Abstract Background Kinesin family member C3 (KIFC3), as reported, plays important roles in several tumor types. Nevertheless, it is unknown whether KIFC3 has effects on non–small cell lung cancer (NSCLC) development. Materials and methods KIFC3 expression was detected by RT‐PCR, and its correlation with prognosis was analyzed by GEPIA website. Small interfering RNA against KIFC3 were adopted for modulating KIFC3 expression in NSCLC cells. KIFC3 effects on NSCLC cell proliferation were determined using the MTT and clone formation assay. Matrigel invasion and wound healing assays were adopted for measuring the invasion and migration capability of NSCLC cells. Western blot was applied for measuring the levels of proteins associated with the phosphatidylinositol‐3‐kinase/protein kinase B (PI3K/Akt) pathway in NSCLC cells. Results KIFC3 was markedly increased in NSCLC samples and cells. KIFC3 knockdown suppressed the proliferation, invasion, and migration in NSCLC. Mechanically, KIFC3 silencing suppressed NSCLC progression through inhibiting the PI3K/Akt pathway. Conclusions KIFC3 lack suppressed the proliferation, invasion, and migration which works, at least partially, by the PI3K/Akt pathway. These findings suggest that targeting KIFC3 via the PI3K/Akt pathway may offer a novel therapeutic strategy for NSCLC.

Abstract Enabling soft shape memory polyurethane (SMPU) with self‐healing is an effective strategy to extend its service life; however, the tradeoff between high mechanical strength and rapid self‐healing to meet the practical applications remains a challenge. Herein, we prepared a polyurethane (PU) with polycarbonate diol as a soft segment, incorporating urea bonds and dynamic disulfide bonds through chain extenders. This material, denoted as PU‐NSS75, exhibited a remarkable tensile strength of 51.4 MPa, attributed to the synergistic energy dissipation effect of urea hydrogen bonding and dynamic disulfide bonds during stretching, leading to enhanced mechanical properties of the PU. The dynamic nature of the bonds, including hydrogen bonding and disulfide bonds, endowed the PU with exceptional self‐healing capabilities. Furthermore, leveraging the reversible mechanism of disulfide bonds under UV irradiation at room temperature, PU‐NSS75 achieved complete self‐healing within 1 min. This photoinduced dynamic chemical bonding also facilitated rapid reconfigure the original shape of the soft SMPU at room temperature, thus broadening its potential applications. This study offers a design approach to enhance soft shape memory polyurethane materials, while the rapid self‐healing and shape reconfiguration provide new avenues for expanding the applications of shape memory polyurethanes.

Abstract Objective To construct the deep learning convolution neural network (CNN) model and machine learning support vector machine (SVM) model of bone remodeling of chronic maxillary sinusitis (CMS) based on CT image data to improve the accuracy of image diagnosis. Methods Maxillary sinus CT data of 1000 samples in 500 patients from January 2018 to December 2021 in our hospital was collected. The first part is the establishment and testing of chronic maxillary sinusitis detection model by 461 images. The second part is the establishment and testing of the detection model of chronic maxillary sinusitis with bone remodeling by 802 images. The sensitivity, specificity and accuracy and area under the curve (AUC) value of the test set were recorded, respectively. Results Preliminary application results of CT based AI in the diagnosis of chronic maxillary sinusitis and bone remodeling. The sensitivity, specificity and accuracy of the test set of 93 samples of CMS, were 0.9796, 0.8636 and 0.9247, respectively. Simultaneously, the value of AUC was 0.94. And the sensitivity, specificity and accuracy of the test set of 161 samples of CMS with bone remodeling were 0.7353, 0.9685 and 0.9193, respectively. Simultaneously, the value of AUC was 0.89. Conclusion It is feasible to use artificial intelligence research methods such as deep learning and machine learning to automatically identify CMS and bone remodeling in MSCT images of paranasal sinuses, which is helpful to standardize imaging diagnosis and meet the needs of clinical application.

Asthma, being the prevailing respiratory ailment globally, remains enigmatic in terms of its pathogenesis. In recent times, the advancement of traditional Chinese medicine pertaining to the intestinal microbiota has yielded a plethora of investigations, which have substantiated the potential of traditional Chinese medicine in disease prevention and treatment through modulation of the intestinal microbiota. Both animal models and clinical trials have unequivocally demonstrated the indispensable role of the intestinal microbiota in the pathogenesis of asthma. This article presents a summary of the therapeutic effects of traditional Chinese medicine in the context of regulating gut microbiota and its metabolites, thereby achieving immune regulation and inhibiting airway inflammation associated with asthma. It elucidates the mechanism by which traditional Chinese medicine modulates the gut microbiota to enhance asthma management, offering a scientific foundation for the utilization of traditional Chinese medicine in the treatment of asthma.

This study analyzed the effect of mite-specific subcutaneous immunotherapy (SCIT) on patients with allergic rhinitis (AR). We enrolled 98 AR patients visiting our hospital from April 2017 to April 2019 and grouped them in a random number table. The control group (n=49) received conventional treatment for three years. The SCIT used a standardized mite allergen injection for the experimental group (n=49) for three years. The study compared total nasal symptom score (TNSS), daily medication score (DMS), total combined score (TCS), visual analog scale (VAS) score, mini-rhinitis quality of life questionnaire (MiniRQLQ) score, and serum immunoglobulin E (sIgE) level before and after treatment. ABSTRACT In China, the duck industry has been severely impacted by the newly emerging duck Tembusu virus (DTMUV). For DTMUV to successfully infect host cells, it employs several strategies that subvert the host's innate immune response. It has been found that several viral proteins encoded by DTMUV have strategically targeted the crucial molecules of the RIG-I-like Receptor (RLR) signaling pathway to antagonize host antiviral responses. However, it is not well known how the host proteins manipulated by DTMUV contribute to innate immune evasion. The present study reports that duck TRIM35 (duTRIM35) antagonizes DTMUV-induced innate immune responses by targeting duck RIG-I (duRIG-I) in duck embryo fibroblasts. A significant increase in duTRIM35 expression occurred during DTMUV infection. DuTRIM35 overexpression suppressed DTMUV-triggered expression of interferon beta (IFN-β) and interferon-stimulated genes (ISGs), promoting viral replication, whereas knockdown of duTRIM35 augments the innate immune response, reducing viral replication. Furthermore, duTRIM35 significantly impaired the IFN-β expression mediated by duRIG-I but not by other RLR signaling molecules. Mechanistically, duTRIM35 interfered with duRIG-I-duTRIM25 interaction and impeded duTRIM25-mediated duRIG-I ubiquitination by interacting with both duRIG-I and duTRIM25. Our findings indicate that duTRIM35 expression induced by DTMUV infection interfered with the duRIG-I-mediated antiviral response, illustrating a novel strategy in which DTMUV can evade the host's innate immunity. IMPORTANCE Duck Tembusu virus (DTMUV), an emerging flavivirus pathogen causing a substantial drop in egg production and severe neurological disorders in duck populations, has led to massive economic losses in the global duck industry. DTMUV has employed various strategies to subvert the host's innate immune response to establish a productive infection in host cells. In this study, we report that duck TRIM35 (duTRIM35) expression was upregulated upon DTMUV infection in vitro and in vivo, and its expression antagonized DTMUV-induced innate immune responses by targeting duck RIG-I (duRIG-I) in duck embryo fibroblasts. Further studies suggest that duTRIM35 interfered with duRIG-I-duTRIM25 interaction and impeded duTRIM25-mediated duRIG-I ubiquitination by interacting with both duRIG-I and duTRIM25. Together, these results revealed that duTRIM35 expression induced by DTMUV infection downregulated duRIG-I-mediated host antiviral response, which elucidated a novel strategy of DTMUV for innate immune evasion.

In industrial production, the effective and reliable performance of hydraulic systems is closely associated with product quality, personal safety, economic efficiency, etc. It is of utmost significance to perform the health status evaluation of systems. In this paper, a least-squares recursive parameter identification algorithm is proposed to realize the graded evaluation of the health status of the hydraulic system under variable operating conditions. First, a nonlinear model of the hydraulic system is established based on a mechanism analysis. Based on the system identifiable model obtained by parameter linearization, the least squares recursive algorithm is used to get the system parameters. Second, the system measurable data are graded and labeled under the same operating condition, and the variable parameter ranges under different health states are obtained by the parameter identification algorithm. Finally, under variable operating conditions, the estimates of variable parameters are compared with the range of health state parameters to complete the system health state graded evaluation. The feasibility of the proposed evaluation method is verified by MATLAB simulation software.

Objective: This study proposed a modified Blumgart anastomosis (m-BA) that uses a firm ligation of the main pancreatic duct with a supporting tube to replace the pancreatic duct-to-jejunum mucosa anastomosis, with the aim of simplifying the complicated steps of the conventional BA (c-BA). Thus, we observe if a difference in the risk of postoperative pancreatic fistula (POPF) exists between the two methods. Methods: The m-BA anastomosis method has been used since 2010. From October 2011 to October 2015, 147 patients who underwent pancreatoduodenectomy (PD) using BA in Tianjin Medical University Cancer Institute and Hospital were enrolled in this study. According to the type of pancreatojejunostomy (PJ), 50 patients underwent m-BA and 97 received c-BA. The two patient cohorts were compared prospectively to some extent but not randomized, and the evaluated variables were operation time, the incidence rate of POPF, and other perioperative complications. Results: The operation time showed no significant difference (P > 0.05) between the two groups, but the time of duct-to-mucosa anastomosis in the m-BA group was much shorter than that in the c-BA group (P < 0.001). The incidence rate of clinically relevant POPF was 12.0% (6/50) in the modified group and 10.3% (10/97) in the conventional group (P > 0.05), which means that the modified anastomosis method did not cause additional pancreatic leakage. The mean length of postoperative hospital stay of the m-BA group was 23 days, and that of the c-BA group was 22 days (P > 0.05). Conclusions: Compared with the conventional BA, we suggest that the modified BA is a feasible, safe, and effective operation method for P J of PD with no sacrifice of surgical quality. In the multivariate analysis, we also found that body mass index (≥ 25 kg/m2) increased the risk of POPF.

Objective: To present a case report highlighting a severe, yet avoidable, complication following small needle-scalpel treatment for cervical spondylosis.Introduction: The small needle-scalpel is a miniature surgical instrument used to create intense and invasive punctures at certain acupoints with a small latch needle. It has been increasingly gaining popularity among clinicians and patients all over the world during the past years. However, severe complications after small needle-scalpel treatment have not previously been reported.Methods: Here we report a 54-year-old man who recently suffered from cervical spondylosis and underwent small needle-scalpel treatment, which was performed by a rural doctor. While there were no new neurologic deficits, the patient experienced delayed functional deterioration until the onset of quadriplegia within 1 month of treatment. Magnetic resonance imaging demonstrated a C2–C7 dorsally placed extradural hematoma with severe cord compression and subcutaneous soft tissue hemorrhage.Results: The patient refused urgent corrective surgery and later died due to respiratory failure.Conclusions: Although small needle-scalpel therapy has many benefits, such as reducing pain, shorter expenditure, shorter period of therapy and better recovery of function, there are also many potentially severe risks, such as cervical extradural bleeding, which requires clinicians to pay more attention to avoid the complications.

Soft error caused by single event upset has been a severe challenge to aerospace-based computing. Silent data corruption (SDC) is one of the results incurred by soft error. SDC occurs when a program generates erroneous output with no indications. SDC is the most insidious type of results and very difficult to detect. To address this problem, we design and implement an invariant-based system called Radish. Invariants describe certain properties of a program; for example, the value of a variable equals a constant. Radish first extracts invariants at key program points and converts invariants into assertions. It then hardens the program by inserting the assertions into the source code. When a soft error occurs, assertions will be found to be false at run time and warn the users of soft error. To increase the coverage of SDC, we further propose an extension of Radish, named Radish_D, which applies software-based instruction duplication mechanism to protect the uncovered code sections. Experiments using architectural fault injections show that Radish achieves high SDC coverage with very low overhead. Furthermore, Radish_D provides higher SDC coverage than that of either Radish or pure instruction duplication.

Acupuncture, as a traditional treatment, has been extensively used in China for thousands of years. According to the World Health Organization (WHO), acupuncture is recommended for the treatment of 77 diseases. And 16 of these diseases are related to inflammatory pain. As a combination of traditional acupuncture and modern electrotherapy, electroacupuncture (EA) has satisfactory analgesic effects on various acute and chronic pain. Because of its good analgesic effects and no side effects, acupuncture has been widely accepted all over the world. Despite the increase in the number of studies, the mechanisms via which acupuncture exerts its analgesic effects have not been conclusively established. A literature review of related research is of great significance to elaborate on its mechanisms and to inform on further research directions. We elucidated on its mechanisms of action on inflammatory pain from two levels: peripheral and central. It includes the mechanisms of acupuncture in the periphery (immune cells and neurons, purinergic pathway, nociceptive ion channel, cannabinoid receptor and endogenous opioid peptide system) and central nervous system(TPRV1, glutamate and its receptors, glial cells, GABAergic interneurons and signaling molecules). In this review, we collected relevant recent studies to systematically explain the mechanisms of acupuncture in treating inflammatory pain, with a view to providing direction for future applications of acupuncture in inflammatory pain and promoting clinical development. Higher capacity and larger scales have always been the top targets for the evolution of optical access networks, driven by the ever-increasing demand from the end users. One thing that started to attract wide attention not long ago, but with at least equal importance as capacity and scale, is energy efficiency, a metric essential nowadays as human beings are confronted with severe environmental issues like global warming, air pollution, and so on. Here, different from the conventional energy consumption analysis of tree-topology networks, we propose an effective energy consumption calculation method to compare the energy efficiency of the tree-topology 10 gigabit ethernet passive optical network (10G-EPON) and ring-topology time- and wavelength-division-multiplexed passive optical network (TWDM-PON), two experimental networks deployed in China. Numerical results show that the ring-topology TWDM-PON networks with 2, 4, 8, and 16 wave-lengths are more energy efficient than the tree-topology 10G-EPON, although 10G-EPON consumes less energy. Also, TWDM-PON with four wavelengths is the most energy-efficient network candidate and saves 58.7% more energy than 10G-EPON when fully loaded. [ABSTRACT FROM AUTHOR]

Tembusu virus (TMUV) is a newly emerged avian flavivirus that has caused severe egg-drop syndrome and fatal encephalitis in domestic ducks. It has spread widely throughout the main duck-producing areas in Asia, resulting in substantial economic losses to the duck industry. Previous studies have reported that TMUV has evolved several strategies to counteract the duck's innate immune responses to successfully establish infection in its host cells. However, the mechanisms underlying this phenomenon have not been elucidated. Here, we discovered that TMUV-encoded NS2B is a negative regulator of poly(I:C)-induced duck interferon-b (IFN-b) expression. Mechanistically, TMUV NS2B was found to interact specifically with the mitochondrial antiviral-signaling protein (duMAVS). Consequently, duMAVS was degraded through the K48-linked ubiquitination and proteasomal pathway, leading to the interruption of the RIG-I-like receptor (RLR) signaling. Further analyses also identified K321, K354, K398, and K411 as crucial residues for NS2B-mediated ubiquitination and degradation of duMAVS. Additionally, we demonstrated that NS2B functions by recruiting the E3 ubiquitin ligase duck membraneassociated RING-CH-type finger 5 (duMARCH5) to modify duMAVS via polyubiquitination, blocking the duMAVS-mediated innate immune response and promoting TMUV replication. Taken together, our findings revealed a novel mechanism by which TMUV evades the duck's antiviral innate immune responses. IMPORTANCE Tembusu virus (TMUV), an emerging pathogenic flavivirus, has spread to most duck farming areas in Asia since 2010, causing significant economic losses to the duck industry. Recently, TMUV has expanded its host range and may pose a potential threat to mammals, including humans. Understanding the interaction between TMUV and its host is essential for the development of effective vaccines and therapeutics. Here, we show that NS2B encoded by TMUV inhibits IFN production by interacting with duck MAVS (duMAVS) to mediate ubiquitination and proteasomal degradation. Further studies suggest that the E3 ubiquitin ligase duck membrane-associated RING-CH-type finger 5 (duMARCH5) is recruited by NS2B to mediate proteasomal degradation of duMAVS. As a result, the innate immune response triggered by the RIG-I-like receptor (RLR) is disrupted, facilitating viral replication. Adjuvant therapy after resection of hepatocellular carcinoma (HCC) is limited. Here, we evaluated the effects of postoperative sorafenib on recurrence and survival in HCC patients. Recurrence-free survival and overall survival were analyzed as the main endpoint, recurrence rate, and mortality rate were analyzed as second endpoint. Furthermore, postrecurrence survival was also analyzed. Clinicopathological factors were compared between sorafenib and control groups. Seventy-eight patients were eligible for final data analysis (46 in control group; 32 in sorafenib group). Sorafenib did not significantly prolong recurrencefree survival (11.0 months in the control group vs. 11.7 months in the sorafenib group, p = 0.702), but significantly prolonged overall survival (32.4 vs. 25.0 months, p = 0.046). Sorafenib did not reduce recurrence rate (67.7% vs. 78.3%, p = 0.737), but significantly reduced mortality rate (28.1% vs. 60.9%, p = 0.004). The increased post-recurrence survival (22.2 vs. 4.4 months, p = 0.003) may have contributed to the survival benefit after recurrence in the sorafenib group. Adjuvant sorafenib did not decrease tumor recurrence, but significantly reduced mortality and prolonged overall survival of HCC patients after curative resection, probably by inhibiting tumor growth after tumor recurrence.
