Introduction: Odontogenic inflammation is a fairly common condition, and depends on the quality of oral health. Unfortunately, the incidence in the Czech population is increasing. It is also sometimes connected with life-threatening complications. Aim: To describe an unusual method of treating phlegmonous inflammation of the mouth floor and neck, complicated by necrotizing fasciitis.Material and Method, Results: The authors demonstrate a strategy of treating odentogennic phlegmonous inflammation of the mouth floor in a patient with a blood coagulation disorder and advanced liver cirrhosis. The course of treatment was complicated by a massive gastric ulcer bleeding and due to the large necrosis of the soft tissue of the neck. Application of local negative pressure was used successfully. Conclusion: The local application of negative pressure in the surgical treatment of necrotizing fasciitis in the head and neck region is not very common. Recently, a number of authors have described the feasibility of this procedure in this anatomical location. The main advantages are, among others, shorter and less costly treatment and reduced stress for medical staff.

While the importance of anaerobic methane oxidation has been reported for marine ecosystems, the role of this process in soils is still questionable. Grasslands used as pastures for cattle overwintering show an increase in anaerobic soil micro-sites caused by animal treading and excrement deposition. Therefore, anaerobic potential methane oxidation activity of severely impacted soil from a cattle winter pasture was investigated in an incubation experiment under anaerobic conditions using 13C-labelled methane. We were able to detect a high microbial activity utilizing CH4 as nutrient source shown by the respiration of 13CO2. Measurements of possible terminal electron acceptors for anaerobic oxidation of methane were carried out. Soil sulfate concentrations were too low to explain the oxidation of the amount of methane added, but enough nitrate and iron(III) were detected. However, only nitrate was consumed during the experiment. 13C-PLFA analyses clearly showed the utilization of CH4 as nutrient source mainly by organisms harbouring 16:1ω7 PLFAs. These lipids were also found as most 13C-enriched fatty acids by Raghoebarsing et al. (2006) after addition of 13CH4 to an enrichment culture coupling denitrification of nitrate to anaerobic oxidation of methane. This might be an indication for anaerobic oxidation of methane by relatives of "Candidatus Methylomirabilis oxyfera" in the investigated grassland soil under the conditions of the incubation experiment.

Short-term diurnal changes in emissions of CO2 and N2O were determined in a cattle overwintering area during three specific periods of the year. Production of both N2O and CO2, as determined with gas chambers buried in soil and spatially distributed changed rapidly, and the general course of fluxes of the two gases was different. CO2 emissions were basically controlled by temperature, and most gas chambers showed the same trends in CO2 flux, indicating low spatial heterogeneity. In contrast, N2O emissions were much more spatially heterogeneous and each chamber had its own time course of emission; therefore, the relationship between flux and temperature was more complicated for N2O than CO2. For estimating gas emissions over long periods, we strongly recommend the use of frequent emission measurements during periods of high gas fluxes.

This paper presents a work in progress on experiments with embedded operating systems for the MicroBlaze processor. Modern embedded systems based on a configurable platform incorporating a similar processor core are gaining importance with the ongoing effort to minimize cost and development time. After an overview of the configurable platform based on this processor core, we devote our attention to uClinux OS. This OS has been successfully ported for the MicroBlaze processor, and we present our current experience with it. At the end of the paper we discuss several possible booting strategies and recommend further development of U-BOOT.

This paper deals with the distance estimation issue in 802.15.4 wireless sensor networks. On a basis of signal strength of received frames a distance between two sensor nodes using the log-normal shadowing radio propagation model (LNSM) is estimated. Basic problems of signal strength based systems are variation of RSSI parameter and correct calibration of coefficients for the LNSM model. For distance estimation in wireless networks, static and dynamic methods of calibration were already introduced. Static calibration has significant drawback in adaptation to the dynamical environment changes. Proposed work deals with an experimental validation of dynamic calibration method for distance estimation in wireless sensor networks. This method for dynamic estimation of radio environment parameters results in the significant improvements in the distance estimation accuracy in comparison with the known static methods.

All known localization techniques for wireless sensor and ad-hoc networks require certain set of reference nodes being used for position estimation. The anchor-free techniques in contrast to anchor-based do not require reference nodes called anchors to be placed in the network area before localization operation itself, but they can establish own reference coordinate system to be used for the relative position estimation. We observed that contemporary anchor-free localization algorithms achieve a low localization error, but dissipate significant energy reserves during the recognition of reference nodes used for the position estimation. Therefore, we have proposed the optimized anchor-free localization algorithm referred to as BRL (Boundary Recognition aided Localization), which achieves a low localization error and mainly reduces the communication cost of the reference nodes recognition phase. The proposed BRL algorithm was investigated throughout the extensive simulations on the database of networks with the different number of nodes and densities and was compared in terms of communication cost and localization error with the known related algorithms such as AFL and CRP. Through the extensive simulations we have observed network conditions where novel BRL algorithm excels in comparison with the state of art.

This paper presents the investigation of energy demands during localization of wireless nodes in ad-hoc networks. We focus on the method based on the received signal strength (RSS) to estimate the distances between the nodes. To deal with the uncertainty of this technique, statistical methods are used. It implies more measurement samples to be taken and consequently more energy to be spent. Therefore, we investigate the accuracy of localization and the consumed energy in the relation to the number of measurement samples. The experimental measurements were conducted with IRIS sensor motes and their results related to the proposed energy model. The results show that the expended energy is not related linearly to the localization error. First, improvement of the accuracy rises fast with more measurement samples. Then, adding more samples, the accuracy increase is moderate, which means that the marginal energy cost of the additional improvement is higher.

A numerical 0D kinetic model is developed here to simulate the detailed kinetics of the ground and excited electronic states of atmospheric gases occurring under streamer discharge conditions. The model is based on an extended kinetic scheme that involves state-to-state vibrational kinetics of the ground electronic states of N2, O2 and NO diatomics, including e–V, V–V and V–T energy transfers, and contains about 104 processes for more than 500 tracked state-specific states (including the higher excited and autoionising states of NI/OI atomic species), and also for state-non-specific species. This scheme takes into account the most important radiative processes occurring in N2 and N2–O2 mixtures that are of diagnostic interest, including the extreme ultraviolet and vacuum ultraviolet radiation produced by many excited and autoionising states of NI/OI. The dependence of the rates of electron-impact processes on the reduced electric field E/N is found by solving the Boltzmann equation for electrons in the two-term approximation using the Boltzmann equation solver Open Source library, BOLOS. We examine several test cases: pure nitrogen at 50 and 200 Torr and synthetic air at 10, 50 and 760 Torr, at a gas temperature of 300 K. The numerical results for pure nitrogen were compared with experimental results recently obtained by laser induced fluorescence for metastables. Good qualitative agreement was observed between numerical simulations and experiments at vibronic levels v  =  2–5, giving important feedback on the appropriateness of the kinetic scheme. Time-resolved microscopic ICCD images, emission waveforms and spectra of triggered single-surface coplanar dielectric barrier micro-discharges were acquired with nanosecond time resolution. The micro-discharges were produced in pure argon by applying periodic high-voltage waveforms to a pair of embedded metallic electrodes at atmospheric pressure, and microscopic images of single micro-discharge events complemented with spectrally resolved emission were acquired with a time resolution of a few nanoseconds. Due to the low jitter of the micro-discharge onset with respect to the high-voltage pulse, we succeeded in separating the very weak emission produced during the course of streamer formation from the emission produced during the transition between the streamer and the subsequent transient glow phase. We identified the characteristic spectrometric signatures of the transition between the streamer and glow phases by analysing the lines belonging to the Ar(3s23p54p → 3s23p54s) multiplet. By linking optical and electrical measurements, we estimated the electron density with a high temporal resolution. The electron densities of the streamer discharge, determined spectroscopically, were in the range of 1019–1020 m−3, while an estimate of 1021 m−3 was obtained using macroscopic electrical analysis combined with ICCD imaging. The evolution of selected population ratios of 2p levels during streamer evolution was determined for the first time, and this may be used in the future for advanced streamer diagnostics. The formation of N2(A3 ) metastable species, produced by cathode-directed streamer discharge, was investigated using the technique of laser-induced fluorescence. A triggered single streamer filament was periodically produced in pure nitrogen (and in nitrogen with admixtures of oxygen) at total pressure of 50 Torr and metastable species were monitored during the streamer channel decay in the centre of the discharge gap. We revealed the dynamics of individual vibrational (v  =  0–8) levels of N2(A3 ) for various oxygen admixtures (0–20%). In pure nitrogen, the observed evolution of the N2(A3 ) during the decaying streamer channel is evidence of initial vibrational relaxation of high vibrational levels towards the v  =  2 and 3 levels, followed by a delayed increase of terminal (v  =  0, 1) levels. A calibration procedure based on the rate of energy-pooling processes was used to place all detected vibronic levels in pure nitrogen on the absolute scale. Population maxima exceeding 1  ×  1014 cm−3 were fixed for the v  =  2 and 3 vibrational levels, while the lowest v  =  0 level reaches only 3  ×  1013 cm−3. Populations of v  =  2–5 vibrational levels were also estimated for N2  +  O2 mixtures after scaling of laser-induced fluorescence signals obtained at various oxygen admixtures. The total N2(A3 ) population in an air-like mixture is formed mainly by v  =  3–4 vibronic levels with the population maximum of ~3  ×  1013 cm−3 fixed at the shortest analyzed delay. This observation, together with the fact that we were unable to detect v  =  0 and 1 levels (fluorescence signals below detection threshold), gives a strong evidence of the inhibition of Δv  =  2 vibrational relaxation towards terminal v  =  0 and 1 levels, causing much lower populations of the lowest v  =  0–1 levels. By analyzing data obtained in pure nitrogen and in nitrogen with three different oxygen admixtures, we have estimated the quenching rate constants of N2(A3 , v)  +  O2 and N2(A3 , v)  +  N2 processes for v  =  2–6 vibrational levels which are consistent with data from the literature. Quenching rate constants for nitrogen reflect the N2(A3 , v)  →  N2(A3 , v  −  2) relaxation process, however the v  =  2 rate constant shows a much larger value compared with the published data. Many studies proved that non-equilibrium discharges generated at atmospheric pressure are highly effective for the bio-decontamination of surfaces of various materials. One of the key processes that leads to a desired result is plasma etching and thus the evaluation of etching rates of organic materials is of high importance. However, the comparison of reported results is rather difficult if impossible as different authors use diverse sources of atmospheric plasma that are operated at significantly different operational parameters. Therefore, we report here on the systematic study of the etching of nine different common polymers that mimic the different structures of more complicated biological systems, bovine serum albumin (BSA) selected as the model protein and spores of Bacillus subtilis taken as a representative of highly resistant micro-organisms. The treatment of these materials was performed by means of atmospheric pressure dielectric barrier discharge (DBD) sustained in open air at constant conditions. All tested polymers, BSA and spores, were readily etched by DBD plasma. However, the measured etching rates were found to be dependent on the chemical structure of treated materials, namely on the presence of oxygen in the structure of polymers. [ABSTRACT FROM AUTHOR]

The initial stages of transient luminous events (TLEs) occurring in the upper atmosphere of the Earth are, in a certain pressure range, controlled by the streamer mechanism. This paper presents the results of the first laboratory experiments to study the TLE streamer phenomena under conditions close to those of the upper atmosphere. Spectrally and highly spatiotemporally resolved emissions originating from radiative states (second positive system) and (first negative system) have been recorded from the positive streamer discharge. Periodic ionizing events were generated in a barrier discharge arrangement at a pressure of 4 torr of synthetic air, i.e. simulating the pressure conditions at altitudes of ≃37 km. Employing Abel inversion on the radially scanned streamer emission and a 2D fitting procedure, access was obtained to the local spectral signatures within the over 106  m s−1 fast propagating streamers. The reduced electric field strength distribution within the streamer head was determined from the ratio of the band intensities with peak values up to 500 Td and overall duration of about 10 ns. The 2D profiles of the streamer head electric fields were used as an experimentally obtained input for kinetic simulations of the streamer-induced air plasma chemistry. The radial and temporal computed distribution of the ground vibrational levels of the radiative states involved in the radiative transitions analyzed (337.1 nm and 391.5 nm), atomic oxygen, nitrogen, nitric oxide and ozone concentrations are vizualized and discussed in comparison with available models of the streamer phase of Blue Jet discharges in the stratosphere. Introduction and aim: Dental models are widely used in dentistry. The transmission of oral cavity situation outside patient's mouth brings us information in the field of diagnostics, treatment planning, and the fabrication planning of dental products. Dental models can be used in any dental field. They are particularly linked to prosthodontics, orthodontics, and maxillofacial surgery. The aim of this article is to report the benefits of the intraoral scanning in conjuction with 3D printing to the reader. Also, it describes their basic principles and presents the most useful technologies of 3D printing for production in dentistry according to the current literature. Materials and methods: The literature search and survey were focused on intraoral scanning and 3D printing. PubMed, Scopus, and Ebsco databases were used to find the articles. Their applicability in dentistry, the inclusion of a control group, and the age of the article within five years were essential for their subsequent selection. Conclusion: The included studies show that the technologies of direct intraoral scanning and 3D printing are already clinically usable today, and in the future we can expect their further development for everyday practice.

The evolution of individual v = 0–10 vibrational levels of N2(A3) metastable species produced by filamentary streamer discharge was investigated by the laser-induced fluorescence technique. Triggered single streamer filament was periodically produced in pure nitrogen at a pressure of 200 torr and metastable species were monitored during the streamer channel decay in the centre of the discharge gap. The observed dynamics of N2(A3) vibrational levels follow two very different scenarios: while higher (v  >  6) vibronic levels decay exponentially in hundreds of nanoseconds, the populations of lower levels (v  ≤  6) definitely increase, first reaching a local maximum on a microsecond timescale and then decreasing afterwards. Population maxima of N2(A3, v  ≤  6) levels occur after the streamer onset with a certain delay, which decreases with increasing vibrational number. Interpretation of experimental observation based on a 0D kinetic model of the post-discharge period takes into account the most important processes redistributing populations between the N2(A3), N2 and N2 vibronic levels. The model reproduces experimental observations fairly well, including observed maxima delays occurring due to the collisional cascade, which transfers metastable species from higher even/odd vibrational levels towards v = 0/v = 1 terminal levels through the Δv = 2 vibrational relaxation mechanism. A calibration procedure based on the rate of energy-pooling processes was used to determine absolute populations of the v = 0 and 1 levels from LIF data, and the model results were utilized to place on an absolute scale all the higher (v  >  1) measured vibronic levels. Vibrational distributions obtained from calibrated LIF data at selected instants show a reasonable qualitative agreement with model predictions. Population maxima exceeding 3   ×   1014 cm−3 were fixed for v = 2 and 3 vibrational levels, while the lowest v = 0 level reaches only 8–9   ×   1013 cm−3. Lastly, we show that the observed rate of the v = 2 level decay is not compatible with published rate constants for the v = 2  →  v = 0 vibrational relaxation. [ABSTRACT FROM AUTHOR]

This paper reviews optical diagnostic methods and approaches applied to study the fundamentals of streamer discharges, considering the peculiarities of streamers developing in atmospheric gases at high (1 bar) as well as low (<10 mbar) pressures. A critical discussion is devoted to the cross-sections for electron-impact excitation/ionization/dissociation processes and corresponding rate constants in relation to methods used to probe streamer properties. The most important spectrometric signatures of radiative transitions of diatomic as well as atomic species are discussed on the basis of their synthetic models with a brief guide on how to simulate the most important emissions. Basic differences between UV–vis–NIR spectra produced by electron-impact and various heavy-particle energy-transfer processes during streamer evolution are presented and possible strategies based on 2D projections of cylindrically symmetric streamers to determine radial distributions of excited species within the streamer channel are discussed. The use of emission techniques to obtain the rotational temperatures and vibrational distributions of excited states of diatomics and laser-induced fluorescence techniques to probe the vibrational manifold of the lowest triplet metastable state of the nitrogen molecule is addressed. The homolytic cleavage of C-C bonds adjacent to specific functional groups has lately emerged as a versatile approach for molecular diversification. Despite the ubiquity and synthetic utility of ketones, radical fragmentation of their α-C-C bonds has proven to be a formidable challenge. Here, we present a broadly applicable deacylative strategy designed to homolytically cleave aliphatic ketones of various complexities, including transformations of cycloalkanones into carboxylic acids tethered to C-centered free radicals that can be engaged in diverse radical-based processes. The method involves ketone activation through treatment with hydrogen peroxide, yielding gem -dihydroperoxides. Subsequent single-electron-transfer reduction mediated by a low-valent metal complex generates alkyl radicals that can be captured selectively with a radicophile of choice, including through catalytic cross-coupling. The logic of our deacylative functionalization is exemplified by the total synthesis of 14 natural products, one analogue, and two drugs starting from readily available natural products, showcasing its transformative power in complex settings. This approach obviates the need for complex reagents and allows the controlled conversion of ketones to reconstructed products, making the process highly applicable across a spectrum of domains.

Background: Speech dysfunction represents one of the initial motor manifestations to develop in Parkinson's disease (PD) and is measurable through smartphone., Objective: The aim was to develop a fully automated and noise-resistant smartphone-based system that can unobtrusively screen for prodromal parkinsonian speech disorder in subjects with isolated rapid eye movement sleep behavior disorder (iRBD) in a real-world scenario., Methods: This cross-sectional study assessed regular, everyday voice call data from individuals with iRBD compared to early PD patients and healthy controls via a developed smartphone application. The participants also performed an active, regular reading of a short passage on their smartphone. Smartphone data were continuously collected for up to 3 months after the standard in-person assessments at the clinic., Results: A total of 3525 calls that led to 5990 minutes of preprocessed speech were extracted from 72 participants, comprising 21 iRBD patients, 26 PD patients, and 25 controls. With a high area under the curve of 0.85 between iRBD patients and controls, the combination of passive and active smartphone data provided a comparable or even more sensitive evaluation than laboratory examination using a high-quality microphone. The most sensitive features to induce prodromal neurodegeneration in iRBD included imprecise vowel articulation during phone calls (P = 0.03) and monopitch in reading (P = 0.05). Eighteen minutes of speech corresponding to approximately nine calls was sufficient to obtain the best sensitivity for the screening., Conclusion: We consider the developed tool widely applicable to deep longitudinal digital phenotyping data with future applications in neuroprotective trials, deep brain stimulation optimization, neuropsychiatry, speech therapy, population screening, and beyond. © 2024 The Author(s). Hyaluronan, a linear glycosaminoglycan comprising D-N-acetylglucosamine and D-glucuronic acid, is the main component of the extracellular matrix. Its influence on cell proliferation, migration, inflammation, signalling, and other functions, depends heavily on its molecular weight and chemical modification. Unsaturated HA oligosaccharides are available in defined length and purity. Their potential therapeutic utility can be further improved by chemical modification, e. g., reduction. No synthesis of such modified oligosaccharides, either stepwise or by hyaluronan cleavage, has been reported yet. Here we show a three-step synthesis (esterification, depolymerization and reduction) of unsaturated even numbered hyaluronan oligosaccharides with carboxylates and the reducing terminus reduced to an alcohol. Particular oligosaccharides were synthesised. The modified oligosaccharides are not cleaved by mammalian or bacterial hyaluronidase and do not affect the growth of mouse and human fibroblasts. Further, MTT and NRU viability tests showed that they inhibit the growth of human colon carcinoma cells HT-29 by 20-50 % in concentrations 500-1000 μg/mL. Interestingly, this effect takes place regardless of CD44 receptor expression and was not observed with unmodified HA oligosaccharides. This study investigates the impact of plasma-seed interaction on germination and early plant development, focusing on Arabidopsis thaliana and Brassica napus . The investigation delves into changes in chemical composition, water absorption, and surface morphology induced by plasma filaments generated in synthetic air. These analyses were conducted using scanning electron microscopy (SEM) and X-ray photoelectron spectroscopy (XPS). Although plasma treatment enhanced water absorption and modified surface chemistry, its impact on germination demonstrated species- and context-dependent variations. Notably, the accelerated germination and morphogenesis of seedlings in microbiome-enriched (MB+) soil could be achieved also in microbiome-deprived (MB-) soil by short-term plasma treatment of seeds. Remarkably, the positive effects of plasma treatment on early developmental events (germination, morphogenesis) and later events (formation of inflorescences) were more pronounced in the context of MB- soil but were accompanied by a slight decrease in disease resistance, which was not detected in MB+ soil. The results underscore the intricate dynamics of plasma-plant interactions and stress the significance of accounting for the soil microbiome while designing experiments with potential field application.

Hyaluronan (HA) has been recently identified as a key component of the densification of thoracolumbar fascia (TLF), a potential contributor to non-specific lower back pain (LBP) currently treated with manual therapy and systemic or local delivery of anti-inflammatory drugs. The aim of this study was to establish a novel animal model suitable for studying ultrasound-guided intrafascial injection prepared from HA with low and high Mw. Effects of these preparations on the profibrotic switch and mechanical properties of TLF were measured by qPCR and rheology, respectively, while their lubricating properties were evaluated by tribology. Rabbit proved to be a suitable model of TLF physiology due to its manageable size enabling both TLF extraction and in situ intrafascial injection. Surprisingly, the tribology showed that low Mw HA was a better lubricant than the high Mw HA. It was also better suited for intrafascial injection due to its lower injection force and ability to freely spread between TLF layers. No profibrotic effects of either HA preparation in the TLF were observed. Extracorporeal membrane oxygenation is the highest form of resuscitation care in management of patients with respiratory failure. In the case of acute respiratory distress syndrome, the veno-venous setting is more often used. ECMO support enables, in case of lung function failure, to obtain the necessary time for the onset of the causal treatment effect or is used as a bridge to transplantation Mortality of the patients varies according to the underlying cause and presence of risk factors (e.g., age, complications or comorbid diseases). The onset of the COVID-19 pandemic has led to a significant increase in the need for ECMO. The quality of life of patients after ECMO is significantly reduced, but most patients do not experience permanent disability.

Popularity of hyaluronan (HA) in the cosmetics and pharmaceutical industries, led to the investigation and development of new HA-based materials, with enzymes playing a key role. Beta-D-glucuronidases catalyze the hydrolysis of a beta-D-glucuronic acid residue from the non-reducing end of various substrates. However, lack of specificity towards HA for most beta-D-glucuronidases, in addition to the high cost and low purity of those active on HA, have prevented their widespread application. In this study, we investigated a recombinant beta-glucuronidase from Bacteroides fragilis (rBfGUS). We demonstrated the rBfGUS's activity on native, modified, and derivatized HA oligosaccharides (oHAs). Using chromogenic beta-glucuronidase substrate and oHAs, we characterized the enzyme's optimal conditions and kinetic parameters. Additionally, we evaluated rBfGUS's activity towards oHAs of various sizes and types. To increase reusability and ensure the preparation of enzyme-free oHA products, rBfGUS was immobilized on two types of magnetic macroporous bead cellulose particles. Both immobilized forms of rBfGUS demonstrated suitable operational and storage stabilities, and their activity parameters were comparable to the free form. Exposure to the sun affects the skin and may eventually result in UV-induced skin damage. It is generally known that hyaluronan (HA) is one of the main structural and functional components of the skin. However, UV-related changes in the HA metabolism in the skin have not yet been elucidated. Using qRT-PCR, confocal microscopy and LC-MS/MS we compared the naturally sun-exposed (SE), sun-protected, experimentally repeatedly UVA + UVB-exposed and acutely (once) UVA + UVB irradiated skin of Caucasian women. The epidermis was harvested by means of suction blistering 24 h after the acute irradiation. In addition, the epidermis was compared with a UV-irradiated in vitro reconstituted 3D epidermis (EpiDerm) and an in vitro 2D culture of normal human keratinocytes (NHEK). The amount of HA was found to be statistically significantly enhanced in the acutely irradiated epidermis. The acute UV evinced the upregulation of HA synthases (HAS2 and HAS3), hyaluronidases (HYAL2 and HYAL3), Cluster of differentiation 44 (CD44), and Cell Migration Inducing Proteins (CEMIP and CEMIP2), while only certain changes were recapitulated in the 3D epidermis. For the first time, we demonstrated the enhanced gene and protein expression of CEMIP and CEMIP2 following UV irradiation in the human epidermis. The ability of hyaluronan as a dietary supplement to increase skin moisture and relieve knee pain has been demonstrated in several clinical studies. To understand the mechanism of action, determining hyaluronan's bioavailability and in vivo fate is crucial. Here, we used 13 C-hyaluronan combined with LC-MS analysis to compare the absorption and metabolism of oral hyaluronan in germ-free and conventional wild-type mice. The presence of Bacteroides spp. in the gut was crucial for hyaluronan absorption. Specific microorganisms cleave hyaluronan into unsaturated oligosaccharides (<3 kDa) which are partially absorbed through the intestinal wall. The remaining hyaluronan fragments are metabolized into short-chain fatty acids, which are only metabolites available to the host. The poor bioavailability (~0.2 %) of oral hyaluronan indicates that the mechanism of action is the result of the systematic regulatory function of hyaluronan or its metabolites rather than the direct effects of hyaluronan at distal sites of action (skin, joints)., Competing Interests: Declaration of competing interest The authors declare no competing interests., (Copyright © 2023 Elsevier Ltd. A cascade of reactions known as the foreign body response (FBR) follows the implantation of biomaterials leading to the formation of a fibrotic capsule around the implant and subsequent health complications. The severity of the FBR is driven mostly by the physicochemical characteristics of implanted material, the method and place of implantation, and the degree of immune system activation. Here we present an in vitro model for assessing new materials with respect to their potential to induce a FBR in the peritoneum. The model is based on evaluating protein sorption and cell adhesion on the implanted material. We tested our model on the free-standing films prepared from hyaluronan derivatives with different hydrophobicity, swelling ratio, and rate of solubilization. The proteomic analysis of films incubated in the mouse peritoneum showed that the presence of fibrinogen was driving the cell adhesion. Neither the film surface hydrophobicity/hydrophilicity nor the quantity of adsorbed proteins were decisive for the induction of the long-term cell adhesion leading to the FBR, while the dissolution rate of the material proved to be a crucial factor. In this paper, the growth requirements, fermentation pattern, and hydrolytic enzymatic activities of anaerobic ciliates collected from the hindgut of the African tropical millipede Archispirostreptus gigas are described. Single-cell molecular analysis showed that ciliates from the millipede hindgut could be assigned to the Nyctotherus velox and a new species named N. archispirostreptae n. sp. The ciliate N. velox can grow in vitro with unspecified prokaryotic populations and various plant polysaccharides (rice starch-RS, xylan, crystalline cellulose20-CC, carboxymethylcellulose-CMC, and inulin) or without polysaccharides (NoPOS) in complex reduced medium with soluble supplements (peptone, glucose, and vitamins). Specific catalytic activity (nkat/g of protein) of α amylase of 300, xylanase of 290, carboxymethylcellulase of 190, and inulinase of 170 was present in the crude protein extract of N. velox . The highest in vitro dry matter digestibility was observed in RS and inulin after 96 h of fermentation. The highest methane concentration was observed in xylan and inulin substrates. The highest short-chain fatty acid concentration was observed in RS, inulin, and xylan. In contrast, the highest ammonia concentration was observed in NoPOS, CMC, and CC. The results indicate that starch is the preferred substrate of the N. velox . Hydrolytic enzyme activities of N. velox showed that the ciliates contribute to the fermentation of plant polysaccharides in the gut of millipedes.

Introduction: Severe respiratory failure is one of the most serious complications of coronavirus disease 2019 (COVID-19). In a small proportion of patients, mechanical ventilation fails to provide adequate oxygenation and extracorporeal membrane oxygenation (ECMO) is needed. The surviving individuals need long-term follow-up as it is not clear what their prognosis is., Aim: To provide a complex clinical picture of patients during follow-up exceeding one year after the ECMO therapy due to severe COVID-19., Methods: All subjects involved in the study required ECMO in the acute stage of COVID-19. The survivors were followed-up for over one year at a specialized respiratory medical center., Results: Of the 41 patients indicated for ECMO, 17 patients (64.7% males) survived. The average age of survivors was 47.8 years, and the average BMI was 34.7 kg·m -2 . The duration of ECMO support was 9.4 days. A mild decrease in vital capacity (VC) and transfer factor (DLCO) was observed on the initial follow-up visit (82.1% and 60%, respectively). VC improved by 6.2% and by an additional 7.5% after 6 months and 1 year, respectively. DLCO improved by 21.1% after 6 months and remained stable after 1 year. Post-intensive care consequences included psychological problems and neurological impairment in 29% of patients; 64.7% of the survivors got vaccinated against SARS-CoV-2 within 12 months of hospitalization and 17.6% experienced reinfection with a mild course., Conclusion: The COVID-19 pandemic has significantly increased the need for ECMO. Patients' quality of life after ECMO is temporarily significantly reduced but most patients do not experience permanent disability.

Hyaluronan is being investigated extensively as a biocompatible and biodegradable material for use in biomedical applications. While the derivatization of hyaluronan broadens its potential therapeutic use, the pharmacokinetics and metabolization of the derivatives must be thoroughly investigated. The fate of intraperitoneally-applied native and lauroyl-modified hyaluronan films with varying degrees of substitution was investigated in-vivo employing an exclusive stable isotope-labelling approach and LC-MS analysis. The materials were gradually degraded in peritoneal fluid, lymphatically absorbed, preferentially metabolized in the liver and eliminated without any observable accumulation in the body. Hyaluronan acylation prolongs its presence in the peritoneal cavity depending on the degree of substitution. The safety of acylated hyaluronan derivatives was confirmed via a metabolic study that revealed its degradation into non-toxic metabolites, i.e. native hyaluronan and free fatty acid. COVID-19 pandemic has massive impact on the whole world, lead to the death of milions of people and required excessive restrictions with large economical, psychological and other impacts. Post-COVID syndrome is defined according to the Czech pneumological and phtizeological society as a complex of symptoms persisting for more than 12 weeks from the acute stage of COVID-19 and which is not possible to explain by other causes. Exact pathopysiological mechanisms and its frequency are still not known, and the data from the literature are not consisent. We present case reports of 2 patients with very different disease and convalescence course. COVID-19 and post-COVID syndrome represent large space for the further research. The prediction of the persisting limitations and other consequencies of the diseases is still not possible. Even though there is usualy higher incidence of post-COVID symptoms in patients surviving severe COVID-19 course, the presented case reports show paradoxically exactly the oposite situation.

Background: Patients with synucleinopathies frequently display language abnormalities. However, whether patients with isolated rapid eye movement sleep behavior disorder (iRBD) have prodromal language impairment remains unknown., Objective: We examined whether the linguistic abnormalities in iRBD can serve as potential biomarkers for conversion to synucleinopathy, including the possible effect of mild cognitive impairment (MCI), speaking task, and automation of analysis procedure., Methods: We enrolled 139 Czech native participants, including 40 iRBD without MCI and 14 iRBD with MCI, compared with 40 PD without MCI, 15 PD with MCI, and 30 healthy control subjects. Spontaneous discourse and story-tale narrative were transcribed and linguistically annotated. A quantitative analysis was performed computing three linguistic features. Human annotations were compared with fully automated annotations., Results: Compared with control subjects, patients with iRBD showed poorer content density, reflecting the reduction of content words and modifiers. Both PD and iRBD subgroups with MCI manifested less occurrence of unique words and a higher number of n-grams repetitions, indicating poorer lexical richness. The spontaneous discourse task demonstrated language impairment in iRBD without MCI with an area under the curve of 0.72, while the story-tale narrative task better reflected the presence of MCI, discriminating both PD and iRBD subgroups with MCI from control subjects with an area under the curve of up to 0.81. A unified approach to meroterpenoids applanatumols B, V, W, X, and Y produced by the medicinal fungus Ganoderma applanatum and 2'- epi -spiroapplanatumine O is presented. The key synthetic sequence consists of a tandem anionic ketone allylation/oxy-Cope rearrangement/α-oxygenation furnishing an α-aminoxy ketone and a persistent radical effect-based 5- exo -trig cyclization leading to the trisubstituted cyclopentane core. The relative configuration of applanatumol V has to be revised. Some compounds display significant cytotoxic and antioxidant properties.

Hyaluronan (HA) comprises a fundamental component of the extracellular matrix and participates in a variety of biological processes. Half of the total amount of HA in the human body is present in the skin. HA exhibits a dynamic turnover; its half-life in the skin is less than one day. Nevertheless, the specific participants in the catabolism of HA in the skin have not yet been described in detail, despite the essential role of HA in cutaneous biology. A deeper knowledge of the processes involved will act to support the development of HA-based topical and implantable materials and enhance the understanding of the various related pathological cutaneous conditions. This study aimed to characterize the distribution and activity of hyaluronidases and the other proteins involved in the degradation of HA in healthy human full-thickness skin, the epidermis and the dermis. Hyaluronidase activity was detected for the first time in healthy human skin. The degradation of HA occurred in lysates at an acidic pH. HA gel zymography revealed a single band corresponding to approximately 50 kDa. This study provided the first comprehensive view of the distribution of canonic HA-degrading proteins (HYAL1 and HYAL2) in human skin employing IHF and IHC. Furthermore, contrary to previous assumptions TMEM2, a novel hyaluronidase, as well as CEMIP, a protein involved in HA degradation, were localized in the human epidermis, as well as in the dermis.

Although the cepstral peak prominence (CPP) and its variant, the cepstral peak prominence smooth (CPPS), are considered to be robust acoustic measures for the evaluation of dysphonia, whether they are sensitive to capture early voice changes in Parkinson's disease (PD) has not yet been explored. This study aimed to investigate the voice changes via the CPP measures in the idiopathic rapid eye movement sleep behavior disorder (iRBD), a special case of prodromal neurodegeneration, and recently diagnosed and advanced-stage Parkinson's disease (AS-PD) patients using different speaking tasks across noise-free and noisy environments. The sustained vowel phonation, reading of passages, and monologues of 60 early stage untreated PD, 30 advanced-stage Parkinson's disease, 60 iRBD, and 60 healthy control (HC) participants were evaluated. Significant differences were found between the PD groups and controls in sustained phonation via the CPP (p < 0.05) and CPPS (p < 0.01) and the monologue via the CPP (p < 0.01), although neither the CPP nor CPPS measures were sufficiently sensitive to capture the possible prodromal dysphonia in the iRBD. The quality of the CPP and CPPS measures was influenced substantially by the addition of ambient noise. It was anticipated that the CPP measures might serve as a promising digital biomarker in assessing the dysphonia from the early stages of PD.

Methanogens represent the final decomposition step in anaerobic degradation of organic matter, occurring in the digestive tracts of various invertebrates. However, factors determining their community structure and activity in distinct gut sections are still debated. In this study, we focused on the tropical millipede species Archispirostreptus gigas (Diplopoda, Spirostreptidae) and Epibolus pulchripes (Diplopoda, Pachybolidae), which release considerable amounts of methane. We aimed to characterize relationships between physicochemical parameters, methane production rates, and methanogen community structure in the two major gut sections, midgut and hindgut. Microsensor measurements revealed that both sections were strictly anoxic, with reducing conditions prevailing in both millipedes. Hydrogen concentration peaked in the anterior hindgut of E. pulchripes . In both species, the intestinal pH was significantly higher in the hindgut than in the midgut. An accumulation of acetate and formate in the gut indicated bacterial fermentation activities in the digestive tracts of both species. Phylogenetic analysis of 16S rRNA genes showed a prevalence of Methanobrevibacter spp. ( Methanobacteriales ), accompanied by a small fraction of so-far-unclassified " Methanomethylophilaceae " ( Methanomassiliicoccales ), in both species, which suggests that methanogenesis is mostly hydrogenotrophic. We conclude that anoxic conditions, negative redox potential, and bacterial production of hydrogen and formate promote gut colonization by methanogens. The higher activities of methanogens in the hindgut are explained by the higher pH of this compartment and their association with ciliates, which are restricted to this compartment and present an additional source of methanogenic substrates. IMPORTANCE Methane (CH 4 ) is the second most important atmospheric greenhouse gas after CO 2 and is believed to account for 17% of global warming. Methanogens are a diverse group of archaea and can be found in various anoxic habitats, including digestive tracts of plant-feeding animals. Termites, cockroaches, the larvae of scarab beetles, and millipedes are the only arthropods known to host methanogens and emit large amounts of methane. Millipedes are ranked as the third most important detritivores after termites and earthworms, and they are considered keystone species in many terrestrial ecosystems. Both methane-producing and non-methane-emitting species of millipedes have been observed, but what limits their methanogenic potential is not known. In the present study, we show that physicochemical gut conditions and the distribution of symbiotic ciliates are important factors determining CH 4 emission in millipedes. We also found close similarities to other methane-emitting arthropods, which might be associated with their similar plant-feeding habits.

There is inconsistent information regarding the size effects of exogenously given hyaluronan on its in vivo fate. The data are often biased by the poor quality of hyaluronan and non-ideal labelling strategies used for resolving exogenous/endogenous hyaluronan, which only monitor the label and not hyaluronan itself. To overcome these drawbacks and establish the pharmacokinetics of intravenous hyaluronan in relation to its M w , 13 C-labelled HA of five M w s from 13.6-1562 kDa was prepared and administered to mice at doses 25-50 mg kg -1 . The elimination efficiency increased with decreasing M w . Low M w hyaluronan was rapidly eliminated as small hyaluronan fragments in urine, while high M w hyaluronan exhibited saturable kinetics and complete metabolization within 48 h. All tested M w s exhibited a similar uptake by liver cells and metabolization into activated sugars. 13 C-labelling combined with LC-MS provides an excellent approach to elucidating in vivo fate and biological activities of hyaluronan., (Copyright © 2021 Elsevier Ltd. Real-world data on regimens for relapsed/refractory multiple myeloma (RRMM) are limited. Daratumumab in combination with bortezomib and dexamethasone is a promising new treatment. The aim of this analysis was to assess the outcomes of daratumumab-bortezomib-dexamethasone (DVd) combination for the treatment of patients with RRMM in a real-world setting. All consecutive RRMM patients who received at least two cycles of DVd treatment between December 2016 and July 2020 were identified. We analyzed the clinical characteristics and survival of 47 patients treated at 7 Slovak centers outside of the clinical trials. The median age was 65 years (range, 35 to 83). The median (range) number of lines of therapy per patient was 3 (2-6). All patients were previously exposed to PIs (proteasome inhibitors) and IMIDs (immunomodulatory drugs), the majority of patients (70.2%) had double refractory (IMIDs and PI) disease and 72.3% of patients were refractory to their last therapy. Most patients presented with high-risk characteristics, including 25.6% adverse cytogenetics and 25.5% extramedullary disease. The majority of patients responded with an overall response rate of 78%, we found complete response in 3, very good partial response in 22, partial response in 12, minor response or stable disease in 9, and progressive disease in 1 patient. After a median follow-up period of 8 months, the median progression-free survival was 10 months. There was a longer progression-free survival in those with 2 vs. >2 prior treatments, with equally good effectivity in standard-risk and high-risk cytogenetic groups. The adverse events were usually mild, none leading to permanent drug interruptions. Daratumumab-bortezomib-based combinations are efficacious and safe regimens in RRMM patients in the real-world setting. This is the first analysis in Slovakia addressing the DVd combination outside of the clinical trial setting.

The aim of this paper is to review chromatographic and mass-spectrometric methods and underline the best analytical approaches for successful analysis of various hyaluronic acid species in different types of samples. Hyaluronan-degrading enzymes and chemical depolymerization produce di- or oligosaccharides suitable for hyaluronan quantification or structural characterization of hyaluronan derivatives. Efficient purification and pre-column derivatization of hyaluronan disaccharides by reductive amination allow subnanogram quantification in biological samples. The chromatographic separation is capable to distinguish all glycosaminoglycans disaccharides and to resolve hyaluronan fragments with 2-40 monomers. Using electrospray ionization or matrix assisted laser desorption ionization, hyaluronan fragments up to 8 kDa or 41 kDa, respectively, can be observed. One- or two-dimensional chromatographic separation with higly sensitive mass-spectrometric detection is an indispensable tool for revealing substituent position, extent of modification and substitution patterns of chemically modified hyaluronan derivatives. Fossorial mammals are supposed to face hypoxic and hypercapnic conditions, but such conditions have been rarely encountered in their natural burrow systems. Gas composition in burrows after heavy rains, deeper burrows and especially nest chambers, where animals usually spend most of the day, could be even more challenging than in shallow burrows. Such situations, however, have been rarely surveyed in the wild. In our study, we determined concentrations of O 2 , CO 2 and CH 4 in active burrows and nests of the giant root-rat Tachyoryctes macrocephalus, a large fossorial rodent endemic to the Afroalpine zone of the Bale Mountains in Ethiopia. We were able to determine the precise location of nests by tracking individuals equipped with radio-collars. To the best of our knowledge, this is the first study that analyses air samples taken directly from the nests of actually occupied burrow systems in any free-living fossorial mammal. We found no evidence for environmental hypoxia in the examined burrows and nests (range 19.7-21.6% O 2 ). Concentrations of CO 2 in the burrows increased after the burrows were plugged in the evening, but did not reach physiologically problematic levels. The highest CO 2 concentrations in burrows were detected in the evening during a wet period (up to 0.44%). In root-rat nest chambers, the highest (but still harmless) CO 2 concentrations (up to 1.31%) were detected in the morning (measured in the late dry season only) together with an elevated concentration (up to 13.5ppm) of CH 4 . Regular surface activity of giant root-rats, combined with the relatively large dimensions of their nest chambers and tunnels, and the absence of heavy soils, may contribute to harmless atmospheres within their burrow systems., (Copyright © 2020 Elsevier GmbH. Fungal diseases seriously affect agricultural production and the food industry. Crop protection is usually achieved by synthetic fungicides, therefore more sustainable and innovative technologies are increasingly required. The atmospheric pressure low-temperature plasma is a novel suitable measure. We report on the effect of plasma treatment on phytopathogenic fungi causing quantitative and qualitative losses of products both in the field and postharvest. We focus our attention on the in vitro direct inhibitory effect of non-contact Surface Dielectric Barrier Discharge on conidia germination of Botrytis cinerea, Monilinia fructicola, Aspergillus carbonarius and Alternaria alternata. A few minutes of treatment was required to completely inactivate the fungi on an artificial medium. Morphological analysis of spores by Scanning Electron Microscopy suggests that the main mechanism is plasma etching due to Reactive Oxygen Species or UV radiation. Spectroscopic analysis of plasma generated in humid air gives the hint that the rotational temperature of gas should not play a relevant role being very close to room temperature. In vivo experiments on artificially inoculated cherry fruits demonstrated that inactivation of fungal spores by the direct inhibitory effect of plasma extend their shelf life. Pre-treatment of fruits before inoculation improve the resistance to infections maybe by activating defense responses in plant tissues.

The effect of hydrazide linkers on the formation and mechanical properties of hyaluronan hydrogels was intensively evaluated. The reaction kinetics of hydrazone formation was monitored by NMR spectroscopy under physiological conditions where polyaldehyde hyaluronan (unsaturated: ΔHA-CHO, saturated: HA-CHO) was reacted with various hydrazides to form hydrogels. Linear (adipic, oxalic dihydrazide) and branched (N,N´,N´´-tris(hexanoylhydrazide-6-yl)phosphoric triamide and 4-arm-PEG hydrazide) hydrazides were compared as crosslinking agents. The mechanical properties of hydrogels were also modified by attaching a hydrophobic chain to HA-CHO; however, it was found that this modification did not lead to an increase in hydrogel stiffness. Cytotoxicity tests showed that all tested hydrazide crosslinkers reduced the viability of cells only slightly, and that the final hyaluronan hydrogels were non-toxic materials., (Copyright © 2019 Elsevier Ltd. A better understanding of in vivo behavior of nanocarriers is necessary for further improvement in their development. Here we present a novel approach, where both the matrix and the drug can be analyzed by LCMS/MS after one sample handling. The developed method was applied for the comparison of pharmacokinetic profile of free and encapsulated doxorubicin (DOX) in oleyl hyaluronan (HA-C18:1) polymeric micelles. The results indicated that nanocarriers were rapidly dissociated upon in vivo administration. Despite this fact, the administration of encapsulated DOX led to its longer circulation time and enhanced tumor targeting. This effect was not observed injecting blank HA-C18:1 micelles followed by unencapsulated DOX. Biodistribution studies and molecular weight estimation of the carrier matrix indicated relatively high stability of HA-C18:1 ester bond in bloodstream and complete elimination of the derivative within 72 h. The present study reports the organocatalytic enantioselective allylic amination of Morita-Baylis-Hillman carbamates efficiently catalyzed by a chiral amine in the presence of a Brønsted acid. Chiral allylic amines were produced in high yields (up to 98 %) and enantioselectivities (up to 97 % ee). Deep sternal wound infection is a feared complication of cardiac surgery due to the negative impact on mortality, morbidity and long-term survival. Its incidence has remained more or less unchanged over the last three decades despite the significant increase in patients´ morbidity and complexity of cardiac surgery. The review summaries strategies to reduce the incidence of deep sternal wound reflecting general surgical site infection prevention and specificities of surgery performed through the median sternotomy. Furthermore, contemporary evidence-based recommendations for prevention of this complication are highlighted in the review. Key words: sternal infection - prevention - cardiac surgery.

PURPOSE OF THE STUDY Outcome analysis of penetrating cardiac and great vessels injury within the 15-year existence of the cardiac surgery service as a part of the major trauma centre of the University Hospital Olomouc MATERIAL AND METHODS Retrospective analysis of a group of a total of 16 patients who underwent a surgery for penetrating cardiac and great vessels injury since II/2002 to XI/2016. The dominant causes of penetrating trauma were stab injuries (15 patients, 94%), in one patient only (6%) it was a gunshot injury. The mean age of the patients included in the group was 42.9 ± 16.1 years, with men significantly prevailing (13 patients, 81%). A total of 7 injured persons (44%) were haemodynamically stable when admitted, 9 injured persons (56%) were unstable or in critical condition. The average transfer distance was 48.8 ± 34.5 km; the injured were admitted on average 115.9 ± 154.8 minutes after being injured. Preoperatively, all the injured suffered from pericardial effusion (>5 mm) confirmed by TTE (81%) or CTA (19%). In 4 patients (25%) pericardial drainage for cardiac tamponade was performed before surgery. RESULTS All the penetrating cardiac and great vessels injuries were repaired by cardiac surgeon, in one case only (6%) the extracorporeal circulation support was used. The injury of coronary arteries was in one case managed by CABG and in the other case by ligation of the peripheral part of the coronary artery. In 4 patients (25%) also a penetrating injury of other organs was simultaneously managed. The mean ICU stay reached 85.8 ± 91.9 hours, on average 5.6 ± 9.3 units of red blood cells were administered during the in-hospital stay which lasted on average 7.1 ± 2.4 days. In the group a nonsignificant increase of left ventricular ejection fraction (44.1 ± 4.7 vs. 49.3 ± 3.2, p = 0.882) was reported at discharge of the injured patients. One patient died on the 78 th day of hypoxic brain damage (6% three-month mortality). The long-term survival analysis showed 94% one-year and 88% five-year cumulative survival in the group. DISCUSSION The incidence of the penetrating cardiac and great vessels injury is directly dependent on the crime level in the respective countries and regions. A cardiac arrest, severe hemodynamic instability, unconsciousness, serious concomitant injury, gunshot injury, multiple or atrial injury represent independent predictors of death in these injuries. The total three-month mortality in penetrating cardiac and great vessels injury ranges from 18 to 42%, the presence of vital signs at the time of hospital admission is associated with 78-92% probability of survival. The surviving patients show excellent long-term results with the exception of those who suffered a severe damage to valve apparatus or with significantly depressed left ventricular function. CONCLUSIONS Our experience proves a high survival rate of patients with penetrating cardiac and great vessels injury. The centralisation of the care into the major trauma centre with a cardiac surgery background, a unified treatment algorithm, and a vital interdisciplinary cooperation are the key goal of successful management of these injuries. Phytomanagement of trace element-contaminated soils can reduce soil toxicity and restore soil ecological functions, including the soil gas exchange with the atmosphere. We studied the emission rate of the greenhouse gases (GHGs) CO 2 , CH 4 , and N 2 O; the potential CH 4 oxidation; denitrification enzyme activity (DEA), and glucose mineralization of a Cu-contaminated soil amended with dolomitic limestone and compost, alone or in combination, after a 2-year phytomanagement with a mixed stand of Populus nigra, Salix viminalis, S. caprea, and Amorpha fruticosa. Soil microbial biomass and microbial community composition after analysis of the phospholipid fatty acids (PLFA) profile were determined. Phytomanagement significantly reduced Cu availability and soil toxicity, increased soil microbial biomass and glucose mineralization capacity, changed the composition of soil microbial communities, and increased the CO 2 and N 2 O emission rates and DEA. Despite such increases, microbial communities were evolving toward less GHG emission per unit of microbial biomass than in untreated soils. Overall, the aided phytostabilization option would allow methanotrophic populations to establish in the remediated soils due to decreased soil toxicity and increased nutrient availability.

Coupling of ion mobility with mass spectrometry has brought new frontiers in separation and quantitation of a wide range of isobaric/isomeric compounds. Ion mobility spectrometry may separate ions possessing the identical molecular formula but having different molecular shapes. The separation space in most commercially available instruments is limited and rarely the mobility resolving power exceeds one hundred. From this perspective, new approaches allowing for extracting individual compound signals out of a more complex mixture are needed. In this work we present a new simple analytical approach based on fitting of arrival time distribution (ATD) profiles by Gaussian functions and generating of ATD functions. These ATD functions well describe even distorted ion mobility peaks of individual compounds and allow for extracting their peaks from mobilograms of mixtures. Contrary to classical integration, our approach works well with irregular overlapping peaks. Using mobilograms of standards to generate ATD functions, poorly separated compounds, e.g. isomers, with identical mass spectra representing a hard to solve task for various chemometric methods can be easily distinguished by our procedure. Alternatively ATD functions can be obtained from ATD profiles of ions unique to individual mixture components (if such ions exist) and mobilograms of standards are not required. On a set of hyaluronan-derived oligosaccharides we demonstrated excellent ATD repeatability enabling the resolution of binary mixtures, including mixtures with minor component level about 5%. A series of peracylated glycosamine-derived thioureas have been synthesized and their behavior as bifunctional organocatalysts has been tested in the enantioselective nucleophilic addition of formaldehyde tert-butyl hydrazone to aliphatic α-keto esters for the synthesis of tertiary azomethyl alcohols. Using the 1,3,4,6-tetra-O-acetyl-2-amino-2-deoxy-β-d-glucosamine derived 3,5-bis-(trifluoromethyl)phenyl thiourea the reaction could be accomplished with high yields (75-98%) and moderate enantioselectivities (50-64% ee). Subsequent high-yielding and racemization-free tranformations of both aromatic- and aliphatic-substituted diazene products in a one pot fashion provide a direct entry to valuable azoxy compounds and α-hydroxy-β-amino esters.

The most of currently produced active pharmaceutical ingredients (APIs) are poorly soluble in the human body. One of the options how to increase their dissolution rate is reducing their particle size. If very small particles of API are desired, traditional milling methods often cause smeared, agglomerated or non-flowing particles due to the forces applied. We tried to compare some of milling methods with the salt-kneading method, which is not typically used in the pharmaceutical industry. Salt-kneading process is driven by several variable parameters (e.g. the amount, hardness and particle size of the salt-kneading material), which influence the degree of size reduction of API particles which are chafed by a surplus of salt-kneading material. A model poorly-soluble API was separately processed with oscillation mill, vibratory mill and kneader; and the morphology, size distribution and solid form of prepared particles were analyzed. Our basic variation of salt-kneading parameters showed the potential of the salt-kneading method, which appears a very effective method of API controlled reduction. The final size can be modified according to the amount and properties of the salt-kneading material. The availability of such a method equips pharmaceutical scientists with a size-reduction method that provides very small, rounded and free-flowing particles of the poorly soluble API and reduces non-preferred needle shape.
