Abstract Circadian rhythms (CRs) are fundamental biological processes that significantly impact human well-being. Disruption of these rhythms can trigger insufficient neurocognitive development, insomnia, mental disorders, cardiovascular diseases, metabolic dysfunctions, and cancer. The field of chronobiology has increased our understanding of how rhythm disturbances contribute to cancer pathogenesis, and how circadian timing influences the efficacy of cancer treatments. As the circadian clock steadily gains recognition as an emerging factor in tumorigenesis, a thorough and comprehensive multi-omics analysis of CR genes/proteins has never been performed. To shed light on this, we performed, for the first time, an integrated data analysis encompassing genomic/transcriptomic alterations across 32 cancer types (n = 10,918 tumors) taken from the PanCancer Atlas, unfavorable prognostic protein analysis, protein–protein interactomics, and shortest distance score pathways to cancer hallmark phenotypes. This data mining strategy allowed us to unravel 31 essential CR-related proteins involved in the signaling crossroad between circadian rhythms and cancer. In the context of drugging the clock, we identified pharmacogenomic clinical annotations and drugs currently in late phase clinical trials that could be considered as potential cancer therapeutic strategies. These findings highlight the diverse roles of CR-related genes/proteins in the realm of cancer research and therapy.

Energy is a conserved quantity. The world demands increased energy supply in the future as a means to support life, in specific eco-friendly electricity. In this study, paraffin wax is doped using CNT and CNT/MgO composite at different composition namely 1, 2 and 3%. Similarly, the composition of CNT with MgO is varied in 50 and 75%. Various tests were performed such as thermal conductivity, zeta potential and electrical conductivity were conducted. Results showed that the thermal conductivity of the PCM with 3% CNT concentration is higher about 60% whereas, using CNT and MgO in the composition of 75/25 the thermal conductivity is enhanced to about 75%. On using MgO and 50% CNT–50% MgO, the thermal conductivity reduced. The average zeta potential of the material containing 50%, 75%, and 100% CNT is 0.298, −0.862, and −0.111 mV, respectively. Because of molecular interaction, the impact of electrical conductivity is lower in 100 percent CNT when compared to the hybrid mixture of both nanoparticles. When contrast to the hybrid mixes, the CNT has a low molecular interaction.

Abstract Many primary-tumor subregions exhibit low levels of molecular oxygen and restricted access to nutrients due to poor vascularization in the tissue, phenomenon known as hypoxia. Hypoxic tumors are able to regulate the expression of certain genes and signaling molecules in the microenvironment that shift it towards a more aggressive phenotype. The transcriptional landscape of the tumor favors malignant transformation of neighboring cells and their migration to distant sites. Herein, we focused on identifying key proteins that participate in the signaling crossroads between hypoxic environment and metastasis progression that remain poorly defined. To shed light on these mechanisms, we performed an integrated multi-omics analysis encompassing genomic/transcriptomic alterations of hypoxia-related genes and Buffa hypoxia scores across 17 pancarcinomas taken from the PanCancer Atlas project from The Cancer Genome Atlas consortium, protein–protein interactome network, shortest paths from hypoxia-related proteins to metastatic and angiogenic phenotypes, and drugs involved in current clinical trials to treat the metastatic disease. As results, we identified 30 hypoxia-related proteins highly involved in metastasis and angiogenesis. This set of proteins, validated with the MSK-MET Project, could represent key targets for developing therapies. The upregulation of mRNA was the most prevalent alteration in all cancer types. The highest frequencies of genomic/transcriptomic alterations and hypoxia score belonged to tumor stage 4 and positive metastatic status in all pancarcinomas. The most significantly associated signaling pathways were HIF-1, PI3K-Akt, thyroid hormone, ErbB, FoxO, mTOR, insulin, MAPK, Ras, AMPK, and VEGF. The interactome network revealed high-confidence interactions among hypoxic and metastatic proteins. The analysis of shortest paths revealed several ways to spread metastasis and angiogenesis from hypoxic proteins. Lastly, we identified 23 drugs enrolled in clinical trials focused on metastatic disease treatment. Six of them were involved in advanced-stage clinical trials: aflibercept, bevacizumab, cetuximab, erlotinib, ipatasertib, and panitumumab.

The most complex and important portion of the body and masticatory system are the temporomandibular joint. The growth of mandibular bone in both sagittal and vertical directions is by the condylar process. This study is done to determine different shapes of mandibular condyles using orthopantomographs (OPGs). The study's goal was to determine the morphology of the mandibular condyle in South Indians. One hundred digital OPGs with no defects were selected and obtained from private dental institutes and hospitals in Chennai for this study. The morphology of the mandibular condyle was studied and tabulated. Using the SPSS software, the Chi-square test was done to find associations between demographics and morphology. The study was an attempt to find the prevalent shape of the condylar head using a panoramic radiograph. Fifty-one percent were oval in shape, 20.5% were bird beak in shape, 13.6% were diamond in shape, and 14.9% were crooked finger in shape. The results suggest an oval–oval shape in more frequency among both genders. OPGs are the most commonly used and interpreted radiographs, due to their low radiation and easy availability among dentists. Hence, the shape of the mandibular condyle can also be determined using OPGs; future studies are to be conducted in large samples to make the context evident.

Sustained liver injuries predominantly promote oxidative stress and inflammation that lead to the progression of chronic liver disease (CLD), including fibrosis, cirrhosis, and hepatocellular carcinoma. Boldine, an alkaloid isolated from Peumus boldus, has been shown to have antioxidant and anti-inflammatory effects. Currently, there is no definitive treatment option available for CLD. Therefore, we investigated the hepatoprotective effect of boldine against carbon tetrachloride (CCl4)-induced chronic liver injury in rats. CCl4 (2mL/kg., b.w., i.p.) was administered twice weekly for 5 weeks to induce chronic liver injury in rats. Separate groups of rats were given boldine (20mg/kg b.w., and 40mg/kg b.w.) and silymarin (100 mg/kg b.w.) orally, daily. Serum transaminases, lipid peroxidation, and antioxidant levels were measured, and nuclear factor-μB (NF-μB), tumor necrosis factor-α (TNF-α), cyclooxygenase-2 (cox-2), interleukin-1 β (IL-1β), and α-smooth muscle actin (α-SMA) gene and protein expressions were evaluated. CCl4 administration increased liver marker enzymes of hepatotoxicity in serum and oxidative stress markers, inflammatory genes and α-smooth muscle actin expression in liver tissue. Boldine concurrent treatment suppressed CCl4-induced elevation of transaminase levels in serum, restored enzymic and non-enzymic antioxidants, and downregulated NF-μB, TNF-α, Cox-2 and IL-1β expressions, thereby suppressing hepatic inflammation. Boldine administration also repressed α-SMA expression. The results of this study demonstrate the antioxidant, anti-inflammatory, and antifibrotic properties of boldine, and it can be a potential therapeutic candidate in the treatment of CLD. Objective: To investigate the association between dermatoglyphics and the DNA repair genetic variants in female breast carcinoma. Methodology: The distinct dermatoglyphic variables include ≥ six whorls, Finger ridge counts, A-B Ridge Count, ATD angle and Pattern intensity Index are analyzed for its association with the DNA repair variants namely XRCC1 Arg194Trp, XRCC3Thr241Met, ERCC4Arg 415Gln, and ERCC5 Asp1104His. The statistical procedure used to analyze the frequency of association is odds ratio and relative risk ratio. Result:The results suggests that the relative risk is about 2 to 4 times with statistical significance for breast cancer and high risk group for the genes XRCC1 Arg194Trp, ERCC4 Arg 415 Gln, ERCC5 Asp1104His in their dominant model in both breast cancer and high risk group for six or more whorls, Pattern Intensity Index, A-B RC. Conclusion: It can be suggested that dermal ridges can be used as an effective biomarker of genomic instability in breast cancer.
