Lanthanides, like erbium and dysprosium, have emerged as powerful platforms for quantum-gas research due to their diverse properties, including a significant large spin manifold in their absolute ground state. However, effectively exploiting the spin richness necessitates precise manipulation of spin populations, a challenge yet to be fully addressed in this class of atomic species. In this work, we present an all-optical method for deterministically controlling the spin composition of a dipolar bosonic erbium gas, based on a clocklike transition in the telecom window at 1299nm. The atoms can be prepared in just a few tens of microseconds in any spin-state composition using a sequence of Rabi-pulse pairs, selectively coupling Zeeman sublevels of the ground state with those of the long-lived clocklike state. Finally, we demonstrate that this transition can also be used to create spin-selective light shifts, thus fully suppressing spin-exchange collisions. These experimental results unlock exciting possibilities for implementing advanced spin models in isolated, clean, and fully controllable lattice systems.

Abstract Temporal Talbot effect, the intriguing phenomenon of the self-imaging of optical pulse trains, is extensively investigated using macroscopic components. However, the ability to manipulate pulse trains, either bright or dark, through the Talbot effect on integrated photonic chips to replace bulky instruments has rarely been reported. Here, we design and experimentally demonstrate a proof-of-principle integrated silicon nitride device capable of imprinting the Talbot phase relation onto in-phase optical combs and generating the two-fold self-images at the output. We show that the GHz-repetition-rate bright and dark pulse trains can be doubled without affecting their spectra as a key feature of the temporal Talbot effect. The designed chip can be electrically tuned to switch between pass-through and repetition-rate-multiplication outputs and is compatible with other related frequencies. The results of this work lay the foundations for the large-scale system-on-chip photonic integration of Talbot-based pulse multipliers, enabling the on-chip flexible up-scaling of pulse trains’ repetition rate without altering their amplitude spectra.

Abstract Background Anti-Jo-1 autoantibodies represent essential markers in the diagnosis of antisynthetase syndrome (ASS). In this retrospective study, we aimed to investigate whether their concentrations and fluctuations could both respectively reflect the severity and evolution of ASS. Methods Between 2015 and 2020, clinical and biological features of ASS patients with at least one positive measure of anti-Jo-1 autoantibody were collected. At each serum sampling, we assessed myositis activity by using the Myositis Intention to Treat Activities Index (MITAX) and compared anti-Jo-1 concentrations with ASS severity, anti-Jo-1 concentrations between patients with and without active disease, and changes in anti-Jo-1 concentrations with disease activity. Results Forty-eight patients with ASS had at least one positive determination of anti-Jo-1 concentration. Among them, twenty-nine patients had at least two determinations of anti-Jo-1 autoantibody in their follow-up. We showed that these autoantibody concentrations were significantly correlated with MITAX (r = 0.4, p = 0.03) and creatine kinase concentration (r = 0.34, p = 0.002) and that they were significantly higher in patients with active disease than in those with inactive disease (91.7 IU/L vs 44.4 IU/L, p = 0.016). Stoichiometric silicon nitride is a highly regarded platform for its favorable attributes, such as low propagation loss and compatibility with complementary metal-oxide-semiconductor technology, making it a prominent choice for various linear and nonlinear applications on a chip. However, due to its amorphous structure, silicon nitride lacks second-order nonlinearity; hence, the platform misses the key functionality of linear electro-optical modulation for photonic integrated circuits. Several approaches have been explored to address this problem, including integration with electro-optic active materials, piezoelectric tuning, and utilization of the thermo-optic effect. In this work, we demonstrate electro-optical modulation in a silicon nitride microring resonator enabled by electric-field poling, eliminating the complexities associated with material integration and providing data modulation speeds up to 75 Mb/s, currently only limited by the electrode design. With an estimated inscribed electric field of 100 V/μm, we achieve an effective second-order susceptibility of 0.45 pm/V. In addition, we derive and confirm the value of the material’s third-order susceptibility, which is responsible for the emergence of second-order nonlinearity. These findings broaden the functionality of silicon nitride as a platform for electro-optic modulation.

ObjectivesTo determine factors associated with COVID-19-related death in people with rheumatic diseases.MethodsPhysician-reported registry of adults with rheumatic disease and confirmed or presumptive COVID-19 (from 24 March to 1 July 2020). The primary outcome was COVID-19-related death. Age, sex, smoking status, comorbidities, rheumatic disease diagnosis, disease activity and medications were included as covariates in multivariable logistic regression models. Analyses were further stratified according to rheumatic disease category.ResultsOf 3729 patients (mean age 57 years, 68% female), 390 (10.5%) died. Independent factors associated with COVID-19-related death were age (66-75 years: OR 3.00, 95% CI 2.13 to 4.22; >75 years: 6.18, 4.47 to 8.53; both vs ≤65 years), male sex (1.46, 1.11 to 1.91), hypertension combined with cardiovascular disease (1.89, 1.31 to 2.73), chronic lung disease (1.68, 1.26 to 2.25) and prednisolone-equivalent dosage >10 mg/day (1.69, 1.18 to 2.41; vs no glucocorticoid intake). Moderate/high disease activity (vs remission/low disease activity) was associated with higher odds of death (1.87, 1.27 to 2.77). Rituximab (4.04, 2.32 to 7.03), sulfasalazine (3.60, 1.66 to 7.78), immunosuppressants (azathioprine, cyclophosphamide, ciclosporin, mycophenolate or tacrolimus: 2.22, 1.43 to 3.46) and not receiving any disease-modifying anti-rheumatic drug (DMARD) (2.11, 1.48 to 3.01) were associated with higher odds of death, compared with methotrexate monotherapy. Other synthetic/biological DMARDs were not associated with COVID-19-related death.ConclusionAmong people with rheumatic disease, COVID-19-related death was associated with known general factors (older age, male sex and specific comorbidities) and disease-specific factors (disease activity and specific medications). The association with moderate/high disease activity highlights the importance of adequate disease control with DMARDs, preferably without increasing glucocorticoid dosages. Caution may be required with rituximab, sulfasalazine and some immunosuppressants.

Abstract We consider a set of elementary compactifications of $$D+1$$ D + 1 to D spacetime dimensions on a circle: first for pure general relativity, then in the presence of a scalar field, first free then with a non minimal coupling to the Ricci scalar, and finally in the presence of gauge bosons. We compute the tree-level amplitudes in order to compare some gravitational and non-gravitational amplitudes. This allows us to recover the known constraints of the U(1), dilatonic and scalar weak gravity conjectures in some cases, and to show the interplay of the different interactions. We study the KK modes pair-production in different dimensions. We also discuss the contribution to some of these amplitudes of the non-minimal coupling in higher dimensions for scalar fields to the Ricci scalar.

Quantum fluctuations stabilize dense self-bound macroscopic quantum states in quantum gases. This work places an Erbium dipolar ultracold atomic gas with dominantly attractive long-range interactions in a 1D periodic lattice, and uses interferometric techniques and numerical modeling to characterize the importance of beyond mean-field effects, revealing the emergence of spatially-extended and single-site localized (2D) droplets and signatures of an anisotropic 2D soliton.

Abstract Despite introduction of biological disease modifying anti-rheumatic drugs (DMARDs) for Rheumatoid arthritis (RA) treatment, therapeutic strategies do not always lead to disease control and remission. Hence, a more efficient patient stratification and monitoring biomarkers and tools are needed to enable a more personalized medicine. We used a whole blood based functional flow cytometry assay to characterize immune cells from RA patients (treated or not), healthy donors and psoriatic arthritis (PsA) patients according to their responses to LPS and/or anti-TNFα (infliximab, IFX). Activation marker expression was measured using a 10-color flow cytometry panel following a no-wash protocol. Naïve-to-treatment RA patients had a stronger inflammatory profile in comparison to healthy donors at basal level. Higher expression of activation markers (CD69 and/or CD11b) on NK, B cells and granulocytes and lower expression of the adhesion molecule CD62L were measured on monocytes, granulocytes and B cells. After LPS, naïve RA patients’ cells were less capable of regulating CD69, CD11b, CD16 or CD62L showing impaired activation capabilities. Upon LPS and IFX co-incubation, hierarchical clustering analysis showed different profiles between cohorts. We believe that this whole blood-based approach should further be assessed for RA patient characterization as it provides new perspectives for stratification and/or monitoring.

Abstract Einstein-Maxwell-dilaton theory with non-trivial dilaton potential is known to admit asymptotically flat and (Anti-)de Sitter charged black hole solutions. We investigate the conditions for the presence of horizons as function of the parameters mass M, charge Q and dilaton coupling strength α. We observe that there is a value of α which separate two regions, one where the black hole is Reissner-Nordström-like from a region where it is Schwarzschild-like. We find that for de Sitter and small non-vanishing α, the extremal case is not reached by the solution. We also discuss the attractive or repulsive nature of the leading long distance interaction between two such black holes, or a test particle and one black hole, from a world-line effective field theory point of view. Finally, we discuss possible modifications of the Weak Gravity Conjecture in the presence of both a dilatonic coupling and a cosmological constant.

IntroductionPsoriatic arthritis (PsA) is a chronic inflammatory disease that frequently develops in patients with psoriasis (PsO) but can also occur spontaneously. As a result, PsA diagnosis and treatment is commonly delayed, or even missed outright due to the manifold of clinical presentations that patients often experience. This inevitably results in progressive articular damage to axial and peripheral joints and entheses. As such, patients with PsA frequently experience reduced expectancy and quality of life due to disability. More recently, research has aimed to improve PsA diagnosis and prognosis by identifying novel disease biomarkers.MethodsHere, we conducted a systematic review of the published literature on candidate biomarkers for PsA diagnosis and prognosis in MEDLINE(Pubmed), EMBase and the Cochrane library with the goal to identify clinically applicable PsA biomarkers. Meta-analyses were performed when a diagnostic bone and cartilage turnover biomarker was reported in 2 or moredifferent cohorts of PsA and control. ResultsWe identified 1444 publications and 124 studies met eligibility criteria. We highlighted bone and cartilage turnover biomarkers, genetic markers, and autoantibodies used for diagnostic purposes of PsA, as well as acute phase reactant markers and bone and cartilage turnover biomarkers for activity or prognostic severity purposes. Serum cartilage oligometrix metalloproteinase levels were significantly increased in the PsA sera compared to Healthy Control (HC) with a standardized mean difference (SMD) of 2.305 (95%CI 0.795-3.816, p=0.003) and compared to osteoarthritis (OA) with a SMD of 0.783 (95%CI 0.015-1.551, p=0.046). The pooled serum MMP-3 levels were significantly higher in PsA patients than in PsO patients with a SMD of 0.419 (95%CI 0.119-0.719; p=0.006), but no significant difference was highlighted when PsA were compared to HC. While we did not identify any new genetic biomarkers that would be useful in the diagnosis of PsA, recent data with autoantibodies appear to be promising in diagnosis, but no replication studies have been published. ConclusionIn summary, no specific diagnostic biomarkers for PsA were identified and further studies are needed to assess the performance of potential biomarkers that can distinguish PsA from OA and other chronic inflammatory diseases.

Abstract Background Anterior uveitis (AU) is the most frequent extra-articular feature of axial spondyloarthritis (axSpA). We aimed to assess and compare the incidence of AU in axSpA patients treated with anti-TNF or anti-IL17A. Methods We systematically reviewed PubMed, EMBase, and Cochrane from inception to May 3, 2020, and searched for placebo-controlled and head-to-head randomized controlled trials (RCTs) assessing anti-TNF monoclonal antibodies (mAb) or soluble receptor fusion protein or anti-IL17A in patients with axSpA according to ASAS criteria and reporting safety data on AU. Data were extracted following a predefined protocol. We did pairwise and network meta-analyses for the primary outcome of AU flares (relapse or de novo) incidence and estimated summary odds ratios (ORs). We assessed the quality of evidence using the Cochrane risk-of-bias 2.0 tool. We ranked treatments according to their effectiveness in preventing AU flare using the P-score. Results We identified 752 citations and included 33 RCTs, comprising 4544 treated patients (anti-TNF mAb 2101, etanercept [ETN] 699, anti-IL17A 1744) and 2497 placebo-receiving patients. Incidence of uveitis was lower with anti-TNF mAb versus placebo (OR = 0.46; CI 95% [0.24; 0.90]) and versus anti-IL17A (OR = 0.34; CI 95% [0.12; 0.92]. According to the P-score, the ranking from the most to the least preventive treatment of uveitis flare was as follows: anti-TNF mAb, ETN, placebo, and anti-IL17A. Conclusion In RCTs assessing anti-TNF and anti-IL17A in axSpA, incident uveitis are rare events. However, this network meta-analysis demonstrates that anti-TNF mAb are associated with a lower incidence of uveitis compared to placebo and anti-IL17A.

Abstract Lactic acid bacteria, in particular Lactococcus lactis, are widely used in the food industry, for the control and/or the protection of the manufacturing processes of fermented food. While L. lactis has been reported to form compact and uniform biofilms it was recently shown that certain strains able to display pili at their surface form more complex biofilms exhibiting heterogeneous and aerial structures. As the impact of those biofilm structures on the biomechanical properties of the biofilms is poorly understood, these were investigated using AFM force spectroscopy and imaging. Three types of strains were used i.e., a control strain devoid of pili and surface mucus-binding protein, a strain displaying pili but no mucus-binding proteins and a strain displaying both pili and a mucus-binding protein. To identify potential correlations between the nanomechanical measurements and the biofilm architecture, 24-h old biofilms were characterized by confocal laser scanning microscopy. Globally the strains devoid of pili displayed smoother and stiffer biofilms (Young Modulus of 4–100 kPa) than those of piliated strains (Young Modulus around 0.04–0.1 kPa). Additional display of a mucus-binding protein did not affect the biofilm stiffness but made the biofilm smoother and more compact. Finally, we demonstrated the role of pili in the biofilm cohesiveness by monitoring the homotypic adhesion of bacteria to the biofilm surface. These results will help to understand the role of pili and mucus-binding proteins withstanding external forces.

Abstract Tiny values for gauge couplings of dark photons allow to suppress their kinetic mixing with ordinary photons. We point out that the weak gravity conjecture predicts consequently low ultraviolet cut-offs where new degrees of freedom might appear. In particular, a mixing angle of $$\mathcal {O}(10^{-15})$$ O ( 10 - 15 ) , required in order to fit the excess reported by XENON1T, corresponds to new physics below $$\mathcal {O}(100)$$ O ( 100 ) TeV, thus accessible at a future circular collider. We show that possible realizations are provided by compactifications with six large extra dimensions and a string scale of order $$\mathcal {O}(100)$$ O ( 100 ) TeV.

Abstract We revisit the scalar weak gravity conjecture and investigate the possibility to impose that scalar interactions dominate over gravitational ones. More precisely, we look for consequences of assuming that, for leading scalar interactions, the corresponding gravitational contribution is sub-dominant in the non-relativistic limit. For a single massive scalar particle, this leads us to compare four-point self-interactions in different type of potentials. For axion-like particles, we retrieve the result of the axion weak gravity conjecture: the decay constant f is bounded by the Planck mass, $$f < {M_{Pl}}$$ f

This article focuses on the study of the mass transfer involved in the application of a bacterial antifouling technique for membrane bioreactors (MBR), via the addition of solid media. These alginate objects can contain a biological system capable of producing an enzyme that degrades the signal molecules responsible for membrane fouling. The objective of this article is to quantify the mass transfer by distinguishing two main types: the transfer from the liquid to the solid media and the transfer from solid media to the liquid phase. For this purpose, a model molecule was chosen, and experiments were specifically developed with an optical device to track the concentration of the dye in the liquid phase, considering three different shapes for the particles (beads, hollow cylinders, and flat sheets). The experiments were first performed in jar tests and then in a lab-scale reactor. The results of this study revealed that the total amount of dye transferred into the sheets was greater than that transferred into the cylinders or the beads, which was attributed to the sheets having a larger exchange area for the same volume. When the dyed media were implemented in the MBR (loading rate of solid media: 0.45% v/v—no biomass), the global transfer coefficient from the sheets to the liquid was found to be greater than for the other shapes, indicating a faster transfer phenomenon. The effect of aeration in the MBR was investigated and an optimal air flowrate for fostering the transfer was found, based on the highest transfer coefficient that was obtained. This study provided key information about mass transfer in MBRs and how it is affected by the particle shapes and the MBR operating conditions.

Intrinsically disordered regions (IDRs) are abundant in the proteome of RNA viruses. The multifunctional properties of these regions are widely documented and their structural flexibility is associated with the low constraint in their amino acid positions. Therefore, from an evolutionary stand point, these regions could have a greater propensity to accumulate non-synonymous mutations (NS) than highly structured regions (ORs, or ‘ordered regions’). To address this hypothesis, we compared the distribution of non-synonymous mutations (NS), which we relate here to mutational robustness, in IDRs and ORs in the genome of potyviruses, a major genus of plant viruses. For this purpose, a simulation model was built and used to distinguish a possible selection phenomenon in the biological datasets from randomly generated mutations. We analyzed several short-term experimental evolution datasets. An analysis was also performed on the natural diversity of three different species of potyviruses reflecting their long-term evolution. We observed that the mutational robustness of IDRs is significantly higher than that of ORs. Moreover, the substitutions in the ORs are very constrained by the conservation of the physico-chemical properties of the amino acids. This feature is not found in the IDRs where the substitutions tend to be more random. This reflects the weak structural constraints in these regions, wherein an amino acid polymorphism is naturally conserved. In the course of evolution, potyvirus IDRs and ORs follow different evolutive paths with respect to their mutational robustness. These results have forced the authors to consider the hypothesis that IDRs and their associated amino acid polymorphism could constitute a potential adaptive reservoir.

ObjectivesRheumatoid arthritis (RA) is associated with HLA-DRB1 genes encoding the shared epitope (SE), a 5-amino acid motive. RA is usually preceded by the emergence of anti-citrullinated protein/peptide antibodies (ACPAs). Citrulline is a neutral amino acid resulting from post-translational modification of arginine involved in peptidic bounds (arginyl residue) by PeptidylArginine Deiminases (PADs). ACPAs recognize epitopes from citrullinated human fibrin(ogen) (hFib) and can be specifically detected by the AhFibA assay. Five citrullinated peptides derived from hFib together represent almost all of the epitopes recognized by patients with ACPA-positive RA, namely: α36–50cit, α171–185cit, α501–515cit, α621–635cit, and β60–74cit. The use of antibody fine specificities as markers of clinical phenotypes has become a major challenge. Our objective was to study whether RA clinical characteristics and HLA-DRB1 genetic background were associated with a specific reactivity against the epitopes borne by the five peptides.Methods184 ACPA-positive RA patients fulfilling the 2010 ACR/EULAR criteria were studied. Patient characteristics including HLA-DRB1 genotype, were collected from their medical files. Anti-CCP2 antibodies, AhFibA, and antibodies against the five citrullinated hFib (hFib-cit) peptides were analyzed by ELISA.ResultsAnti-α505-515cit antibodies were associated with HLA-DRB1*04:01 (OR = 5.52 [2.00 – 13.64]; p = 0.0003). High level anti-α505-515cit antibodies were associated with rheumatoid nodules (OR = 2.71 [1.00 – 7.16], p= 0.044).ConclusionImmune complexes containing anti-α501-515cit antibodies and rheumatoid factors might be involved in the development of rheumatoid nodules on the HLA-DRB1*04:01 background. Apheresis of these epitope-specific antibodies might be a new therapeutic opportunity for patients with rheumatoid nodules.

Abstract It has been observed that an automatic alignment without decoupling is predicted at tree-level in a Two-Higgs Doublet Model (2HDM) with extended supersymmetry in the gauge and Higgs sectors. Moreover, it was found that radiative corrections preserve this alignment to a very good precision. We show that it is the non-abelian global $$SU(2)_R$$ SU(2)R R-symmetry that is at the origin of this alignment. This differs from previously considered Higgs family symmetries as it is present only in the quartic part of the Higgs potential. It can not be imposed to the quadratic part which has to be generated by $$N=1$$ N=1 supersymmetry breaking sectors. This absence of symmetry does not spoil alignment at the minimum of the potential. We show how the (small) misalignment induced by higher order corrections can be described as the appearance of non-singlet representations of $$SU(2)_R$$ SU(2)R in the quartic potential.

Conformational intrinsic disorder is a feature present in many virus proteins. Intrinsically disordered regions (IDRs) have weaker structural requirement than ordered regions and mutations in IDRs could have a lower impact on the virus fitness. This could favor its exploration of adaptive solutions. The potyviral protein VPg contains IDRs with determinants for adaptation to its host plant. To experimentally assess whether IDRs are more resistant to mutations than ordered regions, the biologically relevant interaction between mutant libraries of both VPg and the eukaryotic translation initiation factor 4E (eIF4E) and their respective wild type partner was examined using yeast two hybrid assay. Our data shows that VPg is significantly more robust to mutations than eIF4E and as such belongs to a particular class of intrinsically disordered proteins. This result is discussed from the standpoint of IDRs involvement in the virus adaptive processes.

Preventive actions of probiotics as antidiarrheal agents are well documented, but their mechanisms are poorly understood. Two selected probiotics, Bacillus subtilis CU1 and Lactobacillus plantarum CNCM I-4547, were tested in mouse experimental models of diarrhea and the possible mechanisms of action were investigated. Diarrhea was induced in mice by oral castor oil administration or by i.v. injection of lipopolysaccharide (LPS) of Salmonella enteritis. The antidiarrheal drug loperamide was used as control. Fecal water excretion was quantified for 2 h and paracellular permeability and electrical parameters of the colon were assessed in Ussing chambers. The expression of colonic exchangers or channels and of Toll-like receptor 4 (TLR4) was assessed by immunohistochemistry. Prophylactic treatment with B. subtilis CU1 or with L. plantarum CNCM I-4547 reduced LPS-induced diarrhea. The reduction of water excretion was in the same range as those induced by loperamide. In the castor oil model, this effect was only observed with B. subtilis CU1. The two probiotic treatments abolished the increase in paracellular permeability induced by LPS, but not by castor oil. However, only L. plantarum CNCM I-4547 treatment decreased the colonic expression of TLR-4. After B. subtilis CU1, colonic expression of cystic fibrosis transmembrane conductance regulator (CFTR) was reduced and that of Na+/H+ exchanger 3 (NHE3) increased. B. subtilis CU1 may increase the capacity of the colon to absorb excess of water in diarrheic conditions by acting on CFTR and NHE3 expression. The two probiotics strains showed an impact on diarrhea through limitation of water excretion that may involve paracellular permeability or electrolyte transport for L. plantarum CNCM I-4547 and B. subtilis CU1 respectively.

Numerous in vitro studies have shown the impact of mechanical forces on fibroblasts through mechanotransduction, which consists of the conversion of mechanical signals to biochemical responses. Such responses are characterized by the modulation of gene expression coding not only for extracellular matrix components (collagens, elastin, etc.) but also for degradation enzymes (matrix metalloproteinases [MMPs]) and their inhibitors (tissue inhibitors of metalloproteinases [TIMPs]). A new device providing a mechanical stimulation of the cutaneous and subcutaneous tissue has been used in a simple, blinded, controlled, and randomized study. Materials and methods: Thirty subjects (aged between 35 years and 50 years), with clinical signs of skin sagging, were randomly assigned to have a treatment on hemiface. After a total of 24 sessions with Mécano-Stimulation™, biopsies were performed on the treated side and control area for in vitro analysis (dosage of hyaluronic acid, elastin, type I collagen, MMP9; equivalent dermis retraction; GlaSbox®; n=10) and electron microscopy (n=10). Furthermore, before and after the treatment, clinical evaluations and self-assessment questionnaire were done.Results: In vitro analysis showed increases in hyaluronic acid, elastin, type I collagen, and MMP9 content along with an improvement of the migratory capacity of the fibroblasts on the treated side. Electron microscopy evaluations showed a clear dermal remodeling in relation with the activation of fibroblast activity. A significant improvement of different clinical signs associated with skin aging and the satisfaction of the subjects were observed, correlated with an improvement of the sagging cheek.Conclusion: Mécano-Stimulation is a noninvasive and safe technique delivered by flaps microbeats at various frequencies, which can significantly improve the skin trophicity. Results observed with objective measurements, ie, in vitro assessments and electron microscopy, confirm the firming and restructuring effect clinically observed. We show that a non-abelian global S U ( 2 ) R R-symmetry acting on the quartic part of the two Higgs Doublet Model leads, at tree-level, to an automatic alignment without decoupling. An example of phenomenologically viable model with this feature is the the low energy effective field theory of the Minimal Dirac Gaugino Supersymmetric Model in the limit where the adjoint scalars are decoupled. We discuss here how the S U ( 2 ) R can be identified with the R-symmetry of the N = 2 supersymmetry in the gauge and Higgs sectors. We also review how the radiative corrections lead to a very small misalignment.
