Abstract

The identification of genes involved in replicative stress is key to understanding cancer evolution and to identify therapeutic targets. Here, we show that CDK12 prevents transcription-replication conflicts (TRCs) and the activation of cytotoxic replicative stress upon deregulation of the MYC oncogene. CDK12 was recruited at damaged genes by PARP-dependent DDR-signaling and elongation-competent RNAPII, to repress transcription. Either loss or chemical inhibition of CDK12 led to DDR-resistant transcription of damaged genes. Loss of CDK12 exacerbated TRCs in MYC-overexpressing cells and led to the accumulation of double-strand DNA breaks, occurring between co-directional early-replicating regions and transcribed genes. Overall, our data demonstrate that CDK12 protects genome integrity by repressing transcription of damaged genes, which is required for proper resolution of DSBs at oncogene-induced TRCs. This provides a rationale that explains both how CDK12 deficiency can promote tandem duplications of early-replicated regions during tumor evolution, and how CDK12 targeting can exacerbate replicative-stress in tumors., (© 2024. 30. Emissions of maritime transport: a European reference system.

Abstract

Emissions from ships have recently received more attention since they have become a significant concern for air quality in harbours and port cities. This paper presents the methodology for a comprehensive maritime transport database of activity data, specific energy consumption, emission factors, and total emissions that have been developed within the European EX-TREMIS project. The model is built upon 3 modules: the fleet module, the transport activity module, and the emission module. The fleet module defines the ship categories, the loading capacities, and the engine characteristics of the different vessels by using EUROSTAT data, Sea Web Lloyd's database, and international literature. The transport activity module transforms total cargo handled (mainly based on EUROSTAT data and CEMT statistics) into ship-equivalents. These ship-equivalents are further transformed into ship-hours. The emission module calculates energy uses and CO(2), NO(X), SO(2), CO, HC, CH(4), NMHC, PM emissions from the resulting maritime activities. We have used technology based emission factors to take into account the technological evolution of vessels. To illustrate this new methodology, we present some results (emissions, fuel consumption and emission factors) for different countries. The overall methodology as well as the results and the country specific energy consumption and emission factors per ship type and size class can be extracted from the EX-TREMIS website (www.ex-tremis.eu). Our results contribute to more accurate estimates of emissions and air quality assessments in coastal cities and ports.

Abstract

The main aim of this work is to compare the measurements of paraxial slices of the standard Denta-Scan technique and a new technique (Occlusal technique) in toothless patients. On a more specific level, our goal is to assess the effect of a change in the reference plane of computer axial tomography (CAT), for which the Denta-Scan system is usually employed, by transferring the palate plane to the prosthetic plane, which goes from the center of the external auditory canal to the anterior nasal spine. For the experiment, 15 maxillaries were used from toothless skulls belonging to adult patients who had lost their teeth prior to their death. For each one, a wax rim was made in reference to the Occlusal Plane (Camper's Prosthetic Plane) and a 10 mmx 3.3 mm reference implant was placed. Two tomography scans were made of each skull; one using the conventional technique, and the other using Camper's Prosthetic Plane. The tomograph used was of the helicoid type (Phillips MX8000), with 1.3 mm slices every 0.6 mm. To perform the comparative measurements between the two techniques, a VIS Caliper (Poland), was employed, and the data recorded in a table with the reference of each sample and technique. This was repeated with all 15 skulls. The results obtained were evaluated with the measurement of the reference implant for each technique. It was found that the standard technique (Horizontal Plane) showed 19.20% magnification, as opposed to the technique under examination (Camper's Occlusal Plane), which showed 16.5% magnification. The study showed distortions in each measurement as regards the real measurements of the reference implants. These distortions were less significant in the case of the Occlusal Plane technique than in the Horizontal Technique; however neither of them fully matched the measurements of the reference implant.
