Abstract Background We aimed to develop multiple machine learning models to predict the risk of early intracranial aneurysms (IAs) rupture, evaluate and compare the performance of predictive models. Methods Information related to patients diagnosed with IA by CT angiography and clinicians in Central hospital of Dalian University of Technology from January 2010 to June 2022 was collected, including clinical characteristics, blood indicators and IA morphological parameters. IA with rupture or maximum growth ≥ 0.5 mm within 1 month of first diagnosis was considered unstable. The relevant factors affecting IA stability were screened and predictive models were developed based on the above three levels, including random forest (RF), support vector machine (SVM), and artificial neural network (ANN). Sensitivity, specificity, accuracy and area under curve (AUC) value were used to evaluate the predictive models. Results A total of 989 IA patients were included in the study, including 561 stable patients and 428 unstable patients. For RF models, the training set showed that sensitivity, specificity, accuracy and the AUC values were 72.8–83.7%, 76.9–86.9%, 75.1–84.1% and 0.748 (0.719–0.778)–0.839 (0.814–0.864), respectively; after test set validation, the results were 71.9–78.8%, 75.0–84.0%, 73.6–81.1% and 0.734 (0.688–0.781)–0.809 (0.768–0.850), respectively. For SVM models, the training set were 66.0–80.2%, 76.5–85.5%, 71.7–82.3%, 0.712 (0.682–0.743)–0.913 (0.884–0.924), respectively; the test set were 44.2–78.3%, 63.4–84.4%, 57.9–80.9%, 0.699 (0.651–0.747)–0.806 (0.765–0.848), respectively. For ANN models, the training set were 66.8–83.0%, 75.3–82.3%, 71.6–82.1%, 0.783 (0.757–0.808)–0.897 (0.879–0.914); the test set were 63.1–76.3%, 65.5–84.0%, 64.4–80.6%, 0.680 (0.593–0.694)–0.860 (0.821–0.899). The results of variable importance showed that age, white blood cell count (WBC) and uric acid (UA) played an important role in predicting the stability of IA. Conclusions The predictive stability models of IA based on three artificial intelligence methods shows good clinical application. Age, WBC and UA played an important role in predicting the IA stability, and were potentially important predictors.

Mild cognitive impairment (MCI) is a condition that impairs activities of daily living, and often transforms to dementia. Repetitive transcranial magnetic stimulation (rTMS) and transcranial direct current stimulation (tDCS) show promise in improving cognitive functions in MCI patients. In this meta-analysis, we aimed to compare the effects of rTMS and tDCS on memory functions in MCI patients. We explored eight databases from their inception to March 16, 2024. We obtained 11 studies with 406 patients with MCI. The pollination process of kiwifruit flowers plays a crucial role in kiwifruit yield. Achieving accurate and rapid identification of the four stages of kiwifruit flowers is essential for enhancing pollination efficiency. In this study, to improve the efficiency of kiwifruit pollination, we propose a novel full-stage kiwifruit flower pollination detection algorithm named KIWI-YOLO, based on the fusion of frequency-domain features. Our algorithm leverages frequency-domain and spatial-domain information to improve recognition of contour-detailed features and integrates decision-making with contextual information. Additionally, we incorporate the Bi-Level Routing Attention (BRA) mechanism with C3 to enhance the algorithm’s focus on critical areas, resulting in accurate, lightweight, and fast detection. The algorithm achieves a mAP0.5 of 91.6% with only 1.8M parameters, the AP of the Female class and the Male class reaches 95% and 93.5%, which is an improvement of 3.8%, 1.2%, and 6.2% compared with the original algorithm. Furthermore, the Recall and F1-score of the algorithm are enhanced by 5.5% and 3.1%, respectively. Moreover, our model demonstrates significant advantages in detection speed, taking only 0.016s to process an image. The experimental results show that the algorithmic model proposed in this study can better assist the pollination of kiwifruit in the process of precision agriculture production and help the development of the kiwifruit industry.

Tetrapyrrole compounds play vital roles in the life processes of animals and plants, such as respiration and photosynthesis, due to their functions in light energy capture and transmission, signal transduction, binding and transport of oxygen, their research originated from the study of heme and chlorophyll chemistry. The special structure of tetrapyrrole determines its various properties, which contribute to its antioxidant, anti-inflammatory, antitumoral, antibacterial, neuroprotective and other biological activities. Although many functions of tetrapyrrole compounds have been excavated and utilized, there is a lack of a complete system to summarize them. This review summarizes the biosynthesis, functional evolution, biological activities and optical applications of tetrapyrrole compounds based on existing research on tetrapyrrole compounds.

Post-harvest selection of high-quality Sichuan pepper is a critical step in the production process. To achieve this, a visual system needs to analyze Sichuan pepper with varying postures and maturity levels. To quickly and accurately sort high-quality fresh Sichuan pepper, this study proposes a multi-scale frequency domain feature fusion module (MSF3M) and a multi-scale dual-domain feature fusion module (MS-DFFM) to construct a multi-scale, multi-domain fusion algorithm for feature fusion of Sichuan pepper images. The MultiDomain YOLOv8 Model network is then built to segment and classify the target Sichuan pepper, distinguishing the maturity level of individual Sichuan peppercorns. A selection method based on the average local pixel value difference is proposed for sorting high-quality fresh Sichuan pepper. Experimental results show that the MultiDomain YOLOv8-seg achieves an mAP50 of 88.8% for the segmentation of fresh Sichuan pepper, with a model size of only 5.84 MB. The MultiDomain YOLOv8-cls excels in Sichuan pepper maturity classification, with an accuracy of 98.34%. Compared to the YOLOv8 baseline model, the MultiDomain YOLOv8 model offers higher accuracy and a more lightweight structure, making it highly effective in reducing misjudgments and enhancing post-harvest processing efficiency in agricultural applications, ultimately increasing producer profits.

Abstract Diabetes is one of the most common phenotypes of Wolfram syndrome owing to the presence of the variants of the WFS1 gene and is often misdiagnosed as other types of diabetes. We aimed to explore the prevalence of WFS1-related diabetes (WFS1-DM) and its clinical characteristics in a Chinese population with early-onset type 2 diabetes (EOD). We sequenced all exons of the WFS1 gene in 690 patients with EOD (age at diagnosis ≤ 40 years) for rare variants. Pathogenicity was defined according to the standards and guidelines of the American College of Medical Genetics and Genomics. We identified 33 rare variants predicted to be deleterious in 39 patients. The fasting [1.57(1.06–2.22) ng/ml] and postprandial C-peptide levels [2.8(1.75–4.46) ng/ml] of the patients with such WFS1 variations were lower than those of the patients without WFS1 variation [2.09(1.43–3.05) and 4.29(2.76–6.07) respectively, ng/ml]. Six (0.9%) patients carried pathogenic or likely pathogenic variants; they met the diagnostic criteria for WFS1-DM according to the latest guidelines, but typical phenotypes of Wolfram syndrome were seldom observed. They were diagnosed at an earlier age and usually presented with an absence of obesity, impaired beta cell function, and the need for insulin treatment. WFS1-DM is usually mistakenly diagnosed as type 2 diabetes, and genetic testing is helpful for individualized treatment.

In wireless communication, to fully utilize the spectrum and energy efficiency of the system, it is necessary to obtain the channel state information (CSI) of the link. However, in Frequency Division Duplexing (FDD) systems, CSI feedback wastes part of the spectrum resources. In order to save spectrum resources, the CSI needs to be compressed. However, many current deep-learning algorithms have complex structures and a large number of model parameters. When the computational and storage resources are limited, the large number of model parameters will decrease the accuracy of CSI feedback, which cannot meet the application requirements. In this paper, we propose a neural network-based CSI feedback model, Mix_Multi_TransNet, which considers both the spatial characteristics and temporal sequence of the channel, aiming to provide higher feedback accuracy while reducing the number of model parameters. Through experiments, it is found that Mix_Multi_TransNet achieves higher accuracy than the traditional CSI feedback network in both indoor and outdoor scenes. In the indoor scene, the NMSE gains of Mix_Multi_TransNet are 4.06 dB, 4.92 dB, 4.82 dB, and 6.47 dB for compression ratio η = 1/8, 1/16, 1/32, 1/64, respectively. In the outdoor scene, the NMSE gains of Mix_Multi_TransNet are 3.63 dB, 6.24 dB, 4.71 dB, 4.60 dB, and 2.93 dB for compression ratio η = 1/4, 1/8, 1/16, 1/32, 1/64, respectively.

Abstract Peanut is one of the important oil and economic crops, among which the variegated testa peanut is a unique member. The molecular mechanisms underlying the pigment synthesis in variegated testa are still unclear. Differentially expressed genes (DEGs) in the flavonoid metabolism pathway in pigmented areas indicated that there were 27 DEGs highly related to the synthesis of variegated testa color among 1,050 DEGs. Of these 27, 13 were up-regulated and 14 were down-regulated, including 3 PALs, 1 C4H, 2 CHSs, 1 F3H, 1 F3'H, 2 DFRs, 2 LARs, 2 IAAs, 4 bHLHs, and 9 MYBs. GO (Gene Ontology) analysis indicated that DEGs were similarly enriched in three branches. KEGG (Kyoto Encyclopedia of Genes and Genomes) analysis suggested flavonoid biosynthesis is the most direct metabolic pathway for the synthesis of testa variegation. The liquid chromatography–tandem mass spectrometry (LC–MS/MS) results showed that cyanidin and delphinidin were the primary metabolites that caused the color differences between the pigmented and the non-pigmented areas. Through the verification of 20 DEGs via qPCR, the results were consistent with transcriptome sequencing in four comparison groups. The results in this study lay the foundation for revealing the molecular regulation mechanisms of flavonoid synthesis in variegated testa peanut.

Peanut (Arachis hypogaea L.) is one of the most important economic and oil crops in the world. At present, peanut varieties with rich anthocyanin in testa are rare in the market, but the selection and breeding of varieties with the related traits has always attracted the attention of breeders. In this study, two peanut varieties with the pink and purple testa, G110 (G) and Z18-40 (Z) were used to conduct interaction joint analysis of multi-omics and miRNA-target gene. The anthocyanin content of Z18-40 was 7.49–8.62-folds higher than G110 on 30 DAF (days after flowering) and 45 DAF via Ultraviolet-visible Spectrophotometer (UV-5800, Shanghai, China). And then, a total of 14 candidate genes related with the anthocyanin biosynthesis were identified for correlation in different comparison groups (R2 ≥ 0.80), among of a novel gene Ah21440 related with hydroxycinnamoyl transferase (HCT) biosynthesis was identified. In addition, Cyanidin 3-O-glucoside (Kuromanin, pmb0550) was the only common differentially accumulated metabolite (DAM) identified using multi-omics joint analysis in G1_vs_G2, Z1_vs_Z2, G1_vs_Z1, and G2_vs_Z2, respectively. Correlation analysis of miRNA-target genes and DEGs in the transcriptome shows that, AhmiR2950, AhmiR398, AhmiR50, and AhmiR51 regulated to HCT and chalcone biosynthesis related candidate genes (Ah21440, AhCHS, AhCHI). Lastly, all of 14 candidate genes and 4 differentially expressed miRNAs were validated using quantitative real-time PCR (qRT-PCR), which trends were consistent with that of the former transcriptome data. The results provide important reference for in-depth research on the anthocyanin metabolism mechanism in peanut testa.

Joint surfaces are widely distributed in natural rock mass, and their shear mechanical properties play an important role in determining the safety and stability of rock mass. Previous studies rarely discussed the contribution degree of different joint protrusions to resisting shear stress. In this study, seven irregular dentate joint profiles were proposed to represent the geometric morphology of natural joints. Joint samples were subjected to direct shear tests under constant normal stress using the particle flow code (PFC). First, the reliability of the research scheme was verified by routine test results. Secondly, based on the microcrack tracking module and the force chain analysis, the local failure modes of the joint sample after and during the shearing process are discussed in detail. The relationship between the shear stress and the number of microcracks was studied. Finally, based on the measuring circle function, the variation law of the mean stress at different joint protrusions during the shearing process was tracked. The maximum stress of each protrusion before the shear stress peak was introduced to quantitatively describe the contribution of local protrusions to shear stress. There was an important link between the size of the protrusion and the stress it can withstand. During the shearing process, the local shearing mechanism of the joint surface is controlled by the distribution of joint protrusions. The research results of this paper can provide a good idea for the follow-up joint surface research.

Background: Spinal cord injury (SCI) is currently not completely curable. Exosomes have been widely used in preclinical studies of spinal cord injury. Here, in this meta-analysis, we focused on evaluating the overall efficacy of therapies based on miRNA-modified exosomes on functional recovery in animal models of SCI.Methods: PubMed, embase and Web of Science library databases were searched. Relevant literature was included, and the random effects model was used to assess the overall effect of the intervention, with outcomes expressed as SMD. The primary outcome included motor function scores. Risk of bias (ROB) was assessed using the ROB tool of the Systematic Review Centre for Laboratory Animal Experimentation (SYRCLE). R version 4.1.1software and Review Manager software were used for meta-analysis.Results: A total of 11 preclinical studies were included. The meta-analysis revealed that miRNA-modified exosome therapy was effective in improving motor function scores compared with exosomes alone or control therapy (standardized mean difference: 4.21; 95% confidence interval: 3.39–5.04). There was significant asymmetry in the funnel plot, and trim-and-fill analysis revealed four unpublished studies of motor scores. The quality of all included studies was evaluated with SYRCLE’s ROB tool. The SCI model, administration time and dose had an impact on the effect of the treatment.Conclusion: MiRNA-modified exosomes have shown great potential in the treatment of SCI. Moreover, the efficacy of miRNA-modified exosomes was superior to that of exosomes alone.

This study was conducted to elucidate the molecular mechanisms underlying heat stress (HS)-induced abnormal egg-laying in laying hens. Hy-Line brown laying hens were exposed to HS at 32 °C or maintained at 22 °C (control) for 14 days. In addition, granulosa cells (GCs) from preovulatory follicles were subjected to normal (37 °C) or high (41 °C or 43 °C) temperatures in vitro. Proliferation, apoptosis, and steroidogenesis were investigated, and the expression of estrogen and progesterone synthesis-related genes was detected. The results confirmed that laying hens reared under HS had impaired laying performance. HS inhibited proliferation, increased apoptosis, and altered the GC ultrastructure. HS also elevated progesterone secretion by increasing the expression of steroidogenic acute regulatory protein (StAR), cytochrome P450 family 11 subfamily A member 1 (CYP11A1), and 3b-hydroxysteroid dehydrogenase (3β-HSD). In addition, HS inhibited estrogen synthesis in GCs by decreasing the expression of the follicle-stimulating hormone receptor (FSHR) and cytochrome P450 family 19 subfamily A member 1 (CYP19A1). The upregulation of heat shock 70 kDa protein (HSP70) under HS was also observed. Collectively, laying hens exposed to high temperatures experienced damage to follicular GCs and steroidogenesis dysfunction, which reduced their laying performance. This study provides a molecular mechanism for the abnormal laying performance of hens subjected to HS.

ObjectiveMaturity-onset diabetes of the young caused by hepatocyte nuclear factor-1 alpha (HNF1A) variants (HNF1A-MODY) is a common form of monogenetic diabetes. Although patients with HNF1A-MODY might specifically benefit from sulfonylurea treatment, available methods for screening this specific type of diabetes are not cost-effective. This study was designed to establish an optimized clinical strategy based on multiple biomarkers to distinguish patients with HNF1A-MODY from clinically diagnosed early-onset type 2 diabetes (EOD) for genetic testing in a Chinese population.Research design and methodsA case–control study including 125 non-related young patients with EOD and 15 probands with HNF1A-MODY (cohort 1) was conducted to evaluate reported biomarkers for HNF1A-MODY. A cut-off for the fasting insulin (Fins) level, the 97.5 percentile of 150 healthy subjects with normal components of metabolic syndrome (cohort 2), was used to filter out individuals with obvious insulin resistance (Fins 1.12 mmol/L), the sensitivity and specificity of HNF1A-CSS were 100% and 69.3%, respectively. In the pooled analysis of all young patients, HNF1A-CSS displayed 90.5% sensitivity and 73.6% specificity for identifying patients with HNF1A-MODY among those with clinically diagnosed EOD.ConclusionOur HNF1A-CSS is useful for distinguishing patients with HNF1A-MODY from patients with EOD in a young Chinese population.

Background Glucagon‐like peptide‐1 receptor agonists (GLP‐1RAs) and sodium‐glucose cotransporter‐2 inhibitors (SGLT2is) have shown their beneficial effects on cardiovascular outcomes and multiple cardiovascular risk factors, including hypertension. However, the mechanism of blood pressure (BP)–lowering effects of these agents has not been elucidated. This study aims to evaluate the effect of hemoglobin A1c reduction or body weight reduction with GLP‐1RA treatment and SGLT2i treatment on BP changes in patients with type 2 diabetes mellitus. Abstract To achieve efficient and low energy consumption mining under different cutting depths of a shearer, a multiparameter coupling optimization method for the shearer based on three‐dimensional (3D) characterization of the coal wall was proposed. First, a seven‐axis absolute articulated arm measuring machine was used to obtain 3D point cloud data of the coal wall, and then the 3D of the coal wall surface was reconstructed by using segmentation, filtering, and stitching processing, thereby obtaining the average thickness of different coal wall areas. Second, through the quadratic rotation regression orthogonal combination experiment, the optimal combination of drum speed, traction speed, and cutting depth was obtained, further obtaining the order of primary and secondary influences, and the regression model. Moreover, a particle swarm optimization algorithm was used to obtain the optimal drum speed and finally, the laboratory and field test experiments were conducted to verify the effectiveness of the proposed optimization algorithm in reducing the cutting energy consumption of shearer. The experiment results show that the given optimization algorithm can adaptively optimize the traction speed and drum speed based on the corresponding cutting depth, which significantly reduces the cutting specific energy consumption of the shearer. Thus, it provided an important technical means for the shearer to achieve low energy consumption and efficient mining.

sHSP (small heat-shock protein) IbpB (inclusion-body-binding protein B) from Escherichia coli is known as an ATP-independent holding chaperone which prevents the insolubilization of aggregation-prone proteins by forming stable complexes with them. It was found that the chaperone function of IbpB is greatly modulated by the ambient temperature, i.e. when the temperature increases from normal to heat-shock, the chaperone activity of IbpB is dramatically elevated to a level that allows it to effectively bind the aggregation-prone client proteins. Although it is generally believed that the release and refolding of the client protein from the sHSPs depends on the aid of the ATP-dependent chaperones such as Hsp (heat-shock protein) 70 and Hsp100 when the ambient temperature recovers from heat-shock to normal, the behaviour of the sHSPs during this recovery stage has not yet been investigated. In the present study, we examined the behaviour and properties of IbpB upon temperature decrease from heat-shock to normal. We found that IbpB, which becomes functional only under heat-shock conditions, retains the chaperone activity for an extended period of time after the heat-shock stress condition is removed. A detail comparison demonstrates that such preconditioned IbpB is distinguished from the non-preconditioned IbpB by a remarkable conformational transformation, including a significant increase in the flexibility of the N- and C-terminal regions, as well as enhanced dynamic subunit dissociation/reassociation. Intriguingly, the preconditioned IbpB displayed a dramatic decrease in its surface hydrophobicity, suggesting that the exposure of hydrophobic sites might not be the sole determinant for IbpB to exhibit chaperone activity. Although previous studies have discussed the association between mobile phone use addiction (MPUA) and depressive symptoms in adolescents, studies on Chinese adolescents are limited, with no studies being reported on adolescents from ethnic minority areas within Yunnan. This study aimed to explore the mediating mechanism of insomnia in the association between MPUA and depressive symptoms among Yunnan college students. A sample survey was conducted among 10,121 college students from four colleges in Yunnan province, China, from November to December 2021 via cluster sampling. Insomnia was assessed using the Insomnia Severity Index Scale, MPUA was assessed using the Self-Rating Questionnaire for Adolescent Problematic Mobile Phone Use, and depressive symptoms were assessed using the Patient Health Questionnaire-9 items. A generalized linear model was used to analyze the association between MPUA, insomnia, and depressive symptoms. The structural equation model was used to analyze the mediating effect of insomnia. The positive rates of depressive symptoms, MPUA, and insomnia were 39.2 %, 30.8 %, and 22.3 %, respectively. After adjusting for the demographic and confounding variables, MPUA (β = 0.14, 95 % CI: 0.13–0.15, P < 0.01) and insomnia (β = 0.44, 95 % CI: 0.43–0.46, P < 0.01) were associated with depressive symptoms (P < 0.05). The mediating effect of insomnia accounted for 34.7 % of the total effect in the mediating model of MPUA associated with depressive symptoms. Our findings suggest that insomnia partially mediates the association between MPUA and depressive symptoms among Yunnan college students.
