Abstract

In this work, Bi.sub.2O.sub.3 nanoparticles (NPs) were used for the construction of a modified carbon paste electrode for the non-enzymatic determination of uric acid in human urine samples by differential pulse voltammetry as the quantitative analytical technique which was optimized using a Box-Behnken design. Bi.sub.2O.sub.3-NPs/CPE sensor showed an excellent performance for uric acid oxidation presenting two linear ranges of 5-300 and 300-1000 [mu]M. The good reproducibility, repeatability, low interference in the presence of other chemical species, and statistical evaluation demonstrated that the proposed methodology is as reliable as the enzymatic spectrophotometric technique for the quantification of uric acid in human urine samples. 12. Interfacial host–guest complexation for inverted perovskite solar cells.

Abstract

Perovskite solar cells have demonstrated exceptional development over the past decade, but their stability remains a challenge toward the application of this technology. Several strategies have been used to address this, and the use of host–guest complexation has recently attracted more interest. However, this approach has primarily been exploited in conventional perovskite solar cells based on n-i-p architectures, while its use in inverted p-i-n devices remains unexplored. Herein, we employ representative crown ether, dibenzo-24-crown-8, for interfacial host–guest complexation in inverted perovskite solar cells based on methylammonium and methylammonium-free formamidinium-cesium halide perovskite compositions. Upon post-treatment of the perovskite films, we observed nanostructures on the surface that were associated with the reduced amount of trap states at the interface with the electron transport layer. As a result, we demonstrate improved efficiencies and operational stabilities following ISOS-D-2I and ISOS-L-2I protocols, demonstrating the viability of this approach to advance device stability. Abstract

Trypanosoma brucei is the protozoan parasite responsible for sleeping sickness, a lethal vector-borne disease. T. brucei has a single flagellum (cilium) that plays critical roles in transmission and pathogenesis. An emerging concept is that the flagellum is organized into subdomains, each having specialized composition and function. The overall flagellum proteome has been well studied, but a critical knowledge gap is the protein composition of individual subdomains. We have tested whether APEX-based proximity proteomics could be used to examine the protein composition of T. brucei flagellum subdomains. As APEX-based labeling has not previously been described in T. brucei, we first fused APEX2 to the DRC1 subunit of the nexin-dynein regulatory complex, a well-characterized axonemal complex. We found that DRC1-APEX2 directs flagellum-specific biotinylation, and purification of biotinylated proteins yields a DRC1 "proximity proteome" having good overlap with published proteomes obtained from purified axonemes. Having validated the use of APEX2 in T. brucei, we next attempted to distinguish flagellar subdomains by fusing APEX2 to a flagellar membrane protein that is restricted to the flagellum tip, AC1, and another one that is excluded from the tip, FS179. Fluorescence microscopy demonstrated subdomain-specific biotinylation, and principal-component analysis showed distinct profiles between AC1-APEX2 and FS179-APEX2. Comparing these two profiles allowed us to identify an AC1 proximity proteome that is enriched for tip proteins, including proteins involved in signaling. Our results demonstrate that APEX2-based proximity proteomics is effective in T. brucei and can be used to resolve the proteome composition of flagellum subdomains that cannot themselves be readily purified.IMPORTANCE Sleeping sickness is a neglected tropical disease caused by the protozoan parasite Trypanosoma brucei The disease disrupts the sleep-wake cycle, leading to coma and death if left untreated. T. brucei motility, transmission, and virulence depend on its flagellum (cilium), which consists of several different specialized subdomains. Given the essential and multifunctional role of the T. brucei flagellum, there is need for approaches that enable proteomic analysis of individual subdomains. Our work establishes that APEX2 proximity labeling can, indeed, be implemented in the biochemical environment of T. brucei and has allowed identification of proximity proteomes for different flagellar subdomains that cannot be purified. This capacity opens the possibility to study the composition and function of other compartments. We expect this approach may be extended to other eukaryotic pathogens and will enhance the utility of T. brucei as a model organism to study ciliopathies, heritable human diseases in which cilium function is impaired.

33. The UNAM-MARine Aerosol Tank (UNAM-MARAT): An Evaluation of the Ice-Nucleating Abilities of seawater from the Gulf of Mexico and the Mexican Pacific.

Abstract

Although several studies have shown that sea spray aerosol (SSA) has the potential to act as ice nucleating particles (INP) impacting cloud formation, there is a lack of marine INP studies in tropical latitudes. This is partly due to the unavailability of local oceanographic cruises that perform aerosol-cloud interaction studies in the tropics, as well as the scarcity of appropriate aerosol and cloud microphysics instrumentation. The present study shows the development of the UNAM-MARine Aerosol Tank (UNAM-MARAT), a device that simulates wave breaking to generate SSA particles with the main purpose to characterize their physicochemical properties including their ice nucleating abilities. The UNAM-MARAT was characterized using Instant Ocean Sea Salt and its potential to study ambient sea waters was evaluated with sea seawater samples collected from the Port of Veracruz (PoV) in the Gulf of Mexico, and from the Bay of Acapulco (BoA) and the Bay of Santiago- Manzanillo (BoSM) in the Mexican Pacific Ocean. The portable and automatic UNAM-MARAT is able to generate aerosol particle concentrations as high as 2000 cm-3 covering a wide range of sizes, from 30 nm to 10 μm, similar to those found in the ambient marine boundary layer. The SSA generated from the three natural seawater samples was found to act as INP via immersion freezing, with INP concentrations as high as 130.7 L-1. The particles generated from the BoA seawater samples were the most efficient INPs, reporting the highest ice active site density (ns) values between -20 and -30 °C. Our results also show the direct relationship between particle size and its composition. Larger particles (> 1 μm) were found to be enriched in sodium chloride. In contrast, the fraction of Ca2+, Mg2+, and NO3- was found to increase with decreasing the particle size from 10 μm to 320 nm. 34. A Catch-22—The Test–Retest Method of Reliability Estimation.

Abstract

This article addresses the problems with the traditional reinterview approach to estimating the reliability of survey measures. Using data from three reinterview (or panel) studies conducted by the General Social Survey, we investigate the differences between the two-wave correlational approach embodied by the traditional reinterview strategy, compared to estimates of reliability that take the stability of traits into account based on a three-wave model. Our results indicate that the problems identified with the two-wave correlational approach reflect a kind of "Catch-22" in the sense that the only solution to the problem is denied by the approach itself. Specifically, we show that the correctly specified two-wave model, which includes the potential for true change in the latent variable, is underidentified, and thus, unless one is willing to make some potentially risky assumptions, reliability parameters are not estimable. This article compares the two-wave correlational approach to an alternative model for estimating reliability, Heise's estimates based on the three-wave simplex model. Using three waves of data from the GSS panels, which were separated by 2-year intervals between waves, this article examines the conditions under which the wave-1, wave-2 correlations which do not take stability into account approximate the reliability estimate obtained from three-wave simplex models that do take stability into account. The results lead to the conclusion that the differences between estimates depend on the stability and/or fixed nature of the underlying processes involved. Few if any differences are identified when traits are fixed or highly stable, but for traits involving changes in the underlying traits the differences can be quite large, and thus, we argue for the superiority of reinterview designs that involve more than 2 waves in the estimation of reliability parameters. Abstract

According to the World Health Organization (WHO), breast cancer (BC) is the deadliest and the most common type of cancer worldwide in women. Several factors associated with BC exert their effects by modulating the state of stress. They can induce genetic mutations or alterations in cell growth, encouraging neoplastic development and the production of reactive oxygen species (ROS). ROS are able to activate many signal transduction pathways, producing an inflammatory environment that leads to the suppression of programmed cell death and the promotion of tumor proliferation, angiogenesis, and metastasis; these effects promote the development and progression of malignant neoplasms. However, cells have both non-enzymatic and enzymatic antioxidant systems that protect them by neutralizing the harmful effects of ROS. In this sense, antioxidant enzymes such as superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), glutathione reductase (GR), thioredoxin reductase (TrxR), and peroxiredoxin (Prx) protect the body from diseases caused by oxidative damage. In this review, we will discuss mechanisms through which some enzymatic antioxidants inhibit or promote carcinogenesis, as well as the new therapeutic proposals developed to complement traditional treatments.
