DNA methylation is an important mechanism for epigenetic modifications that have been shown to be associated with responses to plant development. Previous studies found that inverted Populus yunnanensis cuttings were still viable and could develop into complete plants. However, the growth status of inverted cuttings was weaker than that of upright cuttings, and the sprouting time of inverted cuttings was later than that of upright cuttings. There is currently no research on DNA methylation patterns in inverted cuttings of Populus yunnanensis. In this study, we detected genome-wide methylation patterns of stem tips of Populus yunnanensis at the early growth stage and the rapid growth stage by Oxford Nanopore Technologies (ONT) methylation sequencing. We found that the methylation levels of CpG, CHG, CHH, and 6mA were 41.34%, 33.79%, 17.27%, and 12.90%, respectively, in the genome of inverted poplar cuttings, while the methylation levels of the four methylation types were higher in the genome of upright poplar cuttings than in inverted cuttings, 41.90%, 34.57%, 18.09%, and 14.11%, suggesting important roles for DNA methylation in poplar cells. In all comparison groups, CpG-type methylation genes in the Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway were annotated to pathways associated with carbon metabolism, ribosome biogenesis in eukaryotes, glycolysis/gluconeogenesis, pyruvate metabolism, and mRNA detection pathways, suggesting that different biological processes are activated in upright and inverted cuttings. The results show that methylation genes are commonly present in the poplar genome, but only a few of them are involved in the regulation of expression in the growth and development of inverted cuttings. From this, we screened the DET2 gene for significant differences in methylation levels in upright or inverted cuttings. The DET2 gene is a key gene in the Brassinolide (BRs) synthesis pathway, and BRs have an important influence on the growth and development process of plants. The film temperature distribution of the valve plate pair in axial piston pumps affects its lubrication, leakage, and friction. In order to investigate the film temperature distribution of the valve plate pair in axial piston pumps, a test platform was constructed including three displacement sensors for the oil film thickness and eleven thermocouples for the film temperature distribution of the valve plate pair. An accurate film shape model of the valve plate pair was built according to the three-point film thickness test data. Based on the film shape model, the film temperature model of the valve plate pair was developed considering the viscous oil temperature characteristics, the energy loss caused by leakage and viscous friction in the film, and the heat conduction among the oil, cylinder block, and valve plate. The influence of different swash plate tilt angles and operating pressures on the valve plate film temperature was studied. The test results indicate that the film temperature of the valve plate pair increases as the working pressure and swash plate tilt angle increase. The theoretical and experimental absolute errors of the film temperature in the circumferential range [−60°, 60°] of the valve plate high-pressure side are less than 3.5 °C. As the swash plate tilt angle varies from 12° to 16° and working pressure from 3 MPa to 7 MPa, the minimum film thickness position and the maximum temperature point move accordingly in the circumferential range [−15°, 5°] of the valve plate pair. Hydrocephalus, characterized by cerebral ventriculomegaly, is the most common disorder requiring brain surgery in children. Recent studies have implicated SMARCC1 , a component of the BRG1-associated factor (BAF) chromatin remodelling complex, as a candidate congenital hydrocephalus gene. However, SMARCC1 variants have not been systematically examined in a large patient cohort or conclusively linked with a human syndrome. Moreover, congenital hydrocephalus-associated SMARCC1 variants have not been functionally validated or mechanistically studied in vivo. Here, we aimed to assess the prevalence of SMARCC1 variants in an expanded patient cohort, describe associated clinical and radiographic phenotypes, and assess the impact of Smarcc1 depletion in a novel Xenopus tropicalis model of congenital hydrocephalus. To do this, we performed a genetic association study using whole-exome sequencing from a cohort consisting of 2697 total ventriculomegalic trios, including patients with neurosurgically-treated congenital hydrocephalus, that total 8091 exomes collected over 7 years (2016–23). A comparison control cohort consisted of 1798 exomes from unaffected siblings of patients with autism spectrum disorder and their unaffected parents were sourced from the Simons Simplex Collection. Enrichment and impact on protein structure were assessed in identified variants. Effects on the human fetal brain transcriptome were examined with RNA-sequencing and Smarcc1 knockdowns were generated in Xenopus and studied using optical coherence tomography imaging, in situ hybridization and immunofluorescence. SMARCC1 surpassed genome-wide significance thresholds, yielding six rare, protein-altering de novo variants localized to highly conserved residues in key functional domains. Patients exhibited hydrocephalus with aqueductal stenosis; corpus callosum abnormalities, developmental delay, and cardiac defects were also common. Xenopus knockdowns recapitulated both aqueductal stenosis and cardiac defects and were rescued by wild-type but not patient-specific variant SMARCC1. Hydrocephalic SMARCC1-variant human fetal brain and Smarcc1-variant Xenopus brain exhibited a similarly altered expression of key genes linked to midgestational neurogenesis, including the transcription factors NEUROD2 and MAB21L2. These results suggest de novo variants in SMARCC1 cause a novel human BAFopathy we term 'SMARCC1-associated developmental dysgenesis syndrome', characterized by variable presence of cerebral ventriculomegaly, aqueductal stenosis, developmental delay and a variety of structural brain or cardiac defects. These data underscore the importance of SMARCC1 and the BAF chromatin remodelling complex for human brain morphogenesis and provide evidence for a 'neural stem cell' paradigm of congenital hydrocephalus pathogenesis. These results highlight utility of trio-based whole-exome sequencing for identifying pathogenic variants in sporadic congenital structural brain disorders and suggest whole-exome sequencing may be a valuable adjunct in clinical management of congenital hydrocephalus patients. A semi-packed gas chromatographic column has the advantages of high specific surface area and low column pressure. We report that the stagnation regions formed in the adjacent posts along the channel of the semi-packed columns can decrease the area and height of chromatographic peaks, which makes it difficult to detect low-concentration mixed gases. A semi-packed column with staggered elliptic cylindrical post arrays (SC-S) made using a micro-electro-mechanical system technique is presented, and the separation performance of SC-S is compared with that of a semi-packed column with aligned elliptic cylindrical post arrays (SC-A). The simulation results show that the width of stagnation regions in SC-S is 86.89% smaller than that in SC-A. The experimental results indicate that the area and height of chromatographic peaks increased as stagnation regions reduced. In the separation of the alkane mixture from C8 through C10 with 10 ppm concentration, the chromatographic peak of decane was hardly identified in SC-A while the chromatographic peak in SC-S was still clearly visible. The chromatographic peak heights of octane and nonane were increased by 65.06% and 130.00%, respectively, in SC-S. SUMMARY: Rosa roxburghii and Rosa sterilis, two species belonging to the Rosaceae family, are widespread in the southwest of China. These species have gained recognition for their remarkable abundance of ascorbate in their fresh fruits, making them an ideal vitamin C resource. In this study, we generated two high‐quality chromosome‐scale genome assemblies for R. roxburghii and R. sterilis, with genome sizes of 504 and 981.2 Mb, respectively. Notably, we present a haplotype‐resolved, chromosome‐scale assembly for diploid R. sterilis. Our results indicated that R. sterilis originated from the hybridization of R. roxburghii and R. longicuspis. Genome analysis revealed the absence of recent whole‐genome duplications in both species and identified a series of duplicated genes that possibly contributing to the accumulation of flavonoids. We identified two genes in the ascorbate synthesis pathway, GGP and GalLDH, that show signs of positive selection, along with high expression levels of GDP‐d‐mannose 3′, 5′‐epimerase (GME) and GDP‐l‐galactose phosphorylase (GGP) during fruit development. Furthermore, through co‐expression network analysis, we identified key hub genes (MYB5 and bZIP) that likely regulate genes in the ascorbate synthesis pathway, promoting ascorbate biosynthesis. Additionally, we observed the expansion of terpene synthase genes in these two species and tissue expression patterns, suggesting their involvement in terpenoid biosynthesis. Having a spiral grain is considered to be one of the most important wood properties influencing wood quality. Here, transcriptome profiles and metabolome data were analyzed in the straight grain and twist grain of Pinus yunnanensis. A total of 6644 differential expression genes were found between the straight type and the twist type. A total of 126 differentially accumulated metabolites were detected. There were 24 common differential pathways identified from the transcriptome and metabolome, and these pathways were mainly annotated in ABC transporters, arginine and proline metabolism, flavonoid biosynthesis, isoquinoline alkaloid biosynthesis, linoleic acid metabolism, phenylpropanoid, tryptophan metabolism, etc. A weighted gene coexpression network analysis showed that the lightblue4 module was significantly correlated with 2′-deoxyuridine and that transcription factors (basic leucine zipper (bZIP), homeodomain leucine zipper (HD-ZIP), basic helix–loop–helix (bHLH), p-coumarate 3-hydroxylase (C3H), and N-acetylcysteine (NAC)) play important roles in regulating 2′-deoxyuridine, which may be involved in the formation of spiral grains. Meanwhile, the signal transduction of hormones may be related to spiral grain, as previously reported. ARF7 and MKK4_5, as indoleacetic acid (IAA)- and ethylene (ET)-related receptors, may explain the contribution of plant hormones in spiral grain. This paper considers the problem of multiple missiles to attack a maneuvering target simultaneously. Based on a fixed-time fast nonsingular terminal sliding mode surface (FNTSMS), fixed-time distributed cooperative guidance laws are developed. By designing guidance laws along and perpendicular to line of sight (LOS) direction, both relative velocity and range-to-go are consistent. In addition, a saturation function is introduced to avoid the chattering problem caused by the commonly used sign function. Furthermore, a distributed cooperative guidance law with communication failure is considered and proved theoretically, which shows that the proposed guidance law has excellent performance. The mechanosensitive ion channel of large conductance (MscL) is a promising template for the development of new antibiotics due to its high conservation and uniqueness to microbes. Brilliant green (BG), a triarylmethane dye, has been identified as a new antibiotic targeted MscL. However, the detailed binding sites to MscL and the dynamic pathway of BG through the MscL channel remain unknown. Here, the dynamic interactions between BG and MscL were investigated using solid‐state NMR spectroscopy and molecule dynamics (MD) simulations. Residue site‐specific binding sites of BG to the MscL channel were identified by solid‐state NMR. In addition, MD simulations revealed that BG conducts through the MscL channel via residues along the inner surface of the pore sequentially, in which the strong hydrophobic interactions between BG and hydrophobic residues F23 and I27 in the hydrophobic gate region of the MscL channel are major restrictions. Particularly, it was demonstrated that BG activates the MscL channel by reducing the hydrophobicity of the F23 in the gate region by water molecules that are bound to BG. Taken together, these simulations and experimental data provide novel insights into the dynamic interactions between BG and MscL, based on which new hydrophobic antibiotics and adjuvants targeting MscL can be developed. Exploring the phenomenon of surface cracks and the abnormal phenomenon of ground behavior during coal mining under gully landforms, laboratory physical similarity simulation models were established to study the movement characteristics and ground behavior laws of working faces of different mining methods under gully landforms. The results indicate that in cases when a roadway is situated below the top of the slope, the corresponding deformation of the surrounding rock of the roadway is aggravated by the mining stress. Furthermore, when a roadway is located below the base of the gully, it is less affected by mining; thus, it could remain relatively stable. The ground behavior laws of working faces in gully geomorphology areas are associated with the position of the working face and the direction of working: when mining towards the gully, the ground behavior appeared relatively gentle and the surface slipped towards the gully; when mining away from the gully, the ground behavior appeared intense, the pressure was sudden and short, and with the increase in overburden thickness, the interval distance periodically decreased. When a working face passes through a gully, dumping of the hydraulic support should be prevented in the section of mining towards the gully; in sections of mining away from gully, the support strength should be strengthened to prevent the support and other equipment from being crushed. In actual mining, mining towards a gully should be adopted as much as possible in the stoping of the working face. Quantitative yet simple analytical solutions of surface temperature under pulsed laser illumination are presented for a quick estimation in optical spectroscopy studies. Dependence of steady state surface temperature as well as its temporal evolution on laser parameters, such as repetition rate and beam radius, together with medium properties is thoroughly investigated using the analytical solution, which is supported by numerical simulation. It is found that when the pulse number is larger than 100 within the heat diffusion time, the steady-state temperature rise reaches more than 85% of the temperature rise induced by CW laser heating of the same power. We provide a summary of the results to allow their use for a quick estimate of surface temperature evolution from pulse laser heating if laser parameters and medium properties are known. Aquaporins (AQPs) transport water molecules across cell membranes. Although most aquaporins are inhibited by mercury ions, AQP6 was reported to be activated by binding mercury ions to residues C155 and C190. Different from C190 and the other pore-line cysteine residues, C155 is located outside the pore, thus not directly affecting the internal pathway by mercury binding to it. The molecular mechanism of unusual water channel activation by mercury ion binding to the C155 site remains unknown. Here, we investigate the activation of AQP6 by mercury ions binding to C155 by molecular dynamics (MD) simulations. The MD simulation results show that the mercury-induced water permeation activation is derived from the conformational change of a pore-line residue M160, from a point-to-pore conformation before mercury binding to an away-pore conformation after mercury binding. The conformation change of M160 is derived from the reduction of the hydrogen bonding between C155 and S159 in the α-helix with the coordination of C155 to mercury ion altering their conformation significantly. This study reveals the complex mechanism of water channel activation by mercury ion binding to pore-external residues in water channels. • Coordination of Hg2+ ions to C155 and various residues at monomer-monomer interface. • Altered hydrogen bonding between C155 and S159 indirectly affects the M160. • Conformational changes of the pore-internal residue M160 increase the pore radius. Radio frequency (RF) can penetrate most packaging films and has the advantages of pasteurizing prepackaged low-moisture foods and avoiding secondary contamination. The suitable films for prepacking chili powders and the corresponding pasteurization process are unclear. This study aimed to select a suitable film for prepackaging chili powders, optimize the parameters of RF heating prepackaged chili powders, and evaluate the effects of RF-assisted pasteurization on the quality of chili powders. The results showed that the non-woven fabric (NWF) is suitable for prepackaging chili powders by evaluating the influence of RF heating on packaging films (appearance, sealing performance, mechanical properties.). Using NWF, chili powders inoculated with Salmonella enterica Enteritidis PT 30 still achieved 6.81 ± 0.64 log CFU/g reduction, treated by RF heating at an average temperature of 67.06 °C for 7.5 min with an electrode gap of 110 mm, held for 12.5 min at a hot-air convection oven. The pasteurization process had no significant (p > 0.05) effect on the quality (appearance, volatile, and capsaicin) of chili powders. The results indicated that chili powders packed with NWF could still be effectively pasteurized by RF-assisted hot air. An optical limiter was designed and fabricated. The device consists of an organic solution sandwiched between a polymer slab and a transparent relief polymer grating with a triangular groove. At low power the device has a high transmittance because the refractive index of the solution is matched with those of the slab and the grating materials and because the grating does not diffract. However, high power makes the organic solution thermally vaporize and makes the indices of the solution, slab, and grating materials become mismatched, which causes the grating to appear. The incident light is strongly absorbed, scattered, and self-defocused by the organic solution, and the grating suppresses the zero-order diffraction. Thus the transmitted light energy becomes lower than the damage threshold of human eyes or optical sensors. The device is an effective protection for human eyes or optical sensors against broadband pulsed-laser damage.

The recent outbreak of SARS-CoV-2 has had a profound effect on the world. Similar to that in SARS-CoV, the entry receptor of SARS-CoV-2 is ACE2. The binding of SARS-CoV-2 spike protein to ACE2 is the critical to the virus infection. Recently multiple species (human, Chinese chrysanthemum, Malay pangolin and cat) have been reported to be susceptible to the virus infection. However, the binding capacity and the detailed binding mechanism of SARS-CoV-2 spike protein to ACE2 of these species remains unexplored. Herein free energy calculations with MM-GBSA and Potential of Mean Forces together reveal that the Human-SARS-CoV-2 has a higher stability tendency than Human-SARS-CoV. Meanwhile, we uncover that SARS-CoV-2 has an enhanced ability to bind with the ACE2 in humans, pangolins and cats compared to that in bats. Analysis of key residues with energy decomposition and residue contact maps reveal several important consensus sites in ACE2s among the studied species, and determined the more favorable specified residues among the different types of amino acids. These results provide important implications for understanding SARS-CoV-2 host range which will make it possible to control the spread of the virus and use of animal models, targeted drug screening and vaccine candidates against SARS-CoV-2. • The SARS-CoV-2 showed stronger binding to intermediate host than to bat host. • Studied species has the same key active-site residues to association with RBD. • Several specific residue types in ACE2 are susceptible to SARS-CoV-2 infection. • MD results complement previous cryo-EM studies on the SARS-CoV-2 enzyme. Generalization of the concept of band topology from lower-dimensional to higher-dimensional (n > 3) physical systems is expected to introduce new bulk and boundary topological effects. However, theoretically predicted topological singularities in five-dimensional systems—Weyl surfaces and Yang monopoles—have either not been demonstrated in realistic physical systems or are limited to purely synthetic dimensions. We constructed a system possessing Yang monopoles and Weyl surfaces based on metamaterials with engineered electromagnetic properties, leading to the observation of several intriguing bulk and surface phenomena, such as linking of Weyl surfaces and surface Weyl arcs, via selected three-dimensional subspaces. The demonstrated photonic Weyl surfaces and Weyl arcs leverage the concept of higher-dimension topology to control the propagation of electromagnetic waves in artificially engineered photonic media. Spin‐controlled vortex generation and spin‐Hall effect, two distinct effects discovered in optics, have been extensively studied recently. However, while physical origins of two effects are both due to spin‐orbit interactions, their inherent connections remain obscure which also hinders further explorations on the manipulations of them. Here, in studying the scattering of a spin‐polarized light beam at sharp interfaces, an intriguing phase transition between vortex generation and spin‐Hall shift trigged by varying the incidence angle is revealed. After reflection/refraction, the beam contains two components: normal and abnormal modes acquiring spin‐redirection‐Berry phases and Pancharatnam–Berry phases, respectively. Inside the abnormal beam, two classes of wave components gain Pancharatnam–Berry phases with distinct topological natures, generating intrinsic and extrinsic orbital angular momenta (OAM), respectively. Enlarging incidence angle changes the relative portions of these two contributions, making the abnormal beam undergo a phase transition from vortex generation to spin‐Hall shift. Such intriguing effect is experimentally observed at a purposely designed metamaterial slab, exhibiting efficiency enhanced by several‐thousand times compared to that at a conventional slab. These findings unify two previously discovered effects in a single framework, reinterpret previous results with clearer pictures, and shed light on understanding other physical effects involving the competition between intrinsic and extrinsic OAM. Recently, 3D photonic Weyl degeneracies have been realized in both magnetized plasma (MP) and chiral hyperbolic metamaterials (CHM). Both systems can achieve backscattering‐immune transportation of light across sharp corners of translation‐invariant prisms and they also show similar equal‐frequency surfaces (EFSs). Despite this resemblance, their topological properties show striking differences as the Weyl points in MP and CHM arise from broken time‐reversal and spatial‐inversion symmetries, respectively. Here, using the effective medium approach, an interface formed between an MP and a CHM is investigated, which supports two Fermi arc surface states with nearly identical propagation directions. This leads to novel 3D interface configurations for guiding non‐reciprocal surface waves, information multiplexing, and magnetically controllable routing of the signal. In recent years, penetrating weapons have been used more and more to attack increasingly hard targets; therefore, the impact of such a penetrating process has increased to an extremely high level. As an important component of a fuze, the reliability of the ceramic capacitor in high-impact environments is key for the normal working of the fuze. In this paper, we found that a high-impact causes parameter drift of the multilayer ceramic capacitor (MLCC), which further causes the fuze to misfire. This paper mainly studies the internal mechanism of the MLCC's parameter drift during high impact. Firstly, transient physical phenomena, such as capacitance fluctuation and the leakage current increase of the ceramic capacitor under a high acceleration impact, were studied experimentally by a Machete hammer, revealing the relationship between the capacitance change, leakage current change, and acceleration under different working conditions. Secondly, a mechanical model of the ceramic capacitor is established to simulate the change in capacitance value, which shows that the main factor of the capacitance change is the deformation-derived change in the facing area between the electrodes. Lastly, an equivalent circuit model is established to simulate the change in the leakage current, which shows that the main factor of the leakage current change is the piezoelectric resistance of the ceramic dielectric.
