We consider a class of nonequilibrium systems of interacting agents with pairwise interactions and quenched disorder in the dynamics featuring, in the thermodynamic limit, phase transitions. We identify mathematical conditions on the microscopic interaction structure, namely the separability of the interaction kernel, that lead to a dimension reduction of the system in terms of a finite number of reaction coordinates (RCs). Such RCs prove to be proper nonequilibrium thermodynamic variables as they carry information on correlation, memory and resilience properties of the system. Phase transitions can be identified and quantitatively characterised as singularities of the complex valued susceptibility functions associated to the RCs. We study a two-layer energy balance model that allows for vertical exchanges between a surface layer and the atmosphere. The evolution equations of the surface temperature and the atmospheric temperature are coupled by the emission of infrared radiation by one level, that emission being partly captured by the other layer, and the effect of all non-radiative vertical exchanges of energy. Therefore, an essential parameter is the absorptivity of the atmosphere, denoted ε a. The value of ε a depends critically on greenhouse gases: increasing concentrations of CO 2 and CH 4 lead to a more opaque atmosphere with higher values of ϵ a. First, we prove that global existence of solutions of the system holds if and only if ε a ∈ (0 , 2) and blow up in finite time occurs if ε a > 2. (Note that the physical range of values for ε a is (0 , 1 ].) Next, we explain the long time dynamics for ε a ∈ (0 , 2) , and we prove that all solutions converge to some equilibrium point. Finally, motivated by the physical context, we study the dependence of the equilibrium points with respect to the involved parameters, and we prove, in particular, that the surface temperature increases monotonically with respect to ε a. Many natural systems show emergent phenomena at different scales, leading to scaling regimes with signatures of deterministic chaos at large scales and an apparently random behavior at small scales. These features are usually investigated quantitatively by studying the properties of the underlying attractor, the compact object asymptotically hosting the trajectories of the system with their invariant density in the phase space. This multi-scale nature of natural systems makes it practically impossible to get a clear picture of the attracting set. Indeed, it spans over a wide range of spatial scales and may even change in time due to non-stationary forcing. Here, we combine an adaptive decomposition method with extreme value theory to study the properties of the instantaneous scale-dependent dimension, which has been recently introduced to characterize such temporal and spatial scale-dependent attractors in turbulence and astrophysics. To provide a quantitative analysis of the properties of this metric, we test it on the well-known low-dimensional deterministic Lorenz-63 system perturbed with additive or multiplicative noise. We demonstrate that the properties of the invariant set depend on the scale we are focusing on and that the scale-dependent dimensions can discriminate between additive and multiplicative noise despite the fact that the two cases have exactly the same stationary invariant measure at large scales. The proposed formalism can be generally helpful to investigate the role of multi-scale fluctuations within complex systems, allowing us to deal with the problem of characterizing the role of stochastic fluctuations across a wide range of physical systems. The atmospheric meridional energy transport in the Northern Hemisphere midlatitudes is mainly accomplished by planetary and synoptic waves. A decomposition into wave components highlights the strong seasonal dependence of the transport, with both the total transport and the contributions from planetary and synoptic waves peaking in winter. In both winter and summer months, poleward transport extremes primarily result from a constructive interference between planetary and synoptic motions. The contribution of the mean meridional circulation is close to climatology. Equatorward transport extremes feature a mean meridional equatorward transport in winter, while the planetary and synoptic modes mostly transport energy poleward. In summer, a systematic destructive interference occurs, with planetary modes mostly transporting energy equatorward and synoptic modes again poleward. This underscores that baroclinic conversion dominates regardless of season in the synoptic wave modes, whereas the planetary waves can be either free or forced, depending on the season. Plain Language Summary: The atmospheric heat transport from low to high latitudes is the main mechanism through which the climate reequilibrates the latitudinally uneven absorption of solar radiation. The atmospheric transport is fueled by instabilities driven by the presence of temperature differences between low and high latitudes and acts in such a way to reduce such gradient. This is one of the main stabilizing mechanisms of the climate system. In this work, we investigate how motions of different spatial scales contribute to atmospheric heat transports in the Northern Hemisphere. We discover that the relative importance of synoptic and planetary scale atmospheric motions is different in summer and winter. Our analysis delves into the analysis of events associated with extreme heat transport toward high latitudes, where we see a compensating mechanism between synoptic and planetary atmospheric motions. We further study days characterized by very large and very small (or even negative) heat transport toward the high latitudes. These "extreme events" are driven by complex interactions between the different scales. Our results are relevant for elucidating basic dynamical and thermodynamical properties of the atmosphere and can be used to benchmark the performance of climate models. A general circulation model of intermediate complexity with an idealized Earth-like aquaplanet setup is used to study the impact of changes in the oceanic heat transport on the global atmospheric circulation. Focus is on the atmospheric mean meridional circulation and global thermodynamic properties. The atmosphere counterbalances to a large extent the imposed changes in the oceanic heat transport, but, nonetheless, significant modifications to the atmospheric general circulation are found. Increasing the strength of the oceanic heat transport up to 2.5PW leads to an increase in the global mean near-surface temperature and to a decrease in its equator-to-pole gradient. For stronger transports, the gradient is reduced further, but the global mean remains approximately constant. This is linked to a cooling and a reversal of the temperature gradient in the tropics. Additionally, a stronger oceanic heat transport leads to a decline in the intensity and a poleward shift of the maxima of both the Hadley and Ferrel cells. Changes in zonal mean diabatic heating and friction impact the properties of the Hadley cell, while the behavior of the Ferrel cell is mostly controlled by friction. The efficiency of the climate machine, the intensity of the Lorenz energy cycle and the material entropy production of the system decline with increased oceanic heat transport. This suggests that the climate system becomes less efficient and turns into a state of reduced entropy production as the enhanced oceanic transport performs a stronger large-scale mixing between geophysical fluids with different temperatures, thus reducing the available energy in the climate system and bringing it closer to a state of thermal equilibrium. In this paper we assess the snow cover and its dynamics for the western river basins of the Indus River system (IRS) and their sub-basins located in Afghanistan, China, India and Pakistan for the period 2001-2012. First, we validate the Moderate Resolution Imaging Spectroradiometer (MODIS) daily snow products from Terra (MOD10A1) and Aqua (MYD10A1) against the Landsat Thematic Mapper/ Enhanced Thematic Mapper plus (TM/ETM+) data set, and then improve them for clouds by applying a validated non-spectral cloud removal technique. The improved snow product has been analysed on a seasonal and annual basis against different topographic parameters (aspect, elevation and slope). Our results show a decreasing tendency for the annual average snow cover for the westerlies-influenced basins (upper Indus basin (UIB), Astore, Hunza, Shigar and Shyok) and an increasing tendency for the monsoon-influenced basins (Jhelum, Kabul, Swat and Gilgit). Seasonal average snow cover decreases during winter and autumn, and increases during spring and summer, which is consistent with the observed cooling and warming trends during the respective seasons. Sub-basins at relatively higher latitudes/ altitudes show higher variability than basins at lower latitudes/middle altitudes. Northeastern and northwestern aspects feature greater snow cover. The mean end-of-summer regional snow line altitude (SLA) zones range from 3000 to 5000ma.s.l. for all basins. Our analysis provides an indication of a descending end-of-summer regional SLA zone for most of the studied basins, which is significant for the Shyok and Kabul basins, thus indicating a change in their water resources. Such results are consistent with the observed hydro-climatic data, recently collected local perceptions and glacier mass balances for the investigated period within the UIB. Moreover, our analysis shows a significant correlation between winter season snow cover and the North Atlantic Oscillation (NAO) index of the previous autumn. Similarly, the inter-annual variability of spring season snow cover and spring season precipitation explains well the inter-annual variability of the summer season discharge from most of the basins. A larger oceanic heat transport leads to a reduction of all reservoirs and conversions of the Lorenz energy cycle but of different relative magnitude for the individual components. The available potential energy of the zonal mean flow and its conversion to eddy available potential energy are affected most. Both the Hadley and Ferrel cell show a decline for increasing oceanic heat transport, with the Hadley cell being more sensitive. Both cells exhibit a poleward shift of their maxima, and the Hadley cell broadens for larger oceanic transports. The partitioning, by means of the Kuo-Eliassen equation, reveals that zonal mean diabatic heating and friction are the most important sources for changes of the Hadley cell, while the behaviour of the Ferrell cell is mostly controlled by friction. This paper discusses the relative performance of several climate models in providing reliable forcing for hydrological modeling in six representative catchments in the Mediterranean region. We consider 14 Regional Climate Models (RCMs), from the EU-FP6 ENSEMBLES project, run for the A1B emission scenario on a common 0.22º (about 24 km) rotated grid over Europe and the Mediterranean region. In the validation period (1951 to 2010) we consider daily precipitation and surface temperatures from the observed data fields (E-OBS) data set, available from the ENSEMBLES project and the data providers in the ECA&D project. Our primary objective is to rank the 14 RCMs for each catchment and select the four best-performing ones to use as common forcing for hydrological models in the six Mediterranean basins considered in the EU-FP7 CLIMB project. Using a common suite of four RCMs for all studied catchments reduces the (epistemic) uncertainty when evaluating trends and climate change impacts in the 21st century. We present and discuss the validation setting, as well as the obtained results and, in some detail, the difficulties we experienced when processing the data. In doing so we also provide useful information and advice for researchers not directly involved in climate modeling, but interested in the use of climate model outputs for hydrological modeling and, more generally, climate change impact studies in the Mediterranean region. Introduction: Patients with schizophrenia show severe difficulties in interpersonal communication, including impairments in conversation skills, like the turn-taking. To our knowledge, very few studies to date have taken into account conversation analysis in order to investigate turn-taking in schizophrenia patients. Objectives: To investigate the conversational patterns in schizophrenia patients; to assess possible associations between dialogic features, abnormal subjective experiences and symptom dimensions. Methods: Thirty-six patients with Schizophrenia underwent an interview, subsequently analyzed with an innovative semi-automatic analysis. Positive and Negative Syndrome Scale (PANSS) was adopted for the investigation of psychopathology and Examination of Anomalous Self Experience (EASE) for Self-Disorders. Results: Dialogic exchanges are graphically represented in Figure 1. An inverse correlation was found between participant speaking time and PANSS negative symptoms score (r = -0.44, p value < 0.05; Figure 2), whereas no associations were found between conversational variables and PANSS positive or disorganization dimensions. Finally, a positive correlation was found between the EASE item "spazialization of thought" and average pause duration (r = 0.42, p value < 0.05). Conclusions: The finding of a relationship between negative symptoms and conversational patterns suggest that conversational features in schizophrenia are expression of the "core" negative dimension of the disorder. The association with the phenomenon of thought spatialization seems to suggest that the disturbances of the stream of consciousness impact on natural dialogic interactions. In this study, we investigate how PCMDI/CMIP3 general circulation models (GCMs) represent the seasonal properties of the hydrological cycle in four major South and Southeast Asian river basins (Indus, Ganges, and Brahmaputra and Mekong). First, we examine the skill of GCMs by analysing their simulations for the XX century climate (1961-2000) under present-day forcing, and then we analyse the projected changes for the corresponding XXI and XXII century climates under SRESA1B scenario. CMIP3 GCMs show a varying degree of skill in simulating the basic characteristics of the monsoonal precipitation regimes of the Ganges, Brahmaputra and Mekong basins, while the representation of the hydrological cycle over the Indus basin is poor in most cases, with few GCMs not capturing the monsoon signal at all. Although the models' outputs feature a remarkable spread for the monsoonal precipitations, a satisfactory representation of the western mid-latitude precipitation regime is instead observed. Similarly, most of the models exhibit a satisfactory agreement for the basin-integrated runoff in winter and spring, while the spread is large for the runoff during the monsoon season. For future climate scenarios, winter (spring) P - E decreases over all four (Indus and Ganges) basins due to decrease in precipitation associated with the western midlatitude disturbances. Consequently, the spring (winter) runoff drops (rises) for the Indus and Ganges basins. Such changes indicate a shift from rather glacial and nival to more pluvial runoff regimes, particularly for the Indus basin. Furthermore, the rise in the projected runoff along with the increase in precipitations during summer and autumn indicates an intensification of the summer monsoon regime for all study basins. We investigate how the climate models contributing to the PCMDI/CMIP3 dataset describe the hydrological cycle over four major South and Southeast Asian river basins (Indus, Ganges, Brahmaputra and Mekong) for the 20th, 21st (13 models) and 22nd (10 models) centuries. For the 20th century, some models do not seem to conserve water at the river basin scale up to a good degree of approximation. The simulated precipitation minus evaporation (P -E), total runoff (R) and precipitation (P) quantities are neither consistent with the observations nor among the models themselves. Most of the models underestimate P - E for all four river basins, which is mainly associated with the underestimation of precipitation. This is in agreement with the recent results on the biases of the representation of monsoonal dynamics by GCMs. Overall, a modest inter-model agreement is found only for the evaporation and inter-annual variability of P - E. For the 21st and 22nd centuries, models agree on the negative (positive) changes of P - E for the Indus basin (Ganges, Brahmaputra and Mekong basins). Most of the models foresee an increase in the inter-annual variability of P - E for the Ganges and Mekong basins, thus suggesting an increase in large low-frequency dry/wet events. In this paper we explore the potential multistability of the climate for a planet around the habitable zone. We focus on conditions reminiscent to those of the Earth system, but our investigation has more general relevance and aims at presenting a general methodology for dealing with exoplanets. We describe a formalism able to provide a thorough analysis of the non-equilibrium thermodynamical properties of the climate system and explore, using a flexible climate model, how such properties depend on the energy input of the parent star, on the infrared atmospheric opacity, and on the rotation rate of the planet. We first show that it is possible to reproduce the multi-stability properties reminiscent of the paleoclimatologically relevant snowball (SB)-warm (W) conditions. We then characterise the thermodynamics of the simulated Wand SB states, clarifying the central role of the hydrological cycle in shaping the irreversibility and the efficiency of the W states, and emphasizing the extreme diversity of the SB states, where dry conditions are realized. Thermodynamics provides the clue for studying the tipping points of the system and leads us to constructing empirical parametrizations allowing for expressing the main thermodynamic properties as functions of the emission temperature of the planet only. Such empirical functions are shown to be rather robust with respect to changing the rotation rate of the planet from the current terrestrial one to half of it. Furthermore, we explore the dynamical range where the length of the day and the length of the year are comparable. We clearly find that there is a critical rotation rate below which the multi-stability properties are lost, and the ice-albedo feedback responsible for the presence of SB and W conditions is damped. The bifurcation graph of the system suggests the presence of a phase transition in the planetary system. Such critical rotation rate corresponds roughly to the phase-lock 2:1 condition. Therefore, if an Earth-like planet is 1:1 phase-locked with respect to the parent star, only one climatic state would be compatible with a given set of astronomical and astro-physical parameters. The article presents a study which focuses on the reconsideration of the Maximum Entropy Production conjecture (MEP) for the representation of the total material entropy production due at the same time to both horizontal and vertical heat fluxes. It says that a fixed longwave transmissivity was assumed to obtained the MEP solutions. It adds that MEP can predict the thermal meridional structure of the atmosphere once the isolation is given.

The understanding of the statistical properties and of the dynamics of multistable systems is gaining more and more importance in a vast variety of scientific fields. This is especially relevant for the investigation of the tipping points of complex systems. Sometimes, in order to understand the time series of given observables exhibiting bimodal distributions, simple one-dimensional Langevin models are fitted to reproduce the observed statistical properties, and used to investing-ate the projected dynamics of the observable. This is of great relevance for studying potential catastrophic changes in the properties of the underlying system or resonant behaviours like those related to stochastic resonancelike mechanisms. In this paper, we propose a framework for encasing this kind of studies, using simple box models of the oceanic circulation and choosing as observable the strength of the thermohaline circulation. We study the statistical properties of the transitions between the two modes of operation of the thermohaline circulation under symmetric boundary forcings and test their agreement with simplified one-dimensional phenomenological theories. We extend our analysis to include stochastic resonance-like amplification processes. We conclude that fitted one-dimensional Langevin models, when closely scrutinised, may result to be more ad-hoc than they seem, lacking robustness and/or well-posedness. The article discusses a study on a Maximum Entropy Production (MEP) solution for climate model. It mentions the application of its hypothesis to a four-box model of climate which answers for horizontal and vertical material heat fluxes. It also notes that the solution is found with realistic temperature and fluxes along with the involvement of meridional and vertical entropy production terms in the maximization making it applicable to each subsystem.

Using a recent theoretical approach, we study how the impact of global warming of the thermodynamics of the climate system by performing experiments with a simplified yet Earth-like climate model. In addition to the globally averaged surface temperature, the intensity of the Lorenz energy cycle, the Carnot efficiency, the material entropy production and the degree of irreversibility of the system are linear with the logarithm of the CO2 concentration. A statistical methodology is proposed and tested for the analysis of extreme values of atmospheric wave activity at mid-latitudes. The adopted methods are the classical block-maximum and peak over threshold, respectively based on the generalized extreme value (GEV) distribution and the generalized Pareto distribution (GPD). Time-series of the ‘Wave Activity Index’ (WAI) and the ‘Baroclinic Activity Index’ (BAI) are computed from simulations of the General Circulation Model ECHAM4.6, which is run under perpetual January conditions. Both the GEV and the GPD analyses indicate that the extremes of WAI and BAI are Weibull distributed, this corresponds to distributions with an upper bound. However, a remarkably large variability is found in the tails of such distributions; distinct simulations carried out under the same experimental setup provide sensibly different estimates of the 200-yr WAI return level. The consequences of this phenomenon in applications of the methodology to climate change studies are discussed. For the discrete model suggested by Lorenz in 1996, a one-dimensional long-wave approximation with nonlinear excitation and diffusion is derived. The model is energy conserving but non-Hamiltonian. In a low-order truncation, weak external forcing of the zonal mean flow induces avalanchelike breather solutions which cause reversal of the mean flow by a wave-mean flow interaction. Abstract: Global and zonal monthly means of cloud cover fraction for total cloudiness (CF) from the ISCCP D2 dataset are compared to same quantities produced by the 20th century simulations of 21 climate models from the World Climate Research Programme''s (WCRP''s) Coupled Model Intercomparison Project phase 3 (CMIP3). The comparison spans the time frame from January 1984 to December 1999 and the global and zonal averages of CF are studied. It is shown that the global mean of CF for the PCMDI-CMIP3 models, averaged over the whole period, exhibits a considerable variance and generally underestimates the ISCCP value. Large differences among models, and between models and observations, are found in the polar areas, where both models and satellite observations are less reliable, and especially near Antarctica. For this reason the zonal analysis is focused over the 60° S–60° N latitudinal belt, which includes the tropical area and mid-latitudes. The two hemispheres are analysed separately to show the variation of the amplitude of the seasonal cycle. Most models underestimate the yearly averaged values of CF over all the analysed areas, whilst they capture, in a qualitatively correct way, the magnitude and the sign of the seasonal cycle over the whole geographical domain, but overestimate the amplitude of the seasonal cycle in the tropical areas and at mid-latitudes, when taken separately. The interannual variability of the yearly averages is underestimated by all models in each area analysed, and also the interannual variability of the amplitude of the seasonal cycle is underestimated, but to a lesser extent. This work shows that the climate models have a heterogeneous behaviour in simulating the CF over different areas of the Globe, with a very wide span both with observed CF and among themselves. Some models agree quite well with the observations in one or more of the metrics employed in this analysis, but not a single model has a statistically significant agreement with the observational datasets on yearly averaged values of CF and on the amplitude of the seasonal cycle over all analysed areas. Introduction: Individuals with schizophrenia present anomalies in the extension and plasticity of the peripersonal space (PPS), the section of space surrounding the body, shaped through motor experiences. A weak multisensory integration in PPS would contribute to an impairment of self-embodiment processing, a core feature of the disorder linked to specific subjective experiences. In this exploratory study, we aimed at: (1) testing an association between PPS features, psychopathology, and subjective experiences in schizophrenia; (2) describing the PPS profile in individuals with early-onset schizophrenia., Materials and Methods: Twenty-seven individuals with schizophrenia underwent a task measuring the PPS size and boundaries demarcation before and after a motor training with a tool. The Positive And Negative Syndrome Scale (PANSS), the Examination of Anomalous Self Experience scale (EASE) and the Autism Rating Scale (ARS) were used to assess psychopathology. Subsequently, participants were divided into two subgroups, early and adult-onset schizophrenia. The two groups were compared in regard to their PPS and psychopathological profiles., Results: PPS patterns were associated with psychopathology, particularly positively with PANSS negative scale score, and negatively with subjective experiences of existential reorientation (EASE Domain 5 scores) and of social encounters (ARS scores). Only PPS parameters and ARS scores differentiated between early and adult-onset participants., Conclusions: Our results, although preliminary and exploratory, can suggest a link between PPS patterns, negative symptoms, and disturbances of the subjective experience, particularly in the intersubjective domain, in schizophrenia. Background: Psychiatric disorders often emerge during adolescence or young adulthood, leading to significant disability among youth. The transition from Child and Adolescent Mental Health Services (CAMHS) to Adult Mental Health Services (AMHS) is critical for individuals experiencing emerging psychopathology, with delayed access to care negatively impacting long-term outcomes. Accessing mental health services for adolescents and young adults is often complex and delayed due to challenges in service visibility, accessibility and appropriateness., Methods: This study examines the care trajectories of individuals consecutively accessing the early detection and intervention (EDI) centre C'JAAD (Evaluation Centre for Young Adults and Adolescents) in Paris (France) over the year 2021. The main goal was to clarify the role of this EDI centre in the continuity of care and transition to AMHS. Data about their history of care, hospitalisations and referral sources were collected retrospectively., Results: The sample comprised 194 individuals, with 57.2% males and a median age of 20 years. Most patients (67.5%) were ≥18 years old upon arrival, with 31% in a situation of not being in education, employment, or training (NEET). Over one-third (35.2%) had prior psychiatric hospitalisations. Patients were mainly referred to our EDI centre from other hospital departments (42.3%). Regarding care in CAMHS, 50.3% of the total sample had medical follow-up during childhood, of whom 41.9% had discontinued care upon arrival at the EDI centre. The median onset age of care in CAMHS was 14, with a median duration of 12 months. Adult patients experienced an approximately 3-year gap between the end of CAMHS care and assessment at the EDI centre., Discussion: The sample's characteristics resemble those of other EDI centres, but concerns persist regarding referral timing and the NEET status of many youths. Lack of prior medical follow-up and challenges in transitioning to AMHS underscore the need to enhance care continuity and address difficulties in accessing care during the transition to adulthood., (© 2024 The Author(s). Individuals with schizophrenia generally show difficulties in interpersonal communication. Linguistic analyses shed new light on speech atypicalities in schizophrenia. However, very little is known about conversational interaction management by these individuals. Moreover, the relationship between linguistic features, psychopathology, and patients' subjectivity has received limited attention to date. We used a novel methodology to explore dyadic conversations involving 58 participants (29 individuals with schizophrenia and 29 control persons) and medical doctors. High-quality stereo recordings were obtained and used to quantify turn-taking patterns. We investigated psychopathological dimensions and subjective experiences using the Positive and Negative Syndrome Scale for Schizophrenia (PANSS), the Examination of Anomalous Self Experience scale (EASE), the Autism Rating Scale (ARS) and the Abnormal Bodily Phenomena questionnaire (ABPq). Different turn-taking patterns of both patients and interviewers characterised conversations involving individuals with schizophrenia. We observed higher levels of overlap and mutual silence in dialogues with the patients compared to dialogues with control persons. Mutual silence was associated with negative symptom severity; no dialogical feature was correlated with anomalous subjective experiences. Bisphenol-A (BPA), a synthetic compound ubiquitously present in the environment, can act as an endocrine disruptor by binding to both canonical and non-canonical estrogen receptors (ERs). Exposure to BPA has been linked to various cancers, in particular, those arising in hormone-targeted tissues such as the breast. In this study, we evaluated the effect of BPA intake through drinking water on ErbB2/ neu -driven cancerogenesis in BALB- neu T mice, transgenic for a mutated ErbB2/ neu receptor gene, which reproducibly develop carcinomas in all mammary glands. In this model, BPA accelerated mammary cancerogenesis with an increase in the number of tumors per mouse and a concurrent decrease in tumor-free and overall survival. As assessed by immunohistochemistry, BALB- neu T tumors were ER-negative but expressed high levels of the alternative estrogen receptor GPR30, regardless of BPA exposure. On the other hand, BPA exposure resulted in a marked upregulation of progesterone receptors in preinvasive tumors and of Ki67, CD31, and phosphorylated Akt in invasive tumors. Moreover, based on several infiltration markers of immune cells, BPA favored an immunosuppressive tumor microenvironment. Finally, in vitro cell survival studies performed on a cell line established from a BALB- neu T breast carcinoma confirmed that BPA's impact on cancer progression can be particularly relevant after chronic, low-dose exposure.

Tipping points (TP) in climate subsystems are usually thought to occur at a well-defined, critical forcing parameter threshold, via destabilization of the system state by a single, dominant positive feedback. However, coupling to other subsystems, additional feedbacks, and spatial heterogeneity may promote further small-amplitude, abrupt reorganizations of geophysical flows at forcing levels lower than the critical threshold. Using a primitive-equation ocean model, we simulate a collapse of the Atlantic Meridional Overturning Circulation (AMOC) due to increasing glacial melt. Considerably before the collapse, various abrupt, qualitative changes in AMOC variability occur. These intermediate tipping points (ITP) are transitions between multiple stable circulation states. Using 2.75 million years of model simulations, we uncover a very rugged stability landscape featuring parameter regions of up to nine coexisting stable states. The path to an AMOC collapse via a sequence of ITPs depends on the rate of change of the meltwater input. This challenges our ability to predict and define safe limits for TPs.

Focal adhesion plaques (FAPs) play an important role in the communication between cells and the extracellular matrix (ECM) and in cells' migration. FAPs are macromolecular complexes made by different proteins which also interact with matrix metalloproteinases (MMPs). Because of these fundamental properties, FAPs and MMPs are also involved in cancer cells' invasion and in the metastatic cascade. The most important proteins involved in FAP formation and activity are (i) integrins, (ii) a complex of intracellular proteins and (iii) cytoskeleton proteins. The latter, together with MMPs, are involved in the formation of filopodia and invadopodia needed for cell movement and ECM degradation. Due to their key role in cancer cell migration and invasion, MMPs and components of FAPs are often upregulated in cancer and are thus potential targets for cancer therapy. Polyphenols, a large group of organic compounds found in plant-based food and beverages, are reported to have many beneficial healthy effects, including anticancer and anti-inflammatory effects. In this review, we discuss the growing evidence which demonstrates that polyphenols can interact with the different components of FAPs and MMPs, inhibit various pathways like PI3K/Akt, lower focal adhesion kinase (FAK) phosphorylation and decrease cancer cells' invasiveness, leading to an overall antitumoral effect. Finally, here we highlight that polyphenols could hold potential as adjunctive therapies to conventional cancer treatments due to their ability to target key mechanisms involved in cancer progression.

This paper introduces an approach to quantifying ecological resilience in biological systems, particularly focusing on noisy systems responding to episodic disturbances with sudden adaptations. Incorporating concepts from nonequilibrium statistical mechanics, we propose a measure termed "ecological resilience through adaptation," specifically tailored to noisy, forced systems that undergo physiological adaptation in the face of stressful environmental changes. Randomness plays a key role, accounting for model uncertainty and the inherent variability in the dynamical response among components of biological systems. Our measure of resilience is rooted in the probabilistic description of states within these systems and is defined in terms of the dynamics of the ensemble average of a model-specific observable quantifying success or well-being. Our approach utilizes stochastic linear response theory to compute how the expected success of a system, originally in statistical equilibrium, dynamically changes in response to a environmental perturbation and a subsequent adaptation. The resulting mathematical derivations allow for the estimation of resilience in terms of ensemble averages of simulated or experimental data. Finally, through a simple but clear conceptual example, we illustrate how our resilience measure can be interpreted and compared to other existing frameworks in the literature. The methodology is general but inspired by applications in plant systems, with the potential for broader application to complex biological processes.

Aim: Social and role functioning impairments characterize patients along the schizophrenia spectrum, but the existing evaluations tools do not specifically address younger population issues. The Global Functioning Social (GF:S) and Global Functioning Role (GF:R) scales have been specifically designed for that purpose. The aim of this study is to establish the reliability and concurrent validity of the French version of GF:S and GF:R scales., Methods: The two scales GF: Social (GF:S) and Role (GF:R) have first been translated into French and independently back translated and validated by the original authors. Between March 2021 and March 2022, we enrolled 51 participants (20.3 ± 3.7 years old; female = 22/51) amongst help-seekers referring to two different early mental health services in the Île-de-France. In an ecological design, participants met different diagnoses, 7 (13.7%) met the criteria for Ultra-High Risk of psychosis (UHR) using CAARMS criteria., Results: Inter-rater reliability was excellent for scores related to the past month and to the higher levels of functioning over the past year. Both scales showed good to excellent concurrent validity as measured by correlation with the Social and Occupational Functioning Assessment Scale (SOFAS) and the Personal and Social Performance Scale (PSP)., Conclusion: Overall, this study confirms the reliability and validity of the French version of the GF:S and GF:R scales. The use of these scales may improve the evaluation of social and occupational functioning in French-speaking young help-seekers, in a transdiagnostic approach, both in clinical and research settings., (© 2023 John Wiley & Sons Australia, Ltd.)

The polyphenols Curcumin (CUR) and Resveratrol (RES) are widely described for their antitumoral effects. However, their low bioavailability is a drawback for their use in therapy. The aim of this study was to explore whether CUR and RES, used at a bioavailable concentration, could modulate immune responses while retaining antitumor activity and to determine whether CUR and RES effects on the immune responses of peripheral blood mononuclear cells (PBMCs) and tumor growth inhibition could be improved by their combination. We demonstrate that the low-dose combination of CUR and RES reduced the survival of cancer cell lines but had no effect on the viability of PBMCs. Although following CUR + RES treatment T lymphocytes showed an enhanced activated state, RES counteracted the increased IFN-γ expression induced by CUR in T cells and the polyphenol combination increased IL-10 production by T regulatory cells. On the other hand, the combined treatment enhanced NK cell activity through the up- and downregulation of activating and inhibitory receptors and increased CD68 expression levels on monocytes/macrophages. Overall, our results indicate that the combination of CUR and RES at low doses differentially shapes immune cells while retaining antitumor activity, support the use of this polyphenol combinations in anticancer therapy and suggest its possible application as adjuvant for NK cell-based immunotherapies.

Background: Quantitative electroencephalography (EEG) analysis offers the opportunity to study high-level cognitive processes across psychiatric disorders. In particular, EEG microstates translate the temporal dynamics of neuronal networks throughout the brain. Their alteration may reflect transdiagnostic anomalies in neurophysiological functions that are impaired in mood, psychosis, and autism spectrum disorders, such as sensorimotor integration, speech, sleep, and sense of self. The main questions this study aims to answer are as follows: 1) Are EEG microstate anomalies associated with clinical and functional prognosis, both in resting conditions and during sleep, across psychiatric disorders? 2) Are EEG microstate anomalies associated with differences in sensorimotor integration, speech, sense of self, and sleep? 3) Can the dynamic of EEG microstates be modulated by a non-drug intervention such as light hypnosis?, Methods: This prospective cohort will include a population of adolescents and young adults, aged 15 to 30 years old, with ultra-high-risk of psychosis (UHR), first-episode psychosis (FEP), schizophrenia (SCZ), autism spectrum disorder (ASD), and major depressive disorder (MDD), as well as healthy controls (CTRL) (N = 21 × 6), who will be assessed at baseline and after one year of follow-up. Participants will undergo deep phenotyping based on psychopathology, neuropsychological assessments, 64-channel EEG recordings, and biological sampling at the two timepoints. At baseline, the EEG recording will also be coupled to a sensorimotor task and a recording of the characteristics of their speech (prosody and turn-taking), a one-night polysomnography, a self-reference effect task in virtual reality (only in UHR, FEP, and CTRL). An interventional ancillary study will involve only healthy controls, in order to assess whether light hypnosis can modify the EEG microstate architecture in a direction opposite to what is seen in disease., Discussion: This transdiagnostic longitudinal case-control study will provide a multimodal neurophysiological assessment of clinical dimensions (sensorimotor integration, speech, sleep, and sense of self) that are disrupted across mood, psychosis, and autism spectrum disorders. Several attempts have been made to develop targeted therapies for malignant mesothelioma (MM), an aggressive tumour with a poor prognosis. In this study we evaluated whether Curcumin (CUR) potentiated the antitumor activity of the ErbB receptors inhibitor Afatinib (AFA) on MM, employing cell lines cultured in vitro and mice bearing intraperitoneally transplanted, syngeneic MM cells. The rationale behind this hypothesis was that CUR could counteract mechanisms of acquired resistance to AFA. We analysed CUR and AFA effects on MM cell growth, cell cycle, autophagy, and on the modulation of tumour-supporting signalling pathways.This study demonstrated that, as compared to the individual compounds, the combination of AFA + CUR had a stronger effect on MM progression which can be ascribed either to increased tumour cell growth inhibition or to an enhanced pro-apoptotic effect. These results warrant future studies aimed at further exploring the therapeutic potential of AFA + CUR-based combination regimens for MM treatment.
