Abstract Background Septic acute kidney injury (S-AKI) is the leading form of acute kidney failure among hospitalized patients, and the inflammatory response is involved in this process. 4-octyl itaconate (4-OI) is a multi-target itaconate derivative with potent anti-inflammatory action. However, it remains elusive whether and how 4-OI contributes to the regulation of S-AKI. Methods We employed a lipopolysaccharide (LPS)-induced AKI murine model and explored the potential renoprotective effect of 4-OI in vivo. In vitro experiments, BUMPT cells, a murine renal tubular cell line, were conducted to examine the effects of 4-OI on inflammation, oxidative stress, and mitophagy. Moreover, STAT3 plasmid was transfected in BUMPT cells to investigate the role of STAT3 signaling in the 4-OI-administrated state. Results We demonstrate that 4-OI protects against S-AKI through suppressing inflammation and oxidative stress and enhancing mitophagy. 4-OI significantly reduced the levels of Scr, BUN, Ngal as well as the tubular injury in LPS-induced AKI mice. 4-OI restrained inflammation by reducing macrophage infiltration and suppressing the expression of IL-1β and NLRP3 in the septic kidney. 4-OI also reduced ROS levels, as well as cleaved caspase-3 and boosted antioxidants such as HO-1, and NQO1 in mice. In addition, the 4-OI treatment significantly promoted mitophagy. Mechanistically, 4-OI activated Nrf2 signaling and suppressed phosphorylated STAT3 in vivo and vitro. Molecular docking revealed the binding affinity of 4-OI towards STAT3. ML385, a specific Nrf2 inhibitor, partially repressed the anti-inflammatory and anti-oxidative effects of 4-OI and partially restricted the mitophagy induced by 4-OI in vivo and in vitro. Transfected with STAT3 plasmid partially suppressed mitophagy and the anti-inflammatory effect provoked by 4-OI in vitro. Conclusion These data suggest that 4-OI ameliorates LPS-induced AKI by suppressing inflammation and oxidative stress and enhancing mitophagy through the overactivation of the Nrf2 signaling pathway, and inactivation of STAT3. Our study identifies 4-OI as a promising pharmacologic for S-AKI.

The stratification of the atmospheric surface layer (ASL) plays an important role in regulating the water vapor and heat exchange across the lake–air interface. Based on one year of data measured by the eddy covariance technique over Erhai Lake in 2015, the ASL stability ($${\rm{\zeta }}$$) was divided into six ranges, including unstable ($$ - 1 \le {\rm{\zeta }} \lt - 0.1$$), weakly unstable ($$ - 0.1 \le {\rm{\zeta }} \lt - 0.01$$), near-neutral1 ($$ - 0.01 \le {\rm{\zeta }} \lt 0$$), near-neutral2 ($$0 \le {\rm{\zeta }} \lt 0.01$$), weakly stable ($$0.01 \le {\rm{\zeta }} \lt 0.1$$), and stable ($$0.1 \le {\rm{\zeta }} \lt 1$$). The characteristics of ASL stability conditions and factors controlling the latent ($$\rm LE$$) and sensible heat ($$H$$) fluxes under different stability conditions were analyzed in this study. The stability conditions of Erhai Lake have noticeably seasonal and diurnal variation, with the near-neutral and (weakly) stable stratification usually occurring before July, with frequencies of 51.7% and 23.3%, respectively, but most of the (weakly) unstable stratification was observed after July, with a frequency of 59.8%. Large evaporation occurred even in stable atmospheric conditions, due to the coupled effects of the relatively larger lake–air vapor pressure difference and wind speed. The relative controls of $$\rm LE$$ and $$H$$ by different atmospheric variables are largely dependent on the stability conditions. In stable and unstable ranges, $$\rm LE$$ is closely correlated with the vapor pressure difference, whereas in weakly unstable to weakly stable ranges, it is primarily controlled by wind speed. $$H$$ is related to wind speed and the lake–air temperature difference under stable conditions, but shows no obvious relationship under unstable conditions.

Water and carbon exchanges between grassland and the atmosphere are important processes for water balance and carbon balance. Based on eddy covariance observations over a semiarid alpine steppe ecosystem in Bange on the Tibetan Plateau during the growing season from 2014 to 2017, the variations in evapotranspiration (ET), net ecosystem exchange (NEE), and their components and the associated driving factors were analyzed. Linear and nonlinear models were applied to investigate the relationships between fluxes and their controlling factors over different timescales. The results show that the average ET for the growing season ranged from 1.1 to 2.4 mm/d with an average of 2.0 mm/d for the four consecutive years. Drought conditions reduced the surface conductance and hence the Priestley–Taylor coefficient. Mean T/ET was low (0.34) due to low vegetation cover. Plant growth increased the T/ET ratio during the growing season, whereas soil water content (SWC) explained most of the variation of ET and E on daily and monthly scales. The Enhanced Vegetation Index (EVI) was the most important controlling factor for temperature. Transpiration increased with SWC in dry conditions. For the growing season in 2014, 2016, and 2017, Bange was a carbon sink, while it was a carbon source in 2015. The largest CO2 flux was higher and the temperature sensitivity coefficient (Q10) was lower for 2015 than for the other three years. SWC affected these photosynthesis and respiration parameters. The ratio of respiration (Re) to gross primary production (GPP) was the highest during the 2015 growing season. Both on daily and monthly scales, Re was positively and linearly correlated with GPP. The most important controlling factor for the CO2 flux was EVI on daily and monthly scales.

Context: Crescents have been noticed in pathologic changes in patients with diabetic kidney disease (DKD). However, the clinical significance of crescents is still not well recognized. Objective: The main objective was to investigate the association between crescents and the prognoses of type 2 DKD (T2DKD) patients, and, secondly, to analyze the relationship between crescents and clinicopathologic features. Methods: A retrospective cohort study of 155 patients with T2DKD diagnosed by renal biopsy was carried out in a single center. Clinicopathologic features of patients with or without crescents were analyzed. Cox regression models and meta-analysis were used to determine the prognostic values of crescents for T2DKD. A nomogram was constructed to provide a simple estimation method of 1, 3, and 5-year renal survival for patients with T2DKD. Results: Compared with T2DKD patients without crescents, patients with crescents had higher 24-hour proteinuria and serum creatinine levels, as well as more severe Kimmelstiel-Wilson (K-W) nodules, segmental sclerosis (SS), and mesangiolysis (all P < .05). Furthermore, the crescents were positively correlated with serum creatinine, 24-hour proteinuria, K-W nodules, SS, mesangiolysis, and complement 3 deposition. Multivariate Cox models showed that crescents were an independent prognostic risk factor for renal survival (hazard ratio [HR] 2.68, 95% CI 1.27-5.64). The meta-analyzed results of 4 studies on crescents in T2DKD confirmed that patients with crescents had a significantly higher HR for renal progression. Conclusion: Patients with crescents in T2DKD have more severe clinicopathologic changes and worse prognoses. The crescent can serve as an independent risk factor for T2DKD progression. Lake Erhai is a subtropical highland shallow lake on southeast margin of Tibetan Plateau, which is influenced by both South Asian and East Asian summer monsoon. Based on four years continuous eddy covariance (EC) data over Lake Erhai, the monsoon effect on water–atmosphere exchange process is investigated by comparing the energy and CO2 fluxes patterns and their main drivers during pre-monsoon (March–April), monsoon (May–October) and post-monsoon (November–December) periods. The results show the atmospheric properties display a large difference for during three different periods. There is a negative difference between water surface and air temperature (ΔT) during pre-monsoon period, while a positive ΔT during post-monsoon period. The diurnal sensible heat flux (Hs) is larger during post-monsoon period, while the latent heat flux (LE) is larger during monsoon period. The monthly average Hs and heat storage (ΔQ) in the lake remain negative during pre-monsoon period and the early monsoon period, and they become positive in the middle monsoon period, which indicates that the lake absorbs heat at first and releases it subsequently. LE plays a dominating role in the energy partitioning of the lake. The Bowen ratio is higher during post-monsoon period. An uptake of CO2 flux is observed in middle of a day during monsoon and post-monsoon period. The ΔT is the main drivers for Hs and the effect of ΔT is increased as time scales are extended from half-hourly to monthly. The wind speed has a weak effect on Hs but a strong effect on LE and CO2 fluxes. Similar main drivers for Hs are found during pre-monsoon period and post-monsoon period, which is also found for CO2 flux, indicating a large impact of monsoon on the heat and carbon exchange process over Lake Erhai.
