Abstract Three‐phase four‐leg inverters are widely used in power systems due to their inherent ability to handle unbalanced loads. However, with conventional 3D space vector pulse width modulation (3D‐SVPWM), high harmonics of large amplitude exist in the output voltage around the switching frequency and its integer multiples leading to noise in the load motor and limiting the application range of the inverter system. The passband amplification of conventional sinusoidal filters and their cutoff frequency vary with the load, so it is particularly important to investigate the regulation strategy with harmonic suppression. In this study, an improved 3D discontinuous pulse width modulation (M3D‐DPWM) strategy is proposed based on 3D‐SVPWM. By reducing the conventional 3D‐SVPWM switching segments and changing the vector synthesis sequence of the last half switching cycle, the output phase voltage period is reduced to half of the original one, which achieves the purpose of suppressing the output voltage at the switching frequency and its odd multiple high harmonics without changing the fundamental characteristics and adding additional circuits. In contrast, the conventional 3D‐SVPWM switching frequency is increased to 20 kHz to achieve similar performance. Finally, the effectiveness of the proposed strategy is verified by simulation and experiment.

ObjectivesA new Spar-type floating wind turbine platform design scheme is proposed for a 15 MW floating wind turbine, and its performance is analyzed. MethodsBased on potential flow theory, the frequency domain hydrodynamic calculation of the platform is carried out and the wave loads and motion response characteristics of the platform under different wave directions are analyzed. The blade element momentum method is applied to calculate the aerodynamic load of the impeller and make it equivalent to the wind coefficient in order to calculate the time-domain response of the platform under the combined effect of wind, waves and currents under different working conditions, while also analyzing the force situation of the mooring lines. ResultsThe results show that the heave, roll, and pitch of the platform have peak points at the natural frequency of its motion, and the positions of occurrence are all far from the main energy region of the waves. Under the action of wind, waves and currents, compared to the working conditions, the maximum pitch angle of the platform under extreme conditions is smaller, but the amplitude of maximum surge, heave and mooring tension are greater than the working conditions. ConclusionsThe newly designed Spar-type floating wind turbine platform has good stability and hydrodynamic performance. Under the designed environmental loads, its response and mooring force can meet the requirements of the specifications, ensuring the safety and stability of the wind turbine.

Abstract The aim of this study was to develop a dynamic nomogram combining clinical and imaging data to predict malignant brain edema (MBE) after endovascular thrombectomy (EVT) in patients with large vessel occlusion stroke (LVOS). We analyzed the data of LVOS patients receiving EVT at our center from October 2018 to February 2023, and divided a 7:3 ratio into the training cohort and internal validation cohort, and we also prospectively collected patients from another stroke center for external validation. MBE was defined as a midline shift or pineal gland shift > 5 mm, as determined by computed tomography (CT) scans obtained within 7 days after EVT. A nomogram was constructed using logistic regression analysis, and its receiver operating characteristic curve (ROC) and calibration were assessed in three cohorts. A total of 432 patients were enrolled in this study, with 247 in the training cohort, 100 in the internal validation cohort, and 85 in the external validation cohort. MBE occurred in 24% (59) in the training cohort, 16% (16) in the internal validation cohort and 14% (12) in the external validation cohort. After adjusting for various confounding factors, we constructed a nomogram including the clot burden score (CBS), baseline neutrophil count, core infarct volume on CTP before EVT, collateral index, and the number of retrieval attempts. The AUCs of the training cohorts were 0.891 (95% CI 0.840–0.942), the Hosmer–Lemeshow test showed good calibration of the nomogram (P = 0.879). And our nomogram performed well in both internal and external validation data. Our nomogram demonstrates promising potential in identifying patients at elevated risk of MBE following EVT for LVOS.

Numerous offshore wind turbines (OWTs) with bucket foundations have been installed in seismic regions. Compared to the relative development of monopiles (widely installed), seismic design guidelines for bucket-supported OWTs still need to be developed. Moreover, scour around bucket foundations induced by water–current actions also creates more challenges for the seismic design of OWTs. In this study, a simplified seismic analysis method is proposed that incorporates the soil–structure interaction (SSI) for the preliminary design of scoured bucket-supported OWTs, aiming to balance accuracy and efficiency. The dynamic SSI effects are represented using lumped parameter models (LPMs), which are developed by fitting impedance functions of the soil–bucket foundation obtained from the four-spring Winkler model. The water–structure interaction is also considered by the added mass in seismic analysis. Based on the OpenSees 3.3.0 platform, an integral model is established and validated using the three-dimensional finite element method. The results indicate that the bucket-supported OWT demonstrates greater dynamic impedance and first-order natural frequency compared to the monopile-supported OWT, which has an increased seismic response. Seismic spectral characteristics and intensities also play an important role in the responses. Additionally, scour can change the bucket impedance functions and the frequency characteristics of the OWT system, leading to a significant alteration in the seismic response. Scour effects may be advantageous or disadvantageous, depending on the spectral characteristics of seismic excitations. These findings provide insights into the seismic response of bucket-supported OWTs under scoured conditions.

The purpose of this study is to assess effects of two different simulation methods (i.e., interfaces with a single spring-slider system and interfaces with double spring-slider systems) for interactions between reinforcement and the surrounding medium on the performances of geosynthetic-reinforced soil (GRS) structures when conducting numerical analyses. The fundamental difference between these two methods is the number of the spring-slider systems used to connect the nodes of structural elements simulating the geosynthetic reinforcement and the points of solid grids simulating the surrounding medium. Numerical simulation results of pull-out tests show that both methods reasonably predicted the pullout failure mode of the reinforcement embedded in the surrounding medium. However, the method using the interfaces with a single spring-slider system could not correctly predict the interface shear failure mode between the geosynthetics and surrounding medium. Further research shows that these two methods resulted in different predictions of the performance of GRS piers as compared with results of a laboratory load test. Numerical analyses show that a combination of interfaces with double spring-slider systems for reinforcement between facing blocks and interfaces with a single spring-slider system for reinforcement in soil resulted in the best performance prediction of the GRS structures as compared with the test results. This study also proposes and verifies an equivalent method for determining/converting the interface stiffness and strength parameters for these two methods.

Abstract KRAS mutation occurs in nearly 30% of human cancers, yet the most prevalent and oncogenic KRAS(G12D) variant still lacks inhibitors. Herein, we designed a series of potent inhibitors that can form a salt bridge with KRAS’s Asp12 residue. Our ITC results show that these inhibitors have similar binding affinity with both GDP-bound and GTP-bound KRAS(G12D), and our crystallographic studies reveal the structural basis of inhibitor binding-induced switch-II pocket in KRAS(G12D), experimentally confirming the formation of a salt bridge between the piperazine moiety of the inhibitors and the Asp12 residue of the mutant protein. Among KRAS family proteins and mutants, both ITC and enzymatic assays demonstrate the selectivity of the inhibitors for KRAS(G12D); and the inhibitors disrupt the KRAS–CRAF interaction. We also observed the inhibition of cancer cell proliferation as well as MAPK signaling by a representative inhibitor (TH-Z835). However, since the inhibition was not fully dependent on KRAS mutation status, it is possible that our inhibitors may have off-target effects via targeting non-KRAS small GTPases. Experiments with mouse xenograft models of pancreatic cancer showed that TH-Z835 significantly reduced tumor volume and synergized with an anti-PD-1 antibody. Collectively, our study demonstrates proof-of-concept for a strategy based on salt-bridge and induced-fit pocket formation for KRAS(G12D) targeting, which warrants future medicinal chemistry efforts for optimal efficacy and minimized off-target effects.

As a leaf homologous organ, soybean pods are an essential factor in determining yield and quality of the grain. In this study, a recognition method of soybean pods and estimation of pods weight per plant were proposed based on improved YOLOv5 model. First, the YOLOv5 model was improved by using the coordinate attention (CA) module and the regression loss function of boundary box to detect and accurately count the pod targets on the living plants. Then, the prediction model was established to reliably estimate the yield of the whole soybean plant based on back propagation (BP) neural network with the topological structure of 5-120-1. Finally, compared with the traditional YOLOv5 model, the calculation and parameters of the proposed model were reduced by 17% and 7.6%, respectively. The results showed that the average precision (AP) value of the improved YOLOv5 model reached 91.7% with detection rate of 24.39 frames per millisecond. The mean square error (MSE) of the estimation for single pod weight was 0.00865, and the average coefficients of determination R2 between predicted and actual weight of a single pod was 0.945. The mean relative error (MRE) of the total weight estimation for all potted soybean plant was 0.122. The proposed method can provide technical support for not only the research and development of the pod’s real-time detection system, but also the intelligent breeding and yield estimation.

This study conducted plane-strain scaled model tests to investigate the deformation characteristics of geosynthetic reinforced soil (GRS) abutments subjected to vertical loads. Setback distance, i.e., the distance between the back of the abutment facing and the front of the loading plate, was chosen as the investigated influencing factor since it is one of the most frequently used variables by engineers for the design of GRS abutments. This study analyzed the settlements at the top of the abutment, the lateral displacements of the abutment facing, and the volumetric deformations of the abutment under the applied vertical loads. Test results showed that increasing the setback distance could effectively reduce the deformations of the GRS abutment. There existed an optimum setback distance and further increasing the setback distance beyond this optimum value did not have a significant effect on reducing the abutment deformations. The vertical, lateral, and total volumetric deformations of the GRS abutment showed an approximately linear increase with the increase of the applied vertical loads. The lateral volumetric deformations of the GRS abutment were larger than its vertical volumetric deformations and therefore the total volumetric strains of the GRS abutment were not zero based on the test results. However, the theory of zero volume change may still be suitable for the deformation calculation of the GRS abutment since the values of the volumetric strains were minimal. The measured maximum lateral facing displacements were compared with the calculated values using the US Federal Highway Administration (FHWA) method, which assumes zero volume change of the GRS abutment under vertical loads. Comparison results indicated that the FHWA method overestimated the maximum lateral facing displacements of the GRS abutment under vertical loads. An improved method was proposed in this study to calculate the maximum lateral facing displacements under vertical loads based on the theory of zero volume change and a revised distribution of the settlements at the top of the GRS abutment. Results showed that the improved method could better predict the maximum lateral facing displacements as compared to the FHWA method.

The growth process of soybean plants needs a lot of water. The rapid detection of canopy wilting of soybean under drought stress is of great significance for soybean variety breeding, cultivation regulation and fine management. Aiming at the problems of cumbersome and time-consuming when the traditional chemical technology was used to determine soybean wilting index, a calculation method of wilting index for soybean canopy was proposed in this study based on multispectral images’ Fourier transform. Suinong 26, a northeast soybean variety, was taken as the object. First, four kinds of soybean multispectral images of green, red, red-edge and near-infrared channels were acquired by a Sequoia multispectral camera. Second, based on the multispectral reflection image preprocessed by median filter and mean filter, the target area of a multispectral image of the soybean canopy was extracted by the iterative threshold method and affine transformation algorithm, and the effective segmentation rate was 97.02%. In addition, Fourier transform was used to analyze the spectrum characteristics of the soybean canopy’s multispectral image. When the spectrum radius of each channel was 50, the energy reached more than 98% and was concentrated in the low-frequency region of the spectrum center. Finally, according to the difference between the low-frequency DC component and the proportion of total energy in the spectral radius of the multispectral images under normal and drought treatment, a calculation model of the soybean wilting index was constructed based on the energy spectrum of Fourier transform. The results showed that the difference of the wilting index between normal and drought treatment for the four channels (green, near-infrared, red and red-edge) was 2.38, 3.11, 3.56 and 4.11, respectively. The effectiveness of the wilting index was verified and analyzed by using the average leaf inclination angle. The determination coefficient R2 of the four channels between the calculated wilting index and the average leaf inclination angle was more than 0.85. This calculation method can provide a quantitative basis and technical support for the scientific regulation of ecological and morphological phenotypic traits of soybean plants under drought stress.

Reducing the electronic contact resistance at the interfaces of nanostructured materials is a major goal for many kinds of planar and three dimensional devices. In this work, we develop a method to enhance the electronic transport at rough interfaces by inserting a two dimensional flexible and conductive graphene sheet. We observe that an ultra-thin graphene layer with a thickness of 0.35 nm can remarkably reduce the roughness of a sample in a factor of 40%, avoiding the use of thick coatings, leading to a more homogeneous current flow, and extraordinarily increasing the total current compared to the graphene-free counterpart. Due to its simplicity and performance enhancement, this methodology can be of interest to many interface and device designers. [ABSTRACT FROM AUTHOR]
