Caragana korshinskii kom. (CKK) waste, a common forestry byproduct in northwest of China, presents challenges in its transformation into alternative ruminant feed due to its initial nutritional limitations and unappealing palatability. Conventional strategies, such as ensiling and fungal-based solid-state fermentation (SSF) cannot effectively address this issue in practice. Herein, a two-stage bioaugmentation (TBA) process was devised, leveraging the benefits of ensiling and SSF. During the anaerobic ensiling phase, CKK waste was inoculated with Lactiplantibacillus plantarum LP1, effectively suppressing potential animal pathogens such as Aspergillus and Nocardiopsis while enriching the material with potential probiotics like Pediococcus and Lactiplantibacillus, reaching an abundance of 95.7%. In the subsequent aerobic SSF stage, the ensiled CKK underwent inoculation with the white-rot fungus Irpex lacteus F17, which became enriched to 87.9%. Comprehensive multi-omics analysis identified Irpex as the key taxon, possessing an extensive redox enzyme system that led to the improvement in nutrient composition, reduction of astringent phenolic substances, and mitigation of mycotoxins. As a result, the crude protein content of the CKK increased by 39.2%, while lignin, total phenolic substances, and tannic acid content decreased by 24.4%, 52.2%, and 51.4%, respectively. The mycotoxin levels, including aflatoxin B1, zearalenone, and vomitoxin, were rendered negligible, confirming the safety. Overall, this study demonstrates the TBA strategy can successfully transform challenging and unpalatable CKK waste into a nutrient-enriched and safe mycelium-based bioproduct, thereby enabling the valorization of a previously underutilized forestry resource as a promising alternative feed.

Abstract The mucosal epithelium of the head and neck region (including the oral cavity, nasal cavity, pharynx, nasopharynx, and larynx) is the primary site exposed to tobacco smoke, and its presence of nicotinic acetylcholine receptors (nAChRs) has been observed in the mucosal epithelial cells of this area. It remains unclear whether HNSC cells can migrate and invade through nAChR signaling. A model of HNSC cells exposed to nicotine is established. Cell proliferation following nicotine exposure is assessed using the CCK-8 assay, while migration and invasion are evaluated through wound healing and Transwell assays. The effects of CHRNA5 knockdown and overexpression are also investigated. Immunofluorescence staining is used to analyze CHRNA5 expression and localization, and clonogenic assays are performed to measure colony proliferation after CHRNA5 knockdown and overexpression. The interaction between CHRNA5 and CES1 is examined using molecular docking, co-immunoprecipitation, and immunofluorescence. Differentially expressed genes are subjected to pathway enrichment analysis, and MEK/ERK protein expression and phosphorylation are validated via western blot. Tumor formation assays are performed in nude mice using sh-CHRNA5 Cal27 cells, followed by western blot and immunohistochemical staining. Additionally, laryngeal and hypopharyngeal cancer tissues are analyzed through immunohistochemistry. Nicotine significantly enhanced the proliferation, migration, and invasion capabilities of head and neck tumor cells, including Cal27, Fadu, HN6, and Tu686 cells, through the expression of CHRNA5. Knockdown of CHRNA5 can reduce cell migration, invasion, and proliferation, whereas nicotine exposure can reverse this trend. Additionally, the mRNA and protein expression of CES1 decreases with the knockdown of CHRNA5, indicating a regulatory relationship between the two. Transcriptomics revealed that the knockdown of CHRNA5 is associated with the MEK/ERK signaling pathway. Further cellular- and tissue-level evidence confirmed that the levels of p-MEK/MEK, p-ERK/ERK, and CES1 decreased following knockdown of CHRNA5, a trend that nicotine can reverse. Nicotine promotes the proliferation, migration, and invasion of HNSC by upregulating CHRNA5 expression. Knockdown of CHRNA5 reduces these effects, which can be reversed by nicotine. Nicotine exposure activates CHRNA5, regulating CES1 expression via the MEK/ERK pathway, contributing to the recurrence and metastasis of head and neck squamous carcinoma.

Hearing loss, a prevalent sensory impairment, poses significant challenges worldwide. Recent research has shed light on the intricate interplay between ferroptosis, a newly recognized form of regulated cell death characterized by iron-dependent lipid peroxidation, and oxidative stress in the pathogenesis of hearing loss. In this review, we delve into the mechanisms underlying ferroptosis and oxidative stress in various forms of hearing loss, including age-related hearing loss (ARHL), noise-induced hearing loss (NIHL) ototoxic drug-induced hearing loss and genetic hearing loss. We discuss the pivotal role of molecules such as FSP1, ACSL4, LKB1-AMPK, and Nrf2 in modulating these pathways in hearing loss. Furthermore, we explore emerging therapeutic strategies targeting the antioxidant system and ferroptosis, including iron chelators, lipid peroxide inhibitors, and antioxidants, highlighting their potential in mitigating hearing loss progression. By elucidating the molecular mechanisms underlying ferroptosis and oxidative stress, this review offers insights into novel therapeutic avenues for the treatment of hearing loss and underscores the importance of targeting these pathways to preserve auditory function.

Background: Age-related hearing loss (ARHL) — also termed presbycusis — is prevalent among older adults, leading to a range of issues. Although considerable progress in the understanding of ARHL over the decades, available reports lack data from recent years and do not comprehensively reflect the latest advancements and trends. Therefore, our study sought to assess research hotspots and trends in ARHL over the past 5 years to provide the basis for future research. Materials and methods: The Web of Science Core Collection database was searched and screened from January 1, 2019 to October 21, 2023, according to the inclusion criteria. CiteSpace (5.8.R3), VOSviewer (1.6.19), and Microsoft Excel 2019 were employed for bibliometric analysis and visualization. Results: 3084 articles from 92 countries led by the United States and China were included. There has been a steady upward trend in the number of publications from 2019 to 2023. The most productive institutions, authors, and journals are Johns Hopkins University (n = 113), Lin FR (n = 66), and Ear and Hearing (n = 135), respectively. Trend topic analyses revealed that “cochlear synaptopathy” and “dementia” were the predominant foci. Keywords, including “individuals” and “national health”, began to appear. Conclusion: Over the past 5 years, the annual number of publications has increased significantly and will continue to do so. Research on the mechanism of ARHL, represented by “oxidative stress”, is a continuing focus. Emerging topics such as “individual differences” and “national health” may be potential future hotspots in this field.

Non-Saccharomyces yeasts have great potential in improving wine quality, showing personality characteristics, and highlighting the terroir of wine. In this study, we evaluated the impact of simultaneous inoculation with the non-Saccharomyces yeasts Torulaspora delbrueckii or (and) Hanseniaspora uvarum in combination with Saccharomyces cerevisiae (EC1118 or VL3) on the aromatic compounds and sensory quality of Sauvignon blanc wines. The growth of yeast groups in the alcoholic fermentation process was tracked using fluorescence in situ hybridization. The presence of non-Saccharomyces yeast notably impacted the distribution of S. cerevisiae and was related to the species of yeast. The co-fermentation of H. uvarum and S. cerevisiae improved the content of total esters, especially acetate esters. Simultaneous inoculation of T. delbrueckii or (and) H. uvarum significantly increased the content of total terpenes, especially linalool. Similar results were found for some higher alcohols and organic acids. Sensory evaluation showed that the wines mixed fermentation with H. uvarum had significantly tropical fruit aroma characteristics. Citrus and mineral notes, typical aroma characteristics of Sauvignon blanc wine, were enhanced by mixed fermentation strategies with T. delbrueckii or (and) H. uvarum and different S. cerevisiae. Hence, co-fermentation by T. delbrueckii or H. uvarum combined with S. cerevisiae could significantly improve the sensory quality of Sauvignon blanc wine.

Abstract Background While T cell-activating immunotherapies against recurrent head and neck squamous cell carcinoma (HNSCC) have shown impressive results in clinical trials, they are often ineffective in the majority of patients. NK cells are potential targets for immunotherapeutic intervention; however, the setback in monalizumab-based therapy in HNSCC highlights the need for an alternative treatment to enhance their antitumor activity. Methods Single-cell RNA sequencing (scRNA-seq) and TCGA HNSCC datasets were used to identify key molecular alterations in NK cells. Representative HPV-positive ( +) and HPV-negative ( −) HNSCC cell lines and orthotopic mouse models were used to validate the bioinformatic findings. Changes in immune cells were examined by flow cytometry and immunofluorescence. Results Through integration of scRNA-seq data with TCGA data, we found that the impact of IL6/IL6R and CCL2/CCR2 signaling pathways on evasion of immune attack by NK cells is more pronounced in the HPV − HNSCC cohort compared to the HPV + HNSCC cohort. In orthotopic mouse models, blocking IL6 with a neutralizing antibody suppressed HPV − but not HPV + tumors, which was accompanied by increased tumor infiltration and proliferation of CD161+ NK cells. Notably, combining the CCR2 chemokine receptor antagonist RS504393 with IL6 blockade resulted in a more pronounced antitumor effect that was associated with more activated intratumoral NK cells in HPV − HNSCC compared to either agent alone. Conclusions These findings demonstrate that dual blockade of IL6 and CCR2 pathways effectively enhances the antitumor activity of NK cells in HPV-negative HNSCC, providing a novel strategy for treating this type of cancer.

This study proposes a charging efficiency calculation model based on an equivalent internal resistance framework. A data-driven neural network model is developed to predict the charging efficiency of lithium titanate (LTO) batteries for 5% state of charge (SOC) segments under various charging conditions. By considering the impact of entropy change on the open-circuit voltage (OCV) during the charging process, the accuracy of energy efficiency calculations is improved. Incorporating battery data under various charging conditions, and comparing the predictive accuracy and computational complexity of different hyperparameter configurations, we establish a backpropagation neural network model designed for implementation in embedded systems. The model predicts the energy efficiency of subsequent 5% SOC segments based on the current SOC and operating conditions. The results indicate that the model achieves a prediction error of only 0.29% under unknown charging conditions while also facilitating the deployment of the neural network model in embedded systems. In future applications, the relevant predictive data can be transmitted in real time to the cooling system for thermal generation forecasting and predictive control of battery systems, thereby enhancing temperature control precision and improving cooling system efficiency.

This research explored the effect of water-binder ratio (w/b) and curing age on the mechanical properties, microstructure, and damage mechanism of modified recycled aggregate concrete (RAC) with metakaolin (MK). The consequence appeared that the mechanical properties of the specimens were positively correlated with curing age, but negatively correlated with w/b. In addition, the incorporation of MK increased the peak stress and peak strain of the specimens with w/b of 0.65 by an average of 17.46% and 8.42% at six ages. Through microscopic test, it was found that the mechanical properties of the specimen were related to the hydration reaction, and the continuously generated gel products made the matrix structure more compact. Based on the statistical damage theory, the influence of w/b, age and MK on the damage evolution of mesoscopic fracture and yield were quantitatively analyzed, and a correlation between macroscopic mechanical properties and the mesoscopic damage mechanism was established.

Abstract Purpose Stenotrophomonas maltophilia is a Gram-negative pathogen that most commonly causes hospital-acquired infections that can be extremely challenging to treat, contributing to underrecognized mortality throughout the world. The relative benefits of monotherapy as compared to combination therapy in patients diagnosed with S. maltophilia pneumonia, however, have yet to be established. Methods Data from 307 patients diagnosed with S. maltophilia hospital-acquired pneumonia (HAP) across four Chinese teaching hospitals from 2016 to 2022 were retrospectively analyzed. Results Of the analyzed patients, 55.7% (171/307) were administered combination definitive therapy, with a 30-day all-cause mortality rate of 41.0% (126/307). A propensity score weighting analysis revealed that compared with monotherapy, combination definitive therapy was associated with a comparable 30-day mortality risk in the overall patient cohort (OR 1.124, 95% CI 0.707–1.786, P = 0.622), immunocompetent patients (OR 1.349, 95% CI 0.712–2.554, P = 0.359), and patients with APACHE II scores

Abstract A compact microstrip multifunctional reconfigurable filtering power divider (FPD) is proposed based on varactor‐loaded three‐parallel‐coupled line section (TLCS) in this letter. Continuously tunable third‐order filtering power division response with two transmission zeros (TZs) including centre frequency (CF), bandwidth (BW) and power division ration (PDR) is successfully attained by varying the external and internal coupling through loading varactors into a three‐parallel‐coupled line topology. For validation, a prototype is implemented. The results indicate that the CF can be tuned from 0.46 to 0.8 GHz, a tunable 3‐dB BW of 80 to 190 MHz is exhibited with better than 17‐dB return loss and higher than 15‐dB in‐band isolation. In addition to the CF and BW tunability, the power division ratio (PDR) for the presented FPD can also be adjusted from 1:1 to 1:5. Additionally, a compact size is realized with only 0.12λg × 0.16λg.

Background The survival benefit of revascularization of chronic total occlusion (CTO) of the coronary arteries remains a subject of controversy. We measured high sensitivity troponin‐I (hsTn‐I) levels as an estimate of myocardial ischemia in patients with stable coronary artery disease, with the hypothesis that (1) patients with CTO have higher levels of hsTn‐I than patients without CTO, (2) hsTn‐I levels will predict adverse cardiovascular events in patients with CTO, and (3) patients with elevated hsTn‐I levels will have a survival benefit from CTO revascularization. Methods and Results In 428 patients with stable coronary artery disease and CTO undergoing coronary angiography, adverse event rates were investigated. Cox proportional hazards models and Fine and Gray subdistribution hazard models were performed to determine the association between hsTn‐I level and incident event rates in patients with CTO. HsTn‐I levels were higher in patients with compared with those without CTO (median 6.7 versus 5.6 ng/L, P=0.002). An elevated hsTn‐I level was associated with higher adverse event rates (adjusted all‐cause mortality hazard ratio, 1.19 [95% CI, 1.08–1.32]; P=0.030) for every doubling of hsTn‐I level. CTO revascularization was performed in 28.3% of patients. In patients with a high (>median) hsTn‐I level, CTO revascularization was associated with substantially lower all‐cause mortality (adjusted hazard ratio, 0.26 [95% CI, 0.08–0.88]; P=0.030) compared with those who did not undergo revascularization. Abstract Background 9α-hydroxyandrost-4-ene-3,17-dione (9-OHAD) is a significant intermediate for the synthesis of glucocorticoid drugs. However, in the process of phytosterol biotransformation to manufacture 9-OHAD, product degradation, and by-products restrict 9-OHAD output. In this study, to construct a stable and high-yield 9-OHAD producer, we investigated a combined strategy of blocking Δ1‑dehydrogenation and regulating metabolic flux. Results Five 3-Ketosteroid-Δ1-dehydrogenases (KstD) were identified in Mycobacterium fortuitum ATCC 35855. KstD2 showed the highest catalytic activity on 3-ketosteroids, followed by KstD3, KstD1, KstD4, and KstD5, respectively. In particular, KstD2 had a much higher catalytic activity for C9 hydroxylated steroids than for C9 non-hydroxylated steroids, whereas KstD3 showed the opposite characteristics. The deletion of kstDs indicated that KstD2 and KstD3 were the main causes of 9-OHAD degradation. Compared with the wild type M. fortuitum ATCC 35855, MFΔkstD, the five kstDs deficient strain, realized stable accumulation of 9-OHAD, and its yield increased by 42.57%. The knockout of opccr or the overexpression of hsd4A alone could not reduce the metabolic flux of the C22 pathway, while the overexpression of hsd4A based on the knockout of opccr in MFΔkstD could remarkably reduce the contents of 9,21 ‑dihydroxy‑20‑methyl‑pregna‑4‑en‑3‑one (9-OHHP) by-products. The inactivation of FadE28-29 leads to a large accumulation of incomplete side-chain degradation products. Therefore, hsd4A and fadE28-29 were co-expressed in MFΔkstDΔopccr successfully eliminating the two by-products. Compared with MFΔkstD, the purity of 9-OHAD improved from 80.24 to 90.14%. Ultimately, 9‑OHAD production reached 12.21 g/L (83.74% molar yield) and the productivity of 9-OHAD was 0.0927 g/L/h from 20 g/L phytosterol. Conclusions KstD2 and KstD3 are the main dehydrogenases that lead to 9-OHAD degradation. Hsd4A and Opccr are key enzymes regulating the metabolic flux of the C19- and C22-pathways. Overexpression of fadE28-29 can reduce the accumulation of incomplete degradation products of the side chains. According to the above findings, the MF-FA5020 transformant was successfully constructed to rapidly and stably accumulate 9-OHAD from phytosterols. These results contribute to the understanding of the diversity and complexity of steroid catabolism regulation in actinobacteria and provide a theoretical basis for further optimizing industrial microbial catalysts.

Huanglongbing (HLB) is a global devastating citrus disease that is mainly caused by “Candidatus Liberibacter asiaticus” (CLas). It is mostly transmitted by the insect Asian citrus psyllid (ACP, Diaphorina citri) in a persistent and proliferative manner. CLas traverses multiple barriers to complete an infection cycle and is likely involved in multiple interactions with D. citri. However, the protein–protein interactions between CLas and D. citri are largely unknown. Here, we report on a vitellogenin-like protein (Vg_VWD) in D. citri that interacts with a CLas flagellum (flaA) protein. We found that Vg_VWD was upregulated in CLas-infected D. citri. Silencing of Vg_VWD in D. citri via RNAi silencing significantly increased the CLas titer, suggesting that Vg_VWD plays an important role in the CLas–D. citri interaction. Agrobacterium-mediated transient expression assays indicated that Vg_VWD inhibits BAX- and INF1-triggered necrosis and suppresses the callose deposition induced by flaA in Nicotiana benthamiana. These findings provide new insights into the molecular interaction between CLas and D. citri.

A simplified analysis method based on three-dimensional finite element analysis is proposed for the dynamic response of pile foundations under the action of vertically propagating SV waves. This method considers the impact of upper structure inertia force and free field deformation on the internal force of the pile separately. The former is considered using the Equivalent Base Shear Method, while the latter is analyzed using a finite element response acceleration method for underground structures. This study selected three seismic waves and their average values as loads to calculate the dynamic response of pile raft foundations. Through trial calculations, the seismic effect reduction coefficient range (0.35–0.4) of the representative values of the horizontal seismic inertia force corresponding to the upper structure was obtained. The error between the peak shear stress of the pile top obtained by the quasi-static method and the time history analysis results was less than 10%. The research results indicate that the proposed simplified analysis method can accurately obtain the peak shear stress and its distribution pattern of the pile body under horizontal seismic action while significantly improving the analysis efficiency. This method has high accuracy and efficiency in conducting seismic design comparison and analysis of multiple foundation schemes.

A statistical damage model (SDM) of fully-graded concrete was created using statistical damage theory, based on the mechanical properties of axial tension and axial compression of the material. The SDM considers two damage modes, fracture and yield, and explains the intrinsic connection between the mesoscopic damage evolution mechanism and the macroscopic nonlinear mechanical behavior of fully-graded concrete. The artificial bee colony (ABC) algorithm was used to obtain the optimal parameter combination through an intelligent search of parameters εa, εh, εb and H in the constitutive model by taking the test data as the target value, and the sum of the squares of the differences between the target value and the predicted value as the objective function. The SDM numerical simulation model of fully-graded concrete is proposed by compiling subroutines in FORTRAN by constructing two modules of data model and damage analysis. The numerical results under uniaxial and biaxial forces are in agreement with the experimental results, which verifies the accuracy of the program. The model also analyzes the characteristics of mesoscopic damage evolution and predicts the mechanical properties under triaxial forces. The results show that the proposed numerical simulation model can reflect the salient features for fully-graded concrete under uniaxial, biaxial and triaxial loading conditions, and the evolution law of mesoscopic parameters. Therefore, the proposed model serves as a basis for the refined finite element analysis of hydraulic fully-graded concrete structures and reveals the mesoscopic damage mechanism of concrete under different load environments.

Stricter control of the differential settlement is always required in important buildings more than in ordinary buildings. It becomes a necessity to find a simple and efficient optimum design method for pile foundations in terms of performance and economy. In this paper, an adaptive optimization method (AOM) is proposed to reduce the differential settlement of the pile group and piled raft, in which the piles are located in appropriate locations according to the settlement characteristic of the raft. Piled raft foundations with different types of load and different raft shapes are optimized using this method; soil inhomogeneity and nonlinear characteristic are considered during this process. The optimization results show that the reductions of the differential settlement are more than 80%. The overall foundation performances are improved as the maximum settlements of the foundations are reduced. The maximum bearing capacity of the pile group is no more than its ultimate bearing capacity after optimization design, and part of the excess bearing capacity can be translated into economic savings (AOM-ES). By keeping a good optimization effect of the differential settlement, the number of piles can be reduced by AOM-ES compared with the initial design. The AOM is robust and can be applied to the piled foundations of various raft shapes in layered soils under complex vertical loads with no significant impact on optimization efficiency.

Handwriting verification represents an important area in forensic science. Herein, handwritings based on different types and brands pen ink samples made in China were investigated by non-invasive micro-identification methods. Scanning electron microscopy and energy spectrum (SEM-ES) were employed to measure the micro elements and component analysis of the written traces, and spectral mapping (surface scanning) function was focused. The exact assessments of elements in handwriting traces can distinguish the microcosmic differences between different types and brands ink in handwriting, which provides a new means of analysis and research methods of pen handwriting detection in forensic science.

The purpose of this paper is to explore the effect of the baseline control system (BCS) on the fragility of large-scale wind turbine when seismic and wind actions are considered simultaneously. The BCS is used to control the power output by regulating rotor speed and blade-pitch angle in real time. In this study, the fragility analysis was performed and compared between two models using different peak ground acceleration, wind speeds, and specified critical levels. The fragility curves with different wind conditions are obtained using the multiple stripe analysis (MSA) method. The calculation results show that the probability of exceedance specified critical level increases as the wind speed increases in model 1 without considering BCS, while does not have an obvious change in the below-rated wind speed range and has a significant decrease in the above-rated wind speed range in model 2 with considering BCS. The comparison depicts that if the BCS is neglected, the fragility of large-scale wind turbine will be underestimated in around the cut-in wind speed range and overestimated in the over-rated wind speed range. It is concluded that the BCS has a great effect on the fragility especially within the operating conditions when the rated wind speed is exceeded, and it should be considered when estimating the fragility of wind turbine subjected to the interaction of seismic and aerodynamic loads.

In order to investigate the effect of a baseline control system (BCS) on dynamic and fatigue characteristics of modern wind turbines, the simulation results of a 5-MW wind turbine subjected to wind loading without and with considering BCS (No. 1 and No. 2, respectively) were compared. The displacement and moment of tower and blades were compared, and the rainflow counting algorithm was used to analyze the fatigue load expressed in damage equivalent load (DEL). The results indicate that the mean and maximum values of structural dynamic responses in No. 2 have a great reduction with wind speed exceeding the rated speed comparing to No. 1; the 3P (3-times the frequency of rotor speed) of the wind turbine rotor speed will be close to the tower fore-aft (FA) natural frequency with the wind velocity around the cut-in speed in No. 2, which caused larger dynamic responses, number of load cycles, and DELs for the tower compared with No. 1; when wind velocity exceeds the rated wind speed, the BCS caused higher fluctuation of loads, increasing the related DELs. It is concluded that when analyzing the dynamic responses and designing wind turbine tower and blades, taking the BCS into account during analysis is significant.

In this paper, the influence of the number of ground motions on fragility analysis of 5 MW wind turbines under the excitation of aerodynamic and seismic loads is investigated to acquire the sufficient number of records for reasonable fragility estimation. Two scenarios, parked and normal operation, were simulated with the number of actual recorded ground motions ranging from 2 to 48. The fragility function parameters are estimated by utilizing the method of multiple stripe analysis (MSA), and the relationship between the number of ground motions and estimated parameters for each scenario is discussed. In addition, the influence of the number of ground motions on the range of estimated parameters with a 95% confidence interval is also discussed. The simulation results show that there are significant differences between the two scenarios in terms of fragility analysis for 5 MW wind turbines with the interaction of wind and seismic loads, and the lesser number of earthquakes is needed for a normal operation scenario compared to the parked scenario when obtaining the same accuracy fragility estimation. Furthermore, the appropriate number of ground motions to accurately estimate the fragility parameters of 5 MW wind turbines for the two scenarios is presented herein, which is almost unaffected by different wind spectra.

Lipid accumulation in various microalgae has been found induced by nitrogen deprivation, and it controls many different genes expression. Yet, the underlying molecular mechanisms still remain largely unknown. MicroRNA (miRNAs) play a critical role in post-transcriptional gene regulation. In this study, miRNAs were hypothesized involved in lipid accumulation by nitrogen deprivation. A deep-sequencing platform was used to explore miRNAs-mediated responses induced by nitrogen deprivation in Chlamydomonas reinhardtii. The eukaryotic orthologous groups of proteins (KOG) function in the predicted target genes of miRNA with response to nitrogen deprivation were mainly involved in signal transduction mechanisms, including transcription, lipid transport, and metabolism. A total of 109 miRNA were predicted, including 79 known miRNA and 30 novel miRNA. A total of 29 miRNAs showed significantly differential expressions after nitrogen deprivation, and most of them were upregulated. A total of 10 miRNAs and their targeting genes might involve in lipid transport and metabolism biological process. This study first investigates nitrogen deprivation-regulated miRNAs in microalgae and broadens perspectives on miRNAs importance in microalgae lipid accumulation via nitrogen deprivation. This study provides theoretical guidance for the application of microalgae in bio-oil engineering production.

Fiber-reinforced concrete (FRC) is widely used in the field of civil engineering. However, the research on the damage mechanism of FRC under uniaxial tension is still insufficient, and most of the constitutive relations are macroscopic phenomenological. The aim is to provide a new method for the investigation of mesoscopic damage mechanism of FRC under uniaxial tension. Based on statistical damage theory, the damage constitutive model for FRC under uniaxial tension is established. Two kinds of mesoscopic damage mechanisms, fracture and yield, are considered, which ultimately determines the macroscopic nonlinear stress–strain behavior of concrete. The yield damage mode reflects the potential bearing capacity of materials and plays a key role in the whole process. Evolutionary factor is introduced to reflect the degree of optimization and adjustment of the stressed skeleton in microstructure. The whole deformation-to-failure is divided into uniform damage phase and local failure phase. It is assumed that the two kinds of damage evolution follow the independent triangular probability distributions, which could be represented by four characteristic parameters. The validity of the proposed model is verified by two sets of test data of steel fiber-reinforced concrete. Through the analysis of the variation law of the above parameters, the influence of fiber content on the initiation and propagation of micro-cracks and the damage evolution of concrete could be evaluated. The relations among physical mechanism, mesoscopic damage mechanism, and macroscopic nonlinear mechanical behavior of FRC are discussed.

Ensiling has been widely applied to cope with agricultural solid waste to achieve organic waste valorization and relieve environmental pressure and feedstuff shortage. In this study, co-ensiling of cabbage leaf byproduct and rice straw was performed with inoculation of Lactiplantibacillusplantarum (LP) to investigate the effects of inoculation on ensiling performance and microflora profiles. Compared to the control, LP inoculation preserved more dry matter (DM) content (283.4 versus 270.9 g·kg−1 fresh matter (FM) on day 30), increased lactic acid (LA) content (52.1 versus 35.8 g·kg−1 dry matter on day 15), decreased pH (3.55 versus 3.79 on day 15), and caused accumulation of acetic acid (AA), butyric acid (BA), and ammonia. The investigation showed that LP inoculation modified microflora composition, especially resisting potential pathogens and enriching more lactic acid bacteria (LAB) (p < 0.05). Moreover, Lactiplantibacillus and Lacticaseibacillus were identified as the keystone taxa that influenced physicochemical properties and interactions in microflora. They were also the main functional species that directly restrained undesirable microorganisms (p < 0.05), rather than indirectly working via metabolite inhibition and substrate competition (p > 0.05). The results of this present study improve the understanding of the underlying effect of LP inoculation on improving silage quality and facilitate the bio-transformation of cabbage byproduct and rice straw as animal feed.

The fermentation of leaf vegetable waste to produce animal feed reduces the environmental impact of vegetable production and transforms leaf vegetable waste into a commodity. We investigated the effect of exogenous probiotics and lignocellulose enzymes on the quality and microbial community of fermented feed (FF) produced from cabbage waste. The addition of exogenous probiotics resulted in increased crude protein (CP) content (p < 0.05), better odor (moderate organic acid and ethanol, with low ammonia-N, p < 0.05), and a lower relative abundance (RA) of pathogens (below 0.4%, p < 0.05) in FF, compared to without. With the addition of exogenous probiotics, only Pediococcus and Saccharomyces were enriched and symbiotic in FF; these were the keystone taxa to reduce the abundance of aerobic, form-biofilms, and pathogenic microorganisms, resulting in an efficient anaerobic fermentation system characterized by facultative anaerobic and Gram-positive bacterial communities, and undefined saprotroph fungal communities. Thus, inoculation of vegetable waste fermentation with exogenous probiotics is a promising strategy to enhance the biotransformation of vegetable waste into animal feed.

A new elasto-plastic constitutive model for jointed rock mass, which can consider the persistence ratio in different visual angle and anisotropic increase of plastic strain, is proposed. The proposed the yield strength criterion, which is anisotropic, is not only related to friction angle and cohesion of jointed rock masses at the visual angle but also related to the intersection angle between the visual angle and the directions of the principal stresses. Some numerical examples are given to analyze and verify the proposed constitutive model. The results show the proposed constitutive model has high precision to calculate displacement, stress and plastic strain and can be applied in engineering analysis.

Generative artificial intelligence (AI) models have made significant progress in automating the creation of 3D shapes, which has the potential to transform car design. In engineering design and optimization, evaluating engineering metrics is crucial. To make generative models performance-aware and enable them to create high-performing designs, surrogate modeling of these metrics is necessary. However, the currently used representations of 3D shapes either require extensive computational resources to learn or suffer from significant information loss, which impairs their effectiveness in surrogate modeling. To address this issue, we propose a new 2D representation of 3D shapes. We develop a surrogate drag model based on this representation to verify its effectiveness in predicting 3D car drag. We construct a diverse dataset of 4535 high-quality 3D car meshes labeled by drag coefficients computed from computational fluid dynamics simulations to train our model. Our experiments demonstrate that our model can accurately and efficiently evaluate drag coefficients with an R2 value above 0.84 for various car categories. Our model is implemented using deep neural networks, making it compatible with recent AI image generation tools (such as stable diffusion) and a significant step toward the automatic generation of drag-optimized car designs. We study the problem of decomposing a polynomial p into a sum of r squares by minimizing a quadratically penalized objective fp(u)= ...2. This objective is nonconvex and is equivalent to the rank-r Burer--Monteiro factorization of a semidefinite program (SDP) encoding the sum of squares decomposition. We show that for all univariate polynomials p, if r\geq 2, then fp(u) has no spurious second-order critical points, showing that all local optima are also global optima. This is in contrast to previous work showing that for general SDPs, in addition to genericity conditions, r has to be roughly the square root of the number of constraints (the degree of p) for there to be no spurious second-order critical points. Our proof uses tools from computational algebraic geometry and can be interpreted as constructing a certificate using the first- and second-order necessary conditions. We also show that by choosing a norm based on sampling equally spaced points on the circle, the gradient ᐁ fp can be computed in nearly linear time using fast Fourier transforms. Experimentally we demonstrate that this method has very fast convergence using first-order optimization algorithms such as L-BFGS, with near-linear scaling to million-degree polynomials. Steroid drug precursors, including C19 and C22 steroids, are crucial to steroid drug synthesis and development. However, C22 steroids are less developed due to the intricacy of the steroid metabolic pathway. In this study, a C22 steroid drug precursor, 9-hydroxy-3-oxo-4,17-pregadiene-20-carboxylic acid methyl ester (9-OH-PDCE), was successfully obtained from Mycolicibacterium neoaurum by 3-ketosteroid-D¹ -dehydrogenase and enoyl-CoA hydratase ChsH deficiency. The production of 9-OH-PDCE was improved by the overexpression of 17β-hydroxysteroid dehydrogenase Hsd4A and acyl-CoA dehydrogenase ChsE1-ChsE2 to reduce the accumulation of by-products. The purity of 9-OHPDCE in fermentation broth was improved from 71.7% to 89.7%. Hence, the molar yield of 9-OH-PDCE was improved from 66.7% to 86.7%, with a yield of 0.78 g/L. Furthermore, enoyl-CoA hydratase ChsH1-ChsH2 was identified to form an indispensable complex in Mycolicibacterium neoaurum DSM 44704. IMPORTANCE C22 steroids are valuable precursors for steroid drug synthesis, but the development of C22 steroids remains unsatisfactory. This study presented a strategy for the one-step bioconversion of phytosterols to a C22 steroid drug precursor, 9-hydroxy-3-oxo-4,17-pregadiene-20-carboxylic acid methyl ester (9-OH-PDCE), by 3- ketosteroid-D1-dehydrogenase and enoyl-CoA hydratase deficiency with overexpression of 17β-hydroxysteroid dehydrogenase acyl-CoA dehydrogenase in Mycolicibacterium. The function of the enoyl-CoA hydratase ChsH in vivo was revealed. Construction of the novel C22 steroid drug precursor producer provided more potential for steroid drug synthesis, and the characterization of the function of ChsH and the transformation of steroids further revealed the steroid metabolic pathway. Extreme conditions will cause the water level of high fill canal segment to change suddenly, which will affect the velocity and pore pressure of the slope. In this paper, numerical method is used to study the influence of water level sudden change on seepage characteristics of high fill canal segment. HyperMesh software is used to establish the finite element model of typical high fill canal segment under complex foundation conditions. Through the combination of secondary development program and fluid-structure coupling calculation method, the fluid structure coupling effect of canal under sudden change of water level is analyzed in ABAQUS. The results show that when the water level changes suddenly, the pore pressure below the free water surface and the velocity near the free surface will be greatly affected. [ABSTRACT FROM AUTHOR]
