During recent decades, ultrasmall inorganic nanoparticles have attracted considerable interest due to their favorable biodistribution, pharmacokinetics and theranostic properties. In particular, AGuIX nanoparticles made of polysiloxane and gadolinium chelates were successfully translated to the clinics. In an aqueous medium, these nanoparticles are in dynamic equilibrium with polysiloxane fragments due to the hydrolysis of Si–O–Si bonds. Abstract Affinity monolith columns of 375 nL (effective length 8.5 cm, internal diameter 75 μm) were developed for protein-ligand affinity investigations needing only 3 μg of human serum albumin (HSA). To promote specific interactions and avoid non-specific ones, different combinations of monolithic supports and bio-functionalization pathways were evaluated. Silica and glycidylmethacrylate based monoliths were in-situ synthesized and grafted with HSA. Two direct grafting methods epoxy-amine and Schiff Base plus the streptavidin-biotin method were compared. The columns were evaluated by frontal analysis with ligands of known affinity for HSA. It is shown that a classical capillary electrophoresis instrument equipped with an external pressure device can be used to do weak affinity chromatography at low pressure (less than 1.2 MPa) in a fully automated way and with very low reagent consumption. The grafting pathways were compared in terms of (i) total and active amounts of immobilized protein, (ii) non-specific interactions, (iii) protein denaturation. According to these criteria, the organic monoliths combined with the streptavidin-biotin approach provided the best results. This immobilization pathway led to the highest active protein content (40 pmol of HSA per 8.5-cm column) with less than 10% non-specific interactions and 84% protein activity. The target grafting step lasts only 10 min and is UV-monitored, the UV breakthrough curve giving the exact amount of bound protein. This novel approach was validated by K d measurements of 3 known ligands of HSA. Streptavidin generic monolith columns could be stored at 4 °C for 3 months maintaining activity. μg of a biotin modified sensitive protein could be attached to a stable streptavidin monolith for immediate interaction studies avoiding stability problems. This development was subsequently extended to another protein of higher pharmaceutical interest: the N-terminal domain of HSP90. Boronate affinity chromatography (BAC) is commonly used in sample preparation techniques for its specific selectivity for cis-diol-containing compounds. Various chromatographic materials based on phenyl-boronic acid (PBA) functionalization have been designed. This review aims to present the versatility of the underlying BAC mechanisms. The PBA moiety presents secondary/complementary/interfering interactions, i.e., hydrophobic, electrostatic and/or hydrogen-bonding interactions. The comprehensive understandings of these mechanisms are very helpful to manipulate separation selectivity, which is a key feature in biomolecular sample treatment. Each BAC interaction is reviewed and illustrated by specific examples. Special attention is given to the underlying role of PBA supports for sugars, glycans and glycoconjugates recognition. Abstract: There is a need to determine time‐weighted average concentrations of polar contaminants such as pesticides by passive sampling in environmental waters. Calibration data for silicone rubber‐based passive samplers are lacking for this class of compounds. The calibration data, sampling rate (Rs), and partition coefficient between silicone rubber and water (Ksw) were precisely determined for 23 pesticides and 13 candidate performance reference compounds (PRCs) in a laboratory calibration system over 14 d for 2 water flow velocities, 5 and 20 cm s−1. The results showed that an in situ exposure duration of 7 d left a silicone rubber rod passive sampler configuration in the linear or curvilinear uptake period for 19 of the pesticides studied. A change in the transport mechanism from polymer control to water boundary layer control was observed for pesticides with a log Ksw of approximately 3.3. The PRC candidates were not fully relevant to correct the impact of water flow velocity on Rs. We therefore propose an alternative method based on an overall resistance to mass transfer model to adjust Rs from laboratory experiments to in situ hydrodynamic conditions. We estimated diffusion coefficients (Ds) and thickness of water boundary layer (δw) as adjustable model parameters. Log Ds values ranged from −12.13 to −10.07 m2 s−1. Passive sampling techniques have been developed as an alternative method for in situ integrative monitoring of trace levels of neutral pesticides in environmental waters. The objective of this work was to develop a new receiving phase for pesticides with a wide range of polarities in a single step. We describe the development of three new composite silicone rubbers, combining polydimethylsiloxane mechanical and sorption properties with solid-phase extraction sorbents, prepared as a receiving phase for passive sampling. A composite silicone rubber composed of polydimethylsiloxane/poly(divinylbenzene-co- N-vinylpyrrolidone) was selected by batch experiments for its high sorption properties for pesticides with octanol-water partition coefficients ranging from 2.3 to 5.5. We named this composite material 'Polar/Apolar Composite Silicone Rubber'. A structural study by scanning electron microscopy confirmed the homogeneous dispersion of the sorbent particles and the encapsulation of particles within the polydimethylsiloxane matrix. We also demonstrate that this composite material is resistant to common solvents used for the back-extraction of analytes and has a maximal resistance temperature of 350°C. Therefore, the characteristics of the 'Polar/Apolar Composite Silicone Rubber' meet most of the criteria for use as a receiving phase for the passive sampling of pesticides. [ABSTRACT FROM AUTHOR]

Hydrophilic interaction LC was investigated in hydro-organic and nonaqueous elution modes on a titania column by using a set of N-methyl xanthines as neutral polar probes. To get information regarding the mechanisms that are behind the discrimination of these analytes in hydrophilic interaction, we focused our study on the type and amount of organic modifier as a critical yet rarely explored mobile phase parameter. Several alcohols such as methanol, ethanol, and isopropanol were studied as substitutes to acetonitrile in hydro-organic elution mode. Compared to silica, the investigation of the eluotropic series of these alcohols on titania highlighted a different implication in the retention mechanism of the xanthine derivatives. At low amounts of protic solvents, the adsorption mainly characterized the retention of analytes on bare silica; whereas mixed interactions including adsorption and ligand exchange were identified on native titania. To investigate the peculiar behavior of alcohols on the metal oxide, methanol, ethanol, and ethylene glycol were tested in replacement of water in polar-organic elution mode. Distinctive effects on the chromatographic retention and selectivity of xanthines were noticed for the dihydric alcohol, which was found to be a stronger eluting component than water on titania. In this work, the low cost synthesis of polydimethylsiloxane (PDMS) rods is described, and the performances of this new passive sampling device (in laboratory and in situ) are compared to the passive stir bar sorptive extraction (SBSE) for the monitoring of pesticides from different classes (herbicides, insecticides and fungicides) in surface waters. The influence of synthesis parameters of PDMS rods (i.e., heating temperature, heating time and relative amount of curing agent) were assessed regarding their efficiency for the extraction of the target pesticides through a Hadamard's experimental design. This allowed the determination of the effect of the three parameters on the sorption of pesticides within four experiments. Thus, specific conditions were selected for the synthesis of the PDMS rods (heating at 80 °C for 2 h with 10% of curing agent). Laboratory experiments led to similar to lower extraction recovery in the PDMS rods in comparison with passive SBSE, depending on the pesticide. The in situ application demonstrated the efficiency of the PDMS rods for the passive sampling of the target pesticides in river water, although lower amounts of pesticides were recovered in comparison with passive SBSE. So, these very low cost PDMS rods could be used as an alternative to passive SBSE for large-scale monitoring campaigns. [ABSTRACT FROM AUTHOR]

An efficient and reproducible photopolymerized poly(glycidyl methacrylate- co-ethylene dimethacrylate) was synthesized in Teflon-coated fused-silica capillaries (100 μm id) and functionalized by reaction of triethylamine with reactive epoxy groups. We report here the successful transfer of a standard polymerization mixture optimized for the thermally initiated synthesis of glycidyl-based monolith to photo-induced polymerization. The monolith obtained after optimization of the photo-initiation conditions was characterized in reverse-phase chromatography evaluating its suitability in terms of efficiency, retention and hydrodynamic permeability. Reproducibility of the photo-induced procedure was satisfactory with RSD below 6% for retention and efficiency and slightly higher for hydrodynamic permeability (12%). The functionalized generic support was then used in nano-ion-exchange chromatography. Efficiencies up to 75 000 plates/m, ion-exchange capacity of 8 nano-equivalents/cm of monolithic column, with a combination of a satisfactory hydrodynamic permeability allowed to perform fast separations of five inorganic anions in <3 min maintaining baseline resolution. new sol-gel protocol was designed and optimized to produce titanium-dioxide-based columns within confined geometries such as monolithic capillary columns and porous-layer open-tubular columns. A surface pre-treatment of the capillary enabled an efficient anchorage of the monolith to the silica capillary wall during the synthesis. The monolith was further synthesized from a solution containing titanium n-propoxide, hydrochloric acid, N-methylformamide, water, and poly(ethylene oxide) as pore template. The chromatographic application of capillary titania-based columns was demonstrated with the separation of a set of phosphorylated nucleotides as probe molecules using aqueous normal-phase liquid chromatography conditions. Capillary titania monoliths offered a compromise between the high permeability and the important loading capacity needed to potentially achieve miniaturized sample preparations. The specificity of the miniaturized titania monolithic support is illustrated with the specific enrichment of 5′-adenosine mono-phosphate. The monolithic column offered a ten times higher loading capacity of 5′-adenosine mono-phosphate compared with that of the capillary titania porous-layer open-tubular geometry. In recent years, metal oxides such as titania have been commercially available as chromatographic beds that can potentially be used to achieve novel separations of polar compounds. For example β blockers, which are more often encountered in environmental sciences, have a wide range of polarity, and their basic character leads to difficult sample treatment and separation on conventional silica-based sorbents. The contribution of titania to the selective analysis of nine β blockers was evaluated in terms of retention mechanisms observed in hydrophilic interaction LC using acetonitrile/water mobile phases with various additives. The mobile phase additives enabled to control the β blocker charge as well as the titania surface charge. Depending on their respective ionic state, various retention mechanisms were identified at low water contents (<40%), including mainly adsorption mixed with hydrophilic interaction LC partition, ion exchange and ion exclusion. A nano-gravimetric detector (NGD) for gas chromatography is based on a nanoelectromechanical array of adsorbent-coated resonating double clamped beams. NGD is a concentration-sensitive detector and its sensitivity is analyte-dependent based on the affinity of the analyte with the porous layer coated on the NEMS surface. This affinity is also strongly related to the NGD temperature (NGD working temperature can be dynamically set up from 40 to 220 °C), so the sensitivity can be tuned through temperature detector control. An adsorption-desorption model was set up to characterize the NGD response on a large set of n -alkanes from C 10 to C 22 at different NGD temperatures. For fast identification of petroleum mixture based on chromatogram fingerprint, a general strategy for NGD temperature program design was developed leading to a constant relative response factor between 0.96 and 1.03 for all the alkanes, and then chromatograms are very similar to those obtained with a flame ionization detector (FID). The analysis of a real petroleum fluid was also performed and compared to FID results: quantitative results obtained for all the analytes were satisfactory according to precision (<5%) and accuracy (average relative error = 4.3%). Based on such temperature control strategy, NGD sensitivity and the dynamic linear range can be adjusted and detection limits at a picogram level can be easily achieved for all n -alkanes.

Different synthesis routes have been implemented to prepare macroporous monoliths with vinyl pendant groups and micrometric skeletons and through-pore sizes. A standard process combining the synthesis of a widely used (methyltrimethoxysilane/tetramethoxysilane) (MTMS/TMOS) hybrid silica monolith and the postsilanization with vinyltrimethoxysilane (VTMS) was used as reference material (Vgr-MTMS). An alternative "one-pot" procedure was used to obtain vinylized hybrid monoliths. Two VTMS/TMOS hybrid based monoliths were successfully prepared starting from 20% (w) and 80% (w/w) of VTMS, respectively, called 20-VTMS and 80-VTMS. Monoliths were characterized by SEM, nitrogen-adsorption isotherm, and (29)Si MAS NMR spectroscopy. One-pot synthesis allowed to obtain higher vinyl contents (15.9 and 61.5 mol % of Si atoms bonded to vinyl groups respectively for 20-VTMS and 80-VTMS) than for the postgrafted one (7.1%). Accessibility of vinyl groups was determined by the extent of bromination reactions followed by FTIR-ATR spectroscopy. Bromination with reaction yields were higher than 80% for all materials (80%, 85%, and 100% for 80-VTMS, 20-VTMS, and Vgr-MTMS respectively), with no diffusion issues The chemical reactivity of the pendant vinyl groups was investigated through radical-mediated thiol-ene reaction and radical-initiated bisulfite addition. Reaction yields for the two VTMS hybrid monoliths were quite lower (4-6%) than those obtained (about 50%) for the Vgr-MTMS monolith. The difference in reactivity was attributed to the steric hindrance of the vinyl moieties at the surface of hybrid materials. However, the lower reactivity of vinyl groups is compensated by their considerably higher surface density. Thus, hybrid monoliths are advantageous over their grafted counterparts, due to their higher hydrolytic stability and to the greater simplicity of the one-pot process. A new method for monolith synthesis and anchoring inside cyclic olefin copolymer (COC) microchannels in a single step is proposed. It is shown that type I photoinitiators, typically used in a polymerization mixture to generate free radicals during monolith synthesis, can simultaneously act as type II photoinitiators and react with the plastic surface through hydrogen abstraction. This mechanism is used to "photograft" poly(ethylene glycol) methacrylate (PEGMA) on COC surfaces. Contact angle measurements were used to observe the changes in surface hydrophilicity when increasing initiator concentration and irradiation duration. The ability of type I photoinitiators to synthesize and anchor a monolith inside COC microchannels in a single step was proved through SEM observations. Different concentrations of photoinitiators were tried. Finally, electrochromatographic separations of polycyclic aromatic hydrocarbons were realized to illustrate the beneficial effect of anchoring on chromatographic performances. The versatility of the method was demonstrated with two widely used photoinitiators: benzoin methyl ether (BME) and azobisisobutyronitrile (AIBN).

Fused-silica capillaries were packed with Zirchrom-PBD stationary phase for application in CEC, nanoLC and pseudoelectrochromatography (PEC). Acido-basic properties of zirconia can be used to control the EOF even if the zirconia particles were coated by polybutadiene. As for native zirconia, the EOF is pH-dependent and the pI is close to pH 5. The mixed-mode pressure-voltage technique induced a modulation of the mobile-phase velocity as well as an electrophoretic migration of the solutes in order to improve the resolution of the separation. A significant increase of the flow appeared when both hydrodynamic and EOFs were in the same direction. But an important reduction of the electroosmotic velocity was observed when the hydrodynamic flow and EOF were opposed in Zirchrom-PBD columns. This behaviour has been observed at high or low pH on several columns. Separations of neutral and charged compounds have been performed with these columns in PEC mode.

The electroosmotic flow created in zirconia-modified capillaries has been previously investigated. In this paper, we compared the electroosmotic data set with streaming current measurements and we related all these data through zeta-potential. Streaming current measurements give an excellent indication on the direction and the value of the electroosmotic mobility of an electrolyte/capillary system for a large set of experimental conditions: 2 < pH < 12, 0 < ACN < 80 %, 10(-4) M < [SO(2- )4 ] < 4 x 10(-2) M. A good correlation between zeta-potential from streaming current measurements and zeta-potential from electroosmotic mobility measurements was observed (r2 = 0.95). However, the values obtained from streaming current were always slightly lower than the one calculated from electroosmotic mobility (slope = 0.86, sigma = 0.06). In zirconia-coated capillaries the zeta-potential can be tuned from -50 to +100 mV depending on the composition of the electrolyte., (Copyright 2004 WILEY-VCH Verlag GmbH & Co.)
