Mitochondrial DNA (mtDNA) has been used for decades as a predominant tool in population genetics and as a valuable addition to forensic genetic research, owing to its unique maternal inheritance pattern that enables the tracing of individuals along the maternal lineage across numerous generations. The dynamic interplay between evolutionary forces, primarily genetic drift, bottlenecks, and the founder effect, can exert significant influence on genetic profiles. Consequently, the Adriatic islands have accumulated a subset of lineages that exhibits remarkable absence or rarity within other European populations. This distinctive genetic composition underscores the islands' potential as a significant resource in phylogenetic research, with implications reaching beyond regional boundaries to contribute to a global understanding. In the initial attempt to expand the mitochondrial forensic database of the Croatian population with haplotypes from small isolated communities, we sequenced mitogenomes of rare haplogroups from different Croatian island and mainland populations using next-generation sequencing (NGS). In the next step and based on the obtained results, we refined the global phylogeny of haplogroup N1a, HV2, and X by analyzing rare haplotypes, which are absent from the current phylogenetic tree. The trees were based on 16 novel and 52 previously published samples, revealing completely novel branches in the X and HV2 haplogroups and a new European cluster in the ancestral N1a variant, previously believed to be an exclusively African–Asian haplogroup. Acromegaly is a rare disfiguring disease caused by hypersecretion of the growth hormone from the pituitary adenomas. Body proportions, including bones of the hand, have been widely used to estimate anthropometric measures, such as stature. The main aim of this study was to compare stature, hand length, and stature/hand length ratio in healthy and patients with acromegaly and evaluate the accuracy of using hand length for the estimation of stature in patients with acromegaly. A total of 44 patients with acromegaly were analyzed in comparison to the control group of 8944 phenotypically healthy individuals. Measurement data were analyzed using parametric and nonparametric tests, linear categorical regression (in healthy populations, and ordinal regression (in patients with acromegaly). The results show that the criterion stature could be predicted with hand length in both male and female individuals with acromegaly, while the amount of variance explained is something higher in males. Both in males and females, the stature/hand length ratio is higher in healthy individuals compared to acromegalic ones. This information might be of use to endocrinologists, rheumatologists, forensic scientist as well as in research where anthropometric characterization is performed. 5. Linking infant size and early growth with maternal lifestyle and breastfeeding – the first year of life in the CRIBS cohort.

Every third child in Croatia is classed as overweight or obese. Infant growth can represent early warning signs for obesity. To detect early risk factors for obesity by investigating infant size and early growth trajectories and their association with maternal lifestyle and breastfeeding. Ninety-eight mother–child pairs from the Croatian Islands' Birth Cohort Study (CRIBS) cohort were included in the study. Data were collected from questionnaires and medical records. Growth data were converted to Z-scores using World Health Organisation (WHO) standards and used as the primary outcome. Z-score trajectories in the first year of life were in line with WHO standards. A direct link between infant size and maternal socioeconomic status (SES) or breastfeeding was not detected. However, child weight gain in the first 6 months was associated with maternal body mass index (BMI) before pregnancy (p < 0.01). A positive association was also established between breastfeeding and maternal SES and mothers that report an unhealthy diet have heavier children (p < 0.05, respectively). Infant size and early growth in Croatia is in line with WHO standards and risk factors for obesity development were detectable in the first year of life, but not highly pronounced. However, more effective BMI monitoring and promotion of a healthy diet and lifestyle of women before and during pregnancy is needed. [ABSTRACT FROM AUTHOR]

Simple Summary: Human activities such as heavy industry and transport have significantly increased the levels of many poisonous chemicals in the human environment. Among these chemicals are heavy metals, which pose a great risk to human health because they cannot decompose and cannot be eliminated from the human body by metabolic processes. Volcanic stone called clinoptilolite is inert and extremely porous, and can capture heavy metals into its meshed structure. Therefore, it can be used as a food supplement for detox purposes. Several clinical studies have already indicated its detoxifying, antioxidant and anti-inflammatory effects. In our experiment, we fed healthy rats with clinoptilolite, which was milled to fine dust to improve performance, for three months. Two forms were used: tribomechanically activated zeolite and Panaceo-Micro-Activated zeolite. Upon completion of the administration period, we observed the impact of clinoptilolite dust on the metallic composition in different rat tissues and bloodstream. Our results showed that this activated natural stone had indeed released metallic toxicants from the rat organs to the bloodstream, which indicates a detoxification process. Heavy metals are dangerous systemic toxicants that can induce multiple organ damage, primarily by inducing oxidative stress and mitochondrial damage. Clinoptilolite is a highly porous natural mineral with a magnificent capacity to eliminate metals from living organisms, mainly by ion-exchange and adsorption, thus providing detoxifying, antioxidant and anti-inflammatory medicinal effects. The in vivo efficiency and safety of the oral administration of clinoptilolite in its activated forms, tribomechanically activated zeolite (TMAZ) and Panaceo-Micro-Activated (PMA) zeolite, as well as the impact on the metallic biodistribution, was examined in healthy female rats. Concentration profiles of Al, As, Cd, Co, Pb, Ni and Sr were measured in rat blood, serum, femur, liver, kidney, small and large intestine, and brain using inductively coupled plasma mass spectrometry (ICP-MS) after a 12-week administration period. Our results point to a beneficial effect of clinoptilolite materials on the concentration profile of metals in female rats supplemented with the corresponding natural clinoptilolite materials, TMAZ and PMA zeolite. The observed decrease of measured toxicants in the kidney, femur, and small and large intestine after three months of oral intake occurred concomitantly with their most likely transient release into the bloodstream (serum) indicative of a detoxification process. 9. Food Choice Motives and COVID-19 in Belgium.

To investigate the main motives driving dietary intake changes potentially introduced by preventive measures to address the pandemic, an online survey, using a 36-item Food Choice Questionnaire applied for the period before (the year 2019) and during (2020–2021) the pandemic, was distributed between July and October 2021 among adult residents from Belgium. A total of 427 eligible respondents, the majority Dutch-speaking, were included for analyses. The importance of nine motives for food choices, including health, mood, convenience, sensory appeal, natural content, price, weight control, familiarity and ethical concerns, was assessed by scoring from 1 to 5, and comparing mean scores from the during period with the before period. Sensory appeal was the most important food choice motive before (mean score of 4.02 ± 0.51) and during (3.98 ± 0.48) the pandemic. Convenience and health also ranked among the main motives, with health observed to become more important during the pandemic (3.69 ± 0.60 during vs. 3.64 ± 0.59 before). Additionally, mean scores of mood (3.41 ± 0.71 vs. 3.32 ± 0.58), natural content (3.35 ± 0.84 vs. 3.26 ± 0.85) and weight control (3.33 ± 0.79 vs. 3.25 ± 0.76) were significantly higher during as compared to before. The extent of change in the level of importance for natural content was smaller with increasing age, and for health larger for urban areas, but for other motives there were no significant differences across population subgroups. Changes in the level of importance were observed in both directions, while a moderate share of respondents declared no change, suggesting some persistence of food choice motives. Further activities within public health monitoring should be considered to fully understand the COVID-19 implications on food choice motives together with people's food behaviors and consumption. High pre-pregnancy body mass index (BMI) and excessive gestational weight gain (GWG) are significant risk factors for maternal and neonatal health. To assess pre-pregnancy BMI and GWG during pregnancy and their association with different maternal and neonatal characteristics in the transitional Mediterranean population from the Eastern Adriatic islands. Two hundred and sixty-two mother–child dyads from the CRoatian Islands' Birth Cohort Study (CRIBS) were included in the study. Chi-square test, ANOVA, and regression analysis were used to test the association between selected characteristics. In total, 22% of women entered pregnancy as overweight/obese and 46.6% had excessive GWG. Pre-pregnancy overweight and obesity were significantly associated with elevated triglycerides uric acid levels, and decreased HDL cholesterol in pregnancy. Excessive GWG was associated with elevated fibrinogen and lipoprotein A levels. Women with high pre-pregnancy BMI and GWG values were more likely to give birth to babies that were large for gestational age (LGA), additionally confirmed in the multiple logistic regression model. High maternal pre-pregnancy BMI and excessive GWG were both significantly associated with deviated biochemical parameters and neonatal size. More careful monitoring of maternal nutritional status can lead to better pre- and perinatal maternal healthcare. [ABSTRACT FROM AUTHOR]

The aims were to investigate determinants of the quality of life (QoL) of pregnant women. Total of 302 healthy women 18 to 28 weeks of gestation participated in prospective study. WHOQOL-bref, Multidimensional Health Locus of Control scales, Edinburgh Postnatal Depression Scale, and the perceived stress appraisals were administered. Various patterns of predictors for four domains of QoL were identified, for psychological (42% variance explained), social relationship (29%), environmental (29%) and physical health (25%). Depression and hope, together with the extent to which one's health is influenced by powerful other or chance should be targeted in health promotion strategies during pregnancy. Significant mtDNA variation in Southeastern Europe (SEE) reflects the turbulent and complex demographic history of the region, influenced by gene flow from various parts of Eurasia and a long history of intermixing. In this study we present the maternal genetic profile of the Serbian and Montenegrin populations based on the high resolution analysis of 258 mtDNAs, 119 samples from Serbia and 139 samples from Montenegro. Besides the evidence of minor gene flow from distant central/northeastern Asia, the majority of haplogroups place these populations in the broader genetic landscape of Southeastern Europe, close to their neighbors. Objectives The research objective of this study is to enlarge and deepen the Y chromosome research on the Croatian population and enable additional insights into the population diversity and historic events that shaped the current genetic landscape of Croatia and Southeastern Europe (SEE). Materials and Methods A high-resolution phylogenetic and phylogeographic analysis of 66 biallelic (SNPs) and 17 microsatellite (STRs) markers of the Y chromosome was performed using 720 Croatian samples. The obtained results were placed in a wider European context by comparison with ∼4450 samples from a number of other European populations. Results A high diversity of haplogroups was observed in the overall Croatian sample, and all typical European Y chromosome haplogroups with corresponding clinal patterns were observed. Three distinct genetic signals were identifiable in the Croatian paternal gene pool - I2a1b-M423, R1a1a1b1a*-M558, and E1b1b1a1b1a-V13 haplogroups. Discussion The analyses of the dominant and autochthonous I2a1b-M423 lineage (>30%) suggest that SEE had a significant role in the Upper Paleolithic, the R1a1a1b1a*-M558 lineage (19%) represents a signal from present day Slavic populations of Central Europe in the Croatian population, and the phylogeography of the E1b1b1a1b1a-V13 clade (around 9%) implies cultural diffusion of agriculture into Europe via the Balkan Peninsula. Background: Many Croatian islands are examples of genetic isolates, with low level of heterozygosity and high level of inbreeding, due to practice of endogamy. Aim: The aim was to study the genetic structure of two insular and one mainland population through high-resolution phylogenetic analysis of mitochondrial DNA (mtDNA). Subjects and methods: MtDNA polymorphisms were explored in 300 unrelated individuals from Mljet, Lastovo and the coastal city of Dubrovnik, based on SNP polymorphisms. Results: All mtDNA haplogroups found in the sample were of typical European origin. However, the frequency distribution of their subclades differed significantly from other Croatian and European populations. MtDNA haplotype analysis revealed only two possible founder lineages on Mljet and six on Lastovo, accounting for almost half of the sample on both islands. The island of Mljet also has the lowest reported haplotype and nucleotide diversity among Croatian isolates and the island of Lastovo, a new sublineage of a usually quite rare U1b clade. Conclusion: The results can be explained by the effect evolutionary forces have on genetic structure, which is in line with the specific demographic histories of the islands. An additional research value of these two island isolates is the appearance of certain Mendelian disorders, highlighting their importance in epidemiological studies. [ABSTRACT FROM AUTHOR]

34. A Generation Shift in Mediterranean Diet Adherence and Its Association with Biological Markers and Health in Dalmatia, Croatia.

Previous studies have confirmed the beneficial effect of a Mediterranean diet in mitigating health issues related to cardiovascular disease, diabetes and obesity. However, rapid changes in the traditional way of life and the "westernization" of the diet in Mediterranean populations, especially in younger generations, has led to progressive abandonment of healthy dietary patterns. In order to investigate the generation shift in dietary patterns and lifestyle habits in the Mediterranean part of Croatia, we compared two cohorts of 610 women (266 pregnant and 344 non-pregnant) from the same region, but from different age groups. The MDSS score was derived from food frequency questionnaires. The results showed that the young, reproductively active generation (pregnant women) in Dalmatia, Croatia, although having a higher education and socioeconomic status, exhibits a more adverse eating behaviour (lower adherence to the Mediterranean diet) and lifestyle (excessive smoking in pregnancy) than the older population from the same region. Lower MDSS scores across aggregated age groups in both cohorts showed significant association with higher blood lipid levels and higher smoking frequency. In conclusion, Mediterranean diet adherence is associated with biological markers (age, lipid profile) and lifestyle (smoking) in our study, with a more adverse trend observed in the younger generation. 35. Evaluation of the Food Choice Motives before and during the COVID-19 Pandemic: A Cross-Sectional Study of 1232 Adults from Croatia.

The coronavirus disease 2019 pandemic brought changes to almost every segment of our lives, including dietary habits. We present one among several studies, and the first on the Croatian population, aiming at investigating changes of food choice motives before and during the pandemic. The study was performed in June 2021 as an online-based survey, using a 36-item Food Choice Questionnaire applied for both the periods before and during the pandemic. The final sample consisted of 1232 adults living in Croatia. Sensory appeal was ranked as the number one most important food choice motive before, whereas health was ranked as the number one most important food choice motive during the pandemic. Ethical concern was reported as the least important food choice motive both before and during the pandemic. In women, natural content (p = 0.002), health, convenience, price, weight control, familiarity, and ethical concern (all p < 0.001) became more important during the pandemic, while price (p = 0.009), weight control, familiarity, and ethical concern (all p < 0.001) became more relevant for men. All together, these can be considered favorable changes toward optimal diets and may result in beneficial influences on health and lifestyle. Education strategies and efficiently tackling misinformation are prerequisites for informed food choice, which will ensure long-lasting positive effects of such changes. [ABSTRACT FROM AUTHOR]

Maternal nutrition and lifestyle in pregnancy are important modifiable factors for both maternal and offspring's health. Although the Mediterranean diet has beneficial effects on health, recent studies have shown low adherence in Europe. This study aimed to assess the Mediterranean diet adherence in 266 pregnant women from Dalmatia, Croatia and to investigate their lifestyle habits and regional differences. Adherence to the Mediterranean diet was assessed through two Mediterranean diet scores. Differences in maternal characteristics (diet, education, income, parity, smoking, pre-pregnancy body mass index (BMI), physical activity, contraception) with regards to location and dietary habits were analyzed using the non-parametric Mann–Whitney U test. The machine learning approach was used to reveal other potential non-linear relationships. The results showed that adherence to the Mediterranean diet was low to moderate among the pregnant women in this study, with no significant mainland–island differences. The highest adherence was observed among wealthier women with generally healthier lifestyle choices. The most significant mainland–island differences were observed for lifestyle and socioeconomic factors (income, education, physical activity). The machine learning approach confirmed the findings of the conventional statistical method. We can conclude that adverse socioeconomic and lifestyle conditions were more pronounced in the island population, which, together with the observed non-Mediterranean dietary pattern, calls for more effective intervention strategies. 39. Association of pre-pregnancy BMI with biochemical profile during pregnancy, delivery mode and size of neonates in the CRIBS birth cohort.

In the last few decades the prevalence of overweight and obesity has reached epidemic levels worldwide affecting over 2.1 billion people, including a large proportion of women of childbearing age. An adverse intrauterine environment resulting from conditions such as maternal obesity or gestational diabetes, can have long-term negative effect on the child's health later in life. This study focuses on maternal pre-pregnancy BMIs and their association with biochemical profile in pregnancy, delivery mode and size of neonates in the Croatian Islands' Birth cohort Study (CRIBS). A total of 301 mother-child dyads were included in this study. The classification of the pre-pregnancy nutritional status followed WHO BMI cut-offs. Blood samples of women were taken in the second trimester of pregnancy and analysed for fasting blood glucose level, urates, creatinine, uric acid, total protein, bilirubin, triglycerides, total cholesterol, HDL and LDL cholesterol, potassium, sodium, aspartate transaminase (AST), alanine aminotransferase (ALT), gamma-glutamyl transferase (GGT), homocysteine, fibrinogen and lipoprotein A. The neonates' length in cm and weight in gm were measured immediately after delivery at the University Hospital Split. Differences in mean values of biochemical parameters and neonates' size between pre-pregnancy BMI categories were tested using One-way ANOVA (p < 0.05) and differences in prevalence of delivery mode using the Chi-square test. Nutritional status before pregnancy showed that 4% of women were underweight (BMI < 18.5 kg.m
2 ), 75.1 % had normal weight (BMI 18.5–24.9 kg.m2 ), 14.6% were overweight (BMI 25–29.9 kg.m2 ) and 6.3% were obese (BMI ≥ 30.0 kg.m2 ). Pre-pregnancy obesity and overweight were significantly associated with elevated fasting glucose (p = 0.02), triglycerides (p = 0.01), and uric acid (p = 0.001) levels, and with decreased protective HDL cholesterol (p = 0.01) during pregnancy. Also, in comparison with normal weight and underweight women, obese pregnant women more often gave birth to heavier (p = 0.01) and longer babies (p = 0.036) and there was a strong correlation between higher pre-pregnancy BMI and delivery mode (p = 0.004). Obese women had 50% chance of giving birth by caesarean section in comparison with 20.1% in normal weight women. Our study confirmed that high maternal pre-pregnancy BMI could be a significant risk factor for both mother's health during pregnancy and different adverse birth outcomes, such as elevated risk of caesarean section delivery and child macrosomia. Aim To analyze an additional set of ˝Y-chromosome genetic markers to acquire a more detailed insight into the diversity of the Croatian population. Methods A total of 518 Yfiler Plus profiles were genotyped. Allele frequencies, haplotype frequencies, and haplotype diversity were calculated by using the STRAF software v. 2.0.4. Genetic distances were quantified by Rst with AMOVA online tool from the YHRD. The evolutionary history was inferred with the neighbor-joining method of phylogenetic tree construction in the MEGAX software. Whit Athey’s Haplogroup Predictor v. 5 was used for additional comparison with regional and other European populations. Results A total of 507 haplotypes were used for genetic STR analysis. An interpopulation study on 17 Y-STR markers showed the lowest genetic diversity between the Croatian and Bosnian-Herzegovinian populations and the highest between the Croatian and Irish populations. Additional interpopulation comparison with the original 27 Y-STR markers (for the population with available data) was also performed. A total of 518 haplotypes were used in the determination of haplogroup diversity. Haplogroup I with its sublineage I2a expressed the highest prevalence. The second most prevalent haplogroup was R, with its major sublineage R1a, except for the subpopulation of Hvar, where E1b1b was the second most prevalent haplogroup. Rare haplogroups also confirmed in this study were L, T, and Q. G1 was detected for the first time in the Croatian population. Conclusion We obtained a new insight into the differences between examined subpopulations of Croatia and their possible (dis)similarities with neighboring and distant populations. This paper presents the socio-demographic profile of participants in the Croatian Islands' Birth Cohort Study (CRIBS), the first longitudinal study in Croatia to follow pregnant women and their children up to the age of two. The sample consisted of 286 pregnant women (145 from the islands of Brač and Hvar, and 141 from the mainland). The study aimed to: 1) summarise the main characteristics of the CRIBS study sample; 2) compare the sample with the general population of Croatia; 3) analyse the differences between the CRIBS participants according to place of residence (islands vs. mainland) and family type (nuclear vs. extended family). In comparison with women from the mainland, islander women had more often given birth to more than one child and finished only secondary school, while most of the women from the mainland had university degree. The prevalence of different family types also significantly differed between the islands and mainland, with islanders living in extended families and having lower average household income. Aim To determine the sexual dimorphism of the sternum with standard measurements in a contemporary Croatian population sample using multi-slice computed tomography (MSCT) and to compare the data obtained by an automatic with those obtained by a manual approach. Methods Five sternal measurements were obtained from MSCT images of 73 men and 55 women and three sternal indices were calculated. Custom image analysis software was developed for automatic segmentation and calculation of sternal measurements. Measurements of sexual dimorphism were automatically calculated and compared with manual measurements. Results All of the sternal measurements exhibited signifi- cant differences between men and women. The discrepancies between manual and automatic measurements ranged from 2.8% to 3.6% of the mean average values obtained with the automatic approach. The most accurate single-variable discriminant function was sternal body length (82.8%), the most accurate index was sternal area (89.1%), and the discriminant function using three variables was manubrium width, sternal body length, and sternal body width (90.6%). Conclusion Sternal measurements are a reliable sex indicator and can be used in forensic casework. Computer-aided measurement methods can accelerate sex estimation and improve its precision and accuracy. [ABSTRACT FROM AUTHOR]

Aim To perform a comprehensive evaluation of association of common genetic variants in candidate genes in the dopaminergic pathway with schizophrenia in a sample from Croatian population. Methods A case-control association study was performed on 104 unrelated patients with schizophrenia recruited from a psychiatric hospital in Zagreb and 131 phenotypically normal Croatian subjects. Forty-nine tagging single nucleotide polymorphisms (tagSNPs) in 8 candidate genes in the dopaminergic pathway were identified from the HapMap database and tested for association. Genotyping was performed using the SNPlex platform. Statistical analysis was conducted to assess allelic and genotypic associations between cases and controls using a goodness of fit ?2 test and trend test, respectively; adjustment for multiple testing was done by permutation based analysis. Results Significant allele frequency differences between schizophrenia cases and controls were observed at 4 tag- SNPs located in the genes DRD5, HTR1B1, DBH, and TH1 (P < 0.005). A trend test also confirmed the genotypic association (P < 0.001) of these 4 tagSNPs. Additionally, moderate association (P < 0.05) was observed with 8 tagSNPs on SLC6A3, DBH, DRD4, SLC6A4, and COMT. Conclusions Common genetic variants in genes involved in the dopaminergic pathway are associated with schizophrenia in the populations of Caucasian descent. [ABSTRACT FROM AUTHOR]

- Abstract
Food frequency questionnaires (FFQ) are designed to assess habitual diet by asking about the frequency with which food items or specific food groups are consumed over a reference period. In this study, we have developed a FFQ to assess habitual dietary intake of healthy adults to estimate the quality of their nutrition and conducted a study to validate the developed FFQ against a three-day 24-hour recall. Second aim of the study is to establish the correlation between the FFQ to assess habitual dietary intake and a three-day 24-hour recall, in relation with participants' age and education level. FFQ used in our study was adapted from the Harvard Semiquantitative Food Frequency Questionnaire, consisted of 101 food items, divided into 9 groups. The 24-hour recalls were collected during one week (the same week when the FFQ was completed) and on each different day of the week (two weekdays and one weekend day). Both the FFQ and three day 24-hour recall were distributed to 100 healthy, employed subjects, 32 males (aged 39.73 +/- 14.02) and 36 females (aged 34.20 +/- 10.63). The results indicated that the developed FFQ is valid instrument to assess the habitual intake of energy and most of the nutrients in healthy adults in the Republic of Croatia, while men tended to have higher correlation coefficients than women and therefore a higher correlation of the FFQ with the three 24-hour recalls. Statistically significant correlations are not found between energy and nutrient intake based on the education level of the participants.
