Abstract In the race to curb energy and oil consumption, zeroing of wall frictional forces is highly desirable. The turbulent skin friction drag at the solid/liquid interface is responsible for substantial energy losses when conveying liquids through hydraulic networks, contributing approximately 10% to the global electric energy consumption. Despite extensive research, efficient drag reduction strategies effectively applicable in different flow regimes are still unavailable. Here, we use a wall-attached magnetic fluid film to achieve a wall drag reduction of up to 90% in channel flow. Using optical measurements supported by modelling, we find that the strong damping of wall friction emerges from the co-existence of slip and waviness at the coating interface, and the latter is a key factor to obtain almost complete wall drag reduction across laminar and turbulent flow regimes. Our magnetic fluid film is promising and ready to be applied in energy-saving and antifouling strategies in fluid transport and medical devices.

Abstract The hemodynamics in the aorta as well as the durability of aortic valve prostheses vary greatly between different types of devices. Although placement and sizing of surgical aortic valve prostheses are excellent, the valve geometry of common devices cannot be customized to fit the patient’s anatomy perfectly. Similarly, transcatheter aortic valve implantation (TAVI) devices are not customizable and may be orientated unfavorably during implantation. Imperfect fit of an aortic valve prosthesis may result in suboptimal performance and in some cases the need for additional surgery. Leveraging the advent of precision, multi-material 3D-printing, a bioinspired silicone aortic valve was developed. The manufacturing technique makes it fully customizable and significantly cheaper to develop and produce than common prostheses. In this study, we assess the hemodynamic performance of such a 3D-printed aortic valve and compare it to two TAVI devices as well as to a severely stenosed valve. We investigate the blood flow distal to the valve in an anatomically accurate, compliant aorta model via three-dimensional particle tracking velocimetry measurements. Our results demonstrate that the 3D-printed aortic valve induces flow patterns and topology compatible with the TAVI valves and showing similarity to healthy aortic blood flow. Compared to the stenosis, the 3D-printed aortic valve reduces turbulent kinetic energy levels and irreversible energy losses by over 75%, reaching values compatible with healthy subjects and conventional TAVIs. Our study substantiates that the 3D-printed heart valve displays a hemodynamic performance similar to established devices and underscores its potential for driving innovation towards patient specific valve prostheses.

Abstract Countless processes in nature and industry, from rain droplet nucleation to plankton interaction in the ocean, are intimately related to turbulent fluctuations of local concentrations of advected matter. These fluctuations can be described by considering the change of the separation between particle pairs, known as pair dispersion, which is believed to obey a cubic in time growth according to Richardson’s theory. Our work reveals a universal, scale-invariant alignment between the relative velocity and position vectors of dispersing particles at a mean angle that we show to be a universal constant of turbulence. We connect the value of this mean angle to Richardson’s traditional theory and find agreement with data from a numerical simulation and a laboratory experiment. While the Richardson’s cubic regime has been observed for small initial particle separations only, the constancy of the mean angle manifests throughout the entire inertial range of turbulence. Thus, our work reveals the universal nature of turbulent pair dispersion through a geometrical paradigm whose validity goes beyond the classical theory, and provides a framework for understanding and modeling transport and mixing processes.

Abstract Carbon efflux from soils is the largest terrestrial carbon source to the atmosphere, yet it is still one of the most uncertain fluxes in the Earth’s carbon budget. A dominant component of this flux is heterotrophic respiration, influenced by several environmental factors, most notably soil temperature and moisture. Here, we develop a mechanistic model from micro to global scale to explore how changes in soil water content and temperature affect soil heterotrophic respiration. Simulations, laboratory measurements, and field observations validate the new approach. Estimates from the model show that heterotrophic respiration has been increasing since the 1980s at a rate of about 2% per decade globally. Using future projections of surface temperature and soil moisture, the model predicts a global increase of about 40% in heterotrophic respiration by the end of the century under the worst-case emission scenario, where the Arctic region is expected to experience a more than two-fold increase, driven primarily by declining soil moisture rather than temperature increase.

The Hedgehog (HH) pathway is crucial for embryonic development, and adult homeostasis. Its dysregulation is implicated in multiple diseases. Existing cellular models used to study HH signal regulation in mammals do not fully recapitulate the complexity of the pathway. Here we show that Spinal Cord Organoids (SCOs) can be applied to quantitively study the activity of the HH pathway. During SCO formation, the specification of different categories of neural progenitors (NPC) depends on the intensity of the HH signal, mirroring the process that occurs during neural tube development. By assessing the number of NPCs within these distinct subgroups, we are able to categorize and quantify the activation level of the HH pathway. We validate this system by measuring the effects of mutating the HH receptor PTCH1 and the impact of HH agonists and antagonists on NPC specification. SCOs represent an accessible and reliable in-vitro tool to quantify HH signaling and investigate the contribution of genetic and chemical cues in the HH pathway regulation.

Abstract Phototrophic biofilms form complex spatial patterns in streams and rivers, yet, how community patchiness, structure and function are coupled and contribute to larger-scale metabolism remains unkown. Here, we combined optical coherence tomography with automated O2 microprofiling and amplicon sequencing in a flume experiment to show how distinct community patches interact with the hydraulic environment and how this affects the internal distribution of oxygen. We used numerical simulations to derive rates of community photosynthetic activity and respiration at the patch scale and use the obtained parameter to upscale from individual patches to the larger biofilm landscape. Our biofilm landscape approach revealed evidence of parallels in the structure-function coupling between phototrophic biofilms and their streambed habitat.

Hedgehog (HH) signaling is important for embryonic pattering and stem cell differentiation. The G protein-coupled receptor (GPCR) Smoothened (SMO) is the key HH signal transducer modulating both transcription-dependent and transcription-independent responses. We show that SMO protects naive mouse embryonic stem cells (ESCs) from dissociation-induced cell death. We exploited this SMO dependency to perform a genetic screen in haploid ESCs where we identify the Golgi proteins TMED2 and TMED10 as factors for SMO regulation. Super-resolution microscopy shows that SMO is normally retained in the endoplasmic reticulum (ER) and Golgi compartments, and we demonstrate that TMED2 binds to SMO, preventing localization to the plasma membrane. Mutation of TMED2 allows SMO accumulation at the plasma membrane, recapitulating early events after HH stimulation. We demonstrate the physiologic relevance of this interaction in neural differentiation, where TMED2 functions to repress HH signal strength. Identification of TMED2 as a binder and upstream regulator of SMO opens the way for unraveling the events in the ER-Golgi leading to HH signaling activation.

Oct4, along with Sox2 and Klf4 can induce pluripotency, but structurally similar factors like Oct6 cannot. Here, using pluripotency competent and incompetent factors, the authors show that Sox2 plays a dominant role in facilitating chromatin opening at Oct4 bound DNA early during reprogramming to pluripotency.

Zooplankton live in dynamic environments where turbulence may challenge their limited swimming abilities. How this interferes with fundamental behavioral processes remains elusive. We reconstruct simultaneously the trajectories of flow tracers and calanoid copepods and we quantify their ability to find mates when ambient flow imposes physical constrains on their motion and impairs their olfactory orientation. We show that copepods achieve high encounter rates in turbulence due to the contribution of advection and vigorous swimming. Males further convert encounters within the perception radius to contacts and then to mating via directed motion toward nearby organisms within the short time frame of the encounter. Inertial effects do not result in preferential concentration, reducing the geometric collision kernel to the clearance rate, which we model accurately by superposing turbulent velocity and organism motion. This behavioral and physical coupling mechanism may account for the ability of copepods to reproduce in turbulent environments.

The generation of electricity by hydropower plants meets the requirements for intermittent energy demand and for the production of renewable energy. However, the hydropower plant operation has important consequences for the biotic compartment of the river reaches downstream. Intermittent release of water causes sudden variations in discharge that increase bed shear stress and dislodge benthic organisms. Release from high-elevation reservoir can also affect the thermal regime of the river by causing sharp variations in water temperature. Because the hydrodynamic and thermal waves separate while propagating downstream, the benthic community is exposed to two distinct stressors that affect taxa differently based on their sensitivity and adaptations. We investigated separately the effects of a sudden variation in discharge or in water temperature on the small-scale swimming behavior of a widespread species of cyclopoid copepod in a laboratory flume that allows the tracking of organisms both in the water column and in a transparent sediment bed. We gradually varied the discharge or the temperature of the water to mimic the artificial changes caused by hydropower plants. We tracked copepods in three dimensions and quantified the kinematics of their motion. Copepods increased substantially their counter-current swimming effort in response to increasing flow velocity. This behavioral response seems to occur above a threshold in flow velocity of approx. 40 mm/s. It results in a substantial reduction in their downstream transport and hence opposes drift. Copepods reacted differently to warm and cold variations in temperature. Decreasing temperature resulted in a substantially lower counter-current swimming effort, which may therefore increase drift. Rising temperature had no clear effect on their behavior. Our study highlights the importance of understanding the behavioral traits that mediate the response of stream invertebrates to disturbances in the hydraulic and thermal regimes of their environment.

We propose and validate a novel experimental technique to measure two-point statistics of turbulent flows. It consists of spreading rigid fibers in the flow and tracking their position and orientation in time and is therefore named “fiber tracking velocimetry.” By choosing different fiber lengths, i.e., within the inertial or dissipative range of scales, the statistics of turbulence fluctuations at the selected length scale can be probed accurately by simply measuring the fiber velocity at its two ends and projecting it along the transverse-to-fiber direction. By means of fully resolved direct numerical simulations and experiments, we show that these fiber-based transverse velocity increments are statistically equivalent to the (unperturbed) flow transverse velocity increments. Moreover, we show that the turbulent energy-dissipation rate can be accurately measured exploiting sufficiently short fibers. The technique is tested against standard particle tracking velocimetry (PTV) of flow tracers with excellent agreement. Our technique overcomes the well-known problem of PTV to probe two-point statistics reliably because of the fast relative diffusion in turbulence that prevents the mutual distance between particles to remain constant at the length scale of interest. This problem, making it difficult to obtain converged statistics for a fixed separation distance, is even more dramatic for natural flows in open domains. A prominent example is oceanic currents, where drifters (i.e., the tracer-particle counterpart used in field measurements) disperse quickly, but at the same time their number has to be limited to save costs. Inspired by our laboratory experiments, we propose pairs of connected drifters as a viable option to solve the issue.

When non-Newtonian fluids flow through porous media, the topology of the pore space leads to a broad range of flow velocities and shear rates. Consequently, the local viscosity of the fluid also varies in space with a non-linear dependence on the Darcy velocity. Therefore, an effective viscosity μeff is usually used to describe the flow at the Darcy scale. For most non-Newtonian flows the rheology of the fluid can be described by a (non linear) function of the shear rate. Current approaches estimate the effective viscosity by first calculating an effective shear rate mainly by adopting a power-law model for the rheology and including an empirical correction factor. In a second step this averaged shear rate is used together with the real rheology of the fluid to calculate μeff. In this work, we derive a semi-analytical expression for the local viscosity profile using a Carreau type fluid, which is a more broadly applicable model than the power-law model. By solving the flow equations in a circular cross section of a capillary we are able to calculate the average viscous resistance 〈μ〉 directly as a spatial average of the local viscosity. This approach circumvents the use of classical capillary bundle models and allows to upscale the viscosity distribution in a pore with a mean pore size to the Darcy scale. Different from commonly used capillary bundle models, the presented approach does neither require tortuosity nor permeability as input parameters. Consequently, our model only uses the characteristic length scale of the porous media and does not require empirical coefficients. The comparison of the proposed model with flow cell experiments conducted in a packed bed of monodisperse spherical beads shows, that our approach performs well by only using the physical rheology of the fluid, the porosity and the estimated mean pore size, without the need to determine an effective shear rate. The good agreement of our model with flow experiments and existing models suggests that the mean viscosity 〈μ〉 is a good estimate for the effective Darcy viscosity μeff providing physical insight into upscaling of non-Newtonian flows in porous media.

Advancements in optical imaging devices and computer vision algorithms allow the exploration of novel diagnostic techniques for use within engineering systems. A recent field of application lies in the adoption of such devices for non-contact vibrational response recordings of structures, allowing high spatial density measurements without the burden of heavy cabling associated with conventional technologies. This, however, is not a straightforward task due to the typically low-amplitude displacement response of structures under ambient operational conditions. A novel framework, namely Magnified Tracking (MT), is proposed herein to overcome this limitation through the synergistic use of two computer vision techniques. The recently proposed phase-based motion magnification (PBMM) framework, for amplifying motion in a video within a defined frequency band, is coupled with motion tracking by means of particle tracking velocimetry (PTV). An experimental campaign was conducted to validate a proof-of-concept, where the dynamic response of a shear frame was measured both by conventional sensors as well as a video camera setup, and cross-compared to prove the feasibility of the proposed non-contact approach. The methodology was explored both in 2D and 3D configurations, with PTV revealing a powerful tool for the measurement of perceptible motion. When MT is utilized for tracking “imperceptible” structural responses (i.e., below PTV sensitivity), via the use of PBMM around the resonant frequencies of the structure, the amplified motion reveals the operational deflection shapes, which are otherwise intractable. The modal results extracted from the magnified videos, using PTV, demonstrate MT to be a viable non-contact alternative for 3D modal identification with the benefit of a spatially dense measurement grid.

X-ray tomography is a powerful tool giving access to the morphology of biofilms, in 3D porous media, at the mesoscale. Due to the high water content of biofilms, the attenuation coefficient of biofilms and water are very close, hindering the distinction between biofilms and water without the use of contrast agents. Until now, the use of contrast agents such as barium sulfate, silver-coated micro-particles or 1-chloronaphtalene added to the liquid phase allowed imaging the biofilm 3D morphology. However, these contrast agents are not passive and potentially interact with the biofilm when injected into the sample. Here, we use a natural inorganic compound, namely iron sulfate, as a contrast agent progressively bounded in dilute or colloidal form into the EPS matrix during biofilm growth. By combining a very long source-to-detector distance on a X-ray laboratory source with a Lorentzian filter implemented prior to tomographic reconstruction, we substantially increase the contrast between the biofilm and the surrounding liquid, which allows revealing the 3D biofilm morphology. A comparison of this new method with the method proposed by Davit et al (Davit et al., 2011), which uses barium sulfate as a contrast agent to mark the liquid phase was performed. Quantitative evaluations between the methods revealed substantial differences for the volumetric fractions obtained from both methods. Namely, contrast agent-biofilm interactions (e.g. biofilm detachment) occurring during barium sulfate injection caused a reduction of the biofilm volumetric fraction of more than 50% and displacement of biofilm patches elsewhere in the column. Two key advantages of the newly proposed method are that passive addition of iron sulfate maintains the integrity of the biofilm prior to imaging, and that the biofilm itself is marked by the contrast agent, rather than the liquid phase as in other available methods. The iron sulfate method presented can be applied to understand biofilm development and bioclogging mechanisms in porous materials and the obtained biofilm morphology could be an ideal basis for 3D numerical calculations of hydrodynamic conditions to investigate biofilm-flow coupling.

We report an analysis of small-scale enstrophy ?2and rate of strain s2dynamics in the proximity of the turbulent/non-turbulent interface in a flow without strong mean shear. The techniques used are three-dimensional particle tracking (3D-PTV), allowing the field of velocity derivatives to be measured and followed in a Lagrangian manner, and direct numerical simulations (DNS). In both experiment and simulation the Taylor-microscale Reynolds number is Re?= 50. The results are based on the Lagrangian viewpoint with the main focus on flow particle tracers crossing the turbulent/non-turbulent interface. This approach allowed a direct investigation of the key physical processes underlying the entrainment phenomenon and revealed the role of small-scale non-local, inviscid and viscous processes. We found that the entrainment mechanism is initiated by self-amplification of s2through the combined effect of strain production and pressure--strain interaction. This process is followed by a sharp change of ?2induced mostly by production due to viscous effects. The influence of inviscid production is initially small but gradually increasing, whereas viscous production changes abruptly towards the destruction of ?2. Finally, shortly after the crossing of the turbulent/non-turbulent interface, production and dissipation of both enstrophy and strain reach a balance. In this article, we present an experimental setup and data processing schemes for 3D scanning particle tracking velocimetry (SPTV), which expands on the classical 3D particle tracking velocimetry (PTV) through changes in the illumination, image acquisition and analysis. 3D PTV is a flexible flow measurement technique based on the processing of stereoscopic images of flow tracer particles. The technique allows obtaining Lagrangian flow information directly from measured 3D trajectories of individual particles. While for a classical PTV the entire region of interest is simultaneously illuminated and recorded, in SPTV the flow field is recorded by sequential tomographic high-speed imaging of the region of interest. The advantage of the presented method is a considerable increase in maximum feasible seeding density. Results are shown for an experiment in homogenous turbulence and compared with PTV. SPTV yielded an average 3,500 tracked particles per time step, which implies a significant enhancement of the spatial resolution for Lagrangian flow measurements. [ABSTRACT FROM AUTHOR]

Abstract We present a novel stochastic model for quantifying gross solids (GS) physical disintegration under varying turbulent flow conditions and used a unique experimental setup for model calibration and validation. The stochastic deterioration model predicts faeces size evolution over time. It conceptually entails the two main processes of solid fragmentation, namely breakage and erosion. Model parameters were calibrated on synthetic faeces and validated with real human ones. A cylindrical reactor was used, where turbulent flow was forced by an array of water jets and the physical disintegration of the faeces was monitored using a high speed camera. Image analysis of breakage experiments obtained under backlight illumination allowed determination of the evolution of the solids' size over time. The flow field in the reactor was characterised by particle image velocimetry (PIV) using tracer particles seeded into the water. We found different disintegration behaviours depending on turbulence intensity and water content of the solid. In conditions of low shear stress, dense solids hardly disintegrated. Generally, the model predictions mirrored the broad range in the solids disintegration rate imparted by the high variability in flow conditions and in solids characteristics. It is expected that, similar to our experiments, also in real sewer systems both flow conditions and solid characteristics are highly variable and the stochastic model can be tailored to capture this variability. We thus anticipate that the model can be integrated into existing sewer models predicting sewer flows and solids' movement. From these, shear stress, flow velocities and transport of individual solids can be inferred. The integration of the present solids disintegration model may provide better predictions of hot-spots for solids accumulation and blockages in sewers.
