Abstract Conferring drought resistant traits to crops is one of the major aims of current breeding programs in response to global climate changes. We previously showed that exogenous application of acetic acid to roots of various plants could induce increased survivability under subsequent drought stress conditions, but details of the metabolism of exogenously applied acetic acid, and the nature of signals induced by its application, have not been unveiled. In this study, we show that rice rapidly induces jasmonate signaling upon application of acetic acid, resulting in physiological changes similar to those seen under drought. The major metabolite of the exogenously applied acetic acid in xylem sap was determined as glutamine—a common and abundant component of xylem sap—indicating that acetic acid is not the direct agent inducing the observed physiological responses in shoots. Expression of drought-responsive genes in shoot under subsequent drought conditions was attenuated by acetic acid treatment. These data suggest that acetic acid activates root-to-shoot jasmonate signals that partially overlap with those induced by drought, thereby conferring an acclimated state on shoots prior to subsequent drought.

An electron cyclotron resonance (ECR) ion source (ECRIS) can generate an available amount of multicharged ions, thus it is not limited for use in the field of accelerator science, but also in medical/biological fields, such as for heavy ion beam cancer treatment and ion engines. The processes of generating multicharged ions are mainly sequential collisions of a direct ionization process by electrons, and have good ion confinement characteristics. By utilizing this confinement property, we have synthesized iron-encapsulated fullerenes, which are supramolecular and can be expected to have various high functions. Fullerenes and iron ions are vaporized from pure solid materials and introduced into the ECRIS together with the support gas. We investigated conditions under which fullerene ions do not dissociate and iron ions are generated so that both can coexist. Generated ions are extracted from the ECRIS and separated by mass/charge with a dipole magnet, and detected with a Faraday cup. This measurement system is characterized by a wide dynamic range. The charge-state distribution (CSD) of ion currents was measured to investigate the optimum conditions for supramolecular synthesis. As a result, a significant spectrum suggesting the possibility of iron-encapsulated fullerenes was obtained. This paper describes the details of these experimental results.

Radiotherapy using high-energy carbon beams from the Heavy Ion Medical Accelerator in Chiba has been carried out at theNational Institute of Radiological Sciences (NIRS) since June 1994, andmore than 10 000 patients have been treated to date. With the prospective clinical results for the first ten years, we designed a compact accelerator facility for carbon-ion radiotherapy (CIRT), and performed related Research and Development (R&D) works for widespread use of CIRT since 2004. Based on the design and the R&D works, three compact facilities for CIRT were constructed and are in treatment operation, and furthermore two facilities are being constructed in Japan. To further develop the sophisticated CIRT, we constructed a new treatment facility at NIRS. This new facility is equipped with three treatment rooms; two of them have both horizontal and vertical fixed-irradiation-ports, and the other is a rotating-gantry port. For all ports, fast three-dimensional raster-scanning irradiation technology is employed. The rotating gantry equips ten combined-function superconducting magnets, and can deliver carbon ions having the energy of between E = 430 and 50MeV/u to an isocenterwith irradiation angles of over±180°. Construction as well as installation of the superconducting gantry was completed by the end of September 2015, and treatments using the gantry began since May 2015. Presently, we are further designing a next-generation compact facility as a future project; superconducting and laser technologies are to be used for accelerator design. Phosphoenolpyruvate carboxylase (PEPC) is a key enzyme of primary metabolism in bacteria, algae and vascular plants, and it undergoes allosteric regulation by various metabolic effectors. Rice (Oryza sativa) has five plant-type PEPCs, four cytosolic and one chloroplastic. We investigated their kinetic properties using recombinant proteins and found that, like most plant-type PEPCs, rice cytosolic isozymes were activated by glucose 6-phosphate and by alkaline pH. In contrast, no such activation was observed for the chloroplastic isozyme, Osppc4. In addition, Osppc4 showed low affinity for the substrate phosphoenolpyruvate (PEP) and very low sensitivities to allosteric inhibitors aspartate and glutamate. By comparing the isozyme amino acid sequences and three-dimensional structures simulated on the basis of the reported crystal structures, we identified two regions where Osppc4 has unique features that can be expected to affect its kinetic properties. One is the N-terminal extension; replacement of the extension of Osppc2a (cytosolic) with that from Osppc4 reduced the aspartate and glutamate sensitivities to about one-tenth of the wild-type values but left the PEP affinity unaffected. The other is the N-terminal loop, in which a conserved lysine at the N-terminal end is replaced with a glutamate-alanine pair in Osppc4. Replacement of the lysine of Osppc2a with glutamate- alanine lowered the PEP affinity to a quarter of the wildtype level (down to the Osppc4 level), without affecting inhibitor sensitivity. Both the N-terminal extension and the N-terminal loop are specific to plant-type PEPCs, suggesting that plant-type isozymes acquired these regions so that their activity could be regulated properly at the sites where they function. We have been constructing the tandem-type electron cyclotron resonance ion source (ECRIS). Two ion sources of the tandem-type ECRIS are possible to generate plasma individually, and they also confined individual ion species by each different plasma parameter. Hence, it is considered to be suitable for new materials production. As the first step, we try to produce and extract multicharged C60 ions by supplying pure C60 vapor in the second stage plasma because our main target is producing the endohedral fullerenes. We developed a new evaporator to supply fullerene vapor, and we succeeded in observation about multicharged C60 ion beam in tandem-type ECRIS for the first time. [ABSTRACT FROM AUTHOR]

Multicharged metal ion beams are required to be applied in a wide range of fields. We aim at synthesizing iron-endohedral fullerene by transporting iron ion beams from the first stage into the fullerene plasma in the second stage of the tandem-type electron cyclotron resonance ion source (ECRIS). We developed new evaporators by using a direct ohmic heating method and a radiation heating method from solid state pure metal materials. We investigate their properties in the test chamber and produce iron ions on the first stage of the tandem-type ECRIS. As a result, we were successful in extracting Fe+ ion beams from the first stage and introducing Fe+ ion beams to the second stage.We will try synthesizing iron-endohedral fullerene on the tandem-type ECRIS by using these evaporators. Efficient production of multicharged ions has been investigated on the tandem-type ECRIS in Osaka University. According to the consideration of the accessibility conditions of microwaves to resonance and cutoff regions, it was suggested that the upper hybrid resonance (UHR) heating contributed to enhancement of ion beam intensity. In order to enhance multicharged ion beams efficiently, injecting higher frequency microwave with extraordinary (X-mode) toward UHR region has been tried. In this study, 2.45 GHz frequency microwaves are used for conventional ECR discharge, and 9.9 GHz frequency microwaves with X-mode are superimposed for UHR heating. The effects of additive microwave injection are investigated experimentally in terms of plasma parameters and electron energy distribution function (EEDF) measured by Langmuir probe and ion beam current. As the results show, it is confirmed that the electrons in the high energy region are affected by 9.9 GHz X-mode microwave injection from the detailed analysis of EEDF. [ABSTRACT FROM AUTHOR]

Cyanobacteria, algae and plants reduce photosystem contents and modulate photosystem stoichiometry upon acclimation to high-light (HL) conditions to avoid the damage due to excess light energy. In order to elucidate the molecular mechanism of HL acclimation, mRNA levels of genes encoding subunits of photosystems were examined in response to change of photon flux density in the wild-type strain of Synechocystis sp. PCC 6803. Transcript levels and promoter activities of photosystem I (PSI) genes rapidly decreased upon the shift to HL to less than 10% of the initial level within 1 h, whereas responses of photosystem II (PSII) transcript levels were not coordinated. The prompt change in promoter activities of PSI genes, but not PSII genes, seems important for Synechocystis cells to regulate their photosystem contents in response to changes in photon flux density. The mRNA stabilities of PSI genes also decreased during HL incubation. The down-regulation of PSI transcripts under HL did not depend on de novo protein synthesis, contrasting with the requirement of newly synthesized protein factor(s) for the accumulation of PSI transcripts under low light. When gene expression profiles of wild-type cells and a pmgA-disrupted mutant that cannot modulate photosystem stoichiometry under HL were compared, a large increase in the psaAB and psaA transcript levels was observed in the mutant under prolonged exposure to HL for 6 h. Repression of the promoter activity of the psaA gene by a pmgA-dependent mechanism was shown to be essential for the adjustment of photosystem stoichiometry under HL conditions. Solid-state materials suitable for use as proton irradiation targets were investigated for producing high-purity 11CH4 molecules for heavy-ion cancer therapy. The radioactivity of gas produced by proton irradiation was measured for several target materials. Also, the radioactive molecular species of the produced gas were analyzed by radio gas chromatography. We found that 5 x 1012 11C molecules could be collected by proton irradiation on a NaBH4 target. We also found that the 11CH4 molecules were produced and collected directly from the irradiated target, owing to the hydrogen atoms bound in the solid-state NaBH4. [ABSTRACT FROM AUTHOR]
