Archaeological excavations carried out in the town of Palermo revealed important traces of metallurgical activity related to the Punic Panormos. Five samples of iron slags, recovered during the digging, were characterized by micro-structural, mineralogical and chemical investigations as well as by environmental scanning electron microscopy, energy-dispersive X-ray spectroscopy, X-ray powder diffraction and X-ray fluorescence. The studied remains are spongy, rust colored, with a plano-convex shape and their textural and mineralogical features suggest they are metallurgical slags produced during smithing process. Wüstite, magnetite, fayalite, kirschsteinite, hedembergite, cristobalite and quartz are the main identified mineral phases, while goethite, lepidocrocite and calcite occur as minor secondary phases. The iron slags show heterogeneous structures such as hammering clues, layering in wustite crystals and presence of calcium-rich minerals which are consistent with a broad variation of the forging temperature due to the use of different smithing techniques. This assumption is confirmed by the different crystallization temperatures of the detected mineral phases. Skeletal and dendritic shapes are indicative of rapid cooling and, possibly, quenching. The chemical composition is dominated by iron, silicon and calcium and it is almost constant for all the samples suggesting that they were produced with similar raw materials and technology. The high Ca content suggests the use of carbonate-bearing rocks, widely outcropping over the Palermo territory, probably exploited as fluxing ores for the metallurgical process. The multidisciplinary approach adopted in this study provided new data for a better understanding of the metallurgical techniques evolution in the Sicilian territory in ancient times. Since the studied iron slags represent the unique traces so far known of metallurgical activity in the ancient Palermo town, our results open interesting perspectives for future study of metallurgy in ancient Sicily. One of the most impressive Sicilian pottery production is attributed to the so-called Lipari Painter and his followers, whose vessels-found in the archeological site of Lipari (Aeolian Island, Sicily)-are decorated with characteristic blue, red, and white figures. From the archeological point of view, these artworks keep open many questions concerning dating, production technique, and cultural background. In this context, new data on the manufacture procedures and on the raw materials used for the pigments may contribute to a deeper comprehension of this early Hellenistic vase tradition. The preciousness of the vessels, exhibited at the Archeological Museum of Lipari, imposed the use of in situ nondestructive methods to address new insights on the nature of the colored layers. Thus, analyses by Raman and X-ray fluorescence spectroscopy have been performed with portable instruments on a selection of vessels certainly attributed to Lipari Painter and to some others of his followers. The results of this study testify the use of different pigments: kaolin and gypsum, probably supplied locally, for white layers; Egyptian blue for blue hues; red ochre for brown-reddish hues; and cinnabar for pink and red-purple nuances. Ammonium- and Fe( II)-rich fluid flows, known from deep-sea hydrothermal systems, have been extensively studied in the last decades and are considered as sites with high microbial diversity and activity. Their shallow-submarine counterparts, despite their easier accessibility, have so far been under-investigated, and as a consequence, much less is known about microbial communities inhabiting these ecosystems. A field of shallow expulsion of hydrothermal fluids has been discovered at depths of 170-400 meters off the base of the Basiluzzo Islet (Aeolian Volcanic Archipelago, Southern Tyrrhenian Sea). This area consists predominantly of both actively diffusing and inactive 1-3 meters-high structures in the form of vertical pinnacles, steeples and mounds covered by a thick orange to brown crust deposits hosting rich benthic fauna. Integrated morphological, mineralogical, and geochemical analyses revealed that, above all, these crusts are formed by ferrihydrite-type Fe3+ oxyhydroxides. Two cruises in 2013 allowed us to monitor and sampled this novel ecosystem, certainly interesting in terms of shallow-water iron-rich site. The main objective of this work was to characterize the composition of extant communities of iron microbial mats in relation to the environmental setting and the observed patterns of macrofaunal colonization. We demonstrated that iron-rich deposits contain complex and stratified microbial communities with a high proportion of prokaryotes akin to ammonium- and iron-oxidizing chemoautotrophs, belonging to Thaumarchaeota, Nitrospira, and Zetaproteobacteria. Colonizers of iron-rich mounds, while composed of the common macrobenthic grazers, predators, filter-feeders, and tube-dwellers with no representatives of vent endemic fauna, differed from the surrounding populations. Thus, it is very likely that reduced electron donors (Fe2+ and NH4+) are important energy sources in supporting primary production in microbial mats, which form a habitat-specific trophic base of the whole Basiluzzo hydrothermal ecosystem, including macrobenthic fauna. The aim of this work is obtaining information on the provenance of some very peculiar green (or blackgreen) stones used both as façade and floor decorations in the Messina Cathedral. Historical sources and available literature data suggest that these materials could belong to the amphibolite of the Peloritani Mountains. In order to verify the true provenance, some stone samples were studied through various analytical methods (SEM-EDS, XRPD, XRF). The investigated specimens resulted to be metabasites showing pseudomorphic and vein textures, with relicts of olivine replaced by serpentine - forming a mesh textures - and orthopyroxene mostly altered to bastite. Pseudomorphic minerals are serpentine (lizardite and chrysotile), magnetite, amphibole (tremolite-actinolite series) and chlorite (talc-chlorite). These results demonstrate that the analysed green stones cannot belong to the amphibolite outcrop of the Peloritani Mountains. Otherwise, they show mineralogical and petrographic features very similar to those of "verde Calabria", a rock typology largely exposed in the Northern Calabria area - belonging to the ophiolitic series - and widely used in the last century as building material. Adjustable mirrors equipped with piezo actuators are commonly used at synchrotron and free-electron laser (FEL) beamlines, in order to optimize their focusing properties and sometimes to shape the intensity distribution of the focal spot with the desired profile. Unlike them, X-ray mirrors for astronomy are much thinner in order to enable nesting and reduce the areal mass, and the application of piezo actuators acting normally to the surface appears much more difficult. There remains the possibility to correct the deformations using thin patches that exert a tangential strain on the rear side of the mirror: some research groups are already at work on this approach. The technique reported here relies on actively integrating thin glass foils with commercial piezoceramic patches, fed by voltages driven by the feedback provided by X-rays, while the tension signals are carried by electrodes on the back of the mirror, obtained by photolithography. Finally, the shape detection and the consequent voltage signal to be provided to the piezoelectric array will be determined by X-ray illumination in an intra-focal setup at the XACT facility. In the early period, even though professional photographers worked with similar techniques and products, their artistic and commercial aims determined different choices and led them to follow different, often personal, recipes. For this reason, identification of the techniques through date and name of the photographer or through some visual features like colour, tonality and surface of the image layer, often needs further investigation to be proved. Chemical characterization, carried out in a non or micro destructive way, can be crucial to provide useful information about the original composition, degradation process, realization technique, in obtaining an indirect dating of the photograph and/or to choose the most correct conservation treatment. In our case, x-ray fluorescence (XRF) analysis was used to confirm the chemical composition of eleven historical photographs dated between the end of the 19th century and the beginning of the 20th, shot in Palermo (Sicily) by a renowned photographer of the time, and pasted on their original cardboards. The elemental identification, obtained with a non destructive approach, provided important information to distinguish among different photographic techniques in terms of distribution and characterization of chemical elements markers in the photographic surface. Kaposi sarcoma herpesvirus/human herpesvirus-8 (HHV-8) neoplastic and non-neoplastic disease in solid organ transplant (SOT) recipients can be life-threatening. We evaluated the seroprevalence of HHV-8 infection among donors (D) and recipients (R), the incidence of HHV-8 transmission/reactivation, and the clinical characteristics, management, and outcomes of HHV-8-related diseases, including Kaposi sarcoma herpesvirus-associated inflammatory cytokine syndrome (KICS), in consecutive SOT patients from 2011 to 2023. HHV-8 seroprevalence was 3.3% in 1349 donors and 8.4% in 1856 recipients screened (p<0.0001). Among the D+/R- group (n=49), 13 patients developed HHV-8-related diseases: 7 liver recipients had KICS, and 1 lung recipient had Kaposi sarcoma (KS) with subsequent KICS. Four KICS patients treated with rituximab survived, while the 3 patients not treated with rituximab died. Within the D-/R- group, out of 5 (0.3%) patients with non-donor-derived primary HHV-8 infection, 3 liver recipients developed KICS. In the R+ patients (n=155), 3 developed KS. In our cohort, 25/944 (2.6%) liver transplant recipients had a primary HHV-8 infection, and 10 of them (40%) developed KICS; 40% (4/10) of HHV-8 seropositive heart transplant recipients developed reactivation and 2 of them (50%) had fatal KS. Background and Objectives: Few preclinical models of pseudomyxoma peritonei (PMP) have been developed, probably due to the tumor's low incidence and its peculiar characteristics of slow growth. Therefore, there is a need to develop more refined PMP models that better reflect its characteristics. The aim of the study is to develop a culture strategy to generate organoid models derived from PMP patient samples., Methods: We followed a strategy based on combinatorial culture conditions that include the different factors essential for PMP growth and that mimic the microenvironment present in the patients., Results: We cultured PMP samples in the presence of the various factors produced by the niche environment of PMP. We obtained 12 PMP organoid models, each of which grows under specific culture conditions. PMP-derived organoids show long-term expansion capacity and reproduce the genetic landscape and histological phenotype of the tumor of origin., Conclusion: The organoids we developed faithfully reproduce the key features of PMP disease and will allow us to understand the biology of PMP. The archipelago of the Aeolian Islands in the Tyrrhenian Sea is a globally important natural laboratory. The archipelago, declared a UNESCO World Heritage Site for its unique geology and biodiversity, offers a unique opportunity to study plastic pollution. This study presents an initiative to assess the occurrence of plastic pellets on the beaches of five Aeolian Islands. It provides an insight into the polymer composition and the effects of degradation. Collected pellets were analyzed using stereomicroscopy and Fourier transform infrared spectroscopy (FTIR). Hierarchical cluster analysis (HCA) based on the results of the FTIR data has proved to be an effective statistical method in identifying different clusters corresponding to different degradation phases of the collected pellets. The infrared analysis identified polyethylene (80%) as the main polymer, with a small amount of polypropylene (20%). It was found that the surfaces of some pellets undergo changes during weathering that alter the polymer surfaces. By combining data on plastic pellets from the Aeolian Islands and surrounding coastal areas, we are gaining a more comprehensive understanding of the distribution patterns of microplastics. This paper examines the distribution and chemical properties of beached plastic pellets along the Ionian and Tyrrhenian coasts of Southern Italy. Three locations have been sampled: Agnone Bagni (SR) and Paradiso (ME) on the Ionian coast of Sicily, Baia del Tono in Milazzo (ME) on the Sicilian Tyrrhenian coast, and Pizzo Calabro (VV) in Calabria on the Tyrrhenian coast. Variations in shape, size, compactness, color, and other physical features, correlated with residence times and transport, has been highlighted. Raman spectroscopy, used in a portable configuration, enabled rapid identification of polymer types, demonstrating its utility for on-site plastic pollutant monitoring. Polyethylene and polypropylene were the predominant polymers. Principal component analysis of the spectra determined the optimal chemometric classification of pellets by composition, avoiding interference or distortion. Proteolytic release of transmembrane proteins from the cell surface, the so called ectodomain shedding, is a key process in inflammation. Inactive rhomboid 2 (iRhom2) plays a crucial role in this context, in that it guides maturation and function of the sheddase ADAM17 (a disintegrin and metalloproteinase 17) in immune cells, and, ultimately, its ability to release inflammatory mediators such as tumor necrosis factor α (TNFα). Yet, the macrophage sheddome of iRhom2/ADAM17, which is the collection of substrates that are released by the proteolytic complex, is only partly known. In this study, we applied high-resolution proteomics to murine and human iRhom2-deficient macrophages for a systematic identification of substrates, and therefore functions, of the iRhom2/ADAM17 proteolytic complex. We found that iRhom2 loss suppressed the release of a group of transmembrane proteins, including known (e.g. CSF1R) and putative novel ADAM17 substrates. In the latter group, shedding of major histocompatibility complex class I molecules (MHC-I) was consistently reduced in both murine and human macrophages when iRhom2 was ablated. Intriguingly, it emerged that in addition to its shedding, iRhom2 could also control surface expression of MHC-I by an undefined mechanism. We have demonstrated the biological significance of this process by using an in vitro model of CD8 + T-cell (CTL) activation. In this model, iRhom2 loss and consequent reduction of MHC-I expression on the cell surface of an Epstein-Barr virus (EBV)-transformed lymphoblastoid cell line dampened activation of autologous CTLs and their cell-mediated cytotoxicity. A previously undocumented shallow water hydrothermal field from Sicily (Southern Tyrrhenian Sea, Italy) is here described, based on a multidisciplinary investigation. The field, covering an area of nearly 8000 m2 and a depth from the surface to -5 m, was explored in June 2021 to characterise the main physico-chemical features of the water column, describe the bottom topography and features, and identify the main megabenthic and nektonic species. Twenty sites were investigated to characterise the carbonate system. Values of pH ranged between 7.84 and 8.04, ΩCa between 3.68 and 5.24 and ΩAr from 2.41 to 3.44. Geochemical analyses of hydrothermal gases revealed a dominance of CO2 (98.1%) together with small amounts of oxygen and reactive gases. Helium isotope ratios (R/Ra = 2.51) and δ13CCO2 suggest an inorganic origin of hydrothermal degassing of CO2 and the ascent of heat and deep-seated magmatic fluids to the surface. Visual census of fishes and megabenthos (mainly sessile organisms) allowed the identification of 64 species, four of which are protected by the SPA/BIO Protocol and two by the International Union for Conservation of Nature. The macroalgae Halopteris scoparia and Jania rubens and the sponge Sarcotragus sp. were the dominant taxa in the area, while among fishes Coris julis and Chromis chromis were the most abundant species. At present, there is a lack of clinical evidence about the impact and long-term durability of the immune response induced by the third dose of mRNA vaccines. In this study, we followed up the B cell compartment behavior in a cohort of immunocompetent individuals three and six months after the third dose of vaccine. During this period, some subjects contracted the virus. In uninfected vaccinated subjects, we did not report any changes in serum spike-specific IgG levels, with a significant reduction in IgA. Instead, subjects recovered from natural infection showed a significant increase in both specific IgG and IgA. Moreover, we showed a time-related decrease in IgG neutralizing potential to all SARS-CoV-2 variants of concern (VOC) in uninfected compared to recovered subjects, who displayed an increased neutralizing ability, particularly against the omicron variant. Finally, we underlined the presence of a pool of SARS-CoV-2-specific B cells in both groups that are prone to respond to restimulation, as demonstrated by their ability to differentiate into plasma cells and to produce anti-SARS-CoV-2-specific immunoglobulins. These data lead us to assert the long-term effectiveness of the BNT162b2 vaccine in contrasting the severe form of the pathology and prevent COVID-19-associated hospitalization.

Solid organ transplant recipients (SOTRs) show higher rates of COVID-19 breakthrough infection than the general population, and nowadays, vaccination is the key preventative strategy. Nonetheless, SOTRs show lower vaccine efficacy for the prevention of severe COVID-19. Moreover, the emergence of new SARS-CoV-2 variants of concern has highlighted the need to improve vaccine-induced immune responses by the administration of repeated booster doses. In this study, we analyzed the humoral and cellular responses in a cohort of 25 SOTRs, including 15 never-infected SOTRs who received the fourth dose of the mRNA vaccine and 10 SOTRs who contracted SARS-CoV-2 infection after the third dose. We analyzed the serum IgG and IgA levels through CLIA or ELISA, respectively, and the Spike-specific T cells by ELISpot assay. We report a significant increase in anti-Spike IgG and no differences in IgA secretion in both groups of patients before and after the booster dose or the natural infection. Still, we show higher IgA levels in recovered SOTRs compared to the fourth dose recipients. Conversely, we show the maintenance of a positive Spike-specific T-cell response in SOTRs who received the fourth dose, which, instead, was significantly increased in SOTRs who contracted the infection. Our results suggest that the booster, either through the fourth dose or natural infection, in vulnerable poor responder SOTRs, improves both humoral and cellular-specific immune responses against SARS-CoV-2.

To prevent PGI (Protected Geographical Indication) cherry tomato of Pachino (Sicily, Italy) from frauds, an alternative method, which includes chemometric treatments, was proposed. The content of 32 inorganic elements (macro-micronutrients and lanthanides) present in 16 PGI and 24 not PGI cherry tomato samples cv Naomy, and in 16 PGI and 8 not PGI soil samples, was determined by Inductively Coupled Plasma - Mass Spectrometer (ICP-MS). To identify the elements able to differentiate PGI and not PGI cherry tomato samples, Principal Components Analysis (PCA) and Canonical discriminant analysis (CDA) were performed. The first two principal components (PC1-PC2) explain a total variance of 71,41% between PGI and not PGI group, whereas CDA showed Zn, Cd, Mn and Ca as inorganic markers able to correctly classify the 100% of samples. Background: Several studies have indicated that anti-SARS-CoV-2 mRNA vaccinations are less effective in inducing robust immune responses among solid organ transplant recipients (SOTRs) compared with the immunocompetent. The third dose of vaccine in SOTRs showed promising results of immunogenicity, even though clinical studies have suggested that immunocompromised subjects are less likely to build a protective immune response against SARS-CoV-2 resulting in lower vaccine efficacy for the prevention of severe COVID-19. Methods: Serological IgG and IgA were analyzed through CLIA or ELISA, respectively, while Spike-specific T cells were detected by ELISpot assay after the second and third dose of vaccine in 43 SOTRs. Results: The third dose induced an improvement in antibody response against SARS-CoV-2. We also reported a strong correlation between specific humoral and cellular responses after the third dose, even though we did not see significant changes in the magnitude of the SARS-CoV-2-specific T cell response. SOTRs who contracted the SARS-CoV-2 infection after the third dose, despite eliciting a positive IgG response, failed to mount an anti-Spike-S1 IgA response, both after the third dose and after SARS-CoV-2 infection. Conclusions: We can conclude that serum IgA detection can be helpful, along with IgG detection, for the evaluation of vaccine efficacy, principally in fragile subjects at high risk of infection.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), which causes coronavirus disease 2019 (COVID-19), is modifying human activity all over the world with significant health and economic burden. The advent of the SARS-CoV-2 pandemic prompted the scientific community to learn the virus dynamics concerning transmissibility, epidemiology, and usefulness of vaccines in fighting emerging health hazards. Pieces of evidence suggest that the first and second doses of mRNA vaccines induce a significant antibody response in vaccinated subjects or patients who recovered from SARS-CoV-2 infection, demonstrating the importance of the previously formed memory. The aim of this work has been to investigate the effects of BNT162b2 Pfizer-BioNTech mRNA-based vaccine booster dose in a cohort of 11 uninfected immunocompetent (ICs), evaluating the humoral and cellular responses, with more carefulness on memory B and T cells.
