Homeostasis preservation is essential for animal survival, and any event that causes a disturbance in homeostasis is defined as a stressor. Here, we aimed to evaluate the effect of scratch brushes and stages as an environmental enrichment to alleviate stress in dairy goats. Twenty-four mixed-breed goats were divided into two groups according to common physiological conditions in breeding farms: milking and dry (milk-producing and non-milk-producing, respectively). Ten days after exposure to environmental enrichment treatment or not (control), blood was sampled. Following the enrichment, we observed a reduction in reactive oxidative stress metabolites, advanced glycation end products (AGEs), and their binding protein (transferrin) in the dry goats, as determined by an ELISA. In contrast, no change in AGEs, along with an increase in transferrin levels, was observed in the milking goats. Moreover, oxytocin levels decreased in the dry and increased in the milking goats, while serotonin levels increased in the dry and remained unchanged in the milking goats. Additionally, gene expression of the cytokines, IL-6 and IL-1ß, and anti-oxidative proteins, lysozyme and transferrin (in peripheral blood leukocytes), as determined by qPCR, presented the same pattern: down-regulation in the dry or up-regulation in the milking goats. In conclusion, a reliable methodology was developed for measuring husbandry stress in goats and to improve dairy goats’ husbandry practice. Current environmental enrichment produced different responsiveness in goats correlated to their physiological status: beneficial effect in dry goats, detrimental effect in milking goats.

This study evaluated the potential of low-glycoalkaloid potato haulm (LGPH) as a high-quality feed for ruminants. The low-glycoalkaloid potato plants were grown in a net house following standard agricultural practices. Dehaulming was performed a fortnight before the harvest of the tubers, as commercially practiced. Four healthy female lambs (aged 4 months) were employed in a 4×4 Latin square feeding experiment design. The lambs were fed with either a maintenance diet consisting of 73% roughage (control), or treatment diets with supplementation of 10% (P10), 15% (P15), and 20% (P20) LGPH on a dry matter (DM) basis for a period of 21 days, including 14 days of adaption and 7 days of sampling. Refusals and feces were collected for 6 days and pooled followed by a 24 h urine collection on the 7th day. On the 6th day, an hour after morning feeding, blood, and rumen liquid samples were collected. All the samples were analysed, and the data generated were analysed using ANOVA with diet and period as fixed effects and sheep as a random effect. Orthogonal contrasts were used to detect linear and quadratic effects of LGPH in the diet. Linear or polynomial equations were produced to extract the nutrient digestibility and metabolizable energy (ME) of LGPH. No toxicological clinical signs were observed, and the haematology profiles were normal throughout the trial. LGPH did not affect the daily intake of nutrients and energy. However, the apparent digestibility of DM, organic matter (OM), crude protein (CP), and gross energy increased (P= 0.01) linearly (P< 0.01) with supplementation of LGPH in the diet. The inclusion of LGPH had a quadratic effect on neutral detergent fiber (NDF) digestibility (P= 0.027), reaching a peak of 54.8 % at 12.3 % LGPH supplement on a DM basis. The N retention in lambs fed with the P20 diet was nearly twice that of the control diet (P= 0.032; 19.1 vs. 10.2 g/d). The computed digestibility values of DM, OM, CP, and NDF for LGPH were 76.1, 79.7, 84.2, and 72.1 %, respectively, and ME was 2.62 Mcal/kg DM. Conclusively, by significantly reducing the glycoalkaloids’ content in potato haulm, we can safely repurpose the currently wasted foliage as a new source of high-quality roughage feed for ruminants, equivalent to alfalfa hay, without imposing any additional environmental burden. However, further research is necessary to assess impact of LGPH on growth performance, and milk productivity before practical application on commercial farms.

Teff (Eragrostis tef) is a drought-tolerant, multi-harvest, high-quality summer forage crop. We conducted a study aiming at assessing the effect of replacing wheat hay with teff hay in diets on the feed intake, digestibility, and lactational performance of dairy cows. Thirty-four multiparous (≥3rd parity) Israeli Holstein Friesian dairy cows averaging (± SD) 182 days in milk ± 8 days in milk, 45 kg/d ± 4.8 kg/d of milk yield, and a body weight of 647.1 kg ± 51 kg at the beginning of the study were recruited to a 6-week feeding trial. Cows were randomly divided into two balanced groups based on parity, days in milk, and milk yield. Cows were subjected to two low-roughage dietary treatments (~30% roughage): a control wheat hay-based diet and a teff hay-based diet. Production performances, dry matter intake, and nutrient digestibility were measured. Milk samples were analyzed for their composition and fatty acids profile. Blood samples were used to measure metabolite concentrations. The statistical model included fixed effects of dietary treatments, time, and random effects of cows nested in treatment. Production data and feed intake were analyzed as repeated measures using a covariance structure. Dietary treatments did not affect dry matter intake (26 kg/d). However, the teff-fed cows demonstrated higher crude protein digestibility than control cows (61.9% vs. 59.2%). Dietary teff inclusion increased milk yield by 1.5 kg/d. Polyunsaturated fatty acids and omega-3 fatty acids profiles in milk were greater in the teff cows than in the control cows (4.77 g/100 g vs. 4.36 g/100 g and 3.71 g/100 g vs. 3.43 g/100 g, respectively). Non-esterified fatty acids, beta-hydroxybutyrate, and blood urea nitrogen concentrations in circulation were higher in the control group than in the teff group. The acetic-to-propionic-acid ratio in the rumen fluid was higher in control cows than in teff cows (2.90 vs. 2.43). However, the ruminal ammonia-N concentration was higher in the teff cows than in the control cows (18.5 mg/dL vs. 15.8 mg/dL). In conclusion, teff hay inclusion in the rations of high-producing dairy cows increased milk yield, which could be attributed to improved crude protein digestibility and energy partition to production.

Antimicrobial drugs and coccidiostat compounds are commonly used in poultry farming. These compounds are subsequently excreted and released into the environment via broiler litter (BL) and can re-enter the food chain as fertilizer or animal feed. Such residue in animal feed can encourage the appearance of antibiotic-resistant bacteria as well as toxicity. Most analytical methods used to identify and quantitate these drug residues are traditional, and are specific to some antimicrobials and present limitations in assessing complex matrixes like BL. The aim of this study was to develop a multi-residue analytic method for assessing 30 antimicrobial drugs and coccidiostats associated with BL. We investigated the presence and the effects of biotic stack treatment on the degradation of drug residue in BL. Liquid-liquid extraction (LLE) and solid phase extraction (SPE) were replaced by Quick, Easy, Cheap, Effective, Rugged, and Safe (QuEChERS) clean-up steps and detected by liquid chromatography mass spectrometry (LC/MS/MS). Results show that a wide spectrum of residues were detected from 0.4 to 8.9 mg kg−1. Following lab-scale stacking treatment, tilmicosin and eight coccidiostats persisted in BL (26–100%). This research supports the need for better understanding, regulation, and management of the use of BL that might carry a high risk of residue drugs.

The global demand for animal-based products is rising in the face of dwindling feed resources, and yet a huge pool of agro-industrial by-products (AIBPs) are generated, underutilized, and improperly deposited to landfills leading to environmental pollution. Ruminants have a special microbiome that can bioprocess and convert human inedible fibrous material into meat and milk, which appears as a great opportunity to simultaneously reduce pollution while promoting food security.In this study, we collected 15 domestically produced AIBPs from various regions of Israel during both winter and summer seasons to examine their potential as ruminant feed alternatives. We evaluated their storability, nutritional composition, and in vitro digestibility and performed a hierarchical cluster analysis to categorize them based on their distinctive nutritional characteristics. Among the 15 AIBPs, 8 have rich essential nutrients, and minerals, and have excellent in vitro digestibility, but they have less than 6 days of storability and develop off-odours. Out of 15 AIBPs; 8 have low dry matter (DM) content, ranging from 4.7% to 30.45% while the remaining 7 AIBPs have high DM, ranging from 50.6% to 98.6%. The high crude protein (CP) category included 6 AIBPs with CP ranging from 19.7% in beer pulp to 32.1% in jojoba cake. Starch content was high in 3 AIBPs ranging from 33.7% in timorim mix to 65.2% in Irish potato culls. Considerable crude fat content was reported in 4 AIBPs, the highest being yoghurt waste with 42.8%. In terms of neutral detergent fiber (NDF), 5 AIBPs had low NDF content ranging from 0% to 14.1%; 5 AIBPs had moderate concentration ranging from 34.3% to 50.7%, and 5 AIBPs had high levels between 66.6% and 82.8%. Interestingly, 10/15 AIBPs had medium to high in vitro dry matter digestibility (IVDMD). This study, therefore, suggests that recycling AIPBs for livestock nutrition has enormous potential that is still underutilized and offers excellent ways to gain socioeconomic and environmental benefits by expanding animal feed resources and reducing feed-food competition, and landfill burden. However, additional studies are necessary to focus on affordable storage technology to prolong the storability of AIBPs and feeding trials to determine the productive performance of livestock fed an AIBPs-based diet.

Optimal embryonic development and growth of meat-type chickens (broilers) rely on incubation conditions (oxygen, heat, and humidity), on nutrients and on energy resources within the egg. Throughout incubation and according to the embryo’s energy balance, the main energy storage molecules (creatine and glycogen) are continuously utilized and synthesized, mainly in the embryonic liver, breast muscle, and the extraembryonic yolk sac (YS) tissue. During the last phase of incubation, as the embryo nears hatching, dynamic changes in energy metabolism occur. These changes may affect embryonic survival, hatchlings’ uniformity, quality and post hatch performance of broilers, hence, being of great importance to poultry production. Here, we followed the dynamics of creatine and glycogen from embryonic day (E) 11 until hatch and up to chick placement at the farm. We showed that creatine is stored mainly in the breast muscle while glycogen is stored mainly in the YS tissue. Analysis of creatine synthesis genes revealed their expression in the liver, kidney, YS tissue and in the breast muscle, suggesting a full synthesis capacity in these tissues. Expression analysis of genes involved in gluconeogenesis, glycogenesis, and glycogenolysis, revealed that glycogen metabolism is most active in the liver. Nevertheless, due to the relatively large size of the breast muscle and YS tissue, their contribution to glycogen metabolism in embryos is valuable. Towards hatch, post E19, creatine levels in all tissues increased while glycogen levels dramatically decreased and reached low levels at hatch and at chick placement. This proves the utmost importance of creatine in energy supply to late-term embryos and hatchlings.

Wheat and corn silages are widely used as ruminant feed in Israel due to their availability and cost-effectiveness. To ensure long-term preservation without compromising nutritional quality, effective methods must be employed. The inclusion of additives during harvest and ensiling can enhance efficiency and address preservation challenges. In the current study, the effects of microbial inoculum (MI) and urea on the chemical composition, amino acid profiles, aerobic stability, and in vitro digestibility of wheat and corn silages were investigated. Samples of wheat and corn were subjected to four treatments: control, MI, urea and a combination of MI + urea. The treatments were ensiled in anaerobic conditions and opened after 1, 7, 14 or 28 days. The results showed that additives improved the quality parameters of wheat and corn silages. The inclusion of MI produced the most aerobically stable silages. The inclusion of urea in silages decreased aerobic stability. Additives improved in vitro cell wall carbohydrates’ digestibility in both silages and was the best when MI was combined with urea. These results imply that additives could be incorporated in silages to enhance their nutritional value, aerobic stability and digestibility. Nonetheless, increased CP content with additives was not accompanied with a parallel increase in amino acids’ content in corn silage.

The use of antimicrobial drugs and coccidiostats in poultry farming is widespread, with a significant proportion of these drugs being excreted and released into the environment. The residues of such drugs in poultry litter (PL) can result in the development of antibiotic-resistant bacteria. The impact of different biotic treatments (aerobic, anaerobic, and stacking) on broiler litter (BL) before its use as animal feed has not been studied extensively, nor have the differences between antimicrobial-dependent and independent broiler farms been investigated. This study aimed to determine the resistant bacteria in BL used as ruminant feed before and after litter treatment. The results show that the most resistant bacteria before BL treatment were the Enterococcus species. This study also found that the quantity of amoxicillin-resistant Enterococcus detected in samples from antimicrobial-dependent farms was significantly higher than in those from antimicrobial-independent farms. Additionally, 14% of bacteria were multi-resistant to tetracycline, sulfafurazole, and erythromycin in antimicrobial-independent farm litters, significantly lower than those measured in antimicrobial-dependent broiler farm litter. This study highlights the importance of better understanding, regulating, managing, and using animal waste appropriately to reduce the number of antibiotic-resistant bacteria and minimize the use of antimicrobials that carry high risks for animals, humans, and the environment

This study investigated the effect of dietary flavour supplements on the preference, feed efficiency and expression of the sweet taste receptor family 1 members 2 and 3 (T1R2 + T1R3), and sodium-glucose linked transporter 1 (SGLT1) genes in the lambs’ small intestines. Eight, five-month-old, Israeli crossbred Assaf lambs were offered 16 different non-nutritive commercial flavours in rolled barley and ground corn. Capsicum and sucram were the most preferred non-aroma flavours (p = 0.020), while milky (p < 0.001) was the most preferred powder-aroma flavour. For the metabolic and relative gene expression study, eight lambs were randomly assigned to either sucram, capsicum, a mix containing sucram and capsicum at 1:1 ratio or no flavour for control in a 4 × 2 cross-over design. The total collection of urine (females only), faeces and refusals was carried out, and T1R2, T1R3 and SGLT1 relative gene expression evaluated from the proximal jejunum biopsies. Flavour had no significant effect on the feed intake (p = 0.934), but capsicum increased the average daily weight gain per metabolic body weight (p = 0.049). The T1R3 gene was expressed highest in the mix treatment (1.7; p = 0.005). Collectively, our findings indicate that flavours can be used to motivate feed acceptance and improve the weight gain in lambs.

Tef is known as a multi-harvest crop with high production capacity and outstanding fodder quality. Hence, our overall goal is to develop tef as a new multi-harvest summer crop that can maintain high-quality feed and contribute to both field crops and the livestock industry in Israel. In this study, we aimed to evaluate the ability to preserve tef as silage. Four tef genotypes grown under well-watered (100%) and water-limited (75%) irrigation regimes were harvested at grain filling stage and ensiled with either no additives (control, CTL), or with heterofermentative inoculum (HI), molasses (MOL), and both MOL + HI. Our results showed for the first time that tef could be ensiled, although water-soluble carbohydrates (WSC) were lower than those in corn, “the perfect ensiling crop”. Most of the tef silage qualitative parameters were better at water-limited irrigation. Additives HI or MOL or MOL + HI also improved silage parameters, e.g., lowered pH and ammonia nitrogen content, but increased in vitro dry matter digestibility, lactic acid and crude protein content, and lactic acid bacteria counts of tef silage. The current results imply increasing the diversity of local ruminant fodder crops, ensuring high-quality feed supply during the summer.

Leptin, like estrogen, is one of the endo/paracrine factors, which are synthesized in and secreted from mature adipocytes. The roles of the mammary fat pad and mammary adipocytes in the initiation of lactation are not clear. In this study, we showed that combination of prolactin, leptin and estrogen elevated the expression of the milk protein beta-lactoglobulin. We also showed that after prolactin stimulate the secretion of leptin from the mammary fat, leptin upregulated the expression of estrogen receptor alpha in the mammary epithelial cells. Also, prolactin affected aromatase mRNA expression in the bovine mammary fat and we demonstrated that leptin and prolactin can affect cholesterol secretion from explants in culture to the medium. Therefore, we suggest that prolactin initiates estrogen expression (as represented by aromatase mRNA) in the mammary fat pad, whereas leptin stimulates estrogen receptor alpha expression in the mammary epithelial cells. We hypothesize that leptin and estrogen, secreted from the mammary fat regulate lactation after stimulation of prolactin.

The role the mammary epithelial circadian clock plays in gland development and lactation is unknown. We hypothesized that mammary epithelial clocks function to regulate mammogenesis and lactogenesis, and propose the core clock transcription factor BMAL1:CLOCK regulates genes that control mammary epithelial development and milk synthesis. Our objective was to identify transcriptional targets of BMAL1 in undifferentiated (UNDIFF) and lactogen differentiated (DIFF) mammary epithelial cells (HC11) using ChIP-seq. Ensembl gene IDs with the nearest transcriptional start site to ChIP-seq peaks were explored as potential targets, and represented 846 protein coding genes common to UNDIFF and DIFF cells and 2773 unique to DIFF samples. Genes with overlapping peaks between samples (1343) enriched cell-cell adhesion, membrane transporters and lipid metabolism categories. Short-day photoperiod (SDPP) during the dry period increases milk production compared to long-day photoperiod (LDPP) in goats. Photoperiod information is sent to the master clock in the suprachiasmatic nuclei (SCN), which send temporal information to peripheral clocks located in every tissue of the body. The versatile-peroxidase (VP) encoded by mnp4 is one of the nine members of the manganese-peroxidase (MnP) gene family that constitutes part of the ligninolytic system of the white-rot basidiomycete Pleurotus ostreatus (oyster mushroom). VP enzymes exhibit dual activity on a wide range of substrates. As Mn(2+) supplement to P. ostreatus cultures results in enhanced degradation of recalcitrant compounds and lignin, we examined the effect of Mn(2+) on the expression profile of the MnP gene family. In P. ostreatus (monokaryon PC9), mnp4 was found to be the predominantly expressed mnp in Mn(2+)-deficient media, whereas strongly repressed (to approximately 1%) in Mn(2+)-supplemented media. Accordingly, in-vitro Mn(2+)-independent activity was found to be negligible. We tested whether release of mnp4 from Mn(2+) repression alters the activity of the ligninolytic system. A transformant over-expressing mnp4 (designated OEmnp4) under the control of the β-tubulin promoter was produced. Now, despite the presence of Mn(2+) in the medium, OEmnp4 produced mnp4 transcript as well as VP activity as early as 4 days after inoculation. The level of expression was constant throughout 10 days of incubation (about 0.4-fold relative to β-tubulin) and the activity was comparable to the typical activity of PC9 in Mn(2+)-deficient media. In-vivo decolorization of the azo dyes Orange II, Reactive Black 5, and Amaranth by OEmnp4 preceded that of PC9. OEmnp4 and PC9 were grown for 2 weeks under solid-state fermentation conditions on cotton stalks as a lignocellulosic substrate. [(14)C]-lignin mineralization, in-vitro dry matter digestibility, and neutral detergent fiber digestibility were found to be significantly higher (about 25%) in OEmnp4-fermented substrate, relative to PC9. We conclude that releasing Mn(2+) suppression of VP4 by over-expression of the mnp4 gene in P. ostreatus improved its ligninolytic functionality.
