Abstract Background This study investigated the antidiabetic and antioxidant properties of hemp seed oil using various bioanalytical methods. Furthermore, this study determined the suppressive properties of hemp seed oil on α-amylase, acetylcholinesterase and carbonic anhydrase II that purified by the sepharose-4B-L-Tyrosine-sulfanilamide affinity chromatoghraphy, all of which are related to different metabolic diseases. Moreover, the phenolic concentration in the essential oil was quantified through LC–HRMS chromatography. Thirteen distinct phenolic compounds were detected in hemp seed oil. Additionally, both the chemical components and quantity of essential oils within hemp seed oil were assessed through GC–FID and GC/MS analyses. Results The predominant essential oils in hemp seed oil included linoleoyl chloride (34.62%), linoleic acid (33.21%), and 2-4-di-tert-butylphenol (5.79%). Hemp seed oil's ability to scavenge radicals was studied through the use of 2,2’-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) and 1,1-diphenyl-2-picrylhydrazil bioanalytical radical scavenging methods. The results unveiled its potent radical-scavenging properties, with an 46.20 μg/mL for 2,2’-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) radicals and IC50 of 9.76 μg/mL for 1,1-diphenyl-2-picrylhydrazil radicals. The investigation also extended to explore the reducing capabilities of Fe3+-2,4,6-tri(2-pyridyl)-S-triazine, copper (Cu2+), and iron (Fe3+). Hemp seed oil demonstrated notable inhibitory effect against α-amylase (IC50: 545.66 μg/mL), achethylcholinesterase (IC50: 28.00 μg/mL), and carbonic anhydrase II (IC50: 322.62 μg/mL). Conclusions This interdisciplinary research will prove valuable and set the stage for future investigations into the antioxidant characteristics and enzyme inhibition patterns of plants and plants oils that hold medical and industrial significance.

Determining the antioxidant abilities and enzyme inhibition profiles of medicinally important plants and their oils is of great importance for a healthy life and the treatment of some common global diseases. Kiwifruit (Actinidia deliciosa) oil was examined and researched using several bioanalytical methods comprehensively for the first time in this research to determine its antioxidant, antiglaucoma, antidiabetic and anti-Alzheimer’s capabilities. Additionally, the kiwifruit oil inhibitory effects on acetylcholinesterase (AChE), carbonic anhydrase II (CA II), and α-amylase, which are linked to a number of metabolic illnesses, were established. Furthermore, LC-HRMS analysis was used to assess the phenolic content of kiwifruit oil. It came to light that kiwifruit oil contained 26 different phenolic compounds. According to the LC-HRMS findings, kiwifruit oil is abundant in apigenin (74.24 mg/L oil), epigallocatechin (12.89 mg/L oil), caryophyllene oxide (12.89 mg/L oil), and luteolin (5.49 mg/L oil). In addition, GC-MS and GC-FID studies were used to ascertain the quantity and chemical composition of the essential oils contained in kiwifruit oil. Squalene (53.04%), linoleoyl chloride (20.28%), linoleic acid (2.67%), and palmitic acid (1.54%) were the most abundant compounds in kiwifruit oil. For radical scavenging activities of kiwifruit oil, 1,1-diphenyl-2-picryl-hydrazil (DPPH•) and 2,2′-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) (ABTS•+) radicals scavenging techniques were examined. These methods effectively demonstrated the potent radical scavenging properties of kiwifruit oil (IC50: 48.55 μg/mL for DPPH•, and IC50: 77.00 μg/mL for ABTS•+ scavenging). Also, for reducing capabilities, iron (Fe3+), copper (Cu2+), and Fe3+-2,4,6-tri(2-pyridyl)-S-triazine (TPTZ) reducing abilities were studied. Moreover, kiwifruit oil showed a considerable inhibition effect towards hCA II (IC50: 505.83 μg/mL), AChE (IC50: 12.80 μg/mL), and α-amylase (IC50: 421.02 μg/mL). The results revealed that the use of kiwifruit oil in a pharmaceutical procedure has very important effects due to its antioxidant, anti-Alzheimer, antidiabetic, and antiglaucoma effects.

Many chronic diseases such as diabetes and Alzheimer’s disease are related to the type and quality of foods, which are consumed. Particularly, various plant origin products are stated as beneficial against such kind of chronic diseases with secondary metabolites such as their phenolic structures. Satureja cuneifolia is a plant, which is consumed as an herbal tea in some regions of Turkey and that’s why investigate of its biological activity is important. In our study, the anti-diabetic and anti-Alzheimer potentials of the methanol and water extracts of S. cuneifolia plant were measured via some enzymes inhibition experiments as in vitro. The antioxidant ability of the same extracts was measured via radical scavenging and reducing power methods. Also, the total phenolics and flavonoids of the plant were identified. Finally, the extracts were analyzed by the LC-MS/MS analysis and the phenolic content of S. cuneifolia was clarified. In this study, for the first time, the antioxidant and antidiabetic properties of the essential oil from cinnamon (Cinnamomum zeylanicum) leaves were evaluated and investigated using various bioanalytical methods. In addition, the inhibitory effects of cinnamon oil on carbonic anhydrase II (hCA II), acetylcholinesterase (AChE), and α-amylase, which are associated with various metabolic diseases, were determined. Further, the phenolic contents of the essential oil were determined using LC-HRMS chromatography. Twenty-seven phenolic molecules were detected in cinnamon oil. Moreover, the amount and chemical profile of the essential oils present in cinnamon oil was determined using GC/MS and GC-FID analyses. (E)-cinnamaldehyde (72.98%), benzyl benzoate (4.01%), and trans-Cinnamyl acetate (3.36%) were the most common essential oils in cinnamon leaf oil. The radical scavenging activities of cinnamon oil were investigated using 1,1-diphenyl-2-picryl-hydrazil (DPPH•), 2,2′-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid), and (ABTS•+) bioanalytical scavenging methods, which revealed its strong radical scavenging abilities (DPPH•, IC50: 4.78 μg/mL; and ABTS•+, IC50: 5.21 μg/mL). Similarly, the reducing capacities for iron (Fe3+), copper (Cu2+), and Fe3+-2,4,6-tri(2-pyridyl)-S-triazine (TPTZ) were investigated. Cinnamon oil also exhibited highly effective inhibition against hCA II (IC50: 243.24 μg/mL), AChE (IC50: 16.03 μg/mL), and α-amylase (IC50: 7.54μg/mL). This multidisciplinary study will be useful and pave the way for further studies for the determination of antioxidant properties and enzyme inhibition profiles of medically and industrially important plants and their oils.

Cress (Lepidium sativum) is an annual herb from Brassicaceae family, and in some regions. It is known as peppergrass, garden cress, garden peppercress, pepperwort, or poor man’s pepper. Cress is an important medicinal plant in some countries including Turkey. This plant has major scientific and therapeutic significance. Peroxidase (POD, E.C.1.11.1.7) is an oxidoreductase enzyme produced by a number of organisms. In this study, POD enzyme was purified from cress by ammonium sulphate precipitation, gel filtration, and CM-Sephadex ion-exchange chromatographies. Km and Vmax values were calculated from the Lineweaver-Burk graph for H2O2 and guaiacol substrates and the substrate specificity of POD was investigated. The results showed that substrate specificity of H2O2 is better than substrate specificity of guaiacol for this enzyme. The inhibition effects of three cations (Al3+, Cu2+, and Cr3+) and one organic compound of cetyl trimethylammonium bromide were performed and their inhibition types were determined. Finally, optimum pH, optimum temperature, optimum ionic strength, and stable pH were determined.

Objectives Sumac (Rhus coriaria L.) is widely used spice which has several properties such as antioxidant, anti-inflammatory and antimicrobial. The purpose of this animal study was to evaluate the effects of sumac extract on levels of receptor activator of nuclear factor-kappa B ligand (RANKL), osteoprotegerin (OPG) expression, serum oxidative status, and alveolar bone loss in experimental periodontitis. Material and Methods Twenty-four Wistar rats were separated into three groups: non-ligated (NL, n=8), ligature only (LO, n=8), and ligature and treated with sumac extract (S, n=8) (20 mg/kg per day for 11 days). A 4/0 silk suture was placed around the mandibular right first molars subgingivally; after 11 days, the rats were sacrificed, and alveolar bone loss was histometrically measured. The detection of RANKL and OPG were immunohistochemically performed. Levels of serum total antioxidant status (TAS)/total oxidant status (TOS), and oxidative stress index (OSI) were also analyzed. Results Alveolar bone loss was significantly greater in the LO group compared to the S and NL groups (p0.05). Conclusions The present study showed that systemic administration of sumac extract may reduce alveolar bone loss by affecting RANKL/OPG balance, TOS and OSI levels in periodontal disease in rats.

The purpose of this animal study was to evaluate the effects of hawthorn (Crataeus orientalis M Bieber.) extract on serum oxidative status and alveolar bone loss in experimental periodontitis. Twenty-seven Wistar rats were assigned to one of the following groups: non- ligated+placebo (saline) (NL, n = 9), ligature only+placebo (saline) (LO, n = 9), and ligature and treated with hawthorn extract in saline (H, n = 9) (100 mg/kg orogastrically, once a day for 11 days). Periodontitis was induced by submerging a 4/0 silk ligature in the sulcus of the mandibular right first molars of rats, and the animals were sacrificed after 11 days. Micro-CT examinations were performed for linear and volumetric parameter assessment of alveolar bone. Periodontal tissues were histopathologically examined to assess the differences among the study groups. Continuing our work on the sources of natural bioactive compounds, we evaluated the antimicrobial and antioxidant activities of Nepeta trachonitica as well as its major phenolic content using the high-performance liquid chromatography-mass spectrometry/mass spectrometry (HPLC-MS/MS) technique. For antioxidant activity, ferric reducing antioxidant power (FRAP) and cupric ion reducing antioxidant capacity (CUPRAC) methods were performed to measure the reducing power and 1,1-diphenyl-2-picrylhydrazyl (DPPH) assay was employed to evaluate the radical scavenging activity of the sample. For antimicrobial activity, three Gram-positive and four Gram-negative microbial species as well as three fungi species were tested. N. trachonitica appeared to have reasonable antioxidant activity and decent antimicrobial activity as indicated by the inhibition of the organisms’ growth. The most susceptible species were Bacillus subtilis ATCC 6633 and Escherichia coli ATCC 11229 among the organisms tested. Ethanol extract of the plant has the highest effect on Saccharomyces cerevisiae but no effect on Yarrowia lipolytica. The HPLC-MS/MS analysis showed that at least 11 major phenolic compounds of N. trachonitica exist, the major ones being rosmarinic acid, chlorogenic acid and quinic acid. The obtained results suggest that N. trachonitica could be a promising source for food and nutraceutical industries because of its antimicrobial and antioxidant properties and phenolic compounds.

The identification and quantification of the phenolic contents of methanolic extracts of three Salvia L. species namely S. brachyantha (Bordz.) Pobed, S. aethiopis L., and S. microstegia Boiss. and Bal. were evaluated using reverse phase high performance liquid chromatography, UV adsorption, and mass spectrometry (RP-HPLC/MS). In order to determine the antioxidant capacity of these species, cupric ions (Cu2+) reducing assay (CUPRAC) and ferric ions (Fe3+) reducing assay (FRAP) were performed to screen the reducing capacity and 1,1-diphenyl-2-picrylhydrazyl (DPPH) assay was employed for evaluation of the radical scavenging activity for both solvents. In further investigation, the antimicrobial activities of Salvia species were tested using the disc diffusion method against three Gram-positive and four Gram-negative microbial species, as well as three fungi species. The results showed that there is a total of 18 detectable phenols, the most abundant of which was kaempferol in S. microstegia and rosmarinic acids in S. brachyantha and S aethiopis. The other major phenols were found to be apigenin, luteolin, p-coumaric acid, and chlorogenic acid. All species tested showed moderate and lower antioxidant activity than standard antioxidants such as butylated hydroxyanisole (BHA), butylated hydroxytoluene (BHT), and ascorbic acid. The ethanolic extracts of Salvia species revealed a wide range of antimicrobial activity. S. brachyantha and S. microstegia showed the highest antimicrobial activities against B. subtilis, whereas S. aethiopis was more effective on Y. lipolytica. None of the extracts showed anti-fungal activity against S. cerevisiae. Thus these species could be valuable due to their bioactive compounds.

In the first stage of the manuscript, we aimed to examine antioxidant capacity and anticholinergic properties of avocado (Folium perseae) leaves. Avocado leaf was extracted by water (WEFP) and ethyl alcohol (EEFP) and antioxidant activity was determined using by several antioxidant assays including DPPH· and ABTS•+ radical scavenging assays, Cu2+-Cu+ reducing, Fe3+-Fe2+ reducing, and FRAP reducing activities. Avocado leaf extracts demonstrated antioxidant activity and anticholinergic activities, while α-tocopherol, BHT, trolox, and BHA were used as positive antioxidant controls. In the second part of this study, the inhibition effects of WEFP and EEFP were valuated against acetylcholinesterase and butyrylcholinesterase enzymes, which catalyze the breakdown of choline esters (i.e. neurotransmitters). This study obviously showed that avocado leaf extracts had effective antioxidant, antiradical, and anticholinergic influences.

Abstract  Recently, a number of studies on health benefits associated with natural compounds have been demonstrated. Phenolics in fruits, vegetables, herbs and spices possess potent antioxidant, anti-inflammatory, antimutagenic and anticarcinogenic activities. The fringe tree (Chionanthus virginicus) is used as a raw material by pharmaceutical industries for the preparation of homeopathy tinctures. The potential antioxidant activities of two secoiridoids from root bark of fringe tree (Chionanthus virginicus L.) were investigated to evaluate their potential value as the natural products for foods or cosmetic applications. In this study, antioxidant activities were measured by 2,2-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) (ABTS) radical scavenging, 1,1-diphenyl-2-picryl-hydrazyl free radical (DPPH·) scavenging, superoxide anion (O2·−) radical scavenging, total antioxidant activity, reducing activity, hydrogen peroxide (H2O2) scavenging and ferrous metal chelating activity assays. These secoiridoids, as antioxidants neutralized the activities of radicals and inhibited the peroxidation reactions of linoleic acid emulsion. Total antioxidant activity was measured according to ferric thiocyanate method. Butylated hydroxyanisole (BHA), butylated hydroxytoluene (BHT), α-tocopherol and trolox, a water-soluble analog of tocopherol, were used as the reference antioxidant compounds. Ligustroside (3.70 × 10−3 M) and oleuropein (3.80 × 10−3 M) showed 71.9, 82.4, 80.7 and 90.4% inhibition on lipid peroxidation of linoleic acid emulsion, at the concentrations of 10 and 20 μg/mL. On the other hand, 20 μg/mL of standard antioxidant such as α-tocopherol (4.64 × 10−3 M), trolox (7.98 × 10−3 M), BHA (10.08 × 10−3 M) and BHT (9.06 × 10−3 M) exhibited 61.5, 29.8, 74.4 and 71.2% inhibition on peroxidation of linoleic acid emulsion, respectively. In addition, ligustroside and oleuropein had effective DPPH·, ABTS·+ and superoxide anion radicals scavenging, hydrogen peroxide scavenging, total reducing power and metal chelating on ferrous ions activities. Also, those various antioxidant activities were compared to BHA and BHT, α-tocopherol and trolox that are references antioxidants. [ABSTRACT FROM AUTHOR]
