The determination of the endodormancy release and the beginning of ontogenetic development is a challenge, because these are non-observable stages. Changes in protein activity are important aspects of signal transduction. The conversion of threonine to 2-oxobutanoate is the first step towards isoleucine (Ile) biosynthesis, which promote growth and development. The reaction is catalyzed by threonine deaminase/dehydratase (TD). This study on TD activity was conducted at the experimental sweet cherry orchard at Berlin-Dahlem. Fresh (FW), dry weight (DW), water content (WC), and the specific TD activity for the cherry cultivars Summit, Karina and Regina were conducted from flower bud samples between October and April. The content of asparagine (Asn), aspartic acid (Asp), Ile, and valine (Val) were exemplarily shown for Summit. In buds of Summit and Karina, the TD activity was one week after the beginning of the ontogenetic development (t1*), significantly higher compared to samplings during endo- and ecodormancy. Such “peak” activity did not occur in the buds of Regina; TD tended for a longer time (day of year, DOY 6–48) to a higher activity, compared to the time DOY 287–350. For the date “one week after t1*”, the upregulation of TD, the markedly increase of the Ile and Val content, and the increase of the water content in the buds, all this enzymatically confirms the estimated start of the ontogenetic development (t1*) in sweet cherry buds.

The conception of “floral strips” is a strategy to provide more and different food sources for pollinators. The impact of “homogeneous” Phacelia tanacetifolia (“Phacelia”) and “heterogeneous” (flower mix) food sources on the enzyme activity of bumblebees (Bombus terrestris) and honeybees (Apis mellifera) under urban conditions has not been reported. Organisms responding to challenging environmental conditions are known to exhibit increases in oxidative stress parameters which in turn affect both physiological and metabolic parameters. A field study was conducted in Berlin-Dahlem, Germany, using the response of the “marker” enzyme superoxide dismutase (SOD) on food sources for assessment. SOD data is also shown from the wild bee Megachile rotundata (Fabricius 1787), obtained from three different locations in the federal state Brandenburg, Germany. The results demonstrate that the enzyme activity of SOD significantly increased in bumblebees visiting the flower mix compared to the Phacelia. The experimental approach had individual effects at the level of the species, bumblebees and honeybees, respectively. The activity of the biomarker SOD could be successfully used to assess the effects of the compositions of homogeneous and heterogeneous flower fields.

The scientific objective of this study was to answer the question of whether sink limitation is also true for high quality wheat varieties. We examined the incorporation of 15N and 13C during phenological phases into vegetative parts and grains of Elite wheat Triso (E) and Quality wheat Naxos (A) when the spike is halved. Three splits of fertilizer were applied at EC 11, EC 30, EC 59, whereby 10% at EC 30 and EC 59 was 15N, and plants were also labelled with 13CO2. The application of only the third split as 15N, combined with spike-halving, resulted in a significantly higher 15N-content (+11%) of 0.486 mg 15N/g DM, compared to the control (0.437 mg15N/g DM). Labelling whole plants with 13CO2 at EC 59 resulted in a significantly higher 13C-content—40%—(0.223 mg 13C/g DM) of the grains of the control for Triso at the fully-ripe stage (EC 89), compared to Naxos (0.160 mg 13C/g DM). This superiority was reduced to 34%, and was also demonstrated by spike-halving (0.226 mg 13C/g DM, 0.169 mg 13C/g DM). Remobilization of 15N for control and spike-halving treatments were 68.2% and 61.1%, respectively. This clearly demonstrates that the reduction of the sink size by spike-halving leads to a 7% reduction in the remobilization of 15N from vegetative to reproductive tissues.

In a split N-application system, the objective was to quantify N/15N in gluten and non-gluten proteins after the late application of 30 or 60 kg N, whereby 10% of the third split was applied as 15N. This fertilization was combined with a reduced water supply for 21 days (well-watered (ww); water deficit (wd)). German spring wheat cultivars, Elite wheat Taifun, Quality wheat Monsun and cultivars from the Mediterranean territory, Golia, Gönen, were examined. The protein content in gluten was for 30 kg N, ww, similar for Taifun, Golia, and Gönen, but markedly lower in Monsun (231, 245, 247, 194 mg protein/g DM). The water deficit increased the protein content in the gluten of Golia and Gönen and was higher than that of Taifun and Monsun (297, 257, 249, 202 mg protein/g DM). Fertilization of 60 kg N, ww, did not result in any change in the protein content in gluten and differences between the cultivars were not detectable. The 15N protein in gluten was for 30 kg N, ww, markedly higher in Gönen (2.32 mg 15N protein/g DM), compared to Golia and Monsun (1.93, 1.50 mg 15N protein/g DM), and similar in Taifun (1.64 mg 15N protein/g DM). 15N fertilizer uptake into gluten was stimulated by water deficit for 30 and 60 kg N, leading to significantly increased 15N protein in Golia and Gönen, (2.38, 2.99, 4.34, 5.87 mg 15N protein/g DM). Fertilization of 60 kg N led to a proportional two-time increase in the 15N gluten protein of the four cultivars, in ww and wd plants. Assessed on the basis of 15N fertilizer allocation under wd conditions into gluten proteins, Golia and Gönen have a stronger sink activity, compared to Taifun and Monsun.

For several years, there has been a need in phenological modeling to better account for physiological processes during the winter dormancy of woody plants, which is here addressed to the sweet cherry cultivar ‘Summit’ (Prunus avium L.). This study compares three sequential phenology models (M1–M3) for the beginning of ‘Summit’ blossom in the experimental sweet cherry orchard in Berlin-Dahlem (Germany) between 2011/12–2019/20 (model development) and 2020/21–2022/23 (model validation). M1 represents an inverse modeling approach where the chilling and forcing requirements of ‘Summit’ were optimized solely from observed flowering data. In contrast, M2 and M3 are more physiologically based as they already incorporate biological knowledge, so that the model parameters were calculated directly within the specified developmental phases. Here, M2 is a two-phase model that considers experimental data for the date of endodormancy release (t1) of nine years (2011/12–2019/20) to calculate the chilling and forcing requirements. Finally, M3 is a newly developed three-phase model that additionally includes the onset of ontogenetic development (t1*) and the abscisic acid (ABA) content of ‘Summit’ flower buds during the ecodormancy phase (t1 − t1*). The results indicate that the inclusion of ABA-related heat weighting during ecodormancy significantly improves the performance of M3 compared to M1 and M2. While M1 gives satisfactory results in terms of fit and validation, it is considered physiologically unacceptable as it greatly overestimates the chilling requirement of ‘Summit’ by ignoring the ecodormancy phase. M2 accumulates too much heat during ecodormancy as it does not include control by the bud ABA content. The results highlight the need for parameters such as t1, t1*, and the bud ABA content for the physiological modeling of ‘Summit’ blossom. To the best of our knowledge, this is the first study to provide a pathway towards a physiologically based modeling approach.

This study was undertaken to determine the degradation and synthesis of sucrose (Suc) in sweet cherry buds and the twig tissue response to a sequence of environmental temperature changes (cold (orchard) - warm (controlled temperature of ∼22 °C) - cold (orchard)). The results of two years' (2016, 2017) findings were compared with the buds of trees and the buds of twigs in November/December in northeast Germany. The Suc content in the buds of trees and the buds of twigs under natural conditions was stable. Temperatures of ∼22 °C resulted in a significant (Suc) degradation (62%, from 39 to 15 mg/g DW) in the buds of twigs after 21 days (day of the year (DOY) 340). The significant re-synthesis (66%, to 25 mg/g DW after 21 days, DOY 361) in the orchard is noteworthy, and highlights the Suc value as a cryoprotective saccharide. The marked changes in the Suc, glucose, and fructose contents of the twigs exposed to a cold-warm-cold sequence (< DOY 319, DOY 319-340, DOY 340-361), lead to the conclusion that this adaptation is the result of tissue- and cold-specific sucrose invertases/synthases. The effect of low-temperature-active enzymes explains the role of Suc in the buds of trees during the winter rest. When using twigs for plant physiological examinations during the winter rest, results on a metabolite level should be considered when drawing conclusions concerning the overall tree physiology.

Models used to predict the onset of fruit tree blossom under changed climate conditions should be physiologically based as much as possible. Pure optimized phenology models carry the risk of unrealistic predictions due to a misinterpretation of metabolic processes. This was the motivation determining the relevant phases for chill and heat accumulation, which induces cherry blossom (cv. Summit). Investigations are based on 8 years of observational and analytical data, as well as on controlled experiments. For ‘Summit’ buds, to be released from endodormancy, 43 chill portions from 1 September are necessary. After endodormancy release (t1), on average on 30 November, no further chilling is required, because no correlation between chill accumulation during ecodormancy and the subsequent heat accumulation until ‘Summit’ blossom exist. The declining amount of heat, which induces cherry blossom after t1—shown in several forcing experiments—seems to be the result of the declining bud’s abscisic acid (ABA) content, up to ~50% until the beginning of ontogenetic development. Shortly after t1, when the bud’s ABA content is high, a huge amount of heat is necessary to induce cherry blossom under controlled conditions. Heat requirement reduces during ecodormancy along with the reduction in the ABA content. According to these findings, plant development during ecodormancy is suppressed by low temperatures in the orchard and a slowly declining bud’s ABA content. These results should lead to a better consideration of the ecodormancy phase in phenology models.

Many biochemical processes are involved in regulating the consecutive transition of different phases of dormancy in sweet cherry buds. An evaluation based on a metabolic approach has, as yet, only been partly addressed. The aim of this work, therefore, was to determine which plant metabolites could serve as biomarkers for the different transitions in sweet cherry buds. The focus here was on those metabolites involved in oxidation-reduction processes during bud dormancy, as determined by targeted and untargeted mass spectrometry-based methods. The metabolites addressed included phenolic compounds, ascorbate/dehydroascorbate, reducing sugars, carotenoids and chlorophylls. The results demonstrate that the content of phenolic compounds decrease until the end of endodormancy. After a long period of constancy until the end of ecodormancy, a final phase of further decrease followed up to the phenophase open cluster. The main phenolic compounds were caffeoylquinic acids, coumaroylquinic acids and catechins, as well as quercetin and kaempferol derivatives. The data also support the protective role of ascorbate and glutathione in the para- and endodormancy phases. Consistent trends in the content of reducing sugars can be elucidated for the different phenophases of dormancy, too. The untargeted approach with principle component analysis (PCA) clearly differentiates the different timings of dormancy giving further valuable information.
