The Lucaogou Formation, a typical fine-grained mixed formation in the Jimusaer Sag of the Junggar Basin, exhibits considerable potential for hydrocarbon exploration. Accurate brittle prediction is a crucial factor in determining hydraulic fracturing effectiveness. However, the area features complex lithological characteristics, including carbonate rocks, clastic rocks, volcanic rocks, and gypsum interbeds, along with thin layering and sporadic sweet spots. Traditional prediction methods offer limited resolution and there is an urgent need for a seismic brittle prediction method tailored to this complex geological environment. This paper presents a multi-mineral composition equivalent model for complex lithologies that enables the accurate calculation of Vp and Vs These ratios serve as the foundation for pre-stack elastic parameter predictions, which include Poisson’s ratio and Young’s modulus. By comparing the predicted parameters with well-logging measurements, the prediction accuracy is improved to 82%, with particularly high conformity in intervals characterized by high organic matter and clay content. Additionally, a three-dimensional brittle modeling approach reveals that the brittleness of the reservoir exceeds that of the surrounding rock, showing a gradual improvement in brittleness with increasing burial depth from southeast to northwest. The central area exhibits relatively good brittleness, with a stable, blocky distribution pattern.

Simple Summary: Different culturing modes have adopted different living spaces, foods, shelter, substrates, and surroundings for and human actions regarding animals. The Chinese soft-shelled turtle (Pelodiscus sinensis) is one of most economically important turtles, and healthier culturing modes should be applied for high-quality aquatic products to remit serious environmental and health problems. The differences in growth performance, physiological indices, and related gene expression were compared between two culturing modes in the present study. The results indicated that rice–turtle-co-cultured turtles with limited natural foods grew slower than pond-cultured turtles fed with commercial feeds. The creatinine, uric acid, blood urea nitrogen, alkaline phosphatase, acid phosphatase, glutamic oxaloacetic transaminase, and catalase were higher in co-cultured turtles than pond-cultured turtles. More differentially expressed genes were related to environmental information processing, metabolism, and human diseases when comparing pond-cultured turtles and co-cultured turtles. P. sinensis can adapt to different culture modes by adjusting their growth and physiology over a short culture period, and rice–turtle co-culturing proves to be a healthier culturing mode for turtles within the proper culturing strategies. The Chinese soft-shelled turtle (Pelodiscus sinensis) is an important freshwater aquaculture turtle due to its taste and nutritional and medicinal value. More ecological culturing modes, such as rice–turtle co-culture, should be developed to meet the ecological benefit demand. We compared growth, physiological parameters, and transcriptome data to detect the physiological responses and regulatory mechanisms of pond-cultured turtles as compared to co-cultured turtles. The co-cultured turtles grew slower than pond-cultured turtles. The gonadosomatic index of co-cultured male turtles was lower than that of pond-cultured male turtles, and both the mesenteric fat index and limb fat index were lower in co-cultured turtles than in pond-cultured turtles (p < 0.05). The blood GLU of the co-cultured turtles was significantly lower than the GLU of the pond-cultured turtles (p < 0.05), while the values of CRE, UA, BUN, AKP, ACP, GOT, and CAT were higher in the co-cultured turtles than in the pond-cultured turtles (p < 0.05). In total, 246 and 598 differentially expressed genes (DEGs) were identified in the brain and gut from turtles cultured in the two different modes, respectively. More DEGs were related to environmental information processing, metabolism, and human diseases. In the brain, the top enriched pathways of DEGs included the longevity regulating pathway, glycerolipid metabolism, cytokine–cytokine receptor interaction, Toll-like receptor signaling pathway, and PI3K-Akt signaling pathway, while in the gut, the top enriched pathways of DEGs included the cell cycle, DNA replication, cellular senescence, and p53 signaling pathway. Reactivating oil and gas wells, increasing oil and gas production, and improving recovery provide more opportunities for energy supply especially in the extraction of unconventional oil and gas reservoirs. Due to changes caused by well completion and production in pore pressure around oil and gas wells, subsequently leading to changes in ground stress, and the presence of natural and induced fractures in the reservoir, the process of refracturing is highly complex. This complexity is particularly pronounced in shale oil reservoirs with developed weak layer structures. Through true triaxial hydraulic fracturing experiments on Jimsar shale and utilizing micro-CT to characterize fractures, this study investigates the mechanisms and patterns of refracturing. The research indicates: (1) natural fractures and the stress states in the rock are the primary influencing factors in the fracture propagation. Because natural fractures are widely developed in Jimsar shale, natural fractures are the main influencing factors of hydraulic fracturing, especially in refracturing, the existing fractures have a greater impact on the propagation of secondary fracturing fractures. (2) Successful sealing of existing fractures using temporary blocking agents is crucial for initiating new fractures in refracturing. Traditional methods of plugging the seam at the root of existing fractures are ineffective, whereas extensive injection of blocking agents, forming large "sheet-like" blocking bodies in old fractures, yields better sealing effects, promoting the initiation of new fractures. (3) Moderately increasing the pumping rate and viscosity of fracturing fluid is advantageous in forming "sheet-like" temporary blocking bodies, enhancing the complexity of the network of new fractures in refracturing. (4) When there is a high horizontal stress difference, after sealing old fractures, the secondary hydraulic fractures initiate parallel to and extend from the old fractures. In cases of low horizontal stress difference, the complexity of secondary hydraulic fractures increases. When the horizontal stress changes direction, the secondary hydraulic fractures also change direction. It is recommended to use high-viscosity fracturing fluid and moderately increase the pumping rate, injecting blocking agents to seal old fractures, thereby enhancing the complexity of the network of refracturing. Rationale: Currently, research on oligosaccharides primarily focuses on the physiological activity and function, with a few studies elaborating on the spatial distribution characterization and variation in the processing of Rehmannia glutinosa Libosch. Thus, imaging the spatial distributions and dynamic changes in oligosaccharides during the steaming process is significant for characterizing the metabolic networks of R. glutinosa. It will be beneficial to characterize the impact of steaming on the active ingredients and distribution patterns in different parts of the plant. Methods: A highly sensitive matrix‐assisted laser desorption/ionization mass spectrometry image (MALDI‐MSI) method was used to visualize the spatial distribution of oligosaccharides in processed R. glutinosa. Furthermore, machine learning was used to distinguish the processed R. glutinosa samples obtained under different steaming conditions. Results: Imaging results showed that the oligosaccharides in the fresh R. glutinosa were mainly distributed in the cortex and xylem. As steaming progressed, the tetra‐ and pentasaccharides were hydrolyzed and diffused gradually into the tissue section. MALDI‐MS profiling combined with machine learning was used to identify the processed R. glutinosa samples accurately at different steaming intervals. Eight algorithms were used to build classification machine learning models, which were evaluated for accuracy, precision, recall, and F1 score. The linear discriminant analysis and random forest models performed the best, with prediction accuracies of 0.98 and 0.97, respectively, and thus can be considered for identifying the steaming durations of R. glutinosa. Conclusions: MALDI‐MSI combined with machine learning can be used to visualize the distribution of oligosaccharides and identify the processed samples after steaming for different durations. This can enhance our understanding of the metabolic changes that occur during the steaming process of R. glutinosa; meanwhile, it is expected to provide a theoretical reference for the standardization and modernization of processing in the field of medicinal plants. We conducted an 8-week feeding trial to investigate the effect of mixed functional oligosaccharides (chitosan oligosaccharide, fructooligosaccharide, and xylooligosaccharide) extracted from wheat straw on the growth, feeding, physiology, histology, muscle texture, and gut microbiota of Micropterus salmoides. Six diets were formulated by incrementally adding mixed functional oligosaccharides (0.1, 0.5, 1.0, 1.5, and 2.0 mg/kg) to the control diet. 30 individuals with initial body weight of 25.1 ± 3.5 g were randomly allocated to 6 triplicate tanks and fed to apparent satiation twice daily. The fish fed 1.0 mg/kg additives displayed maximum growth, and the feed conversion ratio decreased with the increase in additives from 0 to 1.0 mg/kg (P < 0.05) but did not decline further with the addition of more than 1.0 mg/kg (P > 0.05). The villus height and width were significantly higher in the supplementation groups than in the control group, while the gut and liver structures presented abnormalities with excessive supplementation above 1.5 mg/kg. There were significant differences in muscle texture indices for M. salmoides over the fed additive gradient, and the hardness, gumminess, and chewiness were highest in the 1.0 mg/kg group. High oligosaccharide levels, such as 2.0 mg/kg, decreased the LZM level, while there were no significant differences in the SOD and MDA levels. Gut microbiome analysis revealed no significant differences in richness and diversity for groups fed the functional oligosaccharide gradient; however, the PCoA results showed that the microbial community composition changed markedly in response to different addition levels, and the 0.5 and 1.0 mg/kg supplementation groups were far apart from the lower and higher supplementation groups. The relative abundance of Proteobacteria was lower in the 0.5 and 1.0 mg/kg addition groups, while that of the phyla Fusobacteria and Firmicutes were higher in these two groups. Functional classification showed that microbes related to carbohydrate metabolism were more abundant in the 0.5 and 1.5 mg/kg groups than in the other groups. [ABSTRACT FROM AUTHOR]

Ophiopogonis Radix, also known as "Maidong" (MD) in China, is frequently sulfur-fumigated (SF) in the pretreatment process of MD to improve the appearance and facilitate preservation. However, the process leads to changes in chemical composition, so it is essential to develop an approach to identify the chemical characteristics between nonfumigated and sulfur-fumigated products. This paper provided a practical method based on UPLC-QTOF-MS combined Global Natural Products Social Molecular Networking (GNPS) with multivariate statistical analysis for the characterization and discrimination of MD with different levels of sulfur fumigation, high concentration sulfur fumigation (HS), low concentration sulfur fumigation (LS) and without sulfur fumigation (WS). First, a number of 98 compounds were identified in those MD samples. Additionally, the results of Principal component analysis (PCA) and Orthogonal partial least-squares-discriminant analysis (OPLS-DA) demonstrated that there were significant chemical differences in the chemical composition of MD with different degrees of SF. Finally, fourteen and sixteen chemical markers were identified upon the comparison between HS and WS, LS and WS, respectively. Overall, these results can be able to discriminate MD with different levels of SF as well as establish a solid foundation for further quality control and pharmacological research. Leukemia stem cells (LSCs) constitute the critical barrier to the cure of acute myeloid leukemia (AML) due to their chemoresistance and immune evasion property. Herein, the role of anlotinib, a multiple tyrosine kinase inhibitor, in killing LSCs and regulating chemoresistance and immune evasion was explored. Anlotinib treatment induced apoptosis of LSC-like cells as well as primary AML LSCs, while sparing the normal mononuclear cells in vitro. Moreover, anlotinib could impair the regeneration capacity of LSCs in the patient-derived leukemia xenograft mouse model. Mechanistically, anlotinib inhibited phosphorylation of c-kit, JAK2/STAT3, and STAT5, and downregulated STAT3 and STAT5 expression. In addition, anlotinib downregulated the anti-apoptotic proteins Bcl-2 and Bcl-xL, and upregulated Bax, thereby enhancing the sensitivity of LSCs to idarubicin in vitro. Intriguingly, anlotinib could also partially rescue the interferon-g production of T cells cocultured with LSCs by downregulating PD-L1 expression. In conclusion, anlotinib showed anti-LSC activity and the potential to enhance the sensitivity to idarubicin and inhibit the immunosuppressive feature of LSCs via JAK2/STAT signaling pathway downregulation in the preclinical study. Pearl color is an important factor influencing pearl value, and is affected by the nacre color of the shell in Hyriopsis cumingii. Coproporphyrinogen-III oxidase (CPOX) is a key enzyme in porphyrin synthesis, and porphyrins are involved in color formation in different organisms, including in the nacre color of mussels. In this study, a CPOX gene (HcCPOX) was identified from H. cumingii, and its amino acid sequence was found to contain a coprogen-oxidase domain. HcCPOX mRNA was expressed widely in the tissues of white and purple mussels, and the highest expression was found in the gill, followed by the fringe mantle. The expression of HcCPOX in all tissues of purple mussels (except in the middle mantle) was higher than that of white mussels. Strong hybridization signals for HcCPOX were observed in the dorsal epithelial cells of the outer fold of the mantle. The activity of CPOX in the gill, fringe mantle, and foot of purple mussels was significantly higher than that in white mussels. Moreover, the expression of HcCPOX and CPOX activity were decreased in RNA interference experiments. The red swamp crayfish, Procambarus clarkii (Girard, 1852), is a paradox in China because it has developed as a popular aquaculture species while becoming a notorious invasive species. We investigated the population genetic diversity and population dynamics of wild Chinese populations of P. clarkii based on proPO fragments and the morphological traits across these populations. The results showed that the populations of P. clarkii exhibit high genetic diversity and significant population structure, inferring that the crayfish was artificially transferred rapidly among different locations for different reasons. Results also revealed distinct phenotypic differentiation, with individuals of some populations having a shorter pleon and wider carapace width, whereas other populations have a small length to width ratio. This study provided comprehensive insights into the population dynamics and morphological variations of P. clarkii , which contribute to understanding the rapid evolution of phenotypic traits after its successful invasion. Glaucocalyxin A, a novel potent negative Akt regulator, is a major active constituent of Rabdosia japonica. A simple, specific and sensitive ultra-high pressure liquid chromatography tandem mass spectrometry (UHPLC–MS/MS) method was developed and validated for the quantification of glaucocalyxin A in rat plasma, lung and brain tissues after intravenous administration. Sample preparation was carried out through a simple liqiud–liquid extraction with ethyl acetate using bullatine A as internal standard. The chromatographic separation was achieved by using an Agilent ZORBAX Eclipse Plus C18 column (3.0 mm × 100 mm, 1.8 μm) with a mobile phase of acetonitrile and water containing 0.1% formic acid in a gradient elution. Mass spectrometry analysis was conducted in positive ionization mode with multiple reaction monitoring transitions at m/z 333.2 → 157.1 for glaucocalyxin A and m/z 344.2 → 128.1 for IS. Calibration curves were linear over the ranges of 20.0–4000 ng/mL for both plasma and tissue samples (r 2 > 0.99). The Lower limit of quantification (LLOQ) was 0.284 ng/mL. The intra-day and inter-day precision relative standard deviation (RSD%) were <14.9%, while the accuracy ranged from −12.5 to 17.0% for LLOQ and quality control samples. This paper presents a quadruple-porosity model for multi-stage fractured horizontal well (MFHW) with finite conductive hydraulic fractures in shale gas reservoirs. In this model, free gas, adsorbed gas and dissolved gas co-exist and gas migration in shale incorporates diffusion in kerogen bulk, desorption from the surface of organics and clays, slippage flow in porous kerogen and inorganic matrix, and Darcy flow in natural fractures. Bi-Langmuir theory was introduced to describe gas desorption from the surface of clays and organics. Continuous line source function, Laplace transform and numerical discrete method were employed to solve this new model. Gauss-Jordan elimination method and Stehfest numerical inversion algorithm were applied to calculate the pressure and production responses. Type curves were plotted and flow regimes were identified. Sensitivity analysis of solubility coefficient, diffusion coefficient, inter-porosity coefficient, TOC, clays content, hydraulic fracture conductivity and permeability correction coefficient was performed. Finally, the proposed model was validated by fitting actual production data of a field case and comparing with other models. The matching results showed that quadruple porosity model considering dissolved gas was closer to the actual situation than trilinear-flow model and dual-porosity radial-flow model. To sum up, this presented model considering some key mechanisms further expands the transient pressure models for MFHW in shale gas reservoirs and it can be utilized to analyze well performance in the production life of gas wells. In the present study, ELMAN artificial neural network model was developed to predict the change of NH3-N in aquaculture wafer. The indexes including teed ration , dissolved oxygen in wafer , wafer temperature , air temperature , wafer turbidity , rainfall were recorded and chosen as the input variables , while the NH3-N content in the corresponding pond was chosen as output variable. The above data were collected evetyday from June to October in 2014 and were used to develop model in this test , and the data collected in November of 2014 were chosen to evaluate the developed model. The results showed that the changing trend of NH3-N in aquaculture wafer could be simulated well by the model , the predictive absolute error mean was 0.016 mg/L , and Nash-Sufcliffe efficiency coefficient was 0.74. The prediction model based on ELMAN neural network had a strong ability to describe the nonlinear dynamic changes of NH3-N content in aquaculture wafer , and if showed the good adaptability and accuracy in practical application. Triple Negative Breast Cancer (TNBC) is the malignant tumor with the fastest progression rate in breast cancer. LncRNAs are widely involved in various biological characteristics of tumor. The purpose of this study was to mine LncRNAs that can be used to diagnose and evaluate the prognosis of TNBC. Base on TCGA dataset, we used three R language packages to analyze the differentially expressed (DE) lncRNAs in TNBC. Survival analysis and ROC curve analysis were conducted to estimate the potential diagnostic and prognostic value of LncRNAs for TNBC. Furthermore, CCK-8 and Transwell assays were used to assess the effects of LncRNA on MDA-MB-231 cells proliferation and migration. Additionally, targets mRNAs of candidate LncRNA were predicted by co-expression analysis and multiple target gene prediction databases, then KEGG pathway and GO analysis were conducted using DAVID online tool. 6165 DERNAs and 1258 DElncRNAs were obtained. 40 LncRNAs were significantly correlated with the survival time of TNBC patients. Among them, HAGLROS has the highest HR value. ROC curve analysis also showed that HAGLROS had high sensitivity and specificity. Further in vitro experiments showed that downregulation of HAGLROS inhibited the proliferation and migration of MDA-MB-231 cells. Moreover, by conducting bioinformatics analysis, we found that these target genes of HAGLROS were involved in regulating five signaling pathways. Mechanistic investigations demonstrated that HAGLROS might regulate the expression of PAX5 through miR-330–5p, the effects of miR-330–5p in MDA-MB-231 cells were also analyzed.Conclusion: Our results showed that HAGLROS was significantly overexpressed in TNBC, and high HAGLROS expression predicted poor overall survival. A new spectrofluorimetric method was developed for the determination of trace amounts of histamine in human urine and serum samples. In NaAc-HAc buffer solution of pH 4.0, histamine can react with the acetylacetone-formaldehyde system to produce a fluorescent derivative which emits yellow-green fluorescence at 476 nm, according to the Hantzsch reaction, and the enhanced fluorescence intensity is in proportion to the concentration of histamine. Optimum conditions for the determination of histamine were also investigated. The dynamic range and detection limit for the determination of histamine is 5.96 × 10-8-1.50 × 10-5 mol/L and 4.35 × 10-8mol/L, respectively. This method is practical and can be successfully applied to determination of histamine in human urine and serum samples. A proposal of the reaction pathway is suggested. In this work, the heat transfer characteristics of supercritical pressure CO2 in vertical heating tube with 10 mm inner diameter under high mass flux were investigated by using an SST k-ω turbulent model. The influences of inlet temperature, heat flux, mass flux, buoyancy and flow acceleration on the heat transfer of supercritical pressure CO2 were discussed. Our results show that the buoyancy and flow acceleration effect based on single phase fluid assumption fail to explain the current simulation results. Here, supercritical pseudo-boiling theory is introduced to deal with heat transfer of scCO2. scCO2 is treated to have a heterogeneous structure consisting of vapor-like fluid and liquid-like fluid. A physical model of scCO2 heat transfer in vertical heating tube was established containing a gas-like layer near the wall and a liquid-like fluid layer. Detailed distribution of thermophysical properties and turbulence in radial direction show that scCO2 heat transfer is greatly affected by the thickness of gas-like film, thermal properties of gas-like film and turbulent kinetic energy in the near-wall region. Buoyancy parameters Bu < 10−5, Bu* < 5.6 × 10−7 and flow acceleration parameter Kv < 3 × 10−6 in this paper, which indicate that buoyancy effect and flow acceleration effect has no influence on heat transfer of scCO2 under high mass fluxes. A simple sensitive and quick assay to simultaneously determine Aloe-emodin (AE) and Rhein (RH) has been described based on their natural fluorescence. Due to their similar molecule structures, it is difficult to analysis and determine their contents by conventional fluorometry. Overlapping of fluorescence spectra was resolved by using synchronous fluorometry. The synchronous spectrum, maintaining a constant difference of Δλ=25 nm between the emission and excitation wavelengths, has been selected as optimum to perform the determination. This method was applied to the simultaneous determination of AE and RH in simulated mixtures. Analytical recoveries range from 94.5 to 105.5% for AE and from 98.7 to 107.5% for RH. Pearl color is closely related to the nacre color of shell in Hyriopsis cumingii , and pearl color has a huge impact on its price. The nacre is an important part of the shell, and studies have suggested that mantle exosomes participated in shell formation. Exosomes contain many different proteins that are involved in different biological processes. In this study, exosomes were extracted from mantles of mussels with different nacre color. TMT quantitative proteome sequencing analysis was performed on purple and white mussel mantle exosomes, and 4861 proteins were obtained. Based on the standard of (|log2 (Fold change)| ≥ 1.2 or ≤ 0.83 and p -value ≤ 0.05), a total of 758 differentially expressed proteins were found. Some proteins involved in shell and nacre color formation were predicted with the proteins annotate, GO classification system. Moreover, 14 differentially expressed proteins (including eight up-regulated proteins and six down-regulated proteins) were validated using parallel reaction monitoring (PRM) assays. Overall, this information will be useful to improve our understanding of the molecular mechanisms of shell and nacre color formation in H. cumingii. Mantle epidermis absorb Ca2+, metal ions and nutriments from the living water environment, which are transported by blood or other mechanisms into the mantle and secreted by epithelial cells between the hemolymph and mantle cavity. Patients with myelodysplastic syndromes (MDS) who resist or fail to respond to hypomethylating agents (HMAs) show very poor outcomes and have no effective treatment strategies. Therefore, new therapeutic approaches are urgently needed for MDS patients harboring adverse prognostic factors. Repurposing disulfiram (DSF), an alcohol-abuse drug, with or without Copper (Cu) has attracted considerable attentions as a candidate anti-tumor therapy in diverse malignancies. However, the effect of DSF in the presence or absence of Cu on MDS has not been reported yet. In this study, we found that monotherapy with DSF showed mild cytotoxic effects on MDS preclinical models. However, the anti-tumor activity of DSF was significantly enhanced in the presence of Cu in MDS in vitro and in vivo with minimal safety profiles. DSF/Cu combination blocked MDS cell cycle progression at the G0/G1 phase, accompanied by reduction of the S phase. Accordingly, co-treatment with DSF and Cu downregulated the expression of Cyclin D1 and Cyclin A2, whereas this combination upregulated the level of P21 and P27. Mechanistically, the anti-MDS effectiveness of DSF/Cu was potentially associated with activation of the ER stress-related Bip pathway and inactivation of the Akt pathway. The Chinese soft-shelled turtle ( Pelodiscus sinensis ) is an important freshwater aquaculture turtle due to its taste and nutritional and medicinal value. More ecological culturing modes, such as rice-turtle co-culture, should be developed to meet the ecological benefit demand. We compared growth, physiological parameters, and transcriptome data to detect the physiological responses and regulatory mechanisms of pond-cultured turtles as compared to co-cultured turtles. The co-cultured turtles grew slower than pond-cultured turtles. The gonadosomatic index of co-cultured male turtles was lower than that of pond-cultured male turtles, and both the mesenteric fat index and limb fat index were lower in co-cultured turtles than in pond-cultured turtles ( p < 0.05). The blood GLU of the co-cultured turtles was significantly lower than the GLU of the pond-cultured turtles ( p < 0.05), while the values of CRE, UA, BUN, AKP, ACP, GOT, and CAT were higher in the co-cultured turtles than in the pond-cultured turtles ( p < 0.05). In total, 246 and 598 differentially expressed genes (DEGs) were identified in the brain and gut from turtles cultured in the two different modes, respectively. More DEGs were related to environmental information processing, metabolism, and human diseases. In the brain, the top enriched pathways of DEGs included the longevity regulating pathway, glycerolipid metabolism, cytokine-cytokine receptor interaction, Toll-like receptor signaling pathway, and PI3K-Akt signaling pathway, while in the gut, the top enriched pathways of DEGs included the cell cycle, DNA replication, cellular senescence, and p53 signaling pathway. Employing functional and structured thin films on fiber optic sensors has tremendously improved capabilities in humidity sensing applications. In this paper, we demonstrate fabrication of a fiber optic evanescent wave humidity sensor based on S i O 2 /porous polymethyl methacrylate (PMMA) thin films. With the exposure to moisture, S i O 2 /porous PMMA thin films absorb water molecules. The refractive index and absorption coefficient of thin films change with ambient humidity, resulting in modulation of the light intensity transmitted in fiber. A good linearity is determined between the logarithm of output light intensity and relative humidity (RH). An optimal average sensitivity of 188.3 lux/%RH is achieved with an increase of 11.7 fold in the RH range of 5% to 95%. The response and recovery times are 8 s and 23 s, respectively. Furthermore, the sensor exhibits low hysteresis, and excellent stability and repeatability.

Background: The leaves of L. japonica (LLJ) are widely used as medicine in China. It is rich in caffeoylquinic acids, flavonoids and iridoid glycosides and has strong reducing capacities. Therefore, it can be used as a green material to synthesize silver nanoparticles., Methods: LLJ was used as a reducing agent to produce the LLJ-mediated silver nanoparticles (LLJ-AgNPs). The structure and physicochemical properties of LLJ-AgNPs were characterized by ultraviolet spectroscopy (UV-Vis), scanning electron microscopy (SEM), transmission electron microscopy (TEM), Fourier transform infrared spectroscopy (FTIR), and x-ray powder diffraction (XRD). Antioxidant activity of LLJ-AgNPs was determined by 1,1-diphenyl-2-picrylhydrazyl (DPPH) scavenging. Antibacterial activity was determined by 96 well plates (AGAR) gradient dilution, while the anticancer potential was determined by MTT assay., Results: The results showed LLJ-AgNPs had a spherical structure with the maximum UV-Vis absorption at 400 nm. In addition, LLJ-AgNPs exhibited excellent antioxidant properties, where the free radical scavenging rate of LLJ-AgNPs was increased from 39% to 92% at concentrations from 0.25 to 1.0 mg/mL. Moreover, LLJ-AgNPs displayed excellent antibacterial properties against E. coli and Salmonella at room temperature, with minimum inhibitory values of 10 -6 and 10 -5 g/L, respectively. In addition, the synthetic LLJ-AgNPs exhibited a better inhibition effect in the proliferation of cancer cells (HepG2, MDA-MB -231, and Hela cells)., Conclusion: The present study provides a green approach to synthesize LLJ-AgNPs. A fiber optic humidity sensor based on polyvinyl alcohol (PVA)/Tween 20 film has been fabricated by modulating the intensity of light transmitted in optical fiber. PVA/Tween 20 film was used as the cladding and humidity-sensitive material of optical fiber. The logarithmic of output light intensity exhibited a linear increase with the increase of humidity (22%-82%RH). With the addition of Tween 20 in the formation of film, average sensitivity increased by 13-fold. Fast equilibrium on adsorption and desorption of water molecules were also achieved on the film. The response and recovery times were determined to be 11 s and 9 s, respectively. Moreover, the sensor possesses good repeatability. The sensing mechanism was probably based on the swelling of PVA after adsorbing water molecules, which affected scattering of evanescent waves in the cladding. The output light intensity varied with the decay of evanescent waves.

Bi 5 O 7 I/g-C 3 N 4 p-n junctioned photocatalysts were synthesized by alcohol-heating and calcination in air. The structures, morphologies and optical properties of as-prepared samples were characterized by X-ray diffraction (XRD), scanning electron microscopy (SEM), transmission electron microscopy (TEM), UV-Vis diffuse reflectance spectroscopy (DRS). Photocatalytic activity of the heterojunctioned composites were evaluated by degradation of Rhodamine B (RhB) and tetracycline hydrochloride (TCH) under visible light illumination. The results indicated that the composites exhibited superior efficiencies for photodegradation of RhB and TCH in comparison with pure BiOI, Bi 5 O 7 I and g-C 3 N 4 . An effective built-in electric field was formed by the interface between p-type Bi 5 O 7 I and n-type g-C 3 N 4 , which promoted the efficient separation of photoinduced electron-hole pairs. In addition, 8% Bi 5 O 7 I/g-C 3 N 4 composite showed excellent photostability in a five-cycle photocatalytic experiment. Emerging evidence has illustrated that long noncoding RNA 01234 (LINC01234) has played a pivotal role in the development and progression of human cancer. The regulatory role and underlying mechanisms of LINC01234 in triple-negative breast cancer (TNBC) remains unknown. In this study, we analyzed the expression level of LINC01234 in several breast cancer cell lines. CCK-8, EdU, flow cytometry analysis, wound healing assay, and transwell assay were carried out to investigate the effect of LINC01234 on tumor proliferation, apoptosis, and migration. Bioinformatic analysis and luciferase reporter assays were performed to confirm the molecular binding. We found that LINC01234 was dramatically upregulated in breast cancer cell lines, especially in TNBC. The loss and gain-of functional experiments revealed that LINC01234 significantly promoted proliferation, migration, and suppressed cell apoptosis of MDA-MB-231 cells in vitro and inhibited tumorigenesis in vivo . Mechanistic investigations demonstrated that LINC01234 might act as a competing endogenous RNA (ceRNA) for miR-429 to regulate the SYNJ1 expression. The effects of miR-429 and SYNJ1 in MDA-MB-231 cells were also analyzed. A S i O 2 / T i O 2 bilayer thin-film-based fiber optic humidity sensor was fabricated via a modified dip coating process with enhanced sensitivity. S i O 2 film was coated on the surface of the fiber core, followed by deposition of the T i O 2 layer on S i O 2 . The relative humidity (RH) is measured by modulation in intensity of the transmitted laser at room temperature. The optical fiber humidity sensor based on S i O 2 / T i O 2 film shows two-segmented linearity in measurement with sensitivities of 5.35 and 1.94 µW/% RH at 15%-50% RH and 50%-95% RH, respectively. The response time and recovery time are 25 s and 50 s, respectively. To our knowledge, the superior response time and recovery time of the sensor in our study were achieved over those fiber optic humidity sensors reported with modulation in intensity. Furthermore, this fiber optic humidity sensor has a good reproducibility and long-term stability. The sensing mechanism is attributed to effects of moisture on the refractive index and the light absorption coefficient of S i O 2 film and modulation in the transmission characteristic of evanescent waves in the optical fiber.

Purpose: Breast cancer is the most frequently diagnosed malignancy in women worldwide. MicroRNAs (miRNAs) are thought to serve as potential biomarkers in various cancers, including breast cancer., Methods: We evaluated the miRNA expression profiles in 1,083 breast cancer samples and 104 normal breast tissues from The Cancer Genome Atlas database. We used the edgeR package of R software to analyze the differentially expressed miRNAs in normal and cancer tissues, and screened survival-related miRNAs by Kaplan-Meier analysis. A receiver operating characteristic curve was generated to evaluate the accuracy of these miRNAs as molecular markers for breast cancer diagnosis. Furthermore, the functional role of these miRNAs was verified using cell experiments. Targets of candidate miRNAs were predicted using 9 online databases, and Gene Ontology (GO) functional annotation and pathway analyses were conducted using Database for Annotation, Visualization and Integrated Discovery online tool., Results: A total of 68 miRNAs showed significantly different expression patterns between the groups ( p < 0.001), and 13 of these miRNAs were significantly associated with poor survival ( p < 0.05). Three miRNAs with high specificity and sensitivity, namely, miR-148b-3p, miR-190b, and miR-429, were selected. In vitro experiments showed that the overexpression of these 3 miRNAs significantly promoted the proliferation and migration of MDA-MB-468 and T47D cells and reduced the apoptosis of T47D cells. GO and pathway enrichment analyses revealed that the targets of these dysregulated miRNAs were involved in many critical cancer-related biological processes and pathways., Conclusion: The miR-148b-3p, miR-190b, and miR-429 may serve as potential diagnostic and prognostic markers for breast cancer. A new spectrofluorimetric method was developed for the determination of trace amounts of histamine in human urine and serum samples. In NaAc-HAc buffer solution of pH 4.0, histamine can react with the acetylacetone-formaldehyde system to produce a fluorescent derivative which emits yellow-green fluorescence at 476 nm, according to the Hantzsch reaction, and the enhanced fluorescence intensity is in proportion to the concentration of histamine. Optimum conditions for the determination of histamine were also investigated. The dynamic range and detection limit for the determination of histamine is 5.96 x 10(-8)-1.50 x 10(-5 )mol/L and 4.35 x 10(-8)mol/L, respectively. This method is practical and can be successfully applied to determination of histamine in human urine and serum samples. A proposal of the reaction pathway is suggested.
