We present new unit operations for valving and switching in centrifugal microfluidics that are actuated by a temperature change rate (TCR) and controlled by the rotational frequency. Implementation is realized simply by introducing a comparatively large fluidic resistance to an air vent of a fluidic structure downstream of a siphon channel. During temperature decrease at a given TCR, the air pressure inside the downstream structure decreases and the fluidic resistance of the air vent slows down air pressure compensation allowing a thermally induced underpressure to build up temporarily. Thereby the rate of temperature change determines the time course of the underpressure for a given geometry. The thermally induced underpressure pulls the liquid against a centrifugal counterpressure above a siphon crest, which triggers the valve or switch. The centrifugal counterpressure (adjusted by rotation) serves as an independent control parameter to allow or prevent valving or switching at any TCR. The unit operations are thus compatible with any temperature or centrifugation protocol prior to valving or switching. In contrast to existing methods, this compatibility is achieved at no additional costs: neither additional fabrication steps nor additional disk space or external means are required besides global temperature control, which is needed for the assay. For the layout, an analytical model is provided and verified. The TCR actuated unit operations are demonstrated, first, by a stand-alone switch that routes the liquid to either one of the two collection chambers (n = 6) and, second, by studying the robustness of TCR actuated valving within a microfluidic cartridge for highly integrated nucleic acid testing. Valving could safely be prevented during PCR by compensating the thermally induced underpressure of 3.52 kPa with a centrifugal counterpressure at a rotational frequency of 30 Hz with a minimum safety range to valving of 2.03 kPa. Centrifugal microfluidics shows a clear trend towards a higher degree of integration and parallelization. This trend leads to an increase in the number and density of integrated microfluidic unit operations. The fact that all unit operations are processed by the same common spin protocol turns higher integration into higher complexity. To allow for efficient development anyhow, we introduce advanced lumped models for network simulations in centrifugal microfluidics. These models consider the interplay of centrifugal and Euler pressures, viscous dissipation, capillary pressures and pneumatic pressures. The simulations are fast and simple to set up and allow for the precise prediction of flow rates as well as switching and valving events. During development, channel and chamber geometry variations due to manufacturing tolerances can be taken into account as well as pipetting errors, variations of contact angles, compliant chamber walls and temperature variations in the processing device. As an example of considering these parameters during development, we demonstrate simulation based robustness analysis for pneumatic siphon valving in centrifugal microfluidics. Subsequently, the influence of liquid properties on pumping and valving is studied for four liquids relevant for biochemical analysis, namely, water (large surface tension), blood plasma (large contact angle hysteresis), ethanol/water (highly wetting) and glycerine/water (highly viscous). In a second example, we derive a spin protocol to attain a constant flow rate under varying pressure conditions. Both examples show excellent agreement with experimental validations. Dynamical nuclear polarization holds the key for orders of magnitude enhancements of nuclear magnetic resonance signals which, in turn, would enable a wide range of novel applications in biomedical sciences. However, current implementations of DNP require cryogenic temperatures and long times for achieving high polarization. Here we propose and analyze in detail protocols that can achieve rapid hyperpolarization of 13C nuclear spins in randomly oriented ensembles of nanodiamonds at room temperature. Our protocols exploit a combination of optical polarization of electron spins in nitrogen-vacancy centers and the transfer of this polarization to 13C nuclei by means of microwave control to overcome the severe challenges that are posed by the random orientation of the nanodiamonds and their nitrogen-vacancy centers. Specifically, these random orientations result in exceedingly large energy variations of the electron spin levels that render the polarization and coherent control of the nitrogen-vacancy center electron spins as well as the control of their coherent interaction with the surrounding 13C nuclear spins highly inefficient. We address these challenges by a combination of an off-resonant microwave double resonance scheme in conjunction with a realization of the integrated solid effect which, together with adiabatic rotations of external magnetic fields or rotations of nanodiamonds, leads to a protocol that achieves high levels of hyperpolarization of the entire nuclear-spin bath in a randomly oriented ensemble of nanodiamonds even at room temperature. This hyperpolarization together with the long nuclear-spin polarization lifetimes in nanodiamonds and the relatively high density of 13C nuclei has the potential to result in a major signal enhancement in 13C nuclear magnetic resonance imaging and suggests functionalized and hyperpolarized nanodiamonds as a unique probe for molecular imaging both in vitro and in vivo. Microfluidic systems for polymerase chain reaction (PCR) should be fully closed to avoid vapor loss and to exclude the risk of contaminating the laboratory environment. In closed systems however, the high temperatures of up to 95 °C associated with PCR cause high overpressures up to 100 kPa, dominated by the increase of vapor partial pressure upon evaporation. Such high overpressures pose challenges to the mechanical stability of microfluidic chips as well as to the liquid handling in integrated sample-to-answer systems. In this work, we drastically reduce the pressure increase in fully closed PCR systems by integrating a microchannel that serves as a vapor-diffusion barrier (VDB), separating the liquid-filled PCR chamber from an auxiliary air chamber. In such configurations, propagation of vapor from the PCR chamber into the auxiliary air chamber and as a consequence the increase of pressure is limited by the slow diffusion process of vapor through the VDB. At temperature increase from 23 °C to 95 °C, we demonstrate the reduction of overpressure from more than 80 kPa without the VDB to only 35 kPa with the VDB. We further demonstrate proper function of VDB and its easy integration with downstream processes for PCR based nucleic acid amplification within centrifugal microfluidics. Key importance and the role of sewn seam in the woven fabric are presented. Fabric properties in the places joined with the sewn seam on which garment durability, applicability, appearance and quality depend are highlighted. Seam location is usually the weakest spot on the garment, especially when the great loads are present at these places. Mechanical properties of the sewn fabrics were investigated in different test conditions. The influence of the load on the sewn seam is analyzed in time intervals. Sample with seam have approximately 70% lower breaking properties (breaking force and elongation at break) than the sample without seam. By preloading the seam fabrics with 30%, 50% and 70% of elongation at break, for a 3-h period, the effect on mechanical properties were tested and the differences in pre-stretching intensity were observed. By conducting such test, it is expected that the fabric experience further degradation and that breaking properties further reduce, but the exact opposite occurred (breaking properties, by increasing the preload, increase even more), because of various material properties (structural fabric properties, raw material characteristics, seam characteristics).

Stings in the head region are considered to be a risk factor for severe systemic reactions to hymenoptera stings. We supposed that stings in skin areas, which are well supplied with blood, lead to more severe reactions and tested our hypothesis in 847 patients with confirmed hymenoptera venom allergy. However, symptom severity was independent from sting site: only 16.3% of patients with severe reactions were stung on the head ( P = 0.017). But we confirmed age > 40 years ( P < 0.001) as well as elevated basal tryptase levels ( P = 0.001) as risk factors. Taking antihypertensive drugs seemed to have an influence: 41.7% of patients taking antihypertensive drugs experienced a severe reaction compared to 29.5% of patients, not taking such drugs ( P = 0.019). However, considering patients' age in regression analysis, taking antihypertensive drugs had no effect on symptom severity ( P = 0.342). Two composts produced at different times of the year from garden waste, sewage sludge and wood ash were evaluated for use as ornamental plant substrates. The maturity and lack of phytotoxicity of both composts, as well as the absence of E. coli, were first confirmed by use of laboratory procedures and rapid test kits. A greenhouse experiment was then carried out with two ornamental species, Petunia sp. and Tagetes sp., to evaluate the suitability of the composts as plant substrates. The performance of the composts as well as mixtures of each with 25, 50 and 75% acid peat moss was compared with that of a commercial universal substrate. Addition of the peat to the composts improved some of the physical and chemical properties. However, the results of the pot experiments indicated that under the experimental conditions used, the compost produced from green waste was suitable for use as a plant substrate, without the need for addition of other components; this appears to be an effective way of utilizing the type of urban waste considered in the study. BACKGROUND: In the Lidcombe Program of Early Stuttering Intervention, parents present verbal contingencies for stutter-free and stuttered speech in everyday situations. A previous randomized controlled trial of the programme with preschool-age children from 2005, conducted in two public speech clinics in New Zealand, showed that the odds of attaining clinically minimal levels of stuttering 9 months after randomization were more than seven times greater for the treatment group than for the control group. AIMS: To follow up the children in the trial to determine extended long-term outcomes of the programme. METHODS & PROCEDURES: An experienced speech-language therapist who was not involved in the original trial talked with the children on the telephone, audio recording the conversations using a telephone recording jack. Parental reports were gathered in addition to the children's speech samples in order to obtain a balance of objective data and reports from a wide range of situations. OUTCOMES & RESULTS: At the time of this follow-up, the children were aged 7-12 years, with a mean of 5 years post-randomization in the 2005 trial. Twenty of the 29 children in the treatment arm and eight of the 25 children in the control (no treatment) arm were able to be contacted. Of the children in the treatment group, one (5%) failed to complete treatment and 19 had completed treatment successfully and had zero or near-zero frequency of stuttering. Three of the children (16%) who had completed treatment successfully had relapsed after 2 or more years of speech that was below 1% syllables stuttered. Meaningful comparison with the control group was not possible because an insufficient number of control children were located and some of them received treatment after completing the trial. CONCLUSIONS & IMPLICATIONS: The majority of preschool children are able to complete the Lidcombe Program successfully and remain below 1% syllables stuttered for a number of years. The local anaesthetic drug salicaine hydrochloride (hydroxytetracaine hydrochloride, 4-butylamino-2-hydroxybenzoic acid 2-dimethylaminoethyl ester hydrochloride, SLCHC) and the free-base salicaine (SLC) were characterized by thermal analysis, vibrational and solid-state NMR-spectroscopy, X-ray powder diffraction, X-ray single crystal structure analysis, and water vapor sorption analysis. Additionally, the crystal structures of the anhydrate mod. II° (monoclinic, space group P21/n), the hydrated mod. I (triclinic, space group $P\,\bar 1$), and of the free base (SLC) in the form of the hemihydrate (triclinic, space group $P\,\bar 1$) are discussed. Mod. II° of the polymorphic SLCHC is the thermodynamically stable form at room temperature and is present in commercial products mostly contaminated with a hydrated form that is isomorphic with mod. I. Mod. II° crystallizes from most organic solvents and from the melt below 110°C. Mod. I crystallizes from the melt at temperatures above 110°C, and additionally appears on dehydration of the hydrated mod. I. A third polymorph monotropically related to mod. II° was found by freeze-drying. Previous studies have shown that synovial surfactant could have beneficial roles in the joint, especially as a very effective boundary lubricant capable of high load bearing. This study is aimed at further characterization and identification of the source. Known to be an important minor component of pulmonary surfactant, proteolipid has now been detected in appreciable quantities in bovine synovial fluid and bound to the articular surface. Using standard procedures to separate it from the major component [surface-active phospholipid (SAPL)] by column chromatography, proteolipid :phospholipid ratios were found to be comparable to those in the lung or in lamellar bodies (LBs). LBs are the unequivocal source of surfactant in the lung and we have confirmed an earlier study demonstrating their presence in Type B synoviocytes. Using a fixation procedure specifically designed to preserve the graphite-like structure of SAPL deposited as oligolamellar layers, or coiled as lamellar bodies, we were able to demonstrate these structures in equine joints adjacent to the Golgj apparatus associated with the secretory mechanism of the cell. These results indicate that proteolipid could be facilitating the deposition of the graphite-like surface lining of SAPL providing efficient boundary lubrication just as it promotes surfactant adsorption in the lung and in the formation of myelin. Any deficiency in synovial surfactant, compromising its roles in the joint, is discussed in relation to osteoarthritis and the possible administration of exogenous SAPL to the degenerating joint. We propose to use a dissipatively stabilized nitrogen vacancy (NV) center as a mediator of interaction between two nuclear spins that are protected from decoherence and relaxation of the NV due to the periodical resets of the NV center. Under ambient conditions this scheme achieves highly selective high-fidelity quantum gates between nuclear spins in a quantum register even at large NV-nuclear distances. Importantly, this method allows for the use of nuclear spins as a sensor rather than a memory, while the NV spin acts as an ancillary system for the initialization and readout of the sensor. The immunity to the decoherence and relaxation of the NV center leads to a tunable sharp frequency filter while allowing at the same time the continuous collection of the signal to achieve simultaneously high spectral selectivity and high signal-to-noise ratio. The coherent high-fidelity generation of nuclear spins in long-lived singlet states, which may find application as quantum memory or sensor, represents a considerable experimental challenge. Here, we propose a dissipative scheme that achieves the preparation of pairs of nuclear spins in long-lived singlet states by a protocol that combines the interaction between the nuclei and a periodically reset electron spin of an nirogen-vacancy center with local radio-frequency control of the nuclear spins. The final state of this protocol is independent of the initial preparation of the nuclei, is robust to external field fluctuations, and can be operated at room temperature. We show that a high-fidelity singlet pair of a 13C dimer in a nuclear bath in diamond can be generated under realistic experimental conditions. [ABSTRACT FROM AUTHOR]

Dynamic nuclear polarization (DNP) of molecules in a solution at room temperature has the potential to revolutionize nuclear magnetic resonance spectroscopy and imaging. The prevalent methods for achieving DNP in solutions are typically most effective in the regime of small interaction correlation times between the electron and nuclear spins, limiting the size of accessible molecules. To solve this limitation, we design a mechanism for DNP in the liquid phase that is applicable for large interaction correlation times. Importantly, while this mechanism makes use of a resonance condition similar to solid-state DNP, the polarization transfer is robust to a relatively large detuning from the resonance due to molecular motion. We combine this scheme with optically polarized nitrogen-vacancy (NV) center spins in nanodiamonds to design a setup that employs optical pumping and is therefore not limited by room temperature electron thermal polarization. We illustrate numerically the effectiveness of the model in a flow cell containing nanodiamonds immobilized in a hydrogel, polarizing flowing water molecules 4700-fold above thermal polarization in a magnetic field of 0.35 T, in volumes detectable by current NMR scanners. We report the detection and polarization of nuclear spins in diamond at room temperature by using a single nitrogen-vacancy (NV) center. We use Hartmann-Hahn double resonance to coherently enhance the signal from a single nuclear spin while decoupling from the noisy spin bath, which otherwise limits the detection sensitivity. As a proof of principle, we (i) observe coherent oscillations between the NV center and a weakly coupled nuclear spin and (ii) demonstrate nuclear-bath cooling, which prolongs the coherence time of the NV sensor by more than a factor of 5. Electrocatalytic CO 2 reduction to CO and formate can be coupled to gas fermentation with anaerobic microorganisms. In combination with a competing hydrogen evolution reaction in the cathode in aqueous medium, the in situ, electrocatalytic produced syngas components can be converted by an acetogenic bacterium, such as Clostridium ragsdalei , into acetate, ethanol, and 2,3-butanediol. In order to study the simultaneous conversion of CO, CO 2 , and formate together with H 2 with C. ragsdalei , fed-batch processes were conducted with continuous gassing using a fully controlled stirred tank bioreactor. Formate was added continuously, and various initial CO partial pressures (pCO 0 ) were applied. C. ragsdalei utilized CO as the favored substrate for growth and product formation, but below a partial pressure of 30 mbar CO in the bioreactor, a simultaneous CO 2 /H 2 conversion was observed. Formate supplementation enabled 20-50% higher growth rates independent of the partial pressure of CO and improved the acetate and 2,3-butanediol production. Finally, the reaction conditions were identified, allowing the parallel CO, CO 2 , formate, and H 2 consumption with C. ragsdalei at a limiting CO partial pressure below 30 mbar, pH 5.5, n = 1200 min -1 , and T = 32 °C. Thus, improved carbon and electron conversion is possible to establish efficient and sustainable processes with acetogenic bacteria, as shown in the example of C. ragsdalei.

The need for sustainable alternatives to conventional plastic mulches in agriculture has led to the development of various types of biodegradable mulches made from natural fibres and biopolymers to reduce environmental pollution and mitigate soil pollution caused by conventional plastic mulch usage. Degradation, impact on soil temperature and humidity, and weed suppression properties of needle-punched nonwoven mulches of different mass per unit area, made of jute, hemp, viscose, and PLA biopolymer, are investigated. Their biodegradation is determined by changes in the mulch properties (mass per unit area, thickness, air permeability, tensile properties, microscopic images, and FTIR analyses) during 300 days of exposure to the environmental conditions in the period from May 2022 to February 2023. The change in mass per unit area, thickness, air permeability, and tensile properties of nonwoven mulches did not show a tendency to degrade during exposure to environmental conditions. The microscopic and FTIR analysis showed the degradation of the fibres from the mulches during the exposure time to a certain extent. The environmental conditions influence the change in the dimensions of the mulches (shrinkage and expansion)-which impact periodically tested mass results per unit area-as well as their thickness and air permeability. The nonwoven mulches provide higher temperatures compared to bare soil, though not as high as those observed beneath traditional agricultural foil. When comparing the humidity in bare soil and soil covered by mulches during the plant growth period (June to October), it was found that soil humidity was higher beneath all mulches. The nonwoven mulches provide superior soil moisture retention compared to conventionally used agrofoil. Almost all nonwoven mulches effectively suppressed weed growth, except hemp mulches. The newly produced mulches have the potential to replace traditional agrofoil, offering improved conditions for plant growth, effective weed control, and faster degradation without causing harm to the environment.

This paper investigates the biodegradation properties of cellulose fibers and PLA biopolymer. For that purpose, hemp, jute, and sisal fibers as lignocellulose fibers; viscose fibers (CV) as regenerated cellulose; and polylactide (PLA) as biopolymer were buried in farmland soil for periods of 2, 4, 7, 9 and 11 days under controlled conditions. The influence of their biodegradation on the fiber mechanical properties, bacteria and fungi population, as well as on the soil quality were investigated. After exposure to microorganisms, analyses of the fibers' morphological (SEM), chemical (FTIR), and thermal (TGA) properties were conducted to achieve a comprehensive understanding of their biodegradability. The analysis concluded that lignin and pectin content have a greater impact on the biodegradation of hemp, jute, and sisal fibers than factors like crystallinity and degree of polymerization. The viscose fibers showed lower biodegradability despite their lower degree of polymerization, indicating a resistance to biodegradation due to the "skin" formed during the spinning process. PLA fibers experienced chemical hydrolysis and significant microbial attack, resulting in reduced tenacity. The acquired findings yield valuable insights into the biodegradability of the fibers, thereby facilitating the selection of appropriate fibers for the development of environmentally sustainable products. Notably, a literature review revealed a paucity of research on fiber biodegradability, underscoring the significance of the present study's contributions.

Woven fabrics are widely used for thermal protection due to their porosity, which provides thermal insulation and breathability. This research focuses on investigating the influential parameters in the thermal protective properties of layer interchanging double cloth, including the woven structure and varying yarn fineness. The properties affecting the protective properties and comfort of multilayered woven fabrics include the fabric thickness, fabric porosity, and air permeability. Darcy's law is applicable for determining the effective air permeability of woven fabrics. By understanding and controlling fabric porosity, it becomes possible to develop thermal protective clothing that combines improved comfort, cost-efficiency, and effectiveness. This study represents a novel approach for the clarification of airflow permeability behavior in complex structures of elastic multilayer woven fabrics using Darcy's law. This innovative approach expands the understanding of permeability in fabrics beyond single-layer fabrics with vertical pores or 3D fabrics used in resin injection processes.

The development and application of new types of fibres and their wider application influence the continuous invention of a more economical starching process, as one of the most expensive stages in the technological process of woven fabric production. For example, aramid fibres are increasingly used in clothing with effective protection from mechanical, thermal and abrasion exposure. Simultaneously, comfort and regulation of metabolic heat are extremely important, and this is achieved by using cotton woven fabrics. For such a woven fabrics to satisfy the protective properties and the possibility of all-day wear, fibre is needed, and thus a yarn, that will enable the efficient production of fine, light and comfortable protective woven fabrics. This paper investigates the influence of starching on the mechanical properties of aramid yarns and their comparison to cotton yarns of the same fineness. This will lead to knowledge about the efficiency and necessity of aramid yarn starching. The tests were carried out on an industrial and laboratory starching machine. According to the obtained results, the necessity and the improvement of the physical-mechanical properties of cotton and aramid yarns can be determined, both by industrial and laboratory starching. Finer yarn starched by the laboratory starching process achieves greater efficiency in the yarn's strength and resistance to wear, which indicates the need for starching aramid yarns, especially fineness 16.6 × 2 tex, but also finer ones.

Genomic imprinting is regulated by parental-specific DNA methylation of imprinting control regions (ICRs). Despite an identical DNA sequence, ICRs can exist in two distinct epigenetic states that are memorized throughout unlimited cell divisions and reset during germline formation. Here, we systematically study the genetic and epigenetic determinants of this epigenetic bistability. By iterative integration of ICRs and related DNA sequences to an ectopic location in the mouse genome, we first identify the DNA sequence features required for maintenance of epigenetic states in embryonic stem cells. The autonomous regulatory properties of ICRs further enabled us to create DNA-methylation-sensitive reporters and to screen for key components involved in regulating their epigenetic memory. Besides DNMT1, UHRF1 and ZFP57, we identify factors that prevent switching from methylated to unmethylated states and show that two of these candidates, ATF7IP and ZMYM2, are important for the stability of DNA and H3K9 methylation at ICRs in embryonic stem cells., (© 2022. The proportion of woven fabrics in the broad field of protective textiles is extremely high. By various procedures (surface treatments, fabric lamination, composite production), fabric properties that meet the requirements defined by standards are achieved. However, simultaneously, these procedures cause negative effects in the form of fabric thickness, stiffness, impermeability, non-breathability, and thus, discomfort. Therefore, there are valid and justified reasons to approach the design process of making such woven fabrics using more complex construction solutions-layer interchanging double cloth. In addition, by applying fibres with integrated desired properties and other structural fabric parameters, it is possible to influence the achievement of the properties of multi-purpose multilayer fabrics for protection, which is the aim of this research. The application and combination of aramid and modacrylic/cotton fibres and use of different yarn fineness resulted in different intensities of protection. The correlative values of mentioned parameters and thermal and thermo-physiological properties indicate their strong connection, and thus the effectiveness of the developed woven fabric samples., Competing Interests: The authors declare no conflict of interest.
