In this study, cauda epididymal sperm were collected from Amur leopard cats with various causes of death as well as Tsushima leopard cats that underwent castration surgery, and sperm quality was compared with that in domestic cats. A sufficient number of sperm similar to those in domestic cats could be collected from the cauda epididymis of Amur leopard cats. However, in old leopard cats, no or very few cauda epididymal sperm were recovered, although there were no differences in sperm motility and sperm abnormality. There were no significant differences in sperm quality immediately after collection and after freezing-thawing of cauda epididymal sperm compared with corresponding estimates in domestic cats. Equine and human chorionic gonadotropins were administered to two female Amur leopard cats to induce estrus and ovulation during non-breeding season. Fresh semen collected from male cats was surgically inseminated into the uterine horn of the females. In one animal, two fetal sacs without heartbeats were observed on abdominal ultrasonography 31 days after insemination, which indicated that embryo death had occurred. In the other animal, fetal heartbeats were detected in two fetal sacs 29 days after insemination, which confirmed as pregnancy. This animal delivered two newborns 68 days after insemination; the one of the kittens was assumed to be stillbirth, and the other grew normally. We collected semen from a male Amur leopard cat using the transrectal electroejaculation method and investigated the semen qualities for about four years. In addition, the influence of the season on the spermatogenic function of the Amur leopard cat was investigated with regard to the semen qualities, testicular volume and serum testosterone level. As a result, we could collect semen with good sperm qualities that would be useable for artificial insemination. Some seasonality was noted in the testicular volume and serum testosterone level. Abstract  We develop a hybrid unsteady-flow simulation technique combining direct numerical simulation (DNS) and particle tracking velocimetry (PTV) and demonstrate its capabilities by investigating flows past an airfoil. We rectify instantaneous PTV velocity fields in a least-squares sense so that they satisfy the equation of continuity, and feed them to the DNS by equating the computational time step with the frame rate of the time-resolved PTV system. As a result, we can reconstruct unsteady velocity fields that satisfy the governing equations based on experimental data, with the resolution comparable to numerical simulation. In addition, unsteady pressure distribution can be solved simultaneously. In this study, particle velocities are acquired on a laser-light sheet in a water tunnel, and unsteady flow fields are reconstructed with the hybrid algorithm solving the incompressible Navier–Stokes equations in two dimensions. By performing the hybrid simulation, we investigate nominally two-dimensional flows past the NACA0012 airfoil at low Reynolds numbers. In part 1, we introduce the algorithm of the proposed technique and discuss the characteristics of hybrid velocity fields. In particular, we focus on a vortex shedding phenomenon under a deep stall condition (α = 15°) at Reynolds numbers of Re = 1000 and 1300, and compare the hybrid velocity fields with those computed with two-dimensional DNS. In part 2, the extension to higher Reynolds numbers is considered. The accuracy of the hybrid simulation is evaluated by comparing with independent experimental results at various angles of attack and Reynolds numbers up to Re = 104. The capabilities of the hybrid simulation are also compared with two-dimensional unsteady Reynolds-Averaged Navier–Stokes (URANS) solutions in part 2. In the first part of these twin papers, we demonstrate that the hybrid velocity field approaches the PTV velocity field over time. We find that intensive alternate vortex shedding past the airfoil, which is predicted by the two-dimensional DNS, is substantially suppressed in the hybrid simulation and the resultant flow field is similar to the PTV velocity field, which is projection of the three-dimensional velocity field on the streamwise plane. We attempt to identify the motion that originates three-dimensional flow patterns by highlighting the deviation of the PTV velocity field from the two-dimensional governing equations at each snapshot. The results indicate that the intensive spots of the deviation appear in the regions in which three-dimensional instabilities are induced in the shear layer separated from the pressure side. Air–water two-phase flow in a helically coiled tube is investigated using backlight imaging tomography to elucidate the effect of centrifugal acceleration on phase distribution and interfacial structure. Superficial velocities up to 6 m s-1 in 20 mm diameter tube are tested. We focused on a slug flow regime in which centrifugal acceleration dominates the flow. The interfacial structure is visualized in six directions using a set of originally designed mirror-mounted water jackets. A temporal expansion image is made from line-sampled images and is used to reconstruct phase distribution through a linear backward projection algorithm. The present topography measurement showed various new features of gas–liquid two-phase flow in a helically coiled tube, such as a wall-covering effect in the case of high superficial velocity. Data-assimilation capabilities of hybrid-type simulations integrating time-resolved particle image velocimetry with unsteady computational fluid dynamics (CFD) are characterized, and a series of algorithms developed previously are evaluated in terms of four criteria: (i) compatibility with the governing equations; (ii) completeness of a set of flow quantities; (iii) temporal and spatial filtering functions; and (iv) spatial resolution. This study specifically introduces a hierarchy of three hybrid simulations combining time-resolved particle tracking velocimetry (PTV) and direct numerical simulation (DNS) from low to high fidelities: the proper orthogonal decomposition–Galerkin-projection approach with proportional feedback of PTV data, the DNS solver with similar feedback, and the DNS solver with the extended Kalman filter. By solving a planar-jet problem at , we demonstrate that the resultant hybrid flow fields essentially (i) satisfy the governing equations spatially and approximately temporally, and (ii) can provide instantaneous pressure fields (iii) with the noise levels substantially lower than those of the original PTV data and (iv) the resolution comparable to CFD. The results show that increasing the feedback gain improves replicability, i.e. the agreement between the simulation and the data; however, it degrades temporal compatibility and filtering functions.
