Abstract It is self‐evident that our chests expand and contract during breathing but, surprisingly, exactly how individual alveoli change shape over the respiratory cycle is still a matter of debate. Some argue that all the alveoli expand and contract rhythmically. Others claim that the lung volume change is due to groups of alveoli collapsing and reopening during ventilation. Although this question might seem to be an insignificant detail for healthy individuals, it might be a matter of life and death for patients with compromised lungs. Past analyses were based on static post‐mortem preparations primarily due to technological limitations, and therefore, by definition, incapable of providing dynamic information. In contrast, this study provides the first comprehensive dynamic data on how the shape of the alveoli changes, and, further, provides valuable insights into the optimal lung volume for efficient gas exchange. It is concluded that alveolar micro‐dynamics is nonlinear; and at medium lung volume, alveoli expand more than the ducts.

Abstract Understanding how the alveolar mechanics work in live lungs is essential for comprehending how the lung behaves during breathing. Due to the lack of appropriate imaging tools, previous research has suggested that alveolar morphologies are polyhedral rather than spherical based on a 2D examination of alveoli in fixed lungs. Here, we directly observe high-resolution 3D alveoli in live mice lungs utilizing synchrotron x-ray microtomography to show spherical alveolar morphologies from the live lungs. Our measurements from x-ray microtomography show high sphericity, low packing density, big alveolar size, and low osmotic pressure, indicating that spherical alveolar morphologies are natural in living lungs. The alveolar packing fraction is quite low in live lungs, where the spherical alveoli would behave like free bubbles, while the confinement of alveolar clusters in fixed lungs would lead to significant morphological deformations of the alveoli appearing polyhedral. Direct observations of the spherical alveolar shapes will help understand and treat lung disease and ventilation.

Abstract Synchrotron X-rays can be used to obtain highly detailed images of parts of the lung. However, micro-motion artifacts induced by such as cardiac motion impede quantitative visualization of the alveoli in the lungs. This paper proposes a method that applies a neural network for synchrotron X-ray Computed Tomography (CT) data to reconstruct the high-quality 3D structure of alveoli in intact mouse lungs at expiration, without needing ground-truth data. Our approach reconstructs the spatial sequence of CT images by using a deep-image prior with interpolated input latent variables, and in this way significantly enhances the images of alveolar structure compared with the prior art. The approach successfully visualizes 3D alveolar units of intact mouse lungs at expiration and enables us to measure the diameter of the alveoli. We believe that our approach helps to accurately visualize other living organs hampered by micro-motion.

Abstract Biopolymers are essential building blocks that constitute cells and tissues with well‐defined molecular structures and diverse biological functions. Their three‐dimensional (3D) complex architectures are used to analyze, control, and mimic various cells and their ensembles. However, the free‐form and high‐resolution structuring of various biopolymers remain challenging because their structural and rheological control depend critically on their polymeric types at the submicron scale. Here, direct 3D writing of intact biopolymers is demonstrated using a systemic combination of nanoscale confinement, evaporation, and solidification of a biopolymer‐containing solution. A femtoliter solution is confined in an ultra‐shallow liquid interface between a fine‐tuned nanopipette and a chosen substrate surface to achieve directional growth of biopolymer nanowires via solvent‐exclusive evaporation and concurrent solution supply. The evaporation‐dependent printing is biopolymer type‐independent, therefore, the 3D motor‐operated precise nanopipette positioning allows in situ printing of nucleic acids, polysaccharides, and proteins with submicron resolution. By controlling concentrations and molecular weights, several different biopolymers are reproducibly patterned with desired size and geometry, and their 3D architectures are biologically active in various solvents with no structural deformation. Notably, protein‐based nanowire patterns exhibit pin‐point localization of spatiotemporal biofunctions, including target recognition and catalytic peroxidation, indicating their application potential in organ‐on‐chips and micro‐tissue engineering.

By the use of high-resolution X-ray diffractometry and synchrotron radiation topography, the distribution of dislocations in AlN crystals grown on evaporating SiC substrates is studied. The growth of AlN layer in one process with the evaporation of SiC substrate gave the possibility to prevent the relaxation of thermal stresses through cracking of AlN during cooling the structure. The continuous 0.2-1.5 mm thick plates were used as the model objects for the study of dislocation structure near the AlN/SiC interface. Analysis of the broadening of the Bragg reflection peaks, the shape of scattering maps in reciprocal space and topographs showed that dislocations formed the mosaic structure, different from the distribution of threading dislocations in GaN epilayers. A theoretical model for misfit strain relaxation in the growing AlN layer is suggested. The results enable to clarify the dislocation processes during sublimation growth of industrial quality AlN crystals.

The advanced characteristics of synchrotron light has led in recent years to the development of a series of new experimental techniques to investigate chemical and physical properties on a microscopic scale. Although originally developed for materials science and biomedical research, such techniques find increasing applications in other domains – and could be quite useful for the study and conservation of cultural heritage. Specifically, they can nondestructively provide detailed chemical composition information that can be useful for the identification of specimens, for the discovery of historical links based on the sources of chemical raw materials and on chemical processes, for the analysis of damage, their causes and remedies and for many other issues. Likewise, morphological and structural information on a microscopic scale is useful for the identification, study and preservation of many different cultural and historical specimens. We concentrate here on two classes of techniques: in the first case, photoemission spectromicroscopy. This is the result of the advanced evolution of photoemission techniques like ESCA (Electron Microscopy for Chemical Analysis). By combining high lateral resolution to spectroscopy, photoemission spectromicroscopy can deliver fine chemical information on a microscopic scale in a nondestructive fashion. The second class of techniques exploits the high lateral coherence of modern synchrotron sources, a byproduct of the quest for high brightness or brilliance. We will see that such techniques now push radiology into the submicron scale and the submillisecond time domain. Furthermore, they can be implemented in a tomographic mode, increasing the information and becoming potentially quite useful for the analysis of cultural heritage specimens.

To exploit unique properties of thin films of group III-nitride semiconductors, the production of native substrates is to be developed. The best choice would be AlN; however, presently available templates on sapphire or SiC substrates are defective. The quality of AlN could be improved by eliminating the substrate during the layer growth. In this paper, we demonstrate freestanding AlN layers fabricated by an SiC substrate evaporation method. Such layers were used to investigate dislocation structures near the former AlN–SiC interface. Specimens were characterized by synchrotron radiation imaging, triple-axis diffractometry and transmission electron microscopy (TEM). We found that the evaporation process under non-optimal conditions affected the dislocation structure. When the growth had been optimized, AlN layers showed a uniform distribution of dislocations. The dislocations tended to constitute low-angle subgrain boundaries, which produced out-of-plane and in-plane tilt angles of about 2–3 arc-min. Similar broadening was observed in both symmetric and asymmetric rocking curves, which proved the presence of edge, screws as well as mixed dislocation content. TEM revealed arrays of edge threading dislocations, but their predominance over the other threading dislocations was not supported by present study. To explain the experimental observations, a theoretical model of the dislocation structure formation is proposed.

Single freestanding microtubes of poly(methyl methacrylate)/polypyrrole (PMMA/PPy) are produced based on a meniscus-guided approach. A ring-deposit of nanoparticles is first formed in a meniscus solution of PMMA/PPy nanoparticles by outward liquid flow in fast solvent evaporation. Continuous accumulation of nanoparticles on the ring-deposit is then made by guiding the meniscus upward under the outward flow, thereby forming single composite microtube with controlled outer diameter and wall thickness. The meniscus-guiding enables us to produce an array of freestanding microtubes that are individually controlled in size at the desired positions. We demonstrate individually addressable gas sensors by integrating PMMA/PPy microtubes on electrodes.

The manner in which the nervous system regulates animal behaviors in natural environments is a fundamental issue in biology. To address this question, C. elegans has been widely used as a model animal for the analysis of various animal behaviors. Previous behavioral assays have been limited to two-dimensional (2-D) environments, confining the worm motion to a planar substrate that does not reflect three-dimensional (3-D) natural environments such as rotting fruits or soil. Here, we develop a 3-D worm tracker (3DWT) for freely moving C. elegans in 3-D environments, based on a stereoscopic configuration. The 3DWT provides us with a quantitative trajectory, including the position and movement direction of the worm in 3-D. The 3DWT is also capable of recording and visualizing postures of the moving worm in 3-D, which are more complex than those in 2-D. Our 3DWT affords new opportunities for understanding the nervous system function that regulates animal behaviors in natural 3-D environments.

Heteroepitaxial growth behavior of Mn-doped thin films on Al2O3 by pulsed laser deposition is investigated particularly as a function of Mn content (0.00 less than or equal to x less than or equal to 0.35) using synchrotron x-ray scattering and atomic force microscopy. The superior epitaxial growth of the film with an optimum Mn content was attributed to the formation of much larger grains in it.

Heteroepitaxial growth behavior of SrRuO(sub 3)/SrTiO(sub 3)(001) a thin film during early stages by pulsed laser deposition was investigated using synchrotron x-ray scattering, atomic force microscopy, and high resolution transmission electron microscopy. The 3D islands are coherently strained to the substrate, possessing a very narrow mosaic distribution of 0.007degrees half width at half maximum.

A report is presented of the ion-beam-induced graphitization of ultrathin amorphous carbon films at ambient temperature.

The investigation of the effects of strain energy on the required orientation of TiN thin films shows that the overall energy of the film depends on the surface energy, as its strain energy is very small. The TiN film is usually deposited by plasma-enhanced chemical vapor deposition with a power of 50 watts. However, in rf sputter deposited TiN films, the strain energy caused by the large strain controls the overall energy of the film. The study reveals that the strain energy increases with thickness.

Synchrotron x-ray scattering, field emission scanning electron microscope, and vibrating sample magnetometer are used for examining the inhibition of abnormal acicular grain growth of the Ba-ferrite phase in the crystallization of Ba-ferrite films. The Ba-ferrite phase first nucleated at a higher temperature of 750 degree C in comparison to the alpha-Fe2O3 phase, which nucleated at 700 degree Celsius?

Purpose: This study tests the possibility of using synchrotron radiation (SR) x-ray micro-imaging as a new method to evaluate osseointegration. Materials and Methods: A simple imaging setup with primarily unmonochromatic SR was used to study the interface of a dental implant in contact with bone. The samples were prepared by sectioning the undecalcified specimen of a titanium screw implanted in the tibia of a New Zealand white rabbit. Radiographs of the interface between surrounding tissues and dental titanium implant were imaged using the SR imaging system at PLS (Pohang Light Source) 5C1 beamline, a micro-computed tomography (μCT) system (SkyScan-1072), and a conventional dental x-ray system (Siemens Heliodent MD). Results: The image quality of the osseointegrated titanium implant was compared among the 3 imaging systems. The SR imaging technique showed greater details than other radiographic modalities for evaluation of the healing stage of bone-implant contact. Discussion: The evaluation was especially focused on the image quality of the osseous contact at the bone-to-implant interfaces. Conclusions: This SR imaging technique provides finer details and can be expected to make an impact in the clinical study of osseointegration. [ABSTRACT FROM AUTHOR]

Abstract Background Cells in the human body experience different growth environments and conditions, such as compressive pressure and oxygen concentrations, depending on the type and location of the tissue. Thus, a culture device that emulates the environment inside the body is required to study cells outside the body. Methods A blanket-type cell culture device (Direct Contact Pressing: DCP) was fabricated with an alginate-based hydrogel. Changes in cell morphology due to DCP pressure were observed using a phase contrast microscope. The changes in the oxygen permeability and pressure according to the hydrogel concentration of DCP were analyzed. To compare the effects of DCP with normal or artificial hypoxic cultures, cells were divided based on the culture technique: normal culture, DCP culture device, and artificial hypoxic environment. Changes in phenotype, genes, and glycosaminoglycan amounts according to each environment were evaluated. Based on this, the mechanism of each culture environment on the intrinsic properties of conserving chondrocytes was suggested. Results Chondrocytes live under pressure from the surrounding collagen tissue and experience a hypoxic environment because collagen inhibits oxygen permeability. By culturing the chondrocytes in a DCP environment, the capability of DCP to produce a low-oxygen and physical pressure environment was verified. When human primary chondrocytes, which require pressure and a low-oxygen environment during culture to maintain their innate properties, were cultured using the hydrogel blanket, the original shapes and properties of the chondrocytes were maintained. The intrinsic properties could be recovered even in aged cells that had lost their original cell properties. Conclusions A DCP culture method using a biomimetic hydrogel blanket provides cells with an adjustable physical pressure and a low-oxygen environment. Through this technique, we could maintain the original cellular phenotypes and intrinsic properties of human primary chondrocytes. The results of this study can be applied to other cells that require special pressure and oxygen concentration control to maintain their intrinsic properties. Additionally, this technique has the potential to be applied to the re-differentiation of cells that have lost their original properties.

Abstract Mesenchymal stem cells (MSCs) are increasingly used as regenerative therapies for patients in the preclinical and clinical phases of various diseases. However, the main limitations of such therapies include functional heterogeneity and the lack of appropriate quality control (QC) methods for functional screening of MSC lines; thus, clinical outcomes are inconsistent. Recently, machine learning (ML)-based methods, in conjunction with single-cell morphological profiling, have been proposed as alternatives to conventional in vitro/vivo assays that evaluate MSC functions. Such methods perform in silico analyses of MSC functions by training ML algorithms to find highly nonlinear connections between MSC functions and morphology. Although such approaches are promising, they are limited in that extensive, high-content single-cell imaging is required; moreover, manually identified morphological features cannot be generalized to other experimental settings. To address these limitations, we propose an end-to-end deep learning (DL) framework for functional screening of MSC lines using live-cell microscopic images of MSC populations. We quantitatively evaluate various convolutional neural network (CNN) models and demonstrate that our method accurately classifies in vitro MSC lines to high/low multilineage differentiating stress-enduring (MUSE) cells markers from multiple donors. A total of 6,120 cell images were obtained from 8 MSC lines, and they were classified into two groups according to MUSE cell markers analyzed by immunofluorescence staining and FACS. The optimized DenseNet121 model showed area under the curve (AUC) 0.975, accuracy 0.922, F1 0.922, sensitivity 0.905, specificity 0.942, positive predictive value 0.940, and negative predictive value 0.908. Therefore, our DL-based framework is a convenient high-throughput method that could serve as an effective QC strategy in future clinical biomanufacturing processes.

The stretched exponent, that is a constant in the classical stretched exponential decay, intrinsically changes with time for luminescence decays were found. The time dependence of the stretched exponent was attributed to the lifetime dispersion of localized excitons.

Abstract Background Stem cell transplantation is a fascinating therapeutic approach for the treatment of many neurodegenerative disorders; however, clinical trials using stem cells have not been as effective as expected based on preclinical studies. The aim of this study is to validate the hypothesis that human neural crest-derived nasal turbinate stem cells (hNTSCs) are a clinically promising therapeutic source of adult stem cells for the treatment of Alzheimer’s disease (AD). Methods hNTSCs were evaluated in comparison with human bone marrow-derived mesenchymal stem cells (hBM-MSCs) according to the effect of transplantation on AD pathology, including PET/CT neuroimaging, immune status indicated by microglial numbers and autophagic capacity, neuronal survival, and cognition, in a 5 × FAD transgenic mouse model of AD. Results We demonstrated that hNTSCs showed a high proliferative capacity and great neurogenic properties in vitro. Compared with hBM-MSC transplantation, hNTSC transplantation markedly reduced Aβ42 levels and plaque formation in the brains of the 5 × FAD transgenic AD mice on neuroimaging, concomitant with increased survival of hippocampal and cortex neurons. Moreover, hNTSCs strongly modulated immune status by reducing the number of microglia and the expression of the inflammatory cytokine IL-6 and upregulating autophagic capacity at 7 weeks after transplantation in AD models. Notably, compared with transplantation of hBM-MSCs, transplantation of hNTSCs significantly enhanced performance on the Morris water maze, with an increased level of TIMP2, which is necessary for spatial memory in young mice and neurons; this difference could be explained by the high engraftment of hNTSCs after transplantation. Conclusion The reliable evidence provided by these findings reveals a promising therapeutic effect of hNTSCs and indicates a step forward the clinical application of hNTSCs in patients with AD.

Amyloid-β (Aβ)-peptide production or deposition in the neuropathology of Alzheimer’s disease (AD) was shown to be caused by chronic inflammation that may be induced by infection, but the role of pathogenic-bacteria-related AD-associated Aβ is not yet clearly understood. In this study, we validated the hypothesis that there is a correlation between the Aβ-protein load and bacterial infection and that there are effects of bacteria, Staphylococcus aureus (S. aureus), on the Aβ load in the inflammatory environment of human tonsils. Here, we detected Aβ-peptide deposits in human tonsil tissue as well as tissue similar to tonsilloliths found in the olfactory cleft. Interestingly, we demonstrated for the first time the presence of Staphylococcus aureus (S. aureus) clustered around or embedded in the Aβ deposits. Notably, we showed that treatment with S. aureus upregulated the Aβ-protein load in cultures of human tonsil organoids and brain organoids, showing the new role of S. aureus in Aβ-protein aggregation. These findings suggest that a reservoir of Aβ and pathogenic bacteria may be a possible therapeutic target in human tonsils, supporting the treatment of antibiotics to prevent the deposition of Aβ peptides via the removal of pathogens in the intervention of AD pathogenesis.

Four-dimensional imaging, which indicates imaging in three spatial dimensions as a function of time, provides useful evidence to investigate the interactions of rising bubbles. However, this has been largely unexplored for microbubbles, mostly due to problems associated with strong light scattering and shallow depth of field in optical imaging. Here, tracking x-ray microtomography is used to visualize rising microbubbles in four dimensions. Bubbles are tracked by moving the cell to account for their rise velocity. The sizes, shapes, time-dependent positions, and velocities of individual rising microbubbles are clearly identified, despite substantial overlaps between bubbles in the field of view. Our tracking x-ray microtomography affords opportunities for understanding bubble-bubble (or particle) interactions at microscales – important in various fields such as microfluidics, biomechanics, and floatation. Numerous cell types have shown a remarkable ability to detect and move along gradients in stiffness of an underlying substrate-a process known as durotaxis. The mechanisms underlying durotaxis are still unresolved, but generally believed to involve active sensing and locomotion. Here, we show that simple liquid droplets also undergo durotaxis. By modulating substrate stiffness, we obtain fine control of droplet position on soft, flat substrates. Unlike other control mechanisms, droplet durotaxis works without imposing chemical, thermal, electrical, or topographical gradients. We show that droplet durotaxis can be used to create large-scale droplet patterns and is potentially useful for many applications, such as microfluidics, thermal control, and microfabrication. The X-ray synchrotron is quite different from conventional radiation sources. This technique may expand the capabilities of conventional radiology and be applied in novel manners for special cases. To evaluate the usefulness of X-ray synchrotron radiation systems for real time observations, mouse fetal skeleton development was monitored with a high resolution X-ray synchrotron. A non-monochromatized X-ray synchrotron (white beam, 5C1 beamline) was employed to observe the skeleton of mice under anesthesia at embryonic day (E)12, El4, El5, and El8. At the same time, conventional radiography and mammography were used to compare with X-ray synchrotron. After synchrotron radiation, each mouse was sacrificed and stained with Alizarin red S and Alcian blue to observe bony structures. Synchrotron radiation enabled us to view the mouse fetal skeleton beginning at gestation. Synchrotron radiation systems facilitate real time observations of the fetal skeleton with greater accuracy and magnification compared to mammography and conventional radiography. Our results show that X-ray synchrotron systems can be used to observe the fine structures of internal organs at high magnification. [ABSTRACT FROM AUTHOR]

Colloidal Ag nanoparticles were formed by X-ray irradiation in the presence of a polymer. This new synthesis method is simple, rapid and leads to a high production yield. Compared to the citrate-reduced Ag colloidal, polymer-protected Ag nanoparticles are smaller in size and more stable—and therefore suitable for biomedical application—as verified by TEM observation, XAFS measurement and optical characterization. Ag nanoparticles so produced were also visualized in solution and in real time by a visible light microscope based on dark field light scattering. The color-derived size and distribution of Ag nanoparticles correlates well with the hydrodynamic size data. [ABSTRACT FROM AUTHOR]

Purpose: The purpose of this study is to evaluate the feasibility of phase contrast X-ray microtomography and microradiography, using a polychromatic synchrotron X-ray, for analysis of the mouse lung microstructure. Materials and Methods: Normal mice were used for experiments. Some of the mouse lungs were prepared by the lung fixation-inflation method. The resulting sponge-like inflated lung samples were used for microtomography. The remaining mouse lungs were cut into 10 um sections and were used for microradiography and optical microscopic correlation. The experiments on mouse lung samples were performed at the 7B2 beamline of the Pohang Light Source in Korea. Results: Phase contrast X-ray microtomography of inflated lung samples showed individual alveolar structure on 3-D reconstruction. Phase contrast microradiographs of thin lung samples showed microstructure of lung, such as alveoli and bronchioles, and were well correlated with optical microscopic images. Conclusions: The results indicate that the phase contrast X-ray microtomography and microradiography using polychromatic synchrotron X-ray is feasible for evaluation of microstructure of the lung. Background/aims: There is a need for development of non-invasive methods to improve early diagnosis and screening of suspected malignant lesions. Phase-contrast X-ray microscopy (PCXM) has potential to reveal the structures inside soft tissues, and fine details can be observed without any staining or contrast-enhancing cell preparation. We aimed to investigate the possibility that PCXM can be used to explore the microscopic details of basal cell carcinoma (BCC). Methods: Paraffin blocks of specimens from patients with basal cell carcinoma were cut with 30 μm thickness for PCXM imaging. Experiments were performed at the International Consortium of Phase Contrast Imaging and Radiology (ICPCIR) (7B2) beamline of the Pohang light source in Korea. The PCXM images were achieved by using coherent hard X-rays from a synchrotron source with no monochromatization. Results: We could obtain images with clear edge enhancement by PCXM. The images taken with this technique showed clear anatomic details of organelles in normal skin such as epidermis, dermis and skin appendages. Most of cancerous lesions were clearly differentiated from adjacent normal tissues and the images closely corresponded to those obtained with low-magnification optical microscopy. Conclusion: In this pilot study, we successfully demonstrated that synchrotron PCXM could be used for radiological imaging of BCC with great anatomic details. [ABSTRACT FROM AUTHOR]

In the~ present study the feasibility of applying synchrotron radiation to the morphological study of early-stage lung cancer has been investigated. Lewis lung cancer was implanted and grown in a nude mouse for different periods, and imaged using phase-contrast synchrotron X-rays. Morphological differences were clearly shown between the normal lung and cancerous tissues at this early stage. Irregular and tortuous angiogenesis were found in the periphery region of the developing lung cancer. Results from this study indicate that synchrotron X-rays can be used for imaging cancer development and progression with minimal invasion. [ABSTRACT FROM AUTHOR]

The formation of colloidal unmodified (`naked') gold nanoparticles is investigated by irradiation of a precursor solution with X-rays from a synchrotron source. An interesting morphological evolution as a function of exposure time, from cross-linked network-like structure to individual particles, has been discovered. The particle size decreased with the exposure time and was influenced by the ionic strength of the precursor solution. The authors describe an electroless deposition method for thin films, based on the irradiation by an x-ray beam emitted by a synchrotron source. Specifically, Ni–P films were deposited at room temperature. This synthesis is a unique combination of photochemical and electrochemical processes. The influence of the pH value on the formation and structural properties of the films was examined by various characterization tools including scanning electron microscopy, x-ray diffraction, and x-ray absorption spectroscopy. Real time monitoring of the deposition process by coherent x-ray microscopy reveals that the formation of hydrogen bubbles leads to a self-catalysis effect without a preexisting catalyst. Aim: Having better edge enhancement and penetrating power, refractive index radiology is suitable for the imaging of weakly absorbing objects such as tissue specimens. In this study the potential of refractive index radiology was evaluated for the imaging of renal cell carcinoma (RCC) and prostate cancer (PCA). Methods: Specimens were cut in 3 mm and 4 µm thickness for X-ray radiology and hematoxylin and eosin (HE) staining, respectively. Radiographic images of RCC and PCA were obtained using the synchrotron hard X-rays from the 7B2 beam-line of the Pohang Light Source (PLS). The imaging technique applied was phase-contrast radiology based on the refraction enhancement mechanism. The resulting radiographic images were analyzed in correlation with those of optical microscopy. Results: Using unmonochromatized hard X-rays, it was possible to obtain images with clear edge enhancement and relatively large field of view (6 cm × 6 cm). Even with overlapping signals from thick samples (more than 700-fold thicker than microscopic images), radiographic images clearly showed histological information of organelles in normal kidney such as glomeruli, tubules, and collecting ducts. Histological information of RCC including tumor subtypes and minute changes such as cystic degeneration could be identified without difficulty. The radiographic images of the prostate were comparable with those of low magnification optical microscopy, providing good visualization of normal microstructures such as adenoma, smooth muscle, and normal glands, or differentiation of tiny tumors from surrounding normal tissues. Conclusions: These results suggest the potential of refractive index radiology to provide a new way of imaging biological tissues with low absorption contrast such as RCC and PCA. We evaluated the effect of serum-free and xeno-cultivation (SFXFM) on the characterization, proliferation, and differentiation properties of human nasal stem cells (airway tissue; hTMSCs). hTMSCs were isolated from 10 patients, after which patient samples were separated into two groups, an SFXFM group and a control group. The control group was treated with bovine serum-containing medium. FACS analysis revealed that SFXFM-cultured hTMSCs maintained a characteristic mesenchymal stem cell phenotype. hTMSC proliferation was not influenced by SFXFM. In addition, upregulation of IL-8 and GM-CSF and downregulation of RANTES expression were shown in response to SFXFM. Moreover, two-lineage differentiation properties (osteocyte and adipocyte) of hTMSCs were enhanced under SFXFM. Finally, the genetic stability of SFXFM-cultured hTMSCs was demonstrated by normal karyotype results. SFXFM enables good expansion, multipotentiality, and normal genotype maintenance of MSCs. Moreover, this approach serves as a substitute to conventional media for the cultivation of capable MSCs for upcoming medical applications.

Ionization often changes natural material properties such as wettability; this phenomenon is important for aerosols or tiny droplets in natural and industrial processes. We estimate possible wettability changes by ionization in atmospheric aerosols and cloud droplets by analyzing reliable measurements of charge densities based on a general physical model. This estimation shows that atmospheric aerosols could have charge densities larger than 103 μm-2, indicating significant wettability changes. Our finding would be important to evaluate possible effects of cosmic rays on cloud formation in terms of ionization rates. [ABSTRACT FROM AUTHOR]

Ionization has been proposed to change natural properties of liquids by influencing surface tension; however, microphysical mechanism has not been well established. Our general physical model to link surface tension and ionization rate shows that radiation-induced ionization may significantly lower the surface tension of tiny water droplets (1–100 nm). Our estimation shows that a few electric charges are sufficient to reduce the surface tension, for instance, of a water droplet with 10 nm radius. Ionization-induced surface tension phenomenon may have impact on many industrial and natural situations, for instance, linking a possible microphysical route to cloud formation by cosmic rays. We study degradation behaviors of luminance and voltage in organic light-emitting diodes. We find that normalized luminance and inverse normalized voltage with time, L(t) and V(t)-1, follow the stretched exponential decay. On this basis, we derive a general relation of luminance and voltage with time as L(t)=V(t)-δ(t), where δ(t) indicates a decay exponent, which is attributed to time-dependent space-charge limitation. Here the observation of higher δ(t) at higher initial luminance explains why luminance decay is faster at higher initial luminance. [ABSTRACT FROM AUTHOR]

Micropipe bundling and twisting in SiC crystals was revealed using synchrotron x-ray phase sensitive radiography. The computer simulation of micropipe evolution during the crystal growth suggests that the bundled and twisted micropipes arise under the influence of stress fields from other neighboring micropipes. The annihilation of twisted dipoles is attributed to their transformation into semiloops. Reactions of micropipe coalescence lead to the generation of micropipes and/or the annihilation of initial micropipes, resulting in the decrease in their average density. © 2003 American Institute of Physics. The structural evolution during heteroepitaxial growth of ZnO/sapphire(001) by radio-frequency magnetron sputtering has been studied using real-time synchrotron x-ray scattering. The two-dimensional (2D) ZnO(002) layers grown in the initial stage are highly strained and well aligned to the substrate having a mosaic distribution of 0.01° full width at half maximum (FWHM), in sharp contrast to the reported transition 2D layers grown by molecular-beam epitaxy. With increasing film thickness, the lattice strain is relieved and the poorly aligned (1.25° FWHM) three-dimensional (3D) islands are nucleated on the 2D layers. We report the observation of a strong second-order nonlinear optical response from epitaxial Ba[sub 0.48]Sr[sub 0.52]TiO[sub 3] thin films grown on MgO(001) by magnetron sputtering. The result is unexpected since the crystal structure of Ba[sub 0.48]Sr[sub 0.52]TiO[sub 3] is centrosymmetric under ambient conditions. A second-order nonlinear susceptibility d[sub 33] as large as 11 pm/V at a fundamental wavelength of 1.064 μm is observed in a 300 Å thick film. By studying a series of films with different strains, induced by a film-substrate lattice mismatch, we conclude that the resulting tetragonal distortion, presumably accompanied by a loss of inversion symmetry, is responsible for the second-harmonic generation. © 2000 American Institute of Physics. When a liquid drop impacts a solid surface, air is generally entrapped underneath. Using ultrafast x-ray phase-contrast imaging, we directly visualized the profile of an entrapped air film and its evolution into a bubble during drop impact. We identified a complicated evolution process that consists of three stages: inertial retraction of the air film, contraction of the top air surface into a bubble, and pinch-off of a daughter droplet inside the bubble. Energy transfer during retraction drives the contraction and pinch-off of a daughter droplet. The wettability of the solid surface affects the detachment of the bubble, suggesting a method for bubble elimination in many drop-impact applications. [ABSTRACT FROM AUTHOR]
