm model (RNM) analysis of number born alive (NBA) in purebred Landrace pigs, where breeding value changes according to maximum temperature at mating day, using public meteorological observation data in Japan. We analysed 52,668 NBA records obtained from 10,320 Landrace sows. Pedigree data contained 99,201 animals. Off-farm daily temperature data at the nearest weather station from each of the farms were downloaded from the Japan Meteorological Agency website. A plateau-linear RNM analysis based on daily maximum temperature on mating day (threshold temperature of 16.6°C) was performed. The percentage of the records with daily maximum temperatures at mating days of ≤16.6, ≥25.0, ≥30.0 and ≥35.0°C were 34.3%, 33.6%, 14.0% and 0.8%, respectively. The value of Akaike's information criterion for the plateau-linear RNM was lower than that for a simple repeatability model (RM). With the plateau-linear RNM, estimated value of heritability ranged from 0.14 to 0.15, while that from the RM analysis was 0.15. Additive genetic correlation between intercept and slope terms was estimated to be -0.52 from the plateau-linear RNM analysis. Estimated additive genetic correlations were >0.9 between NBA at different temperatures ranging from 16.6 to 37.6°C. For the 10,320 sows, average values of prediction reliability of the intercept and slope terms for breeding values in the plateau-linear RNM were 0.47 and 0.16, respectively. Increasing weight for slope term in linear selection index could bring positive genetic gain in the slope part, but prediction accuracy would decrease. Our results imply that genetically improving heat tolerance in sows reared in Japan focusing on NBA using RNM is possible, while RNM is more complex to implement and interpret. Maternal nutrition is one of the main environmental factors regulating gene expression during fetal development through epigenetic modifications. Some nutrients, such as the amino acid l-arginine, are added to maternal diets to modulate gene expression, improve the reproductive performance of females, and enhance conceptus development. This study investigated the hypothesis that supplementation of pregnant gilts with l-arginine regulates gene expression in conceptuses through epigenetic mechanisms. For this, fetal programming phenotypic markers, the expression of key epigenetic genes, and the abundance of DNA methylation proteins (DNMT3A and DNMT1) were evaluated in 25- and 35-day conceptuses from gilts supplemented (ARG) or not (CON) with 1.0 % l-arginine during early gestation. At 25 days, there were no significant differences in phenotypic markers between CON and ARG embryos (P > 0.05). Similarly, no differences were found between CON and ARG fetuses at 35 days (P > 0.05). Maternal supplementation with l-arginine did not influence the expression of the evaluated key epigenetic genes in pig embryos or fetuses, nor DNMT3A protein abundance (P > 0.05); on the other hand, DNMT1 protein abundance was lower in ARG fetuses (P = 0.002). It is concluded that supplementation of l-arginine in pregnant gilts affects epigenetic mechanisms, such as DNA methylation, in 35-day fetuses through regulation of DNMT1 levels. The premise was tested that the additional genetic gain was achieved in the overall breeding objective in a pig breeding program using genomic selection (GS) compared to a conventional breeding program, however, some traits achieved larger gain than other traits. GS scenarios based on different reference population sizes were evaluated. The scenarios were compared using a deterministic simulation model to predict genetic gain in scenarios with and without using genomic information as an additional information source. All scenarios were compared based on selection accuracy and predicted genetic gain per round of selection for objective traits in both sire and dam lines. The results showed that GS scenarios increased overall response in the breeding objectives by 9% to 56% and 3.5% to 27% in the dam and sire lines, respectively. The difference in response resulted from differences in the size of the reference population. Although all traits achieved higher selection accuracy in GS, traits with limited phenotypic information at the time of selection or with low heritability, such as sow longevity, number of piglets born alive, pre- and post-weaning survival, as well as meat and carcass quality traits achieved the largest additional response. This additional response came at the expense of smaller responses for traits that are easy to measure, such as back fat and average daily gain in GS compared to the conventional breeding program. Sow longevity and drip loss percentage did not change in a favourable direction in GS with a reference population of 500 pigs. With a reference population of 1000 pigs or onwards, sow longevity and drip loss percentage began to change in a favourable direction. The present experiment was carried out to investigate the role of Oxyrase in preserving the in vitro quality, redox status and in vivo fertility of crossbred boar spermatozoa. A total of 24 ejaculates from 6 crossbred (n = 4 from each boar) boars were collected and extended in Beltsville Thawing Solution (BTS) in 1:2 ratio and divided into three aliquots. The first aliquot served as a control (without Oxyrase). Rest of the two aliquots were supplemented with 0.125 (T1) and 0.25 IU/mL Oxyrase (T2). Semen samples were preserved at 15°C for 5 days and kinematics of spermatozoa by CASA, semen quality parameters and oxidative stress status were evaluated at 0, 72 and 120 h of storage. The findings of studies revealed that supplementation of Oxyrase at 0.25 IU/mL resulted in higher (p < 0.05) total motility, progressive motility, plasma membrane integrity, acrosome integrity and functional integrity of plasma membrane at 72 and 120 h in comparison to the control group. Mitochondrial membrane potential (MMP) was higher (p < 0.05) at 72 and 120 h, whereas higher (p < 0.05) DNA integrity was observed at 120 h in T2. The lipid peroxidation (LPO) was lower (p < 0.05) and superoxide dismutase activity (SOD) and total antioxidant capacity (TAC) were higher (p < 0.05) in the T2 group at 120 h as compared to control. In vivo fertility trials indicated a higher (p < 0.05) litter size in T2 in comparison to other groups. The number of piglets born alive is frequently greater than the functional teats, and some farms are equalizing litters with surplus piglets, limiting piglet udder access. Removing the barrier among farrowing crates may allow piglets to socialize and seek other sows. Thus, we evaluated the effects litter size at cross-fostering and socialization on the sow and piglet performance during lactation. Two factors were included in a 2 × 2 factorial design: socialization and litter size relative to sow functional teat number at cross-fostering. Litters (n = 189) were then assigned to one of four groups: CONT+0 (litters not co-mingled with no additional piglet), CONT+1 (not co-mingled with one additional piglet), Co-M+0 (co-mingled with no additional piglet), Co-M+1 (co-mingled with one additional piglet). Piglets were cross-fostered from 12-16 h after birth, and 24 h later, Co-M groups were socialized by removing the barrier between two adjacent pens. During lactation, sows lost more caliper units in the + 1 group than in the + 0 group (P = 0.04). The number of functional teats did not differ among groups, but Co-M sows had a higher udder lesion score at weaning than CONT (6.55 vs 4.83; P < 0.01). Furthermore, +1 sows had fewer vacant teats throughout lactation (P < 0.01). Milk yield did not differ among the groups (P ≥ 0.13). Regarding the number of weaned piglets, no difference was observed for socialization (P = 0.84), but + 1 sows weaned 0.67 more piglets than + 0 (P < 0.01). Although CONT+0 had the heaviest piglets at weaning (P < 0.01), litter weight did not differ among the groups (P ≥ 0.08). Facial and joint lesions were frequently observed in Co-M (P < 0.01) than CONT. Piglet loss rate did not differ among treatments (overall rate = 12.6%; P ≥ 0.26). The removal rate, however, was more frequent in + 1 litters than in  + 0 (P < 0.01). Death due to starvation was higher in CONT+1 than CONT+0 (P < 0.01) but did not differ between the Co-M groups (P = 0.99). Litters formed with one additional piglet relative to functional teat number weaned more piglets, albeit with lower individual weight. Rooting is a strongly motivated, species-specific behaviour of pigs. Most housing systems do not provide appropriate materials that enable the full expression of this behaviour, and it remains unclear whether straw is suitable to entirely fulfil the rooting motivation of pigs. We therefore investigated the suitability of small (minimal) and large (deep) amounts of straw as well as large amounts of compost to satisfy rooting motivation in pigs. Fifty-seven growing-finishing pigs were housed in three pens, each providing permanent access to one of the three treatment substrates. Eight pigs per group were tested individually in a classical preference test (PT) and another eight pigs in a conditioned place preference test (CPPT). In the tests, pigs could show their preference to consume freely available feed ("feed") or feed hidden in sawdust ("root"). In the CPPT, feed was only present during training but not during testing. Pigs were exposed to the test situation twice, with approximately 72 kg and 115 kg BW. In both tests, the following variables were measured and used as outcome variables in linear mixed effect models: first decision to choose one of the two stimuli ("feed" or "root"), duration of time spent in proximity to "root", number of changes between stimuli, and latency to the first decision. Overall, the pigs' first decision (by tendency; P = 0.076) and the duration in proximity to "root" (P = 0.034) varied among treatments: Pigs housed with minimal straw tended to be more likely to choose "root" first (posthoc comparison; P = 0.090) and spent more time in proximity to "root" (P = 0.030) than pigs housed with compost, whereas pigs housed with deep straw were intermediate. Interestingly, the patterns of response to the treatment differed depending on the behavioural tests for both, first decision (interaction; P = 0.032) and duration in proximity to "root" (interaction; by tendency; P = 0.006). In addition, pigs in the PT changed more often between stimuli than pigs in the CPPT (P < 0.001). There was a tendency for an interactive effect between test and treatment for latency to first decision (interaction; P = 0.082), though pairwise comparisons did not reveal any differences. The main principles of organic farming as presented by the European organisation for organic food and farming are health, ecology, fairness and care, but the intrinsic quality of products is also important for consumers. Pig genotype was tested as a lever to improve animal welfare and pork quality (meat tenderness, processing ability) of organic, non-castrated males while controlling the risk for boar taint. Non-castrated Large White × Duroc (D, n = 47) or Large White × Pietrain NN (P, n = 34) males were involved in two batches, each including one group of pigs per genotype. Each group was reared in a pen from the same building on deep straw bedding (1.3 m 2 /pig), with a feeding zone (0.2 m 2 /pig) and an outdoor area (1.0 m 2 /pig), from 28 kg BW until slaughter at ca. 125 kg BW. All pigs received ad libitum the same growing and finishing diets, and hay. Overall, health and welfare indicators showed few problems, but the proportions of pigs with skin scratches, and tail lesions at the end of the finishing period, were lower in D than in P pigs (P < 0.05). Growth rate and final BW did not differ between genotypes. The D pigs had lower carcass lean meat content (P < 0.001) and relative proportions of ham and loin (P ≤ 0.01), and higher proportions of belly and backfat (P ≤ 0.001) than P pigs. Compared to P, loin (Longissimus muscle) of D pigs was less light and exudative and had higher chroma (P < 0.05), but pH 24 h and glycolytic potential did not differ. Loin meat of D pigs had higher intramuscular fat content (P < 0.001) and tended to have a lower shear force (P = 0.09), but cooking loss did not differ. In the ham muscles, D pigs had higher chroma than P pigs in the Gluteus medius, whereas pH 24 h did not differ in the Gluteus medius and Semimembranosus. D pigs had higher backfat concentrations of androstenone (P < 0.001), and skatole and indole (P < 0.05) than P pigs, suggesting a higher risk of rejection by consumers due to boar taint. However, only one D carcass was detected as tainted by human nose test. Altogether, organic farming of non-castrated Duroc crossbred males appears to be favourable for animal welfare, technological and several sensory pork properties provided that the risk of undesirable odours is limited through management practices., (Copyright © 2024 The Author(s). While several countries impose minimum light requirements for pig housing, it remains unknown whether these requirements are beneficial for pig welfare. Therefore, we aim to review the current knowledge on the effects of light on pig welfare. In this paper, we explain concepts defining light, discuss the relevance of vision for pigs and systematically review the effects of light on pig welfare. Systematic literature searches were performed in two databases to find studies about light and welfare-related topics, including behaviour, health, hormonal secretions and productivity. After screening, 63 studies were reviewed. According to literature, light is relevant in pigs' lives as they are diurnal animals and use vision in combination with other senses to, for example, locate food and interact with conspecifics. Throughout this paper, the investigated light parameters are photoperiod, intensity and spectrum. Pigs seem to have p for a certain light intensity and spectrum, but these preferences vary over production phases. Photoperiod influences feed intake and growth, especially in piglets, but no conclusion can be drawn because of contradictory results. Furthermore, pigs' activity patterns adapt to the provided light schedule and show a diurnal rhythm with higher activity during lit hours. Photoperiod also plays a role in the diurnal secretion of hormones. Cortisol secretion increases shortly before the moment of light onset, and melatonin secretion is influenced by the light and dark contrast with a nocturnal rise after light offset. Some behaviours are impacted by light intensity; for instance, dim conditions are associated with resting and bright conditions with elimination behaviour. Moreover, a few studies showed that in dimmer conditions, more negative social interactions occur, while brighter conditions lead to more positive interactions. Lastly, even though light spectrum is the least explored light parameter, several studies showed that UV B light can activate the cutaneous synthesis of vitamin D 3 . A limitation in the current literature is that several studies tested light treatments differing in more than one light parameter, making the interpretation of each light parameter difficult. Moreover, most studies do not provide information on other light parameters not targeted by the study, particularly on light spectrum. This review aimed to enlighten aspects of welfare from the farm animal-centred point of view rarely addressed such as those anatomical and physiological alterations induced in farm animals to obtain high performance. Hence, the major working hypothesis was that high-producing farm animals developed an imbalance between body structural and functional capacities and the genetic procedures applied to obtain industrial production of animal protein. This is called "disproportionality", a feature which cannot be compensated by feeding and management approaches. Consequences of disproportionality are the insidious development of disturbances of the metabolism, low-grade systemic inflammation and as a final stage, production diseases, developing throughout the productive life span of a farm animal and affecting animal welfare. Based on scientific evidence from literature, the review discusses disproportional conditions in broilers, laying hens, sows, piglets, dairy cows, bulls and calves as the most important farm animals for production of milk, meat, foetuses and eggs. As a conclusion, farm animal welfare must consider analysing issues from an animal-centered point of view because it seems evident that, due to genetics and management pressures, most of farm animals are already beyond their physiological limitations. Animal welfare from an animal-centered point must be addressed as an ethical step to establish limits to the strength placed on the animal's anatomical and physiological functionality. Efficient management of pig reproduction is paramount for the sustainability and productivity of the global pork industry. Modern artificial insemination (AI) breeding programs have greatly benefited from the integration of advanced selection methods and biomarkers to enhance the reproductive performance of boars. While traditional selection methods have relied soley on boar phenotype, such as growth rate and conformation, modern pig breeding has shifted more and more toward molecular and genetic tools, which are still complemented by phenotypic traits. These methods encompass genomics, transcriptomics, and proteomics. Biomarkers serve as critical indicators of boar reproductive capacity. They can help to identify individuals with superior fertility and aid in the early identification of potential fertility issues, allowing for proactive management strategies. This review summarizes current knowledge of various biomarkers associated with semen quality, sperm function, and overall reproductive fitness in boars. Furthermore, we explore advanced technologies and their potential applications in uncovering novel selection methods and biomarkers for predicting boar fertility. The optimization of processes associated with artificial insemination (AI) is of great importance for the success of the pig industry. Over the last two decades, great reproductive performance has been achieved, making further significant progress limited. Optimizing the AI program, however, is essential to the pig industry's sustainability. Thus, the aim is not only to reduce the number of sperm cells used per estrous sow but also to improve some practical management in sow farms and boar studs to transform the high reproductive performance to a more efficient program. As productivity is mainly influenced by the number of inseminated sows, guaranteeing a constant breeding group and with healthy animals is paramount. In the AI studs, all management must ensure conditions to the health of the boars. Some strategies have been proposed and discussed to achieve these targets. A constant flow of high-quality and well-managed breeding groups, quality control of semen doses produced, more reliable technology in the laboratory routine, removal of less fertile boars, the use of intrauterine AI, the use of a single AI with control of estrus and ovulation (fixed-time AI), estrus detection based on artificial intelligence technologies, and optimization regarding the use of semen doses from high genetic-indexed boars are some strategies in which improvement is sought. In addition to these new approaches, we must revisit the processes used in boar studs, semen delivery network, and sow farm management for a more efficient AI program. Livestock management is evolving into a new era, characterized by the analysis of vast quantities of data (Big Data) collected from both traditional breeding methods and new technologies such as sensors, automated monitoring system, and advanced analytics. Artificial intelligence (A-In), which refers to the capability of machines to mimic human intelligence, including subfields like machine learning and deep learning, is playing a pivotal role in this transformation. A wide array of A-In techniques, successfully employed in various industrial and scientific contexts, are now being integrated into mainstream livestock management practices. In the case of swine breeding, while traditional methods have yielded considerable success, the increasing amount of information requires the adoption of new technologies such as A-In to drive productivity, enhance animal welfare, and reduce environmental impact. Current findings suggest that these techniques have the potential to match or exceed the performance of traditional methods, often being more scalable in terms of efficiency and sustainability within the breeding industry. Climate change has been linked to increasing temperatures and weather extremes. Certain regions around the world become more susceptible to environmental hazards that limit pig production and reproductive fertility. Environmental measures that link to pig fertility are needed to assess change, risk and develop solutions. Sub-populations of pigs display lower fertility in summer and are susceptible to heat stress. In the context of a warming climate, elevated temperatures and number of heat stress days increase body temperature and change the physiology, behavior, feed intake, and stress response of the pig. These changes could alter follicle development, oocyte quality, estrus expression, conception and litter size. In boars, sperm quality and production are reduced in response to summer heat stress. Nevertheless, while temperature increases have occurred over the years in some warmer locations, other regions have not shown those changes. Perhaps this involves the measures used for heat stress assessment or that climate is buffered in more temperate areas. Reductions in pig fertility are not always evident, and depend upon climate, year, genotype and management. This could also involve selection, as females more susceptible to heat stress and fertility failure, are subsequently culled. In the years from 1999 to 2020 when increases in global temperature from baseline occurred, measures of female fertility improved for farrowing rate and litter size. Despite decades of research and handling of semen for use in artificial insemination (AI) and other assisted reproductive technologies, 5-10% of selected boar sires are still considered sub-fertile, escaping current assessment methods for sperm quality and resilience to preservation. As end-product, the ejaculate (emitted spermatozoa sequentially exposed to the composite seminal plasma, the SP) ought to define the homeostasis of the testes, the epididymis, and the accessory sexual glands. Yet, linking findings in the ejaculate to sperm production biology and fertility is suboptimal. The present essay critically reviews how the ejaculate of a fertile boar can help us to diagnose both reproductive health and resilience to semen handling, focusing on methods -available and under development- to identify suitable biomarkers for cryotolerance and fertility. Bulk SP, semen proteins and microRNAs (miRNAs) have, albeit linked to sperm function and fertility after AI, failed to enhance reproductive outcomes at commercial level, perhaps for just being components of a complex functional pathway. Hence, focus is now on the interaction sperm-SP, comparing in vivo with ex vivo, and regarding nano-sized lipid bilayer seminal extracellular vesicles (sEVs) as priority. sEVs transport fragile molecules (lipids, proteins, nucleic acids) which, shielded from degradation, mediate cell-to-cell communication with spermatozoa and the female internal genital tract. Biosecurity is a major concern in the global pig production. The separation in time of semen collection, processing and insemination in the pig farm is a few days for chilled semen but it can be indefinite when using cryopreserved semen. Field fertility results of boar cryopreserved semen are close to chilled semen, which makes it a valuable resource for the establishment of semen genebanks, long-distance semen trade, and the implementation of other technologies such as the sex-sorted semen. But cryopreserved semen is far from being routine in pig farms. The most recent research efforts to facilitate its implementation include the use of additives before freezing, or in the thawing extender. Long-term preserved semen trade is a biosecurity challenge. To harmonize international trade of germplasm, the World Organization of Animal Health (WOAH) established a regulatory framework for all member countries. The present paper aims to review the latest advances of boar semen cryopreservation with special focus on the benefits of its inclusion as a routine tool in the pig industry. We also review recently reported field fertility results of cryopreserved semen, its international trade compared to chilled semen, and the regulatory framework involved. Boar cryopreserved semen is a valuable tool to control biosecurity risk, implement other technologies, and facilitate international trade. Research already demonstrated good field fertility results, but it still represents less than 0.1 % of the international trade. The last decades of research have revealed that many other factors besides gamete genomes are able to determine the reproductive outcomes. Indeed, paternal factors have been observed to be capable of modulating multiple crucial features of the reproductive process, such as sperm physiology, the maternal environment and, even, the offspring health. These recent advances have been encompassed with the emergence of OMICS technologies, as they comprehensively characterise the molecular composition of biological systems. The present narrative review aimed to take a closer look at the potential of these technologies in the field of reproductive biology. This literature revision shows that most studies up to date have followed a non-targeted approach to screen mammalian seminal plasma (SP) and sperm metabolite composition through different metabolome platforms. These studies have proposed metabolites of multiple natures as potential in vivo fertility biomarkers. Yet, targeted approaches can be used to answer specific biological question, and their power is exemplified herein. For instance, metabolomic studies have uncovered not only that glycolysis is the main ATP energy source of pig sperm, but also that sperm metabolism can trigger DNA damage, hence compromise embryo development. In conclusion, this review shows the potential of both non-targeted and targeted metabolomics for the discovery of cell pathways that govern the reproductive process. The importance of boar reproductive traits, including semen quality, in the sustainability of pig production system is increasingly being acknowledged by academic and industrial sectors. Research is needed to understand the biology and genetic components underlying these traits so that they can be incorporated into selection schemes and managerial decisions. This article reviews our current understanding of genome biology and technologies for genome, transcriptome and epigenome analysis which now facilitate the identification of causal variants affecting phenotypes more than ever before. Genetic and transcriptomic analysis of candidate genes, Genome-Wide Association Studies, expression microarrays, RNA-Seq of coding and noncoding genes and epigenomic evaluations have been conducted to profile the molecular makeups of pig sperm. These studies have provided insightful information for a several semen-related parameters. Nonetheless, this research is still incipient. The spermatozoon harbors a reduced transcriptome and highly modified epigenome, and it is assumed to be transcriptionally silent for nuclear gene expression. For this reason, the extent to which the sperm's RNA and epigenome recapitulate sperm biology and function is unclear. Hence, we anticipate that single-cell level analyses of the testicle and other male reproductive organs, which can reveal active transcription and epigenomic profiles in cells influencing sperm quality, will gain popularity and markedly advance our understanding of sperm-related traits. Cryopreservation is the most efficient procedure for long-term preservation of mammalian sperm; however, its use is not currently dominant for boar sperm before its use for artificial insemination. In fact, freezing and thawing have an extensive detrimental effect on sperm function and lead to impaired fertility. The present work summarises the basis of the structural and functional impact of cryopreservation on pig sperm that have been extensively studied in recent decades, as well as the molecular alterations in sperm that are related to this damage. The wide variety of mechanisms underlying the consequences of alterations in expression levels and structural modifications of sperm proteins with diverse functions is detailed. Moreover, the use of cryotolerance biomarkers as predictors of the potential resilience of a sperm sample to the cryopreservation process is also discussed. Regarding the proteins that have been identified to be relevant during the cryopreservation process, they are classified according to the functions they carry out in sperm, including antioxidant function, plasma membrane protection, sperm motility regulation, chromatin structure, metabolism and mitochondrial function, heat-shock response, premature capacitation and sperm-oocyte binding and fusion. Special reference is made to the relevance of sperm membrane channels, as their function is crucial for boar sperm to withstand osmotic shock during cryopreservation. In pig production, the optimization of artificial insemination (AI) efficiency significantly relies on the accurate assessment of semen quality and fertility of boars. Traditional methods such as conventional seminogram techniques, although long-standing, exhibit limited sensitivity in predicting boar fertility, warranting the exploration of novel molecular markers. This review synthesizes the current knowledge on the utilization of molecular markers for semen quality evaluation and male fertility prediction in boars, providing an in-depth examination of molecular markers in this context. Specifically, the present work delves into the potential of OMICs technologies, encompassing genetic and genomic approaches, transcriptomics, proteomics, and metabolomics. A diverse array of molecular markers, including genomic regions associated with sperm quality and male fertility, chromatin integrity, mitochondrial DNA content, mRNA and non-coding RNA signatures, as well as proteins and metabolites in sperm and seminal plasma, are identified as promising molecular markers for fertility prediction in boars. Furthermore, the need of validating biomarkers and their practical implementation in AI centres is here emphasized. Addressing these considerations and integrating molecular markers within the swine breeding field holds the potential to enhance reproductive management practices and optimize productivity in boar breeding programs. Feeding of breeding boars in artificial insemination (AI) centers is critical to maintaining and improving breeding quality and performance in agriculture. Modern feeding strategies for AI boars are aiming towards maximizing their lifetime semen dose output. Given the high growth potential of modern swine genetics, AI boars should be controlled in their daily gain to reduce stress factors for the locomotion system, final body weights, and improve their survivability but also the ease of handling boars. The feeding program should be designed in such a way that young boars (up to 7 months of age) are limited to a daily gain of 400-600 g. Mature boars should be fed towards a body condition score of '2'. Aside from energy intake, protein sources should provide 0.62% SID lysine. As far as minerals and vitamins, special attention should be given to Calcium and Phosphorus as they play a crucial role in bone mineralization. A standardized total tract digestible Calcium-Phosphorus ratio between 1.75:1 and 1.82:1 seems to be most favorable. While certain nutritional requirements are needed to enable the production of ejaculates eligible for AI, considerations should be given to the mitigation of risk factors like mycotoxins, herbicides, and pesticides. The objective of this study was to investigate how the cumulative expulsion interval, litter size, and birth order influence the characteristics of newborn piglets from hyperprolific sows, as well as their intake of colostrum. Analysis was conducted on the temporal aspects of parturition in 254 sows, including 3920 piglets. Newborn piglet characteristics examined encompassed delivery time, birth order, birth status, umbilical cord rupture, meconium staining, delivery ease, birth weight, blood oxygen saturation, and heart rate. From the raw data, colostrum intake, birth interval, and cumulative birth interval of each piglet were computed. Piglets were sorted based on the cumulative expulsion interval into nine categories: 0-30, 31-60, 61-90, 91-120, 121-150, 151-180, 181-210, 211-240, and >240 min. Litter sizes were categorized into three groups: 10-13, 14-17, and ≥18 piglets per litter. The birth-order ranking referred to the piglet's position within a litter and was categorized into four quartiles: 1, 2, 3, and 4. On average, the expulsion interval, cumulative expulsion interval, and colostrum consumption were 14.8 ± 21.8 min, 138.6 ± 155.7 min, and 407.6 ± 162.1 g, respectively. In litters with ≥18 piglets, the percentage of piglets with inadequate colostrum intake (<300 g) was higher compared to litters with 10-13 piglets (33.1 % vs. 12.9 %, P < 0.001) and 14-17 piglets (23.9 %, P < 0.001). The occurrence of stillbirths in the fourth quartile of the litter was higher than in the first (16.0 % vs. 2.2 %, P < 0.001), second (4.0 %, P < 0.001), and third (7.8 %, P < 0.001) quartiles. Among litters with ≥18 piglets, those born in the third quartile exhibited lower colostrum consumption compared to those born in the first quartile (346.0 ± 9.9 vs. 391.4 ± 8.6 g, P = 0.028). Colostrum intake decreased with longer cumulative expulsion intervals, evident from a notable decline starting at 90 min post-farrowing compared to piglets born within the first 30 min (P = 0.004). Interestingly, colostrum consumption exhibited a negative correlation with the cumulative expulsion interval (P < 0.001), but not with individual piglet expulsion intervals (P = 0.460). Piglets born with meconium staining were most prevalent among those with a cumulative expulsion interval of >240 min. In summary, both the cumulative expulsion interval and birth-order ranking significantly impact the physiological characteristics of newborn piglets and their colostrum intake. Colostrum consumption decreases and newborn piglet characteristics change with higher cumulative expulsion intervals. The recognition of animals as sentient beings has raised societal awareness of the welfare of farmed animals. This has been instrumental in increasing the legislative pressure towards welfare-conscious farming practices and, more lately, greater consumer demand for ethically sourced products. Besides, improved animal welfare is a key driver for enhanced performance, particularly in breeding boars, as welfare conditions strongly influence reproductive outcomes. The stressors associated with confinement impact testicular physiology and semen quality and the efforts to improve the welfare of breeding boars have so far focused on mitigating the negative consequences associated with stressors. The Five Freedoms framework and the European Food Safety Authority (EFSA)'s 'welfare consequences' approach prioritise the alleviation of suffering. In breeding boars, key welfare negative consequences include movement restriction, isolation stress, inability to engage in exploratory behaviours, locomotory disorders, and prolonged hunger. The negative consequences, which can be mitigated by improved housing and management practices, should be tackled in any commitment to improve the welfare of breeding boars. Animal welfare science, however, has recently shifted towards cultivating positive welfare experiences and a life worth living, beyond just alleviating suffering. The Five Domain Model systematically evaluates animal welfare, considering both negative and positive aspects. Encouraging positive welfare states involves facilitating species-specific behaviours, such as exploration and play, and fostering positive human-animal interactions. In breeding boars, strategies promoting positive welfare include providing enriching environments, encouraging exploration, and cultivating positive interactions with caretakers. Single Layer Centrifugation (SLC) through a low density colloid offers an alternative solution to antibiotic use in boar semen extenders, with lower costs compared to high density colloids. The aim of this study was to explore the reproductive performance of sows when using SLC-prepared semen doses without antibiotics, employing low density Porcicoll to prepare semen doses for artificial insemination in a commercial swine herd in Thailand. Ejaculates were divided into two equal parts to create insemination doses, with each dose containing 3000 × 10 6 sperm/80 ml for intra-uterine insemination in individual sows. The sows were inseminated twice, with the interval between the two inseminations ranging from 8 to 16 h. The CONTROL group consisted of 206 semen doses treated with antibiotics, prepared for insemination in 103 sows, while the SLC group comprised 194 SLC-prepared semen doses without antibiotics for inseminating 97 sows. Fertility and fecundity traits, including non-return rate, conception rate, farrowing rate, and litter traits (i.e., the total number of piglets born per litter, number of piglets born alive per litter, number of stillborn piglets, and number of mummified fetuses), were compared between groups. Furthermore, data on piglet characteristics, including live-born and stillborn piglets (i.e., the prevalence of stillbirth (yes, no), birth weight, crown-rump length, body mass index (BMI), and ponderal index (PI)), were determined. No significant differences in non-return rate (75.7 % vs. 77.3 %), conception rate (73.8 % vs. 73.2 %), and farrowing rate (71.8 % vs. 73.2 %) were observed between the CONTROL and SLC groups, respectively (P > 0.05). Nevertheless, the total number of piglets born per litter in the SLC group was higher than in the CONTROL group (14.6 ± 0.9 vs. 12.3 ± 0.6, respectively, P = 0.049). Interestingly, the prevalence of stillbirth in the SLC group was lower than in the CONTROL group (6.2 % vs. 11.6 %, respectively, P < 0.001). Moreover, the newborn piglets in the SLC group exhibited higher birth weight and BMI compared to those in the CONTROL group (1.36 ± 0.03 vs. 1.26 ± 0.02 kg, P = 0.005, and 18.3 ± 0.3 vs. 17.3 ± 0.2 kg/m 2 , P = 0.003). In conclusion, employing sperm doses after SLC through a low density colloid in artificial insemination within a commercial breeding operation did not have a detrimental impact on either fertility or fecundity traits but showed potential benefits in increasing the total number of piglets born per litter. Moreover, improvements were observed in the birth weight and body indexes of piglets, and the percentage of stillbirths was reduced. Our findings introduce new possibilities for antibiotic alternatives in semen extenders to reduce the risk of antimicrobial resistance in the swine industry. Melatonin, a hormone synthesized in various tissues, plays a crucial role in modulating sperm characteristics, yet its protective function on boar sperm remains poorly understood. This study aimed to investigate the expression and localization of melatonin-related proteins (AANAT, ASMT, MT1, MT2, and NQO2) in pig tissues, assess the impact of melatonin on pig sperm motility parameters and quality, and elucidate the underlying molecular mechanisms. Our results revealed widespread expression of AANAT, ASMT, MT1, MT2, and NQO2 proteins in pig tissues, particularly in the testis. Specific localization patterns were observed in Leydig cells, reproductive epithelium, and columnar epithelium cells in the testis and cauda epididymis. Additionally, melatonin membrane receptors MT1 and MT2 were detected in boar sperm. Melatonin treatment significantly enhanced boar sperm motility parameters and quality, particularly with 10 nM melatonin treatment. Inhibition of the MT1 receptor, but not the MT2 receptor, resulted in decreased sperm motility, highlighting the pivotal role of the MT1 receptor in mediating melatonin's effects on boar sperm. Metabolomic analysis revealed significant alterations in sperm metabolites following melatonin supplementation, particularly in amino acid metabolism. Decreased oocyte quality is a significant contributor to the decline in female fertility that accompanies aging in mammals. Oocytes rely on mRNA stores to support their survival and integrity during the protracted period of transcriptional dormancy as they await ovulation. However, the changes in mRNA levels and interactions that occur during porcine oocyte maturation and aging remain unclear. In this study, the mRNA expression profiles of porcine oocytes during the GV, MII, and aging (24 h after the MII stage) stages were explored by transcriptome sequencing to identify the key genes and pathways that affect oocyte maturation and postovulatory aging. The results showed that 10,929 genes were coexpressed in porcine oocytes during the GV stage, MII stage, and aging stage. In addition, 3037 genes were expressed only in the GV stage, 535 genes were expressed only in the MII stage, and 120 genes were expressed only in the aging stage. The correlation index between the GV and MII stages (0.535) was markedly lower than that between the MII and aging stages (0.942). A total of 3237 genes, which included 1408 upregulated and 1829 downregulated genes, were differentially expressed during porcine oocyte postovulatory aging (aging stage vs. MII stage). Key functional genes, including ATP2A1, ATP2A3, ATP2B2, NDUFS1, NDUFA2, NDUFAF3, SREBF1, CYP11A1, CYP3A29, GPx4, CCP110, STMN1, SPC25, Sirt2, SYCP3, Fascin1/2, PFN1, Cofilin, Tmod3, FLNA, LRKK2, CHEK1/2, DDB1/2, DDIT4L, and TONSL, and key molecular pathways, such as the calcium signaling pathway, MAPK signaling pathway, TGF-β signaling pathway, PI3K/Akt signaling pathway, FoxO signaling pathway, gap junctions, and thermogenesis, were found in abundance during porcine postovulatory aging. These genes are mainly involved in the regulation of many biological processes, such as oxidative stress, calcium homeostasis, mitochondrial function, and lipid peroxidation, during porcine oocyte postovulatory aging. There are few existing publications that describe transvaginal ultrasound-guided Ovum Pick-Up (OPU) in sows, and the impacts of the procedure for the welfare of the animals are unknown. In this study, we evaluated the effects of OPU, performed following restraint in a claw-trimming chute, on the animal welfare and reproductive health of second parity hybrid sows. The study utilized a generalized randomized block design at a commercial sow pool. We assessed salivary cortisol levels before, during, and after the procedure to compare the physiological stress response between OPU and restraint chute procedures (control group). We found a significant increase in salivary cortisol caused by the physical restraint procedure, and that the salivary cortisol level at the end of the procedure did not differ between OPU and control groups (p = 0.51). Furthermore, we conducted a novel approach-aversion test for sows, designed to assess if a feed reward would motivate the animals to willingly participate in the OPU-procedure. The animals were trained daily to enter the chute to access a feed reward. Ten animals in each group failed to complete the training period and did not voluntarily enter the restraint chute on the experimental day. This indicates that even the short daily restraint procedure during the four-day long training period was aversive to some animals. There was no difference in aversion towards the restraint chute between OPU and control groups one day after the procedure. The reproductive performance of the animals was subsequently evaluated through oestrus synchronization and insemination of the sows after the experiment. There was no observed difference in the farrowing rate (p = 0.72) and total number of born piglets (p = 0.84) between OPU and control sows. On average, we retrieved 9.0 ± 5.9 oocytes during the OPU-sessions (N = 26). Our results show that a majority of the sows prioritize the motivation for feed over their aversion to the OPU procedure. However, the physical restraint procedure is unpleasant for the animals and elicits a temporary stress response. We suggest that transvaginal OPU may be used for the recovery of oocytes from live sows, but refinements are needed to avoid stress during the lifting procedure. Dielectric barrier discharge (DBD) plasma regulates the levels of reactive oxygen species (ROS), which are critical for sperm quality. MicroRNAs (miRNAs) are non-coding single-stranded RNA molecules encoded by endogenous genes, which regulate post-transcriptional gene expression in animals. At present, it is unknown whether DBD plasma can regulate sperm ROS levels through miRNAs. To further understand the regulatory mechanism of DBD plasma on sperm ROS levels, miRNAs in fresh boar spermatozoa were detected using Illumina deep sequencing technology. We found that 25 known miRNAs and 50 novel miRNAs were significantly upregulated, and 14 known miRNAs and 74 novel miRNAs were significantly downregulated in DBD plasma-treated spermatozoa. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis showed that target genes of differentially expressed miRNAs were involved in many activities and pathways associated with antioxidants. We verified that DBD plasma significantly increased boar sperm quality and reduced ROS levels. These results suggest that DBD plasma can improve sperm quality by regulating ROS levels via miRNAs. Reduced piglet feed intake immediately postweaning (pw) leads to disruption of small intestine structure and function and reduced growth. Our objective was to evaluate the effect of providing supplemental milk or liquid starter diet for either 4 or 11 days pw, on intestinal parameters of newly weaned piglets and growth to slaughter. At weaning (28 ± 0.6 days old), five hundred and eighty-seven piglets ((Large White × Landrace) × Duroc) were divided into 59 pen groups, each containing 9-10 same sex (entire male or female) piglets. The pen groups were blocked by sex and weaning weight and provided with ad-libitum access to one of five dietary treatments: (1) Dry pelleted starter diet (control; CON); (2) CON+liquid milk replacer for 4 days pw (M4); (3) CON+liquid milk replacer for 11 days pw (M11); (4) CON+liquid starter diet for 4 days pw (S4) and (5) CON+liquid starter diet for 11 days pw (S11). Pen groups were weighed at weaning, days 11, 20, 28, and 47 pw and at target sale weight. Feed disappearance (on a DM basis) was recorded on each weighing day. On day 7 pw, 10 piglets per treatment were euthanised to collect small intestine tissue samples for determination of villus height (VH), crypt depth and brush-border membrane enzyme activity. Data were analysed using SAS-version 9.4. Between days 0 and 11 pw, M11 increased average daily feed intake by 48% and average daily gain (ADG) by 57% compared to CON (P < 0.05), and increased ADG by 54% (P < 0.05) compared to S4. Piglets on M11 also had improved feed conversion efficiency compared with CON piglets between days 0 and 11 pw. Treatment did not affect growth performance after day 28 pw, or carcass parameters at slaughter. At day 7 pw, M11 piglets had 37% higher jejunal VH than CON piglets (P < 0.05) and S11 piglets had 28% higher ileal VH than S4 piglets (P < 0.05). M11 piglets had up to 150% higher ileal sucrase activity than M4, S4 and S11 piglets (P < 0.05) and 180% higher ileal maltase activity than S4 piglets (P < 0.05). In conclusion, M11 reduced the immediate negative effects of weaning, as it was associated with increased feed intake, growth, brush-border membrane enzyme activity and improved intestinal structure early pw. Objective: To evaluate the agreement between measurements of invasive blood pressure (IBP) and oscillometric blood pressure from the tongue (OBPton) using a multiparameter monitor., Study Design: Unblinded, prospective, experimental study., Animals: A total of 12 female Large White crossbreed pigs., Methods: Pigs undergoing experimental procedures that required arterial cannula placement were recruited. A blood pressure cuff with the closest width to 40% of the circumference of the tongue was placed rostral to the lingual frenulum. Systolic, mean and diastolic IBP and OBPton were measured simultaneously at 5 minute intervals. Agreement between paired measurements was examined using Bland-Altman analysis. Mean bias, precision (standard deviation of mean bias), 95% limits of agreement, correlation coefficients and percentage of measurements within 10 and 20 mmHg of IBP were calculated., Results: The total numbers of paired measurements recorded were 124, 126 and 124 for systolic, mean and diastolic blood pressures, respectively. The mean bias, precision and 95% limits of agreement for systolic OBPton were 11.5, 11.5 (-11.1 to 34.2), for mean OBPton 5.6, 5.7 (-5.7 to 16.8) and for diastolic OBPton 7.6, 10.1 (-12.1 to 27.4) mmHg. Correlation coefficients were greater than 0.9 for mean OBPton only. More than 50% of measurements were within 10 mmHg of IBP and 80% of measurements were within 20 mmHg of IBP for mean and diastolic OBPton only., Conclusions and Clinical Relevance: The tongue as a cuff site for oscillometric blood pressure measurement is a useful site for measuring mean arterial, but not systolic or diastolic blood pressure in anaesthetized Large White crossbreed pigs. The aim of the present work was to gain insight into the metabolism of pigs derived from assisted reproductive technologies during their adulthood. Approximately 4h after feeding, a blood sample was taken from 3.5 year old sows born by artificial insemination (AI group, n = 7) and transfer of in vitro produced embryos (IVP group, n = 11) to determine the physiological concentrations of the main biomarkers of carbohydrates (glucose and lactate), proteins (albumin, creatinine and urea) and lipids (cholesterol and triglycerides). Four weeks later, an oral glucose tolerance test (OGTT; 1.75g glucose/kg body weight) was performed after an overnight fast and 1h of water withdrawal. Blood samples were obtained prior (T = 0 min; fasting conditions) and 15, 30, 45, 60, 90, 120, 150, 180, 210 and 240 min after glucose intake. At each time point, glycemia was measured immediately using glucometer test strips, and serum was collected to determine the above metabolites along with insulin and glucagon. After OGTT, the area under the curve (AUC) between sampling times and homeostasis model assessment of insulin resistance (HOMA) indices were calculated. Under physiological conditions, the concentration of metabolites studied was similar between AI and IVP sows. In both groups, fasting decreased cholesterol and increased triglycerides and urea (P < 0.001). However, creatinine and lactate were similar in both groups under physiological and fasting conditions. The expected increase in albuminemia and decrease in glycaemia after fasting was only observed in IVP sows. OGTT revealed a different glucose curve pattern (monophasic in AI and biphasic in IVP group), a lower mean concentration of cholesterol, glucose, lactate, triglycerides in IVP compared to AI pigs (P < 0.01), and a higher mean concentration of albumin, creatinine and insulin in IVP compared to AI group (P < 0.05). On the contrary, no differences were found between groups for mean serum glucagon and urea levels, nor for glucose homeostasis indices HOMA-IR and HOMA-%B. The AUC differed between groups at several time points with larger AUC for creatinine, and smaller AUC for glucose, glucagon, and triglycerides, in IVP pigs than in AI pigs at 180-210 min (P < 0.05). In conclusion, under physiological conditions the metabolic profile of fully-grown AI and IVP sows is similar and within normal ranges. Extracellular vesicles (EVs) are abundant in reproductive tract fluids and serve as important mediators of paracrine communication, influencing the function of gametes. Sperm undergo development in the male reproductive tract and exert their function within the female reproductive tract, engaging in interactions with various types of EVs present throughout the reproductive system. Previous studies have demonstrated that both male and female reproductive tract EVs can impact sperm function by transferring regulatory cargoes to them. Nevertheless, inconsistencies of previous research regarding the effects of EVs on sperm function, coupled with a lack of investigation into the influence of female reproductive tract EVs on sperm fertilization, have left the true role and underlying mechanisms of reproductive tract EVs on sperm function largely unexplored. Given that pigs represent significant economic livestock and serve as an ideal biomedical model for human diseases, this review aims to provide a comprehensive summary of the current knowledge regarding reproductive tract EVs and their influence on boar sperm function, while highlighting their potential roles. Reproductive success requires considerable energy investment. Research has shown that some adipokines, i.e. the hormones produced in adipose tissue, affect reproductive functions by influencing all structures of the hypothalamic-pituitary-ovarian axis. Apelin is a recently identified member of the adipokine family. To the best of the authors' knowledge, this is the first study to investigate the gene and protein expression of the apelin system (the apelin hormone and the apelin receptor, APJ) in the anterior (AP) and posterior (PP) pituitary lobes of the domestic pig during different phases of the oestrous cycle (days 2 to 3, 10 to 12, 14 to 16, and 17 to 19) and in early pregnancy (days 10 to 11, 12 to 13, 15 to 16, and 27 to 28). It was also assumed that apelin participates in the regulation of luteinizing hormone (LH) and follicle-stimulating hormone (FSH) secretion and influences Akt, MAPK/Erk1/2, and AMPK signalling pathways in the AP during the oestrous cycle. Apelin, APJ mRNAs and proteins were detected in both pituitary lobes. Apelin was identified in gonadotropes, somatotropes, lactotropes, and thyrotropes. The study also revealed that apelin and APJ mRNA/protein levels fluctuate during the oestrous cycle and early gestation. Apelin affects basal, GnRH- and/or insulin-stimulated gonadotropin secretion in some phases of the cycle, as well as the phosphorylation of Akt, MAPK/Erk1/2, and AMPK proteins in AP cells. These findings suggest that apelin may be produced locally in the pituitary and that this gland is receptive to apelin's action. Alternative farrowing systems that have been developed in recent years could have a positive effect on the welfare of sows during farrowing and lactation. Oxytocin measurements in saliva may provide information about positive animal welfare status. The purpose of this study was to evaluate the changes in salivary oxytocin concentrations in sows during the lactation period in three different farrowing systems and in two different seasons. Crossbred Duroc sows (n = 34, average parity = 3.6 ± 1.80) were housed in conventional farrowing crates (FC) (n = 10) or in farrowing pens with temporary crating (TC), including SWAP (n = 12) and JFL15 (n = 12) in two different seasons: summer and winter. Saliva samples were collected for six days during lactation: days 2, 4, 12, 23, 25 (i.e., 1-day post-weaning) and 26 (i.e., 2-day post-weaning) after farrowing. Moreover, behavioral data from sows was recorded on days 2, 4, 12 and 23 after farrowing, using a 30-s scan sampling method for 3 min per pen to record the behaviors which were assessed by the same observer. The results showed that the salivary oxytocin concentrations were 472.5 pg/mL and 399.4 pg/mL higher in both TC (SWAP and JLF15, respectively) than in the FC in early-lactation period, and these differences were more pronounced in summer and at the end of lactation in winter. In terms of behavior, higher number of mother-young interactions were observed in TC than FC in early- and mid-lactation period. Skeleton bones, distinguished by trabecular and cortical bone tissue content, exhibit varied growth and composition, in response to modified dietary calcium and phosphorus levels. The study investigated how gilts adapt their individual bone and bone region mineralisation kinetics in response to changing intake of Ca and P. A total of 24 gilts were fed according to a two-phase (Depletion (D) 60-95 and Repletion (R) 95-140 kg BW, respectively). During the D phase, gilts were fed either 60% (D60) or 100% (D100) of the estimated P requirement. Subsequently, during the R phase, half of the gilts from each D diet were fed either 100% (R100) or 160% (R160) of the estimated P requirement according to a 2 × 2 factorial arrangement. Bone mineral content (BMC) was assessed in the whole body, individual bones (femur and lumbar spine L2-L4), and bone regions (head, front legs, trunk, pelvis, femur, and hind legs) every 2 weeks using dual-energy X-ray absorptiometry (DXA). At 95 kg BW, gilts fed D60 showed reduced BMC and BMC/BW ratio in all studied sites compared to those fed D100 (P < 0.001). During the depletion phase, the allometric BW-dependent regressions slopes for BMC of D100 gilts remained close to 1 for all sites and did not differ from each other. In contrast, the slopes were lower in D60 gilts (P < 0.05), with an 18% reduction in the whole body, except for the front and hind legs, femur, and pelvis, which exhibited higher reductions (P < 0.05). At 140 kg BW, BMC and BMC/BW ratio of all studied sites were similar in gilts previously fed D60 and D100, but higher in R160 than in R100 gilts (P < 0.05), except for front and hind legs. During the repletion phase, the allometric BW dependent regressions slopes for BMC were lower (P < 0.05) in R100 than in R160 gilts (for whole body -10%; P < 0.01) except for front and hind legs, femur, and pelvis. In conclusion, bone demineralisation and recovery followed similar trends for all measured body sites. However, the lumbar spine region was most sensitive whereas the hind legs were least sensitive. This study assessed the effects of dexamethasone treatment on farrowing performance and piglet traits in the first 5 days of life in multiparous sows, a high-risk group for stillbirths and prolonged farrowing. In this study, 185 multiparous sows (parity 4.25 ± 0.14) were selected on the day of farrowing and divided into three treatments: CON - control, without dexamethasone treatment; DexaPF - treatment with dexamethasone (20 mg im per female) at the time of copious colostrum secretion (pre-farrowing); and DexaFO - treatment with dexamethasone (20 mg im per female) when the first piglet was born (farrowing onset). All sows and their litters were monitored for farrowing duration, obstetric interventions, colostrum yield and intake, newborn piglet traits, and piglet performance until 5 d of age. A subsample of 106 females (∼35 per treatment) had their blood glucose concentration checked hourly shortly after the first piglet was born until the end of farrowing. Additionally, blood samples from 42 litters were collected for immunocrit evaluation. The results showed no differences regarding farrowing duration (CON = 258.02 ± 13.81 min; DexaPF = 251.29 ± 13.60 min; DexaFO = 294.92 ± 13.89 min; P = 0.06) and obstetric intervention rates among treatments (CON = 36.58 ± 6.78 %; DexaPF = 42.16 ± 6.89 %; DexaFO = 48.05 ± 7.08 %; P = 0.45). The blood glucose concentration during farrowing was higher in DexaPF (94.56 ± 1.57 mg/dL; P < 0.001) than in CON (73.50 ± 1.72 mg/dL) and DexaFO (87.94 ± 1.80 mg/dL). No differences were observed regarding total piglets born and born alive, stillborn, newborn piglet vitality, colostrum intake, immunocrit, colostrum yield, and glycemia and rectal temperature at 24 h of age (P ≥ 0.13). Regarding meconium staining, higher percentages of piglets born without meconium staining were observed in DexaFO (54.77 ± 5.21 %; P = 0.02) compared with CON (48.58 ± 5.26 %), and no difference was observed for the DexaPF group (53.23 ± 5.21 %). In addition, a higher unbroken umbilical cord rate was observed in DexaFO (92.41 ± 1.31 %; P < 0.01) than the CON or DexaPF (86.91 ± 1.97 % and 89.31 ± 1.67 %, respectively). However, the treatments did not affect piglet performance (weight gain and survival) until 5 d of age (P ≥ 0.15). International interest in loose-housed farrowing is growing and there are ongoing discussions within the European Union (EU) on new legal requirements. However, there is a lack of empirical data on loose-housed farrowing pen sizes and sow dimensions in commercial production. The aim of this study was to map and describe sow size and loose-housing farrowing pen size on commercial piglet-producing farms in Sweden. The study included 146 sows and 51 pen types on 35 medium sized to large Swedish piglet-producing farms (ranging from 106 to 1300 sows in production). Sow length ranged from 129 to 238 cm (mean ± SD 191.3 ± 19.3 cm) and sow height from 74 to 133 cm (86.7 ± 7.7 cm). Floor space occupied by the sow when lying down (length x height) ranged from 1.0 to 3.2 m 2 (1.7 ± 0.3 m 2 ). Pen length ranged from 259 to 415 cm (315.1 ± 24.3 cm), pen width from 188 to 245 cm (207.0 ± 10.7 cm), total pen area from 5.7 to 8.9 m 2 (6.5 ± 0.5 m 2 ), piglet corner area from 0.5 to 1.8 m 2 (1.1 ± 0.4 m 2 ) and area available for the sow (total area - piglet corner area) from 3.9 to 6.4 m 2 (5.4 ± 0.6 m 2 ). These results show that there is substantial variation in sow, pen and piglet corner size on commercial piglet-producing farms in Sweden. This poses a risk of mismatches between sow and pen size (pens too short in relation to sow dimensions), especially for older sows. This study evaluated the efficiency of prostaglandin F2α (PGF) to hasten ovulation in weaned sows. In experiment I, weaned sows detected in estrus (0 h) received: no hormone (Control; n = 56); 0.5 mg PGF IM at 0 h and 2 h (PGF0; n = 56); or 0.5 mg PGF IM at 24 h and 26 h (PGF24; n = 55). In experiment II, weaned sows that did not express estrus signs until 72 h after weaning (0 h) were assigned to: no hormone (Control; n = 45); 10 µg buserelin acetate IM at 0 h (Buserelin; n = 43); 0.5 mg PGF IM at 34 h and 36 h (PGF; n = 44); or 10 µg buserelin acetate IM at 0 h plus 0.5 mg PGF IM at 34 h and 36 h (Buserelin + PGF; n = 45). In experiment I, no effect of PGF on the interval treatment onset to ovulation was observed (P > 0.05), and no treatment effect was observed on the relative or cumulative proportion of females that ovulated post-treatment onset (P > 0.05). In experiment II, treatment onset to ovulation interval was shorter for Buserelin group than for PGF group (P < 0.05), and a higher cumulative percentage of Buserelin treated sows ovulated up to 48 h compared to PGF and Control groups (P < 0.01), with no differences from Buserelin + PGF. Treatments did not affect total number of piglets born in both experiments (P > 0.05). Reducing the dietary crude protein (CP) could effectively reduce pressure on protein ingredient supplies. However, few data have been reported about the extent to which CP can be reduced and whether limiting the use of soybean meal leads to electrolyte imbalance. In this experiment, using the low protein (LP) diet [2% lower than NRC (2012)], seventy-two piglets (35 days old) were randomly divided into 2 groups with 6 replicates of 6 piglets each: CON group (CP = 18.5%) and LP group (CP = 16.5%), to investigate the effect of the LP diet on electrolyte balance, acid-base balance, intestinal structure and amino acid transport in piglets. The results revealed that the LP diet decreased the average daily gain and dietary CP digestibility, and damaged the villi structure of the small intestine. Compared with the CON diet, the potassium content decreased and the chlorine content increased in the LP diet, and similar trends were shown in piglet serum. The arterial pH, pCO 2 , HCO 3 - , and base excess of piglets in the LP group were lower than those in the CON group, while pO 2 was higher than those in the CON group. Interestingly, the LP diet significantly increased the lysine content in piglet serum and significantly decreased the levels of arginine, leucine, and glutamic acid. Furthermore, the LP diet significantly affected the expression of some amino acid transport vectors (B 0 AT1, EAAC1, and y + LAT1). In summary, these findings suggested that the LP diet leads to acid-base imbalance, amino acid transport disorder and amino acids imbalance in piglets, and the dietary electrolyte may be a key factor in the impact of the LP diet on piglet growth performance and intestinal health., (© 2024 Wiley‐VCH GmbH. The utilization of dietary components to support gut function and the health of young animals is an important factor for improved performance. The influence of high dietary fat levels in a low or high energy density diet on the performance of weaned piglets in relation to intestinal absorptive function, amino acid utilization, oxidative stress, and microbial metabolites was assessed in this study. The study examined the effect of two different diets containing either a low energy density/high-fat level or a high energy density/high-fat level. A total of 16 healthy weaners (9.60 ± 0.13 kg) were allocated to one of the two dietary treatments. There were eight weaners per treatment. Results showed that feed intake and body weight gain of weaners were increased by the diet of high energy density/high-fat level (p < 0.05), but the feed efficiency showed an increased tendency of significance (p = 0.05). In the duodenum, the villus height (VH) and VH/crypt depth (CD) ratio (VH:CD) were increased by dietary high energy density/high fat. In the jejunum and ileum, the CD was increased by low energy density/high-fat diet, while the goblet cell count and VH:CD were increased by dietary high energy density/high-fat level. Methionine, lysine and phenylalanine concentrations were increased by high energy density/high-fat diet while low energy density/high-fat diet showed an increased tendency to increase citrulline and ornithine concentrations in the piglet. Oxidative stress marker, lactase enzyme activity and serum calcium concentration were increased by a high energy density/high-fat diet. Increased dietary fat in all diets induced diarrhoea in the weaners (p < 0.01). Since 2022, European specifications for organic pig farming have evolved to distribute 100 % organic feed and to reinforce the link to the soil with feed resources that should primarily be obtained from the farm or the same region. Feeding strategy acts as a lever to improve various quality dimensions of organic (as well as conventional) pork, including intrinsic dimensions (carcass composition, nutritional, organoleptic, technological, sanitary qualities) and extrinsic dimensions related to animal farming (image). Diet may also influence the risk of undesirable odours or flavours that may be found in pork from non-castrated male pigs. This study aimed at evaluating the effects of a specific feeding strategy on several quality dimensions of organic meat from non-castrated male pigs. The experiment was conducted with 77 organic non-castrated male pigs (Piétrain NN × Large White) reared according to organic specifications and distributed in two batches. Within litters, male littermates were allocated at around 33 kg of live weight to either a Control group which received a Control feed (C) corresponding to the organic specifications or in a Bio+ group which received an organic test feed based mainly on French raw materials and which contained more fibres (faba bean and access to forages) and omega-3 fatty acids (linseed, camelina). All pigs were reared in the same building on deep straw bedding (1.3 m 2 /pig) with free outdoor access (1.0 m 2 /pig) using one pen per experimental group. Pigs were fed ad libitum until slaughter at about 125 kg live weight. Average daily gain, carcass weight and lean meat content did not differ significantly between C and Bio+ pigs. Compared to C, Bio+ pigs had higher (P < 0.05) ultimate pH in the loin (Longissimus muscle) and ham (Gluteus medius, and Semimembranosus) muscles, associated with a lower Longissimus muscle glycolytic potential (P < 0.001). Loin and ham meat from Bio+ vs. C pigs was lighter (P < 0.05) and had a more intense red colour (P < 0.10). The Bio+ strategy led to a lower Longissimus muscle n-6:n-3 fatty acid ratio (P < 0.001), indicating an improvement in pork nutritional value. Backfat skatole concentration was lower in Bio+ than in C pigs whereas backfat androstenone was higher in Bio+ than in C pigs (P < 0.05). Background: Longitudinal records of automatically-recorded vaginal temperature (T V ) could be a key source of data for deriving novel indicators of climatic resilience (CR) for breeding more resilient pigs, especially during lactation when sows are at an increased risk of suffering from heat stress (HS). Therefore, we derived 15 CR indicators based on the variability in T V in lactating sows and estimated their genetic parameters. We also investigated their genetic relationship with sows' key reproductive traits., Results: The heritability estimates of the CR traits ranged from 0.000 ± 0.000 for slope for decreased rate of T V (Slope De ) to 0.291 ± 0.047 for sum of T V values below the HS threshold (HSU B ). Moderate to high genetic correlations (from 0.508 ± 0.056 to 0.998 ± 0.137) and Spearman rank correlations (from 0.431 to 1.000) between genomic estimated breeding values (GEBV) were observed for five CR indicators, i.e. HS duration (HSD), the normalized median multiplied by normalized variance (Nor&#95;medvar), the highest T V value of each measurement day for each individual (Max Tv ), and the sum of the T V values above (HSU A ) and below (HSU B ) the HS threshold. These five CR indicators were lowly to moderately genetically correlated with shoulder skin surface temperature (from 0.139 ± 0.008 to 0.478 ± 0.048) and respiration rate (from 0.079 ± 0.011 to 0.502 ± 0.098). The genetic correlations between these five selected CR indicators and sow reproductive performance traits ranged from - 0.733 to - 0.175 for total number of piglets born alive, from - 0.733 to - 0.175 for total number of piglets born, and from - 0.434 to - 0.169 for number of pigs weaned. The individuals with the highest GEBV (most climate-sensitive) had higher mean skin surface temperature, respiration rate (RR), panting score (PS), and hair density, but had lower mean body condition scores compared to those with the lowest GEBV (most climate-resilient)., Conclusions: Most of the CR indicators evaluated are heritable with substantial additive genetic variance. Five of them, i.e. HSD, Max Tv , HSU A , HSU B , and Nor&#95;medvar share similar underlying genetic mechanisms. In addition, individuals with higher CR indicators are more likely to exhibit better HS-related physiological responses, higher body condition scores, and improved reproductive performance under hot conditions. Fibres, as abundant in agricultural by-products, exhibit a large range of physicochemical properties that can influence digestive processes such as digesta mean retention time (MRT), thereby affecting nutrient digestion kinetics. In this study, we investigated the effects of particle size of insoluble fibres, and gelation of soluble fibres on MRT of liquids, fine solids, and fibrous particles in the different segments of the gastrointestinal tract (GIT) of pigs. Twenty-four boars (51.6 ± 4.90 kg) were allocated to four diets; two diets contained 15% wheat straw, either coarsely chopped or finely ground (1-mm screen), two diets contained 27% wheat bran without or with the addition of 10% low-methylated pectin. After 14 days of adaptation to the diet, a total collection of faeces was performed to determine the total tract digestibility of nutrients. Thereafter, pigs were fed diets supplemented with tracers for at least 5 days and dissected following a frequent feeding procedure to approach steady-state passage of digesta. The MRT of liquids (Co-EDTA), fine solids (TiO 2 ), and fibrous particles (Chromium-mordanted fibres) in the different segments of the GIT were quantified. In the stomach, particle size reduction of straw decreased the MRT of fine solids by 02:39 h, and fibrous particles by 07:21 h (P < 0.10). Pectin addition to the wheat bran diet reduced the MRT of fine solids by 03:09 h, and fibrous particles by 07:10 h (P < 0.10), but not of liquids, resulting in less separation between digesta phases in the stomach compared with the bran diet (P < 0.05). In the mid-small intestine (SI), pectin addition reduced the MRT of fibrous particles and the separation between fibrous particles and fine solids. No further effects of particle size reduction of straw nor pectin addition on MRT and digestibility of starch, nitrogen, or fat were observed in the SI. In the large intestine (LI), particle size reduction of straw reduced separation between fibrous particles and fine solids (P < 0.10), while pectin addition had no effects. Total tract, non-starch polysaccharide degradation of straw was poor (∼31%), and unaffected by particle size reduction (P > 0.10). The complete fermentation of pectin did not influence the degradation of wheat bran fibres (∼51%). In conclusion, the effects of particle size of insoluble fibres and gelling properties of soluble fibres on the passage of digesta phases were most pronounced in the stomach, but less prominent in distal segments of the GIT., (Copyright © 2024 The Author(s). Farrowing crates are widely used as the main housing system for farrowing and lactating sows on commercial farms although they have a negative impact on sow welfare and behaviour. One of the main reasons for using farrowing crates is to reduce piglet mortality during farrowing and lactation. We compared the effects of temporary crating (TC) versus free farrowing (FF) with different nest-building materials on production and welfare measures. Sows were investigated using a 2 × 3 factorial design. Sows were allocated to two farrowing treatments: FF sows were not crated around farrowing, and TC sows were crated from 2 days before expected farrowing until 3 days after farrowing, when the crate was opened. In both farrowing treatments, the nest-building material was given from 7 days prior to farrowing as follows: (1) 1 L of straw twice per day, (2) 1 sheet of newspaper twice per day or (3) fastened jute fabric to the farrowing crate. The study consisted of two parts; part 1 included a total of 87 sows for data on piglet mortality and growth, and part 2 included data on nest-building and farrowing behaviour from a subset of these sows (n = 34). Farrowing treatment affected piglet mortality; FF sows had a lower number of stillborn piglets than TC sows (P = 0.04), but the number of crushed piglets was higher (P < 0.01). Nest-building material tended to have an impact on total number of crushed piglets (P = 0.08) and piglets that died during 3 days of lactation (P = 0.09). Litter growth was better in the FF group than in the TC group from piglet age of 1 day to 3 weeks (P = 0.04). Overall, little nest-building-related behaviour occurred, probably due to the small amount of nest-building material. The usability of the nest-building material varied with farrowing treatment; FF sows used newspaper most for nest-building, while TC sows used straw and jute fabric (P = 0.01). There were no differences in the piglet birth intervals or the farrowing duration. In conclusion, free farrowing can have positive effects on piglet growth but negative effects on piglet survival in early lactation. To avoid a high body protein mobilization in modern lean sows during lactation, an adequate dietary amino acid (AA) supply and an efficient AA utilization are crucial. This study evaluated the effects of dietary CP and in vitro protein digestion kinetics on changes in sow body condition, litter weight gain, milk composition, blood metabolites, protein utilization efficiency and subsequent reproductive performance. We hypothesized that a slower digestion of dietary protein would improve AA availability and utilization. In total, 110 multiparous sows were fed one of four lactation diets in a 2 × 2 factorial design, with two CP concentrations: 140 g/kg vs 180 g/kg, and two protein digestion kinetics, expressed as a percentage of slow protein (in vitro degradation between 30 and 240 min): 8 vs 16% of total protein. Feeding sows the high CP diets reduced sow weight loss (Δ = 7.6 kg, P < 0.01), estimated body fat loss (Δ = 2.6 kg, P = 0.02), and estimated body protein loss (Δ = 1.0 kg, P = 0.08), but only at a high percentage of slow protein. A higher percentage of slow protein increased litter weight gain throughout lactation (Δ = 2.6 kg, P = 0.04) regardless of CP concentrations, whereas a higher CP only increased litter weight gain during week 3 of lactation (Δ = 1.2 kg, P = 0.01). On Day 15 postfarrowing, serial blood samples were taken from a subsample of sows fed with the high CP diets. In these sows, a high percentage of slow protein resulted in higher plasma AA concentrations at 150 and 180 min after feeding (Δ = 0.89, P = 0.02, Δ = 0.78, P = 0.03, mmol/L, respectively) and lower increases in urea at 90 and 120 min after feeding (Δ = 0.67, P = 0.04, Δ = 0.70, P = 0.03, mmol/L, respectively). The higher dietary CP concentration increased total nitrogen loss to the environment (Δ = 604 g, P < 0.01) with a reduction of protein efficiency (Δ = 14.8%, P < 0.01). In the next farrowing, a higher percentage of slow protein increased subsequent liveborn litter size (Δ = 0.7, P < 0.05). In conclusion, feeding sows with a high dietary CP concentration alleviated maternal weight loss during lactation when the dietary protein digestion rate was slower, but lowered protein efficiency. A slower protein digestion improved litter weight gain, possibly by reducing AA oxidation and improving plasma AA availability, thus, improving protein efficiency., (Copyright © 2024 The Authors.
