Abstract The identification of effective drug targets and the development of bioactive molecules are areas of high need in cancer therapy. The phosphatidylinositol transfer protein alpha/beta isoform (PITPα/β) has been reported to play an essential role in integrating phosphoinositide trafficking and lipid metabolism in diverse cellular processes but remains unexplored as a potential target for cancer treatment. Herein, data analysis of clinical cancer samples revealed that PITPα/β expression is closely correlated with the poor prognosis. Target identification by chemical proteomic methods revealed that microcolin H, a naturally occurring marine lipopeptide, directly binds PITPα/β and displays antiproliferative activity on different types of tumour cell lines. Furthermore, we identified that microcolin H treatment increased the conversion of LC3I to LC3II, accompanied by a reduction of the level of p62 in cancer cells, leading to autophagic cell death. Moreover, microcolin H showed preeminent antitumour efficacy in nude mouse subcutaneous tumour models with low toxicity. Our discoveries revealed that by targeting PITPα/β, microcolin H induced autophagic cell death in tumours with efficient anti-proliferating activity, which sheds light on PITPα/β as a promising therapeutic target for cancer treatment.

Corneal blindness is a common phenomenon, and corneal transplantation is an effective treatment for corneal defects. However, there is usually a mismatch between the corneal repair material and the degree of the patient’s corneal defect. Therefore, patients with different corneal defects need suitable corneal repair materials with a specific microstructure for personalized treatment. In this research, collagen films with bionic structures were fabricated through ethanol evaporation technique by regulating the volume ratios of collagen solution: ethanol = 10:0(Col)/9:1(CC91)/8:2(CC82)/CC73(CC73). Under various preparation conditions, the obtained collagen films contain layered structures of different density. SEM photos show that the CC73 film with a dense layer arrangement has a microstructure similar to that of the corneal epithelial layer, whereas the Col film has a loose layered structure similar to that of the corneal stroma layer. Four kinds of collagen films showed different optical properties and water absorption ability. A more ordered arrangement of internal layer structure leads to better mechanical properties of the collagen film. In view of this, we think that these collagen films with different microstructures and different interlayer spacing may have huge potential applications for personalized corneal repair.

Abstract Pressure ulcers (PU) are caused by persistent long-term pressure, which compromises the integrity of the epidermis, dermis, and subcutaneous adipose tissue layer by layer, making it difficult to heal. Platelet products such as platelet lysate (PL) can promote tissue regeneration by secreting numerous growth factors based on clinical studies on skin wound healing. However, the components of PL are difficult to retain in wounds. Gelatin methacrylate (GelMA) is a photopolymerizable hydrogel that has lately emerged as a promising material for tissue engineering and regenerative medicine. The PL liquid was extracted, flow cytometrically detected for CD41a markers, and evenly dispersed in the GelMA hydrogel to produce a surplus growth factor hydrogel system (PL@GM). The microstructure of the hydrogel system was observed under a scanning electron microscope, and its sustained release efficiency and biological safety were tested in vitro. Cell viability and migration of human dermal fibroblasts, and tube formation assays of human umbilical vein endothelial cells were applied to evaluate the ability of PL to promote wound healing and regeneration in vitro. Real-time polymerase chain reaction (PCR) and western blot analyses were performed to elucidate the skin regeneration mechanism of PL. We verified PL’s therapeutic effectiveness and histological analysis on the PU model. PL promoted cell viability, migration, wound healing and angiogenesis in vitro. Real-time PCR and western blot indicated PL suppressed inflammation and promoted collagen I synthesis by activating STAT3. PL@GM hydrogel system demonstrated optimal biocompatibility and favorable effects on essential cells for wound healing. PL@GM also significantly stimulated PU healing, skin regeneration, and the formation of subcutaneous collagen and blood vessels. PL@GM could accelerate PU healing by promoting fibroblasts to migrate and secrete collagen and endothelial cells to vascularize. PL@GM promises to be an effective and convenient treatment modality for PU, like chronic wound treatment.

Abstract The aim of this work is to examine the effects of vitamin E addition to water on the structure of the gill tissue and energy metabolism of crucian carp (Carassius auratus) under cooling stress. The crucian carp were chilled using a cold acclimation intelligent chilling equipment from 20 °C to 5 °C. They were divided into three groups: the control group (E1), the negative control group (E2), and the 100 mg/L vitamin E (E3) solution. Three different temperature points (20 °C, 10 °C, and 5 °C) were used to collect, test, and analyze the samples. The findings demonstrated that in the E3 treatment group, phosphoenolpyruvate carboxykinase, acetyl coenzyme A carboxylase, total cholesterol, urea nitrogen, triglyceride, and fatty acid synthase contents were significantly lower under cooling stress than those in the E1 and E2 treatment groups (P

Background:. In this study, the clinical efficacy of a new framework construction technique for costal cartilage rhinoplasty was investigated. Methods:. From January 2020 to February 2022, patients who underwent rhinoplasty in the department of plastic and reconstructive surgery of Zhejiang Provincial People’s Hospital were enrolled. The sixth costal cartilage was made into the nasal columellar support graft (strut), the nasal tip graft integrated scaffold, and the septal extension grafts. The strut and septal extension grafts were sutured to construct the framework. Adobe Photoshop 6.0 was used to measure a series of aesthetic indices preoperatively and at 9 months postoperatively, including the nasal length, tip projection, nasofrontal angle, columellar/lobular angle, and nasolabial angle. The visual analog scale score and rhinoplasty outcome evaluation score were used to assess patient satisfaction. A paired t test was used for data analysis, and a P value of less than 0.05 was considered to indicate statistical significance. Results:. A total of 65 patients aged 19–43 years (average age 26.0 y) were enrolled. No short-term complications occurred. All patients were followed up for 9–23 months. There were statistically significant differences in all measurements (P < 0.05), which suggested that the aesthetic defects of the nose were corrected and that no obvious deflection or rotation of the nasal tip occurred. The visual analog scale score and rhinoplasty outcome evaluation score indicated a significant improvement in patient satisfaction (P < 0.05). Conclusions:. This new framework construction technique for costal cartilage rhinoplasty can reduce the risk of framework deflection and nasal tip rotation and provide a satisfactory nasal columellar shape.

Currently, “being alive but not fresh” is a common problem existing in the aquatic products and no guiding prediction can be given for the quality of the shelf life due to the lack of systematic studies. In this connection, the life vitality indication and nutritional quality changes of Patinopecten yessoensis during the period of anhydrous storage and transportation at ice temperatures were probed into in this study, with findings showing that indicators such as the edge shrinkage rate, stimulus response time, and shell closure force can all be used to characterize the post-capture status of scallops and are associated linearly with their shelf life. Specifically, regarding nutritional quality, with the extension of anhydrous storage and transportation at ice temperatures, the water content of soft tissues decreased significantly and water loss became serious, during which glycogen as energy storage material fluctuated greatly. Meanwhile, the bitter amino acid content decreased significantly after scallops were out of water, and the fresh and sweet taste was thus revealed. The brightness of the adductor muscle played a dominant role in the change of color difference, which showed an upward trend within 5 d after leaving water, and decreased with the fluctuation of vitality in the later period. In addition, electronic nose and electronic tongue can effectively distinguish scallop samples with different shelf life based on differences in odor and taste. The main aroma sources of fresh seafood, that is, aldehydes (trans-2-octenal and trans-2-undecylenic aldenyde) and esters (methyl butyrate) showed a downward trend, while the contents of non-pleasant flavor substances (ethanethiol, methanethiol, isopropanol, n-propanol, etc.) showed an upward trend. In terms of taste, CTS (salty), ANS (sweet), and SCS (bitter) taste values were negatively correlated with storage and transportation time. It is concluded that the quality of the Patinopecten yessoensis can be maintained within 5 days of anhydrous storage and transportation at ice temperatures.

Owing to the high hydrocarbon content and photosynthetic efficiency of phytoplankton, planktonic microalgae account for the main source of petroleum hydrocarbons in marine and continental petroliferous basins. Therefore, the presence of algal residues in such basins is considered an important indicator of high-quality hydrocarbon source rocks. The southwestern region of the Qaidam Basin, characterized by lacustrine deposits from the Paleogene, has substantial petroleum resources. However, studies on the development of source rocks and the mechanisms involved in their organic matter enrichment are insufficient, thus hampering ongoing shale oil exploration efforts. Herein, abundant Botryococcus fossils were discovered in the main source rocks of the upper member of Xiaganchaigou Formation of Paleogene in Southwest Qaidam, accounting for >50 % of the total organic matter content. The palynofacies assemblages reflected relatively distal and oxygen-rich sedimentary environments. The algae-rich rocks usually exhibited distinctive laminated structures alternately composed of quartz-feldspathic, clay and carbonate laminae, indicating periodic climate fluctuations. The Botryococcus, which was mainly preserved in the coarse-grain quartz-feldspathic laminae, probably reflected heavy precipitation conditions and subsequently high nutrient inputs. Finally, the oxygen-rich, low salinity and eutrophic water was likely taking form which benefit the growth of these hydrobiontic algae. These algal-rich shales exhibited typically high hydrogen index (IH) and total organic carbon (TOC) values, indicating their high hydrocarbon-generation potential. Thus, they are important marker beds for high-quality source rocks in petroliferous basins. Concurrently, the coarse-grained detrital mineral laminae displayed excellent reservoir physical properties, probably providing sufficient reservoir space for planktonic algae-derived liquid hydrocarbon. Therefore, these Botryococcus-rich source rocks might represent important targets for ongoing shale oil exploration.

The establishment of the ecological protection redline (EPR) in China plays a crucial role in safeguarding key ecological functional areas and sustaining biodiversity. Besides their significance, the impact of these redlines on regional carbon sequestration remains unclear due to current inconsistencies in their implementation and adjustment. This paper conducts a thorough analysis of the evolution of two EPR versions on Hainan Island and employs the InVEST model to assess the effects of two EPR areas on regional carbon storage between 2000 and 2020. Additionally, a spatial autoregressive model is integrated to explore the socioeconomic and environmental driving mechanisms behind changes in regional carbon storage, thereby revealing complex interactions between ecological protection policy adjustments and ecosystem services of carbon storage. Key findings include: (1) The 2022 EPR area has improved the management of ecological protection boundaries, permanent cultivated land, and urban development boundaries, outperforming the 2016 version, particularly in control of construction land and water resource management. (2) The EPR2022 exhibits higher carbon storage per area, indicating a more effective zoning system. The increase in carbon storage is primarily observed in transitions from cultivated land to forest, grassland to forest, and water bodies to grassland. (3) Elevation exhibits a relatively strong positive correlation with carbon storage within the EPRs, with high-altitude areas typically encompassing the core zones of tropical rainforest national parks that possess diverse vegetation and stable ecosystems, conducive to maintaining or increasing carbon storage. Evaluating the efficacy of the EPR in enhancing regional carbon storage is critical for refining management strategies and achieving carbon neutrality goals. Future strategies should incorporate a comprehensive analysis of ecosystem services of carbon storage into the EPR framework to enhance multiple ecosystem functions, thereby establishing a resilient and functionally stable ecological landscape. This research introduces an insightful estimation method crucial for developing more effective ecological protection policies and provides valuable insights for managing ecological protection areas globally.

As a Free Trade Port in China, the protection of ecological environment and high-quality tourism development on Hainan Island have become a national strategy. Exploring the coupling relationship of the two subsystems holds great practical significance for promoting the island’s international demonstration area and high-quality development. Based on the characteristics of international islands, a comprehensive index system is established to explore the coupling coordination evolutionary process and interactive stress relationship of tourism development and the ecological environment in Hainan Island from 2005 to 2020. The results show: (1) From 2005 to 2019, the comprehensive evaluation index of the two subsystems of Hainan Island generally showed a continuous growth trend. (2) From 2005 to 2019, the coupling degree of the two subsystems of Hainan Island strengthened continuously, the coupling coordination degree increased steadily from a moderate imbalance to good coordination, and tourism development and the ecological environment entered a stage of parallel and benign interactive development. (3) In 2020, affected by COVID-19, the tourism development indicators declined rapidly, while the ecological environment still maintained a rising trend. (4) Based on the inverted U-shaped curve model, when the tourism development index reaches 0.7269, the ecological environment quality reaches the maximum value of 0.7956, which is the best state between the two subsystems. The ecological environment and tourism development of Hainan Island are generally in a benign interaction, but there are signs of interactive stress starting in 2017. In the future, the pressure on the ecological environment of Hainan Island will increase.

Mangrove restoration is recognized as an effective strategy for enhancing the carbon storage capacity of natural ecosystems, advancing toward the “carbon neutrality” goal. The carbon storage effects of ecological restoration efforts remain insufficiently understood as previous studies have focused on carbon storage dynamics in ecosystems, yet the specific impacts of targeted mangrove restoration are less explored. This study utilizes multi-temporal remote sensing data and actual restoration data from Dongzhai Harbor Hainan Island to identify the mangrove wetland coverage and quantify the spatiotemporal evolution of carbon storage under various restoration efforts using the InVEST model. Additionally, we employed the PLUS model to simulate and compare carbon storage potential under multiple development goals. The findings reveal the following: (a) Mangrove restoration significantly increased the area of land with high carbon sink capability, resulting in a regional carbon storage increase of 210,001.68 tons from 2015 to 2021, with 97% of this increase attributable to ecological restoration. (b) Mangrove coverage is crucial for regional carbon storage, with an average of 443 tons of carbon stored per hectare. Decreases in carbon storage occurred mainly during the conversion of mangroves to aquaculture, and forests/agriculture to residential areas. Increases in carbon storage were seen in the reverse transitions. (c) Comparing the scenarios focused solely on mangrove protection with cultivated land protection, the carbon storage in Dongzhai Harbor is projected to reach its maximum by 2045 under the carbon storage priority scenario. Our findings build a scientific foundation for formulating effective mangrove conservation and restoration strategies.

Eugenol is a widely used fishery anesthetic. This study investigated the effects of various concentrations of eugenol on blood physiological and biochemical indexes, and muscle flavor, in crucian carp (Carassius auratus). To determine the appropriate concentration of eugenol anesthetic for use in crucian carp transportation and production operations, we evaluated seven anesthesia groups of 20, 30, 40, 50, 60, 70, and 80 mg/L and one control group (without eugenol) to determine the effects on blood physiological and biochemical indexes, and muscle flavor. The red blood cells and platelets of crucian carp decreased significantly (p < 0.05) with eugenol treatment. With increasing eugenol concentration, the white blood cells and hemoglobin did not change significantly, whereas lactate dehydrogenase, alkaline phosphatase, alanine aminotransferase, and aspartate aminotransferase increased significantly (p < 0.05). The content of phosphorus, magnesium, and sodium increased after anesthesia, whereas the content of potassium decreased with increasing eugenol concentration. After anesthesia, the content of albumin and total protein in the serum decreased with increasing eugenol concentration (p < 0.05); triglyceride first increased and subsequently decreased (p < 0.05); blood glucose content first increased and then decreased (p < 0.05); and no significant difference was observed in total cholesterol content (p > 0.05). No significant difference was observed in muscle glycogen and liver glycogen content after eugenol anesthesia (p > 0.05). The eugenol-based anesthesia test did not indicate major liver histomorphological effects, but the very small number of gill sheet edema cases observed requires further study. Analysis of electronic nose data indicated that eugenol treatment affected the flavor of the fish. The anesthesia concentration of 20–80 mg/L had some effect on the physiology and biochemistry of crucian carp, thus providing a reference for the application of eugenol in crucian carp transportation and experimental research.

In this paper, the quality change of Yesso scallop (Patinopecten yessoensis) in the process of anhydrous storage and transportation after cold acclimation and induced dormancy was studied, and the regulation mechanism of quality degradation during storage and transportation in the process of gradient chilling stress and drying exposure was further explored. The results show that, when transferred from hydrous to anhydrous states, the breathing pattern of the scallops changed from aerobic to anaerobic. Their gill filaments were altered and their apparent vitality constantly declined, which was reflected by the edge shrinkage of the pallium and the direct proportions of the edge reduction rate and the stimulus response period. After being in the anhydrous state for 4 d, the AEC value dropped to 67.59%. At this time, if they were placed under hydration again, the scallops resumed a good growth state. By proteomics analysis, it was revealed that cold acclimation and dry exposure mainly led to changes in biological functions and pathways, such as mitochondrial inner membrane and ATP hydrolysis activity. In addition, it can be seen from the functional annotation and enrichment analysis of the metabolite KEGG that cold acclimation promoted the purine metabolism of scallops, while dry exposure inhibited the metabolism of saturated fatty acids. In this study, the infrared sensing mode was used for the first time, too, in order to record the heart-rate changes of the scallops during circulation, which shows that non-destructive vitality monitoring of Lamellibranchia is feasible.

The anaesthetic effect of vanillin on crucian carp was investigated using different concentrations of vanillin, with a nonvanillin control. The effective concentration range of vanillin anaesthesia was determined from the behavioural characteristics of crucian carp during the anaesthesia onset and recovery phases. Physiological and biochemical indices, and the electronic nose response to the fish muscle, were measured over the range of effectiveanaestheticc concentrations. An increased concentration of vanillin shortened the time taken to achieve deep anaesthesia but increased the recovery time. The levels of white blood cells, red blood cells, haemoglobinn, platelets, alanine aminotransferase, alkaline phosphatase, lactate dehydrogenase, phosphorus, potassium, magnesium, total protein, and serum albumin were lower than the control in the vanillin treatment group. Triglycerides and total cholesterol were not significantly affected. Histology showed no effect of vanillin on the liver, except at 1.00 g/L vanillin. Vanillin resulted in a nondose-responsive effect on the gill tissue, increasing the width and spacing of the gill lamellae. E-Nose analysis of the carp-muscle flavour volatiles was able to distinguish between different vanillin treatment concentrations. GC-IMS identified 40 flavour compounds, including 8 aldehydes, 11 alcohols, 10 ketones, 2 esters, and 1 furan. Vanillin had aanaestheticic effect on crucian carp and these findings provide a theoretical basis for improving the transport and experimental manipulation of crucian carp.

The rapid expansion of cities in tropical regions has triggered a series of problems such as the destruction of rare natural resources and decreases in the environmental resource carrying capacity and ecological security, which seriously threaten the sustainable development of tropical cities. In this study, the city of Sanya, Hainan, China, is taken as an example. A bottom-line ecological security pattern is constructed based on the remote sensing ecological index (RSEI) and the patch-generating land use simulation (PLUS) for urban growth boundary (UGB) delineation. The results show that Sanya has a good ecological background, but the overall ecological quality decreased from 2014 to 2018 due to the expansion of construction in hot spot areas. Under the natural growth scenario, the urban built-up area in Sanya in 2030 will be 73.81% greater than in 2018, mainly occupying a large amount of agricultural and ecological space, and urban expansion will not be effectively suppressed. Delineation of the UGBs combined with the ecological constraints can effectively protect the regional ecological security and control the urban sprawl, which is relatively consistent with the current planning. The results of this study demonstrate that the RSEI-PLUS-based UGB delineation perspective has a high scientific and applicability, and they provide a reference for the coordinated ecological–economic sustainable development of ecologically fragile cities in tropical areas.
