Abstract

In BCR-ABL1 lymphoblastic leukemia, treatment heterogeneity to tyrosine kinase inhibitors (TKIs), especially in the absence of kinase domain mutations in BCR-ABL1, is poorly understood. Through deep molecular profiling, we uncovered three transcriptomic subtypes of BCR-ABL1 lymphoblastic leukemia, each representing a maturation arrest at a stage of B-cell progenitor differentiation. An earlier arrest was associated with lineage promiscuity, treatment refractoriness and poor patient outcomes. A later arrest was associated with lineage fidelity, durable leukemia remissions and improved patient outcomes. Each maturation arrest was marked by specific genomic events that control different transition points in B-cell development. Interestingly, these events were absent in BCR-ABL1+ preleukemic stem cells isolated from patients regardless of subtype, which supports that transcriptomic phenotypes are determined downstream of the leukemia-initialing event. Overall, our data indicate that treatment response and TKI efficacy are unexpected outcomes of the differentiation stage at which this leukemia transforms.

4. The short stem GHEs in total hip replacement – experience after 380 implantations

Abstract

Introduction: Standard straight stems have been recognized as a gold standard implant in the field of hip replacement surgery. However, lately uncemented bone-preserving short stems started to gain more and more popularity. This was reflected in the increasing variety of available models. Up till now, short and mid-term results are available.Patients and methods: In 2002, the cementless short stemmed GHEs was introduced. 380 patients were included in our study between 2002 and 2008. Only GHEs short stems were implanted. The clinical and radiological evaluations were performed in the Orthopaedic Department, Leipzig University Hospitals, on the average of 24 months (3 to 60 months) postoperatively.Results: 365 primary implantations and 15 revision implantations were carried out. Average age 60 years. Favourable clinical and radiological outcome was seen in 361/380 patients (95%). Postoperative complications were seen in 19/380 patients (5%): 8 fissures/fractures (2.1%), 5 infections (1.3%), 4 aseptic loosenings (1.1%), 2 dislocations (0.5%).Conclusions: Short stem implants, including our own experience with GHEs model, are satisfying and promising. They represent a valuable supplementation of the treatment modalities in hip replacement surgery. However, long term results are still awaited.

Abstract

KnowWhereGraph is one of the largest fully publicly available geospatial knowledge graphs. It includes data from 30 layers on natural hazards (e.g., hurricanes, wildfires), climate variables (e.g., air temperature, precipitation), soil properties, crop and land-cover types, demographics, and human health, various place and region identifiers, among other themes. These have been leveraged through the graph by a variety of applications to address challenges in food security and agricultural supply chains; sustainability related to soil conservation practices and farm labor; and delivery of emergency humanitarian aid following a disaster. In this paper, we introduce the ontology that acts as the schema for KnowWhereGraph. This broad overview provides insight into the requirements and design specifications for the graph and its schema, including the development methodology (modular ontology modeling) and the resources utilized to implement, materialize, and deploy KnowWhereGraph with its end-user interfaces and public query SPARQL endpoint. Abstract

Wikibase – which is the software underlying Wikidata – is a powerful platform for knowledge graph creation and management. However, it has been developed with a crowd-sourced knowledge graph creation scenario in mind, which in particular means that it has not been designed for use case scenarios in which a tightly controlled high-quality schema, in the form of an ontology, is to be imposed, and indeed, independently developed ontologies do not necessarily map seamlessly to the Wikibase approach. In this paper, we provide the key ingredients needed in order to combine traditional ontology modeling with use of the Wikibase platform, namely a set of axiom patterns that bridge the paradigm gap, together with usage instructions and a worked example for historical data.
