Hepatocellular carcinoma (HCC) is a common malignant primary tumor that is usually diagnosed at an advanced stage; thus, there is an urgent need for efficient and sensitive novel diagnostic markers to determine the prognosis and halt disease progression in patients with HCC. Disulfidptosis is a recently discovered form of programmed cell death, essentially an abnormal accumulation of intracellular bisulfides. Therefore, our study aimed to investigate the role of disulfidptosis-related genes (DRGs) in the pathogenesis of HCC. Based on public databases, our work demonstrates the relationship between DRG and expression, immunity, mutation/drug sensitivity, and functional enrichment in HCC. We also revealed the significant heterogeneity of HCC in different DRGs sub-clusters and in differentially expressed genes (DEGs), respectively. Subsequently, the most relevant candidate gene, SLC7A11, was screened by machine learning to further validate the significance of SLC7A11 in the clinical features, prognosis, nomogram pattern, and immune infiltration of HCC. Our study, which elucidates the potential mechanisms of DRGs and HCC, reveals that SLC7A11 can serve as a novel prognostic biomarker and provides opportunities and challenges for individualized cancer immunotherapy strategies.

AIM: To investigate the choice of surgical approach, therapeutic effect and complications of orbital cavernous hemangioma(OCH)in different positions of orbit.METHODS: The clinical data of 128 patients of 128 eyes with OCH whose were surgically removed and pathologically diagnosed in the department of ophthalmology of the Second Affiliated Hospital of Air Force Military Medical University from January 2016 to August 2021 were retrospectively analyzed. The position of OCH in the orbit was determined by preoperative imaging examination(CT/MRI), so as to select different surgical approaches and analyze the postoperative curative effect and the incidence of complications.RESULTS: The location of OCH in the orbit: 82 eyes in the muscle cone and 46 eyes outside the muscle cone. According to the quadrant of the orbit where the tumor was located, there were 24 eyes in the upper-outer quadrant, 38 eyes in the lower-outer quadrant, 28 eyes in the upper-inner quadrant, 12 eyes in the lower-inner quadrant, and 26 eyes the intraconal central space. The selection of surgical approach: 1)OCH in the muscle cone: conjunctival approach surgery in 53 eyes, lateral orbital approach surgery in 22 eyes, lateral combined medial conjunctiva orbital approach surgery in 5 eyes, skin approach surgery in 1 eye, transnasal approach under nasal endoscope surgery in 1 eye; 2)OCH outside the muscle cone: skin approach in 29 eyes, conjunctival approach in 12 eyes and lateral orbital approach in 5 eyes. Postoperative efficacy: except for 1 eye of postoperative tumor residue, the other 127 eyes were completely removed. Postoperative complications: 1)Ocular motility disorder: 16 eyes, including conjunctival approach surgery in 11 eyes, lateral orbital approach surgery in 4 eyes, lateral combined medial conjunctiva orbital approach surgery in 1 eye; 2)Visual acuity decreased in 9 eyes: conjunctival approach surgery in 3 eyes, lateral orbital approach surgery in 6 eyes; 3)Mydriasis occurred in 9 eyes, including 4 eyes via conjunctival approach surgery and 5 eyes via lateral orbital approach surgery; 4)Intraorbital hemorrhage occurred in 3 eyes: all occurred through conjunctival approach surgery; 5)Visual acuity was lost in 2 eyes, including conjunctival approach surgery in 1 eye and lateral orbital opening approach surgery in 1 eye; 6)There were 2 eyes of ptosis, including conjunctival approach surgery in 1 case and skin approach surgery in 1 eye; 7)Tumor residue occurred in 1 eye: multiple intraorbital tumors were treated by lateral orbital approach surgery.CONCLUSION: The accurate location of OCH combined with imaging examination and the selection of appropriate surgical approaches according to different locations can successfully remove the tumor and reduce the incidence of complications.

Esophageal cancer (ESCA) is a common malignant tumor with poor prognosis. Accumulating evidence indicates an important role of lysosomal-associated membrane protein 2 (LAMP2) in the progression and development of various cancers. In this study, we obtained RNA-sequencing raw count data and the corresponding clinical information for ESCA samples from The Cancer Genome Atlas and Gene Expression Omnibus databases. We comprehensively investigated the expression and prognostic significance of LAMP2 and relationships between LAMP2 expression and prognosis, different clinicopathological parameters, and immune cell infiltration in ESCA. We also obtained the differentially expressed genes between the high LAMP2 expression and low LAMP2 expression groups in ESCA and performed a functional enrichment analysis of the 250 linked genes most positively related to LAMP2 expression. Moreover, we performed the pan-cancer analysis of LAMP2 to further analyze the role of LAMP2 in 25 commonly occurring types of human cancer. We also verified and compared the expression of LAMP2 in 40 samples of human ESCA tissue and adjacent tissues. The results indicated that LAMP2 expression was significantly upregulated in ESCA and various human cancers. In addition, LAMP2 expression was associated with certain clinicopathological parameters, prognosis, and immune infiltration in ESCA and the other types of cancer. Our study represents a comprehensive pan-cancer analysis of LAMP2 and supports the potential use of the modulation of LAMP2 in the management of ESCA and various cancers.

The long-term prognosis of patients with lung cancer remains poor and thus it is imminent to further elucidate the molecular mechanism for the oncogenesis of lung cancer. In this study, we observed that surfactant protein C (SFTPC) expression was downregulated in human lung adenocarcinoma tissues and cell lines, and low SFTPC expression correlated with poor overall survival of lung adenocarcinoma patients. Moreover, we found that overexpression of SFTPC could inhibit lung cancer cell proliferation in vitro and in vivo, but downregulation of SFTPC showed the opposite results. Besides, it was observed that miR-629-3p expression was upregulated in human lung adenocarcinoma tissues and cell lines. More importantly, we found that miR-629-3p could downregulate SFTPC expression by directly binding to the SFTPC 3'-UTR and inhibit the regulatory effect of SFTPC on lung adenocarcinoma cell proliferation. In conclusion, these data suggested that miR-629-3p-meditated downregulation of SFTPC may promote lung adenocarcinoma progression.

Abstract Background Retinoblastoma is the most common intraocular malignancy occurring in children. It can metastasize to the regional lymph nodes, central nervous system and distant organs usually the bones and bone marrow and very rarely to the soft tissue. Here, we report a case of unilateral retinoblastoma in a 4-year-old girl accompanied by a large metastasis of the parotid and submandibular glands that developed about 6 months previously and gradually increased in size 5 months after enucleation of the left eye. Case presentation A 4-year-old girl with a history of unilateral retinoblastoma presented with a large, painful and worsening mass (about 20 × 23 cm) of the left side of the neck. Following surgery, the orbital tumour was completely resected, and the large tumour invasion range in the left side of the neck was not resected completely. Histopathological examination revealed retinoblastoma of the orbit and the parotid and submandibular glands. After chemotherapy and additional local radiotherapy on the parotid and submandibular glands, the tumour was inactive and stable. Conclusions Delayed detection and inappropriate management contribute to poor outcomes. Fundus examinations, education regarding the early signs of RB, and optimization of the therapeutic strategy for RB may play important roles in ocular health.

Purpose: To compare the clinical efficacy and complications of limited internal fixation combined with external fixation (LIFEF) and open reduction and internal fixation (ORIF) in the treatment of Pilon fracture. Methods: We searched databases including Pubmed, Embase, Web of science, Cochrane Library and China Biology Medicine disc for the studies comparing clinical efficacy and complications of LIFEF and ORIF in the treatment of Pilon fracture. The clinical efficacy was evaluated by the rate of nonunion, malunion/delayed union and the excellent/good rate assessed by Mazur ankle score. The complications including infections and arthritis symptoms after surgery were also investigated. Results: Nine trials including 498 pilon fractures of 494 patients were identified. The meta-analysis found no significant differences in nonunion rate (RR = 1.60, 95% CI: 0.66 to 3.86, p = 0.30), and the excellent/good rate (RR = 0.95, 95% CI: 0.86 to 1.04, p = 0.28) between LIFEF group and ORIF group. For assessment of infections, there were significant differences in the rate of deep infection (RR = 2.18, 95% CI: 1.34 to 3.55, p = 0.002), and the rate of arthritis (RR = 1.26, 95% CI: 1.03 to 1.53, p = 0.02) between LIFEF group and ORIF group. Conclusion: LIFEF has similar effect as ORIF in the treatment of pilon fractures, however, LIFEF group has significantly higher risk of complications than ORIF group does. So LIFEF is not recommended in the treatment of pilon fracture.

AIM: To investigate the effects of lentivirus (LV) mediated integrin-linked kinase (ILK) RNA interference (RNAi) on biological behaviors of human lens epithelial cells (LECs). METHODS: Human cataract LECs and immortalized human LEC line, human lens epithelial (HLE) B-3 cells were transfected by lentiviral vector expressing ILK-specific short hairpin RNA (shRNA) and then stimulated by transforming growth factor- (TGF-, the silencing of ILK gene and protein was identified by reverse transcription-polymerase chain reaction (RT-PCR) and Western blot methods; biological behaviors including cell cycle and apoptosis, cell morphology, -smooth muscle actin (SMA) stress fiber formation and cell migration were examined. RESULTS: Remarkable decreases of ILK protein expression were detected in LECs carrying lentiviral ILK-shRNA vector; flow cytometry revealed arresting of cell cycle progression through the G1/S transition and higher apoptosis rate in ILK-RNAi-LV transfected cells. Less -SMA stress fiber formation and migration was observed in ILK-RNAi-LV transfected LECs. CONCLUSION: The present study demonstrated that ILK was an important regulator for LECs proliferation and migration. LV mediated ILK RNAi is an effective way to decrease ILK-regulated cell growth by arresting cell cycle progression and increasing cell apoptosis, as well as, to prevent cell migration by inhibiting TGF- induced -SMA stress fiber formation. Thus, LV mediated ILK RNAi might be useful to prevent posterior capsular opacification.

AIM: To evaluatethe laser photocoagulation of different ranges for diabetic retinopathy(DR)prognosis.METHODS: A total of 98 cases(153 eyes)with proliferative diabetic retinopathy treated by laser photocoagulation from November 2010 to November 2012 were enrolled in this study. The eyes were divided into three groups, Group A of 38 eyes(25%)receiving pan-retinal photocoagulation, group B of 55 eyes(36%)receiving quadrant retinal laser photocoagulation and group C 60 eyes(39%)receiving retinal laser photocoagulation only in the ischemic area. Blood glucose, refraction, intraocular pressure and vision were measured preoperatively and postoperatively. And all patients underwent VEP, ERG, fundus fluorescein angiography(FFA)and optical coherence tomography(OCT)examinations before and after operation. Patients were followed up for 3 months. RESULTS: After 3 months, decrease of macular thickness in group A and B was more significant than that in group C(PPCONCLUSION: Quadrant retinal laser photocoagulation can significantly improve retinal hypoxia, prevent retinal neovascularization and protect visual function in macular area.

Objective To investigate the effect of heparanase over-expression on the biological function of gallbladder carcinoma cell line GBC-SD. Methods The heparanase over-expression plasmid vector was constructed and transfected into GBC-SD cells by liposome method. The transfection efficiency was verified by RT-qPCR and Western blot. CCK-8 assay, flow cytometry and scratch healing experiment were used to detect the cell proliferation, apoptosis and migration.The expression of syndecan-1, FGF-2 and E-cadherin protein was detected by Western blot. Results The expression of heparanase mRNA(P＜0.01) and protein(P＜0.05) in gallbladder cancer cell line GBC-SD was significantly increased after transfection.Compared with the control group,the over-expression of heparanase significantly up-regulated the proliferation and migration ability of GBC-SD cells in vitro(P＜0.05), down-regulated the level of apoptosis(P＜0.05),and increased the expression of FGF-2 protein, while decreasing the expression of syndecan-1(P＜0.01) and E-cadherin(P＜0.05). Conclusions Over-expression of heparanase promotes the proliferation and migration of gallbladder cancer cell line GBC-SD, and the mechanism may be related to its regulation of E-cadherin through FGF-2.

Metal coatings with good conductivity and hydrophobicity property were prepared on wood surface via a simple electroless nickel (Ni) approach. The variation of crosssectional area for coatings on wood surface was investigated. The cross-sectional area of the coatings increased as the number of deposition steps was increased. The resistance, conductivity, and hydrophobicity property of the coatings were obviously enhanced with the increase in the number of depositions. The composite structure was characterised with scanning electron microscopy (SEM) images. The conductivity and hydrophobicity property of the ideal coatings on wood surface can be greatly improved. Here, the resistance of longitudinal coatings was as large as 28.4 mΩ and the conductivity can be up to 706.7 S/cm. The resistance of the coatings was decreased 5 times compared with that of coatings obtained via a single deposition. The contact angle of coatings varied in angle between 90.7 and 116.89°. The cross-sectional area change of coatings had a direct influence on the properties of coatings. [ABSTRACT FROM AUTHOR]

Hollow composite coatings were prepared on the surface of cellulose fibres by a simple electroless Ni-P/nano-SiC approach. The metal coated extent, dispersion extent of micro/nano cellulose fibres and crystalline structure of Ni-P/nano-SiC composite coatings were investigated. The homogeneous hollow composite coatings and metal coated extent of micro/nano cellulose fibres were improved with increase in concentration of nano-SiC in the coatings, and the ideal composite coatings were obtained within the range of 0.2 to 0.35 g (0.6 g/l) nano-SiC in the solution bath. The metallisation for cellulose fibres enhanced the dispersion extent of micro-nano cellulose fibres. A uniform coating, consisting of the hollow coating on cellulose fibres surface, could be obtained. At the same time, metallisation did not damage the original structure and surface functional groups of the cellulose fibres. The concentration of nano-SiC, ultrasonic time, ultrasonic power and cellulose concentration had a direct influence on the metal coated extent on the surface of cellulose fibres. The coated extent, dispersion extent and crystalline structure of metallisation cellulose fibres would exhibit ideal characteristics. Here, the concentration of cellulose fibres, ultrasonic time, ultrasonic power and the concentration of nano-SiC were 1.2 g/l, 120 min, 960 W and 0.6 g/l, respectively. [ABSTRACT FROM AUTHOR]

This study aims to observe the improvement of non-alcoholic steatohepatitis(NASH) after using water extracts of Polygoni Multiflori Radix and Polygoni Multiflori Radix Praeparata and explore their preliminary mechanism. Mice were fed with methionine-choline-deficent diet(MCD) for 6 weeks for modeling, and mice were orally given with 50, 100, 200 mg·kg~(-1) of Polygoni Multiflori Radix water extract(PMRWE) or Polygoni Multiflori Radix Praeparata water extract(PMRPWE) at the last 4 weeks. During the whole experimental procedure, the body weight changes of the mice were monitored and recorded. Serum alanine aminotransferase(ALT) and aspartate aminotransferase(AST) activities were detected; liver histopathological evaluation and NAFLD activity score(NAS) calculation were conducted, and the levels of reactive oxygen species(ROS) in liver tissues were analyzed. The contents of triglyceride(TG) and non-esterified fatty acids(NEFA) in liver tissues were detected, and oil red O staining of the liver tissues was conducted and observed. Quantitative polymerase chain reaction(qPCR) was used to detect hepatic mRNA expression of β-oxidation-related genes in mice. The results showed that PMRWE(100, 200 mg·kg~(-1)) and PMRPWE(50, 100, 200 mg·kg~(-1)) alleviated liver damage in MCD-induced NASH in mice. PMRWE(100, 200 mg·kg~(-1)) and PMRPWE(50, 100, 200 mg·kg~(-1)) reduced hepatic li-pid accumulation in mice with NASH. Different doses of PMRPWE inversed the decreased hepatic mRNA expression of β-oxidation-related genes in mice with NASH. This study indicated that PMRPWE and PMRWE could ameliorate MCD-induced NASH in mice by promoting fatty acid β oxidation, reducing liver lipid accumulation, and alleviating liver damage. Moreover, the protective effect of PMRPWE against MCD-induced NASH was better than PMRWE.
