Newcastle Disease Virus (NDV) genotype VII (GVII) is becoming the predominant strain of NDV in the poultry industry. It causes high mortality even in vaccinated chickens with a common NDV genotype II vaccine (GII-vacc). To overcome this, the killed GVII vaccine has been used to prevent NDV outbreaks. However, the debate about vaccine differences remains ongoing. Hence, this study investigated the difference in chickens' responses to the two vaccines at the molecular level. The spleen transcriptomes from vaccinated chickens reveal that GVII-vacc affected the immune response by downregulating neuroinflammation. It also enhanced a synaptogenesis pathway that operates typically in the nervous system, suggesting a mechanism for the neurotrophic effect of this strain. We speculated that the down-regulated immune system regulation correlated with protecting the nervous system from excess leukocytes and cytokine activity. In contrast, GII-vacc inhibited apoptosis by downregulating PERK/ATF4/CHOP as part of the unfolded protein response pathway but did not affect the expression of the same synaptogenesis pathway. Thus, the application of GVII-vacc needs to be considered in countries where GVII is the leading cause of NDV outbreaks. Background and Aim: African swine fever (ASF) causes disease in pigs with up to 100% mortality rates. There is no effective vaccine to protect against it. This study aimed to perform in silico docking of ASF virus (ASFV) pNP868R protein with potential flavonoid ligands to identify ligands that interfere with mRNA cap formation. Materials and Methods: The ASFV pNP868R protein was tested with hyperoside, isoquercetin, quercetin, and quercitrin in this in silico simulation. ASFV pNP868R protein was extracted from the Research Collaboration for Structural Bioinformatics P rotein Data Bank (RCSB PDB) database with PDB ID 7D8U (https://www.rcsb.org/structure/7D8U). Standard ligands were separated from proteins using UCSF Chimera 1.13. The standard ligand was redocked to protein using AutoDockTools 1.5.6 with the AutoDock4 method for validation. In the docking process, the grid box size was 40 × 40 × 40 Å3 with x, y, and z coordinates of 16.433, -43.826, and -9.496, respectively. The molecular docking process of the proposed ligand-protein complex can proceed if the standard ligand position is not significantly different from its original position in the viral protein's pocket. The root mean square deviation (RMSD), root mean square fluctuation (RMSF), and radius of gyration (RoG) of the hyperoside with the lowest energy binding need to be analyzed with molecular dynamics using Groningen machine for chemical simulation 5.1.1. Results: Molecular docking and dynamic simulation revealed that hyperoside had the most stable and compact binding to the pNP868R protein. Hyperoside binds to the protein at the minimum energy of -9.07 KJ/mol. The RMSD, RMSF, and RoG values of 0.281 nm, 0.2 nm, and 2.175 nm, respectively, indicate the stability and compactness of this binding. Conclusion: Hyperoside is the most likely antiviral candidate to bind to the pNP868R protein in silico. Therefore, it is necessary to test whether this flavonoid can inhibit mRNA capping in vitro and elicit the host immune response against uncapped viral mRNA. [ABSTRACT FROM AUTHOR]

Conception Rate (CR) and Service Conception (S/C) are two parameters for assessing the reproductive performance of cattle. This research was conducted to determine the value of S/C and CR in crossbreeding cattle in the working area of the Public Health Center. The material used in this study was 72 crossbreed cattle aged 3-10 years. Semi-intensive rearing system with AI mating. The results the S/C in this study was 3.9 higher than the ideal standard of 1.6–2. The CR in this study was 58.6%. CR numbers are still relatively low from the ideal, namely 65 -75%. Newcastle Disease Virus (NDV) is an RNA virus that causes death in chickens, even those vaccinated with a commercial vaccine derived from genotype II (GII). Therefore, the NDV genotype VII (GVII) vaccine has been developed, but the host response at the molecular level is obscure. This study used three-week-old twenty chickens and divided them into two groups. The first group was injected with the GVII vaccine on day 14 and day 28 and the second group was the negative control. On day 42, the spleen was harvested. Spleen transcriptomic results showed that GVII vaccine stimulated organism growth and development group consisting of fibroblast growth factor (FGF) signaling, sperm motility, white adipose tissue browning pathway, canonical Wnt or Wnt/B cateninsignaling, and non-canonical Wnt, which comprise of the Wnt/Ca2+ pathway and PCP pathway. Background and Aim: Cysticercosis is a zoonotic disease with a global concern. Estimation of the prevalence and identification of potential risk factors are necessary for the prevention and control of the disease. This study aimed to estimate the seroprevalence of cysticercosis and the correlation of the increased prevalence with several potential risk factors. Materials and Methods: The seroprevalence of cysticercosis was conducted using an enzyme-linked immunosorbent assay (ELISA), developed by the Institute of Tropical Medicine Antwerp, to detect Cysticercus cellulosae. This study used serum samples from 62 pigs taken from two regencies on Timor Island. The data analysis was performed using SPSS software 20.0 (IBM Corp., NY, USA) to evaluate ELISA results and the strength of the relationship between risk factors and the prevalence of disease using the odds ratio (OR). Results: Serum samples from 18 out of the 62 pigs were found to be positive; the seroprevalence of cysticercosis was 29%. The results showed that an extensive farming system led to a higher prevalence of cysticercosis compared to an intensive farming system, namely, 10 out of 18 (56.6%), and that the possibility of identifying cysticercosis in pigs in an extensive farming system was 5 times greater than that in pigs in an intensive farming system. In addition, the results showed that nine out of 18 households who did not have toilet facilities were found to be seropositive, indicating a significant relationship between the risk factor of toilet availability with cysticercosis in pigs, with an OR of 4.5. In addition, the results showed that there was no significant relationship between the risk factor of the feed source and the prevalence of cysticercosis in pigs. Conclusion: It can be concluded that the seroprevalence of cysticercosis was 29% in domestic pigs of Timor Island. The risk factors of an extensive pig farming system and toilet availability in community houses were significantly related to the possibility of cysticercosis on Timor Island. [ABSTRACT FROM AUTHOR]

The genomes of two newly emerged Newcastle disease virus strains, chicken/Indonesia/Mega/001WJ/2013 and chicken/Indonesia/Cimanglid/002WJ/2015, from disease outbreaks in chickens in Indonesia are reported. Phylogenetic analysis of different genotypes of Newcastle disease virus using the F gene coding sequences suggests that these two strains belong to genotype VII.2, in class II of avian paramyxoviruses., (Copyright © 2020 Doan et al.)

Here, we report two genomes of newly emerged strains of Newcastle disease virus (NDV), Chicken/Indonesia/Tangerang/004WJ/14 and Chicken/Indonesia/VD/003WJ/11, from disease outbreaks in chickens in Indonesia.
