Abstract Methylerythritol cyclodiphosphate (MEcPP) is an intermediate in the biosynthesis of isoprenoids in plant plastids and in bacteria, and acts as a stress signal in plants. Here, we show that MEcPP regulates biofilm formation in Escherichia coli K-12 MG1655. Increased MEcPP levels, triggered by genetic manipulation or oxidative stress, inhibit biofilm development and production of fimbriae. Deletion of fimE, encoding a protein known to downregulate production of adhesive fimbriae, restores biofilm formation in cells with elevated MEcPP levels. Limited proteolysis-coupled mass spectrometry (LiP-MS) reveals that MEcPP interacts with the global regulatory protein H-NS, which is known to repress transcription of fimE. MEcPP prevents the binding of H-NS to the fimE promoter. Therefore, our results indicate that MEcPP can regulate biofilm formation by modulating H-NS activity and thus reducing fimbriae production. Further research is needed to test whether MEcPP plays similar regulatory roles in other bacteria.

Abstract Protein-encoding circular RNAs (circRNAs) are newly identified RNA molecules characterized by intense interaction with translating ribosome. Emerging evidence has implicated physiological and pathological significance of these non-canonical RNAs, yet a large body of them remains unidentified. Due to limited tools at hand, we developed CircProPlus, an automated computational pipeline for de novo detection of translated circRNAs. In comparison to previously established CircPro, CircProPlus adjusts the overall workflow and integrates more robust implements for achieving easier accessibility, higher flexibility and productivity. In present study, we tested the performance of CircProPlus when using different circRNA-detecting implements (i.e., CIRI2, CirComPara2) in the evaluation of coding ability of circRNAs. Results showed that CirComPara2, a state-of-the-art algorithm, consistently outperformed CIRI2 when coupled with CircProPlus in testing real data collected from different RNA libraries and species, which highlighted its potency in data mining of circRNAs with protein-coding potential.

ABSTRACT: This study aims to identify lactic acid bacteria (LAB) isolates possessing physiological characteristics suitable for use as probiotics in yogurt fermentation. Following acid and bile salt tolerance tests, Lactiplantibacillus plantarum (NUC08 and NUC101), Lacticaseibacillus rhamnosus (NUC55 and NUC201), and Lacticaseibacillus paracasei (NUC159, NUC216, and NUC351) were shortlisted based on intraspecies distribution for further evaluation. Their physiological probiotic properties, including transit tolerance, adhesion, autoaggregation, surface hydrophobicity, biofilm formation, and antibacterial activity, were assessed. Principal component analysis indicated that Lactiplantibacillus plantarum NUC08 was the preferred choice among the evaluated strains. Subsequent investigations revealed that co-culturing Lactiplantibacillus plantarum NUC08 with 2 yogurt starter strains resulted in a cooperative and synergistic effect, enhancing the growth of mixed strains and increasing their tolerance to simulated gastric and intestinal conditions. Additionally, when Vibrio harveyi bioluminescent reporter strain was used, the 3 cocultured strains cooperated to induce the activity of a quorum sensing (QS) molecule autoinducer-2 (AI-2), hinting a potential connection between phenotypic traits and QS in the cocultured strains. Importantly, LAB viable counts were significantly higher in yogurt co-fermented with Lactiplantibacillus plantarum NUC08, consistently throughout the storage period. In conclusion, the study demonstrates that the probiotic strain Lactiplantibacillus plantarum NUC08 can be employed in synergy with yogurt starter strains, affirming its potential for use in the development of functional fermented dairy products.

Most prior studies on the insulation effect of off-wall insulation have neglected the natural convection of the air lining; this assumption is inconsistent with reality. In this study, the effect of natural air convection on the heat transfer mechanism in off-wall insulation lining was studied via experiments and numerical simulation. The thermal insulation effect of the air lining was evident. The temperature field distribution of the surrounding rock is affected by the natural convection in the air lining and is thus clearly related to the azimuth. The temperature disturbance of the surrounding rock in the direction of θ = 0° is the minimum, whereas that in the direction of θ = 180° is the maximum. The most significant factors affecting the thermal insulation effect of the off-wall insulation lining are the thermal conductivity of the rock and thermal resistance of the insulation layer, followed by thickness of the air lining and temperature difference between the original rock temperature and the air flow. The relationships between the average unstable heat transfer coefficient and various parameters were obtained via fitting. The fitting formula can be used in engineering practice to calculate the heat transfer from the surrounding rock to the tunnel airflow under various conditions.

Abstract The outlet temperature of the heat recovery reservoir is an important parameter in the design of refrigeration with heat recovery systems. In this paper the second law of thermodynamics has been applied to an irreversible Carnot refrigerator with heat recovery (CRHR) coupled to variable-temperature heat reservoirs. The refrigerating rate, input power, refrigeration coefficient, heat recovery coefficient, comprehensive coefficient of performance and exergy efficiency are chosen as the objective functions. The design rule chosen for this study is that the heat transfer area should be constrained. The mathematical expressions for assessing performance parameters with respect to area ratio, were derived for this study. These expressions are transcendental equations. The numerical solution method was employed to calculate the approximate solutions of the optimum performance parameters in a numerical example. The results indicate that the increase in the outlet temperature of heat recovery reservoir could lead to a rise in the maximum value of refrigerating rate and minimum value of input power; also it will lead to the decline in the maximum value of refrigeration coefficient, heat recovery coefficient, comprehensive coefficient and the exergy efficiency. When the ratio of heat recovery heat exchanger area to the summation of high temperature heat exchanger area and the heat recovery heat exchanger area is 1.0, the performance coefficients would attain their limit values and all of the condensing heat could be recycled. Our findings are helpful to the design and optimization to inform preparation of standard relating to the development of refrigerator with heat recovery.

The moment of inertia of the primary flywheel and the secondary flywheel in a dual mass flywheel (DMF) directly affects the vibration damping performance in an automotive driveline. To enable better minimization of vibration and noise by changing the moment of inertia of the DMF to adjust the frequency characteristics of the automotive driveline, a new variable inertia DMF structure is proposed by introducing electromagnetic devices. The finite element simulation model of the electromagnetic field of an electromagnetic device is established, the electromagnetic field characteristics in the structure are analyzed, and the variation in the electromagnetic force under different air gaps and current conditions is obtained. The electromagnetic force test system of the electromagnetic device is constructed, and the validity of the finite element simulation analysis of the electromagnetic field of the electromagnetic device is verified. A mechanical model of the electromagnetic device is established to analyze the characteristics of the displacement of the moving mass in the structure as well as the variation in the moment of inertia of the DMF at different rotational speeds and currents. The maximum adjustable proportion of its moment of inertia can reach 15.07%. A torsional model of the automotive driveline is established to analyze the effect of variable inertia DMF on the resonance frequency of the system under different currents. The results show that the electromagnetic device introduced in the DMF can realize the active adjustment of the moment of inertia and enable the resonance frequency to decrease with increasing rotational speed, which expands the idea of optimizing the vibration damping performance of the DMF and provides a reference for better control of the torsional vibration of the automobile or other mechanical transmission systems.

BackgroundEpithelioid trophoblastic tumor (ETT) is the rarest gestational trophoblastic tumor, with poor response to chemotherapy. Hysterectomy, as the cornerstone therapy for early ETT, is particularly challenging in reproductive-age women who often have a strong desire for fertility preservation. The management of extra-uterine ETT could be even more complicated and inconsistent. Here we reported a case of isolated ETT lesions in lungs managed with thoracic surgery without hysterectomy.Case presentationA 32-year-old woman presented with amenorrhea for 2 months. Her serum β- human chorionic gonadotropin (hCG) levels fluctuated between 52 and 75 mIU/mL. The patient underwent removal of intrauterine device and suction and curettage, but only proliferative endometrium was found. Methotrexate was given for a provisional diagnosis of ectopic pregnancy of unknown location, while β-hCG had no significant decline. She complained of mild chest pain during the past half year, and the chest computed tomography (CT) result showed two mixed ground-glass nodules of 24 mm × 14.2 mm in right upper lobe and 10 mm × 8 mm in the right lower lobe and a thin-walled cavity in the posterior segment of the left lower lobe. Right upper wedge resection and right lower segmentectomy were performed 3 months later. The result of the pathological examination of pulmonary mass indicated an epithelioid trophoblastic tumor. She was diagnosed with ETT at stage III (with right lung metastasis) according to FIGO 2000. Her menstrual cycle recovered within 1 month after the first thoracic surgery. However, β-hCG was elevated again to 9 mIU/mL, and the positron emission tomography/computed tomography (PET/CT) scans revealed the consolidation of the nodule in the left lower lobe which enlarged to about 1.0 cm × 1.7 cm. Her second pulmonary surgery without hysterectomy was conducted. Followed for 12 months for postoperative monitoring, the patient was found to be disease-free with negative results of serial serum β-hCG and chest CT.ConclusionOur case highlights the efficacy of fertility-sparing surgery for isolated ETT in lungs. The surgical management of pulmonary isolated ETT could be individualized under long-term supervision. Sporadic reports on the favorable outcome of extra-uterine ETT with fertility-sparing surgery were described in the last decades. The safety of this surgical strategy might be warranted only if enough reliable data is accumulated.

This study investigated the effects of nisin combined with ε-polylysine on microorganisms and the refrigerated quality of fresh-cut jackfruit. After being treated with distilled water (control), nisin (0.5 g/L), ε-polylysine (0.5 g/L), and the combination of nisin (0.1 g/L) and ε-polylysine (0.4 g/L), microporous modified atmosphere packaging (MMAP) was carried out and stored at 10 ± 1°C for 8 days. The microorganisms and physicochemical indexes were measured every 2 days during storage. The results indicated that combined treatment (0.1 g/L nisin, 0.4 g/L ε-polylysine) had the best preservation on fresh-cut jackfruit. Compared with the control, combined treatment inhibited microbial growth (total bacterial count, mold and yeast), reduced the weight loss rate, respiratory intensity, polyphenol oxidase and peroxidase activities, and maintained higher sugar acid content, firmness, and color. Furthermore, it preserved higher levels of antioxidant compounds, reduced the accumulation of malondialdehyde and hydrogen peroxide, thereby reducing oxidative damage and maintaining high nutritional and sensory qualities. As a safe application of natural preservatives, nisin combined with ε-polylysine treatment has great application potential in the fresh-cut jackfruit industry.

Minimally invasive techniques have been widely applied in general thoracic surgery. Compared with video-assisted thoracoscopic surgery (VATS), due to its theoretic superiority, robotic surgery is challenging the traditional position of VATS. With its unique advantages, including 3D vision and a high-freedom endowrist, it leads to easier lymph node dissection, more convenient blood vessel dissection, a shorter learning curve and competence for the completion of complex surgery. However, as a new surgical technology, the safety and efficacy of robotic-assisted thoracoscopic surgery (RATS) still need to be further verified. Thus, in this article, we review and summarize the application of RATS versus VATS in general thoracic surgery.

Melatonin treatment was investigated for the sensory quality and senescence in postharvest cattails (Typha latifolia L.) during cold storage. The 0.75 mM melatonin treatment reduced surface browning and delaying lignification of Cattails stored at 4 °C. The results showed that melatonin treatment slowed weight loss and firmness, maintained sensory quality and reducing sugar content. Melatonin treatment reduced browning by inhibiting the increase of MDA and H2O2 contents and POD activity. Melatonin treatment maintained high non-enzymatic antioxidant components (Vitamin C and total phenolic content) and antioxidant enzyme activities (SOD, CAT, and APX), thereby alleviating the browning and senescence of postharvest cattails. These findings indicate that melatonin treatment can maintain postharvest cattails quality.

Abstract Background Individuals with certain chronic inflammatory lung diseases have a higher risk of developing lung cancer (LC). However, the underlying mechanisms remain largely unknown. Here, we hypothesized that chronic exposure to house dust mites (HDM), a common indoor aeroallergen associated with the development of asthma, accelerates LC development through the induction of chronic lung inflammation (CLI). Methods The effects of HDM and heat-inactivated HDM (HI-HDM) extracts were evaluated in two preclinical mouse models of LC (a chemically-induced model using the carcinogen urethane and a genetically-driven model with oncogenic Kras G12D activation in lung epithelial cells) and on murine macrophages in vitro. Pharmacological blockade or genetic deletion of the Nod-like receptor family pyrin domain-containing protein 3 (NLRP3) inflammasome, caspase-1, interleukin-1β (IL-1β), and C–C motif chemokine ligand 2 (CCL2) or treatment with an inhaled corticosteroid (ICS) was used to uncover the pro-tumorigenic effect of HDM. Results Chronic intranasal (i.n) instillation of HDM accelerated LC development in the two mouse models. Mechanistically, HDM caused a particular subtype of CLI, in which the NLRP3/IL-1β signaling pathway is chronically activated in macrophages, and made the lung microenvironment conducive to tumor development. The tumor-promoting effect of HDM was significantly decreased by heat treatment of the HDM extract and was inhibited by NLRP3, IL-1β, and CCL2 neutralization, or ICS treatment. Conclusions Collectively, these data indicate that long-term exposure to HDM can accelerate lung tumorigenesis in susceptible hosts (e.g., mice and potentially humans exposed to lung carcinogens or genetically predisposed to develop LC).

The first quantum revolution has brought us the classical Internet and information technology. Today, as technology advances rapidly, the second quantum revolution quietly arrives, with a crucial moment for quantum technology to establish large-scale quantum networks. However, solid-state quantum bits (such as superconducting and semiconductor qubits) typically operate in the microwave frequency range, making it challenging to transmit signals over long distances. Therefore, there is an urgent need to develop quantum transducer chips capable of converting microwaves into optical photons in the communication band, since the thermal noise of optical photons at room temperature is negligible, rendering them an ideal information carrier for large-scale spatial communication. Such devices are important for connecting different physical platforms and efficiently transmitting quantum information. This paper focuses on the fast-developing field of optomechanical quantum transducers, which has flourished over the past decade, yielding numerous advanced achievements. We categorize transducers based on various mechanical resonators and discuss their principles of operation and their achievements. Based on existing research on optomechanical transducers, we compare the parameters of several mechanical resonators and analyze their advantages and limitations, as well as provide prospects for the future development of quantum transducers.

The vibration frequency characteristics of a rotor system are directly related to its moment of inertia. In this paper, a moment of inertia adjustment device is proposed to adjust the frequency characteristics of a rotor system and better reduce vibration by changing the moment of inertia. First, a mathematical model of the moment of inertia and the temperature field are established. A finite element simulation model of the electromagnetic field of the electromagnetic control unit in the device is established. The influence of current and air gap on the electromagnetic forces is discussed. Then, the validity of the finite element simulation for the electromagnetic control unit is verified using experimental results. In addition, the variations in the displacement and force of the moving mass and the moment of inertia of the device with speed are analyzed. The results show that the proposed moment of inertia adjustment device can be used to significantly adjust the moment of inertia, which provides a reference for better controlling vibrations in rotor systems. Finally, a finite element simulation model for an electromagnetic field analysis of the electromagnetic control unit in the device is established. The results show that the maximum temperature of the electromagnetic control device is 332 K in 60 min, which is in accordance with the requirements.

In this paper, an automatic colony counting system based on an improved image preprocessing algorithm and convolutional neural network (CNN)-assisted automatic counting method was developed. Firstly, we assembled an LED backlighting illumination platform as an image capturing system to obtain photographs of laboratory cultures. Consequently, a dataset was introduced consisting of 390 photos of agar plate cultures, which included 8 microorganisms. Secondly, we implemented a new algorithm for image preprocessing based on light intensity correction, which facilitated clearer differentiation between colony and media areas. Thirdly, a U2-Net was used to predict the probability distribution of the edge of the Petri dish in images to locate region of interest (ROI), and then threshold segmentation was applied to separate it. This U2-Net achieved an F1 score of 99.5% and a mean absolute error (MAE) of 0.0033 on the validation set. Then, another U2-Net was used to separate the colony region within the ROI. This U2-Net achieved an F1 score of 96.5% and an MAE of 0.005 on the validation set. After that, the colony area was segmented into multiple components containing single or adhesive colonies. Finally, the colony components (CC) were innovatively rotated and the image crops were resized as the input (with 14,921 image crops in the training set and 4281 image crops in the validation set) for the ResNet50 network to automatically count the number of colonies. Our method achieved an overall recovery of 97.82% for colony counting and exhibited excellent performance in adhesion classification. To the best of our knowledge, the proposed “light intensity correction-based image preprocessing→U2-Net segmentation for Petri dish edge→U2-Net segmentation for colony region→ResNet50-based counting” scheme represents a new attempt and demonstrates a high degree of automation and accuracy in recognizing and counting single-colony and multi-colony targets.

The two-point and multipoint analyses were calculated by Merlin and SimWalk2 software to obtain the nonparametric linkage (NPL) value, corresponding P value, and parameter linkage limit of detection (LOD) value.Genome-wide linkage analysis yielded a significant linkage signal located on the short arm of chromosome 19. Background: Cryopreservation of ovarian tissue is a promising method for preserving fertility. Transmission electron microscopy (TEM) is an evaluation system for cryo-injury during the cooling and warming process which is very laborious and needs to be optimized. Objective: In this study, we evaluated that serum 17β-oestradiol (E2) may be used as an indicator of vitrified ovarian tissue. Methods: Immunodeficient nude mice were used as hosts for xenografting of vitrified-warmed human ovarian tissues. A total of 54 mice were divided into two group: vitrified ovarian xenotransplant (VOX) group (n ​= ​45) and non-transplant control group (n ​= ​9). The transplanted mice were grouped into vitrified/warmed grafted-4 weeks (VOX-4w, n ​= ​15), vitrified/warmed grafted-6weeks (VOX-6w n ​= ​15) and vitrified/warmed grafted-12 weeks (VOX-12w n ​= ​15) according to the time after transplantation.The viable and functional recovery of grafted ovarian tissue was assessed by light microscopy, transmission electron microscopy, and hormone (E2) assays. Results: Serum E2 concentration was significantly higher in VOX-6w (group (21.07 ​pg/ml) than that of VOX-12w group (15.59 ​pg/ml). VOX-12w group showed a lower value (12.61 ​pg/ml) for E2 concentration. The trend for E2 concentration was consistent with the morphological identification of the grafts. Conclusion: In vivo serum hormone E2 released by cortical biopsies can be used as a functional marker for xenotransplanted vitrified-warmed human ovarian tissue reserve.

Summary Antibiotic resistance in Helicobacter pylori has been growing worldwide with current treatment regimens. Development of new compounds for treatment of H. pylori infections is urgently required to achieve a successful eradication therapy in the future. Armeniaspirols, a novel class of natural products isolated from Streptomyces armeniacus, have been previously identified as antibacterial agents against Gram‐positive pathogens. In this study, we found that armeniaspirol A (ARM1) exhibited potent antibacterial activity against H. pylori, including multidrug‐resistant strains, with MIC range values of 4–16 μg ml‐1. The underlying mechanism of action of ARM1 against H. pylori involved the disruption of bacterial cell membranes. Also, ARM1 inhibited biofilm formation, eliminated preformed biofilms and killed biofilm‐encased H. pylori in a dose‐dependent manner. In a mouse model of multidrug‐resistant H. pylori infection, dual therapy with ARM1 and omeprazole showed efficient in vivo killing efficacy comparable to the standard triple therapy, and induced negligible toxicity against normal tissues. Moreover, at acidic pH 2.5, ARM1 exhibited a much more potent anti‐H. pylori activity than metronidazole. Thus, these findings demonstrated that ARM1 is a novel potent anti‐H. pylori agent, which can be developed as a promising drug lead for treatment of H. pylori infections.

Abstract Background Congenital anatomic abnormalities of fallopian tubes and ovaries are rarely reported. Herein, we describe four cases of undescended ovary during laparoscopic surgery with abnormal anatomy of fallopian tube, yet without abnormal uterine development and urinary system abnormalities, which are analyzed by their clinical features and effects on reproductive function. Case presentation For the patients with undescended ovary, the location of unilateral or bilateral upper poles of the ovaries were usually much higher than that of the bifurcation of the common iliac vessel, and the fallopian tubes at the same side opened in the para-colonic sulcus. Among these four patients, two patients had primary infertility, one patient had tubal pregnancy rupture and bleeding, and one patient had uterine leiomyoma. The development of uterus was normal in all cases, and there was no abnormal development of urinary system. During the infertility examination, the fact that fallopian tubes lifted up in hysterosalpingography (HSG) might be regarded as an indicator of possible undescended ovary. The pelvic ultrasonography examination was of limited use in diagnosing undescended ovary. Conclusion Laparoscopy is the gold standard for the diagnosis of undescended ovary. When there is periodic post-sacral spinal pain, MRI or HSG can be used for diagnosis of undescended ovary.

Helicobacter pylori FabX, a dehydrogenase/isomerase flavoprotein, is required for unsaturated fatty acid synthesis. Here, the authors characterize FabX substrate recognition and catalytic mechanism, and reveal that it contains an atypical [4Fe-4S] cluster, which is essential and participates in the catalytic cycle.

Abstract Background Isoprenoids are the most ancient and essential class of metabolites produced in all organisms, either via mevalonate (MVA)-and/or methylerythritol phosphate (MEP)-pathways. The MEP-pathway is present in all plastid-bearing organisms and most eubacteria. However, no comprehensive study reveals the origination and evolutionary characteristics of MEP-pathway genes in eukaryotes. Results Here, detailed bioinformatics analyses of the MEP-pathway provide an in-depth understanding the evolutionary history of this indispensable biochemical route, and offer a basis for the co-existence of the cytosolic MVA- and plastidial MEP-pathway in plants given the established exchange of the end products between the two isoprenoid-biosynthesis pathways. Here, phylogenetic analyses establish the contributions of both cyanobacteria and Chlamydiae sequences to the plant’s MEP-pathway genes. Moreover, Phylogenetic and inter-species syntenic block analyses demonstrate that six of the seven MEP-pathway genes have predominantly remained as single-copy in land plants in spite of multiple whole-genome duplication events (WGDs). Substitution rate and domain studies display the evolutionary conservation of these genes, reinforced by their high expression levels. Distinct phenotypic variation among plants with reduced expression levels of individual MEP-pathway genes confirm the indispensable function of each nuclear-encoded plastid-targeted MEP-pathway enzyme in plant growth and development. Conclusion Collectively, these findings reveal the polyphyletic origin and restrict conservation of MEP-pathway genes, and reinforce the potential function of the individual enzymes beyond production of the isoprenoids intermediates.

Glycogen can be metabolized via glycogenolysis and the pentose phosphate pathway as well as into the production of UDP glucose, which when secreted can bind the P2Y14 receptor. Here the authors show how these glycogen metabolism pathways contribute to proinflammatory macrophage activation and susceptibility to sepsis.

Abstract Endometriosis (EMS) is a multifactorial disease that affects 10%–15% women of reproductive age and is associated with chronic pelvic pain and infertility. The pathogenesis of EMS has not been consistently explained until now. In this study, we involved 36 endometriosis patients and 14 control subjects who performed laparoscopic surgery due to gynecological benign tumor. The samples from lower third of vagina (CL), posterior vaginal fornix (CU), cervical mucus (CV), endometrium (ET) and peritoneal fluid (PF), were collected and sequenced by 16S rRNA amplicon. The continuous change of the microbiota distribution was identified along the reproductive tract. The flora in lower reproductive tract (CL, CU) were dominated by Lactobacillus. Significant difference of the community diversity began showing in the CV of EMS patients and gradually increased upward the reproductive tract. It indicates the microbiota in cervical samples is expected to be an indicator for the risk of EMS. This study also highlights the decreasing of Lactobacillus in vaginal flora and the increasing of signature Operational Taxonomic Units (OTUs) in transaction zone (CV) and upper reproductive tract (ET, PF) of EMS patients, which reflect the alteration of microbial community associated with EMS, participation of specific colonized bacteria in the EMS pathogenesis and relationship between microbiota and development of disease.

MEcPP is an evolutionarily conserved metabolite that acts as a plastid-to-nucleus retrograde signal to regulate adaptive responses to fluctuating light. Here the authors show that MEcPP regulates seedling development by stabilizing the phyB photoreceptor in an auxin and Ca2+ dependent manner.

With the development of computer technology, screening cancer biomarkers based on public databases has become a common research method. Here, an eight-gene prognostic model, which could be used to judge the prognosis of patients with lung adenocarcinoma (LUAD), was developed through bioinformatics methods. This study firstly used several gene datasets from GEO database to mine differentially expressed genes (DEGs) in LUAD tissue and healthy tissue via joint analysis. Later, enrichment analysis for the DEGs was performed, and it was found that the DEGs were mainly activated in pathways involved in extracellular matrix, cell adhesion, and leukocyte migration. Afterward, a TCGA cohort was used to perform univariate Cox, least absolute shrinkage and selection operator method, and multivariate Cox regression analyses for the DEGs, and a prognostic model consisting of eight genes (GPX3, TCN1, ASPM, PCP4, CAV2, S100P, COL1A1, and SPOK2) was established. Receiver operation characteristic (ROC) curve was then used to substantiate the diagnostic efficacy of the prognostic model. The survival significance of signature genes was verified through the GEPIA database, and the results exhibited that the risk coefficients of the eight genes were basically congruous with the effects of these genes on the prognosis in the GEPIA database, which suggested that the results were accurate. Finally, combined with clinical characteristics of patients, the diagnostic independence of the prognostic model was further validated through univariate and multivariate regression, and the results indicated that the model had independent prognostic value. The overall finding of the study manifested that the eight-gene prognostic model is closely related to the prognosis of LUAD patients, and can be used as an independent prognostic indicator. Additionally, the prognostic model in this study can help doctors make a better diagnosis in treatment and ultimately benefit LUAD patients.

To study the nonlinear dynamic characteristics of the dual-mass flywheel (DMF) under the conformal contact action between the friction damping ring and primary flywheel pressure plate, the contact action model is established and analyzed based on Winkler model. Through analysis and calculation, the contact deformation, contact pressure at different contact positions, and equivalent torsional contact stiffness are obtained. The nonlinear dynamic analysis model of three-degree-of-freedom (3DOF) which takes the conformal contact into account and two-degree-of-freedom (2DOF) without considering conformal contact is established. The approximate analytical solution of the nonlinear frequency characteristics of the system at steady state is derived. By comparing with the results obtained from numerical method, the theoretical analysis process is proved to be valid. And it is found that the overall amplitude and angular displacement transmissibility of the 3DOF model are smaller than the 2DOF model, especially at resonance frequency. The effects of the friction damping ring moment inertia, stiffness of DMF, and axial friction torque on the frequency characteristics of system and angular displacement transmissibility are analyzed. The forced vibration response analysis of the 3DOF model is conducted, through which the torsional angle variations of the primary flywheel, friction damping ring, and secondary flywheel with time are obtained. The results show that the amplitude of the secondary flywheel is much smaller than that of the primary flywheel, indicating that the DMF has prominent damping performance.

Nuclear factor erythroid-2–related factor-2 (NFE2L2/Nrf2) is a transcription factor that regulates the expression of antioxidant genes. Both Kelch-like ECH-associated protein 1 (Keap1) mutations and Nrf2 mutations contribute to the activation of Nrf2 in non-small cell lung cancer (NSCLC). Nrf2 activity is associated with poor prognosis in NSCLC. Metabolic reprogramming represents a cancer hallmark. Increasing studies reveal that Nrf2 activation promotes metabolic reprogramming in cancer. In this review, we discuss the underlying mechanisms of Nrf2-mediated metabolic reprogramming and elucidate its role in NSCLC. Inhibition of Nrf2 can alter metabolic processes, thus suppress tumor growth, prevent metastasis, and increase sensitivity to chemotherapy in NSCLC. In conclusion, Nrf2 may serve as a therapeutic target for the treatment of NSCLC.

Human urine is traditionally considered to be sterile, and whether the urine harbours distinct microbial communities has been a matter of debate. Potential links between female urine and reproductive tract microbial communities is currently not clear. Here, we collected urine samples from 147 Chinese women of reproductive age and explored the nature of colonization by 16S rRNA gene amplicon sequencing, quantitative real-time PCR, and live bacteria culture. To demonstrate the utility of this approach, the intra-individual Spearman’s correlation was used to explore the relationship between urine and multiple sites of the female reproductive tract. PERMANOVA was also performed to explore potential correlations between the lifestyle and various clinical factors and urinary bacterial communities. Our data demonstrated distinct bacterial communities in urine, indicative of a non-sterile environment. Streptococcus-dominated, Lactobacillus-dominated, and diverse type were the three most common urinary bacterial community types in the cohort. Detailed comparison of the urinary microbiota with multiple sites of the female reproductive tract microbiota demonstrated that the urinary microbiota were more similar to the microbiota in the cervix and uterine cavity than to those of the vagina in the same women. Our data demonstrate the potential connectivity among microbiota in the female urogenital system and provide insight and resources for exploring diseases of the urethra and genital tract.

Long noncoding RNA (lncRNA) plays crucial roles in various biological processes of different cancers, especially acting as a competing endogenous RNA (ceRNA). However, the role of lncRNA-mediated ceRNA in Wilms tumor (WT), which is the most common malignant kidney cancer in children, remains unknown. In present study, RNA sequence profiles and clinical data of 125 patients with WT consisting of 119 tumor and 6 normal tissues from Therapeutically Applicable Research To Generate Effective Treatments database were analyzed. A total of 1833 lncRNAs, 156 microRNAs (miRNAs), and 3443 messenger RNAs (mRNAs) were identified as differentially expressed (DE) using “DESeq2” package. The lncRNA-miRNA-mRNA ceRNA regulatory network involving 748 DElncRNAs, 33 DEmiRNAs, and 189 DEmRNAs was constructed based on miRcode, Targetscan, miRTarBase, and miRDB database. Gene Ontology term and Kyoto Encyclopedia of Genes and Genomes pathway analyses revealed that DEmRNAs were mainly enriched in cell proliferation-related processes and tumor-related pathways, respectively, and 13 hub genes were identified by a protein–protein interaction network. Survival analysis detected 48 lncRNAs, 7 miRNAs, and 16 mRNAs to have significant impact on the overall survival of patients with WT. Additionally, we found that 6 DElncRNAs with potential prognostic value were correlated with tumor stage ( DENND5B-AS1 ) and histologic classification ( TMPO-AS1 , RP3-523K23.2 , RP11-598F7.3 , LAMP5-AS1 , and AC013275.2 ) of patients with WT. Our research provides a great insight into understanding the molecular mechanism underlying occurrence and progression of WT, as well as the potential to develop targeted therapies and prognostic biomarkers.

Angiosperms (flowering plants) are the most diverse and species-rich group of plants. The vast majority (∼99.95%) of angiosperms form a clade called Mesangiospermae, which is subdivided into five major groups: eudicots, monocots, magnoliids, Chloranthales, and Ceratophyllales. The relationships among these Mesangiospermae groups have been the subject of long debate. In this study, we assembled a phylogenomic dataset of 1594 genes from 151 angiosperm taxa, including representatives of all five lineages, to investigate the phylogeny of major angiosperm lineages under both coalescent- and concatenation-based methods. We dissected the phylogenetic signal and found that more than half of the genes lack phylogenetic information for the backbone of angiosperm phylogeny. We further removed the genes with weak phylogenetic signal and showed that eudicots, Ceratophyllales, and Chloranthales form a clade, with magnoliids and monocots being the next successive sister lineages. Similar frequencies of gene tree conflict are suggestive of incomplete lineage sorting along the backbone of the angiosperm phylogeny. Our analyses suggest that a fully bifurcating species tree may not be the best way to represent the early radiation of angiosperms. Meanwhile, we inferred that the crown-group angiosperms originated approximately between 255.1 and 222.2 million years ago, and Mesangiospermae diversified into the five extant groups in a short time span (∼27 million years) at the Early to Late Jurassic.

Abstract Background Flowering time is a key trait for regional adaption and seed production in rice (Oryza sativa L.). Forward and reverse genetic studies have characterized a number of flowering-time genes. However, co-expression analysis has not been used to identify the flowering-time genes. Results We predicted a G2-like family transcription factor, OsPHL3, by co-expression networks analysis with photoperiodic flowering pathway genes. OsPHL3 contains a MYB-CC domain, and was localized in the nucleus with transcriptional activation potential. OsPHL3 was mainly expressed in the leaves and exhibited a circadian rhythmic expression pattern. Rice lines overexpressing OsPHL3 showed a delayed flowering time in the genetic background of TP309 under both long-day (Beijing) and short-day (Hainan) conditions. By contrast, the knockout rice lines of OsPHL3 by CRISPR/Cas9 technology promoted flowering time regardless of genetic backgrounds (i.e. Nipponbare and TP309) or day length. Further analysis indicated that OsPHL3 delayed flowering time by down-regulating the expression of Hd3a and RFT1 through promoting Hd1 under long-day conditions (LDs), or suppressing Ehd1/Hd1 under short-day conditions (SDs). Conclusions Our results suggested that co-expression analysis is a useful strategy for identifying novel flowering-time genes in rice.

Multiple sets of spring, spring seat structures are uniformly arranged in circumferential short spring dual mass flywheel (DMF), which generally have a symmetrical structure. The internal springs and spring seats are constrained by the shape of the primary flywheel and the secondary flywheel. At different rotational speeds, the springs and spring seats have different centrifugal forces. To study the dynamic characteristics of the DMF including torque and stiffness with considering the influence of centrifugal force and friction, the discrete method is used to analyze the mechanical actions of the transmission parts in DMF. The torque action between the spring seat and the secondary flywheel is deduced. The dynamic characteristics of the DMF are obtained through analyzing and calculating. Due to the symmetry of the structure, the torque transmitted and the stiffness of the DMF also have specific symmetrical characteristics. Namely, at two relative rotational angles of the same magnitude and opposite direction, the magnitude of the transmitted torque is the same, the direction is opposite. The magnitude and direction of stiffness are the same. The influence of speed, friction coefficient, spring mass, and spring seat mass on the torque and stiffness characteristics are analyzed. Finally, the theoretical analysis is proved to be valid by the torque characteristics test of DMF.

Whether the female reproductive tract harbours distinct microbiomes beyond the vagina has been a matter of debate. Here, the authors show a subject-specific continuity in microbial communities at six sites along the female reproductive tract, indicative of a non-sterile environment.

Background: Genomic DNA methylation plays an important role in both the occurrence and development of bladder cancer. Kaempferol (Kae), a natural flavonoid that is present in many fruits and vegetables, exhibits potent anti-cancer effects in bladder cancer. Similar to other flavonoids, Kae possesses a flavan nucleus in its structure. This structure was reported to inhibit DNA methylation by suppressing DNA methyltransferases (DNMTs). However, whether Kae can inhibit DNA methylation remains unclear. Methods: Nude mice bearing bladder cancer were treated with Kae for 31 days. The genomic DNA was extracted from xenografts and the methylation changes was determined using an Illumina Infinium HumanMethylation 450 BeadChip Array. The ubiquitination was detected using immuno-precipitation assay. Results: Our data indicated that Kae modulated DNA methylation in bladder cancer, inducing 103 differential DNA methylation positions (dDMPs) associated with genes (50 hyper-methylated and 53 hypo-methylated). DNA methylation is mostly relied on the levels of DNMTs. We observed that Kae specifically inhibited the protein levels of DNMT3B without altering the expression of DNMT1 or DNMT3A. However, Kae did not downregulate the transcription of DNMT3B. Interestingly, we observed that Kae induced a premature degradation of DNMT3B by inhibiting protein synthesis with cycloheximide (CHX). By blocking proteasome with MG132, we observed that Kae induced an increased ubiquitination of DNMT3B. These results suggested that Kae could induce the degradation of DNMT3B through ubiquitin-proteasome pathway. Conclusion: Our data indicated that Kae is a novel DNMT3B inhibitor, which may promote the degradation of DNMT3B in bladder cancer.

Counter-current chromatography (CCC) is a unique liquid–liquid partition chromatography and largely relies on the partition interactions of solutes and solvents in two-phase solvents. Usually, the two-phase solvents used in CCC include a lipophilic organic phase and a hydrophilic aqueous phase. Although a large number of partition interactions have been found and used in the CCC separations, there are few studies that address the role of water on solvents and solutes in the two-phase partition. In this study, we presented a new insight that H2O (water) might be an efficient and sensible hydrophobic agent in the n-hexane-methanol-based two-phase partition and CCC separation of lipophilic compounds, i.e., anti-cancer component mollugin from Rubia cordifolia. Although the n-hexane-methanol-based four components solvent systems of n-hexane-ethyl acetate-methanol-water (HEMWat) is one of the most popular CCC solvent systems and widely used for natural products isolation, this is an interesting trial to investigate the water roles in the two-phase solutions. In addition, as an example, the bioactive component mollugin was targeted, separated, and purified by MS-guided CCC with hexane-methanol and minor water as a hydrophobic agent. It might be useful for isolation and purification of lipophilic mollugin and other bioactive compounds complex natural products and traditional Chinese medicines.

This paper mainly studies the heat transfer performance of backplane micro-channel heat pipes by establishing a steady-state numerical model. Compared with the experimental data, the heat transfer characteristics under different structure parameters and operating parameters were studied, and the change of heat transfer coefficient inside the system, the air outlet temperature of the back plate and the influence of different environmental factors on the heat transfer performance of the system were analyzed. The results show that the overall error between simulation results and experimental data is less than 10%. In the range of the optimal filling rate (FR = 64.40%−73.60%), the outlet temperature at the lowest point and the highest point of the evaporation section is 22.46 °C and 19.60 °C, the temperature difference does not exceed 3 °C, and the distribution gradient in vertical height is small and the air outlet temperature is uniform. The heat transfer coefficient between the evaporator and the condenser is larger than the heat transfer coefficient under the conditions of low and high liquid charge rate. It increases gradually along the flow direction, and decreases gradually with the flow rate of the condenser. When the width of the flat tube of the evaporator increases from 20 mm to 28 mm, the internal pressure drop of the evaporator decreases by 45.83% and the heat exchange increases by 18.34%. When the number of evaporator slices increases from 16 to 24, the heat transfer increases first and then decreases, with an overall decrease of 2.86% and an increase of 87.67% in the internal pressure drop of the evaporator. The inclination angle of the corrugation changes from 30° to 60°, and the heat transfer capacity and pressure drop increase. After the inclination angle is greater than 60°, the heat transfer capacity and resistance decrease. The results are of great significance to system optimization design and engineering practical application.

This paper deals with the heat transfer performance of a micro-channel backplane heat pipe air conditioning system. The optimal range of the filling rate of a micro-channel backplane heat pipe air conditioning system was determined in the range of 65−75%, almost free from the interference of working conditions. Then, the influence of temperature and air volume flow rate on the heat exchange system were studied. The system maximum heat exchange is 7000−8000 W, and the temperature difference between the inlet and outlet of the evaporator and the condenser is almost 0 °C. Under the optimum refrigerant filling rate, the heat transfer of the micro-channel heat pipe backplane system is approximately linear with the temperature difference between the inlet air temperature of the evaporator and the cooling distribution unit (CDU) inlet water temperature in the range of 18−28 °C. The last part compares the heat transfer characteristics of two refrigerants at different filling rates. The heat transfer, pressure, and refrigerant temperature of R134a and R22 are the same with the change of filling rate, but the heat transfer of R134a is lower than that of R22. The results are of great significance for the operational control and practical application of a backplane heat pipe system.

In order to improve the working performance and service life of the in-wheel motor for micro-electric vehicle, the outer rotor in-wheel motor of micro-electric vehicle is designed and the model is analyzed, which are based on the theory of electromagnetic field and working principle of the motor. Besides, the size of the micro-electric vehicle wheel is optimized and designed, and the length of the internal and external diameters of inner stator and outer rotor of the outer rotor in-wheel motor are designed and calculated. According to the air-gap selection principles, the parameter of air gap is designed and determined. Some important parameters are derived and calculated, such as the pole-arc coefficient, air-gap magnetic flux density, gap coefficient, and the coefficient of air-gap flux waveforms. The finite element model outer rotor of in-wheel motor is established by Ansoft Maxwell software, and the finite element mesh subdivision is carried on. Magnetic flux densities, magnetic field strength, and magnetic line of force are analyzed by dynamic simulation, and the starting characteristics and the change of torque curve are studied and analyzed. The final simulation analysis results are in good agreement with the design calculation results, which indicate that design scheme of outer rotor in-wheel motor is useful that can meet the performance requirements of outer rotor in-wheel motor for micro-electric vehicle.

The influences of urbanization on weather in Guangdong Province, China, were studied using the Weather Research and Forecasting model from 31 December 2009 through 3 January 2010. Model outputs were compared with extensive monitoring of meteorological data to examine the simulation ability. Model results between tests (with and without land-use change) show that the urbanization had major effects on meteorological fields across nearly the entire Pearl River Delta region and particularly in urban areas. Studied fields (wind speed, temperature, precipitation, and sensible and latent heat fluxes) were affected by the urbanization of the PRD region. The major influences occurred in urban areas, where wind speeds decreased greatly, while the daytime surface upward sensible heat flux clearly increased. Unlike the sensible heat flux, the latent heat flux had a nonmonotonic increase or decrease. As a consequence of the two heat fluxes, 2-m temperature varied with location and time. Change of precipitation was complex. The main rain band became more concentrated, while precipitation decreased upwind of the urban area and increased downwind.

In obesity, CD11c+ innate immune cells are recruited to adipose tissue and create an inflammatory state that causes both insulin and catecholamine resistance. We found that ablation of Gnas, the gene that encodes Gαs, in CD11c expressing cells protects mice from obesity, glucose intolerance, and insulin resistance. Transplantation studies showed that the lean phenotype was conferred by bone marrow-derived cells and did not require adaptive immunity. Loss of cAMP signaling was associated with increased adipose tissue norepinephrine and cAMP signaling, and prevention of catecholamine resistance. The adipose tissue had reduced expression of catecholamine transport and degradation enzymes, suggesting that the elevated norepinephrine resulted from decreased catabolism. Collectively, our results identified an important role for cAMP signaling in CD11c+ innate immune cells in whole-body metabolism by controlling norepinephrine levels in white adipose tissue, modulating catecholamine-induced lipolysis and increasing thermogenesis, which, together, created a lean phenotype. Introduction: Video-assisted thoracic surgery (VATS) has been widely accepted in the diagnosis and treatment of thoracic diseases for the past three decades due to its small incision, minimal trauma, and rapid recovery after surgery. A growing number of patients with concurrent pulmonary nodules and mediastinal lesions have been detected. Simultaneous ipsilateral resection of coexisting lesions is a preferred procedure. Aim: To introduce our technique and preliminary experience in performing uniportal video-assisted thoracic surgery (VATS) for the simultaneous resection of pulmonary and mediastinal lesions. Material and methods: We retrospectively analysed 8 consecutive patients who underwent simultaneous uniportal VATS resection of coexisting lesions of the lung and mediastinum in our centre. The clinical data were recorded and analysed. Results: A total of 8 patients were enrolled, and all patients successfully underwent surgery through a single incision; no perioperative deaths occurred. The average single incision length was 3.9 ±0.3 cm, the operative time was 102.3 ±54.4 min, and the bleeding volume was 27.5 ±17.9 ml. The thoracic drainage time was 3.0 ±2.3 days, with a mean volume of 390.6 ±361.3 ml. The length of postoperative hospital stay was 4.0 ±1.9 (range: 3-9) days. No serious complications were observed in the hospital or during postoperative follow-up. Conclusions: Uniportal VATS is feasible and safe for the simultaneous resection of pulmonary and mediastinal lesions in selected patients and offers cosmetically pleasing incisions and promising clinical application prospects. The objective of this study is to investigate on real time the mineralization process of hydroxyap-atite/chitosan (HAP/CS) composite in a simulated body fluid using the piezoelectric quartz crystal impedance (PQCI) technique and electrochemical impedance spectroscopy (EIS) to reveal its kinetics processes for bone tissue engineering. The morphology and composition for the corresponding stages of mineralization products were characterized by Fourier transformed infrared (FT-IR), X-ray diffraction (XRD) and scanning electrode microscopy (SEM). A decrease of the frequency (f) and an increase of the charge transfer resistance (Rct) at the gold electrode/solution interface were observed due to the mineralization of HAP/CS composite. According to the data of the frequency and static capacity (Cs), the correlative kinetic equations and parameters were obtained by nonlinear regression. The results show that the mineralization process of HAP/CS is divided into two stages: the nucleation stage and the crystal growth stage. The experiments show that the proposed method could provide real time and multidimensional information to the monitoring of bio-material mineralization process. [ABSTRACT FROM AUTHOR]
