Social isolation (SI) is a notable model of prolonged mild stress, characterized by multiple neurochemical and behavioral alterations, that appears particularly suitable for studying different aspects of the interplay between stress and ethanol (EtOH) consumption in order to characterize potential molecular mechanisms, including changes in the function of inhibitory GABAergic synapses, underlying such interaction. In C57BL/6J mice, SI is associated with an altered hippocampal concentration of the neuroactive steroids 3α-hydroxy-5α-pregnan-20-one (3α-5α-THP), an increased expression of the α4 and δ subunit of γ-aminobutyric acid type A receptors (GABAARs) in the dentate gyrus (DG), and a parallel enhancement of the stimulatory action of 4,5,6,7-tetrahydroisoxazolo[5,4-c] pyridin-3-ol (THIP) on GABAergic tonic currents recorded in voltage-clamped DG granule cells (DGGCs). In addition, SI in C57BL/6J mice determines an increase in voluntary EtOH consumption and EtOH preference when compared to group-housed (GH) control animals. Furthermore, in hippocampal slices of SI mice we also observed a marked reduction of both cellular excitability and long term potentiation (LTP) in pyramidal neurons of the CA1 hippocampal sub-region, effects that were prevented by the long term treatment of SI mice with the neuroactive steroid precursor progesterone. In this article, we summarize some of our recent findings on the effects of SI in C57BL/6J mice on voluntary EtOH intake, regulation of GABAARs gene expression and function and hippocampal long term synaptic plasticity.

The relation between changes in brain and plasma concentrations of neurosteroids and the function and structure of [Gamma]-aminobutyric acid type A ([GABA.sub.A]) receptors in the brain during pregnancy and after delivery was investigated in rats. In contrast with plasma, where all steroids increased in parallel, the kinetics of changes in the cerebrocortical concentrations of progesterone, allopregnanolone (AP), and allotetrahydrodeoxycorticosterone (THDOC) diverged during pregnancy. Progesterone was already maximally increased between days 10 and 15, whereas AP and allotetrahydrodeoxycorticosterone peaked around day 19. The stimulatory effect of muscimol on 36[Cl.sup.-] uptake by cerebrocortical membrane vesicles was decreased on days 15 and 19 of pregnancy and increased 2 days after delivery. Moreover, the expression in cerebral cortex and hippocampus of the mRNA encoding for [Gamma]2L [GABA.sub.A] receptor subunit decreased during pregnancy and had returned to control values 2 days after delivery. Also [Alpha]1,[Alpha]2, [Alpha]3, [Alpha]4, [Beta]1 [Beta]2,[Beta]3, and [Gamma]2S mRNAs were measured and failed to change during pregnancy. Subchronic administration of finasteride, a 5[Alpha]-reductase inhibitor, to pregnant rats reduced the concentrations of AP more in brain than in plasma as well as prevented the decreases in both the stimulatory effect of muscimol on 36[Cl.sup.-] uptake and the decrease of [Gamma]2L mRNA observed during pregnancy. These results indicate that the plasticity of [GABA.sub.A] receptors during pregnancy and after delivery is functionally related to fluctuations in endogenous brain concentrations of AP whose rate of synthesis/metabolism appears to differ in the brain, compared with plasma, in pregnant rats.

The progressive increase of marine macro-litter on the bottom of the Mediterranean Sea is an urgent problem that needs accurate information and guidance to identify those areas most at risk of accumulation. In the absence of dedicated monitoring programs, an important source of opportunistic data is fishery-independent monitoring campaigns of demersal resources. These data have long been used but not yet extensively. In this paper, MEDiterranean International Trawl Survey (MEDITS) data was supplemented with 18 layers of information related to major environmental (e.g. depth, sea water and wind velocity, sea waves) and anthropogenic (e.g. river inputs, shipping lanes, urban areas and ports, fishing effort) forcings that influence seafloor macro-litter distribution. The Random Forest (RF), a machine learning approach, was applied to: i) model the distribution of several litter categories at a high spatial resolution (i.e. 1 km2); ii) identify major accumulation hot spots and their temporal trends. Results indicate that RF is a very effective approach to model the distribution of marine macro-litter and provides a consistent picture of the heterogeneous distribution of different macro-litter categories. The most critical situation in the study area was observed in the north-eastern part of the western basin. In addition, the combined analysis of weight and density data identified a tendency for lighter items to accumulate in areas (such as the northern part of the Tyrrhenian Sea) with more stagnant currents. This approach, based on georeferenced information widely available in public databases, seems a natural candidate to be applied in other basins as a support and complement tool to field monitoring activities and strategies for protection and remediation of the most impacted areas. Abstract The global plastic economy demands to reduce the flow of plastic into oceans and promote remedial actions for already accumulated seafloor litter. In this perspective, baseline levels of contamination and tools for dating litter items in order to assess the efficacy of those actions, are thus needed. In this note we discuss the utility of introducing the acquisition of shelf-life and labels features from litter items into already established standardized protocols such as the one proposed by MEDiterranean International Trawl Survey (MEDITS). Our investigation was conducted on 612 high resolution images of litter items collected in each haul, which was retrieved during 6 years of surveys (2013–2018) around Sardinian sea (central western Mediterranean). While for the majority of items (89%) expiration date or labels were not present or legible, over 50% of dated items were likely dumped at sea from a period <5 years. In this perspective, this sort of data could represent a useful tool for monitoring the effectiveness of input reduction actions that implicitly rely on the gradual reduction of recently dumped items. Highlights • Tools for estimating the efficacy of measures aimed at reducing the input of litter in worlds' oceans are needed. • Expiration dates and label features were retrieved from MEDITS survey photo material from 2013 to 2018 in order to back date seafloor macro-litter items. • A total of 54 items were back dated, including aluminum cans produced in 1980's. • Plastic items dumped within 5 years from retrieval are the most numerous and identifiable seafloor macro-litter items. • This practice could be introduced in operation onboard to monitor the redution of this fraction of seafloor litter across years and thus the efficacy of input reduction measures. Abstract The aim of the present study was to evaluate, through a Collaborative Fisheries Research (CFR) project, the efficacy of artificial dens as a possible integrative action for the management of O. vulgaris in the wild. Artificial dens, anchored on rocky substrates at a depth of 38–42 m in a temporary Fully Protected Area (FPA) off the western coast of Sardinia (central western Mediterranean Sea), showed their effectiveness as a temporary and/or safe site for Octopus vulgaris spawning. The suitability of these artificial dens was demonstrated by the presence of egg strings and females in parental care, and by the fact that all of the brooding-phases until the hatching had taken place inside. The presence of abundant cobbles, appeared to be a key factor in the building of a solid barrier for protection at the entrance of the spawning artificial dens, similar to those seen in natural dens. In addition, our results demonstrate the potential coexistence of artificial dens with natural ones, suggesting their potential use as integrative tool for the management of O. vulgaris. Close collaboration with fishermen in the framework of a CFR project within FPA may increase the success of this management action, through the enforcement of the area. Brooding details and laid egg features revealed by the monitoring are reported and discussed. Abstract This study reports data on benthic litter abundance, composition and distribution obtained during deep-sea trawl surveys conducted along the Sardinian continental margin down to the bathyal plain, at depths comprised from 740 to 1740 m. None of the investigated sites was litter free. Density ranged from a minimum of 4 to a maximum of >1300 litter items per km2, with a mean value of 258 ± 59 items. Plastic accounted for 56% of the total collected items, followed by glass (24%), metal (10%). Most items, irrespectively of the category, were single-use items. Fish abundance in all of the investigated catches was significantly higher than the number of litter items, the weight of which was similar to the reared fish biomass. Our results confirm that anthropogenic waste has reached the deep Mediterranean Sea, and that the most recent EU legislation banning single-use plastic tools represents a timely and necessary measure. By means of ROV surveys, we assessed the quantity, composition and bathymetric distribution of marine litter in 17 sites along the Sardinian continental margin (Central Western Mediterranean) at depths ranging from 100 to 480 m. None of the investigated sites was litter free, but the mean density of litter (0.0175 ± 0.0022 items m − 2 ) was lower than that reported from other Tyrrhenian regions. The difference in the total litter density among sites was negligible, but the density of derelict fishing gear (DFG) items (most of which ascribable to small scale fishery) in submarine canyons was higher in submarine canyons than in other habitats. Our result suggest that submarine canyons (known to be highly vulnerable ecosystems) act as major repositories of DFGs, and, therefore, we anticipate the need of specific measures aimed at minimizing the loss and abandonment of DFGs in submarine canyons. The present work compares microplastics (MPs) contamination in two charismatic crustaceans: European spiny lobster Palinurus elephas and langoustine Nephrops norvegicus. Samples (P. elephas n = 14; N. norvegicus n = 15) were collected between 76 and 592 m depth, from four sites in west Sardinia, Italy. An extraction protocol was applied on stomachs and intestines, separately, and over 500 particles were further characterized through μFT-IR. We document 100 % occurrence in specimens from both species, with P. elephas being significantly more contaminated (9.1 ± 1.75 vs. 3.2 ± 0.45 MPs individual−1), ingesting larger MPs with different polymeric composition. The scavenging-based feeding strategy of both species could explain such exposure to MPs, mostly derived by single-use plastic. The overall results highlight that both species are clearly affected by plastic pollution, being valuable bioindicators and charismatic species that could thus represent excellent flagship species for raising awareness toward the global issue of plastic in the marine environment. • We compared microplastic contamination in Palinurus elephas and Nephrops norvegicus. • We report 100 % occurrence, polyester and polyethylene as dominant polymers (>65 %). • Spiny lobsters ingest more and larger microplastics than langoustines. • Langoustines were confirmed to fragment microplastics during digestion. • Both species are efficient bioindicators and flagship species of plastic impact. Background. The shortnose greeneye, Chlorophthalmus agassizi Bonaparte, 1840, is a species with a circumglobal distribution and is among the most abundant commercial fishes in some Mediterranean areas. The knowledge of the biology and ecology of this species is poor and geographically limited, then the aim of this study is to provide a contribution to the knowledge on the reproductive biology of this monoecious deep sea fish in Sardinian waters. Materials and Methods. In this paper the morphology and the development of the gonads, the mean size at maturity, the monthly evolution in the percent frequency of the maturity stages, and of the indices related to reproduction of the shortnose greeneye were examined. Individuals were caught by trawls, between 270 and 504 m of depth in the Sardinian seas. Results. The ovarian pattern is of an asynchronous type, characterized by releasing of eggs in successive batches. Seven stages of development for the ovary and four for the testis were identified on the basis of macroscopic and histological features. The female portion is the most evident component and shows a later maturation than the male portion. The spawning period is unique and takes place from May to September. Conclusion. The identification of spawning period and the adopted reproductive strategy is essential to obtain a better understanding of its biology and a good management of its fisheries. A single adult female specimen of Halosaurus ovenii Johnson, 1864 was captured by trammel nets at a depth of about 200 m off the coast of Arbatax (Sardinia, Italy) in early April 2007. Macroscopic and microscopic analysis of the gonad showed a postspawning ovary. This is the fourth documented capture of this fish in the Mediterranean Sea, representing the north-easternmost record for this species in this geographic area. Neuroactive steroids such as 3α,5α‐THP are reduced metabolites of progesterone and are considered to play an important physiological role to locally modulate neuronal excitability by “fine‐tuning” the action of GABA acting at GABAA receptors (GABAA‐Rs). In different brain regions, such as the hippocampus, different subpopulations of nerve cells exhibit two components of inhibitory GABAergic transmission: a phasic component mediated by synaptic GABAA‐Rs, and a tonic component mediated by “ambient” GABA acting at extrasynaptic GABAA‐Rs mainly containing the δ subunit and endowed with a higher sensitivity to neuroactive steroids compared to synaptic receptors. It is also well accepted that fluctuations in brain neuroactive steroid levels may result in plastic changes of GABAA‐Rs. In this article we review some of our results obtained with the model of pregnancy in rats. Pregnancy, in fact, is characterized by a marked and progressive increase in plasma and brain levels of neuroactive steroids which could contribute to changes in mood, anxiety as well as other psychiatric conditions. Such elevation in brain neuroactive steroid concentrations during pregnancy, in turn, is accompanied by alterations in both gene expression and function of synaptic and extrasynaptic GABAA‐Rs in the hippocampus as well as other areas. In the coastal karstic region of SW Sardinia, from Capo Pecora in the north to Nebida in the south, many interesting geological monuments, ecotopes and several places of social and cultural interest that deserve preservation and valorization can be found. This mostly rocky coastline is characterized by the presence of a Cambrian sequence prevalently composed of limestones and dolostones and only locally of phyllites. A correct use of the many environmental and cultural monuments is necessary both to preserve and to valorize them; therefore they have to be properly managed in order to preserve their original natural conditions. Here, we describe and classify these monuments with the aim to propose an integrated model of sustainable development of this exceptional coastal landscape. Our model is based on several thematic maps according to which we propose the establishment of a natural reserve with specific geological and geomorphologic sites and the creation of several touristic circuits that are the basis for a further valorization of this territory. Marine litter is an ever-increasing problem that demands immediate reduction plans and mitigation actions that should act synergically to efficiently meet ambitious goals. Since the seafloor has been recognized as the major sink for marine debris, the study of litter accumulation dynamics represent a fundamental tool to evaluate possible removal actions. We analysed a 7 years (2013–2019) standardized data series collected along Sardinian fishing grounds through MEDiterranean International Trawl Survey, for which estimates of density and weight of seafloor macro-litter were calculated for over 707 hauls. Results show the absence of any temporal trend in seafloor macro-litter density and weight, but rather indicate a spatial and bathymetric segregation of different litter categories. Our data showed how different sources and physical features of macro-litter items (i.e. , plastic and fishing gear, rubber, glass, metals, and cloth) led to spatially segregated accumulation hotspots. We also point out here how the identification of seafloor macro-litter hotspots using aggregated data that include plastic items could obscure the identification of other segregated but yet relevant hotspots of other macro-litter categories accumulated in the marine environment. These hotspots often occurred at shallower depths and closer to coastlines, thus representing potential spots where eventual future litter removal action could be prioritized. • We investigated temporal trends in seafloor macro-litter abundance in 7 years study. • Macro-litter abundance and density did not show any temporal trend. • Accumulation hotspots showed a segregated distribution according to litter typologies. • More accessible areas for mitigation actions have been identified. • Aggregated data may blur the identification of other minor accumulation hotspots.
