Grapevine trunk diseases (GTDs) are currently limiting grapevine productivity in many vineyards worldwide. As no chemical treatments are registered to control GTDs, biocontrol agents are being tested against these diseases. Esquive® WP, based on the fungus Trichoderma atroviride I-1237 strain, is the first biocontrol product registered in France to control GTDs. In this study, we determine whether, following grapevine pruning wound treatments with Esquive® WP, changes occurred or not in the indigenous microbial communities that are colonizing grapevine wood. Over a 6-year period, Esquive® WP was applied annually to pruning wounds on three grapevine cultivars located in three different regions. Wood samples were collected at 2 and 10 months after the Esquive® WP treatments. Based on MiSeq high-throughput sequencing analyses, the results showed that specific microbial communities were linked to each 'region/cultivar' pairing. In certain cases, a significant modification of alpha diversity indexes and the relative abundance of some microbial taxa were observed between treated and non-treated grapevines 2 months after Esquive® WP treatment. However, these modifications disappeared over time, i.e., 10 months post-treatment. This result clearly showed that Esquive® WP pruning wood treatment did not induce significant changes in the grapevine wood's microbiome, even after 6 years of recurrent applications on the plants. Despite an increasing number of studies being carried out on the biocontrol of grapevine trunk diseases (GTDs), no commercial bacterial products have yet been developed to control GTDs. Knowledge of the precise modes of action (MOA) and the different application methods (AM) for biocontrol agents is crucial if they are to be successful in the field. In light of this, the present study aimed at selecting the most appropriate AM for eight bacterial strains with high potential for controlling Neofusicoccum parvum. These strains were applied on one-year-old grapevine in pots (grown from cuttings) using three methods: co-inoculation at stem level, preventive inoculation at stem level and preventive inoculation at the soil surface. The inhibitory activity of the bacterial strain against N. parvum was significantly dependent on the AM. Application of bacterial strains to stems, especially in a preventive way, was much more efficient than inoculation in the soil. When performing preventive inoculation on stems, the inhibition of N. parvumwood necrosis reached 50 and 65 % for Pantoea agglomerans (S1) and Paenibacillus sp. (S19) respectively. To decipher the underlying processes linked to fungal inhibition, the way in which several MOA affected the antagonistic capacity of these two strains was studied via in vitro and in planta assays. While P. agglomerans (S1) inhibited N. parvum by the secretion of antifungal volatile compounds, Paenibacillus sp. (S19) mainly inhibited this pathogen by antibiosis. In addition, both bacterial strains induced systemic defenses in grapevine. However, this affect tended to be higher at 15-dpi after inoculation with P. agglomerans (S1) than after innoculation with Paenibacillus sp. (S19) (three defense genes repressed versus five respectively). Finally, P. agglomerans (S1) and Paenibacillus sp. (S19) were shown to be potential biocontrol candidates for fighting N. parvum in grapevine, due to the combination of direct control via their antifungal activity and indirect control via their ability to activate the grapevine defense system.

Vine training and pruning are cultural strategies that can be deployed to manage grapevine trunk diseases (GTDs). Forty-year-old commercial vineyards in the Cognac region, France, trained to either Guyot-Arcure (severe pruning) or Guyot-Poussard (minimal pruning), were studied to determine how the two systems affected trunk disease symptomatology. Effects of pruning practices on the pathobiome and mycobiome of asymptomatic grapevines were also assessed, using culture-and amplicon-based Illumina sequencing approaches. The hypothesis examined was that severe pruning of Guyot-Arcure increases trunk diseases incidence and severity, and causes higher pathogen load and microbial diversity, compared to Guyot-Poussard. Numbers of symptomatic and asymptomatic vines for the two training systems were recorded over 3 years, including numbers of vines with esca foliar symptoms, and partially unproductive and dead vines. Six asymptomatic vines from each pruning method were selected, and culturing and sequencing data were obtained from 27 samples per vine. Fungi in the Phaeomoniellaceae, Togniniaceae, and Botryosphaeriaceae were the most frequently identified. The data indicated that severe pruning increased risk of pathogen infections, with Phaeomoniella chlamydospora, Phaeoacremonium minimum and Diplodia sp. being the most commonly identified fungi. Greater numbers of dead or dying vines were recorded in the severely pruned vineyard, indicating that this strategy shortens vine longevity. Results also showed that severe pruning increased endophytic microbial diversity, and that the pruning methods influenced mycobiome community composition. In central Australia, an apparently coeval gneiss dome (Entia Dome) developed adjacent to a thrust belt (Arltunga Nappe Complex) within an intracratonic setting. Here we employ a combination of fieldwork, geochronology, and numerical modeling to investigate the structure and tectonic evolution of these features. We present a structural model linking an extensional domain comprising the Entia Dome, across a transitional zone containing the Bruna décollement zone and the Illogwa shear zone, into a contractional zone comprising thrusts and duplexes of the Arltunga Nappe Complex. Supported by numerical modeling, we propose a tectonic model in which the dome and nappe complex formed synchronously because of the convergent gravitational collapse of the 30-40-km-deep Paleozoic Harts Range rift. Grapevine trunk diseases (GTDs) are currently among the most important health challenges for viticulture in the world. Esca, Botryosphaeria dieback, and Eutypa dieback are the most current GTDs caused by fungi in mature vineyards. Their incidence has increased over the last two decades, mainly after the ban of sodium arsenate, carbendazim, and benomyl in the early 2000s. Since then, considerable efforts have been made to find alternative approaches to manage these diseases and limit their propagation. Biocontrol is a sustainable approach to fight against GTD-associated fungi and several microbiological control agents have been tested against at least one of the pathogens involved in these diseases. In this review, we provide an overview of the pathogens responsible, the various potential biocontrol microorganisms selected and used, and their origins, mechanisms of action, and efficiency in various experiments carried out in vitro, in greenhouses, and/or in vineyards. Lastly, we discuss the advantages and limitations of these approaches to protect grapevines against GTDs, as well as the future perspectives for their improvement. [ABSTRACT FROM AUTHOR]

Two major diseases that affect grapevine leaves and berries are controlled by the oomycete Pythium oligandrum. As the efficacy of biocontrol agents strongly depends on factors such as the trophic behaviors of pathogens and cultivar susceptibility, a two-disease approach was implemented to evaluate the activity of P. oligandrum against Botrytis cinerea (the necrotrophic fungus of gray mold) and Plasmopara viticola (the biotrophic oomycete of downy mildew) on two grapevine cultivars with different susceptibilities to these two pathogens. The results show that grapevine root inoculation with P. oligandrum significantly reduced P. viticola and B. cinerea infection on the leaves of the two cultivars, but with differences. This was observed when the relative expression of 10 genes was measured in response to each pathogen, and could be attributed to their lifestyles, i.e., biotrophic or necrotrophic, which are related to the activation of specific metabolic pathways of the plant. In response to P. viticola infection, genes from the jasmonate and ethylene pathways were mainly induced, whereas for B. cinerea, the genes induced were those of the ethylene–jasmonate pathway. Dynamic subsidence and uplift of plates are often explained by the vertical motion of density anomalies in the mantle. Such models predict surface vertical motion rates of less than 100 m Myr−1 at long‐wavelengths with a timespan of tens of Myr. However, during periods of relative sea‐level stability, some of the phases of vertical motion on stable portions of plates have occurred at rates greater than 100 m Myr−1 during episodes that may last only a few Myr. Here, we show that vertical surface motions, with rates greater than 100 m Myr−1 and durations less than a few Myr, can be explained by changes in basal shear stress caused by variation in horizontal motion of a viscous plate relative to the asthenosphere. Stresses acting on cold, thick and negatively buoyant oceanic lithosphere are thought to be crucial to the initiation of subduction and the operation of plate tectonics (1,2), which characterizes the present-day geodynamics of the Earth. Because the Earth's interior was hotter in the Archaean eon, the oceanic crust may have been thicker, thereby making the oceanic lithosphere more buoyant than at present (3), and whether subduction and plate tectonics occurred during this time is ambiguous, both in the geological record and in geodynamic models (4). Here we show that because the oceanic crust was thick and buoyant (5), early continents may have produced intra-lithospheric gravitational stresses large enough to drive their gravitational spreading, to initiate subduction at their margins and to trigger episodes of subduction. Our model predicts the co-occurrence of deep to progressively shallower mafic volcanics and arc magmatism within continents in a self-consistent geodynamic framework, explaining the enigmatic multimodal volcanism and tectonic record of Archaean cratons (6). Moreover, our model predicts a petrological stratification and tectonic structure of the sub-continental lithospheric mantle, two predictions that are consistent with xenolith (5) and seismic studies, respectively, and consistent with the existence of a mid-lithospheric seismic discontinuity (7). In the context of continental extension, transient compressional episodes (stress inversion) and phases of uplift (depth inversion) are commonly recorded with no corresponding change in plate motion. Changes in gravitational potential energy during the rifting process have been invoked as a possible source of compressional stresses, but their magnitude, timing, and relationship with depth inversions remain unclear. Using high-resolution two-dimensional numerical experiments of the full rifting process, we track the dynamic interplay between the far-field tectonic forces, loading and unloading of the surface via surface processes, and gravitational body forces. Our results show that rift basins tend to localize compressive stresses; they record transient phases of compressional stresses as high as 30 MPa and experience a profound depth inversion, 2 km in magnitude, when sediment supply ceases, providing an additional driver for the breakup unconformity, a well-documented phase of regional uplift typically associated with continental breakup. Grapevine curettage was re-introduced in France in the early 2000s, and is important for facilitating recovery of plants from esca disease. This surgical practice involves removal of deadwood of vines with leaf symptoms, focusing on white rot generally observed at the centres of grapevine trunks. Assessment of the efficacy of this practice was initiated in the Bordeaux region in 2014. One 'Sauvignon Blanc' vineyard severely affected by esca was initially surveyed in the summer of 2014, to identify and treat vines with esca foliar symptoms. Annually thereafter, from 2014 to 2018, selected vine stocks were curetted. Two other 'Sauvignon Blanc' vineyards also displaying high levels of esca damage were added to the study in 2015 and 2016. Curettage treatments ceased in 2018, resulting in 11 trials (vineyard × year combinations). In total, 856 vines (422 curetted and 434 control vines) were then surveyed annually up to 2021, for assessments and comparisons of esca development. At each site, plants with esca symptoms recovered well after curettage: on average 85% of all curetted vines became asymptomatic the year immediately after the treatment. Six years after treatment, for curettage campaigns carried out in 2014 and 2015, more than half of the curetted vines were symptom-free, whereas <12% of the control vines were asymptomatic, and gradual loss of efficacy was observed at each site. The mean annual proportion of efficacy erosion was approx. 8% per year. This study highlights the possible short- and midterm benefits of trunk surgery to enable recovery of esca-affected vines, and for them to recover and remain leaf-asymptomatic for several years. Pruning experimental studies have been performed in different vineyards, in France, USA and Australia. This article investigates and models the effects of pruning quality on the installation of desiccation cones and wood necrotization. Two different modalities of pruning, short and high pruning, were performed at the same period each year on three cultivars in Bordeaux (Cabernet Sauvignon and Sauvignon Blanc) and Charente (Ugni Blanc) wine regions. In the short typ of pruning, the diaphragm was damaged but, in the high one, a 2–3 cm woody length was left immediately above the diaphragm. None of the three cultivars showed any correlation between necrosis length and spur diameter (R2 < 0.1925). Analysis of the Ugni Blanc, 8 months after pruning, showed significantly more necrosis length (>60%) than either Cabernet Sauvignon (31–41%) or Sauvignon Blanc (25–55%). Desiccation cone necrotization rates also varied with the vintage, particularly for Ugni Blanc. 4 or 8 months after pruning, the newly-installed desiccation cones could then be analysed. High pruning stopped the desiccation cones at the diaphragm, which ensured that the sap flow path remained unaffected. [ABSTRACT FROM AUTHOR]

Long‐lived high to ultra‐high temperature (HT‐UHT) granulitic terranes formed throughout Earth's history. Yet, the detailed processes involved in their formation remain unresolved and notably the sequence of appearance and duration of migmatisation and granulites conditions in the orogenic cycle. These processes can be evaluated by analytical and numerical models. First, solving the steady‐state heat equation allows underlining the interdependency of the parameters controlling the crustal geotherm at thermal equilibrium. Second, performing two‐dimensional thermo‐mechanical experiments of an orogenic cycle, from shortening to gravitational collapse, allows to consider non‐steady‐state geotherms and understand how deformation velocity may affect the relative timing of migmatite and granulite formation. These numerical experiments with elevated radiogenic heat production and slow shortening rates allow the formation of large volumes of prograde migmatites and granulites going through the sillimanite field as observed in many HT‐UHT terranes. Finally, the interplay between these parameters can explain the difference in predicted pressure‐temperature‐time paths that can be compared with the natural rock archive. [ABSTRACT FROM AUTHOR]

This paper explores the secular evolution of the height of an isostatically balanced collisional mountain belt in the context of ongoing convergence. We show that until the Neoarchean, continents were unable to sustain topography >2500 m. During the Neoarchean the continental lithosphere evolved through a rheological threshold, allowing for the development of significant topography. The consequence of the strengthening of the continental lithosphere is fundamental for the coupling of the Earth's geochemical reservoirs. The Neoarchean was a period of global changes during which exogenic envelopes recorded major shifts in composition toward modern values. We propose that during the Neoarchean the exogenic envelopes, which were until then coupled to the mantle through hydrothermal processes and volcanism, also became coupled to the continental crust through reliefgenerating tectonics processes and erosion, hence changing the balance between mantle versus crustal interaction with the exogenic Earth. Rootstocks are the link between the soil and scion in grapevines, can provide tolerance to abiotic and biotic stresses, and regulate yield and grape quality. The vascular system of grapevine rootstocks in nurseries is still an underexplored niche for research, despite its potential for hosting beneficial and pathogenic microorganisms. The purpose of this study was to investigate the changes in the composition of fungal communities in 110 Richter and 41 Berlandieri rootstocks at four stages of the grapevine propagation process. Taxonomic analysis revealed that the fungal community predominantly consisted of phylum Ascomycota in all stages of the propagation process. The alpha-diversity of fungal communities differed among sampling times for both rootstocks, with richness and fungal diversity in the vascular system decreasing through the propagation process. The core microbiome was composed of the genera Cadophora, Cladosporium, Penicillium and Alternaria in both rootstocks, while the pathogenic genus Neofusicoccum was identified as a persistent taxon throughout the propagation process. FUNguild analysis showed that the relative abundance of plant pathogens associated with trunk diseases increased towards the last stage in nurseries. Fungal communities in the vascular system of grapevine rootstocks differed between the different stages of the propagation process in nurseries. Numerous genera associated with potential biocontrol activity and grapevine trunk diseases were identified. Understanding the large diversity of fungi in the rootstock vascular tissue and the interactions between fungal microbiota and grapevine will help to develop sustainable strategies for grapevine protection. Summary: Fungi are considered to cause grapevine trunk diseases such as esca that result in wood degradation. For instance, the basidiomycete Fomitiporia mediterranea (Fmed) is overabundant in white rot, a key type of wood‐necrosis associated with esca. However, many bacteria colonize the grapevine wood too, including the white rot. In this study, we hypothesized that bacteria colonizing grapevine wood interact, possibly synergistically, with Fmed and enhance the fungal ability to degrade wood. We isolated 237 bacterial strains from esca‐affected grapevine wood. Most of them belonged to the families Xanthomonadaceae and Pseudomonadaceae. Some bacterial strains that degrade grapevine‐wood components such as cellulose and hemicellulose did not inhibit Fmed growth in vitro. We proved that the fungal ability to degrade wood can be strongly influenced by bacteria inhabiting the wood. This was shown with a cellulolytic and xylanolytic strain of the Paenibacillus genus, which displays synergistic interaction with Fmed by enhancing the degradation of wood structures. Genome analysis of this Paenibacillus strain revealed several gene clusters such as those involved in the expression of carbohydrate‐active enzymes, xylose utilization and vitamin metabolism. In addition, certain other genetic characteristics of the strain allow it to thrive as an endophyte in grapevine and influence the wood degradation by Fmed. This suggests that there might exist a synergistic interaction between the fungus Fmed and the bacterial strain mentioned above, enhancing grapevine wood degradation. A lower crust with a petrological heterogeneity, deformation of extensions and reduced-viscosity in lower crust having a minimal viscosity difference between silicic and mafic rocks are essential for the lower crustal layering to be placed in a widespread region. Phanerozoic regions with lower crust imply that metamorphism of medium-pressure granulite facies are associated with the significant event recorded by the layered lower crust. Granulite facies metamorphism of the crust and the upper crust extensions determining deposits on the sedimentary basin occurred in the same period.

Although sodium arsenite was widely used in Europe until its ban in 2003, its effects on microorganisms is not clearly understood. To improve our understanding of sodium arsenite curative effect on GTDs, grapevines displaying esca-foliar symptoms from different French regions (Alsace, Champagne, Languedoc) were treated or not with sodium arsenite, and analyzed for their wood microbiota. Using metabarcoding, we identified the fungal and bacterial taxa composition of microbiota colonizing woody trunk tissues. Large differences in fungal microbiota composition between treated and untreated grapevines were observed while no major impacts were observed on bacteria microbiota. The main fungal species detected in untreated necrotic woody tissues was Fomitiporia mediterranea (63–94%), a fungal pathogen associated with esca. The relative abundance of this fungal species significantly decreased after sodium arsenite treatment in the three vineyards, in particular in white-rot necrotic tissues and their borders (−90%). F. mediterranea was the most sensitive to sodium arsenite among fungi from grapevine woody tissues. These results strongly suggest that the effect of sodium arsenite on GTDs is due to its ability to efficiently and almost specifically eliminate F. mediterranea from white-rot necrotic tissues, allowing saprobic fungi to colonize the tissues previously occupied by this pathogenic fungus. Fungal species involved in Esca cause the formation of grapevine wood necroses. It results in the deterioration of vascular network transport capacity and the disturbance of the physiological processes, leading to gradual or sudden grapevine death. Herein, for two consecutive growing seasons, a detailed analysis of the structural (wood necrosis and leaf discoloration) and physiological parameters related to the water use of healthy and esca-symptomatic grapevines was conducted. Measurements were carried out on 17-year-old grapevines that expressed, or not, Esca-leaf symptoms in a vineyard of the Bordeaux region (France). Whole-plant transpiration was recorded continuously from pre-veraison to harvest, using noninvasive sap flow sensors. Whole-plant transpiration was systematically about 40–50% lower in Esca-diseased grapevines compared with controls, and this difference can be observed around 2 weeks before the first Esca-foliar symptoms appeared in the vineyard. Unlike grapevine sap flow disruption, structural (e.g., leaf discolorations), functional (e.g., stomatal conductance, photosynthetic activity, phenolic compounds), and genetic (e.g., expression of leaf-targeted genes) plant responses were only significantly impacted by Esca at the onset and during leaf symptoms development. We conclude that sap flow dynamic, which was related to a high level of a white-rot necrosis, provides a useful tool to predict plant disorders due to Esca-grapevine disease. [ABSTRACT FROM AUTHOR]

BACKGROUND Black‐foot and Petri diseases are the main fungal diseases associated with young grapevine decline. Two field experiments were established to evaluate the preventive effect of two potential biocontrol agents (BCAs), that is Streptomyces sp. E1 + R4 and Pythium oligandrum Po37, and three BCA‐commercial products containing Trichoderma atroviride SC1, Trichoderma koningii TK7 and Pseudomonas fluorescens + Bacillus atrophaeus on fungal infection in grafted plants and plant growth parameters. RESULTS: The effectiveness of some BCA in reducing the incidence and severity of both diseases was dependent on the plant part analyzed and the plant age. No single BCA application was able to control both diseases. Streptomyces sp. E1 + R4 were able to reduce significantly the infection of the most prevalent black‐foot disease fungi while P. oligandrum Po37 and Trichoderma spp. were able to reduce significantly Phaeomoniella chlamydospora and Phaeoacremonium minimum (Petri disease) infection. BCA treatments had no effect on the shoot weight, and root weight was significantly lower in all BCA treatments with respect to the control. CONCLUSIONS: The combination of the disease‐suppressive activity of two or more beneficial microbes in a biocontrol preparation is required to prevent infection by black‐foot and Petri disease fungi in vineyards. Summary: Esca, a major grapevine trunk disease in old grapevines, is associated with the colonization of woody tissues by a broad range of plant pathogenic fungi. To identify which fungal and bacterial species are involved in the onset of this disease, we analysed the microbiota from woody tissues of young (10‐year‐old) grapevines at an early stage of esca. Using meta‐barcoding, 515 fungal and 403 bacterial operational taxonomic units (OTUs) were identified in woody tissues. In situ hybridization showed that these fungi and bacteria co‐inhabited in grapevine woody tissues. In non‐necrotic woody tissues, fungal and bacterial microbiota varied according to organs and seasons but not diseased plant status. Phaeomoniella chlamydospora, involved in the Grapevine trunk disease, was the most abundant species in non‐necrotic tissues from healthy plants, suggesting a possible non‐pathogenic endophytic behaviour. Most diseased plants (70%) displayed cordons, with their central white‐rot necrosis colonized essentially by two plant pathogenic fungi (Fomitiporia mediterranea: 60%–90% and P. chlamydospora: 5%–15%) and by a few bacterial taxa (Sphingomonas spp. and Mycobacterium spp.). The occurrence of a specific association of fungal and bacterial species in cordons from young grapevines expressing esca‐foliar symptoms strongly suggests that that microbiota is involved in the onset of this complex disease. In orogens worldwide and throughout geologic time, large volumes of deep continental crust have been exhumed in domal structures. Extension‐driven ascent of bodies of deep, hot crust is a very efficient mechanism for rapid heat and mass transfer from deep to shallow crustal levels and is therefore an important mechanism in the evolution of continents. The dominant rock type in exhumed domes is quartzofeldspathic gneiss (typically migmatitic) that does not record its former high‐pressure (HP) conditions in its equilibrium mineral assemblage; rather, it records the conditions of emplacement and cooling in the mid/shallow crust. Mafic rocks included in gneiss may, however, contain a fragmentary record of a HP history, and are evidence that their host rocks were also deeply sourced. An excellent example of exhumed deep crust that retains a partial HP record is in the Montagne Noire dome, French Massif Central, which contains well‐preserved eclogite (garnet+omphacite+rutile+quartz) in migmatite in two locations: one in the dome core and the other at the dome margin. Both eclogites record P ~ 1.5 ± 0.2 GPa at T ~ 700 ± 20°C, but differ from each other in whole‐rock and mineral composition, deformation features (shape and crystallographic preferred orientation, CPO), extent of record of prograde metamorphism in garnet and zircon, and degree of preservation of inherited zircon. Rim ages of zircon in both eclogites overlap with the oldest crystallization ages of host gneiss at c. 310 Ma, interpreted based on zircon rare earth element abundance in eclogite zircon as the age of HP metamorphism. Dome‐margin eclogite zircon retains a widespread record of protolith age (c. 470–450 Ma, the same as host gneiss protolith age), whereas dome‐core eclogite zircon has more scarce preservation of inherited zircon. Possible explanations for differences in the two eclogites relate to differences in the protolith mafic magma composition and history and/or the duration of metamorphic heating and extent of interaction with aqueous fluid, affecting zircon crystallization. Differences in HP deformation fabrics may relate to the position of the eclogite facies rocks relative to zones of transpression and transtension at an early stage of dome development. Regardless of differences, both eclogites experienced HP metamorphism and deformation in the deep crust at c. 310 Ma and were exhumed by lithospheric extension—with their host migmatite—near the end of the Variscan orogeny. The deep crust in this region was rapidly exhumed from ~50 to <10 km, where it equilibrated under low‐P/high‐T conditions, leaving a sparse but compelling record of the deep origin of most of the crust now exposed in the dome.
