An efficient one-pot synthesis of novel pyrazolo[3,4- b][1,8]naphthyridine and pyrazolo[3,4- d]pyrimido[1,2- a]pyrimidine derivatives has been investigated from the reaction of 2-amino (pyrimidine or pyridine), aromatic aldehydes and 3-methyl-1-phenyl-2-pyrazolin-5-one. All synthesized compounds were evaluated for in vivo anti-inflammatory activity using carrageenan-induced rat paw edema assay. Compounds 4a, 4e, 4g, 5a, 5b (80%) and 5c (86%) showed good to excellent results when compared with the anti-inflammatory active standard drug diclofenac Na (93%). On the basis of the structure-activity relationship, the anti-inflammatory activity of pyrazolo[3,4- d]pyrimido[1,2- a]pyrimidine derivatives has been found to be much better than pyrazolo[3,4- b][1,8]naphthyridine derivatives. The present study describes the synthesis and in vitro anticancer activity of novel piritrexim analogs against human breast cancer cell line MCF-7 and a normal Vero monkey cell line. The scheme involves the formation of 3-(4-nitrophenyl)-4-phenyl-1,8-naphthyridin-2-amine accomplished by the condensation reaction of 4-nitrophenyl acetonitrile, aromatic aldehyde, and 2-aminopyridine using acetic acid as a catalyst. The selected synthesized compounds have been screened against a human breast cancer cell line MCF-7 and a normal Vero monkey cell line, out of which the compounds 4b, 4e, 4f exhibited higher potency (GI [<10 µg/mL)], and compounds 4a, 4c show moderate to good activity (GI 41.4 and 12.4 µg/mL ) towards human breast cancer cell line MCF-7. Docking study on dihydrofolate reductase enzyme receptor (DHFR) and epidermal growth factor receptor reveals that compounds 4a, 4b, 4c, and 4e show moderate to good binding along with acceptable glide scores. An efficient P2O5 mediated protocol for the synthesis of pyrido[2,3‐d:6,5‐d]dipyrimidines has been developed through one‐pot three‐component condensation of aromatic aldehydes, barbituric acid/thiobarbituric acid, and 1,3‐dimethyl‐6‐amino‐uracil in ethanol. All the synthesized products were screened for their in vitro antitubercular activity, and results reveal that most of the compounds exhibited moderate to good activity against Mycobacterium tuberculosis H37RV strain. Fused pyrazolopyranopyrimidines containing both biologically active pyranopyrazole and pyranopyrimidine compounds were synthesized using a one-pot, four-component reaction of ethyl aceto acetate, hydrazine hydrate, benzaldehydes, and thiobarbituric acid without catalyst in ethanol. All target compounds obtained in very good to excellent yields over short reaction times by adapting a simple workup procedure. All the synthesized compounds exhibited good to excellent antituberculosis activities. The results shown that 4-(1H-imidazol-2-yl)-3-methyl-7-thioxo-4,6,7,8-tetrahydropyrazolo[4’,3′:5,6]pyrano[2,3-d] pyrimidin-5(1H)-one (5a) is a good antitubercular agent, as good as the standard streptomycin drug, based on minimum inhibitory concentration (MIC). An ecofriendly route has been investigated for the synthesis of 4-(4-nitro-phenyl)-2-phenyl-1,4-dihydro-benzo[4,5]imidazo[1,2-a]pyrimidine-3-carboxylic acid ethyl ester derivatives by one-pot, three-component condensation of ethyl benzoylacetate, aromatic aldehydes, and 2-amino benzimidazole using 260 mol% of citric acid as reaction mediator. Citric acid is an inexpensive, nontoxic, and green medium with smoothly activates the rate of reaction. The synthesized compounds were assessed for in vitro antimycobacterial activity againstMycobacterium tuberculosisH37RV strain using the microplate alamar blue assay (MABA). The results indicate that among all the synthesized compound series, P-4 and P-9 compounds illustrate effective activity with a minimum inhibitory concentration of 25 µg/ml. [ABSTRACT FROM PUBLISHER]
