Abstract Background In the normal process of bioethanol production, biomass is transported to integrated large factories for degradation to sugar, fermentation, and recovery of ethanol by distillation. Biomass nutrient loss occurs during preservation and degradation. Our aim was to develop a decentralized ethanol production system appropriate for farm or co-operative level production that uses a solid-state fermentation method for producing bio-ethanol from whole crops, provides cattle feed, and produces no wastes. The idea is to incorporate traditional silage methods with simultaneous saccharification and fermentation. Harvested, fresh biomass is ensiled with biomass-degrading enzymes and yeast. Multiple parallel reactions for biomass degradation and ethanol and lactic acid production are induced in solid culture in hermetically sealed containers at a ranch. After fermentation, ethanol is collected on site from the vapor from heated fermented products. Results The parallel reactions of simultaneous saccharification and fermentation were induced efficiently in the model fermentation system. In a laboratory-scale feasibility study of the process, 250 g of freshly harvested forage rice with 62% moisture was treated with 0.86 filter paper units/g dry matter (DM) of cellulase and 0.32 U/g DM of glucoamylase. After 20 days of incubation at 28°C, 6.4 wt.% of ethanol in fresh matter (equivalent to 169 g/kg DM) was produced. When the 46 wt.% moisture was gathered as vapor from the fermented product, 74% of the produced ethanol was collected. Organic cellular contents (such as the amylase and pronase degradable fractions) were decreased by 63% and organic cell wall (fiber) content by 7% compared to silage prepared from the same material. Conclusions We confirmed that efficient ethanol production is induced in nonsterilized whole rice plants in a laboratory-scale solid-state fermentation system. For practical use of the method, further study is needed to scale-up the fermentation volume, develop an efficient ethanol recovery method, and evaluate the fermentation residue as an actual cattle feed.

We developed a rapid and simple method for evaluating the degradation of solid biodegradable plastics (BPs). Dye-containing BP films were used as substrates and the release of dye caused by the degradation of BPs was confirmed by a color change in the enzyme solution after a reaction time of 24 h. Abstract: Silage production from rice straw and whole-plant forage paddy rice is increasing in Japan because of decrease in rice consumption. One potential use for this silage is bioethanol production. In this study, we analyzed the effectiveness of three different commercially available cellulases at saccharification of sun-dried rice straw, ensiled rice straw, and rice whole-crop silage (WCS). Furthermore, the ethanol productivity of the simultaneous saccharification and ethanol fermentation process (SSF) from the same plant substrates was analyzed. Among the three kinds of cellulases tested (Novozymes NS50013, Genencor GC220, and Acremonium cellulase), Acremonium cellulase showed the highest ethanol production for the three plant substrates, and the WCS produced the highest ethanol level. Analysis of the enzymatic degradation activity of the cellulases revealed that Acremonium cellulase contained remarkably high glucoamylase and pectinase side activities relative to the other cellulase preparations. The addition of glucoamylase and pectinase to the other two cellulases significantly increased ethanol productivity to levels observed for the Acremonium cellulase preparation, which showed little enhanced performance with the addition of the same enzymes. Finally, we tested whether milling and sterilization had an effect on ethanol production and found that sterilized silage produced higher ethanol levels but that the milling process had no significant effect. These results show that (i) silage made from whole-plant rice can be used for bioethanol production and (ii) the proper selection and combination of commercially available enzymes can make SSF more cost efficient by removing the need for a pre-treatment step. Esterases from the yeast Pseudozyma antarctica (PaE) and the fungus Paraphoma sp. B47-9 (PCLE) can degrade biodegradable plastics (Shinozaki et al., 2013; Suzuki et al., 2014). The degradation profiles of plastic films composed of poly(butylene succinate), poly(butylene succinate- co -adipate), or poly(butylene adipate) by these enzymes were characterized by liquid chromatography-mass spectroscopy in terms of the molecular structures and molecular weights of the degradation products. Monomers and oligomers with molecular weights corresponding to dimers to octamers were identified as products of degradation by PaE in an aqueous reaction solution, irrespective of the type of biodegradable plastic film. Size-exclusion chromatography indicated that the number-average molecular weight of degraded films decreased with reaction time, suggesting that PaE degraded polyester films randomly into monomer units (endo-type degradation). 4-Hydroxy-3-methyl-2(1 H )-quinolone ( 1 ), a molecule known for a long time and recently discovered from a Brassicaceae plant Isatis tinctoria without providing sufficient evidence to support the structure, was isolated from a fermentation extract of Burkholderia sp. 3Y-MMP isolated from a soil by a Zn 2+ enrichment culture. Detailed spectroscopic analyses by MS and NMR, combined with 13 C chemical shift comparison with literature values of the related compounds and a synthetic preparation of 1 , allowed its first full NMR characterization and identification of 2-quinolone but not 2-quinolinol ( 2 ) as the preferred tautomer for this heterocyclic system. While the metal-chelating activity was negligible, compound 1 at 10 μM, a concentration lower than that in liquid production cultures, quenched hydroxy radical-induced chemiluminescence emitted by luminol by 86%. Because some Burkholderia species are pathogenic to plants and animals, the above result suggests that 1 is a potential antioxidant to counteract reactive oxygen species-based immune response in the host organisms., (Copyright © 2020, Li et al.; licensee Beilstein-Institut.)

Siderophores are considered to have a good potential as decontamination agents owing to their metal-chelating abilities. In order to confirm whether siderophores can be used in the recovery of metal ions, a siderophore (or metallophore) exhibiting Co 2+ -chelating activity was screened to demonstrate its ability to recover Co 2+ from an aqueous solution. A siderophore-producing bacterium, Pandoraea sp. HCo-4B, was identified from a screen of Co 2+ -resistant bacteria grown in an aerobic enrichment culture with a Co 2+ -supplemented medium. After incubation of the crude extracted siderophore in a Co 2+ -containing solution, the Co 2+ -siderophore complex was adsorbed on to a C 18 column. The bound Co 2+ was eluted from the column by the addition of 10 mM H 2 SO 4 . The recovered amount of Co 2+ was proportional to the amount of the added siderophore. We observed that the siderophore identified in this study binds to Co 2+ at a 1:1 ratio.

To improve the productivity of Paraphoma-like fungal strain B47-9 for biodegradable plastic (BP)-degrading enzyme (PCLE), the optimal concentration of emulsified poly(butylene succinate-co-adipate) (PBSA) in the medium was determined. Emulsified PBSA was consumed as a sole carbon source and an inducer of PCLE production by strain B47-9. Among the various concentrations of emulsified PBSA [0.09-0.9% (w/v)] used in flask cultivation, 0.27% yielded the maximum enzyme activity within a short cultivation period. To evaluate the residual concentration of emulsified PBSA in culture, emulsified PBSA in aliquots of culture supernatant was digested in vitro, and the concentration of released monomerised succinic acid was determined. Regardless of the initial concentration of emulsified PBSA in medium, PCLE activity was detected after residual succinic acid decreased below 0.04 mg/mL in culture broth. Jarfermentation was performed at a 0.27% PBSA concentration. Among the various airflow rates tested, 1 LPM resulted in a PCLE production rate of 1.0 U/mL/day. The enzyme activity in the resulting culture filtrate (4.2 U/2 mL) was shown to degrade commercial BP films (1 × 1 cm, 20 µm thickness) within 8 hours.

Background: In an attempt to reduce environmental loading during ethanol production from cellulosic plant biomass, we have previously proposed an on-site solid state fermentation (SSF) method for producing ethanol from whole crops, which at the same time provides cattle feed without producing wastes. During the ensiling of freshly harvested plant biomass with cellulase and glucoamylase, the added yeast and lactic acid bacteria induced simultaneous saccharification and production of ethanol and lactic acid in hermetically sealed containers on-farm. In a previous study, laboratory-scale SSF (using 250 g of fresh rice crop biomass) yielded 16.9 weight % ethanol in dry matter (DM) after 20 days of incubation. In this study, the fermentation volume was scaled up to a normal-sized round bale and the fermentation process (ethanol concentrations of the products) was monitored. The ethanol produced was recovered and the recovery efficiency was evaluated., Results: SSF tests with forage rice round bales using polyethylene-wrapped whole plant materials (cultivar Leaf Star, average of 125.2 kg dry weight) were monitored in the field without temperature control. They yielded 14.0 weight % ethanol and 2.9 weight % lactic acid in DM after six months of incubation, and the ethanol ratio in the bale remained stable for 14 months after processing. SSF tests with three different rice cultivars were conducted for three years. Ethanol recovery from a fermented whole bale (244 kg fresh matter (FM) containing about 12.4 kg ethanol) by one-step distillation using vacuum distillation equipment yielded 86.3% ethanol collected from distilled solution (107 kg of 10.0 weight % ethanol). In addition, an average of 1.65 kg ethanol in 40.8 kg effluent per bale was recovered. Relative nitrogen content was higher in SSF products than in silage made from the same plant material, indicating that fermentation residue, whose quality is stabilized by the lactic acid produced, can be used as cattle feed., Conclusions: We have successfully demonstrated an efficient on-site ethanol production system with non-sterilized whole rice crop round bale. However, issues concerning the establishment of the ethanol recovery procedure on-site and evaluation of the fermentation residue as cattle feed have to be addressed.

A 41-year-old woman with idiopathic thrombocytopenic purpura and dyslipidemia abruptly developed vertigo, truncal ataxia and divergence paralysis. Cranial magnetic resonance imaging demonstrated the presence of infarction in the left superior paramedian mesencephalic artery involving the vicinity of the periaqueductal gray matter. The symptoms rapidly resolved under the administration of anti-platelet agents. The precise location of the hypothetical divergence center of the ocular motor system remains unclear because the lesions responsible for divergence paralysis are rarely identified on neuroimaging. We emphasize that this is a first reported case of DP caused by acute midbrain infarction and speculate that the mesencephalic reticular formation may be partially involved.

A 71-year-old man with hyperlipidemia abruptly developed left-sided isolated shoulder palsy. Cranial magnetic resonance imaging demonstrated infarction of the cortical branch of the right middle cerebral artery (MCA). In the primary motor cortex, there is broad somatotopic representation of various body parts in a particular arrangement, and the area corresponding to the shoulder is very small. Consequently, there have been only 3 reported cases of isolated shoulder palsy due to cerebral infarction, and its vascular supply remains uncertain. The present case indicates that the corresponding area to the shoulder receives its blood from the cortical branch of the MCA.
