The crosstalk between clathrin-mediated endocytosis (CME) and the autophagy pathway has been reported in mammals; however, the interconnection of CME with autophagy has not been established in plants. Here, we report that the Arabidopsis CLATHRIN LIGHT CHAIN (CLC) subunit 2 and 3 double mutant, clc2-1 clc3-1, phenocopies Arabidopsis AUTOPHAGY-RELATED GENE (ATG) mutants in both autoimmunity and nutrient sensitivity. Accordingly, the autophagy pathway is significantly compromised in the clc2-1 clc3-1 mutant. Interestingly, multiple assays demonstrate that CLC2 directly interacts with ATG8h/ATG8i in a domain-specific manner. As expected, both GFP-ATG8h/GFP-ATG8i and CLC2-GFP are subjected to autophagic degradation, and degradation of GFP-ATG8h is significantly reduced in the clc2-1 clc3-1 mutant. Notably, simultaneous knockout of ATG8h and ATG8i by CRISPR-Cas9 results in enhanced resistance against Golovinomyces cichoracearum, supporting the functional relevance of the CLC2-ATG8h/8i interactions. In conclusion, our results reveal a link between the function of CLCs and the autophagy pathway in Arabidopsis.

Time-lapse elastic full waveform inversion is used to monitor the spatio-temporal evolution of the CO2 plume during and after supercritical CO2 injection based on a series of time-lapse (repeated) cross-well seismic monitoring datasets obtained at the Nagaoka Carbon Capture and Storage (CCS) site in Japan. The full waveform inversion method successfully estimates the time-lapse velocity decrease of up to 30% within a thin 12 m layer, which is consistent with the magnitude and thickness of the well-log measurements. After the second monitoring survey, the velocity decrease becomes stable and gradually extends down dip along pre-existing geological structures. The full waveform inversion results starkly contrast with the previous estimates based on traveltime tomography. The previous traveltime tomography applications only used the traveltime-delays and resulted in low resolution with few percentage change which was not adequate to correctly resolve CO2 injection changes. The datasets pose significant challenges due to background noise, tube waves, apparent non-isotropic source radiation patterns, apparent reservoir velocity anisotropy and missing key acquisition parameters such as the number of stacks per shot point. To overcome these obstacles, we meticulously perform careful data preprocessing integrating both the body waves and tube waves. We develop waveform-based source mechanism estimation to represent non-isotropic source excitation, and then conduct forward modeling studies to constrain the anisotropy model.

Knowledge of collocations facilitates second language (L2) learning by enhancing accuracy and fluency. However, acquiring L2 collocations is often challenging for learners. One factor contributing to this difficulty is incongruency between first and second languages (e.g., "draw distinctions" in English corresponds to do "distinctions" in Japanese). One possible solution is to facilitate semantic restructuring, where learners develop their understanding of L2 words' semantic possibilities. For the restructuring of semantic knowledge to occur, repeated exposure to multiple collocations containing the same node word may be necessary (e.g., "draw attention," "draw a conclusion"). However, how to facilitate semantic restructuring through repeated exposure is still unknown. Particularly, to what extent temporal distribution (i.e., spacing) of multiple collocations affects learning is an important issue to be examined. Based on recent research on collocation learning, the present article argues that a combination of massing and spacing can promote L2 collocation learning from the viewpoint of semantic restructuring. It also discusses some practical implications for effective L2 collocation learning.

SUMMARY: As the high-frequency analogue to field-scale earthquakes, acoustic emissions (AEs) provide a valuable complement to study rock deformation mechanisms. During the load-stepping creep experiments with CO2-saturated water injection into a basaltic sample from Carbfix site in Iceland, 8791 AE events are detected by at least one of the seven piezoelectric sensors. Here, we apply a cross-correlation-based source imaging method, called geometric-mean reverse-time migration (GmRTM) to locate those AE events. Besides the attractive picking-free feature shared with other waveform-based methods (e.g. time-reversal imaging), GmRTM is advantageous in generating high-resolution source images with reduced imaging artefacts, especially for experiments with relatively sparse receivers. In general, the imaged AE locations are found to be scattered across the sample, suggesting a complicated fracture network rather than a well-defined major shear fracture plane, in agreement with X-ray computed tomography imaging results after retrieval of samples from the deformation apparatus. Clustering the events in space and time using the nearest-neighbour approach revealed a group of ‘repeaters’, which are spatially co-located over an elongated period of time and likely indicate crack, or shear band growth. Furthermore, we select 2196 AE events with high signal-to-noise-ratio (SNR) and conduct moment tensor estimation using the adjoint (backpropagated) strain tensor fields at the locations of AE sources. The resulting AE locations and focal mechanisms support our previously assertion that creep of basalt at the experimental conditions is accommodated dominantly by distributed microcracking.

Abstract: The westernmost Nankai Trough, southwest Japan, exhibits a rapid along-strike reduction in plate coupling in the proximity to the subducting Kyushu-Palau ridge. Yet how and to what extent the ridge subduction impacts physical properties at the megathrust have not been investigated. Here we present high-resolution seismic P-wave velocity models along the forearc wedge in the western Nankai Trough derived from full-waveform inversion analyses of seismic refraction data. The velocity models show that where the plate coupling is weak and the plate boundary presumably hosts slow earthquakes, the upper plate exhibits lower seismic velocities indicating higher degree of fracturing over a ~ 100 km length along trough. Intriguingly, the extent of the upper-plate low-velocity features is significantly larger than the surficial width of the Kyushu-Palau ridge, and this low-velocity zone is underthrust by the slab with increased crustal thickness by 2–4 km. Seismic reflection images consistently reveal that the thicker slab crust has appreciable basement roughness extending ~ 60 km from the eastern margin of the Kyushu-Palau ridge beneath the western Shikoku basin. We suggest that such a thicker and rugged slab crust, together with the main body of the Kyushu-Palau ridge, can cause significant fracture zones in the overriding plate, decrease the interplate coupling and produce preferable conditions for shallow slow earthquakes to occur when subducted. The results may also provide structural constraints on the western limit of future megathrust earthquakes in the Nankai Trough. Graphical Abstract

Humans can coordinate their behavior with others through interactions; however, not all pairs can coordinate. From the perspective of predictive processing, different social interaction patterns can be explained by the diversity of individuals' belief strength. To investigate the relationship between coordination and belief strength, we conducted an interaction experiment using a Simon electronic light-sequence game in which participants memorize the order of color sequences. The results of our experiment, involving 23 pairs of participants, revealed diversity in the degree of coordination within pairs and the strength of belief between individuals. Our analysis supports the hypothesis that belief strength explains the success or failure of coordination: Coordination fails when both individuals in a pair have weak beliefs, whereas it succeeds when one person becomes the leader and the other becomes the follower because of the different strengths of their beliefs. Our findings suggest that predictive processing theory can be applied to situations involving social interactions.

For cosmic microwave background (CMB) polarization observations, calibration of detector polarization angles is essential. We have developed a fully remote controlled calibration system with a sparse wire grid that reflects linearly polarized light along the wire direction. The new feature is a remote-controlled system for regular calibration, which has not been possible in sparse wire grid calibrators in past experiments. The remote control can be achieved by two electric linear actuators that load or unload the sparse wire grid into a position centered on the optical axis of a telescope between the calibration time and CMB observation. Furthermore, the sparse wire grid can be rotated by using a motor. A rotary encoder and a gravity sensor are installed on the sparse wire grid to monitor the wire direction. They allow us to achieve detector polarization angle calibration with an expected systematic error of 0.08°. The calibration system will be installed in small-aperture telescopes at Simons Observatory.

The energy transition towards a more sustainable and renewable future is a pivotal global endeavor. Central to this shift for the United States is the critical role of domestically sourced lithium, a key mineral in the production of high-performance batteries essential for electric vehicles and renewable energy storage systems. This has driven the United States to invest heavily in a domestic supply chain for battery-grade lithium to enhance energy security, reduce supply chain vulnerabilities, and foster economic growth by tapping into local resources. A notable example is the Biden Administration’s “American Battery Materials Initiative,” which was included in the $2.8-billion Bipartisan Infrastructure Law (The White House, 2022). The “Salton Sea Known Geothermal Resource Area” in Imperial County, California has been identified as a potential domestic U.S. resource of lithium due to the brine-hosted lithium in the deep subsurface geothermal reservoir. An analysis funded by the U.S. Department of Energy provides an overview of opportunities and challenges associated with developing the lithium resource in the Salton Sea geothermal reservoir, as well as potential environmental and societal impacts to the county and surrounding region. The geologic history of the region suggests that lithium in the subsurface brines could have come from multiple sources, including water and sediments from the Colorado River, which have been periodically deposited over the past several million years; rocks from the mountain ranges surrounding the Imperial Valley; and lithium-bearing volcanic rocks and igneous intrusions from past geologic events. Further, several processes may have concentrated lithium in the brine over time, including evaporative concentration of lithium-bearing water that flowed into the basin and leaching of lithium from the sediments and rocks by the circulating geothermal brines. Geothermal brine production at the Salton Sea Geothermal Field, the area with existing geothermal power plants, has averaged just over 120 million metric tons per year since 2004. Using an approximate lithium brine concentration of 198 parts per million (ppm), the amount of dissolved lithium contained in these produced brines is estimated to be 127,000 metric tons of lithium carbonate equivalent (LCE) per year. The total dissolved lithium content in the well-characterized portion of the Salton Sea Geothermal Reservoir is estimated at 4.1 million metric tons of LCE, and the estimated total resource increases to 18 million metric tons of LCE if assumptions for porosity and total reservoir size are increased to reflect the probable resource extent. Analysts measured lithium concentrations in the reservoir rocks, which were shown to vary with depth and mineralogy. These data were used to help refine conceptual and computer models of the reservoir; specifically, two complementary computer models of the reservoir were developed. Analysts used the first model to simulate the approximate 30-year history of geothermal power production in the area using historical production and reinjection data, then used that model to simulate a 30-year forecasting period. This forecast assumed continued production and reinjection rates at current levels but removes 95% of the lithium from the produced geothermal brine starting January 1, 2024. The model found that lithium recovery declines by more than half, from 0.8 to 0.3 kilograms per second (kg/s). Forecast scenarios that are optimized to both recover lithium and harness geothermal energy are expected to sustain lithium production rates much more effectively.The second model included more detailed simulations of the movement of brine and chemical reactivity of lithium within the reservoir. It showed that the reactions of relatively stable lithium-bearing minerals are slow, and that the primary replenishment mechanism for lithium in the brines is the upward flux of convecting lithium-rich brine from below the producing reservoir. However, these replenishment rates are not fast enough to produce significant increases in lithium, which could limit the long-term sustainability of the lithium resource. It is important to note that these models are preliminary and are based on current understanding of fluid replenishment rates, the minerals present in the geothermal system, and their chemical properties and reactivity. Further work should be undertaken to improve them and the associated predictions. The report also considered potential impacts on regional water resources, air quality, chemical use, and solid waste disposal needs, as well as the seismic risk associated with geothermal power production and lithium extraction activity. These investigations highlighted the need to proceed with good monitoring and verification systems and with appropriate mitigation technologies. However, the analysis illustrates that if these things are done properly, lithium development is not likely to create significant negative environmental impacts.Specifically, expanding geothermal energy production and lithium extraction will have a modest impact on water availability in the region. Initial estimates suggested that ~3% of historically available water supply for the region would be needed for currently proposed geothermal energy and lithium recovery operations; the majority of current water usage is for agriculture. It is not anticipated that expanding geothermal capacity or lithium production would impact the availability or quality of water used for human consumption and will not directly affect the water quality of the Salton Sea. However, the long-term drought conditions in the western United States may restrict future availability of water to the region, which is sourced from the Colorado River.In terms of regional air emissions of all pollutants identified in the analysis (particulate matter, hydrogen sulfide, ammonia, and benzene, expanding geothermal energy and adding lithium extraction overall have a small impact. Chemical use involved in geothermal power production and lithium extraction is consistent with chemical use in industrial settings, and the analysis did not identify any persistent organic pollutants or acutely toxic chemicals among those currently being used. Moving fluids within the subsurface can impact subsurface pressures and stresses, potentially triggering seismic activity. Early in geothermal energy production, increasing seismicity rates in the Salton Sea Geothermal Field correlated strongly with energy production activity; however, that correlation weakened after 1996. Even following the onset of geothermal energy production, seismic hazard in the Salton Sea Geothermal Field has not increased beyond that of the surrounding region.In addition to technical outcomes from the analysis, the report describes an initial effort to incorporate community engagement into lithium research by understanding the local context and priorities and identifying how to effectively communicate to share information and gather feedback. The report includes information about the social and historical context of the region to enable a more holistic understanding of the resource and its potential impact, and identifies key community questions by observing public meetings, visiting the region, and consulting with local organizations. The report provides recommendations about how future research efforts can address community concerns and implement more community-engaged practices. These include developing formal partnerships with local organizations and establishing a community advisory board to facilitate ongoing dialogue and opportunities for feedback. The future work will build on and further refine the models and scenarios presented in the report and strive to deepen engagement with local communities.

Abstract Background Despite recent reports, the effectiveness of postoperative oral nutritional supplementation (ONS) on body weight loss and malnutrition after gastrectomy remains controversial. We aimed to elucidate the effectiveness of ONS especially in octogenarian patients undergoing oncological gastrectomy. Abstract Background Complete endoscopic resection of superficial non-ampullary duodenal epithelial tumors (SNADETs) is technically difficult, especially with an extremely high risk of adverse event (AE), although various endoscopic resection methods including endoscopic mucosal resection (EMR), underwater EMR (UEMR), and endoscopic submucosal dissection (ESD) have been tried for SNADETs. Accordingly, a novel simple resection method that can completely resect tumors with a low risk of AEs should be developed. Aims A resection method of Noninjecting Resection using Bipolar Soft coagulation mode (NIRBS) which has been reported to be effective and safe for colorectal lesions is adapted for SNADETs. In this study we evaluated its effectiveness, safety, and simplicity for SNADETs measuring ≤ 20 mm. Results This study included 13 patients with resected lesions with a mean size of 7.8 (range: 3–15) mm. The pathological distributions of the lesions were as follows: adenomas, 77% (n = 10) and benign and non-adenomatous lesions, 23% (n = 3). The en bloc and R0 resection rate was 100% (n = 13). The median procedure duration was 68 s (32–105). None of the patients presented with major AEs including bleeding and perforation. Conclusions Large studies such as prospective, randomized, and controlled trials should be conducted for the purpose of validating effectiveness, safety, and simplicity of the NIRBS for SNADETs measuring ≤ 20 mm suggested in this study.

Abstract Maintaining eye contact is a fundamental aspect of non-verbal communication, yet current remote communication tools, such as video calls, struggle to replicate the natural eye contact experienced in face-to-face interactions. This study presents a cutting-edge vision-sharing system for android avatars that enables remote eye contact by synchronizing the eye movements of the operator and the avatar. Our innovative system features an eyeball integrated with a wide-angle lens camera, meticulously designed to mimic human eyes in both appearance and functionality. This technology is seamlessly integrated into android avatars, enabling operators to perceive the avatar’s surroundings as if physically present. It provides a stable and immersive visual experience through a head-mounted display synchronized with the avatar’s gaze direction. Through rigorous experimental evaluations, we demonstrated the system’s ability to faithfully replicate the operator’s perspective through the avatar, making eye contact more intuitive and effortless compared with conventional methods. Subjective assessments further validated the system’s capacity to reduce operational workload and enhance user experience. These compelling findings underscore the potential of our vision-sharing system to elevate the realism and efficacy of remote communication using android avatars.

Abstract Background Pancreatic ductal adenocarcinoma tumors exhibit resistance to chemotherapy, targeted therapies, and even immunotherapy. Dendritic cells use glucose to support their effector functions and play a key role in anti-tumor immunity by promoting cytotoxic CD8+ T cell activity. However, the effects of glucose and lactate levels on dendritic cells in pancreatic ductal adenocarcinoma are unclear. In this study, we aimed to clarify how glucose and lactate can impact the dendritic cell antigen-presenting function and elucidate the relevant mechanisms. Methods Glycolytic activity and immune cell infiltration in pancreatic ductal adenocarcinoma were evaluated using patient-derived organoids and resected specimens. Cell lines with increased or decreased glycolysis were established from KPC mice. Flow cytometry and single-cell RNA sequencing were used to evaluate the impacts on the tumor microenvironment. The effects of glucose and lactate on the bone marrow-derived dendritic cell antigen-presenting function were detected by flow cytometry. Results The pancreatic ductal adenocarcinoma tumor microenvironment exhibited low glucose and high lactate concentrations from varying levels of glycolytic activity in cancer cells. In mouse transplantation models, tumors with increased glycolysis showed enhanced myeloid-derived suppressor cell infiltration and reduced dendritic cell and CD8+ T cell infiltration, whereas tumors with decreased glycolysis displayed the opposite trends. In three-dimensional co-culture, increased glycolysis in cancer cells suppressed the antigen-presenting function of bone marrow-derived dendritic cells. In addition, low-glucose and high-lactate media inhibited the antigen-presenting and mitochondrial functions of bone marrow-derived dendritic cells. Conclusions Our study demonstrates the impact of dynamic glycolytic reprogramming on the composition of immune cells in the tumor microenvironment of pancreatic ductal adenocarcinoma, especially on the antigen-presenting function of dendritic cells.

Abstract Background The micro-axial flow pump Impella, a new mechanical circulatory device for cardiogenic shock, is still only available in a limited number of hospitals, due to the facility certification requirements and insufficient evidence of the benefit of introducing Impella in hospitals. This study aimed to evaluate the impact of introducing Impella in hospitals on in-hospital mortality of patients treated with extracorporeal membrane oxygenation (ECMO). Methods Using a nationwide Japanese inpatient database, we identified patients who received ECMO during hospitalization between 1 April 2014 and 31 March 2021. A hospital-level propensity score–matched cohort was created matching hospitals that introduced Impella (exposure group) to those that did not introduce Impella (control group). The inclusion period in each hospital was divided into two time periods according to the time of Impella introduction in the exposure group and the corresponding hospital in the control group (before and after exposure). The primary outcome was in-hospital mortality. Uncontrolled and controlled interrupted time-series analyses involved before–after exposure comparison and exposure–control comparison. Results Out of 34,379 eligible patients, we created a matched cohort of 8351 patients from 86 hospitals with Impella introduction (exposure group) and 7230 patients from 86 hospitals without Impella introduction (control group). In-hospital mortality before and after exposure was 62.5% and 59.3, respectively, in the exposure group; and 66.8% and 63.7%, respectively, in the control group. Uncontrolled interrupted time-series analysis showed no significant level change or trend change in the before–after exposure comparison in both the exposure and the control groups. Controlled interrupted time-series analysis also showed no significant level change (−0.01%; 95% confidence intervals −5.36% to + 5.33%) or trend change (+ 0.10%, −0.30% to + 0.40%) after exposure in the exposure–control comparison. Conclusions This nationwide inpatient database study showed no association between Impella introduction in hospitals and in-hospital mortality of patients who underwent ECMO. Because this study confined itself to analze of the impact of the introduction of Impella solely at the hospital level, further detailed studies are warranted to assess its efficacy at the patient level.

Urticarial vasculitis is often triggered by medications, infections, and autoimmune disorders. However, vaccinations against viral and bacterial pathogens have recently been documented to induce urticarial vasculitis. We describe the case of a 67-year-old woman who was presented with an extensive erythematous and purpuric rash without systemic symptoms 3 days after an influenza vaccination. She was diagnosed with normocomplementemic urticarial vasculitis based on clinical findings, normal complement levels, and histopathological findings of leukocytoclastic vasculitis. After receiving oral histamines, she showed complete resolution 3 months after receiving the influenza vaccination. Although vaccination-associated vasculitis is common, urticarial vasculitis following vaccinations is rare. We reviewed 13 cases of urticarial vasculitis following a wide range of vaccines, including those against Bacillus Calmette–Guérin, serogroup B meningococcus, influenza, and coronavirus disease. We conducted a comprehensive review of various aspects, including age, sex, past medical history, type of vaccination, number of vaccinations, onset time, cutaneous symptoms, place of eruption, systemic symptoms, laboratory disorders, treatment period, and treatment of urticarial vasculitis. Two patients developed hypocomplementemic urticarial vasculitis after vaccination, and both experienced systemic symptoms such as arthralgia and fever. In this review, no significant differences were found in the data, which may be attributed to the small number of cases. The mechanisms underlying the induction of urticarial vasculitis by vaccines remain unknown; however, in addition to immune complex deposition and complement activation due to vaccine components, molecular mimicry may trigger urticarial vasculitis by producing vaccine-derived pathogenic antigen antibodies. This case study emphasizes the need for heightened awareness and further investigation of urticarial vasculitis as a rare adverse effect of vaccination.Keywords: vaccination, vasculitis, anticardiolipin antibody, molecular mimicry

Abstract Background Tumor-associated neutrophils (TANs) constitute an abundant component among tumor-infiltrating immune cells and have recently emerged as a critical player in pancreatic ductal adenocarcinoma (PDAC) progression. This study aimed to elucidate the pro-tumor mechanisms of TAN and identify a novel target for effective immunotherapy against PDAC. Methods Microarray and cytokine array analyses were performed to identify the mechanisms underlying the function of TANs. Human and mouse TANs were obtained from differentiated HL-60 cells and orthotopically transplanted PDAC tumors, respectively. The interactions of TANs with cancer and cytotoxic T-cells were evaluated through in vitro co-culture and in vivo orthotopic or subcutaneous models. Single-cell transcriptomes from patients with PDAC were analyzed to validate the cellular findings. Results Increased neutrophil infiltration in the tumor microenvironment was associated with poor survival in patients with PDAC. TANs secreted abundant amounts of chemokine ligand 5 (CCL5), subsequently enhancing cancer cell migration and invasion. TANs subpopulations negatively correlated with cytotoxic CD8+ T-cell infiltration in PDAC and promoted T-cell dysfunction. TANs upregulated the membranous expression of Nectin2, which contributed to CD8+ T-cell exhaustion. Blocking Nectin2 improved CD8+ T-cell function and suppressed tumor progression in the mouse model. Single-cell analysis of human PDAC revealed two immunosuppressive TANs phenotypes: Nectin2+ TANs and OLR1+ TANs. Endoplasmic reticulum stress regulated the protumor activities in TANs. Conclusions TANs enhance PDAC progression by secreting CCL5 and upregulating Nectin2. Targeting the immune checkpoint Nectin2 could represent a novel strategy to enhance immunotherapy efficacy in PDAC.

Abstract Trace elements released into the atmosphere by human activities, such as fossil fuel combustion, flow into forest ecosystems via wet and dry deposition and can flow out via mountainous stream water. The Kajikawa catchment site in Niigata Prefecture, located on the Sea of Japan side in central Japan, suffers from large amounts of acidic substance deposition due to its geographical conditions and meteorological seasonality and is affected by transboundary air pollution originating from the Asian continent due to northwesterly seasonal winds in winter. This study determined the concentrations of trace elements dissolved in precipitation and stream water to comprehensively evaluate the effects of transboundary air pollution on forest ecosystems. The results showed that the concentrations of trace elements and major inorganic ions in precipitation tended to be high in winter and low in summer. The fluxes of many trace elements also increased in winter, reflecting high precipitation amount. Except for Sr, the enrichment factors of the highly enriched trace elements did not show clear seasonality. Therefore, they were continuously influenced by anthropogenic activities. Furthermore, the stable Pb isotope ratio changed significantly during the observation period; however, this was not explained by seasonal changes. This study revealed that trace elements are transported to the Sea of Japan side in central Japan, especially in winter; however, their quantity and content change in response to changes in local and transboundary air pollution. Primary extraskeletal osteosarcoma of the lung is exceedingly rare and associated with a poor prognosis. This case report presents a patient with circumferential pulmonary ossification secondary to lung extraskeletal osteosarcoma with compressive mediastinal shift who underwent extrapleural pneumonectomy that led to resolution of symptoms. This case offers an approach to the operative management of primary thoracic osteosarcoma and suggests that even patients with advanced disease may be surgical candidates, particularly for symptom relief.

Calcification of the mitral valve annulus is common in patients on dialysis. The growing number of individuals receiving dialysis has been accompanied by an increase in cases necessitating surgical intervention for mitral valve annulus calcification. In this report, we present a severe case characterized by bulky calcification of the mitral annulus, which was managed with mechanical mitral valve replacement. A 61-year-old man on dialysis presented with chest pain upon exertion that had persisted for 3 months. Cardiac echocardiography revealed severe mitral stenosis and regurgitation, accompanied by cardiac dysfunction. During surgery, an ultrasonic aspiration system was employed to remove the calcification of the mitral valve annulus to the necessary extent. Subsequently, a mechanical mitral valve was sutured into the supra-annular position. To address the regurgitation, the area surrounding the valve was sewn to the wall of the left atrium. Postoperative assessments indicated an absence of perivalvular leak and demonstrated improved cardiac function. The patient was discharged on postoperative day 22. We describe a successful mitral mechanical valve replacement in a case of extensive circumferential mitral annular calcification. Even with severe calcification extending into the left ventricular myocardium, we were able to minimize the decalcification process. This approach enabled the performance of mitral mechanical valve replacement in a high-risk patient on dialysis, thus expanding the possibilities for cardiac surgery.

Abstract The genome is constantly exposed to DNA damage from endogenous and exogenous sources. Fine modulation of DNA repair, chromatin remodeling, and transcription factors is necessary for protecting genome integrity, but the precise mechanisms are still largely unclear. We found that after ionizing radiation (IR), global trimethylation of histone H3 at lysine 4 (H3K4me3) was decreased at an early (5 min) post-IR phase but increased at an intermediate (180 min) post-IR phase in both human and mouse hematopoietic cells. We demonstrated that PTIP, a component of the MLL histone methyltransferase complex, is required for H3K4me3 upregulation in the intermediate post-IR phase and promotes cell cycle arrest by epigenetically inducing a cell cycle inhibitor, PRDM1. In addition, we found that PTIP expression is specifically downregulated in acute myeloid leukemia patients. These findings collectively suggest that the PTIP-PRDM1 axis plays an essential role in proper DNA damage response and its deregulation contributes to leukemogenesis.

Abstract Odours used by insects for foraging and mating are carried by the air. Insects induce airflows around them by flapping their wings, and the distribution of these airflows may strongly influence odour source localisation. The flightless silkworm moth, Bombyx mori, has been a prominent insect model for olfactory research. However, although there have been numerous studies on antenna morphology and its fluid dynamics, neurophysiology, and localisation algorithms, the airflow manipulation of the B. mori by fanning has not been thoroughly investigated. In this study, we performed computational fluid dynamics (CFD) analyses of flapping B. mori to analyse this mechanism in depth. A three-dimensional simulation using reconstructed wing kinematics was used to investigate the effects of B. mori fanning on locomotion and pheromone capture. The fanning of the B. mori was found to generate an aerodynamic force on the scale of its weight through an aerodynamic mechanism similar to that of flying insects. Our simulations further indicate that the B. mori guides particles from its anterior direction within the ~ 60° horizontally by wing fanning. Hence, if it detects pheromones during fanning, the pheromone can be concluded to originate from the direction the head is pointing. The anisotropy in the sampling volume enables the B. mori to orient to the pheromone plume direction. These results provide new insights into insect behaviour and offer design guidelines for robots for odour source localisation.

Abstract Aims Plasma volume status (PVS), a measure of plasma volume, has been evaluated as a prognostic marker for chronic heart failure. Although the prognostic value of PVS has been reported, its significance in patients with acute decompensated heart failure (ADHF) admitted to the cardiovascular intensive care unit (CICU) remains unclear. In this study, we examined the relationship between PVS and long‐term mortality in patients with ADHF admitted to the CICU. Methods Between January 2018 and December 2020, 363 consecutive patients with ADHF were admitted to the Nippon Medical School Hospital CICU. Of the 363 patients, 206 (mean age, 74.9 ± 12.9 years; men, 64.6%) were enrolled in this study. Patients who received red blood cell transfusions, underwent dialysis, were discharged from the CICU or died in the hospital were excluded from the study. We measured the PVS of the patients at admission, transfer to the general ward (GW) and discharge using the Kaplan–Hakim formula. The patients were assigned to four groups according to the quartiles of their PVS measured at each of the three abovementioned timepoints. We examined the association between PVS and all‐cause mortality during the observation period (1134 days). The primary endpoint of this study was all‐cause mortality. Results The Kaplan–Meier analysis showed that the high PVS group had a significantly higher mortality rate at admission, transfer to the GW and discharge than the other groups (log‐rank test: P = 0.016, P = 0.005 and P

The language and cultural priorities in Australian Indigenous education have been priority areas since the inaugural national Indigenous education policy was launched in 1989. For over thirty years, these priorities have sat awkwardly in the largely non-Indigenous teaching profession and classroom teachers continue to struggle with how to embed these priorities into the education process, despite the efforts of Australian Curriculum, Assessment and Reporting Authority and their elaborations for application at curriculum and practice levels. In this article, I suggest that these language and cultural priorities are at cross-purposes with education priorities, and neither have helped to curb the demise of our Indigenous languages nor improved Indigenous students' educational outcomes.

SUMMARY: Active or man-made seismic sources are often used to delineate subsurface geological structures via seismic imaging techniques. Although conventional controlled seismic sources have provided high-resolution subsurface images, the high cost of data acquisition necessitates improved use of cheaper alternative seismic sources for subsurface imaging, such as traffic noise, quarry blasts and earthquakes. Here, we explore the potential use of quarry blasts as an active seismic source for subsurface imaging. The goal of our study is to demonstrate the utility of quarry blasts as seismic sources for investigating subsurface structure with a field experiment in Mill Creek, southern Oklahoma. We deployed a 7455-m-long receiver line of 72 receivers (105 m spacing) extending away from a quarry, with a recording time of 28 d. We analyse similarities between blasts, wave types, origin time and excitation location of quarry blasts, which is important information for using such signal as an active source. Given a relative homogeneous geological condition in the study area, we could not observe clear reflected waves. However, the use of P- and S-wave velocities led to the delineation of a major basement damage zone in the study area, which further demonstrates the viability of quarry blasts as a useful seismic source for subsurface geological imaging.

We investigate the effects of site response on source parameter estimates using earthquakes recorded by the LArge-n Seismic Survey in Oklahoma (LASSO). While it is well known that near-surface unconsolidated sediments can cause an apparent breakdown of earthquake self-similarity, the influence of laterally varying site conditions remains unclear. We analyze site conditions across the 1825-station array on a river plain within an area of 40 km by 23 km using vertical ground motions from 14 regional earthquakes. While the source radiation pattern controls P-wave ground motions below 8 Hz, the surface geology correlates with P-wave ground motions above 8 Hz and S-wave ground motions at 2–21 Hz. Stations installed in alluvial sediments have vertical ground motions that can exceed three times the array median. We use the variation of ground motion of regional earthquakes across the array as a proxy for site effects. The corner frequencies and stress drops of local earthquakes (ML = 0.01–3) estimated using a standard single-spectra approach show negative correlations with the site-effect proxy, while the seismic moments show positive correlations. In contrast, the spectral-ratio approach effectively shows no correlation. The overall bias is small as expected for this relatively homogeneous structure; accurate estimation of site-related biases requires at least 30 stations. Correcting for site-related biases reduces the standard deviations of the source parameters by less than 13% of the total variations. Remaining variations are partially associated with source directivity and model misfits— as small earthquakes can have complex ruptures.

In shallow subduction zones, fluid behavior impacts various geodynamic processes capable of regulating slip behaviors and forming mud volcanoes. However, evidence of structures that control the fluid transfer within an overriding plate is limited and the physical properties at the source faults of slow earthquakes are poorly understood. Here we present high-resolution seismic velocity models and reflection images of the Hyuga-nada area, Japan, where the Kyushu-Palau ridge subducts. We image distinct kilometer-wide columns in the upper plate with reduced velocities that extend vertically from the seafloor down to 10-13 km depth. We interpret the low-velocity columns as damaged zones caused by seamount subduction and suggest that they serve as conduits, facilitating vertical fluid migration from the plate boundary. The lateral variation in upper-plate velocity and seismic reflectivity along the plate boundary correlates with the distribution of slow earthquakes, indicating that the upper-plate drainage system controls the complex pattern of seismic slip at subduction faults.
