Conventional commonly used titanium stamping techniques like hot forming and superplastic forming each have distinct disadvantages. Hot forming usually leads to limited formability and significant springback, whilst superplastic forming often results in long cycle times, high energy use, and costly tooling. A novel, energy-efficient, and cost-effective Heat Stamping process emerges as a promising solution. Two types of Ti6Al4V components, cup-shaped and U-shaped, were produced using the novel Heat Stamping process, and their properties were examined. A defect-free cup-shaped component achieving a considerably high draw ratio of up to 1.8, was successfully fabricated. A moderate enhancement in the hardness of the component indicated a superior post-form strength achieved by Heat Stamping. Further analysis of the U-shaped components shows that the one produced using the novel method demonstrated a notable reduction in springback angle, from 4.8° to 0.7°, highlighting the potential of H eat Stamping in achieving high shape accuracy for near-net-shape forming.

Immune checkpoint blockade (ICB) immunotherapy remains hampered by insufficient immunogenicity and a high‐lactate immunosuppressive tumor microenvironment (TME). Herein, a nanobody‐engineered NIR‐II nanoadjuvant with targeting metabolic reprogramming capability is constructed for potentiating NIR‐II photothermal‐ferroptosis immunotherapy. Specifically, the nanoadjuvant (2DG@FS‐Nb) is prepared by metallic iron ion‐mediated coordination self‐assembly of D‐A‐D type NIR‐II molecules and loading of glycolysis inhibitor, 2‐deoxy‐D‐glucose (2DG), followed by modification with aPD‐L1 nanobody (Nb), which can effectively target the immunosuppressive TME and trigger in situ immune checkpoint blockade. The nanoadjuvants responsively release therapeutic components in the acidic TME, enabling the precise tumor location by NIR‐II fluorescence/photoacoustic imaging while initiating NIR‐II photothermal‐ferroptosis therapy. The remarkable NIR‐II photothermal efficiency and elevated glutathione (GSH) depletion further sensitize ferroptosis to induce severe lipid peroxidation, provoking robust immunogenic cell death (ICD) to trigger anti‐tumor immune response. Importantly, the released 2DG markedly inhibits lactate generation through glycolysis obstruction. Decreased lactate efflux remodels the immunosuppressive TME by suppressing M2 macrophage proliferation and downregulating regulatory T cell levels. This work provides a new paradigm for the integration of NIR‐II phototheranostics and lactate metabolism regulation into a single nanoplatform for amplified anti‐tumor immunotherapy combined with ICB therapy. The objective of this study was to develop a food 3D printing gel and investigate the effects of whey protein isolate (WPI), sodium alginate (SA), and water-bath heating time on the 3D printing performance of the gel. Initially, the influence of these three factors on the rheological properties of the gel was examined to determine the suitable formulation ranges for 3D printing. Subsequently, the formulation was optimized using response surface methodology, and texture analysis, scanning electron microscopy (SEM), and Fourier-transform infrared (FTIR) spectroscopy were conducted. The rheological results indicated that gels with WPI concentrations of 6–7 g, SA concentrations of 0.8–1.2 g, and water-bath heating times of 10–12 min exhibited lower yield stress and better self-supporting properties. The optimized formulation, determined through response surface methodology, consisted of 1.2 g SA, 6.5 g WPI, and a heating time of 12 min. This optimized formulation demonstrated enhanced extrusion capability and superior printing performance. SEM analysis revealed that the optimized gel possessed good mechanical strength, and FTIR spectroscopy confirmed the successful composite formation of the gel. Chimeric antigen receptor (CAR) T cell therapy shows promise in treating malignant tumors. However, the use of human epidermal growth factor receptor-2 (HER2) CAR-T cells carries the risk of severe toxicity, including cytokine release syndrome, due to their "on-target off-tumor" recognition of HER2. Enhancing the quality and functionality of HER2 CARs could greatly improve the therapeutic potential of CAR-T cells. In this study, we developed a novel anti-HER2 monoclonal antibody, Ab8, which targets domain III of HER2, distinct from the domain IV recognition of trastuzumab. Although two anti-HER2 mAbs induced similar levels of antibody-dependent cellular cytotoxicity, trastuzumab-based CAR-T cells exhibited potent antitumor activity against HER2-positive cancer cells. In conclusion, our findings provide scientific evidence that antibody recognition of the membrane-proximal domain promotes the anti-tumor response of HER2-specific CAR-T cells. The food industry holds immense promise for 3D printing technology. Current research focuses mainly on optimizing food material composition, molding characteristics, and printing parameters. However, there is a notable lack of comprehensive studies on the shape changes of food products, especially in modeling and simulating deformations. This study addresses this gap by conducting a detailed simulation of the starch gel printing and deformation process using COMSOL Multiphysics 6.2 software. Additive manufacturing (AM) technology is widely acclaimed for its user-friendly operation and cost-effectiveness. The 3D printing process may lead to changes in part dimensions and mechanical properties, attributable to the accumulation of residual stresses. Studies require a significant amount of time and effort to discover the optimal composition of the printed material and the most effective deformed 3D structure. There is a risk of failure, which can lead to wasted resources and research delays. To tackle this issue, this study thoroughly analyzes the physical properties of the gel material through COMSOL Multiphysics 6.2 software, It simulates the heat distribution during the 3D printing process, providing important insights into how materials melt and solidify. Three-part models with varying aspect ratios were meticulously designed to explore shape changes during both the printing process and exposure to an 80 °C environment, employing NMR and rheological characterization. Using the generalized Maxwell model for material simulation in COMSOL Multiphysics, the study predicted stress and deformation of the parts by analyzing solid heat transfer and solid mechanics physical fields. Simulation results showed that among three models utilizing a gel-PET plastic membrane bilayer structure, Model No. 1, with the largest aspect ratio, exhibited the most favorable deformation under an 80 °C baking environment. It displayed uniform bending in the transverse direction without significant excess warpage in the edge direction. In contrast, Models No. 2 and No. 3 showed varying degrees of excess warpage at the edges, with Model No. 3 exhibiting a more pronounced warpage. Extrusion-based 3D food printing is one of the most common ways to manufacture complex shapes and personalized food. A wide variety of food raw materials have been documented in the last two decades for the fabrication of personalized food for various groups of people. This review aims to highlight the most relevant and current information on the use of protein raw materials as functional 3D food printing ink. The functional properties of protein raw materials, influencing factors, and application of different types of protein in 3D food printing were also discussed. This article also clarified that the effective and reasonable utilization of protein is a vital part of the future 3D food printing ink development process. The challenges of achieving comprehensive nutrition and customization, enhancing printing precision and accuracy, and paying attention to product appearance, texture, and shelf life remain significant. We report a superconducting kinetic inductance memory (SKIM) element, which can be controlled exclusively by the bias supercurrent, without involving magnetic fields and heating elements. The SKIM is nonvolatile memory. The device is made of two Nb Dayem bridges, and it can operate reliably up to 2.8 K. The achieved error rate is as low as one in 105 operations. The purified polysaccharides from Hericium erinaceus fermented mycelium entitled with PHEB was analyzed and it was mainly composed of six glycosidic bonds. It has been confirmed to show the relieving activity against Alzheimer's Disease (AD)- just as behaviors of B6C3-Tg (APPswePSEN1d E9)/Nju double transgenic [Genotype: (Appswe)T, (Psen1) T] (APP/PS1) mice. Six-week PHEB administration significantly improved the cognitive behavior of mice. Brain injury, amyloid beta deposition and tau hyperphosphorylation were alleviated in PHEB-treated AD mice without changes in other tissues. PHEB alleviated the oxidative stress in brains of AD mice via regulation the Nrf2 and its downstream kinase, which further improved the cholinergic system function. Proteomics and bioinformatics analysis showed that the therapeutic effect of PHEB is achieved by regulating calcium homeostasis mediated by oxidative stress. Furthermore, PHEB regulated the CaMK II/IV to achieve the calcium homeostasis in brains; and ultimately to show the anti-AD property. • The polysaccharide (PHEB) is first purified from Hericium erinaceus mycelium. • Clear structure of PHEB with the molecular weight of 36.1 kDa has been confirmed. • PHEB has the anti-Alzheimer's disease activities in APP/PS1 mice. • Oxidative stress-mediated calcium homeostasis is related to anti-AD effect of PHEB. Improving the activity and stability of electrocatalysts for methanol oxidation reaction (MOR) is key important for the large-scale application of direct methanol fuel cells (DMFCs). Herein, we provide a novel scheme to firstly build Bi-decorated Pt aerogel through the structural reconstruction of Pt/Bi 2 O 3 interface, which demonstrates an excellent mass activity of 7.99 A mg-1 Pt in MOR (8-fold higher than that of commercial Pt/C). Experimental results show that the synergistic effect between Bi0 species and Pt aerogel is the major cause of the improvement of the catalytic activity. In-suit Fourier transform infrared (FTIR) Spectroscopy results indicated that the generation and transformation of HCOO– intermediate are accelerated. Density functional theory (DFT) calculations indicate that the decorated Bi enhances the adsorption of OH species on the adjacent Pt site, which can facilitate the dehydrogenation of methanol and the removal of poisonous CO species. Thus, the electro-catalytic performance can be improved. • CQDs and GNPs were synthesized as fluorescence and colorimetric nanomaterials, respectively. • CQDs-GNPs-based dual-mode nanosensor was applied for malathion detection. • Standard arrays were fabricated by a smartphone and used for visual detection of malathion. • Cabbages adulterated by malathion were detected by the established calibration curves and standard arrays. A dual-mode fluorescence/colorimetric sensor based on carbon quantum dots (CQDs) and gold nanoparticles (GNPs) was developed for visual detection of malathion in cabbage. The CQDs-GNPs nanocomposite exhibited emission wavelength at 527 nm and absorption wavelength at 524 nm. The fluorescence intensity increased and absorption decreased with addition of malathion. Fluorescence and colorimetric calibration curves were established based on fluorescence intensity (R2 = 0.9914) and absorbance (R2 = 0.9608) in the range of 1 × 10−9–1 × 10−2 M, respectively. Furthermore, fluorescence and colorimetric standard arrays were prepared for visual detection of malathion according to the change of fluorescence brightness and color. Finally, the approximate concentrations of malathion in cabbage samples were estimated by the standard arrays and naked eyes. The calibration curves were used for accurate detection in cabbage samples with recoveries of 89.9%–103.4% (fluorescence) and 88.7%–107.6% (colorimetric). Tobacco ( Nicotiana tabacum L.) shows promise for remediating Cd-contaminated soil due to its significant Cd accumulation capabilities. Although various tobacco varieties exhibit distinct Cd bioaccumulation capacities, a comprehensive understanding of the underlying mechanisms is lacking. This study, conducted using hydroponics, explores differences in Cd accumulation and tolerance mechanisms between two tobacco varieties, Basma and Yunyan 87. The results showed that Cd stress reduced the dry weight, tolerance index, and root morphology for both varieties. Basma exhibited a relatively smaller decline in these indices compared to Yunyan 87. Moreover, Basma demonstrated a higher Cd bioconcentration factor (BCF), concentration, and accumulated content, signifying its superior tolerance and bioaccumulation capacity to Cd compared to Yunyan 87. The Carbonyl Cyanide3-ChloroPhenylhydrazone (CCCP) addition resulted in reduced Cd accumulation and BCFs in both tobacco species. This effect was more pronounced in Basma, suggesting that Basma relies more on an active transport process than Yunyan 87. This could potentially explain its enhanced bioaccumulation ability. Subcellular Cd distribution analysis revealed Basma's preference for distributing Cd in soluble fractions, while Yunyan 87 favoured the cell wall fractions. Transmission electron microscope showed that Basma's organelles were less damaged than Yunyan 87's under Cd stress, possibly contributing to the superior tolerance of Basma. Therefore, these results provided a theoretical foundation for development of Cd-contaminated soil tobacco remediation technology.

Flavonoid metabolism in Camptotheca acuminate remained an untapped area for years. A tandem MS approach was used and focused on the mining and characterizing of flavonoids in mature C. acuminate . Fifteen new flavonoids and forty-three known flavonoids, including fifteen flavone analogs, sixteen flavonol analogs, seven flavanone analogs, six chalcone analogs, four xanthone analogs, ten flavane analogs were mined and identified based on their MS/MS fragments. Fifty-three of them were firstly characterized in C. acuminate. Eight biosynthetic precursors for these flavonoids were also identified. We constructed a specific metabolic map for flavonoids according to their relative contents in the flowers, fruits, stems, and leaves of C. acuminate . Furthermore, the most probable genes involved in chalcone biosynthesis, flavonoid hydroxylation, methylation, and glycosylation were further mined and fished in the gene reservoir of C. acuminate according to their conserved domains and co-expression analysis. Gold (Au) can be used as an ideal metal electrocatalyst for ethanol and glucose oxidation reactions due to its high performance-to-cost ratio. In this paper, the Au aerogel with high-index facets was synthesized by using the laser ablation in liquid technology, which can improve the electrocatalytic activity of Au. The as-prepared Au aerogel showed excellent mass activity and specific activity toward ethanol oxidation reaction, which are 4.6 times and 2.1 times higher than Au/C, respectively. The 3D porous nature and rich defect of the Au aerogel provide more active sites. In addition, the high-index facets with under-coordinated atoms enhance the adsorption of ethanol and glucose molecules, thus improving the intrinsic catalytic activity of Au aerogel. The effect of high-index facets has also been investigated by density functional theory calculations. Furthermore, the Au aerogels also show good electrocatalytic activity and stability toward glucose oxidation reaction. This paper describes a near-infrared quantum dot (CuInS 2 QD)/antibiotic (vancomycin) nanoparticle-based assay for the Staphylococcus aureus and iron(iii) detection. CuInS 2 QDs with good biological tissue permeability and biocompatibility are combined with vancomycin through covalent interaction to form a detection system for two harmful factors. The detection principle of Staphylococcus aureus is mainly the fluorescence quenching caused by the accumulation of CuInS 2 @Van QDs on the surface of Staphylococcus aureus. The detection principles of the iron(iii) ion are mainly ascribed to the aggregation of quantum dots and the transfer of charges, which cause the fluorescence signal to change. The linear range of S. aureus and the Fe 3+ ion is 10 3 to 10 8 CFU mL -1 and 10-90 μM, respectively. Their detection limits are 665 CFU mL -1 and 3.5 μM, respectively. The procedure was validated by the quantitation of Staphylococcus aureus and iron(iii) in spiked samples, and was found to demonstrate the feasibility of this method.

Understanding bacterial adhesion on a surface is a crucial step to design new materials with improved properties or to control biofilm formation and eradication. Sum Frequency Generation (SFG) vibrational spectroscopy has been employed to study in situ the conformational response of a self-assembled monolayer (SAM) of octadecanethiol (ODT) on a gold film to the adhesion of hydrophilic and hydrophobic ovococcoid model bacteria. The present work highlights vibrational SFG spectroscopy as a powerful and unique non-invasive biophysical technique to probe and control bacteria interaction with ordered surfaces. Indeed, the SFG vibrational spectral changes reveal different ODT SAM conformations in air and upon exposure to aqueous solution or bacterial adhesion. Furthermore, this effect depends on the bacterial cell surface properties. The SFG spectral modeling demonstrates that hydrophobic bacteria flatten the ODT SAM alkyl chain terminal part, whereas the hydrophilic ones raise this ODT SAM terminal part. The aim of this study was to establish whether early prepubertal Sannen dairy goat could provide a large source of ova for SCNT. The effects of different hormonal treatments including untreated control, FSH alone, estadiol plus progesterone (E2-P4), and E2-P4 and FSH (E2-P4-FSH) on ovary size, follicle number and size were studied using the early prepubertal goats aged 39-60 days. Then prepubertal goats aged 39-120 days were categorized into three groups to study the effect of age on recruited follicle number. The meiotic competence of oocytes derived from > or =3 mm follicles recovered from the early prepubertal goats treated with E2-P4-FSH was compared with those treated with FSH alone. Finally, the development competence of the ova from the early goats treated with E2-P4-FSH was evaluated by SCNT. The E2-P4-FSH treatment produced the largest ovaries, the highest numbers of total follicles and follicles > or =3 mm diameter compared with the other treatments. The number of > or =3 mm follicles per goat treated with E2-P4-FSH was significantly higher for those in the age groups 39-60 days than those in the age groups 61-90 days and 91-120 days. The FSH alone treatment resulted in a lower proportion matured ova in vitro within 27 hr than from those goats treated with E2-P4-FSH. Ova derived from the early prepubetal goats resulted in lower rate of blastocyst in SCNT (15.3% versus 22.1%, P < 0.01) than that of adult goat However, the number of ova recovered per goat was substantially greater for the prepubertal goats (108 +/- 10.3 Versus 28 +/- 5.0). Consequently, the younger goats produced significantly more blastocyst (7.1 +/- 2.7 versus 4.2 +/- 1.4) per head. It was concluded that early pubertal goats treated with E2-P4-FSH could provide a relatively high number of developmentally competent ova for SCNT studies.
