To comprehensively describe the operation of photo-induced force microscopy (PiFM), we have developed a model based on coupled harmonic oscillators. This model features two point masses connected by massless elastic wires, offering greater intuitiveness compared to existing PiFM models. It simplifies these models into a unified theoretical framework. By solving the equations of motion using adjusted oscillator parameters, we have successfully replicated all dynamic features from previous theories. These features include resonance frequencies and shapes of eigenmodes, as well as the responses to various external forces in the two PiFM modes: direct coupling and sideband coupling. Furthermore, by integrating our model with a recently developed photo-induced thermal expansion force model, which covers both tip-enhanced and global expansions, we have managed to uncover the underlying physical mechanism responsible for the unique signal behaviors observed in sideband coupling mode, where the signal plot, as a function of sample thickness, unexpectedly exhibits a peak followed by a valley, rather than a proportionally increasing signal. Our study has the potential to enhance the comprehension of various other physical phenomena associated with PiFM in the future.

When light illuminates the junction formed between a sharp metal tip and a sample, different mechanisms can contribute to the measured photoinduced force simultaneously. Of particular interest are the instantaneous force between the induced dipoles in the tip and in the sample, and the force related to thermal heating of the junction. A key difference between these 2 force mechanisms is their spectral behavior. The magnitude of the thermal response follows a dissipative (absorptive) Lorentzian line shape, which measures the heat exchange between light and matter, while the induced dipole response exhibits a dispersive spectrum and relates to the real part of the material polarizability. Because the 2 interactions are sometimes comparable in magnitude, the origin of the chemical selectivity in nanoscale spectroscopic imaging through force detection is often unclear. Here, we demonstrate theoretically and experimentally how the light illumination gives rise to the 2 kinds of photoinduced forces at the tip-sample junction in the midinfrared. We comprehensively address the origin of the spectroscopic forces by discussing cases where the 2 spectrally dependent forces are entwined. The analysis presented here provides a clear and quantitative interpretation of nanoscale chemical measurements of heterogeneous materials and sheds light on the nature of light-matter coupling in optomechanical force-based spectronanoscopy.

This study aimed to investigate the impact of plastic particles on cell viability and tissue toxicity. The cells were subjected to incubation with plastic particles immobilized in agar gels, resulting in minimal cell death and cytotoxicity. Furthermore, direct exposure of various cell types to suspended plastic particles showed negligible effects on cell viability, with no observed lysis or growth inhibition. Staining techniques revealed minimal plastic particle‐induced cell death, with the majority of cells remaining viable. However, histological evaluations of mouse tissues demonstrated that the nondegradable low‐density poly(ethylene) (LDPE) groups exhibited severe irritation, characterized by an excessive presence of macrophages, neutrophils, fibrosis, fat infiltration, and increased blood vessel formation in subcutaneous tissue. In contrast, biodegradable neat poly(butylene succinate) (KRICT‐PBS) and cellulose nanocrystals/PBS nanocomposite (CNC‐PBS) groups induced minimal toxicity. Interestingly, the intravenous injection of KRICT‐PBS and CNC‐PBS degradate solutions in mice exhibited mild toxicity, suggesting no significant damage to normal kidney and liver function. We use a hemispheric sapphire lens in combination with an off-axis parabolic mirror to demonstrate high-resolution vibrationally resonant sum-frequency generation (VR-SFG) microscopy in the mid-infrared range. With the sapphire lens as an immersed solid medium, the numerical aperture (NA) of the parabolic mirror objective is enhanced by a factor of 1.72, from 0.42 to 0.72, close to the theoretical value of 1.76 ( = nsapphire). The measured lateral resolution is as high as 0.64 μm. We show the practical utility of the sapphire immersion lens by imaging collagen-rich tissues with and without the solid immersion lens.

Coherent Raman scattering (CRS) microscopy is gaining acceptance as a valuable addition to the imaging toolset of biological researchers. Optimal use of this label-free imaging technique benefits from a basic understanding of the physical principles and technical merits of the CRS microscope. This tutorial offers qualitative explanations of the principles behind CRS microscopy and provides information about the applicability of this nonlinear optical imaging approach for biological research.

The photoinduced dipole force (PiDF) is an attractive force arising from the Coulombic interaction between the light-induced dipoles on the illuminated tip and the sample. It shows extreme sample-tip distance and refractive index dependence, which is promising for nanoscale infrared (IR) imaging of ultrathin samples. However, the existence of PiDF in the mid-IR region has not been experimentally demonstrated due to the coexistence of photoinduced thermal force (PiTF), typically one to two orders of magnitude higher than PiDF. In this study, we demonstrate that, with the assistance of surface phonon polaritons, the PiDF of c-quartz can be enhanced to surpass its PiTF, enabling a clear observation of PiDF spectra reflecting the properties of the real part of permittivity. Leveraging the detection of the PiDF of phonon polaritonic substrate, we propose a strategy to enhance the sensitivity and contrast of photoinduced force responses in transmission images, facilitating the precise differentiation of the heterogeneous distribution of ultrathin samples. To selectively target and treat murine melanoma B16BL6 tumors expressing αvβ3 integrin receptors, we engineered tumor-specific functional extracellular vesicles (EVs) tailored for the targeted delivery of antitumor drugs. This objective was achieved through the incorporation of a pH-responsive adjuvant, cyclic arginine-glycine-aspartic acid peptide (cRGD, serving as a tumor-targeting ligand), and 5-fluorouracil (5-FU, employed as a model antitumor drug). The pH-responsive adjuvant, essential for modulating drug release, was synthesized by chemically conjugating 3-(diethylamino)propylamine (DEAP) to deoxycholic acid (DOCA, a lipophilic substance capable of integrating into EVs' membranes), denoted as DEAP-DOCA. The DOCA, preactivated using N-(2-aminoethyl)maleimide (AEM), was chemically coupled with the thiol group of the cRGD-DOCA through the thiol–maleimide click reaction, resulting in the formation of cRGD-DOCA. Subsequently, DEAP-DOCA, cRGD-DOCA, and 5-FU were efficiently incorporated into EVs using a sonication method. The resulting tumor-targeting EVs, expressing cRGD ligands, demonstrated enhanced in vitro/in vivo cellular uptake specifically for B16BL6 tumors expressing αvβ3 integrin receptors. The ionization characteristics of the DEAP in DEAP-DOCA induced destabilization of the EVs membrane at pH 6.5 through protonation of the DEAP substance, thereby expediting 5-FU release. Consequently, an improvement in the in vivo antitumor efficacy was observed for B16BL6 tumors. Based on these comprehensive in vitro/in vivo findings, we anticipate that this EV system holds substantial promise as an exceptionally effective platform for antitumor therapeutic delivery. In this study, we developed nanoclusters that respond to singlet oxygen and can be adjusted in size to enhance the efficacy of tumor therapy. Specifically, we engineered nanoclusters, denoted as dHAc‐PSDA‐Ce6, through the covalent conjugation of hyaluronate dots (dHA) with 3,3‐(propane‐2,2‐diylbis(sulfanediyl))dipropionic acid (PSDA) as a cleavable crosslinker mediated by singlet oxygen, and chlorin e6 (Ce6) as a representative model drug for photodynamic therapy. The hyaluronic acid (HA) component within dHAc‐PSDA‐Ce6 was harnessed for the specific targeting of CD44 receptors that are overexpressed on tumor cells. Upon subjecting the vicinity of tumor cells to light irradiation, the PSDA crosslinker within dHAc‐PSDA‐Ce6 undergoes cleavage, likely facilitated by the singlet oxygen generated by Ce6. This cleavage process leads to the formation of dHA‐Ce6 molecules. We hypothesized that a nanocluster, circulating safely within the bloodstream, would undergo a transformation into individual dHA‐Ce6 molecules upon light irradiation at the localized tumor site. This transformation, in turn, enhances the photodynamic antitumor effect. Our in vitro investigations have demonstrated the remarkable efficacy of dHAc‐PSDA‐Ce6 in inducing tumor cell death upon light irradiation. Furthermore, our in vivo biodistribution studies have revealed the substantial accumulation of these nanoclusters within the tumor site. We anticipate that this singlet oxygen‐responsive nanocluster harbors substantial promise for enhancing tumor accumulation and fortifying the effectiveness of tumor eradication. [ABSTRACT FROM AUTHOR]
