Safflower (Carthamus tinctorius L.) is a promising oilseed crop with potential applications in the food, pharmaceutical, and industrial sectors. Understanding the oil content and fatty acid composition of safflower germplasm is crucial for breeding programs aimed at enhancing its agronomic and nutritional traits. This study assessed the oil content and fatty acid composition in 87 safflower accessions. Significant variations were observed, with the oil content ranging from 36.88% to 18.44%. Genotype Egypt 1 exhibited the highest oil content. Among fatty acids, China 1 had the highest myristic acid (0.170%) content, while Remzibey had the lowest (0.100%). Palmitic acid ranged from 6.13% to 8.20%, with Egypt 3 and Bangladesh 3 at the extremes. For palmitoleic acid, Jordan 5 had the highest content (0.53%) and Bangladesh 2/Portugal 2 the lowest (0.03%). Linoleic acid varied from 37.7% (China 7) to 77.73% (Iran 1). A correlation analysis indicated strong positive correlations between protein and oil content, as well as between palmitic and myristic acids, and between palmitic and linoleic acids. Conversely, protein exhibited highly negative correlations with myristic, palmitic, and palmitoleic acids. The protein percentage showed a high heritability but a low genetic advance, while palmitic acid, oil percentage, stearic acid, linoleic acid, palmitoleic acid, and oleic acid showed a high heritability and a moderate genetic advance as a percentage of the mean. These findings can aid in developing cultivars with enhanced fatty acids, oil quality, and nutritional value, facilitating sustainable production for a wide range of industrial applications. Apple (Malus domestica [Suckow] Borkh) holds global economic and cultural value, particularly in temperate regions. This study investigates the genetic variations among 52 apple accessions from three research centre's in Kazakhstan, employing the iPBS-retrotransposons marker system. Of the 35 markers initially screened, 12 highly polymorphic markers were selected for PCR amplification, producing 280 bands, 279 of which were polymorphic, yielding a polymorphism rate of 99.64%. Genetic diversity indices revealed notable variability with the effective number of alleles (ne = 1.655), Shannon's information index (I = 0.373), gene diversity (h = 0.549), and an average genetic distance of 1.2. Analysis of molecular variance (AMOVA) demonstrated that 97% of the genetic variation occurred within the population. STRUCTURE analysis divided the germplasm into two distinct populations and one unclassified population based on collection centers. Both the neighbor-joining tree and principal coordinate analysis (PCoA) supported these results, confirming the genetic separation into two groups. This study highlighted the significant genetic diversity among apple accessions, demonstrating the effectiveness of the iPBS-retrotransposons marker system. Additionally, the highest genetic distance (1.2) observed between the Tyulpan and Red Chief samples, positions these accessions as suitable and promising candidates for future breeding initiatives. In modern plant breeding, molecular markers have become indispensable tools, allowing the precise identification of genetic loci linked to key agronomic traits. These markers provide critical insight into the genetic architecture of crops, accelerating the selection of desirable traits for sustainable agriculture. This review focuses on the advancements in quantitative trait locus (QTL) mapping and genome-wide association studies (GWASs), highlighting their effective roles in identifying complex traits such as stress tolerance, yield, disease resistance, and nutrient efficiency. QTL mapping identifies the significant genetic regions linked to desired traits, while GWASs enhance precision using larger populations. The integration of high-throughput phenotyping has further improved the efficiency and accuracy of QTL research and GWASs, enabling precise trait analysis across diverse conditions. Additionally, next-generation sequencing, clustered regularly interspaced short palindromic repeats (CRISPR) technology, and transcriptomics have transformed these methods, offering profound insights into gene function and regulation. Single-cell RNA sequencing further enhances our understanding of plant responses at the cellular level, especially under environmental stress. Despite this progress, however, challenges persist in optimizing methods, refining training populations, and integrating these tools into breeding programs. Future studies must aim to enhance genetic prediction models, incorporate advanced molecular technologies, and refine functional markers to tackle the challenges of sustainable agriculture. ABSTRACT Micronutrient deficiencies, such as manganese (Mn), pose significant global health risks, affecting millions of people worldwide and leading to serious health conditions. Biofortifying crops, notably common beans, offer a sustainable solution to combat these deficiencies. This study aimed to uncover the genetic basis associated with Mn content in Turkish common bean (Phaseolus vulgaris) germplasm through a genome‐wide association study (GWAS), which is crucial for developing nutrient‐rich bean varieties. Here, we examined variation among 183 common bean accessions, collected from 19 provinces of Turkey and identified the genetic basis linked to seed Mn content. Genotype by environment interaction significantly influenced Mn content (p < 0.05). The mean Mn content was observed 31.69 mg kg−1 across the germplasm. Bingol‐16 had the lowest, while Malatya‐59 had the highest Mn contents. Stability analysis was performed using the ‘STABILITYSOFT’ method and found 10 stable accessions. The cluster constellation plot was generated using JMP statistical software. A total of 7900 DArTseq markers were used for association analysis, identifying 16 markers across four chromosomes (Pv2, Pv5, Pv7 and Pv11). Notably, markers DArT‐3374915 and DArT‐3375187 exhibited consistent associations across different environments, making them promising candidates for Mn‐focused breeding programmes. Gene annotation and interactome analysis, including BLAST searches and protein–protein interaction analysis, revealed associations of candidate genes with Mn concentration regulation, shedding light on potential mechanisms underlying Mn accumulation in common beans. With the development of new technologies in recent years, researchers have made significant progress in crop breeding. Modern breeding differs from traditional breeding because of great changes in technical means and breeding concepts. Whereas traditional breeding initially focused on high yields, modern breeding focuses on breeding orientations based on different crops' audiences or by-products. The process of modern breeding starts from the creation of material populations, which can be constructed by natural mutagenesis, chemical mutagenesis, physical mutagenesis transfer DNA (T-DNA), Tos17 (endogenous retrotransposon), etc. Then, gene function can be mined through QTL mapping, Bulked-segregant analysis (BSA), Genome-wide association studies (GWASs), RNA interference (RNAi), and gene editing. Then, at the transcriptional, post-transcriptional, and translational levels, the functions of genes are described in terms of post-translational aspects. This article mainly discusses the application of the above modern scientific and technological methods of breeding and the advantages and limitations of crop breeding and diversity. Industrial hemp (Cannabis sativa L.) is a highly recalcitrant plant under in vitro conditions that can be overcome by employing external stimuli. Hemp seeds were primed with 2.0–3.0% hydrogen peroxide (H2O2) followed by culture under different Light Emitting Diodes (LEDs) sources. Priming seeds with 2.0% yielded relatively high germination rate, growth, and other biochemical and enzymatic activities. The LED lights exerted a variable impact on Cannabis germination and enzymatic activities. Similarly, variable responses were observed for H2O2 × Blue-LEDs combination. The results were also analyzed by multiple regression analysis, followed by an investigation of the impact of both factors by Pareto chart and normal plots. The results were optimized by contour and surface plots for all parameters. Response surface optimizer optimized 2.0% H2O2 × 918 LUX LEDs for maximum scores of all output parameters. The results were predicted by employing Multilayer Perceptron (MLP), Random Forest (RF), and eXtreme Gradient Boosting (XGBoost) algorithms. Moreover, the validity of these models was assessed by using six different performance metrics. MLP performed better than RF and XGBoost models, considering all six-performance metrics. Despite the differences in scores, the performance indicators for all examined models were quite close to each other. It can easily be concluded that all three models are capable of predicting and validating data for cannabis seeds primed with H2O2 and grown under different LED lights.Key message: Chemical priming of H2O2 with LED lights regulates the cannabis plant growth. Rice is an important diet source for the majority of the world's population, and meeting the growing need for rice requires significant improvements at the production level. Hybrid rice production has been a significant breakthrough in this regard, and the floral traits play a major role in the development of hybrid rice. In grass species, rice has structural units called florets and spikelets and contains different floret organs such as lemma, palea, style length, anther, and stigma exsertion. These floral organs are crucial in enhancing rice production and uplifting rice cultivation at a broader level. Recent advances in breeding techniques also provide knowledge about different floral organs and how they can be improved by using biotechnological techniques for better production of rice. The rice flower holds immense significance and is the primary focal point for researchers working on rice molecular biology. Furthermore, the unique genetics of rice play a significant role in maintaining its floral structure. However, to improve rice varieties further, we need to identify the genomic regions through mapping of QTLs (quantitative trait loci) or by using GWAS (genome-wide association studies) and their validation should be performed by developing user-friendly molecular markers, such as Kompetitive allele-specific PCR (KASP). This review outlines the role of different floral traits and the benefits of using modern biotechnological approaches to improve hybrid rice production. It focuses on how floral traits are interrelated and their possible contribution to hybrid rice production to satisfy future rice demand. We discuss the significance of different floral traits, techniques, and breeding approaches in hybrid rice production. Seed traits are very important agronomic traits that contribute significantly into crop yield and are considered useful indicators to monitor the inheritance of quantitative traits influenced by genotype and environmental conditions. To investıgate genomic regions useful for breeding high-yielding and highly nutritious common bean (Phaseolus vulgaris L.) cultivars, a mini core collection of Turkish common bean germplasm was screened under five environments and two locations for seed traits (seed width, seed yield/plant (SYP), and hundred seed weight). The results of analysis of variance (ANOVA) across the environments revealed significant effects of genotypes and genotype by environment interaction (G × E) for all studied traits except hundred seed weight (HSW), where G × E was found nonsignificant. During this study, mean seed width, SYP, and HSW were 7.038 mm, 21.815 g, and 42.1 g, respectively. Pearson's correlation coefficient revealed a significant and positive correlation (p = 0.01) of seed width with SYP and HSW. At the provinces level, minimum and maximum HSW was reflected by Elazığ and Bolu provinces, respectively. Studied germplasm was grouped into two populations in constellation analysis based on their HSW. A total of 7900 DArTseq markers identified through genotyping by sequencing (GBS) were used for the investigation of marker-trait association. DArT-3373385 marker present on chromosome Pv08 showed a significant association for seed width. DArT-8215545 marker showed a significant association for SYP and this marker was also present on chromosome Pv08. A total of four DArTseq markers were determined to be associated with HSW, and DArT-3372052 contributed maximum (11.32%) to phenotypic variations. Faba bean is protein-enriched legume crop consumed in the form of pods, and cooked seeds and is a part of the traditional food of many Mediterranean countries. It shows variations in its phenotypes, especially in seed size and color. Therefore, it is very important to investigate faba bean genotypes having less cooking time. Keeping this in view, a total of 399 faba bean accessions including three commercial varieties were obtained from International Center for Agricultural Research in the Dry Areas (ICARDA) and Aegean Agricultural Research Institute National Gene Bank for the investigation of cooking traits variations. The field experiments were conducted according to the augmented block design in three provinces (Sivas, Adana, and İzmir) of Türkiye for two consecutive years (2017 and 2018). A good range of variations was observed for all studied traits and the mean cooking time was 114.16 min. İzmir10 had better cooking properties due to its shorter cooking time than other cultivars and landraces. Higher values of the swelling index revealed the higher swelling ability of Balıkesir74. Correlation analysis was performed and swelling capacity showed significant and positive correlations with swelling index and hydration capacity at a significance level of p = 0.05. As this germplasm was collected from 45 provinces, the landrace from Şanlıurfa reflected minimum cooking time and maximum swelling capacity. Principal component analysis (PCA) was performed and the 1st five PCs accounted for a total of 100.00% variations. Biplot analysis resulted in a total of 63.02% variations and the studied germplasm was divided based on cooking time, hydration capacity, and swelling capacity. İzmir10 showed minimum cooking time, it should be also used as a candidate parent to develop faba bean cultivars requiring lesser cooking time to save time and energy. Spring durum wheat occupies over 0.5 M ha in Kazakhstan and represents an important domestic and export commodity. This study aimed to characterize 151 durum wheat cultivars and advanced lines originating from eight breeding programs of the Kazakhstan–Siberia Spring Wheat Improvement Network (KASIB) between 2003 and 2018. The phenotypic characterization was performed in two contracting evaluation sites more than 1000 km apart (Almaty in the Southeast and Shortandy in the North) for two years and a total of 11 agronomic traits were recorded. Field trials at both locations followed regional agronomy practices, including sowing, harvesting, and genotype evaluation using a randomized complete block design (RCBD). The growing season was longer in Almaty, resulting in a higher number of grains per spike. Though grains are smaller in size with an overall higher yield, 243 g/m2 versus 170 g/m2, there was no correlation between germplasm performance at the two sites. Molecular characterization was performed with 10 iPBS-retrotransposons primers that resulted in a total of 345 bands and showed a mean polymorphism of 91.9%. Mean values of gene diversity (0.251), Shannon's information index (0.388), and expected heterozygosity (0.233) revealed a relatively high level of genetic diversity in the KASIB set. AMOVA revealed higher genetic variations due to differences within the populations. Marker-based cluster analysis, including STRUCTURE and neighbor-joining algorithms, divided the material into two populations with clear differences in geographic origin. Sorghum is emerging as a model crop for functional genetics and genomics of tropical grasses with abundant uses, including food, feed, and fuel, among others. It is currently the fifth most significant primary cereal crop. Crops are subjected to various biotic and abiotic stresses, which negatively impact on agricultural production. Developing high-yielding, disease-resistant, and climate-resilient cultivars can be achieved through marker-assisted breeding. Such selection has considerably reduced the time to market new crop varieties adapted to challenging conditions. In the recent years, extensive knowledge was gained about genetic markers. We are providing an overview of current advances in sorghum breeding initiatives, with a special focus on early breeders who may not be familiar with DNA markers. Advancements in molecular plant breeding, genetics, genomics selection, and genome editing have contributed to a thorough understanding of DNA markers, provided various proofs of the genetic variety accessible in crop plants, and have substantially enhanced plant breeding technologies. The world is facing rapid climate change and a fast-growing global population. It is believed that the world population will be 9.7 billion in 2050. However, recent agriculture production is not enough to feed the current population of 7.9 billion people, which is causing a huge hunger problem. Therefore, feeding the 9.7 billion population in 2050 will be a huge target. Climate change is becoming a huge threat to global agricultural production, and it is expected to become the worst threat to it in the upcoming years. Keeping this in view, it is very important to breed climate-resilient plants. Legumes are considered an important pillar of the agriculture production system and a great source of high-quality protein, minerals, and vitamins. During the last two decades, advancements in OMICs technology revolutionized plant breeding and emerged as a crop-saving tool in wake of the climate change. Various OMICs approaches like Next-Generation sequencing (NGS), Transcriptomics, Proteomics, and Metabolomics have been used in legumes under abiotic stresses. The scientific community successfully utilized these platforms and investigated the Quantitative Trait Loci (QTL), linked markers through genome-wide association studies, and developed KASP markers that can be helpful for the marker-assisted breeding of legumes. Gene-editing techniques have been successfully proven for soybean, cowpea, chickpea, and model legumes such as Medicago truncatula and Lotus japonicus. A number of efforts have been made to perform gene editing in legumes. Moreover, the scientific community did a great job of identifying various genes involved in the metabolic pathways and utilizing the resulted information in the development of climate-resilient legume cultivars at a rapid pace. Keeping in view, this review highlights the contribution of OMICs approaches to abiotic stresses in legumes. Cysteine-rich poly comb-like protein (CPP) is a member of cysteine-rich transcription factors that regulates plant growth and development. In the present work, we characterized twelve CPP transcription factors encoding genes in soybean (Glycine max). Phylogenetic analyses classified CPP genes into six clades. Sequence logos analyses between G. max and G. soja amino acid residues exhibited high conservation. The presence of growth and stress-related cis-acting elements in the upstream regions of GmCPPs highlight their role in plant development and tolerance against abiotic stress. Ka/Ks levels showed that GmCPPs experienced limited selection pressure with limited functional divergence arising from segmental or whole genome duplication events. By using the PAN-genome of soybean, a single nucleotide polymorphism was identified in GmCPP-6. To perform high throughput genotyping, a kompetitive allele-specific PCR (KASP) marker was developed. Association analyses indicated that GmCPP-6-T allele of GmCPP-6 (in exon region) was associated with higher thousand seed weight under both water regimes (well-water and water-limited). Common bean is considered a recalcitrant crop for in vitro regeneration and needs a repeatable and efficient in vitro regeneration protocol for its improvement through biotechnological approaches. In this study, the establishment of efficient and reproducible in vitro regeneration followed by predicting and optimizing through machine learning (ML) models, such as artificial neural network algorithms, was performed. Mature embryos of common bean were pretreated with 5, 10, and 20 mg/L benzylaminopurine (BAP) for 20 days followed by isolation of plumular apice for in vitro regeneration and cultured on a post-treatment medium containing 0.25, 0.50, 1.0, and 1.50 mg/L BAP for 8 weeks. Plumular apice explants pretreated with 20 mg/L BAP exerted a negative impact and resulted in minimum shoot regeneration frequency and shoot count, but produced longer shoots. All output variables (shoot regeneration frequency, shoot counts, and shoot length) increased significantly with the enhancement of BAP concentration in the post-treatment medium. Interaction of the pretreatment × posttreatment medium revealed the need for a specific combination for inducing a high shoot regeneration frequency. Higher shoot count and shoot length were achieved from the interaction of 5 mg/L BAP × 1.00 mg/L BAP followed by 10 mg/L BAP × 1.50 mg/L BAP and 20 mg/L BAP × 1.50 mg/L BAP. The evaluation of data through ML models revealed that R2 values ranged from 0.32 to 0.58 (regeneration), 0.01 to 0.22 (shoot counts), and 0.18 to 0.48 (shoot length). On the other hand, the mean squared error values ranged from 0.0596 to 0.0965 for shoot regeneration, 0.0327 to 0.0412 for shoot count, and 0.0258 to 0.0404 for shoot length from all ML models. Among the utilized models, the multilayer perceptron model provided a better prediction and optimization for all output variables, compared to other models. The achieved results can be employed for the prediction and optimization of plant tissue culture protocols used for biotechnological approaches in a breeding program of common beans. Cadmium (Cd) is a non-essential heavy metal having toxic effects on all living organisms. Durum wheat (Triticum durum Desf.) is widely used in human diets but has the potential to accumulate Cd. It also has a high level of genetic diversity, which may be exploited to develop cultivars with low Cd content. We aimed to perform marker-assisted selection and validate previously identified Cd markers in durum wheat germplasm for use in the investigation of accessions that accumulate low grain Cd content. We assessed 130 durum wheat accessions phenotypically and using three different molecular markers. Grain Cd contents of the studied germplasm varied 4.91-fold (26.2–128.7 μg/kg) with an average of 58.2 μg/kg. Landraces showed lower average values of grain Cd content than cultivars. Three molecular markers (usw47, Cad-5B and KASP marker Cad-5B) were used to differentiate high and low Cd accumulating lines. Results showed high correlation and successfully classified the accessions to the expected high or low Cd level; 87 accessions showed the low Cd alleles, and 43 accessions the high Cd alleles, except for five accessions with the usw47 marker that showed heterozygous status. A significant correlation coefficient (r = 0.944*) was observed among the three molecular markers. Based on molecular markers, 96.2% of the accessions were classified accurately. The KASP assay was highly effective in successfully separating low from high Cd content accessions and could be used as a molecular tool in durum wheat breeding programs, with less cost and time, targeting reduced grain Cd levels. The results of this study will allow durum wheat breeders to accelerate their progress to select suitable genotypes with the desired alleles. Drought, salinity, extreme temperatures, and ultraviolet (UV) radiation are major environmental factors that adversely affect plant growth and crop production. As a protective shield covering the outer epidermal cell wall of plant aerial organs, the cuticle is mainly composed of cutin matrix impregnated and sealed with cuticular waxes, and greatly contributes to the plant adaption to environmental stresses. Past decades have seen considerable progress in uncovering the molecular mechanism of plant cutin and cuticular wax biosynthesis, as well as their important roles in plant stress adaptation, which provides a new direction to drive strategies for stressresilient crop breeding. In this review, we highlighted the recent advances in cuticle biosynthesis in plant adaptation to drought, salinity, extreme temperatures, and UV radiation stress, and discussed the current status and future directions in harnessing cuticle biosynthesis for crop improvement. Magnesium (Mg) is the fourth most abundant element in the human body and plays the role of cofactor for more than 300 enzymatic reactions. In plants, Mg is involved in various key physiological and biochemical processes like growth, development, photophosphorylation, chlorophyll formation, protein synthesis, and resistance to biotic and abiotic stresses. Keeping in view the importance of this element, the present investigation aimed to explore the Mg contents diversity in the seeds of Turkish common bean germplasm and to identify the genomic regions associated with this element. A total of 183 common bean accessions collected from 19 provinces of Turkey were used as plant material. Field experiments were conducted according to an augmented block design during 2018 in two provinces of Turkey, and six commercial cultivars were used as a control group. Analysis of variance depicted that Mg concentration among common bean accessions was statistically significant (p < 0.05) within each environment, however genotype x environment interaction was nonsignificant. A moderate level (0.60) of heritability was found in this study. Overall mean Mg contents for both environments varied from 0.33 for Nigde-Dermasyon to 1.52 mg kg-1 for Nigde-Derinkuyu landraces, while gross mean Mg contents were 0.92 mg kg-1. At the province level, landraces from Bolu were rich while the landraces from Bitlis were poor in seed Mg contents respectively. The cluster constellation plot divided the studied germplasm into two populations on the basis of their Mg contents. Marker-trait association was performed using a mixed linear model (Q + K) with a total of 7,900 DArTseq markers. A total of six markers present on various chromosomes (two at Pv01, and one marker at each chromosome i.e., Pv03, Pv07, Pv08, Pv11) showed statistically significant association for seed Mg contents. Among these identified markers, the DArT-3367607 marker present on chromosome Pv03 contributed to maximum phenotypic variation (7.5%). Additionally, this marker was found within a narrow region of previously reported markers. Durum wheat (Triticum turgidum L.) is the 10th most cultivated crop and is a vital food source for human consumption. Genetic diversity in durum wheat is still an unexplored subject, and gene banks are filled with accessions waiting to be screened. Here, four different groups of durum wheat, Turkish and foreign cultivars, and in situ and ex situ landraces were evaluated for population structure and genetic diversity. A collection of 129 durum wheat accessions were genotyped, with a total of 6,357 markers (SilicoDArT, SNP [single‐nucleotide polymorphism], and simple sequence repeats [SSRs]). The highest mean allele number per marker (1.562) was obtained with SNP markers in ex situ landraces; the lowest mean allele number per marker (0.788) was obtained with SSR markers in the foreign cultivars. The minimum percentage of polymorphic loci was 38.70% observed with SSR markers in foreign cultivars; the maximum was 76.20% in SilicoDArT markers in ex situ landraces. According to Neighbor‐Joining analyses, the studied collection was divided into groups of cultivars and landraces, with some level of admixtures. Structure and principal coordinate analysis obtained distinct clusters for ex situ landraces and foreign cultivars, supported by Neighbor‐Joining analysis. A significant level of intergroup and intragroup variation was observed, and Turkish ex situ landraces were found as the most diverse and distinct group in the collection. Turkish landraces may be a promising source of novel allelic diversity that can be used in durum wheat breeding worldwide. Simple Summary: Invertase genes are among the important genes responsible for carbon metabolism in plants, significantly contributing to plant development and stress responses. In this study, for the first time, we performed genome-wide analysis for Invertase gene family in potato, identified and conducted expression profiling in different tissues by RNA seq analysis and validated it by Q-PCR. We also performed invertase family genes expression profiling under drought, salt and heat stress to elucidate their involvement in stress responses. Findings of this study will be helpful for future functional and genetic studies not only in potato but also in other plants. The potato is one of the most important and valuable crops in terms of consumption worldwide. However, abiotic stressors are the critical delimiters for the growth and productivity of potato. Invertase genes play key roles in carbon metabolism, plant development, and responses to stress stimuli. Therefore, a comprehensive genome-wide identification, characterization and expression analysis of invertase genes was performed in the potato. The current study identified 19 invertase genes, randomly distributed throughout the potato genome. To further elucidate their evolutionary, functional and structural relationship within family and with other plant species, we performed sequence and phylogenetic analysis, which segregated invertase genes into two main groups based on their sequence homology. A total of 11 genes are included in acidic invertases and 8 genes are in neutral or alkaline invertases, elucidating their functional divergence. Tissue specific expression analyses (RNA sequencing and qRT-PCR) of different plant tissues showed differential expression pattern. Invertase genes have higher expression in flower, leaf, root and shoot tissues, while under abiotic stress conditions, the expression of the invertase gene is significantly upregulated. Results of this study revealed that vacuolar and cell wall destined invertases are mainly the functional member genes of the invertase family.
