Current article presents results of a qualitative research dedicated to the phenomenology of intersubjective relationships in person’s infantile, realistic, value life-worlds. The characteristics of the anthropological image of the Other as well as the basic attitudes that define relationship between the subjects life-world and the Other are described.

Abstract Gelatinous drop-like corneal dystrophy (GDLD) is a rare autosomal recessive eye disease. GDLD is characterized by the loss of barrier function in corneal epithelial cells (CECs) and amyloid deposition due to pathogenic variants in the TACSTD2 gene. Limbal stem cell transplantation (LSCT) has been suggested as an effective therapeutic alternative for patients with GDLD. However, despite LSCT, amyloid deposition recurs in some patients. The pathogenesis of recurrence is poorly studied. We present the case of a patient with GDLD. Genetic analysis revealed a homozygous deletion, NM_002353.3:c.653del, in the TACSTD2 gene. Functional analysis in a cell model system revealed the loss of the transmembrane domain and subcellular protein mislocalization. The patient with GDLD underwent direct allogeneic LSCT with epithelial debridement followed by deep anterior lamellar keratoplasty 10 months later due to amyloid deposition and deterioration of vision. Taken together, the results of transcriptome analysis and immunofluorescence staining of post-LSCT corneal sample with amyloid deposits obtained during keratoplasty demonstrated complete restoration of wild-type TACSTD2 expression, indicating that donor CECs replaced host CECs. Our study provides experimental evidence that amyloid deposition can recur after LSCT despite complete restoration of wild-type TACSTD2 expression.

Purpose. The article is devoted to the problem of motivational readiness towards the professional activity of cadets in the universities under the Ministry of Internal Affairs. The relevance of the chosen topic is due to the need to improve the training quality in the field of public order protection, protection of citizens' interests and control over compliance with the rule of law in all spheres of society. The subject of the analysis is the motivational readiness formation peculiarities towards the professional activity among cadets of universities under the Ministry of Internal Affairs (on the example of the Federal State Public Educational Establishment of Higher Training «Nizhniy Novgorod Academy of the Ministry of the Interior of the Russian Federation» cadets). Methodology. The research is based on the methodology of the study motivation at the University by T.I. Ilyina and the methodology of value diagnosing orientations in the career "Career Anchors" by E. Shein. Results. The obtained research results allowed us to identify the dynamics of the leading motives, as well as to identify a group of the most significant value orientations of students of the Nizhniy Novgorod academy of the Ministry of the Interior of Russia, underlying their mastery of the future profession: service orientation (the need to benefit people, society), integration of lifestyles (a successful combination of personal life and career), professional competence (constant self-improvement and improvement of one's professional level) and challenge (willingness to act in extreme situations, take responsibility for solving complex tasks). Practical implications. The results of the study can be applied in the process of preparing and conducting classes in educational organizations of the Ministry of Internal Affairs of Russia, as well as by practicing employees of the Ministry of Internal Affairs.

High-chromium martensitic steels are a promising material for the production of elements of boilers and steam pipelines, as well as blades and rotors of steam turbines for new coal-burning thermal generating units. The use of such materials will give an opportunity for the transition to ultra-supercritical steam parameters (temperature of 600–620 °C and pressure of 25–30 MPa), which will allow increasing the efficiency of generating units to 45 %. Modifications of the chemical composition of high-chromium steels have led to significant improvements of high-temperature properties such as 100,000 h creep strength and 1 % creep limit, while resistance to softening due to low-cycle fatigue remains understudied in this field. This work covers the study of low-cycle fatigue at room temperature with different amplitudes of deformation of martensitic high-chromium 10%Cr–3%Co–2%W–0.5%Mo–0.2%Cu–0.2%Re–0.003%N–0.01%B steel. The steel was pre-subjected to normalizing at 1050 °С followed by tempering at 770 °С. After heat treatment, the steel structure was a tempered martensitic lath structure stabilised by the particles of secondary phases of M23C6 carbides, NbX carbonitrides, and M6C carbides. The average width of martensite laths was 380 nm, and the dislocation density was 1.4×1014 m−2. At low-cycle fatigue, with an increase in the strain amplitude from 0.2 to 1 %, the number of cycles before failure significantly decreases, and the value of plastic deformation in the middle of the number of loading cycles significantly increases. Maximum softening (18 %) is observed at a strain amplitude of 1 % in the middle of the number of loading cycles. In general, the steel structure after low-cycle fatigue tests does not undergo significant changes: the width of the laths increases by 18 % at a strain amplitude of more than 0.3 %, while the dislocation density remains at a rather high level (about 1014 m−2) at all strain amplitudes.

Abstract Background Prostate adenocarcinoma (PRAD) is the second leading cause of cancer-related deaths in men. High variability in DNA methylation and a high rate of large genomic rearrangements are often observed in PRAD. Results To investigate the reasons for such high variance, we integrated DNA methylation, RNA-seq, and copy number alterations datasets from The Cancer Genome Atlas (TCGA), focusing on PRAD, and employed weighted gene co-expression network analysis (WGCNA). Our results show that only single cluster of co-expressed genes is associated with genomic and epigenomic instability. Within this cluster, TP63 and TRIM29 are key transcription regulators and are downregulated in PRAD. We discovered that TP63 regulates the level of enhancer methylation in prostate basal epithelial cells. TRIM29 forms a complex with TP63 and together regulates the expression of genes specific to the prostate basal epithelium. In addition, TRIM29 binds DNA repair proteins and prevents the formation of the TMPRSS2:ERG gene fusion typically observed in PRAD. Conclusion Our study demonstrates that TRIM29 and TP63 are important regulators in maintaining the identity of the basal epithelium under physiological conditions. Furthermore, we uncover the role of TRIM29 in PRAD development.

Introduction. The work updates the situation with the enhanced sustainability of industrial enterprises in the Arctic territories of the Russian Federation since they are characterized with resources’ high potential associated with high environmental risks due to their exploration. Purpose. The paper aims at comparing the key indicators for sustainable development of the leading industrial enterprises in the Russian Federation in terms of their environmental impact. Materials and Methods. The authors reviewed modern studies related to the assessment of industrial development in the Arctic from the standpoint of technology, environmental friendliness, the formation of a data system, which reflect the trends in environmental load on the territory from the activities of industries. The identified trends in the sustainable development of industrial enterprises in the Arctic zone were analyzed and analytically interpreted. Methodologically, the paper synthesizes the methods of retrospective analysis and identifies the trends by analyzing rated key sustainability indicators defined by the data of industrial enterprises. Results. The paper proves the need for an in-depth analysis and detailed analysis of indicators of industrial load on the environment in the Arctic territories. The database for comparing key indicators was compiled, and the key trends in the sustainable development of industrial enterprises were identified. The analysis of their dynamics showed the multidirectionality in the development of industrial enterprises. The trajectories of enterprises’ efforts in achieving sustainability parameters were described: when the growth of expenditures is associated with a decrease in emissions and waste from production activities; when production and environmental indicators show multidirectional dynamics; when the enterprise growth rate decreases, environmental indicators improve. Conclusions. Thus, the authors substantiated the need to develop and implement a mechanism for regulating environmental policy at the state level, which would provide for measures aimed at active and systematic reduction of the environmental load on the environment in the Arctic territories. The animal models used in biomedical research cover virtually every human disease. RatDEGdb, a knowledge base of the differentially expressed genes (DEGs) of the rat as a model object in biomedical research is a collection of published data on gene expression in rat strains simulating arterial hypertension, age-related diseases, psychopathological conditions and other human afflictions. The current release contains information on 25,101 DEGs representing 14,320 unique rat genes that change transcription levels in 21 tissues of 10 genetic rat strains used as models of 11 human diseases based on 45 original scientific papers. RatDEGdb is novel in that, unlike any other biomedical database, it offers the manually curated annotations of DEGs in model rats with the use of independent clinical data on equal changes in the expression of homologous genes revealed in people with pathologies. The rat DEGs put in RatDEGdb were annotated with equal changes in the expression of their human homologs in affected people. In its current release, RatDEGdb contains 94,873 such annotations for 321 human genes in 836 diseases based on 959 original scientific papers found in the current PubMed. RatDEGdb may be interesting first of all to human geneticists, molecular biologists, clinical physicians, genetic advisors as well as experts in biopharmaceutics, bioinformatics and personalized genomics. The article investigates the use of Russian architecture images in contemporary projects in Russia. Recent examples demonstrate a trend towards embodying local memories and Russian architectural identities: many of the contemporary architects and designers incorporate traditional Russian art motifs into their works, using materials and techniques that allow them to create a unique and culturally significant aesthetics. The theoretical aspects of the phenomenon of memory and continuity in traditional Russian architecture are considered. Examples of contemporary projects with characteristic reminiscences – images of traditional housing, complex spatial compositions, borrowings of individual forms and techniques – are analyzed. Conclusions are drawn about the use of the cultural memory phenomenon in contemporary projects.

The article presents the results of a study of personal distress and its relationship with indicators of psychological well-being and professional burnout among teachers with different experience of working with students with learning disabilities. The study involved 293 respondents (N=293) from 20 to 65 years old, (average age 34.6±13.2; SE=0.774), with work experience from 6 months to 45 years (average — 11.16; SD=11.13; SE=0.649) of who 94.3% were women. In the empirical sample, three groups of teachers were represented in accordance with the experience of working with students with learning disabilities: 111 people working in a special school (37.8%); teachers working in an inclusive school — 103 people (35.15%); and teachers who have no experience working with students with learning disabilities — 79 people (26.96%). The "Scale of psychological well-being" by K. Riff in the adaptation of P.P. Fesenko, T.D. Shevelenkova, the questionnaire of professional burnout by K. Maslach, S. Jackson (MBI), the multifactorial empathy questionnaire by M. Davis (IRI) methods were used. The results obtained make it possible to say that personal distress is significantly higher among teachers working in inclusive education. Empathic care and decentration are significantly higher in groups of teachers of special and inclusive education compared to teachers without work experience in inclusion. At the same time, there were no differences between groups of teachers in terms of psychological well-being. Emotional exhaustion, empathic care, self-acceptance and autonomy turned out to be significant predictors of the personal distress of teachers with different experience of working with students with learning disabilities.

The problem of the development of emotional self-regulation as a process of becoming an independent experiencing in its cultural sign-symbolic mediation is discussed. The results of the study are presented: the process of becoming an independent experience in adolescents with developmental delay. The sample consisted of 42 normotypic adolescents and 25 adolescents with developmental delay. Methods: psychodiagnostic – drawing “My emotional state”, author's questionnaire, composition “The meaning of music in my life”; methods of qualitative data analysis: methods of grounded theory, content analysis. Methods of mathematical statistics: frequency analysis, criterion – Fisher angular transformation. A generalizing conclusion is made that the mediation of the experience of a teenager with developmental delay is largely difficult. Music plays the role of “activation” – strengthening the actual state of a teenager, but the experiencing remains incomplete, since there is only saturation of the immediate bodily-perceived level of it. Low cognitive and reflexive abilities of adolescents with developmental delay do not allow using an artistic image as a means-incentive for “mastering oneself”.

Given the high growth rates of cognitive decline among the elderly population and the lack of effective etiological treatments, early diagnosis of cognitive impairment progression is an imperative task for modern science and medicine. It is of particular interest to identify predictors of an unfavorable subsequent course of cognitive disorders, specifically, rapid progression. Our study assessed the informative role of various risk factors on the dynamics of cognitive impairment among mild cognitive impairment (MCI) patients. The study included patients with MCI (N = 338) who underwent neuropsychological assessment, magnetic resonance imaging (MRI) examination, blood sampling for general and biochemical analysis, APOE genotyping, and polygenic risk score (PRS) evaluation. The APOE ε4/ε4 genotype was found to be associated with a diminished overall cognitive scores initial assessment and negative cognitive dynamics. No associations were found between cognitive changes and the PRS. The progression of cognitive impairment was associated with the width of the third ventricle and hematological parameters, specifically, hematocrit and erythrocyte levels. The absence of significant associations between the dynamics of cognitive decline and PRS over three years can be attributed to the provided suitable medical care for the prevention of cognitive impairment. Adding other risk factors and their inclusion in panels assessing the risk of progression of cognitive impairment should be considered.

Emotional stress is one of the health risk factors in the modern human lifestyle. Stress exposure can provoke the manifestation of various pathological conditions, one of which is a sharp increase in the blood pressure level. In the present study, we analyzed changes in the transcriptome profiles of the hypothalamus of hypertensive ISIAH and normotensive WAG rats exposed to a single short-term restraint stress (the rat was placed in a tight wire-mesh cage for 2 h). This type of stress can be considered emotional stress. The functional annotation of differentially expressed genes allowed us to identify the most significantly altered biological processes in the hypothalamus of hypertensive and normotensive rats. The study made it possible to identify a group of genes that describe a general response to stress, independent of the rat genotype, as well as a hypothalamic response to stress specific to each strain. The alternatively changing expression of the Npas4 (neuronal PAS domain protein 4) gene, which is downregulated in the hypothalamus of the control WAG rats and induced in the hypothalamus of hypertensive ISIAH rats, is suggested to be the key event for understanding inter-strain differences in the hypothalamic response to stress. The stress-dependent ISIAH strain-specific induction of Fos and Jun gene transcription may play a crucial role in neuronal activation in this rat strain. The data obtained can be potentially useful in the selection of molecular targets for the development of pharmacological approaches to the correction of stress-induced pathologies related to neuronal excitability, taking into account the hypertensive status of the patients.

Introduction. The Upper-Kama dialect is one of the varieties of the Komi language, which is rather common in the Afanasyevsky area of the Kirov region. The dialect is an intermediate idiom between the Komi-Permian and Komi-Zyryan languages. The purpose of the paper is to analyze the Komi-Permian and Komi-Zyryan components in the lexical system of the dialect. Materials and Methods. The material for the study was collected during dialectological expeditions to the area of residence of the Kirov Permians in 2002–2012. It was collected from native speakers with a high level of language competence. The research was carried out using descriptive, comparative and statistical methods. Results and Discussion. The vocabulary of the Upper-Kama dialect is heterogeneous. The most numerous group of words common for the Komi-Permian and Komi-Zyryan dialects is distinguished. Lexemes in the Upper Kama dialects and at a larger territory in the Komi-Zyryan language, but are absent in the Komi-Permian one. There is also a group of words which correspondences are recorded in the southern Komi-Zyryan dialects: Mid-Sysola, Luza-Letka and mainly in the Upper-Sysola one. On the territory of the Upper-Kama idiom there are the lexemes, which isoglosses cover the Upper-Kama and Komi-Permian dialects. However, there are not so many similar Komi-Permian-Upper-Kama isoglosses; most of them can be found in the Zyryan dialects. As a result of the analysis of the texts in the Upper-Kama dialect, it turned out that the overwhelming majority of lexemes are found in all varieties of Komi languages. Conclusion. The analysis of the lexical composition of the Upper-Kama dialect has shown that the specified idiom cannot be unconditionally attributed to the Komi-Permian language, since the correlation of the Komi-Zyryan and Komi-Permian lexical features in it is approximately the same.

The article investigates the interaction between the memory phenomenon and avant-garde architecture by reviewing international and Soviet examples of how memory worked in the 1920-30s architecture, reminiscences of the avant-garde in the 20th-21st century architecture, and the latest inclusions of modernist and avant-garde architecture in current memory. Today, the avant-garde architecture occupies a place in the history of architecture not simply as a practice of denying the past and pursuing innovation, but as an era of fascination with great ideas based on a strong cultural foundation that made it possible to dream of the future. The avant-garde era played a great role in the formation of the entire architecture of the Modern Movement, and definition of the values and paradigms of modernity. The contemporary architectural avant-garde of the 21st century, for which one of the main tasks is the reception and translation of memory, serves as an embodiment of search for harmonious interaction between the past and the present. Examples are considered and conclusions are drawn about the nature of the interaction between avant-garde architecture and the phenomenon of memory.

The article is devoted to the journey of the writer-ethnographer S. V. Maksimov in the Vladimir province, which took place in the summer of 1855 during his first large expedition — in the Vladimir, Nizhny Novgorod and Vyatka provinces. Despite the fact that Maximov’s impressions were not immediately reflected and formed into a framed text, they had a significant impact on the writer’s work. Comparison of the first published archival manuscript of Maximov, dedicated to the crafts of Sudogodsky district, and the books “In the East” (1864), “Siberia and penal servitude” (section “Prison Dictionary”) (1871), “Wandering Russia for Christ’s Sake” (1877), “Winged Words. It is not for nothing and not for nothing that the word is said and will not break until the century” (1899), as well as the involvement of sources used by Maximov, demonstrates the features of the writer’s creative laboratory, the transformation of factual material into an artistic text. The study of the results of Maksimov’s journey allows to speak about the significant role of the trip both for Maksimov’s creativity (in terms of the accumulation of aterial to which he will repeatedly refer, the formation of the principles of writing and ethnographic work, etc.) and for the local history of the Vladimir Region.

Problem and goal. The training of future teachers in the field of education involves the formation of competencies in the development of software for various purposes, the design of automated control systems for real objects. The development of relevant qualities in the conditions of higher education is complicated by a number of problems of various nature. The authors suggest to improve the training of future specialists to organize project activities of students to create dialogue programs (training bots). Methodology. Project-based training is used for the formation of competencies in the field for informatization of education, obtaining in-demand soft skills, team building. The construction of a chatbot takes place in teamwork. The experiment involved 43 students of the North Caucasus Federal University in the direction of training 44.03.05 “Pedagogical Education with Two Training Profiles.” The assessment of the quality of educational results was carried out using the author's testing of 10 tasks (“credited”/“not credited”). Fischer's criterion was used to establish statistically significant differences. Results. Students studied the services for creating chatbots, the didactic potential and functionality of the received programs, used them to solve problems. Statistically significant differences were revealed in the changes that occurred in the system in terms of the quality of vocational training. The features of the presented variant of the organization of educational activities for the creation of chatbots are described: teamwork, the use of templates and scripts, and the choice of project topics. Conclusion. Problematic questions are formulated, the answers to which allow us to determine the directions of work on the construction of chatbots: discussion of the development goal and didactic goal, the use of blocks and actions, modeling interaction scenarios, etc.
